Effects of Different Nitrogen Application Methods on Enzyme Activities in Leaves at Late Growth Stage of Spring Maize 被引量:2

[Objective] This paper aimed at exploring the effects of different nitrogen application methods on the enzyme activities of leaves at ear position in late growth stage of maize. [Method] By pot experiment, the superoxide dismutase (SOD) activity, peroxidase (POD) activity, malonyl dialdehyde (MDA) content and soluble protein content were determined. [Result] At the filling stage and ripening stage, with the increasing of nitrogen application rate, MDA content gradually decreased, while SOD activity, POD activity and soluble protein content increased. MDA content with two topdressing nitrogen was lower than that with one top dressing nitrogen at the same nitrogen application rate, while SOD activity, POD activity and soluble protein content with two topdressing nitrogen were higher than that with one topdressing nitrogen. [Conclusion] Different nitrogen application methods have relatively significant effects on the MDA content, SOD activity, POD activity and soluble protein content, which is of certain directive significance for preventing spring maize prematuration. 展开更多

[Objective] This paper aims to study a new method of extracting pumpkin polysaccharide from pumpkin. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Conclusion] This study provided basic data for research and application of Pumpkin polysaccharide. 展开更多

The influences of different sampiing methods and sampling parts on detection results were compared. [ Results] The positive rate of standard sampling was 18.18% higher than that of non-stand- ard sampling, and the positive rate of samples collected from cowpea tail was 16.67% higher than that collected from other parts. Microbially induced calcite precipitation(MICP)and Enzyme induced calcite precipitation(EICP)techniques were implemented to reinforce the large-scale calcareous sand in this study.Then a coupled numerical model to predict the biochemical reactions and hydraulic characteristics of MICP and EICP reactions was proposed and verified by physical experiments.Results showed that:This model could describe the variations of bacteria,calcium,calcite,permeability over time reasonably.It is necessary to consider the influence of the calculation domain scale when simulating the convection-diffusionreaction in the multi-process of MICP and EICP reactions.The numerical and experimental values of calcite content are 0.841 g/cm^(3) and 0.861 g/cm^(3) for MICP-reinforced sand,0.263 g/cm^(3) and 0.227 g/cm^(3) for EICP-reinforced sand after 192 h of reaction.The reaction rate k_(rea) is an important parameter to control the calcite content.Accordingly,the permeability coefficient of MICP and EICP reinforced calcareous sand decreases by 32%and 18%.Due to the influence of substance transportation and calcite precipitation,the calcite shows a trend of decreasing firstly and then increasing with the enhancing of the initial permeability coefficient in biochemical reactions.The optimal injecting ratio q11:q12 in this study is 100:300 mL/min.The process for the application of MICP and EICP coupled numerical model is also recommended,which provides reference for engineering projects in ground improvement. 展开更多

[Objective] The technology of using c-amylase and glucoamylase to prepare microporeus potato starch was studied.[ Method ] Taking potato starch as raw materials, starch hydrolysis rate and the oil absorption as a measure of index, the influences of the reaction temperature, two enzymes proportion, the quantity of enzyme, the chroma of substrate, buffer solution pH and reaction time on microporous potato starch were inves- tigated. [ Result] The experimental results showed that the best technological conditions were reaction temperature 45 ℃, enzyme ratio ( glucoamy- lase/α-amylase)6, the quantity of enzyme (amount of enzyme and starch quality than) is 1.0%, the substrate quantity chroma of 0.14 g/ml, buffer solution pH 4, the reaction time 8 h. In such process condition, the oil adsorption rate of hydrolyzed potato starch was as high as 70.2%, starch hydrolytic ratio was 34.16%. [ Condmion] The study provided a basis for the development and utilization of microporous starch. During the course of study, we found that both poly (N-isopropylacrylamide ) (PNIP) and PNIP-Ab (enzyme-labelled antibody)could be adhere tightly to a cellulose acetale-nitrate membrane, and that the retention of PNIP-Ab on the membrane increased over 30-fold when compared with the unconjugated Ab.Thus we used this characteristic to develop a novel immunoassay method-polymer enzyme linked immunoassay method: homogeneous antigen-antibody reaction and heterogeneous separation process. Enzymes play essential roles in the biological processes of sludge treatment.In this article,the ultrasound method to extract enzymes from sludge flocs was presented.Results showed that using ultrasound method at 20 kHz could extract more types of enzymes than that at 40 kHz and ethylenediamine tetraacetic acid(EDTA)methods.The optimum parameters of ultrasound extraction at 20 kHz were duration of 10 min and intensity of 552 W/g TSS.Under the optimum condition,ultrasound could break the cells and extract both the extracellular and a small part of intercellular enzymes.Ultrasound intensity was apparently more susceptive to enzyme extraction than duration,suggesting that the control of intensity during ultrasound extraction was more important than that of duration.The Pearson correlation analysis between enzyme activities and cation contents revealed that the different types of enzymes had distinct cation binding characteristics. 展开更多

