A reduced model is proposed and analyzed for the simulation of vortexinduced vibrations (VIVs) for turbine blades. A rotating blade is modelled as a uniform cantilever beam, while a van der Pol oscillator is used to represent the time-varying characteristics of the vortex shedding, which interacts with the equations of motion for the blade to simulate the fluid-structure interaction. The action for the structural motion on the fluid is considered as a linear inertia coupling. The nonlinear characteristics for the dynamic responses are investigated with the multiple scale method, and the modulation equations are derived. The transition set consisting of the bifurcation set and the hystere- sis set is constructed by the singularity theory and the effects of the system parameters, such as the van der Pol damping. The coupling parameter on the equilibrium solutions is analyzed. The frequency-response curves are obtained, and the stabilities are determined by the Routh-Hurwitz criterion. The phenomena including the saddle-node and Hopf bifurcations are found to occur under certain parameter values. A direct numerical method is used to analyze the dynamic characteristics for the original system and verify the va- lidity of the multiple scale method. The results indicate that the new coupled model is useful in explaining the rich dynamic response characteristics such as possible bifurcation phenomena in the VIVs. 展开更多

To control the jumping phenomena and the unstable region of the nonlinear oscillator, feedback controllers were designed. Bifurcation control equations were obtained by using the multiple scales method. And through the numerical analysis, good controller could be obtained by changing the feedback control gain. Then a feasible way of further research of saddle-node bifurcation was provided. Finally, an example shows that the feedback control method applied to the hanging bridge system of gas turbine is doable. 展开更多

The study of dynamical behavior of water or gas flows in broken rock is a basic research topic among a series of key projects about stability control of the surrounding rocks in mines and the prevention of some disasters such as water inrush or gas outburst and the protection of the groundwater resource. It is of great theoretical and engineering importance in respect of promo- tion of security in mine production and sustainable development of the coal industry. According to the non-Darcy property of seepage flow in broken rock dynamic equations of non-Darcy and non-steady flows in broken rock are established. By dimensionless transformation, the solution diagram of steady-states satisfying the given boundary conditions is obtained. By numerical analysis of low relaxation iteration, the dynamic responses corresponding to the different flow parameters have been obtained. The stability analysis of the steady-states indicate that a saddle-node bifurcaton exists in the seepage flow system of broken rock. Consequently, using catastrophe theory, the fold catastrophe model of seepage flow instability has been obtained. As a result, the bifurcation curves of the seepage flow systems with different control parameters are presented and the standard potential function is also given with respect to the generalized state variable for the fold catastrophe of a dynamic system of seepage flow in broken rock. 展开更多

非小细胞肺癌组织中SETDB1、KDM1A、CMTM6表达水平及临床意义

目的探讨非小细胞肺癌(NSCLC)组织中SET结构域分支型组蛋白赖氨酸甲基转移酶1(SETDB1)、赖氨酸特异性组蛋白去甲基化酶1(KDM1A)、趋化因子样因子样MARVEL跨膜结构域含蛋白6(CMTM6)表达与临床病理特征及与预后的关系。方法选择2021年8月... 目的探讨非小细胞肺癌(NSCLC)组织中SET结构域分支型组蛋白赖氨酸甲基转移酶1(SETDB1)、赖氨酸特异性组蛋白去甲基化酶1(KDM1A)、趋化因子样因子样MARVEL跨膜结构域含蛋白6(CMTM6)表达与临床病理特征及与预后的关系。方法选择2021年8月至2022年8月安徽医科大学第一附属医院胸外科收治的102例NSCLC患者。收集癌组织及癌旁组织标本,采用免疫组化法检测癌组织及癌旁组织SETDB1、KDM1A、CMTM6的表达;根据预后情况将患者分为生存组(n=78)和死亡组(n=24),多因素Cox回归分析影响NSCLC患者发生死亡的影响因素,ROC分析SETDB1、KDM1A、CMTM6对NSCLC的诊断价值,Kaplan-Meier法分析SETDB1、KDM1A、CMTM6表达与患者预后的关系。结果NSCLC癌组织SETDB1、KDM1A、CMTM6阳性表达率较癌旁组织均升高(P<0.05);与组织中高分化、TNM分期Ⅰ-Ⅱ期、无淋巴结转移的患者比较,组织低分化、TNM分期Ⅲ-Ⅳ期、有淋巴结转移的患者癌组织中SETDB1、KDM1A、CMTM6阳性表达率均升高(P<0.05);SETDB1、KDM1A、CMTM6三者联合对NSCLC有较高的诊断价值。SETDB1、KDM1A、CMTM6是NSCLC患者发生死亡的危险因素(P<0.05)。SETDB1阳性表达患者生存率低于SETDB1阴性表达患者(P=0.025),KDM1A阳性表达患者生存率低于KDM1A阴性表达患者(P=0.044),CMTM6阳性表达患者生存率低于CMTM6阴性表达患者(P=0.008)。结论NSCLC患者癌组织中SETDB1、KDM1A、CMTM6阳性表达率升高,三者的表达水平与组织分化程度、TNM分期、淋巴结转移以及生存率有关。 展开更多

