In this study,a straightforward one-step hydrothermal method was successfully utilized to synthesize the solid solution Na_(0.9)Mg_(0.45)Ti_(3.55)O_(8)-Na_(2)Ni_(2)Ti_(6)O_(16)(NNMTO-x),where x denotes the molar perce...In this study,a straightforward one-step hydrothermal method was successfully utilized to synthesize the solid solution Na_(0.9)Mg_(0.45)Ti_(3.55)O_(8)-Na_(2)Ni_(2)Ti_(6)O_(16)(NNMTO-x),where x denotes the molar percentage of Na_(2)Ni_(2)Ti_(6)O_(16)(NNTO)within Na_(0.9)Mg_(0.45)Ti_(3.55)O_(8)(NMTO),with x values of 10,20,30,40,and 50.Both XPS(X-ray Photoelectron Spectroscopy)and EDX(Energy Dispersive X-ray Spectroscopy)analyses unequivocally validated the formation of the NNMTO-x solid solutions.It was observed that when x is below 40,the NNMTO-x solid solution retains the structural characteristics of the original NMTO.However,beyond this threshold,significant alterations in crystal morphology were noted,accompanied by a noticeable decline in photocatalytic activity.Notably,the absorption edge of NNMTO-x(x<40)exhibited a shift towards the visible-light spectrum,thereby substantially broadening the absorption range.The findings highlight that NNMTO-30 possesses the most pronounced photocatalytic activity for the reduction of CO_(2).Specifically,after a 6 h irradiation period,the production rates of CO and CH_(4)were recorded at 42.38 and 1.47μmol/g,respectively.This investigation provides pivotal insights that are instrumental in the advancement of highly efficient and stable photocatalysts tailored for CO_(2)reduction processes.展开更多

years,traditional Chinese medicine compounds and probiotics have shown good potential in regulating the endocrine system and improving the intestinal microecology,providing new ideas for the treatment of breast hyperplasia.OBJECTIVE:To explore the effects and mechanisms of traditional Chinese medicine compounds and fermented probiotic compounds on breast hyperplasia in mice,providing new theoretical and experimental bases for the clinical treatment and prevention of breast hyperplasia.METHODS:(1)Network pharmacology tools were used to predict the anti-breast-hyperplasia activity of Herba Gueldenstaedtiae(Euphorbia humifusa),as well as its potential targets and signaling pathways.The databases included:TCMSP,OMIM,GeneCards database,UniProt website,Venny2.1.0 website,Metascape,HERB website,and STRING database,all of which are open-access databases.Network pharmacology can predict and screen key information such as the targets corresponding to the active ingredients of traditional Chinese medicine,disease targets,and action pathways through network analysis and computer-system analysis.Therefore,it has been increasingly widely used in the research of traditional Chinese medicine.(2)A breast hyperplasia model was induced in mice by injecting estrogen and progesterone.Mice in the normal blank group were injected intraperitoneally with normal saline every day.Mice in the model group and drugadministration groups were injected intraperitoneally with estradiol benzoate injection at a concentration of 0.5 mg/kg every day for 25 days.From the 26th day,the injection of estradiol benzoate injection was stopped.Mice in the normal blank group were injected intramuscularly with normal saline every day,and mice in the model group and drug-administration groups were injected intramuscularly with progesterone injection at a concentration of 5 mg/kg for 5 days.After the model was established,each group was given drugs respectively.The normal blank group and the model group were gavaged with 0.2 mL/d of normal saline;the positive blank group(Xiaozheng Pill group)was gavaged with an aqueous solution of Xiaozheng Pill at 0.9 mg/g;the low-,medium-and high-dose groups of Compound Herba Gueldenstaedtiae were gavaged with an aqueous solution of the compound medicine at 0.75,1.5,and 3.0 mg/(g·d)respectively;the low-,medium-and high-dose groups of traditional Chinese medicine-bacteria fermentation were gavaged with an aqueous solution of the compound medicine at 0.75,1.5,and 3.0 mg/(g·d)respectively.The administration was continuous for 30 days.RESULTS AND CONCLUSION:(1)The results of network pharmacology research showed that the Compound Herba Gueldenstaedtiae(Euphorbia humifusa)contained 46 active ingredients,which were related to 1213 potential targets.After comparison with 588 known breast-hyperplasia targets,it was speculated that 50 of these targets might be related to the direct effect of the compound on breast hyperplasia.(2)After drug intervention,there was no significant change in the high-dose group of Compound Herba Gueldenstaedtiae compared with the normal blank group.The liver indicators of the other intervention groups all significantly decreased(P<0.05).(3)In terms of kidney and uterine indicators,the medium-dose group of Compound Herba Gueldenstaedtiae decreased significantly compared with the normal blank group(P<0.05).In terms of the uterine index,the model group increased significantly compared with the normal blank group(P<0.01).(4)After 1-month drug treatment,the number of lobules and acini in the breast tissue of the Xiaozheng Pill group,the low,medium,and high-dose group of Compound Herba Gueldenstaedtiae,the low,medium,and highdose groups of traditional Chinese medicine-bacteria fermentation decreased,and the duct openings narrowed.With the increase of drug dose,diffuse hyperplasia of breast tissue was significantly improved.(5)The ELISA results showed that compared with the model group,the estrogen level was lower in the medium-dose group of traditional Chinese medicine-bacteria fermentation after the intervention(P<0.05).In addition,the follicle-stimulating hormone level in the low-dose group of Compound Herba Gueldenstaedtiae was lower than that of the model group(P<0.05).(6)The intervention in the mouse model led to changes in the abundance of short chain fatty acids and intestinal flora in all groups.To conclude,the Compound Herba Gueldenstaedtiae and its probiotic fermentation products significantly improved mammary gland hyperplasia in mice by regulating hormone levels,improving the structure of the gut microbiota,and increasing the content of shortchain fatty acids,providing new ideas and potential Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to enviro...Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to environmental changes upon entry into oral biofilms are reviewed.Supra-gingival biofilms are characterized by acidic pH,the presence of bacterial enzymes,and the development of hypoxia in deeper layers.Sub-gingival biofilms are slightly alkaline,with hypoxia occurring over their entire depth.Upon entering biofilms,negatively charged,pH-and/or hypoxia-responsive nanocarriers become positively charged.This charge reversal leads to electrostatic double-layer attraction between positively charged nanocarriers towards negatively charged,waterfilled channel walls in biofilms,enhancing their accumulation in a biofilm.Degradation of bacterial enzyme-responsive nanocarriers causes in-biofilm release of antimicrobial cargo,yielding higher local antimicrobial concentrations than can be achieved through their direct,oral administration without harming soft tissues.Enhanced antibiofilm activity after in-biofilm antimicrobial release from biofilm-responsive micelles and liposomes has been demonstrated in vitro towards single-species Streptococcus mutans and Staphylococcus aureus biofilms or in vivo using specific-pathogen-free rodents inoculated with selected pathogens.This preferential antibacterial activity regulated the microbial composition of ex vivo human oral biofilm towards a more healthy microbiome composition.Although clinical confirmation is limited,the potential benefits of stimuli-responsive,antimicrobial-loaded nanocarriers for oral biofilm control and microbiome restoration are worth further investigation towards clinical translation.展开更多

