Rosa roxburghii is an underutilized natural fruit that contains very high levels of Vc. However, the Vc content of R. roxburghii varies considerably during plant development and ripening. To better understand the molecular mechanisms that underlie fluctuations in Vc content of R. roxburghii fruit at different developmental stages, we performed transcriptomic and metabolomic analyses and identified two significant gene networks/modules and 168 transcription factors directly involved in Vc synthesis. Promoter analysis of two core genes involved in Vc synthesis, RrGGP and RrGalUR, revealed the presence of a retroviral long terminal repeat(LTR) insert in the RrGalUR promoter. Using yeast one-hybrid and dual-luciferase assays, we demonstrated that the transcription factors RrHY5H and RrZIP9 bind to the promoter of RrGGP to promote its expression. RrZIP6 and RrWRKY4 bind to the LTR in the RrGalUR promoter to promote its expression. Our results reveal a molecular mechanism that controls Vc synthesis and accumulation in R. roxburghii fruit.展开更多
Anoectochilus roxburghii from different planting sources was used as the research object,and the biological activity of its extracts was compared.The results showed that the whole plant extracts of A.roxburghii plante...Anoectochilus roxburghii from different planting sources was used as the research object,and the biological activity of its extracts was compared.The results showed that the whole plant extracts of A.roxburghii planted in bag culture (BAG),greenhouse cultivation (GRE) and tissue culture seedlings (TIS) were rich in active ingredients such as A.roxburghii polysaccharides and A.roxburghii glycosides.In vitro experiments showed that Anoectochilus roxburghii extracts from different planting sources had good antioxidant capacity,tyrosinase inhibition ability and hyaluronidase inhibition ability.Among them,the GRE Anoectochilus roxburghii whole plant extract had the strongest scavenging ability on DPPH free radical,ABTS free radical and hydroxyl free radical,and the inhibitory effect on tyrosinase activity and hyaluronidase activity was significantly higher than that of BAG and TIS.At the same time,it also verified that the extract of Anoectochilus roxburghii has antioxidant,whitening,anti-inflammatory and other skin care effects,which provides a theoretical basis for the development and application of Anoectochilus roxburghii extract in the field of cosmetics.展开更多
Solid-phase extraction ultrahigh-performance liquid chromatography-tandem mass spectrometry(SPE-UHPLC-MS/MS)was used to evaluate the contamination of ochratoxin A(OTA)and assessed the human exposure risk of OTA in Ras...Solid-phase extraction ultrahigh-performance liquid chromatography-tandem mass spectrometry(SPE-UHPLC-MS/MS)was used to evaluate the contamination of ochratoxin A(OTA)and assessed the human exposure risk of OTA in Rasa roxburghii.A more suitable method for OTA extraction and purification of R.roxburghii was obtained.Treated 25 mL of R.roxburghii juice with enzymatic hydrolysis at a concentration of 0.06 mg/mL,filtered the resulting mixture and concentrated the filtrate to dry,then redissolved with 0.2 mL of methanol and diluted with 0.4 mL of ultra-pure water.Added sample solution to the activated hydrophilic-lipophilic balance(HLB)column,washed with 6 mL of ultra-pure water and purified by eluting with 6 mL of methanol.The eluent was collected and dried using nitrogen at 40℃,then redissolved in 1 mL of methanol and filtered for detection.The hazard quotient(HQ)values of all R.roxburghii fruit and juice,which were storage at room temperature,4 and−20℃from 0 day to 63 days,ranged from 0.077%to 35.792%,which were within the allowable limits for human consumption.From the perspective of OTA contamination,the results indicated that the maximal storage time of R.roxburghii fruit were 14 days at room temperature,35 days at 4℃and 63 days at−20℃.And the maximal storage time of R.roxburghii juice were 7 days for sealed storage and the same day for open storage at room temperature,14 days for sealed storage and 7 days for open storage at 4℃,and 63 days for sealed storage and 56 days for open storage at−20℃during the experiment period.And all thirty samples randomly sampled from the market were OTA negative.The results of this study can lay a foundation for the formulation of OTA limit standards in fruits and juice in the future,and provide a reference for consumers to consume R.roxburghii more healthily.展开更多