Due to the increasing use of passive absorbers to control unwanted vibrations,many studies have been done on energy absorbers ideally,but the lack of studies of real environmental conditions on these absorbers is felt.The present work investigates the effect of viscoelasticity on the stability and bifurcations of a system attached to a nonlinear energy sink(NES).In this paper,the Burgers model is assumed for the viscoelasticity in an NES,and a linear oscillator system is considered for investigating the instabilities and bifurcations.The equations of motion of the coupled system are solved by using the harmonic balance and pseudo-arc-length continuation methods.The results show that the viscoelasticity affects the frequency intervals of the Hopf and saddle-node branches,and by increasing the stiffness parameters of the viscoelasticity,the conditions of these branches occur in larger ranges of the external force amplitudes,and also reduce the frequency range of the branches.In addition,increasing the viscoelastic damping parameter has the potential to completely eliminate the instability of the system and gradually reduce the amplitude of the jump phenomenon. 展开更多

Relying upon some substitution, a parametrically constrained bifurcation is transformed to an unconstrained bifurcation about new variables. A general form of transition sets of the parametrically constrained bifurcation is derived. The result indicates that only the constrained bifurcation set is influenced by parametric constraints, while other transition sets are the same as those of the corresponding nonparametrically constrained bifurcation. Taking parametrically constrained pitchfork bifurcation problems as examples, effects of parametric constraints on bifurcation classification are discussed. 展开更多

In this paper, we examine a discrete-time Host-Parasitoid model which is a non-dimensionalized Nicholson and Bailey model. Phase portraits are drawn for different ranges of parameters and display the complicated dynamics of this system. We conduct the bifurcation analysis with respect to intrinsic growth rate <em>r</em> and searching efficiency <em>a</em>. Many forms of complex dynamics such as chaos, periodic windows are observed. Transition route to chaos dynamics is established via period-doubling bifurcations. Conditions of occurrence of the period-doubling, Neimark-Sacker and saddle-node bifurcations are analyzed for <em>b≠a</em> where <em>a,b</em> are searching efficiency. We study stable and unstable manifolds for different equilibrium points and coexistence of different attractors for this non-dimensionalize system. Both the period-doubling bifurcation and saddle-node bifurcation of periodical solutions are computed since the observed floquet multiplier overpass the unit circle by DDE-Biftool software in Matlab. The continuation of saddle-node bifurcation line or period-doubling curve is carried out as varying free parameters and time delays. 为了解脂肪组织中SET结构域分叉组蛋白赖氨酸甲基转移酶1(SET domain bifurcated 1,SETDB1)的主要生理生化功能,以野生小鼠(Setdb1F/F小鼠)为对照,检测了脂肪组织特异性敲除Setdb1小鼠(Fabp4-Setdb1^(-/-)小鼠)的甘油三酯(triglyceride,... 为了解脂肪组织中SET结构域分叉组蛋白赖氨酸甲基转移酶1(SET domain bifurcated 1,SETDB1)的主要生理生化功能,以野生小鼠(Setdb1F/F小鼠)为对照,检测了脂肪组织特异性敲除Setdb1小鼠(Fabp4-Setdb1^(-/-)小鼠)的甘油三酯(triglyceride, TG)含量、脂肪组织和生殖器官的形态以及生殖组织VASA(DEAD-box helicase 4, DDX4)蛋白表达。结果表明:Fabp4-Setdb1^(-/-)小鼠脂肪组织SETDB1蛋白表达减少;Fabp4-Setdb1^(-/-)雄鼠的肝脏和血清中TG含量无显著变化,Fabp4-Setdb1^(-/-)雌鼠血清TG含量显著低于Setdb1F/F雌鼠(P<0.05);而Fabp4-Setdb1^(-/-)小鼠体内脂肪沉积增多,且脂肪细胞单位面积显著增加(P<0.001)。另外,Fabp4-Setdbl^(-/-)雄鼠睾丸生精小管的管壁薄,生精细胞间隙大,VASA表达下降;Fabp4-Setdb1^(-/-)雌鼠卵巢内的卵泡数量减少。研究结果提示脂肪组织特异性敲除Setdb1可明显改变小鼠生殖器官结构和功能。 展开更多