Understanding the aerodynamic and dynamic characteristics of unloaded freight trains in crosswinds is pivotal for ensuring their operational safety and reliability.The dynamic performance of unloaded gondola cars unde...Understanding the aerodynamic and dynamic characteristics of unloaded freight trains in crosswinds is pivotal for ensuring their operational safety and reliability.The dynamic performance of unloaded gondola cars under varying windbreak heights is therefore investigated in this study,revealing distinct differences in lateral stability and safety indicators,and enabling the determination of an optimal windbreak height.A 3D unsteady aerodynamic model was developed using the improved delayed detached eddy simulation(IDDES)method and an overset numerical mesh.Also leveraging a multi-body dynamics(MBD)model of a three-wagon freight car configuration,we investigate time-averaged aerodynamic forces,transient flow field distributions,and nonlinear dynamic responses.Parametric analyses reveal a non-monotonic relationship between the height of the windbreak and the stability of the train.A windbreak with a critical height of 2 m(0.74 relative to the car body height)results in 76%,64%,and 81%lower values of the derailment coefficient CD,wheel unloading ratio R,and overturning coefficient C_(O),respectively.Notably,when the height of the windbreak exceeds 2 m,vortices within the gondola induce an adverse pressure coefficient distribution(C_(p)=−2.17)on the leeward internal wall,intensifying the lateral force and overturning moment.Furthermore,frequency-domain analysis reveals that the lateral sway and overturning vibration mode are associated with low-frequency(1.61 Hz)lateral vibrations under crosswind conditions.This study provides a theoretical foundation for the design and optimization of railway windbreaks.展开更多
Reinforced concrete(RC)columns are often subjected to off-central explosion due to the uncertainty of blast locations.However,few studies have focused on the dynamic response of RC columns under offcentral explosions....Reinforced concrete(RC)columns are often subjected to off-central explosion due to the uncertainty of blast locations.However,few studies have focused on the dynamic response of RC columns under offcentral explosions.A field blast experiment was conducted under close-in explosion with varying detonation offset distances(0 m,0.5 m,and 1 m),the overpressure load and dynamic responses of the full-scale RC columns were measured.Compared with the centrally detonated condition,a relative offset distance of 1.67 decreases the maximum and residual deflections of the RC column by 16.8%and 21.4%,respectively,while increasing the maximum and residual support rotations by 24.7%and 17.8%.Based on the experimental results,a theoretical model was proposed that considers the detonation location and charge mass,boundary conditions,axial compression ratio and material properties.The theoretical model exhibited good agreement with the experimental results,with prediction errors below 10%for both maximum and residual deflection.The effects of parameters were analyzed,and it indicated that an increase in offset distance results in decreased maximum and residual deflections but an increased support angle,thereby exacerbating damage.Higher axial load ratio,span-depth ratio,and longitudinal reinforcement ratio reduce both deflections and support angle.Additionally,a rapid method to predict the maximum and residual deflection of RC columns under off-central blast loading was also proposed based on the Generalized Regression Neural Network(GRNN).Eleven features which related to the RC column properties and the blast characteristics were used in the training process of GRNN,and accurate predictions were achieved with prediction errors within 20%.This study fills the gap in predicting the dynamic response of RC columns under off-central explosion,providing valuable references for blast-resistant design.展开更多
Eucalyptus(Eucalyptus camaldulensis Dehnh.)is an important exotic species in northern Nigeria commonly used for poles and timber.Sustainable management of this resource would require quantifying its volume.Stem taper ...Eucalyptus(Eucalyptus camaldulensis Dehnh.)is an important exotic species in northern Nigeria commonly used for poles and timber.Sustainable management of this resource would require quantifying its volume.Stem taper equations are one of the main and most efficient methods for estimating stem volume to any merchantable limit of a species.There is currently no taper equation for Eucalyptus species in Nigeria.Therefore,this study developed taper equations for E.camaldulensis in northern Nigeria.Data for this study were obtained from a private plantation in Jalingo Local Government Area,Taraba State,Nigeria.68 trees were felled and sectioned into 1-m bolt across the stem to a merchantable limit of 5 cm,which were used as the fitting dataset.An additional 22 trees were felled and used to validate the taper equations for stem volume estimation.Seven taper equations were initially fitted to the dataset using nonlinear least squares.The best taper equation was then refitted using a nonlinear mixed-effects approach and calibrated using diameters of one to five sections from the butt end.The taper equations were numerically integrated to obtain the stem volume,which was compared with empirical volume equations.The result shows that the Kozak(Can J For Res 27(5):619-629.10.1139/x97-011,1997)equation,which included eight parameters,provided the best fit for predicting section diameters for under and over bark.The mixed-effects taper equation(NLME-TE)explained most stem diameter variations in the fitting dataset(pseudo-R2:0.986-0.987;RMSE:0.547-0.578 cm)without substantial residual trends.The validation showed that the prediction accuracy of the integrated NLME-TE improved as the number of sectional diameter measurements increased,with at least a 35%reduction in volume estimate error.For practical implementation,two calibration sectional diameter measurements taken from the butt end per tree are recommended.This approach would reduce measurement effort and cost while improving model performance.展开更多