This study mainly investigated the regulatory effect of Rosa roxburghii Tratt fruit juice fermented by Lacticaseibacillus paracasei SR10-1(LAB-RRTJ)on modulating gut microbiota in dextran sulfate sodium(DSS)-induced u...This study mainly investigated the regulatory effect of Rosa roxburghii Tratt fruit juice fermented by Lacticaseibacillus paracasei SR10-1(LAB-RRTJ)on modulating gut microbiota in dextran sulfate sodium(DSS)-induced ulcerative colitis in mice.Compared to control group,DSS induction decreased body weight of mice,indexes of Shannon,Simpson,Chao1 and Faith_pd,and increased disease activity index(DAI)and levels of interleukin 1β(IL-1β),IL-6,tumor necrosis factorα(TNF-α)and interferon-γ(IFN-γ);And this induction also led to an increase in Proteobacteria,Verrucomicrobia and Actinobacteria at phylum level,harmful bacterial species richness at genus level,and relative richness of S.sciuri,Desulfovibrio C21_c20,R.gnavus and Akkermansia muciniphila at species level,and a decrease in Firmicutes at phylum level and relative richness of B.acidifaciens in mice.LAB-RRTJ increased body weight of mice with DSS induced ulcerative colitis(UC)and indexes of Shannon,Simpson,Chao1 and Faith_pd,reduced DAI and the content of four infl ammatory factors and improved gut microbiota imbalance in DSS induced UC mice.Besides,the number of operational taxonomic units(OTUs)increased,α-diversity andβ-diversity were restored and similar to those in mice in the control group after LAB-RRTJ treatment.Compared with the positive drug treatment group,LAB-RRTJ has a better effect on regulating gut microbiota diversity in colitis mice.Correlation analysis showed that infl ammatory factors were positively correlated with harmful bacteria and negatively correlated with beneficial bacteria which commonly found in some colitis mice.Taken together,our study demonstrated that LAB-RRTJ could alleviate DSS-induced colitis in mice through the modulation of infl ammatory cytokines and gut microbiota composition.展开更多

In this study, we employed transcriptomics and metabolomics to study changes related to the flavonoids at five different stages of R. roxburghii fruit development. Flavonoids and the genes related to their biosynthesis were found to undergo significant changes in abundance across different developmental stages, and numerous quercetin derivatives were identified. We found three gene expression modules that were significantly associated with the abundances of the different flavonoids in R. roxburghii and identified three structural UDP-glycosyltransferase genes directly involved in the synthesis of quercetin derivatives within these modules. In addition, we found that RrBEH4, RrLBD1 and RrPIF8could significantly increase the expression of downstream quercetin derivative biosynthesis genes. Taken together,these results provide new insights into the metabolism of flavonoids and the accumulation of quercetin derivatives in R. roxburghii.展开更多

Rosa roxburghii Tratt.is a well-known commercial horticultural crop in China with nutritional and medicinal value.Wild germplasms of this species are mainly distributed in Southwest China but the population is decreas...Rosa roxburghii Tratt.is a well-known commercial horticultural crop in China with nutritional and medicinal value.Wild germplasms of this species are mainly distributed in Southwest China but the population is decreasing due to continuous exploitation,habitat destruction,and fragmentation.Therefore,assessing the genetic diversity and phylogeography is essential for efficient conservation.Herein,two chloroplast intergenic spacers(trnL-trnF and accD-psaI)were investigated in 255 individuals from 29 R.roxburghii populations and 18 haplotypes(H1–H18)were identified.High levels of haplotype diversity(Hd=0.829)and nucleotide diversity(π=1.3×10^(−3))were detected in these populations.Also,the genetic variation representing 86.4%of the total variation was detected by an analysis of molecular variance.A significant correlation was established between genetic divergence and geographic distance by the Mantel test(r=0.204,P=0.04,9999 permutations),suggesting the isolation-by-distance model.A significantly higher Nst than Gst(Nst=0.257,Gst=0.136,P<0.05)indicated the phylogeographic structure of R.roxburghii.Further phylogeographic analysis revealed rapid range expansion in the population,probably between 647073 and 217848 years ago.The primary processes shaping the genetic patterns of the R.roxburghii populations included restricted gene flow with isolation distance within clades 1-8,2-3,and overall,contiguous expansion within clades 1-3 and 3-2,past fragmentation,and/or long-distance colonization within clades 1-9 and 2-2.Conservation priority should be given to the core populations GZ,FQ,DF,DS,xy,AL,LC,PB,and XY in the Yunnan-Guizhou Plateau,NZ and MX in the Qingling-Bashan mountains,and MN in the Hengduan mountains,where an in situ preservation and management strategy should be applied.展开更多

