BACKGROUND Validation of the reference gene(RG)stability during experimental analyses is essential for correct quantitative real-time polymerase chain reaction(RT-qPCR)data normalisation.Commonly,in an unreliable way,...BACKGROUND Validation of the reference gene(RG)stability during experimental analyses is essential for correct quantitative real-time polymerase chain reaction(RT-qPCR)data normalisation.Commonly,in an unreliable way,several studies use genes involved in essential cellular functions[glyceraldehyde-3-phosphate dehydro-genase(GAPDH),18S rRNA,andβ-actin]without paying attention to whether they are suitable for such experimental conditions or the reason for choosing such genes.Furthermore,such studies use only one gene when Minimum Information for Publication of Quantitative Real-Time PCR Experiments guidelines recom-mend two or more genes.It impacts the credibility of these studies and causes dis-tortions in the gene expression findings.For tissue engineering,the accuracy of gene expression drives the best experimental or therapeutical approaches.We cultivated DPSCs under two conditions:Undifferentiated and osteogenic dif-ferentiation,both for 35 d.We evaluated the gene expression of 10 candidates for RGs[ribosomal protein,large,P0(RPLP0),TATA-binding protein(TBP),GAPDH,actin beta(ACTB),tubulin(TUB),aminolevulinic acid synthase 1(ALAS1),tyro-sine 3-monooxygenase/tryptophan 5-monooxygenase activation protein,zeta(YWHAZ),eukaryotic translational elongation factor 1 alpha(EF1a),succinate dehydrogenase complex,subunit A,flavoprotein(SDHA),and beta-2-micro-globulin(B2M)]every 7 d(1,7,14,21,28,and 35 d)by RT-qPCR.The data were analysed by the four main algorithms,ΔCt method,geNorm,NormFinder,and BestKeeper and ranked by the RefFinder method.We subdivided the samples into eight subgroups.RESULTS All of the data sets from clonogenic and osteogenic samples were analysed using the RefFinder algorithm.The final ranking showed RPLP0/TBP as the two most stable RGs and TUB/B2M as the two least stable RGs.Either theΔCt method or NormFinder analysis showed TBP/RPLP0 as the two most stable genes.However,geNorm analysis showed RPLP0/EF1αin the first place.These algorithms’two least stable RGs were B2M/GAPDH.For BestKeeper,ALAS1 was ranked as the most stable RG,and SDHA as the least stable RG.The pair RPLP0/TBP was detected in most subgroups as the most stable RGs,following the RefFinfer ranking.CONCLUSION For the first time,we show that RPLP0/TBP are the most stable RGs,whereas TUB/B2M are unstable RGs for long-term osteogenic differentiation of human DPSCs in traditional monolayers.展开更多
To implement the multi-way phase shifting maintaining the compact size and simplicity in structure,the uniform reference line concept was proposed for the differential phase shifter.However,the performance in bandwidt...To implement the multi-way phase shifting maintaining the compact size and simplicity in structure,the uniform reference line concept was proposed for the differential phase shifter.However,the performance in bandwidth and phase range deteriorates with the additional requirements considered.To solve this problem,a quarter wavelength coupled line section loaded with open/short stubs is proposed as the basic element to implement the main line and also reference line.According to the theoretical analysis on this basic element,the loading stubs can be used to control the phase shift and also the phase slope of the basic element without affecting the amplitude property.With the predetermined parameters of the uniform reference line,only two parameters are required for the implementation of different differential phase shifts.This demonstrates the high simplicity of the proposed structure.For demonstration,an eight-way differential phase shifter operating at 3.5 GHz was implemented using the vertically installed planar structure.The prototype was further fabricated and measured.Good agreement between simulation and measurement can be observed.The implemented phase shifter can provide a wide range of phase shifting values from 45°to 315°with reference to the uniform reference line over a relative bandwidth of 62.3%.展开更多