Objective:To investigate the spatial gradient of intraoperative impedance across the cochlear electrode array in pediatric cochlear implant recipients and assess its potential as a physiological indicator for the elec...Objective:To investigate the spatial gradient of intraoperative impedance across the cochlear electrode array in pediatric cochlear implant recipients and assess its potential as a physiological indicator for the electrode-neural interface.Methods:A prospective observational study involving 56 pediatric patients underwent cochlear implantation with Cochlear Nucleus devices.Intraoperative polarized impedance and electrically evoked compound action potential(ECAP)threshold were recorded across all 1232 electrodes using AutoNRT software.Eight electrodes with open-or short-circuit were excluded,leaving 1,224 for analysis.Impedance values were categorized by cochlear region(basal,middle,apical),and electrodes with elevated impedance(10-20 kΩ)were analyzed for regional distribution and clinical relevance.Data were analyzed for spatial patterns and correlation with the ECAP threshold profiles.Results:A consistent basal-to-apical increase in impedance was observed(7.7±1.9,9.2±1.4,10.8±1.5 kΩ;p<0.001).Impedance and ECAP threshold were weakly correlated(ρ=-0.20,p<0.001;β=-1.26,p<0.001),with a positive association in the apical region(ρ=0.12,p=0.048).Electrodes with higher impedance(1020 kΩ)were less likely to show elevated or absent TNRT(OR=0.175,p=0.02).The impedance gradient persisted across age groups and was significantly correlated with ECAP threshold patterns.Conclusion:Intraoperative impedance monitoring reveals a strong and physiologically consistent gradient,with higher values in apical electrodes.This gradient reflects anatomical and tissue interface variations,which may offer a valuable physiological indicator for intraoperative electrode positioning and neural interface integrity.展开更多

BACKGROUND:To investigate the mechanism underlying the basal forebrain-DMN/solv cholinergic projection-induced attenuation of brain injury after cardiopulmonary resuscitation(CPR).METHODS:Forty-six male Sprague-Dawley...BACKGROUND:To investigate the mechanism underlying the basal forebrain-DMN/solv cholinergic projection-induced attenuation of brain injury after cardiopulmonary resuscitation(CPR).METHODS:Forty-six male Sprague-Dawley rats were randomly divided into five groups:the sham group(n=6),the return of spontaneous circulation(ROSC) group(n=10),the optogenetic activation(CHAT-Light-CHR2) group(n=10),the optogenetic inhibition(CHAT-Light-NpHR) group(n=10),and the optogenetic activation combined with left cervical vagotomy(CHAT-Light-CHR2 + Lc VGX) group(n=10).Excitatory(CHR2) or inhibitory(NpHR) optogenetic viruses were injected into the basal forebrain(BF) of rats,followed by the implantation of ceramic ferrules.After three weeks of viral expression,the sham group received tracheotomy and catheterization only,the ROSC group underwent tracheotomy,asphyxial cardiac arrest,and CPR,and the CHR2 and NpHR groups received post-ROSC optogenetic activation or inhibition,respectively.The CHAT-Light-CHR2 + LcVGX group was pretreated with left cervical vagotomy followed by post-ROSC activation.Post-ROSC assessments included neurofunctional deficit score(NDS),histopathology(HE/Nissl/TUNEL staining and CD11b microglial activation) in the hippocampal CA1/prefrontal cortex,serum cytokines(IL-1β,IL-6 and TNF-α),and whole-brain immunofluorescence(c-Fos/CHAT) for neuronal activation mapping.RESULTS:Compared with the rats of sham group,rats of the ROSC group presented reduced NDS,neuronal loss,increased apoptosis,elevated CD11b expression,and increased cytokine levels.CHAT-CHR2 activation improved NDS,reduced neuronal loss and apoptosis,and decreased CD11b expression and TNF-α levels.CHAT-NpHR inhibition caused no improvement in NDS but exacerbated neuronal loss and CD11b expression elevation.CHAT-CHR2+LcVGX reversed these protective effects.Whole-brain immunofluorescence staining revealed that optogenetic activation of cholinergic neurons in the BF of the CHAT-Light-CHR2 group excited neurons in the DMN/solv region,which were identified as dopaminergic neurons.CONCLUSION:Cholinergic projections from the basal forebrain-DMN/solv may alleviate systemic and neuroinflammatory responses through the cholinergic anti-inflammatory pathway and mitigate brain injury following CPR.展开更多
Tiller number represents a critical agronomic trait determining sugarcane yield.Strigolactones(SLs) are plant hormones regulating plant architecture.D27,an essential enzyme in the SL biosynthetic pathway,catalyzes a r...Tiller number represents a critical agronomic trait determining sugarcane yield.Strigolactones(SLs) are plant hormones regulating plant architecture.D27,an essential enzyme in the SL biosynthetic pathway,catalyzes a reversible isomerization reaction.ScD27.2,the D27 homolog in sugarcane,contains abiotic stress-responsive elements in its promoter,suggesting its potential importance in SL biosynthesis and stress tolerance.ScD27.2 potentially optimizes sugarcane agronomic traits,particularly tiller number and yield.Understanding its mechanisms will advance the development of high-yielding,stresstolerant sugarcane varieties.To investigate the role of D27 in sugarcane tillering,the key carotene isomerase gene ScD 27.2 was silenced(via RNAi) and overexpressed(OE) in sugarcane cultivar ‘XTT22' plantlets.ScD27-RNAi-2 sugarcane exhibited decreased ScD27.2 expression and increased tiller numbers compared to the wild type ‘XTT22'.Conversely,overexpression lines(ScD27-OE-1,ScD27-OE-5,and ScD27-OE-9) showed increased ScD27.2 expression and decreased tiller numbers.ScD27-OE-9 demonstrated notable lateral bud germination,while ScD27-RNAi-2 exhibited reduced drought tolerance.Under normal light and water management conditions,transgenic sugarcane plants showed significant differences in tiller number and plant height.During extended drought conditions,ScD27-RNAi-2 height was significantly reduced compared to wild type ‘XTT22' and ScD27-OE-9,manifesting a dwarf,multi-tiller phenotype.Additionally,ScD27-RNAi-2 showed significantly reduced SLs content.These findings demonstrate that ScD27.2 regulates tillering under drought stress,likely through SL biosynthesis,and that its drought response may be mediated by the transcription factor ScMYB44.展开更多