D. Shi). [Methods] Nitric acid was used as the digestion system for microwave digestion. Inductively coupled plasma mass spectrometry(ICP-MS) was used to determine the trace elements in thorn pear and golden thorn pear. [Results] Be, Ti, V, Cr, Mn, Co, Ni, As, Sr, Mo, Cd, Sb and Hg were determined by ICP-MS method in thorn pear and golden thorn pear. The correlation coefficients of the regression equations of the elements were higher than 0.996; the precision RSD was lower than 2.87%; and the recovery ranged from 94.3% to 109.8%. [Conclusions] The method has the advantages of high sensitivity, good accuracy and simultaneous detection of multiple elements, and provides a reference method and theoretical basis for determining trace elements in thorn pear and golden thorn pear and other fruits.展开更多

Sequence analysis showed 79-92% identity in nucleotide sequence and 75-87% identity in amino acid sequence to that of strawberry and Arabidopsis thaliana, etc. Northern blot showed that the expression of GalLDH was significantly different in different organs. The transcription level of GalLDH in fruit was significantly higher than that in leaf, stem and root respectively. Furthermore, this expression mode was highly correlated with AsA levels.展开更多

The result showed that contents of GA 1+3, IAA and ZRs in the flower receptacle of young fruit were high and then decreased, contents of GA 1+3and IAA in seeds increased with its development until maturity. The contents of ZRs and ABA in the flower receptacle of young fruits and seeds were high, and then had a tendency of descent with the development of fruits. The contents of GA 1+3, IAA, ZRs and ABA in flower receptacle of young fruit with normal fertilization were much higher than those of in non-fertilization fruits. In the flower receptacle of malformed fruits, the contents of GA 1+3, IAA and ZRs in normally developing flower receptacle were significantly higher than those in malformed fruits. The growth and development of fruits depended largely on the seeds, development because the seeds in the fruits with non-fertilization failed to be formed or even formed but irreproducible, which would lead to the decrease of GA 1+3, IAA contents and the abnormal development of fruits. Pinus roxburghii Sargent (Pinaceae) commonly known as "chir pine" is widely used in traditional and folkloric systems of medicine. The all parts of the plant are believed to possess medicinal qualities in Ayurvedic and Unani systems of medicine. In these traditional systems of medicine, the plant is used to heal many diseases, including afflictions of the eyes, ears, throat, blood, and skin. The plant parts are rich in various bioactive compounds such as a-pinene, abietic acid, quercetin and xanthone. Resin acids and flavanoid form a major portion of these bioactive compounds. This review presents examples of traditional medicinal uses for P. roxburghii, and subsequently explores the current understanding of the chemical, pharmacological, and biochemical properties of the extracts and the main active constituents found in each tissue of the plant. Clinical trial information is also included where available. Careful evaluation of these data may be helpful for scientists and researchers to discover and evaluate the specific chemical entities responsible for the traditional medicinal uses of P. roxburghii.展开更多

