BACKGROUND In patients with colorectal cancer(CRC),tumour metastasis is the leading cause of death.The search for key genes involved in metastasis of CRC is imperative for improved prognoses and treatments.SPDL1 has been implicated in the deve-lopment of CRC,however,its mechanism of action remains unclear.AIM To investigate the role and mechanism of action by which SPDL1 inhibits the development and metastasis of CRC.METHODS In this study,we examined the relationship between SPDL1 expression and CRC prognosis using immunohistochemistry.Survival analyses were performed using Kaplan-Meier analysis and log-rank test.After knocking down SPDL1 in the HCT116 cancer cell line changes in cell viability,migration,invasion,and gene expression were examined using a cell counting kit 8 assay,Transwell assay,and Western blot.The effect of SPDL1 on the cell cycle was assessed using flow cy-tometry.RNA sequencing was used to analyse the effect of SPDL1 on gene expression of CRC cells.The mechanism of action of SPDL1 in CRC was further clarified using U0126,an inhibitor of the mitogen-activated protein kinase signaling pathway.RESULTS SPDL1 is expressed at low levels in tissues of patients with CRC,and this reduced expression is associated with poor prognosis.Functionally,low expression of SPDL1 in CRC promotes cell proliferation,migration,invasion,and affects the cell cycle.Mechanistically,SPDL1 affects the progression of CRC through its regulation of the process of epithelial-mesenchymal transition(EMT)and of the epidermal growth factor receptor(EGFR)/extracellular signal-regulated kinase(ERK)signaling pathways.CONCLUSION This study showed that the loss of SPDL1 may induce EMT and promote cell migration and invasion in CRC through the EGFR/ERK pathway. 展开更多

血清sPDL1水平和外周血NLR在判断晚期胆管癌患者生存预后中的意义 被引量:3

目的探究血清可溶性细胞程序性死亡配体1(soluble programmed death-ligand 1,sPDL1)水平和中性粒与淋巴细胞比值(neutrophil-to-lymphocyte ratio,NLR)在晚期胆管癌(biliary tract cancer,BTC)患者中生存预后的意义。方法 2017年2月~2... 目的探究血清可溶性细胞程序性死亡配体1(soluble programmed death-ligand 1,sPDL1)水平和中性粒与淋巴细胞比值(neutrophil-to-lymphocyte ratio,NLR)在晚期胆管癌(biliary tract cancer,BTC)患者中生存预后的意义。方法 2017年2月~2019年3月收集西安市第三医院144例晚期BTC且进行姑息化疗的患者临床资料,包括sPDL1水平,NLR,血小板与淋巴细胞比值(platelet-tolymphocyte ratio,PLR)以及全身免疫炎症指数(systemic immune-inflammation index, SII)。采用单纯随机抽样方法验证每项标志物的分界值,将患者分为发展组与稳定组。比较不同组患者的sPDL1水平及总生存时间(overall survival,OS),分析影响患者生存预后的因素。结果所有患者OS为9.12个月(95%CI 8.24~11.41),sPDL1中值为1.19(0.04~7.31)ng/ml,平均为1.45±1.18 ng/ml。NLR,PLR及SII的中位值分别为2.58,141.78和583.95。高水平sPDL1(≥0.96 ng/ml)患者的OS少于低水平s PDL1患者,差异具有统计学意义(P<0.001)。高水平sPDL1(HR=2.048,95%CI=1.26~3.34,P=0.004)及NLR(HR=1.565,95%CI=1.07~2.30,P=0.023)是患者预后较差的独立风险因素。结论血清sPDL1水平可以测定,且sPDL1及NLR在晚期BTC患者生存预后判断中具有重要临床意义。 展开更多

腹膜纤维化与血浆/腹透液中分泌型PDL1水平的相关性研究

目的探讨免疫检查点程序性细胞死亡配体1(PDL1)的分泌形式可溶性细胞程序性死亡配体1(sPDL1)与腹膜纤维化(PF)进展的相关性。方法对不同腹膜透析(PD)时长、不同PF程度的PD患者进行观察,检测PD脱落细胞PDL1表达、血浆和PD液中的sPDL1分... 目的探讨免疫检查点程序性细胞死亡配体1(PDL1)的分泌形式可溶性细胞程序性死亡配体1(sPDL1)与腹膜纤维化(PF)进展的相关性。方法对不同腹膜透析(PD)时长、不同PF程度的PD患者进行观察,检测PD脱落细胞PDL1表达、血浆和PD液中的sPDL1分泌情况;使用腹膜间皮细胞(HPMC)建立高糖纤维化模型并用慢病毒进行PDL1下调处理,评估PDL1/sPDL1与间皮细胞转分化的相关性。结果PD治疗之后sPDL1显著上调;与PD液sPDL1相比,血浆sPDL1与原发疾病等因素具有相关性;PD液sPDL1与PF水平呈正相关;高水平sPDL1患者更易进入腹膜功能下一阶段;PD液sPDL1与脱落细胞PDL1表达呈正相关;在高糖刺激下,PDL1及sPDL1在HPMC中的表达显著升高,随后α-SMA和snail的表达上调;慢病毒下调PDL1及sPDL1后,间皮细胞α-SMA及snail相应下调。结论PD液sPDL1与PD治疗的相关性较高;PD液sPDL1的增高可能为纤维化的早期征兆,提示腹膜溶质转运功能的升高;高糖诱导下,sPDL1在HPMC中过表达,促进细胞转分化,诱导PF的进展。 展开更多