Reference evapotranspiration (ETo) is often used to estimate actual evapotranspiration in water balance studies. In this study, the present and future spatial distributions and temporal trends of ETo in the Xiangjia...Reference evapotranspiration (ETo) is often used to estimate actual evapotranspiration in water balance studies. In this study, the present and future spatial distributions and temporal trends of ETo in the Xiangjiang River Basin (XJRB) in China were analyzed. ETo during the period from 1961 to 2010 was calculated with historical meteorological data using the FAO Penman-Monteith (FAO P-M) method, while ETo during the period from 2011 to 2100 was downscaled from the Coupled Model Intercomparison Project Phase 5 (CMIP5) outputs under two emission scenarios, representative concentration pathway 4.5 and representative concentration pathway 8.5 (RCP45 and RCP85), using the statistical downscaling model (SDSM). The spatial distribution and temporal trend of ETo were interpreted with the inverse distance weighted (IDW) method and Mann-Kendall test method, respectively. Results show that: (1) the mean annual ETo of the XJRB is 1 006.3 mm during the period from 1961 to 2010, and the lowest and highest values are found in the northeast and northwest parts due to the high latitude and spatial distribution of climatic factors, respectively; (2) the SDSM performs well in simulating the present ETo and can be used to predict the future ETo in the XJRB; and (3) CMIP5 predicts upward trends in annual ETo under the RCP45 and RCP85 scenarios during the period from 2011 to 2100. Compared with the reference period (1961-1990), ETo increases by 9.8%, 12.6%, and 15.6% under the RCP45 scenario and 10.2%, 19.1%, and 27.3% under the RCP85 scenario during the periods from 2011 to 2040, from 2041 to 2070, and from 2071 to 2100, respectively. The predicted increasing ETo under the RCP85 scenario is greater than that under the RCP45 scenario during the period from 2011 to 2100.展开更多
In order to solve the problems that the novel wide area differential method on the satellite clock and ephemeris relative correction(CERC)in the non-geostationary orbit satellite constellation,a virtual reference sate...In order to solve the problems that the novel wide area differential method on the satellite clock and ephemeris relative correction(CERC)in the non-geostationary orbit satellite constellation,a virtual reference satellite(VRS)differential principle using relative correction of satellite ephemeris errors is proposed.It is referred to be as the VRS differential principle,and the elaboration is focused on the construction of pseudo-range errors of VRS.Through qualitative analysis,it can be found that the impact of the satellite’s clock and ephemeris errors on positioning can basically be removed and the users’positioning errors are near zero.Through simulation analysis of the differential performance,it is verified that the differential method is universal in all kinds of satellite navigation systems with geostationary orbit(GEO)constellation,Medium orbit(MEO)constellation or hybrid orbit constellation,and it has insensitivity to abnormal aspects of a satellite ephemeris and clock.Moreover,the real time positioning accuracy of differential users can be maintained within several decimeters after the pseudo-range measurement noise is effectively weakened or eliminated.展开更多

Against a background of deepened globalization, the socio-spatial distribution characteristics of the employed population are one of the important perspectives that reflect the impact of globalization on urban social ...Against a background of deepened globalization, the socio-spatial distribution characteristics of the employed population are one of the important perspectives that reflect the impact of globalization on urban social space. Based on data of the population census of Shanghai in 2000 and 2010, using location quotient and index of dissimilarity, this paper analyzes the residential differentiation evolution in Shanghai from 2000 to 2010. The results show that except the high-end service industry, the residential differentiation of the employees in the other three types of industries has intensified but is not serious. In addition, the proportion of employees in high-end service industry within the Middle Ring has increased, while that of employees in the middle-and low-end service industry beyond the Middle Ring has increased, which further proves that globalization has an increasingly obvious impact on the socio-spatial differentiation in Shanghai. However, the adjustment in spatial development made by the Shanghai municipal government in response to globalization makes the socio-spatial evolution of Shanghai greatly different from the suburbanization of social elites in the West.展开更多

Compared with the conventional references, the reference with the technique is mainly characterized by a two cascaded stages current mode signal differentiator. In the differentiator, a zero OTA G_m is proposed, to achieve the wide-band differential characteristic. With the technique, the PSRR beyond the pole's corresponding frequency can be significantly improved with the minimum supply voltage only about V_(GS_PMOS) +(V_(GS_NMOS-VTH)). Fabricated with a 0.18 μm CMOS process, with the 0.9 V supply voltage, the PSRR @ 20 MHz of the reference is achieved at-54 dB. Moreover, the power dissipation is 19 μW.展开更多