[Results]MS + 0. 5 mg/L NAA + 2 mg/L BA + 20 g/L sucrose + 6 g/L agar was suitable for induction of stem segments of A. roxburghii; MS + 0. 5 mg/L NAA + 2 mg/L BA + 1 mg/L KT + 25 g/L sucrose + 6 g/L agar was most suitable for proliferation of cluster buds of A. roxburghii; MS + 1. 0 mg/L IBA + 1. 0 mg/L NAA + 1 g/L activated carbon + 50 g/L mashed banana + 25 g/L sucrose + 6 g/L agar was most suitable for rooting and seedling hardening of A. roxburghii; using peat soil: fine sand( 3∶ 1) as transplantation matrix,the survival rate was the highest. OBJECTIVE To evaluate antioxidant,anti-inflammatory,antimicrobial activities of Pinus Roxburghii stem bark in vitro and evaluation of in vivo anti-osteoporotic activity in female rats.METHODS The antioxidant activity ...OBJECTIVE To evaluate antioxidant,anti-inflammatory,antimicrobial activities of Pinus Roxburghii stem bark in vitro and evaluation of in vivo anti-osteoporotic activity in female rats.METHODS The antioxidant activity of plant extract was assessed using DPPH,nitric oxide and H2O2 radical scavenging methods.In vitro anti-inflammatory activity was evaluated using albumin denaturation,membrane stabilization methods at various concentrations.The antibacterial and anti-fungal activity of extracts was also done against micro-organisms:Pseudomonas aurignosa,Escherichia coli,Staphylococcus aureus,Bacillus subtilis and Candida albicans.Quantitative analysis of total phenolics,flavonoids and tannins were also estimated.Female Wistar albino rats(220-260g)were employed and ovaries were surgically removed bilaterally in anaesthesia which causes post-menopausal syndrome in all the groups except sham operated rats.Extracts of Pinus roxburghii stem at 100 and 200mg·kg-1,po.and tamoxifen as standard 1mg·kg-1,po.were given.Acute toxicity study was also done.RESULTS Treatment with Pinus roxburghii extract in two doses significantly(P<0.05)ameliorated surgical ovariectomy induced biochemical and biomechanical changes like decrease in bone density,bone strength,alkaline phosphatase,serum calcium,serum phosphorous and increase in urine hydroxylproline levels.Plant extract also has significant(P<0.05)antioxidant,antimicrobial and anti-inflammatory activities.CONCLUSION The assessments done in present investigation may conclude that the constituents of Pinus roxburghii has significant potential in ameliorating oxidative,inflammatory reactions and possesses anti-microbial;osteobalstic and osteoclastic activities used for calcium homeostatis and improved bone strength in female rats.展开更多

Qiime and Mothur software packages were used to sort and count the number of sequences and operational taxonomic units (OTUs) and analyze the species abundance and distribution, Alpha diversity and differences of species abundance among samples. [Result] The number of effective sequences/OTUs for each sample was 110485/247 (stem) and 98524/259 (leaf) , respectively. The rarefaction curve showed that adequate sampling was performed. The number of OTUs had almost reached a saturation point. Endophytic bacteria in A. roxburghii mainly belonged to the genera Acidovorax, Pseudomonas, Sphingomonas and Halomonas. [Conclusions] Illumi-na MiSeq high-throughput sequencing technology provides more accurate and scientific data resources for the study of endophytic bacteria in A. roxburghii.展开更多
With growing concerns about the negative impact of environmental pollution on skin health,there is an increasing demand for natural active ingredients to combat free radicals and excessive melanin production.In this c...With growing concerns about the negative impact of environmental pollution on skin health,there is an increasing demand for natural active ingredients to combat free radicals and excessive melanin production.In this context,Rosa roxburghii Tratt(RR)has emerged as a potential source of skin-lightening compounds.Vitamin C(Vc)and superoxide dismutase(SOD)-rich extracts were extracted from RR,namely RRVC and RRSOD,using macroporous resin column chromatography and graded ammonium sulfate precipitation.The antioxidant and skin-lightening effects of these extracts were evaluated.RRVC and RRSOD showed strong inhibitory activity against various radicals and tyrosinase.When formulated together,their antioxidant and skin-lightening abilities were significantly enhanced,with the optimal ratio of RRVC and RRSOD(RRVS)of 5:32.In vivo studies on C57BL/6J mice revealed that these two extracts and their formulations effectively lowered oxidative stress levels in a model of d-galactose-induced aging in mice.Furthermore,they inhibited melanogenesis and tyrosinase activity in B16 cells and reduced ultravioletradiation-induced melanin deposition in mouse ears.The combination of RRVC and RRSOD had a more potent melanin-scavenging effect compared with the individual extracts.These findings suggest that RRVC and RRSOD,especially when combined at the optimal ratio,can whiten skin,increasing the value of RR products and offering natural alternatives for skin lightening.展开更多

文摘Anoectochilus roxburghii from different planting sources was used as the research object,and the biological activity of its extracts was compared.The results showed that the whole plant extracts of A.roxburghii planted in bag culture (BAG),greenhouse cultivation (GRE) and tissue culture seedlings (TIS) were rich in active ingredients such as A.roxburghii polysaccharides and A.roxburghii glycosides.In vitro experiments showed that Anoectochilus roxburghii extracts from different planting sources had good antioxidant capacity,tyrosinase inhibition ability and hyaluronidase inhibition ability.Among them,the GRE Anoectochilus roxburghii whole plant extract had the strongest scavenging ability on DPPH free radical,ABTS free radical and hydroxyl free radical,and the inhibitory effect on tyrosinase activity and hyaluronidase activity was significantly higher than that of BAG and TIS.At the same time,it also verified that the extract of Anoectochilus roxburghii has antioxidant,whitening,anti-inflammatory and other skin care effects,which provides a theoretical basis for the development and application of Anoectochilus roxburghii extract in the field of cosmetics.