Polyfluoroalkyl substances(PFAS)have emerged as persistent environmental contaminants because of their chemical stability,degradation-resistance and bioaccumulation potential.However,current studies mainly focus on th...Polyfluoroalkyl substances(PFAS)have emerged as persistent environmental contaminants because of their chemical stability,degradation-resistance and bioaccumulation potential.However,current studies mainly focus on the toxicity of single PFAS such as perfluorooctanoic acid(PFOA)and perfluorobutanoic acid(PFBA),the knowledge of their combined effects is relatively limited.In this study,we explored the immune response of the gut in large yellow croaker(Larimichthys crocea)under the combined stress of PFOA and PFBA.Histologicalanalyses revealed that the combined effect induced intestinal vacuolization and decreased the length of intestinal villi.And it significantly activated pro-inflammatory pathways with marked upregulation of tnfα,il1β,il6 and myd88 expressions,particularly after 14 days of exposure.Gut microbiota analysis revealed substantial dysbiosis,including 1)reduced alpha diversity,2)increased abundance of potential pathogenic taxa(Proteobacteria and Spirochaetota),and 3)depletion of beneficial Firmicutes.PICRUSt-based functional prediction indicated temporal metabolic shifts,with upregulation of DNA repair pathways at day 3 and enhanced bacterial motility protein activity at days 7 and 14 of post-exposure.The Pearson correlation analysis further indicated that these immune genes had significant positive correlations with Vibrio and Brevinema,and negative correlations with Streptococcus.Our present study will provide novel insights into the microbiome-mediated immunomodulation in the larger yellow croaker exposed to combined PFAS,which will be helpful for healthy farming of economically important marine species.展开更多
Tree growth variability is a key determinant of forest stabilities.Previous studies have shown that recent climate change has increased variability in tree growth,while others have challenged this viewpoint,leading to...Tree growth variability is a key determinant of forest stabilities.Previous studies have shown that recent climate change has increased variability in tree growth,while others have challenged this viewpoint,leading to ongoing debate in this field.Moreover,gaps remain in understanding the climatic mechanisms driving increased tree growth variability,particularly for species simultaneously limited by multiple climate factors.In this study,we assessed the temporal trends in variability of Picea purpurea radial growth and its linkage with growth-climate sensitivity utilizing dendrochronological methods.Our results revealed a significant increase in P.purpurea radial growth variability from 1960 to 2020,as indicated by continuous rises in the standard deviation,coefficient of variation,and mean sensitivity of tree-ring width indices.The increased frequency of extreme growth declines further supported this finding.Furthermore,moisture condition in July was identified as a key limiting factor of P.purpurea growth.Notably,the strengthening relationship between tree-ring width indices and vapor pressure deficit(VPD)suggests that the moisture sensitivity for P.purpurea growth has increased over the period 1960-2020.This enhanced sensitivity to VPD,whose interannual variability has also increased synchronously,may have contributed to the rise in P.purpurea growth variability.Additionally,the maximum temperature in May was positively correlated with P.purpurea growth;however,there is little evidence that this factor contributed to the observed increase in growth variability.These findings provide new insights into P.purpurea growth trends and improve our understanding of the potential future impacts of climate change on forest ecosystems.展开更多
Field-effect nanofluidic transistors(FENTs),biomimicking the structure and functionality of neuron,act as biological transistors with the ability to gate switching responses to external stimuli.The switching ratio has...Field-effect nanofluidic transistors(FENTs),biomimicking the structure and functionality of neuron,act as biological transistors with the ability to gate switching responses to external stimuli.The switching ratio has been verified to evaluate the performance of FENTs,but until recently,the response time,another crucial indicator,has been ignored.Employing finite-element method,we investigated the relationship among gate charge,switching ratio and response time by divisionally manipulating gate charge,including entrance surface and the surface of confinement space,for ion transport to optimize switching capability.The dual-split gate charge on FENTs exhibits synergistic effect on switching response.Based on the two regional gate charge on FENTs,multivalence ions in lower concentration,high aspect ratio and single channel show higher switching ratio but longer response time compared to monovalent ions.The findings highlight the necessity of balancing these two signals in FENTs and offer insights for optimizing their design and expanding applications to dual-signal-detection iontronics.展开更多

文摘In this study,a straightforward one-step hydrothermal method was successfully utilized to synthesize the solid solution Na_(0.9)Mg_(0.45)Ti_(3.55)O_(8)-Na_(2)Ni_(2)Ti_(6)O_(16)(NNMTO-x),where x denotes the molar percentage of Na_(2)Ni_(2)Ti_(6)O_(16)(NNTO)within Na_(0.9)Mg_(0.45)Ti_(3.55)O_(8)(NMTO),with x values of 10,20,30,40,and 50.Both XPS(X-ray Photoelectron Spectroscopy)and EDX(Energy Dispersive X-ray Spectroscopy)analyses unequivocally validated the formation of the NNMTO-x solid solutions.It was observed that when x is below 40,the NNMTO-x solid solution retains the structural characteristics of the original NMTO.However,beyond this threshold,significant alterations in crystal morphology were noted,accompanied by a noticeable decline in photocatalytic activity.Notably,the absorption edge of NNMTO-x(x<40)exhibited a shift towards the visible-light spectrum,thereby substantially broadening the absorption range.The findings highlight that NNMTO-30 possesses the most pronounced photocatalytic activity for the reduction of CO_(2).Specifically,after a 6 h irradiation period,the production rates of CO and CH_(4)were recorded at 42.38 and 1.47μmol/g,respectively.This investigation provides pivotal insights that are instrumental in the advancement of highly efficient and stable photocatalysts tailored for CO_(2)reduction processes.

