Given the ...Epilepsy is a leading cause of disability and mortality worldwide. However, despite the availability of more than 20 antiseizure medications, more than one-third of patients continue to experience seizures. Given the urgent need to explore new treatment strategies for epilepsy, recent research has highlighted the potential of targeting gliosis, metabolic disturbances, and neural circuit abnormalities as therapeutic strategies. Astrocytes, the largest group of nonneuronal cells in the central nervous system, play several crucial roles in maintaining ionic and energy metabolic homeostasis in neurons, regulating neurotransmitter levels, and modulating synaptic plasticity. This article briefly reviews the critical role of astrocytes in maintaining balance within the central nervous system. Building on previous research, we discuss how astrocyte dysfunction contributes to the onset and progression of epilepsy through four key aspects: the imbalance between excitatory and inhibitory neuronal signaling, dysregulation of metabolic homeostasis in the neuronal microenvironment, neuroinflammation, and the formation of abnormal neural circuits. We summarize relevant basic research conducted over the past 5 years that has focused on modulating astrocytes as a therapeutic approach for epilepsy. We categorize the therapeutic targets proposed by these studies into four areas: restoration of the excitation–inhibition balance, reestablishment of metabolic homeostasis, modulation of immune and inflammatory responses, and reconstruction of abnormal neural circuits. These targets correspond to the pathophysiological mechanisms by which astrocytes contribute to epilepsy. Additionally, we need to consider the potential challenges and limitations of translating these identified therapeutic targets into clinical treatments. These limitations arise from interspecies differences between humans and animal models, as well as the complex comorbidities associated with epilepsy in humans. We also highlight valuable future research directions worth exploring in the treatment of epilepsy and the regulation of astrocytes, such as gene therapy and imaging strategies. The findings presented in this review may help open new therapeutic avenues for patients with drugresistant epilepsy and for those suffering from other central nervous system disorders associated with astrocytic dysfunction.展开更多

文摘Enzyme-Induced Carbonate Precipitation(EICP)is an innovative technique to improve soil strength and reduce permeability. However, the use of EICP for reinforcing underwater sand beds remains largely unexplored. To advance EICP implementation in various geotechnical applications, this paper develops a model box system to investigate the effectiveness of the EICP technique in reinforcing underwater sand beds.An"injection-extraction"system is designed to facilitate the flow of the EICP solution through underwater sand layers.Key parameters, including conductivity,pH,and Ca^(2+)concentration of the solution, are measured and analyzed.Electrical resistivity tomography(ERT)is utilized to evaluate the reinforcement effect in the underwater sand bed.The permeability of the model is tested to verify the feasibility of EICP technology for strengthening underwater sands.Furthermore, scanning electron microscope(SEM)is performed to investigate the growth mechanisms of calcium carbonate(CaCO_(3))crystals.The results show that the permeability of the model decreases from 1.28×10^(-2)m/s to 9.66×10^(-5)m/s,representing a reduction of approximately three orders of magnitude. This verifies that the EICP technology can greatly reduce the permeability of underwater sand beds. With increasing grouting cycles, the resistivity of the underwater sand initially decreases and then increases. This variation in sand resistivity is significantly influenced by the ion concentration in the solution, resulting in marked differences in resistivity at various depths and positions within the sand. The findings from this study offer a theoretical basis for the application of EICP technology in reinforcing seabed foundations and supporting marine infrastructure such as offshore pipelines, wind turbines, and oil platforms.

文摘Copper ions are essential for cellular function but can induce cytotoxic effects when dysregulated. This review explores the multifaceted role of copper in cancer metabolism with a focus on the novel concept of cuproptosis,a regulated form of cell death triggered by copper accumulation. The mechanisms underlying copper homeostasis are detailed, including dietary absorption, systemic distribution, and intracellular utilization.Key transporters, such as copper transporter 1(CTR1)and ATPase copper transporting alpha/b(ATP7A/B),are highlighted.Cancer cells often exhibit elevated copper levels, supporting proliferation and metastasis through pro-tumorigenic pathways.Recent studies have shown that disrupting copper homeostasis can induce cuproptosis, which is characterized by the aggregation of lipoylated mitochondrial proteins and disruption of iron-sulfur cluster biogenesis.Advances in copper-based nanotechnology have enabled targeted delivery of copper to tumors, enhancing therapeutic efficacy through synergistic effects with reactive oxygen species(ROS)generation and immunomodulation. However, the hypoxic tumor microenvironment poses significant challenges by upregulating copper-sequestering proteins and downregulating key cuproptosis mediators.Future directions include integrating multi-omics approaches to identify novel therapeutic targets and developing combination therapies to overcome hypoxia-induced resistance. This review provides a comprehensive overview of copper metabolism in cancer, emphasizing the potential of cuproptosis induction as a powerful strategy for oncologic intervention.