A closed form of an analytical expression of concentration in the single-enzyme, single-substrate system for the full range of enzyme activities has been derived. The time dependent analytical solution for substrate, enzyme-substrate complex and product concentrations are presented by solving system of non-linear differential equation. We employ He’s Homotopy perturbation method to solve the coupled non-linear differential equations containing a non-linear term related to basic enzymatic reaction. The time dependent simple analytical expressions for substrate, enzyme-substrate and free enzyme concentrations have been derived in terms of dimensionless reaction diffusion parameters ε, λ1, λ2 and λ3 using perturbation method. The numerical solution of the problem is also reported using SCILAB software program. The analytical results are compared with our numerical results. An excellent agreement with simulation data is noted. The obtained results are valid for the whole solution domain. 展开更多

In this paper, an approximate analytical method to solve the non-linear differential equations in an immobilized enzyme film is presented. Analytical expressions for concentrations of substrate and product have been derived for all values of dimensionless parameter. Dimensionless numbers that can be used to study the effects of enzyme loading, enzymatic gel thickness, and oxidation/ reduction kinetics at the electrode in biosensor/biofuel cell performance were identified. Using the dimensionless numbers identified in this paper, and the plots representing the effects of these dimensionless numbers on concentrations and current in biosensor/biofuel cell are discussed. Analytical results are compared with simulation results and satisfactory agreement is noted. 展开更多

Approximate analytical solutions of non-linear reaction equations containing non-linear terms related to enzymatic reaction mechanism are solved using Homotopy perturbation method. The relevant analytical expression for the substrate and product concentration profiles is discussed in terms of dimensionless reaction diffusion parameters α, β, γE, and γS. Numerical solution is also obtained using Matlab program. Our analytical expression compared with numerical estimation and good agreement is noted. 展开更多

In this paper, mathematical model of Martens and Hall (Analytical chemistry 66, 2763-2770 (1994)) for an immobilized oxidase enzyme electrode is discussed. The model involves the system of non-linear reaction diffusion equations under the steady state conditions. A simple and closed-form of approximate analytical expressions for the concentrations of the immobilization of three enzyme substrates has been derived by solving the system of non-linear reaction diffusion equations using new approach of homotopy perturbation method. Approximate polynomial expression of concentration of substrate, oxygen and oxidized mediator and current was obtained in terms of the Thiele moduli and the small values of parameters B<sub>s</sub>, B<sub>o</sub> and B<sub>m</sub> (normalized surface concentration of substrate, oxygen and oxidized mediator). Furthermore, in this work the numerical simulation of the problem is also reported using Matlab program. An agreement between analytical expressions and numerical results is noted. 展开更多

