Therefore, we reviewed the possibility of introducing an automated analyzer to standardize the Ki-67 evaluation method. Methods: We retrospectively reviewed a clinical database of patients who underwent surgery for early breast cancer at Tokyo-West Tokushukai Hospital. Among them, those who underwent preoperative core needle biopsy (CNB) were enrolled. The concordance rates of estrogen receptor (ER), progesterone receptor (PgR), human epidermal growth factor receptor 2 (HER2), and Ki-67 by local pathologists were reviewed (valuations made by local pathologists), and nonmatching cases (from August 2008 to October 2011) were reassessed both by central review and using an automated analyzer with virtual slides. The results were compared with the evaluations made by local pathologists, and we reexamined the concordance rate by using central review and the automated analyzer. Results: The concordance rate of Ki-67 evaluations made by local pathologists in the preoperative CNB and surgical specimens was 78.7% in 287 cases pathologically assessed from October 2008 to March 2013. This rate was significantly lower (p < 0.01) than that of ER (95.6%), PgR (88.5%), and HER2 (91.6%). Reassessment of the 37 cases of nonmatching Ki-67 values from 2008 to October 2011 using central review and an automated analyzer resulted in clear improvement in matching of 22 (92.1%) and 24 (93.1%) of 37 cases, respectively. Conclusion: The concordance rate of Ki-67 in preoperative CNB and surgical specimens was lower than that of other biological markers;however, they were nearly equal by reassessment using central review and an automated analyzer.展开更多

Based on the theory that sliding force is greater than the shear resistance (resisting force) at the potential slip surface is the necessary and sufficient condition to occur the landslide as the sliding criterion, the principle and method for sliding force remote monitoring is presented, and the functional relationship between the human mechanical quantity and the natural sliding force is derived, hereby, the natural sliding force can be calculated according to the human mechanical quantity. Based on above principle and method, a new system of landslide remote monitoring is designed and 53 systems are installed on the landslide body in the Luoshan mining area, which make up the landslide remote monitoring network. According to the results of field test around 8 months, monitoring curves between sliding force and time are obtained, which can describe and forecast the develop trend of landslide. According to above analysis, the results show that this system has some following advantages: (1) real-time monitoring; (2) remote intelligent transmission; (3) landslides early warning.展开更多

The study on shallow landslide is not only helpful to the local government in disaster prevention, but also the theoretical basis of regional early warning technology. To determine the whole-process characteristics and failure mechanisms of flow-slide failure of granite residual soil slopes, we conducted a detailed hazard investigation in Minqing County, Fujian Province, which was impacted by Typhoon Lupit-induced heavy rainfall in August 2021. Based on the investigation and preliminary analysis results, we conducted indoor artificial rainfall physical model tests and obtained the whole-process characteristics of flow-slide failure of granite residual soil landslides. Under the action of heavy rainfall, a granite residual soil slope experiences initial deformation at the slope toe and exhibits development characteristics of continuous traction deformation toward the middle and upper parts of the slope. The critical volumetric water content during slope failure is approximately 53%. Granite residual soil is in a state of high volumetric water content under heavy rainfall conditions, and the shear strength decreases, resulting in a decrease in stability and finally failure occurrence. The new free face generated after failure constitutes an adverse condition for continued traction deformation and failure. As the soil permeability(cm/h) is less than the rainfall intensity(mm/h), and it is difficult for rainwater to continuously infiltrate in short-term rainfall, the influence depth of heavy rainfall is limited. The load of loose deposits at the slope foot also limits the development of deep deformation and failure. With the continuous effect of heavy rainfall, the surface runoff increases gradually, and the influence mode changes from instability failure caused by rainfall infiltration to erosion and scouring of surface runoff on slope surface. Transportation of loose materials by surface runoff is an important reason for prominent siltation in disaster-prone areas.展开更多