文摘Herein,antibacterial silver‑doped fluorescent carbon dots(Ag‑CDs)were synthesized through a stepwise hydrothermal method,with polyethyleneimine(PEI),citric acid(CA),and silver nitrate(AgNO3)serving as precursors.The applicability and antimicrobial efficacy of these nanomaterials were systematically investigated for metal ion sensing.Experimental evidence demonstrated that the Ag‑CDs exhibited a pronounced fluorescence quenching response toward ferric ions(Fe^(3+)),enabling their quantitative determination via a linear concentration‑dependent relationship.These Ag‑CDs exhibited significant inhibitory effects on biofilm growth and disruption for both Escherichia coli and Staphylococcus aureus.Mechanism investigations indicate that Ag‑CDs induced the death of Escherichia coli and Pseudomonas aeruginosa by disrupting their bacterial morphology and structure,triggering the generation of intracellular reactive oxygen species(ROS),and impairing their antioxidant defense system.
文摘BACKGROUND:Breast hyperplasia is a common benign breast disease mainly caused by endocrine disorders,manifested as abnormal hyperplasia of breast tissue.In recent years,traditional Chinese medicine compounds and probiotics have shown good potential in regulating the endocrine system and improving the intestinal microecology,providing new ideas for the treatment of breast hyperplasia.OBJECTIVE:To explore the effects and mechanisms of traditional Chinese medicine compounds and fermented probiotic compounds on breast hyperplasia in mice,providing new theoretical and experimental bases for the clinical treatment and prevention of breast hyperplasia.METHODS:(1)Network pharmacology tools were used to predict the anti-breast-hyperplasia activity of Herba Gueldenstaedtiae(Euphorbia humifusa),as well as its potential targets and signaling pathways.The databases included:TCMSP,OMIM,GeneCards database,UniProt website,Venny2.1.0 website,Metascape,HERB website,and STRING database,all of which are open-access databases.Network pharmacology can predict and screen key information such as the targets corresponding to the active ingredients of traditional Chinese medicine,disease targets,and action pathways through network analysis and computer-system analysis.Therefore,it has been increasingly widely used in the research of traditional Chinese medicine.(2)A breast hyperplasia model was induced in mice by injecting estrogen and progesterone.Mice in the normal blank group were injected intraperitoneally with normal saline every day.Mice in the model group and drugadministration groups were injected intraperitoneally with estradiol benzoate injection at a concentration of 0.5 mg/kg every day for 25 days.From the 26th day,the injection of estradiol benzoate injection was stopped.Mice in the normal blank group were injected intramuscularly with normal saline every day,and mice in the model group and drug-administration groups were injected intramuscularly with progesterone injection at a concentration of 5 mg/kg for 5 days.After the model was established,each group was given drugs respectively.The normal blank group and the model group were gavaged with 0.2 mL/d of normal saline;the positive blank group(Xiaozheng Pill group)was gavaged with an aqueous solution of Xiaozheng Pill at 0.9 mg/g;the low-,medium-and high-dose groups of Compound Herba Gueldenstaedtiae were gavaged with an aqueous solution of the compound medicine at 0.75,1.5,and 3.0 mg/(g·d)respectively;the low-,medium-and high-dose groups of traditional Chinese medicine-bacteria fermentation were gavaged with an aqueous solution of the compound medicine at 0.75,1.5,and 3.0 mg/(g·d)respectively.The administration was continuous for 30 days.RESULTS AND CONCLUSION:(1)The results of network pharmacology research showed that the Compound Herba Gueldenstaedtiae(Euphorbia humifusa)contained 46 active ingredients,which were related to 1213 potential targets.After comparison with 588 known breast-hyperplasia targets,it was speculated that 50 of these targets might be related to the direct effect of the compound on breast hyperplasia.(2)After drug intervention,there was no significant change in the high-dose group of Compound Herba Gueldenstaedtiae compared with the normal blank group.The liver indicators of the other intervention groups all significantly decreased(P<0.05).(3)In terms of kidney and uterine indicators,the medium-dose group of Compound Herba Gueldenstaedtiae decreased significantly compared with the normal blank group(P<0.05).In terms of the uterine index,the model group increased significantly compared with the normal blank group(P<0.01).(4)After 1-month drug treatment,the number of lobules and acini in the breast tissue of the Xiaozheng Pill group,the low,medium,and high-dose group of Compound Herba Gueldenstaedtiae,the low,medium,and highdose groups of traditional Chinese medicine-bacteria fermentation decreased,and the duct openings narrowed.With the increase of drug dose,diffuse hyperplasia of breast tissue was significantly improved.(5)The ELISA results showed that compared with the model group,the estrogen level was lower in the medium-dose group of traditional Chinese medicine-bacteria fermentation after the intervention(P<0.05).In addition,the follicle-stimulating hormone level in the low-dose group of Compound Herba Gueldenstaedtiae was lower than that of the model group(P<0.05).(6)The intervention in the mouse model led to changes in the abundance of short chain fatty acids and intestinal flora in all groups.To conclude,the Compound Herba Gueldenstaedtiae and its probiotic fermentation products significantly improved mammary gland hyperplasia in mice by regulating hormone levels,improving the structure of the gut microbiota,and increasing the content of shortchain fatty acids,providing new ideas and potential sources of drugs for the treatment of breast hyperplasia.

