In this paper,we propose a second-order moving-water equilibria preserving nonstaggered central scheme to solve the Ripa model via flux globalization.To maintain the moving-water steady states,we use the discrete source terms proposed by Britton et al.(J Sci Comput,2020,82(2):Art 30)by incorporating the expression of the source terms as a whole into the flux gradient,which directly avoids the discrete complexity of the source terms in order to maintain the well-balanced properties of the scheme.In addition,since the nonstaggered central scheme requires re-projecting the updated values of the nonstaggered cells onto the staggered cells,we modify the calculation of the global variables by constructing ghost cells and alternating the values of the global variables with the water depths obtained from the solution through the nonlinear relationship between the global flux and the water depth.In order to maintain the second-order accuracy of the scheme on the time scale,we incorporate a new Runge-Kutta type time discretization in the evolution of the numerical solution for the nonstaggered cells.Meanwhile,we introduce the"draining"time step technique to ensure that the water depth is positive and that it satisfies mass conservation.Numerical experiments verify that the scheme is well-balanced,positivity-preserving and robust. 展开更多

目的:探讨Ripa裂解液蛋白提取法和Invent离心管柱蛋白提取法对慢性应激模型大鼠海马组织PI3K/Akt/mTOR信号通路相关蛋白表达量的影响。方法:空白组和模型组大鼠各3只,取双侧海马组织,每个样品分别采用Ripa裂解液、Invent离心管柱蛋白提... 目的:探讨Ripa裂解液蛋白提取法和Invent离心管柱蛋白提取法对慢性应激模型大鼠海马组织PI3K/Akt/mTOR信号通路相关蛋白表达量的影响。方法:空白组和模型组大鼠各3只,取双侧海马组织,每个样品分别采用Ripa裂解液、Invent离心管柱蛋白提取法进行蛋白提取,分为空白+Invent组、空白+Ripa组、模型+Invent组、模型+Ripa组。采用Western Blot法检测PI3K/Akt/mTOR信号通路相关蛋白表达情况,析因设计方法分析各因素的效果。结果:大鼠海马PI3K、p-Akt/Akt、p-mTOR/mTOR表达均受组别和提取方法的影响,组别和提取方法存在交互作用,运用Invent离心管柱蛋白提取法提取蛋白,模型组的PI3K、p-Akt/Akt、p-mTOR/mTOR表达较空白组减少,差异有统计学意义(P<0.05)。结论:慢性应激抑制大鼠海马PI3K/Akt/mTOR信号通路, Invent离心管柱蛋白提取法操作便捷,蛋白提取率高,尤其对信号通路中磷酸化蛋白的提取具有明显的优势。 展开更多

背景:RIPA Buffer对不同组织细胞总蛋白的提取效率有差异,并不通适于所有的组织样本。目的:筛选适合酸中毒小鼠骨骼肌组织蛋白提取的裂解液,为研究骨骼肌萎缩奠定基础。方法:3月龄健康雄性C57BL/6小鼠20只,体质量25-30 g,由山西医科大... 背景:RIPA Buffer对不同组织细胞总蛋白的提取效率有差异,并不通适于所有的组织样本。目的:筛选适合酸中毒小鼠骨骼肌组织蛋白提取的裂解液,为研究骨骼肌萎缩奠定基础。方法:3月龄健康雄性C57BL/6小鼠20只,体质量25-30 g,由山西医科大学实验动物中心提供。麻醉后脱臼处死小鼠,分离获取下肢腓肠肌。实验分为2组:HCl诱导酸中毒组每日定时供给10 g混合10 mL 0.4 mol/L HCl的饲料;对照组饲料中混合同等量的H2O,连续7 d。比较RIPA Buffer、Original Buffer和JP Buffer对酸中毒小鼠骨骼肌组织蛋白的提取效果;蛋白免疫印迹实验检测AKT、p-AKT(Thr308)、rp S6和p-rp S6(Ser235/236)的表达;RT-qPCR定量分析GLUT4的表达。结果与结论:①3种裂解液对骨骼肌组织蛋白的提取能力有差异,JP Buffer提取蛋白产量高,但是目的蛋白信号并不强;RIPA Buffer提取蛋白产量相对偏低;Original Buffer提取足量骨骼肌组织蛋白,蛋白免疫印迹显示目的蛋白条带清晰;②蛋白免疫印迹实验评分显示,Original Buffer总分高于其他2种裂解液;③蛋白免疫印迹实验结果显示,酸中毒组和对照组AKT、rpS6磷酸化程度均无变化;④RT-qPCR定量分析显示,酸中毒组和对照组RNA水平GLUT4表达无变化; ⑤结果提示,OriginalBuffer是提取骨骼肌组织蛋白的最佳裂解液;短期HCl诱导酸中毒组AKT信号通路未激活,延长酸中毒时间是否会激活该信号通路值得研究。针对不同实验样本选择合适的裂解液是保证蛋白免疫印迹实验结果可信度的前提。 展开更多