However, there is no unified classification standard for the types of loess landslides in Tianshui. In this study, we explored the landslide distribution and failure characteristics by means of field investigation, remotesensinginterpretation, geological mapping, drilling exploration and shearwave velocity tests, and established a database of Tianshui landslides. Our analysis shows that shear zones in mudstone usually develop in weak intercalated layers. Landslides occur mainly along the West Qinling faults on slopes with gradients of 10° to 25° and on southeast-and southwest-facing slopes. These landslides were classified into five types: loess landslides, loess–mudstone interface landslides, loess flow-slides, loess–mudstone plane landslides and loess–mudstone cutting landslides. We discussed the evolution and failure process of each landslide type and analyzed the formation mechanism and motion characteristics of large-scale landslides. The analysis results show that the landslides in the study area are characterized by a gentle slope, long runout and high risk. The relationship between the runout L and the vertical drop H of the large-scale landslides in the study area is L > 4 H. There are good correlations between the equivalent friction coefficient of largescale landslides and their maximum height, runout, area and volume. The sliding zone of large-scale landslides often develops in the bedrock contact zone or in a weak interlayer within mudstone. From microstructure analysis, undisturbed mudstone consists mainly of small aggregates with dispersed inter-aggregate pores, whereas sheared clay has a more homogeneous structure. Linear striations are well developed on shear surfaces, and the clay pores in those surfaces have a more uniform distribution than those in undisturbed clay.展开更多

They are widely distributed and caused tremendous damages. The sliding mechanism and characteristics of these earthquake-induced landslides are different from those of conventional gravity landslides. Their occurrences are apparently controlled by the powerful earthquake, and they are characterized by high potential energy sliding and ejection sliding. In this article, the earthquake-induced landslides are classified, the characteristics of the sliding and destruction of these landslides are described, and the rela- tionship between the earthquake and the landslides is analyzed. The Donghekou (东河口) landslide is used as an example to characterize fast-moving long-distance earthquake-induced ejection slippage and landslide. This research suggests that many phenomena and issues related to earthquake-induced land- slide are beyond current recognition and knowledge, and new methodologies should be adopted to consider the effect of the earthquake as the paramount factor in the development of landslides. Furthermore, the study of fast-moving long-distance earthquake-induced landslides can also shed light on the formation of old and ancient landslides.展开更多

To accurately understand the distribution of groundwater and its impact on the landslide hazard, a combination of Electrical Resistivity Tomography(ERT), Induced Polarization(IP) and Surface Nuclear Magnetic Resonance(SNMR) methods were used in this study. By conducting a comprehensive analysis, the characteristics of water-bearing structure in vertical and groundwater distribution in horizontal were detected;and the influence of the groundwater on plateau and landslides was also identified. The results indicate that the groundwater occurs in the loess aquifer with a three-layer structure in vertical.Horizontally, the aquifer has a unified water table over the plateau, with a low water level in the north and high one in the south. The high resistivity bedrock uplift belt in the middle of the plateau forms a watershed, with the north side of the uplift belt being a relatively stable slope area with stable water content and fewer geological disasters. In contrast, the south side of the uplift belt is a disaster-prone region with vertical fissures well developed in the loess aquifers. The southern landslides are characterized by the interphase distribution of high and low electrical resistivity. The infiltration and discharge of groundwater result in the formation of a collapse belt in the low resistivity water-bearing structure of landslide, which causes the entire block with high resistivity and stable bedrock to slide. There was a newly formed landslide in a larger range at the landslide' s trailing edge. This study provides a scientific basis for the study of landslides mechanisms and disaster prevention by identifying the distribution characteristics of groundwater and analyzing its influence from a geophysical perspective in Heifangtai.展开更多

Their sliding velocity and runout distance are two major parameters for quantifying and analyzing the risk of submarine landslides.Currently, commercial calculation programs such as BING have limitations in simulating underwater soil movements. All of these processes can be consistently simulated through a smoothed particle hydrodynamics(SPH) depth integrated model. The basis of the model is a control equation that was developed to take into account the effects of soil consolidation and erosion. In this work, the frictional rheological mode has been used to perform a simulation study of submarine landslides. Time-history curves of the sliding body's velocity, height, and length under various conditions of water depth, slope gradient, contact friction coefficient, and erosion rate are compared; the maximum sliding distance and velocity are calculated; and patterns of variation are discussed. The findings of this study can provide a reference for disaster warnings and pipeline route selection.展开更多

Conclusion: The concordance rate of Ki-67 in preoperative CNB and surgical specimens was lower than that of other biological markers;however, they were nearly equal by reassessment using central review and an automated analyzer.