文摘Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to environmental changes upon entry into oral biofilms are reviewed.Supra-gingival biofilms are characterized by acidic pH,the presence of bacterial enzymes,and the development of hypoxia in deeper layers.Sub-gingival biofilms are slightly alkaline,with hypoxia occurring over their entire depth.Upon entering biofilms,negatively charged,pH-and/or hypoxia-responsive nanocarriers become positively charged.This charge reversal leads to electrostatic double-layer attraction between positively charged nanocarriers towards negatively charged,waterfilled channel walls in biofilms,enhancing their accumulation in a biofilm.Degradation of bacterial enzyme-responsive nanocarriers causes in-biofilm release of antimicrobial cargo,yielding higher local antimicrobial concentrations than can be achieved through their direct,oral administration without harming soft tissues.Enhanced antibiofilm activity after in-biofilm antimicrobial release from biofilm-responsive micelles and liposomes has been demonstrated in vitro towards single-species Streptococcus mutans and Staphylococcus aureus biofilms or in vivo using specific-pathogen-free rodents inoculated with selected pathogens.This preferential antibacterial activity regulated the microbial composition of ex vivo human oral biofilm towards a more healthy microbiome composition.Although clinical confirmation is limited,the potential benefits of stimuli-responsive,antimicrobial-loaded nanocarriers for oral biofilm control and microbiome restoration are worth further investigation towards clinical translation.

文摘The interplay between gut microbiota and host health has attracted significant interest in the animal science community.Maintaining gut microbiota homeostasis by supplementing probiotics to treat clinical conditions like calf diarrhea is an emerging area of research nowadays because of increased concerns regarding antimicrobial resistance(AMR)and drug residues in animal products.Probiotics reduce the incidence of calf diarrhea by increasing the gut microbiota diversity and richness with more commensal bacteria such as Lactobacillus and Bifidobacterium that produce antimicrobial compounds,as well as modulating the immune response by increasing cytokines,Interleukin-2(IL-2),IL-4,IL-6,IL-10,and reducing tumor necrosis factor-α(TNF-α),by increasing production of antibodies,especially immunoglobulin E(Ig E),also Ig G,differentiating naive Th lymphocytes(Tho)into Th1,hence stimulate innate immunity and prime the adaptive immune response.Specific probiotic strains of bacteria and yeast(Saccharomyces cerevisiae)derived probiotics maintain the integrity of the intestinal barrier.In this review,data are being organized to address the role of probiotics in treating calf diarrhea by modulating gut microbiota and stimulating an immune response against notorious pathogens,to present animal and veterinary scientists and nutritionists with a new concept to treat infectious diseases from the perspective of the gut microbiota,increasing animal health,performance,and welfare.In conclusion,health status and gut microbiome are strongly interlinked.Research data indicated a significant reduction in the incidence of diarrhea after probiotic administration.If interrelations between probiotics and existing gut microbiota are explored more quantitatively,novel antibiotic substitutes can emerge in the future.

文摘Understanding the aerodynamic and dynamic characteristics of unloaded freight trains in crosswinds is pivotal for ensuring their operational safety and reliability.The dynamic performance of unloaded gondola cars under varying windbreak heights is therefore investigated in this study,revealing distinct differences in lateral stability and safety indicators,and enabling the determination of an optimal windbreak height.A 3D unsteady aerodynamic model was developed using the improved delayed detached eddy simulation(IDDES)method and an overset numerical mesh.Also leveraging a multi-body dynamics(MBD)model of a three-wagon freight car configuration,we investigate time-averaged aerodynamic forces,transient flow field distributions,and nonlinear dynamic responses.Parametric analyses reveal a non-monotonic relationship between the height of the windbreak and the stability of the train.A windbreak with a critical height of 2 m(0.74 relative to the car body height)results in 76%,64%,and 81%lower values of the derailment coefficient CD,wheel unloading ratio R,and overturning coefficient C_(O),respectively.Notably,when the height of the windbreak exceeds 2 m,vortices within the gondola induce an adverse pressure coefficient distribution(C_(p)=−2.17)on the leeward internal wall,intensifying the lateral force and overturning moment.Furthermore,frequency-domain analysis reveals that the lateral sway and overturning vibration mode are associated with low-frequency(1.61 Hz)lateral vibrations under crosswind conditions.This study provides a theoretical foundation for the design and optimization of railway windbreaks.

文摘Reinforced concrete(RC)columns are often subjected to off-central explosion due to the uncertainty of blast locations.However,few studies have focused on the dynamic response of RC columns under offcentral explosions.A field blast experiment was conducted under close-in explosion with varying detonation offset distances(0 m,0.5 m,and 1 m),the overpressure load and dynamic responses of the full-scale RC columns were measured.Compared with the centrally detonated condition,a relative offset distance of 1.67 decreases the maximum and residual deflections of the RC column by 16.8%and 21.4%,respectively,while increasing the maximum and residual support rotations by 24.7%and 17.8%.Based on the experimental results,a theoretical model was proposed that considers the detonation location and charge mass,boundary conditions,axial compression ratio and material properties.The theoretical model exhibited good agreement with the experimental results,with prediction errors below 10%for both maximum and residual deflection.The effects of parameters were analyzed,and it indicated that an increase in offset distance results in decreased maximum and residual deflections but an increased support angle,thereby exacerbating damage.Higher axial load ratio,span-depth ratio,and longitudinal reinforcement ratio reduce both deflections and support angle.Additionally,a rapid method to predict the maximum and residual deflection of RC columns under off-central blast loading was also proposed based on the Generalized Regression Neural Network(GRNN).Eleven features which related to the RC column properties and the blast characteristics were used in the training process of GRNN,and accurate predictions were achieved with prediction errors within 20%.This study fills the gap in predicting the dynamic response of RC columns under off-central explosion,providing valuable references for blast-resistant design.
文摘Eucalyptus(Eucalyptus camaldulensis Dehnh.)is an important exotic species in northern Nigeria commonly used for poles and timber.Sustainable management of this resource would require quantifying its volume.Stem taper equations are one of the main and most efficient methods for estimating stem volume to any merchantable limit of a species.There is currently no taper equation for Eucalyptus species in Nigeria.Therefore,this study developed taper equations for E.camaldulensis in northern Nigeria.Data for this study were obtained from a private plantation in Jalingo Local Government Area,Taraba State,Nigeria.68 trees were felled and sectioned into 1-m bolt across the stem to a merchantable limit of 5 cm,which were used as the fitting dataset.An additional 22 trees were felled and used to validate the taper equations for stem volume estimation.Seven taper equations were initially fitted to the dataset using nonlinear least squares.The best taper equation was then refitted using a nonlinear mixed-effects approach and calibrated using diameters of one to five sections from the butt end.The taper equations were numerically integrated to obtain the stem volume,which was compared with empirical volume equations.The result shows that the Kozak(Can J For Res 27(5):619-629.10.1139/x97-011,1997)equation,which included eight parameters,provided the best fit for predicting section diameters for under and over bark.The mixed-effects taper equation(NLME-TE)explained most stem diameter variations in the fitting dataset(pseudo-R2:0.986-0.987;RMSE:0.547-0.578 cm)without substantial residual trends.The validation showed that the prediction accuracy of the integrated NLME-TE improved as the number of sectional diameter measurements increased,with at least a 35%reduction in volume estimate error.For practical implementation,two calibration sectional diameter measurements taken from the butt end per tree are recommended.This approach would reduce measurement effort and cost while improving model performance.