探讨不同培肥措施对褐土农田养分和冬小麦产量的影响,解析土壤胞外酶和微生物群落的变化特征,为高效提升褐土农田土壤肥力和冬小麦产量提供理论依据.基于2017~2024年连续7a的定位试验,包括5种不同培肥措施:不施肥处理(CK)、常规施肥处理... 探讨不同培肥措施对褐土农田养分和冬小麦产量的影响,解析土壤胞外酶和微生物群落的变化特征,为高效提升褐土农田土壤肥力和冬小麦产量提供理论依据.基于2017~2024年连续7a的定位试验,包括5种不同培肥措施:不施肥处理(CK)、常规施肥处理(NPK)、优化施肥处理(NPKM)、单施有机肥处理(OM)和常规施肥+秸秆直接还田处理(NPKS),利用高通量测序技术和生态网络方法,综合分析了土壤养分含量、胞外酶活性、细菌和真菌群落结构和功能.结果表明:①不同培肥措施(NPK、NPKM、NPKS和OM)均可以提高土壤肥力、小麦产量和胞外酶活性,其中NPKS处理提升效果最为显著.②不同培肥措施改变了土壤细菌群落结构组成,不同培肥措施均提高了变形菌门和拟杆菌门的相对丰度,降低了酸杆菌门的相对丰度;SOC、UE和TP是影响细菌群落组成的重要影响因素.③土壤真菌优势门为子囊菌门、担子菌门和被孢霉门;均提高了子囊菌门和被孢霉门相对丰度;DHA、AP和UE是影响真菌群落组成的重要影响因素.④不同培肥措施增加了土壤细菌群落网络的复杂性,降低了土壤真菌群落网络的复杂性.⑤FAPROTAX细菌功能预测结果表明,不同培肥措施提高了土壤碳氮循环相关功能;FUNGuild真菌功能预测表明,秸秆还田可以抑制土壤病原菌的生长,而施加有机肥则会促进腐生真菌生长.不同培肥措施均可提高土壤肥力和胞外酶活性,增加有益菌群丰度,显著改变土壤微生物群落结构和组成,促进土壤碳氮循环,秸秆还田可以抑制土壤病原菌的生长.有机无机配施(NPKS和NPKM)提升效果优于单施化肥(NPK)和单施有机肥(OM),有机无机配施有利于改善土壤质量,促进碳和氮循环,提高作物产量,从而实现旱作农业的可持续发展. 展开更多

目的:探究生物学方法对档案馆古籍纸张霉菌污染的治理效果。方法:首先从霉菌污染的纸板上提取菌种并进行培养,随后将其沾染到宣纸上,模拟档案馆古籍纸张霉菌污染的环境,制备出多个样品试件。通过霉菌分类鉴定、除霉剂配制和清洗处理,完... 目的:探究生物学方法对档案馆古籍纸张霉菌污染的治理效果。方法:首先从霉菌污染的纸板上提取菌种并进行培养,随后将其沾染到宣纸上,模拟档案馆古籍纸张霉菌污染的环境,制备出多个样品试件。通过霉菌分类鉴定、除霉剂配制和清洗处理,完成试件霉菌污染的治理,并与两种对照组方法的污染治理效果进行对比。结果:采用生物学方法能够准确鉴定出污染菌种为黑曲霉、青霉、木霉和黄曲霉四种。基于此,配制了含有几丁质酶、纤维素酶、葡聚糖酶的清洗溶液,最终将纸张上的霉菌OD均值从1.28、1.4、1.1和1.2,显著降低至0.2以下。结论:生物学方法在治理档案馆古籍纸张霉菌污染治理方面不但可行,而且治理效果相较于现有方法更优。 展开更多