文摘The plant pathogenic fungus Sclerotinia sclerotiorum is the causative agent of Sclerotinia stem rot(SSR)disease in most dicotyledons.Among the various proteins involved in drug efflux or substance transport,ATP-binding cassette(ABC)transporters constitute a superfamily of membrane-bound proteins that may play a crucial role in the survival of S.sclerotiorum.However,the expression patterns and functions of ABC transporter genes in S.sclerotiorum remain largely uncharacterized. This study characterized a highly expressed S.sclerotiorum ABC transporter gene during inoculation on host plants,Ss BMR1.Silencing Ss BMR1 resulted in a significant reduction in hyphal growth, infection cushion development, sclerotia formation, and virulence.Moreover,host-induced gene silencing(HIGS)of Ss BMR1 significantly enhanced plant resistance.Transcriptome and metabolomics analyses suggested that Ss BMR1 is involved in antioxidant and toxin transport, thereby influencing fungal defense and cell rescue mechanisms. In comparison to the wild-type strain,Ss BMR1 gene-silenced transformants exhibited a diminished response to extracellar oxidative stress and a decreased exporting of antioxidant glutathione.Tolerance assays further demonstrated the crucial role of Ss BMR1 in conferring resistance to the plant antifungal substances, camalexin and brassinin,as well as certain fungicides.Furthermore,Ss BMR1 gene-silenced transformants showed enhanced repression on virulence when sprayed with camalexin and brassinin on the leaves. Thus,Ss BMR1 likely contributes to virulence by facilitating the export of antioxidant and providing resistance against antifungal agents. The findings of this study provide valuable insights that could contribute to the development of novel management techniques for SSR.

文摘Epilepsy is a leading cause of disability and mortality worldwide. The findings presented in this review may help open new therapeutic avenues for patients with drugresistant epilepsy and for those suffering from other central nervous system disorders associated with astrocytic dysfunction.

文摘In this study,an inverse design framework was established to find lightweight honeycomb structures(HCSs)with high impact resistance. The hybrid HCS,composed of re-entrant(RE)and elliptical annular re-entrant(EARE)honeycomb cells, was created by constructing arrangement matrices to achieve structural lightweight. The machine learning(ML)framework consisted of a neural network(NN)forward regression model for predicting impact resistance and a multi-objective optimization algorithm for generating high-performance designs. The surrogate of the local design space was initially realized by establishing the NN in the small sample dataset, and the active learning strategy was used to continuously extended the local optimal design until the model converged in the global space. The results indicated that the active learning strategy significantly improved the inference capability of the NN model in unknown design domains.By guiding the iteration direction of the optimization algorithm, lightweight designs with high impact resistance were identified. The energy absorption capacity of the optimal design reached 94.98%of the EARE honeycomb, while the initial peak stress and mass decreased by 28.85%and 19.91%,respectively.Furthermore,Shapley Additive Explanations(SHAP)for global explanation of the NN indicated a strong correlation between the arrangement mode of HCS and its impact resistance.By reducing the stiffness of the cells at the top boundary of the structure, the initial impact damage sustained by the structure can be significantly improved.Overall, this study proposed a general lightweight design method for array structures under impact loads, which is beneficial for the widespread application of honeycomb-based protective structures.

文摘High-entropy oxides(HEOs)have emerged as a promising class of memristive materials, characterized by entropy-stabilized crystal structures, multivalent cation coordination, and tunable defect landscapes. These intrinsic features enable forming-free resistive switching, multilevel conductance modulation, and synaptic plasticity, making HEOs attractive for neuromorphic computing. This review outlines recent progress in HEO-based memristors across materials engineering, switching mechanisms, and synaptic emulation.Particular attention is given to vacancy migration, phase transitions, and valence-state dynamics—mechanisms that underlie the switching behaviors observed in both amorphous and crystalline systems.Their relevance to neuromorphic functions such as short-term plasticity and spike-timing-dependent learning is also examined. While encouraging results have been achieved at the device level, challenges remain in conductance precision, variability control, and scalable integration.Addressing these demands a concerted effort across materials design, interface optimization, and task-aware modeling. With such integration,HEO memristors offer a compelling pathway toward energy-efficient and adaptable brain-inspired electronics.