文摘Colorectal cancer remains one of the leading causes of morbidity and mortality worldwide.Despite notable advances in early detection and therapeutic strategies,the molecular mechanisms underlying tumor survival,chemotherapy resistance,and metastasis are not yet fully understood.MicroRNAs(miRNAs)have emerged as pivotal regulators of cancer development,as they modulate gene expression and orchestrate key signaling pathways.However,the epigenetic mechanisms that control miRNA expression and their downstream gene targets remain largely unclear.In this review,we highlight the critical role of the colorectal cancer microenvironment in influencing miRNA expression and discuss how this regulation contributes to tumorigenesis.A better understanding of these processes may lead to the identification of novel therapeutic targets and strategies to prevent recurrence.
文摘Objective:To investigate the spatial gradient of intraoperative impedance across the cochlear electrode array in pediatric cochlear implant recipients and assess its potential as a physiological indicator for the electrode-neural interface.Methods:A prospective observational study involving 56 pediatric patients underwent cochlear implantation with Cochlear Nucleus devices.Intraoperative polarized impedance and electrically evoked compound action potential(ECAP)threshold were recorded across all 1232 electrodes using AutoNRT software.Eight electrodes with open-or short-circuit were excluded,leaving 1,224 for analysis.Impedance values were categorized by cochlear region(basal,middle,apical),and electrodes with elevated impedance(10-20 kΩ)were analyzed for regional distribution and clinical relevance.Data were analyzed for spatial patterns and correlation with the ECAP threshold profiles.Results:A consistent basal-to-apical increase in impedance was observed(7.7±1.9,9.2±1.4,10.8±1.5 kΩ;p<0.001).Impedance and ECAP threshold were weakly correlated(ρ=-0.20,p<0.001;β=-1.26,p<0.001),with a positive association in the apical region(ρ=0.12,p=0.048).Electrodes with higher impedance(1020 kΩ)were less likely to show elevated or absent TNRT(OR=0.175,p=0.02).The impedance gradient persisted across age groups and was significantly correlated with ECAP threshold patterns.Conclusion:Intraoperative impedance monitoring reveals a strong and physiologically consistent gradient,with higher values in apical electrodes.This gradient reflects anatomical and tissue interface variations,which may offer a valuable physiological indicator for intraoperative electrode positioning and neural interface integrity.
文摘This study examines the dynamic response of two adjacent 9-and 20-story benchmark steel buildings subjected to six near-fault earthquake records.Two-dimensional numerical models were employed to account for the complexities of structure-soil-structure interaction(SSSI).The research focuses on the separation gap between the buildings and the effects of pounding while considering Fixed Base(FB)and SSSI models,evaluated according to UBC 94 and ASCE 7-16 seismic codes.Key findings reveal that pounding occurs with the UBC 94 separation gap when earthquake frequency aligns with system frequency,leading to increased column stresses in the 9-story building.In contrast,the ASCE 7-16 standard effectively prevents pounding in both the FB and SSSI models.Additionally,drifts and displacements of lower floors in SSSI models exceed the allowable limits of ASCE 7-16,underscoring the impact of soil-structure interaction on seismic response.

文摘BACKGROUND:To investigate the mechanism underlying the basal forebrain-DMN/solv cholinergic projection-induced attenuation of brain injury after cardiopulmonary resuscitation(CPR).METHODS:Forty-six male Sprague-Dawley rats were randomly divided into five groups:the sham group(n=6),the return of spontaneous circulation(ROSC) group(n=10),the optogenetic activation(CHAT-Light-CHR2) group(n=10),the optogenetic inhibition(CHAT-Light-NpHR) group(n=10),and the optogenetic activation combined with left cervical vagotomy(CHAT-Light-CHR2 + Lc VGX) group(n=10).Excitatory(CHR2) or inhibitory(NpHR) optogenetic viruses were injected into the basal forebrain(BF) of rats,followed by the implantation of ceramic ferrules.After three weeks of viral expression,the sham group received tracheotomy and catheterization only,the ROSC group underwent tracheotomy,asphyxial cardiac arrest,and CPR,and the CHR2 and NpHR groups received post-ROSC optogenetic activation or inhibition,respectively.The CHAT-Light-CHR2 + LcVGX group was pretreated with left cervical vagotomy followed by post-ROSC activation.Post-ROSC assessments included neurofunctional deficit score(NDS),histopathology(HE/Nissl/TUNEL staining and CD11b microglial activation) in the hippocampal CA1/prefrontal cortex,serum cytokines(IL-1β,IL-6 and TNF-α),and whole-brain immunofluorescence(c-Fos/CHAT) for neuronal activation mapping.RESULTS:Compared with the rats of sham group,rats of the ROSC group presented reduced NDS,neuronal loss,increased apoptosis,elevated CD11b expression,and increased cytokine levels.CHAT-CHR2 activation improved NDS,reduced neuronal loss and apoptosis,and decreased CD11b expression and TNF-α levels.CHAT-NpHR inhibition caused no improvement in NDS but exacerbated neuronal loss and CD11b expression elevation.CHAT-CHR2+LcVGX reversed these protective effects.Whole-brain immunofluorescence staining revealed that optogenetic activation of cholinergic neurons in the BF of the CHAT-Light-CHR2 group excited neurons in the DMN/solv region,which were identified as dopaminergic neurons.CONCLUSION:Cholinergic projections from the basal forebrain-DMN/solv may alleviate systemic and neuroinflammatory responses through the cholinergic anti-inflammatory pathway and mitigate brain injury following CPR.