目的探讨组蛋白赖氨酸N端甲基转移酶SET结构域分支型1(SETDB1)对人肾癌细胞增殖、凋亡的影响。方法体外培养肾癌ACHN细胞及正常肾小管上皮HK-2细胞,qRT-PCR及Western blot分别检测两种细胞系中SETDB1 mRNA及蛋白的表达。采用转染SETDB1... 目的探讨组蛋白赖氨酸N端甲基转移酶SET结构域分支型1(SETDB1)对人肾癌细胞增殖、凋亡的影响。方法体外培养肾癌ACHN细胞及正常肾小管上皮HK-2细胞,qRT-PCR及Western blot分别检测两种细胞系中SETDB1 mRNA及蛋白的表达。采用转染SETDB1特异性干扰性小核糖核酸(siRNA)敲低ACHN细胞中SETDB1表达,并采用实时定量聚合酶链反应(qRT-PCR)、Western blot检测SETDB1的敲低效果,CCK-8法、溴脱氧尿嘧啶核苷(Brd U)结合试验检测细胞活力与增殖,流式细胞仪、caspase3/7活性分析检测细胞凋亡的变化。采用qRTPCR、Western blot检测沉默SETDB1后p53 mRNA及蛋白的表达水平。结果 SETDB1在肾癌ACHN细胞中表达水平明显高于正常肾小管上皮HK-2细胞(t=14.435,P<0.01);转染SETDB1特异性siRNA沉默SETDB1表达后,ACHN细胞中SETDB1 mRNA(t=13.091,P<0.01)及其蛋白表达水平明显降低;ACHN细胞活力显著降低(P<0.05);细胞内DNA复制活性明显降低(P<0.01);细胞凋亡率增加(t=6.765,P<0.01);Csapase3/7酶活性提高(t=5.763,P<0.01);同时p53 mRNA(t=9.241,P<0.01)及蛋白的表达水平显著提高。结论 SETDB1在肾癌细胞中表达升高,下调SETDB1能够通过促进p53表达而发挥抗肿瘤生长作用。 展开更多

