A new chemically mutagenic mutant of Synechococcus PCC7942 named high-CO2 re-quiring mutant, which could grow at 4% CO2 but could not grow at air levels of CO2, was isolated. Com-parative studies on some physiological aspects of the mutant and high-cO2 growing cells (growing at 4%CO2 ) were conducted. The result showed that the mutant had lower growing rate, about 1/40th photosyn-thetic affinity to inorganic carbon, 25% lower carbonic anhydrase (CA) activity, lower quenching rate ofchlorophyll fluoresence, and about 1/2 alkalinization rate of the medium. The CA activity responses ofthe two types of cells to different concentrations of CO2 were determined. Upon the addition of inorganiccarbon (Ci), the rate of active Ci uptake described by the rate of chlorophyll fluorescence quenching ofthe mutant was obviously lower compared with that of the high-CO2 growing cells; the size of the internalinorganic carbon pool size determined by the extent of fluorescence quenching Of the mutant was alsosmaller. 展开更多

Vitamin B(VB1),including thiamin,thiamin monophosphate(TMP),and thiamin pyrophosphate(TPP),is an essential micronutrient for all living organisms.Nevertheless,the precise function of VB1 in rice remains unclear.Here,we described a VB1 auxotrophic mutant,chlorotic lethal seedling(cles)from the mutation of OsTH1,which displayed collapsed chloroplast membrane system and decreased pigment content.OsTH1 encoded a phosphomethylpyrimidine kinase/thiamin-phosphate pyrophosphorylase,and was expressed in various tissues,especially in seedlings,leaves,and young panicles.The VB1 content in cles was markedly reduced,despite an increase in the expression of VB1 synthesis genes.The decreased TPP content affected the tricarboxylic acid cycle,pentose phosphate pathway,and de novo fatty acid synthesis,leading to a reduction in fatty acids(C16:0 and C18:0)and sugars(sucrose and glucose)of cles.Additionally,irregular expression of chloroplast membrane synthesis genes led to membrane collapse.We also found that alternative splicing and translation allowed OsTH1 to be localized to both chloroplast and cytosol.Our study revealed that OsTH1 was an essential enzyme in VB1 biosynthesis and played crucial roles in seedling growth and development by participating in fatty acid and sugar metabolism,providing new perspectives on VB1 function in rice. 展开更多

Stability of base-exposed backfill roof in underhand drift-and-fill mining is crucial for the safety of those working beneath.Given the commonly used primary-and-secondary mining sequence,interfaces are formed between adjacent filled drifts,which can weaken the integrity of the backfill roof.These interfaces also lead to two common drift layouts:aligned drifts and staggered drifts.However,less attention has been paid to the interfaces and the two drift layouts were not adequately distinguished in previous studies.In this paper,the interfaces between filled drifts were firstly considered to investigate the stability of backfill roof.Failure modes and strength requirements of backfill roof in aligned and staggered drifts are comprehensively investigated by FLAC3D,with a focus on considerations of varied shear parameters of the interfaces.Results show that failure modes in aligned drifts transition from block sliding to top caving,bottom caving or sloughing as the interface cohesion increases from zero to at least half of the backfill cohesion.Further increases in interface cohesion allow aligned drifts to behave as if there are no interfaces between them.The critical stability conditions of backfill roof in aligned drifts were mostly determined by the interface strength instead of the backfill strength.However,the stability of backfill roof in staggered drifts is barely affected by the interface strength.The outcomes are expected to provide references for mining engineers to optimize drift layouts and perform cost-effective backfill roof strength design at mines using underhand drift-and-fill mining method. 展开更多

Avulsion injury of one or more spinal ventral roots induces a critical loss of motoneurons,followed by irreversible locomotor function impairment ranging from inadequate limb movement to complete paralysis of the limb.Recent surgical techniques facilitate improvement of limb function,but it remains to be determined exactly how many motoneurons are needed to survive and grow new axons to achieve sufficient muscle reinnervation.The aim of this study was to determine the minimum motoneuron quantity required to reinnervate the denervated skeletal muscles of the limb and produce a functionally satisfactory locomotor pattern.Since none of the commercially available methods and equipment were able to provide a quantifiable and in-depth analysis of the motor pattern of the entire hind limb,we have developed and applied a sensitive movement recording and analyzing system in order to determine the threshold of satisfactory functional reinnervation;we combined video-based footprint analysis and hind limb motion analysis to achieve a new and reliable assessment.Sprague-Dawley rats underwent a lumbar 4-5 ventral root avulsion,and their L4 ventral roots were subsequently reimplanted.The animals received different doses of riluzole treatment in order to rescue incremental numbers of the damaged motoneuron pool.We were able to assess one rear-view and six lateral parameters of the hind limb movement pattern by measuring specific joint angles,footprint,and gait parameters in single video frames.Four months after the operation,we performed Fast Blue retrograde tracing to label and count the reinnervating motoneurons.We then compared the numbers of reinnervating motoneurons and the functional improvement.Our results confirmed a strong relationship between functional restoration of the original movement pattern and morphological reinnervation;approximately 30%of the original motor pool was able to produce a useful locomotor pattern.We believe that our knowledge of the minimal motoneuron numbers required to reinnervate target muscles may help plan the segmental redistribution of the motoneuron pools for reinnervation surgeries. 展开更多