文摘The development of highly efficient and multifunctional nanozymes holds promise for addressing the challenges posed by drugresistant bacteria. Here, copper single-atom-loaded MoS_(2) nanozymes(CuSAs/MoS_(2))were developed to effectively combat drug-resistant bacteria by synergistically integrating the triple strategies of oxidative damage,cuproptosis-like death and disruption of cell wall synthesis.Density functional theory revealed that each Cu center coordinated with three sulfur ligands, enhancing the adsorption of H_2O_2,which reduced the activation energy of the key step by 17%,thereby improving peroxidase-like(PODlike)activity.The generation of reactive oxygen species in combination with CuSAs/MoS_(2) glutathione peroxidase-like(GSH-Px-like)for glutathione scavenging resulted in an imbalance in redox homeostasis within bacteria.CuSAs/MoS_(2),which act as nanopioneers, drive oxidative stress to initiate the process of cuproptosis-like death, leading to abnormal aggregation of lipoylated proteins and inactivation of iron-sulfur cluster proteins.Moreover,CuSAs/MoS_(2) inhibited the biosynthesis of the peptidoglycan synthesis precursors D-glutamate and m-diaminopimelic acid and disrupted the peptidoglycan cross-linking process mediated by penicillin-binding proteins, effectively blocking the compensatory cell wall remodeling pathway ofβ-lactam-resistant bacteria.Overall,CuSAs/MoS_(2) with multiple functions can not only efficiently kill bacteria but also decelerate the development of bacterial resistance to combat drug-resistant bacterial infections.

文摘Aqueous zinc metal batteries(AZMBs)are promising candidates for renewable energy storage, yet their practical deployment in subzero environments remains challenging due to electrolyte freezing and dendritic growth.Although organic additives can enhance the antifreeze properties of electrolytes, their weak polarity diminishes ionic conductivity, and their flammability poses safety concerns, undermining the inherent advantages of aqueous systems.Herein,we present a cost-effective and highly stable Na_(2)SO_(4)additive introduced into a Zn(ClO_(4))2-based electrolyte to create an organic-free antifreeze electrolyte.Through Raman spectroscopy,in situ optical microscopy, densityfunctional theory computations, and molecular dynamics simulations,we demonstrate that Na+ions improve low-temperature electrolyte performance and mitigate dendrite formation by regulating uniform Zn^(2+)deposition through preferential adsorption and electrostatic interactions. As a result, the Zn||Zn cells using this electrolyte achieve a remarkable cycling life of 360 h at-40℃ with 61% depth of discharge, and the Zn||PANI cells retained an ultrahigh capacity retention of 91%even after 8000 charge/discharge cycles at-40℃.This work proposes a cost-effective and practical approach for enhancing the long-term operational stability of AZMBs in low-temperature environments.

文摘We investigated the effects of fly ash(FA)content on the mechanical properties of recycled aggregate concrete(RAC)and its regeneration potential under freeze and thaw(F-T)cycles.The physical properties of second-generation recycled concrete aggregates(RCA)were used to analyze the regeneration potential of RAC after F-T cycles.Scanning electron microscopy was used to study the interfacial transition zone microstructure of RAC after F-T cycles. Results showed that adding 20%FA to RAC significantly enhanced its mechanical properties and frost resistance. Before the F-T cycles, the compressive strength of RAC with 20%FA reached 48.3 MPa,exceeding research strength target of 40 MPa.A majority of second-generation RCA with FA had been verified to attain class Ⅲ,which enabled their practical application in non-structural projects such as backfill trenches and road pavement. However, the second-generation RCA with 20%FA can achieve class Ⅱ,making it ideal for 40 MPa structural concrete.

文摘Head-splitting is a prevalent physiological disorder in cabbage that causes substantial economic losses. However, the genetic factors and molecular mechanisms underlying head-splitting resistance remain largely unexplored. This study identified a genomic region(q NLQ3.1) for head-splitting resistance on chromosome C03 through the combination of QTLseq and GPS analysis in an F_(2) population derived from hybridizing two cabbage inbred lines,‘Dazhengfu'(ZF,susceptible) and ‘103'(resistant).Traditional genetic linkage analysis narrowed q NLQ3.1 to a 74.6 kb region.Furthermore, comparative analysis of the two parental lines using transcriptomic and metabolic profiling demonstrated the crucial role of hormones in regulating head-splitting resistance.Bol028000,encoding a homologue of Arabidopsis Cytokinin Response Factor 3(CRF3),emerged as a promising candidate for head-splitting resistance and was subsequently validated through Sanger sequencing and quantitative RT-PCR(qRT-PCR).Subcellular localisation analysis revealed that Bol028000 was mainly expressed in the nucleus.Additionally, one kompetitive allele-specific PCR(KASP) marker from Bol028000 was developed and utilized to screen 42 inbred lines. These findings enhance the theoretical understanding of head-splitting resistance and provide valuable insights for the molecular breeding of head-splitting resistant cabbages.