目的分析SET结构域分叉组蛋白赖氨酸甲基转移酶2(SETDB2)、半乳糖凝集素3(Gal-3)、pleckstrin同源结构域S1(PLEKHS1)在甲状腺乳头状癌组织中的表达情况,以及三者对患者预后的预测价值。方法(1)选择118例甲状腺乳头状癌患者作为研究组,4... 目的分析SET结构域分叉组蛋白赖氨酸甲基转移酶2(SETDB2)、半乳糖凝集素3(Gal-3)、pleckstrin同源结构域S1(PLEKHS1)在甲状腺乳头状癌组织中的表达情况,以及三者对患者预后的预测价值。方法(1)选择118例甲状腺乳头状癌患者作为研究组,45例甲状腺良性结节患者作为对照组。采用免疫组化法检测甲状腺乳头状癌组织和甲状腺良性结节组织中SETDB2、Gal-3、PLEKHS1蛋白表达情况。比较研究组与对照组之间,以及不同特征甲状腺乳头状癌患者之间病灶组织中SETDB2、Gal-3、PLEKHS1蛋白的阳性表达率。(2)随访1年,记录甲状腺乳头状癌患者的生存情况。分析SETDB2、Gal-3、PLEKHS1蛋白阳性表达和阴性表达患者的生存率;比较预后良好和预后不良患者SETDB2、Gal-3、PLEKHS1蛋白的阳性表达率。采用受试者工作特征(ROC)曲线分析SETDB2、Gal-3、PLEKHS1对甲状腺乳头状癌患者预后的预测价值。结果(1)与对照组相比,研究组病灶组织中SETDB2蛋白阳性表达率较低,Gal-3、PLEKHS1蛋白阳性表达率较高(P<0.05)。在甲状腺乳头状癌患者中,Ⅲ~Ⅳ期、浸润深度≥1/2、组织中低分化、淋巴结转移、肿瘤直径>1 cm的患者病灶组织中SETDB2蛋白阳性表达率更低,Gal-3、PLEKHS1蛋白阳性表达率更高(P<0.05)。(2)在甲状腺乳头状癌患者中,SETDB2蛋白阳性表达患者生存率高于SETDB2蛋白阴性表达患者,Gal-3、PLEKHS1蛋白阳性表达患者生存率低于Gal-3、PLEKHS1蛋白阴性表达患者(P<0.05);预后良好患者的SETDB2蛋白阳性表达率高于预后不良患者,Gal-3、PLEKHS1蛋白阳性表达率低于预后不良患者(P<0.05)。ROC曲线分析结果显示,SETDB2、Gal-3、PLEKHS1蛋白总评分预测甲状腺乳头状癌患者预后不良的曲线下面积分别为0.826、0.841、0.859。结论在甲状腺乳头状癌中,SETDB2呈低表达,而Gal-3、PLEKHS1呈高表达,三者均与患者的病情严重程度相关,且对预后具有一定的预测价值。 展开更多

目的:探讨KRAS基因突变与SETDB1(set domain bifurcated 1)蛋白表达在结直肠癌中的临床意义及两者间的关系。方法:收集2017年1月至2017年12月于西南医科大学附属医院住院接受诊治的122例结直肠癌患者的组织标本及病例资料,采用二代测序... 目的:探讨KRAS基因突变与SETDB1(set domain bifurcated 1)蛋白表达在结直肠癌中的临床意义及两者间的关系。方法:收集2017年1月至2017年12月于西南医科大学附属医院住院接受诊治的122例结直肠癌患者的组织标本及病例资料,采用二代测序技术(NGS)检测KRAS基因突变情况,免疫组织化学染色检测SETDB1蛋白的表达情况。结果:122例结直肠癌组织中KRAS基因突变率为41.8%,KRAS基因突变状态与肿瘤部位、术前血清CEA水平相关(P<0.05),与年龄、性别、分化程度、肿瘤大小、pT-NM分期、淋巴结转移等无关(P>0.05)。KRAS基因突变状态与SETDB1蛋白表达呈正相关(r=0.232,P=0.008)。SETDB1蛋白水平与肿瘤分化程度、大小及术前血清CEA水平相关(P<0.05),与年龄、性别、肿瘤部位、pTNM分期、淋巴结转移等无关(P>0.05)。结论:SETDB1蛋白在KRAS突变型结直肠癌组织中呈现高表达,KRAS基因突变可能影响SETDB1蛋白表达。 展开更多