文摘Solid-phase extraction ultrahigh-performance liquid chromatography-tandem mass spectrometry(SPE-UHPLC-MS/MS)was used to evaluate the contamination of ochratoxin A(OTA)and assessed the human exposure risk of OTA in Rasa roxburghii.A more suitable method for OTA extraction and purification of R.roxburghii was obtained.Treated 25 mL of R.roxburghii juice with enzymatic hydrolysis at a concentration of 0.06 mg/mL,filtered the resulting mixture and concentrated the filtrate to dry,then redissolved with 0.2 mL of methanol and diluted with 0.4 mL of ultra-pure water.Added sample solution to the activated hydrophilic-lipophilic balance(HLB)column,washed with 6 mL of ultra-pure water and purified by eluting with 6 mL of methanol.The eluent was collected and dried using nitrogen at 40℃,then redissolved in 1 mL of methanol and filtered for detection.The hazard quotient(HQ)values of all R.roxburghii fruit and juice,which were storage at room temperature,4 and−20℃from 0 day to 63 days,ranged from 0.077%to 35.792%,which were within the allowable limits for human consumption.From the perspective of OTA contamination,the results indicated that the maximal storage time of R.roxburghii fruit were 14 days at room temperature,35 days at 4℃and 63 days at−20℃.And the maximal storage time of R.roxburghii juice were 7 days for sealed storage and the same day for open storage at room temperature,14 days for sealed storage and 7 days for open storage at 4℃,and 63 days for sealed storage and 56 days for open storage at−20℃during the experiment period.And all thirty samples randomly sampled from the market were OTA negative.The results of this study can lay a foundation for the formulation of OTA limit standards in fruits and juice in the future,and provide a reference for consumers to consume R.roxburghii more healthily.
文摘Cosmetics made from natural ingredients are becoming more and more common because they contain bioactive compounds that provide many health benefits,are more environmentally friendly and sustainable.Anoectochilus roxburghii(AR)is a precious Chinese herb in China,possessing bioactive components including flavonoids,polysaccharides,volatile oils,kinsenoside,etc.Due to its unique bioactivity and high safety,the prospect of development and utilization is very broad.In this paper,the extraction,separation and purification techniques of AR’s flavonoid,polysaccharides,volatile oils and kinsenoside are briefly summarized.Meanwhile,we discusses their conformational relationships in cosmetics with antioxidant,whitening,antibacterial and soothing effect,and looks forward to the study of Anoectochilus roxburghii in the field of cosmetics,with a view to providing references to the development and utilization of this resource in the field of cosmetics.

文摘This study mainly investigated the regulatory effect of Rosa roxburghii Tratt fruit juice fermented by Lacticaseibacillus paracasei SR10-1(LAB-RRTJ)on modulating gut microbiota in dextran sulfate sodium(DSS)-induced ulcerative colitis in mice.Compared to control group,DSS induction decreased body weight of mice,indexes of Shannon,Simpson,Chao1 and Faith_pd,and increased disease activity index(DAI)and levels of interleukin 1β(IL-1β),IL-6,tumor necrosis factorα(TNF-α)and interferon-γ(IFN-γ);And this induction also led to an increase in Proteobacteria,Verrucomicrobia and Actinobacteria at phylum level,harmful bacterial species richness at genus level,and relative richness of S.sciuri,Desulfovibrio C21_c20,R.gnavus and Akkermansia muciniphila at species level,and a decrease in Firmicutes at phylum level and relative richness of B.acidifaciens in mice.LAB-RRTJ increased body weight of mice with DSS induced ulcerative colitis(UC)and indexes of Shannon,Simpson,Chao1 and Faith_pd,reduced DAI and the content of four infl ammatory factors and improved gut microbiota imbalance in DSS induced UC mice.Besides,the number of operational taxonomic units(OTUs)increased,α-diversity andβ-diversity were restored and similar to those in mice in the control group after LAB-RRTJ treatment.Compared with the positive drug treatment group,LAB-RRTJ has a better effect on regulating gut microbiota diversity in colitis mice.Correlation analysis showed that infl ammatory factors were positively correlated with harmful bacteria and negatively correlated with beneficial bacteria which commonly found in some colitis mice.Taken together,our study demonstrated that LAB-RRTJ could alleviate DSS-induced colitis in mice through the modulation of infl ammatory cytokines and gut microbiota composition.