文摘Tiller number represents a critical agronomic trait determining sugarcane yield.Strigolactones(SLs) are plant hormones regulating plant architecture.D27,an essential enzyme in the SL biosynthetic pathway,catalyzes a reversible isomerization reaction.ScD27.2,the D27 homolog in sugarcane,contains abiotic stress-responsive elements in its promoter,suggesting its potential importance in SL biosynthesis and stress tolerance.ScD27.2 potentially optimizes sugarcane agronomic traits,particularly tiller number and yield.Understanding its mechanisms will advance the development of high-yielding,stresstolerant sugarcane varieties.To investigate the role of D27 in sugarcane tillering,the key carotene isomerase gene ScD 27.2 was silenced(via RNAi) and overexpressed(OE) in sugarcane cultivar ‘XTT22' plantlets.ScD27-RNAi-2 sugarcane exhibited decreased ScD27.2 expression and increased tiller numbers compared to the wild type ‘XTT22'.Conversely,overexpression lines(ScD27-OE-1,ScD27-OE-5,and ScD27-OE-9) showed increased ScD27.2 expression and decreased tiller numbers.ScD27-OE-9 demonstrated notable lateral bud germination,while ScD27-RNAi-2 exhibited reduced drought tolerance.Under normal light and water management conditions,transgenic sugarcane plants showed significant differences in tiller number and plant height.During extended drought conditions,ScD27-RNAi-2 height was significantly reduced compared to wild type ‘XTT22' and ScD27-OE-9,manifesting a dwarf,multi-tiller phenotype.Additionally,ScD27-RNAi-2 showed significantly reduced SLs content.These findings demonstrate that ScD27.2 regulates tillering under drought stress,likely through SL biosynthesis,and that its drought response may be mediated by the transcription factor ScMYB44.

文摘Polyfluoroalkyl substances(PFAS)have emerged as persistent environmental contaminants because of their chemical stability,degradation-resistance and bioaccumulation potential.However,current studies mainly focus on the toxicity of single PFAS such as perfluorooctanoic acid(PFOA)and perfluorobutanoic acid(PFBA),the knowledge of their combined effects is relatively limited.In this study,we explored the immune response of the gut in large yellow croaker(Larimichthys crocea)under the combined stress of PFOA and PFBA.Histologicalanalyses revealed that the combined effect induced intestinal vacuolization and decreased the length of intestinal villi.And it significantly activated pro-inflammatory pathways with marked upregulation of tnfα,il1β,il6 and myd88 expressions,particularly after 14 days of exposure.Gut microbiota analysis revealed substantial dysbiosis,including 1)reduced alpha diversity,2)increased abundance of potential pathogenic taxa(Proteobacteria and Spirochaetota),and 3)depletion of beneficial Firmicutes.PICRUSt-based functional prediction indicated temporal metabolic shifts,with upregulation of DNA repair pathways at day 3 and enhanced bacterial motility protein activity at days 7 and 14 of post-exposure.The Pearson correlation analysis further indicated that these immune genes had significant positive correlations with Vibrio and Brevinema,and negative correlations with Streptococcus.Our present study will provide novel insights into the microbiome-mediated immunomodulation in the larger yellow croaker exposed to combined PFAS,which will be helpful for healthy farming of economically important marine species.

文摘Tree growth variability is a key determinant of forest stabilities.Previous studies have shown that recent climate change has increased variability in tree growth,while others have challenged this viewpoint,leading to ongoing debate in this field.Moreover,gaps remain in understanding the climatic mechanisms driving increased tree growth variability,particularly for species simultaneously limited by multiple climate factors.In this study,we assessed the temporal trends in variability of Picea purpurea radial growth and its linkage with growth-climate sensitivity utilizing dendrochronological methods.Our results revealed a significant increase in P.purpurea radial growth variability from 1960 to 2020,as indicated by continuous rises in the standard deviation,coefficient of variation,and mean sensitivity of tree-ring width indices.The increased frequency of extreme growth declines further supported this finding.Furthermore,moisture condition in July was identified as a key limiting factor of P.purpurea growth.Notably,the strengthening relationship between tree-ring width indices and vapor pressure deficit(VPD)suggests that the moisture sensitivity for P.purpurea growth has increased over the period 1960-2020.This enhanced sensitivity to VPD,whose interannual variability has also increased synchronously,may have contributed to the rise in P.purpurea growth variability.Additionally,the maximum temperature in May was positively correlated with P.purpurea growth;however,there is little evidence that this factor contributed to the observed increase in growth variability.These findings provide new insights into P.purpurea growth trends and improve our understanding of the potential future impacts of climate change on forest ecosystems.

文摘Field-effect nanofluidic transistors(FENTs),biomimicking the structure and functionality of neuron,act as biological transistors with the ability to gate switching responses to external stimuli.The switching ratio has been verified to evaluate the performance of FENTs,but until recently,the response time,another crucial indicator,has been ignored.Employing finite-element method,we investigated the relationship among gate charge,switching ratio and response time by divisionally manipulating gate charge,including entrance surface and the surface of confinement space,for ion transport to optimize switching capability.The dual-split gate charge on FENTs exhibits synergistic effect on switching response.Based on the two regional gate charge on FENTs,multivalence ions in lower concentration,high aspect ratio and single channel show higher switching ratio but longer response time compared to monovalent ions.The findings highlight the necessity of balancing these two signals in FENTs and offer insights for optimizing their design and expanding applications to dual-signal-detection iontronics.