蜂斗菜素调控circSETDB1/miR-345-5p对肝癌细胞生物学行为的影响

目的本研究旨在评估蜂斗菜素对肝细胞癌细胞系2(hepatocellular carcinoma cell line 2,HepG2)的抗肿瘤活性,并探讨蜂斗菜素通过调控环状SET结构域蛋白1(circular RNA SET domain bifurcated histone lysine methyltransferase 1,circSE... 目的本研究旨在评估蜂斗菜素对肝细胞癌细胞系2(hepatocellular carcinoma cell line 2,HepG2)的抗肿瘤活性,并探讨蜂斗菜素通过调控环状SET结构域蛋白1(circular RNA SET domain bifurcated histone lysine methyltransferase 1,circSETDB1)和微小RNA-345-5p(microRNA-345-5p,miR-345-5p)的mRNA表达对细胞增殖、凋亡和迁移的影响。方法将HepG2细胞分为多个处理组。对照组不做任何处理;蜂斗菜素处理包括蜂斗菜素L组(25 mol/L)、蜂斗菜素M组(50 mol/L)、蜂斗菜素H组(100 mol/L)、蜂斗菜素组(100 mol/L);siRNA干扰包含环状SET结构域蛋白1小干扰RNA(small interfering RNA targeting circular SET domain bifurcated histone lysine methyltransferase 1,si-circSETDB1)组、小干扰RNA阴性对照(small interfering RNA negative control,si-NC)组;过表达环状SET结构域蛋白1过表达质粒(overexpression vector for circular SET domain bifurcated histone lysine methyltransferase 1,pcDNA-circSETDB1)组和蜂斗菜素+pcDNA-circSETDB1组。采用qRT-PCR检测肝癌组织及癌旁组织的circSETDB1和miR-345-5p的mRNA表达水平;CCK-8检测细胞增殖活性;流式细胞术检测细胞凋亡情况;Western blot检测半胱氨酸天冬氨酸特异性蛋白酶-3(cleaved cysteinyl aspartate-specific proteinase-3,Cleaved-Caspase-3)蛋白表达水平;克隆形成实验检测细胞集落形成;划痕实验和Transwell实验分别检测细胞愈合率和迁移;双荧光素酶报告基因实验验证circSETDB1与miR-345-5p的靶向关系。结果与癌旁组织相比,肝癌组织中circSETDB1 mRNA表达升高(P<0.05),miR-345-5p mRNA表达降低(P<0.05)。与对照组相比,蜂斗菜素各剂量组HepG2细胞的细胞活性、集落形成数降低(P<0.05),细胞凋亡率和Cleaved-Caspase-3蛋白表达升高(P<0.05),且呈剂量依赖性(P<0.05)。与si-NC组相比,si-circSETDB1组HepG2细胞的circSETDB1表达、细胞活性、集落形成数、迁移细胞数和划痕愈合率降低(P<0.05),miR-345-5p表达、细胞凋亡率和Cleaved-Caspase-3蛋白表达升高(P<0.05)。与蜂斗菜素组相比,蜂斗菜素+pcDNA-circSETDB1组HepG2细胞中circSETDB1表达、细胞活性、集落形成数、迁移细胞数和划痕愈合率升高(P<0.05),miR-345-5p表达、细胞凋亡率和Cleaved-Caspase-3蛋白表达降低(P<0.05)。结论蜂斗菜素可抑制肝癌细胞HepG2增殖和迁移,并诱导细胞凋亡,其机制可能与抑制circSETDB1表达而上调miR-345-5p表达有关。 展开更多

为实现流屑角突变现象的节能利用,设计完成180组6061铝合金工件外圆车削实验,获得180组流屑角突变现象的平衡点坐标数据。应用突变理论将实际状态参数(流屑角)到正则状态参数(突变正则模型中的状态参数)之间的映射函数假设为系数待定的... 为实现流屑角突变现象的节能利用,设计完成180组6061铝合金工件外圆车削实验,获得180组流屑角突变现象的平衡点坐标数据。应用突变理论将实际状态参数(流屑角)到正则状态参数(突变正则模型中的状态参数)之间的映射函数假设为系数待定的线性多项式,将实际控制参数(进给量和背吃刀量)到正则控制参数(突变正则模型中的控制参数)之间的映射函数分别假设为系数待定的线性多项式、二次多项式和指数函数;利用该组映射函数将实测平衡点坐标转换为正则平衡点坐标,并代入尖点型正则突变平衡曲面方程左边的多项式,计算出多项式的值;以多项式值的平方和最小作为目标函数,优化出全部假定映射函数的系数,并最终确定采用所获得的与线性多项式假设对应的平衡曲面方程和分叉集方程作为流屑角突变的平衡曲面实验方程和分叉集实验方程。验证实验显示,所建方程的预测精度优于前人建立的理论方程。 展开更多