文摘Rosa roxburghii fruit is rich in flavonoids, but little is known about their biosynthetic pathways. In this study, we employed transcriptomics and metabolomics to study changes related to the flavonoids at five different stages of R. roxburghii fruit development. Flavonoids and the genes related to their biosynthesis were found to undergo significant changes in abundance across different developmental stages, and numerous quercetin derivatives were identified. We found three gene expression modules that were significantly associated with the abundances of the different flavonoids in R. roxburghii and identified three structural UDP-glycosyltransferase genes directly involved in the synthesis of quercetin derivatives within these modules. In addition, we found that RrBEH4, RrLBD1 and RrPIF8could significantly increase the expression of downstream quercetin derivative biosynthesis genes. Taken together,these results provide new insights into the metabolism of flavonoids and the accumulation of quercetin derivatives in R. roxburghii.
文摘This study employed single factor tests and an orthogonal deign to optimize the process for extracting triterpenoids from Anoectochilus roxburghii with ultrasound-assisted technology.The results showed that the optimal extraction conditions were ultrasound treatment time of 30 min,a solid-to-liquid ratio of 1∶40,an ethanol volume fraction of 70%,and extraction for 40 min.Under these conditions,the yield of triterpenoids extracted from A.roxburghii was 1.621%,with a relative deviation of 0.0335%.The results provided a basis for the extraction and utilization of medicinal components in A.roxburghii.

文摘Rosa roxburghii Tratt.is a well-known commercial horticultural crop in China with nutritional and medicinal value.Wild germplasms of this species are mainly distributed in Southwest China but the population is decreasing due to continuous exploitation,habitat destruction,and fragmentation.Therefore,assessing the genetic diversity and phylogeography is essential for efficient conservation.Herein,two chloroplast intergenic spacers(trnL-trnF and accD-psaI)were investigated in 255 individuals from 29 R.roxburghii populations and 18 haplotypes(H1–H18)were identified.High levels of haplotype diversity(Hd=0.829)and nucleotide diversity(π=1.3×10^(−3))were detected in these populations.Also,the genetic variation representing 86.4%of the total variation was detected by an analysis of molecular variance.A significant correlation was established between genetic divergence and geographic distance by the Mantel test(r=0.204,P=0.04,9999 permutations),suggesting the isolation-by-distance model.A significantly higher Nst than Gst(Nst=0.257,Gst=0.136,P<0.05)indicated the phylogeographic structure of R.roxburghii.Further phylogeographic analysis revealed rapid range expansion in the population,probably between 647073 and 217848 years ago.The primary processes shaping the genetic patterns of the R.roxburghii populations included restricted gene flow with isolation distance within clades 1-8,2-3,and overall,contiguous expansion within clades 1-3 and 3-2,past fragmentation,and/or long-distance colonization within clades 1-9 and 2-2.Conservation priority should be given to the core populations GZ,FQ,DF,DS,xy,AL,LC,PB,and XY in the Yunnan-Guizhou Plateau,NZ and MX in the Qingling-Bashan mountains,and MN in the Hengduan mountains,where an in situ preservation and management strategy should be applied.

文摘[Objectives] This study was conducted to establish an analytical method for the establishment of 13 trace elements in Anshun thorn pear(Rosa roxburghii) and golden thorn pear(Rosa sterilis S. D. Shi). [Methods] Nitric acid was used as the digestion system for microwave digestion. Inductively coupled plasma mass spectrometry(ICP-MS) was used to determine the trace elements in thorn pear and golden thorn pear. [Results] Be, Ti, V, Cr, Mn, Co, Ni, As, Sr, Mo, Cd, Sb and Hg were determined by ICP-MS method in thorn pear and golden thorn pear. The correlation coefficients of the regression equations of the elements were higher than 0.996; the precision RSD was lower than 2.87%; and the recovery ranged from 94.3% to 109.8%. [Conclusions] The method has the advantages of high sensitivity, good accuracy and simultaneous detection of multiple elements, and provides a reference method and theoretical basis for determining trace elements in thorn pear and golden thorn pear and other fruits.