急性阑尾炎是最常见的急腹症之一,及时、准确地诊断急性阑尾炎是降低病死率、减少住院时间和治疗费用的最佳方法。急性阑尾炎的诊断主要基于患者的临床症状、各种炎症指标及影像学检查结果,在临床实践中大多依赖影像学检查。临床评分系... 急性阑尾炎是最常见的急腹症之一,及时、准确地诊断急性阑尾炎是降低病死率、减少住院时间和治疗费用的最佳方法。急性阑尾炎的诊断主要基于患者的临床症状、各种炎症指标及影像学检查结果,在临床实践中大多依赖影像学检查。临床评分系统对患者的症状、体征和实验室指标进行赋值,然后进行风险分层来指导临床决策,可指导临床医生有针对性地使用辅助检查,减少对辅助检查的依赖,节约医疗资源。目前,有关临床评分系统诊断有阑尾炎的临床应用及验证研究在国内鲜见报告。本文就急性阑尾炎相关临床评分系统概况和常见的临床评分系统在急性阑尾炎诊断中的应用研究进展进行综述,以期为急性阑尾炎的诊断病情评估提供参考。 展开更多

[目的]基于改良的对比蛋白提取结果的方法,探讨两种不同总蛋白提取方法对不同丰度蛋白提取得率的影响。[方法]小胶质细胞株N9总蛋白及商品化预染蛋白标准品,行聚丙烯酰胺凝胶电泳(SDS-PAGE),三种常用图像软件定量免疫印迹结果,比较线性相关系数R^2;选择R^2值最高的软件,灰密度定量不同丰度p-IκBα经RIPA裂解液法和Minute试剂盒法蛋白提取后免疫印迹检测的p-IκBα蛋白得率。[结果]三种软件定量分析同一免疫印迹结果显示,Image Studio Lite的上样量与条带灰密度值的R^2最大(R^2=0.998);运用Image Studio Lite定量免疫印迹检测p-IκBα高丰度组,发现使用RIPA裂解液法比Minute法的p-IκBα提取得率高出7.5%,而p-IκBα低丰度组,前者p-IκBα的得率只有后者的41%,差异显著(P<0.05)。[结论]Minute试剂盒法在免疫印迹检测中对低丰度蛋白具有高提取得率的优越性。 展开更多

目的探讨超声检测加速时间/射血时间(AT/ET)、胎儿肺动脉阻力指数(RIPA)联合总肺容积(TLV)诊断胎儿肺成熟度的价值。方法选取2021年1月至2022年5月在金水区总医院进行产前检查的159例孕妇(孕周≥38周)为研究对象,根据胎儿羊水或气管抽... 目的探讨超声检测加速时间/射血时间(AT/ET)、胎儿肺动脉阻力指数(RIPA)联合总肺容积(TLV)诊断胎儿肺成熟度的价值。方法选取2021年1月至2022年5月在金水区总医院进行产前检查的159例孕妇(孕周≥38周)为研究对象,根据胎儿羊水或气管抽吸液中板层小体(LB)是否≥50×10^(9)/L分为肺发育成熟组(124例,LB≥50×10^(9)/L)和肺发育不成熟组(35例,LB<50×10^(9)/L)。比较两组一般临床资料、胎儿AT/ET、RIPA、TLV,Logistic回归分析胎儿肺发育不成熟的影响因素,受试者工作特征(ROC)曲线分析AT/ET、RIPA、TLV联合检测对胎儿肺发育不成熟的预测价值。结果肺发育不成熟组产妇营养状况、羊水指数与肺发育成熟组比较,差异有统计学意义(P<0.05);与肺发育成熟组比较,肺发育不成熟组胎儿AT/ET、TLV指标较低,RIPA指标较高(P<0.05);Logistic回归分析显示,产妇营养不良、羊水指数(≤8.76 cm)、胎儿AT/ET(≤0.32)、RIPA(>0.83)、TLV(≤86.63 cm3)是胎儿肺发育不良的危险因素(P<0.05);胎儿AT/ET、RIPA、TLV指标联合预测胎儿肺发育不成熟的AUC、敏感度、特异度最大。结论AT/ET、RIPA、TLV指标与胎儿肺发育程度有关,联合检测对临床预测胎儿肺发育不成熟具有数据指导作用。 展开更多

The innate immune response is the first line of host defense to eliminate viral infection. Pattern recognition receptors in the cytosol, such as RIG-I-like receptors (RLR) and Nod-like receptors (NLR), and membrane bound Toll like receptors (TLR) detect viral infection and initiate transcription of a cohort of antiviral genes, including interferon (IFN) and interferon stimulated genes (ISGs), which ultimately block viral replication. Another mechanism to reduce viral spread is through RIPA, the RLR-induced IRF3-mediated pathway of apoptosis, which causes infected cells to undergo pre- mature death. The transcription factor IRF3 can mediate cellular antiviral responses by both inducing antiviral genes and triggering apoptosis through the activation of RIPA. The mechanism of IRF3 activation in RIPA is dis- tinct from that of transcriptional activation; it requires linear polyubiquitination of specific lysine residues of IRF3. Using RIPA-active, but transcriptionally inactive, IRF3 mutants, it was shown that RIPA can prevent viral replication and pathogenesis in mice. 展开更多

In this work,we present a high-order discontinuous Galerkin method for the shallow water equations incorporating horizontal temperature gradients(also known as the Ripa model),which exactly maintains the lake at rest steady state.Herein,we propose original numerical fluxes defined on the basis of the hydrostatic reconstruction idea and a simple source term approximation.This novel approach allows us to achieve the well-balancing of the discontinuous Galerkin method without complication.Moreover,the proposed method retains genuinely high-order accuracy for smooth solutions and it shows good resolution for discontinuous solutions at the same time.Rigorous numerical analysis as well as extensive numerical results all verify the good performances of the proposed method. 展开更多