文摘BACKGROUND Histopathologically stained archived tissue slides are stored in hospital archives for years to decades. They are the largest available source of biological materials and are a potentially useful resource that can be used for retrospective epidemiological studies.DNA recovered from the slides can be used for several downstream molecular processes including polymerase chain reaction, single nucleotide polymorphism analysis, and whole genome sequencing. The DNA from these slides can be utilized to compare gene signatures of normal and diseased tissues. However, extraction of high-quality DNA from archived stained hematoxylin and eosin(H&E)slides remains challenging.AIM To standardize a new protocol for extracting DNA from archived H&E-stained tissue slides for further molecular assays.METHODS A total of 100 archived H&E-stained cancer slides were subjected to a total of five methods of DNA extraction. Methods were varied in the deparaffinization step, tissue rehydration, duration of lysis, and presence or absence of proteinase K.The extracted DNA was quantified using a NanoDrop spectrophometer and the quality was analyzed by agarose gel electrophoresis. Then each sample was subjected to polymerase chain reaction(PCR)to amplify the internal control gene GAPDH,thereby confirming the DNA intactness, which could be further utilized for other downstream applications.RESULTS Of the five different methods tested, the third method wherein xylene was used for tissue deparaffinization followed by 72 h of digestion and without proteinase K inactivation yielded the highest amount of DNA with good purity. The yield was significantly higher when compared to other methods. In addition,90%of the extracted DNA showed amplifiable GAPDH gene.CONCLUSION Here we present a step-by-step,cost-effective,and reproducible protocol for the extraction of PCR-friendly DNA from archived H&E-stained cancer tissue slides that can be used for further downstream molecular applications.

文摘With the scale extending of mining, the landslide disaster in the earth’s surface will become more and more serious, and these landslide disasters are being threatened to the sustainable safe mining of the underground mine and the open-pit mine. According to above analysis, the results show that this system has some following advantages: (1) real-time monitoring; (2) remote intelligent transmission; (3) landslides early warning.

文摘Affected by typhoons over years, Fujian Province in Southeast China has developed a large number of shallow landslides, causing a long-term concern for the local government. Transportation of loose materials by surface runoff is an important reason for prominent siltation in disaster-prone areas.

文摘Landslides in Tianshui Basin, Gansu Province, Northwest China, severely affect the local population and the economy;therefore, understanding their evolution and kinematics is of great interest for landslide risk assessment and prevention. Linear striations are well developed on shear surfaces, and the clay pores in those surfaces have a more uniform distribution than those in undisturbed clay.
文摘The great Wenchuan (汶川) earthquake induced a large quantity of landslides. Furthermore, the study of fast-moving long-distance earthquake-induced landslides can also shed light on the formation of old and ancient landslides.

文摘The occurrence of landslides in Heifangtai plateau is primarily caused by the rise in water levels due to irrigation. This study provides a scientific basis for the study of landslides mechanisms and disaster prevention by identifying the distribution characteristics of groundwater and analyzing its influence from a geophysical perspective in Heifangtai.

文摘Effects of heat softening on the initiation of slide surface(shear banding) in clayey slopes during fast deformation were discussed.Controlling equations considering heat, pore pressure and mechanical movement were presented.By perturbation method, the instability condition of localized zone(i.e.criterion for initiation of shear banding) for thermal related soils, such as clayey slope, was obtained. It is shown that slide surface initiates once the thermal-softening effects overcome the strain-hardening effects whether it is adiabatic or not.Without strain hardening effects, strain rate hardening obviously plays a role in initiation of shear band.During initiating process, heat is trapped inside the shear band, which leads rapidly to a pore pressure increase and fast loss of strength. The localized shear strain is concentrated in a narrow zone with a width of several centimeters at most and increases fast. This zone forms the sliding surface.Temperature can increase more than 2?C,pore pressure can increase 160% in about 0.1s inside this zone. These changes cause the fast decrease in friction-coefficient by about 36% over the initial value. That is how shear band initiated and developed in clayey slopes.

文摘Submarine landslides can cause severe damage to marine engineering structures. The findings of this study can provide a reference for disaster warnings and pipeline route selection.