文摘A 855 bp cDNA encoding L-galactono-1,4-lactone dehydrogenase (GalLDH) fragment was cloned from fruit of R. roxburghii Tratt by the method of RT-PCR, on the basis of the homologous genes of Arabidopsis thaliana, cauliflower, sweet potato, strawberry, etc. in GenBank. Sequence analysis showed 79-92% identity in nucleotide sequence and 75-87% identity in amino acid sequence to that of strawberry and Arabidopsis thaliana, etc. Northern blot showed that the expression of GalLDH was significantly different in different organs. The transcription level of GalLDH in fruit was significantly higher than that in leaf, stem and root respectively. Furthermore, this expression mode was highly correlated with AsA levels.
文摘The fruit of cultivar Guinong5 (Rosa roxburghii Tratt.) was used to investigate the regularity of its growth and development, and the changes of endogenous hormone contents during the period of the growth and development. The result showed that contents of GA 1+3, IAA and ZRs in the flower receptacle of young fruit were high and then decreased, contents of GA 1+3and IAA in seeds increased with its development until maturity. The contents of ZRs and ABA in the flower receptacle of young fruits and seeds were high, and then had a tendency of descent with the development of fruits. The contents of GA 1+3, IAA, ZRs and ABA in flower receptacle of young fruit with normal fertilization were much higher than those of in non-fertilization fruits. In the flower receptacle of malformed fruits, the contents of GA 1+3, IAA and ZRs in normally developing flower receptacle were significantly higher than those in malformed fruits. The growth and development of fruits depended largely on the seeds, development because the seeds in the fruits with non-fertilization failed to be formed or even formed but irreproducible, which would lead to the decrease of GA 1+3, IAA contents and the abnormal development of fruits. Therefore the fruits deformed and dropped inevitably.
文摘Traditional medicine is a blend of information gathered over generations from various communities and cultures. Pinus roxburghii Sargent (Pinaceae) commonly known as "chir pine" is widely used in traditional and folkloric systems of medicine. The all parts of the plant are believed to possess medicinal qualities in Ayurvedic and Unani systems of medicine. In these traditional systems of medicine, the plant is used to heal many diseases, including afflictions of the eyes, ears, throat, blood, and skin. The plant parts are rich in various bioactive compounds such as a-pinene, abietic acid, quercetin and xanthone. Resin acids and flavanoid form a major portion of these bioactive compounds. This review presents examples of traditional medicinal uses for P. roxburghii, and subsequently explores the current understanding of the chemical, pharmacological, and biochemical properties of the extracts and the main active constituents found in each tissue of the plant. Clinical trial information is also included where available. Careful evaluation of these data may be helpful for scientists and researchers to discover and evaluate the specific chemical entities responsible for the traditional medicinal uses of P. roxburghii.

文摘[Objectives]To screen the tissue culture rapid propagation formula suitable for each tissue culture stage of Anoectochilus roxburghii( Wall) Lind. [Methods] The stem segments of Fujian A. roxburghii were used as explant to study the tissue culture rapid propagation and transplantation techniques. The comparative experiment was carried out to study the effects of different hormone concentrations on the induction of stem segments,proliferation of cluster buds,rooting and seedling hardening of A. roxburghii,and study the effects of transplantation matrix on the transplantation of A. roxburghii. [Results]MS + 0. 5 mg/L NAA + 2 mg/L BA + 20 g/L sucrose + 6 g/L agar was suitable for induction of stem segments of A. roxburghii; MS + 0. 5 mg/L NAA + 2 mg/L BA + 1 mg/L KT + 25 g/L sucrose + 6 g/L agar was most suitable for proliferation of cluster buds of A. roxburghii; MS + 1. 0 mg/L IBA + 1. 0 mg/L NAA + 1 g/L activated carbon + 50 g/L mashed banana + 25 g/L sucrose + 6 g/L agar was most suitable for rooting and seedling hardening of A. roxburghii; using peat soil: fine sand( 3∶ 1) as transplantation matrix,the survival rate was the highest. [Conclusions] The experiment results are expected to provide references for factory production of A. roxburghii.