针对桑椹药渣中残留多糖未被充分利用的问题,优化超声辅助复合酶法提取工艺,并探究其体外抗氧化活性。以桑椹药渣为原料,通过单因素试验筛选纤维素酶与果胶酶作为复合酶,优化酶解时间、料液比(g/mL)、酶浓度、pH值、反应温度等参数,结... 针对桑椹药渣中残留多糖未被充分利用的问题,优化超声辅助复合酶法提取工艺,并探究其体外抗氧化活性。以桑椹药渣为原料,通过单因素试验筛选纤维素酶与果胶酶作为复合酶,优化酶解时间、料液比(g/mL)、酶浓度、pH值、反应温度等参数,结合正交试验确定最佳提取工艺;采用苯酚-硫酸法测定多糖含量,并通过DPPH自由基和ABTS+自由基清除实验评价抗氧化活性。结果显示正交试验优化后最佳工艺为pH值4、酶解温度40℃、复合酶浓度1%,多糖得率达443.15 mg/g。体外抗氧化实验表明,桑椹药渣多糖对DPPH自由基和ABTS+自由基清除率最高分别为49.37%(10 mg/mL)和79.39%(5 mg/mL),且清除能力随浓度增加而增强。因此,超声辅助复合酶法可高效提取桑椹药渣多糖,其抗氧化活性显著,为桑椹资源综合利用及中药废弃物高值化开发提供了理论依据。 展开更多

针对甜菜红素摩尔吸光系数ε易受溶剂、温度等因素影响而导致测定结果不准确的问题,开展了甜菜红素含量的分光光度与酶标定量检测方法比对研究。建立了基于动态标准曲线的甜菜红素检测体系,在535 nm波长下分光光度法线性范围覆盖0〜15 mg... 针对甜菜红素摩尔吸光系数ε易受溶剂、温度等因素影响而导致测定结果不准确的问题,开展了甜菜红素含量的分光光度与酶标定量检测方法比对研究。建立了基于动态标准曲线的甜菜红素检测体系,在535 nm波长下分光光度法线性范围覆盖0〜15 mg/L(r^(2)=0.9999),酶标法线性范围覆盖0〜30 mg/L(r^(2)=0.9997);当取样量为0.25 g,提取溶液体积为20 mL,分光光度法检出限为0.0001 mg/g,定量限为0.0004 mg/g;酶标法检出限为0.0080 mg/g,定量限为0.0319 mg/g;分光光度法重复性RSD为0.029%,再现性RSD为4.341%,加标回收率为98.45%,酶标法重复性RSD为0.7832%,再现性RSD为5.092%,加标回收率为102.6%,均符合国家相关标准法规对准确度(95%≤回收率≤105%)和精密度(3.8%≤RSD≤5.3%)的要求,为甜菜红素定量提供了可靠的技术方案。 展开更多

2024—2025年西宁市生鲜果蔬农药残留快速检测分析

目的 调查研究2024—2025年西宁市生鲜果蔬农产品的农药残留情况。方法 本研究采用酶抑制法对西宁市某农产品交易市场和产地采购的样品, 185个生鲜蔬菜和125个生鲜水果,其中生鲜蔬菜13464批次,生鲜水果5712批次,共计310个品种19176批次... 目的 调查研究2024—2025年西宁市生鲜果蔬农产品的农药残留情况。方法 本研究采用酶抑制法对西宁市某农产品交易市场和产地采购的样品, 185个生鲜蔬菜和125个生鲜水果,其中生鲜蔬菜13464批次,生鲜水果5712批次,共计310个品种19176批次样品,进行有机磷和氨基甲酸酯类农药残留快速检测与分析。结果 共有30个蔬菜品种、7个水果品种, 60批次样品检测出农药残留超标。其中,超标率前5的果蔬是黄瓜、青笋、香笋、旱瓜和线茄。农药残留超标样品的检测值在51.30%~99.89%之间,总体定性初筛阳性率为0.33%,合格率在98.92%~99.94%之间,整体样品合格率为99.67%,合格率较高。结论 2024—2025年西宁地区生鲜果蔬农产品中农药残留情况处于相对安全的水平,但仍存在农药残留超标的情况。建议持续加强检测与监测,进一步保障市民的食品安全问题。 展开更多