文摘OBJECTIVE To evaluate antioxidant,anti-inflammatory,antimicrobial activities of Pinus Roxburghii stem bark in vitro and evaluation of in vivo anti-osteoporotic activity in female rats.METHODS The antioxidant activity of plant extract was assessed using DPPH,nitric oxide and H2O2 radical scavenging methods.In vitro anti-inflammatory activity was evaluated using albumin denaturation,membrane stabilization methods at various concentrations.The antibacterial and anti-fungal activity of extracts was also done against micro-organisms:Pseudomonas aurignosa,Escherichia coli,Staphylococcus aureus,Bacillus subtilis and Candida albicans.Quantitative analysis of total phenolics,flavonoids and tannins were also estimated.Female Wistar albino rats(220-260g)were employed and ovaries were surgically removed bilaterally in anaesthesia which causes post-menopausal syndrome in all the groups except sham operated rats.Extracts of Pinus roxburghii stem at 100 and 200mg·kg-1,po.and tamoxifen as standard 1mg·kg-1,po.were given.Acute toxicity study was also done.RESULTS Treatment with Pinus roxburghii extract in two doses significantly(P<0.05)ameliorated surgical ovariectomy induced biochemical and biomechanical changes like decrease in bone density,bone strength,alkaline phosphatase,serum calcium,serum phosphorous and increase in urine hydroxylproline levels.Plant extract also has significant(P<0.05)antioxidant,antimicrobial and anti-inflammatory activities.CONCLUSION The assessments done in present investigation may conclude that the constituents of Pinus roxburghii has significant potential in ameliorating oxidative,inflammatory reactions and possesses anti-microbial;osteobalstic and osteoclastic activities used for calcium homeostatis and improved bone strength in female rats.

文摘[Objectives]To compare the differences in the total flavonoids between leaves and whole plants of Anoectochilus roxburghii.[Methods]The total flavonoids in the tissue cultured leaves and whole plants of A.roxburghii were extracted by microwave-assisted extraction.The content of total flavonoids in the extracts was determined by UV spectrophotometry.[Results]The results of the content determination showed that there were significant differences in the flavonoid content between the leaves and the whole plants of A.roxburghii.[Conclusions]The content of total flavonoids in tissue cultured leaves was higher than that of whole plants.Thus,in the development and utilization of A.roxburghii,tissue cultured leaves can be used to take the place of the whole plants.

文摘[Objective] This study aimed to investigate the diversity of community composition and diversity of endophytic bacteria in Anoectochilus roxburghii.[Method] By using Illumina MiSeq high-throughput sequencing technology, the 16S rDNA-V4 region of endophytic bacteria in ^4. roxburghii was sequenced. Qiime and Mothur software packages were used to sort and count the number of sequences and operational taxonomic units (OTUs) and analyze the species abundance and distribution, Alpha diversity and differences of species abundance among samples. [Result] The number of effective sequences/OTUs for each sample was 110485/247 (stem) and 98524/259 (leaf) , respectively. The rarefaction curve showed that adequate sampling was performed. The number of OTUs had almost reached a saturation point. Endophytic bacteria in A. roxburghii mainly belonged to the genera Acidovorax, Pseudomonas, Sphingomonas and Halomonas. [Conclusions] Illumi-na MiSeq high-throughput sequencing technology provides more accurate and scientific data resources for the study of endophytic bacteria in A. roxburghii.

文摘With growing concerns about the negative impact of environmental pollution on skin health,there is an increasing demand for natural active ingredients to combat free radicals and excessive melanin production.In this context,Rosa roxburghii Tratt(RR)has emerged as a potential source of skin-lightening compounds.Vitamin C(Vc)and superoxide dismutase(SOD)-rich extracts were extracted from RR,namely RRVC and RRSOD,using macroporous resin column chromatography and graded ammonium sulfate precipitation.The antioxidant and skin-lightening effects of these extracts were evaluated.RRVC and RRSOD showed strong inhibitory activity against various radicals and tyrosinase.When formulated together,their antioxidant and skin-lightening abilities were significantly enhanced,with the optimal ratio of RRVC and RRSOD(RRVS)of 5:32.In vivo studies on C57BL/6J mice revealed that these two extracts and their formulations effectively lowered oxidative stress levels in a model of d-galactose-induced aging in mice.Furthermore,they inhibited melanogenesis and tyrosinase activity in B16 cells and reduced ultravioletradiation-induced melanin deposition in mouse ears.The combination of RRVC and RRSOD had a more potent melanin-scavenging effect compared with the individual extracts.These findings suggest that RRVC and RRSOD,especially when combined at the optimal ratio,can whiten skin,increasing the value of RR products and offering natural alternatives for skin lightening.