Inertial confinement fusion(ICF)requires a constant search for the most effective materials to improve the efficiency of compression of the capsule and of laser-to-target energy transfer.Foams could provide a solution,but they require further experimental and theoretical investigation.The new 3D-printing technologies,such as two-photon polymerization,are opening a new era in the production of foams,allowing fine control of material morphology.Very few detailed studies of the interaction of foams with high-power lasers in regimes relevant for ICF have been described in the literature to date,and more investigation is needed.In this work,we present the results of an experimental campaign performed at the ABC laser facility at ENEA Centro Ricerche Frascati in which 3D-printed microstructured materials were irradiated at high power.3D simulations of the laser-target interaction performed with the FLASH code reveal that the laser is scattered by plasma density gradients and channeled into the structure when the center of the focal spot is on the through hole.The time required for the laser to completely ablate the structure given by the simulations is in good agreement with the experimental measurement.Measurements of the reflected and transmitted laser light indicate that scattering occurred during the irradiation,in accordance with the simulations.Two-plasmon decay has also been found to be active during irradiation. 展开更多

To meet the challenge of mismatches between power supply and demand,modern buildings must schedule flexible energy loads in order to improve the efficiency of power grids.Furthermore,it is essential to understand the effectiveness of flexibility management strategies under different climate conditions and extreme weather events.Using both typical and extreme weather data from cities in five major climate zones of China,this study investigates the energy flexibility potential of an office building under three short-term HVAC management strategies in the context of different climates.The results show that the peak load flexibility and overall energy performance of the three short-term strategies were affected by the surrounding climate conditions.The peak load reduction rate of the pre-cooling and zone temperature reset strategies declined linearly as outdoor temperature increased.Under extreme climate conditions,the daily peak-load time was found to be over two hours earlier than under typical conditions,and the intensive solar radiation found in the extreme conditions can weaken the correlation between peak load reduction and outdoor temperature,risking the ability of a building’s HVAC system to maintain a comfortable indoor environment. 展开更多

Advanced OER/HER electrocatalytic alternatives are crucial for the wide adaptation of green hydrogen energy.Herein,Ru/NiMnB spherical cluster pillar(SCP),denoted as Ru/NiMnB,is synthesized using a combination of electro-deposition and hydrothermal reaction.Systematic investigation of Ru doping in the NiMnB matrix revealed significant improvements in electrocatalytic performance.The Ru/NiMnB SCPs demonstrate superior OER/HER activity with low overpotentials of 150 and 103 mV at 50mA/cm^(2)in 1 M KOH,making them highly competitive with state-of-the-art electrocatalysts.Remarkably,the Ru/NiMnB SCPs exhibit a low 2-E cell voltage of 2.80 V at ultra-high current density of 2,000 m A/cm^(2)in 1 M KOH,outperforming the standard benchmark electrodes of RuO_(2)||Pt/C,thereby positioning Ru/NiMnB as one of the best bifunctional electrocatalysts.These SCPs exhibit exceptional high-current characteristics,stability and corrosion resistance,as evidenced by continuous operation at 1,000 mA/cm^(2)high-current density for over 150 h in 6 M KOH at elevated temperatures under harsh industrial conditions.Only a small amount of Ru incorporation significantly enhances the electrocatalytic performances of NiMnB,attributed to increased active sites and improved intrinsic properties such as conductivity,adsorption/desorption capability and reaction rates.Consequently,Ru/NiMnB SCPs present a promising bi-functional electrode concept for efficient green H_(2)production. 展开更多

In the gas-coal integrated mining field,the conventional design method of pipeline coal pillars leads to a large amount of coal pillars being unrecovered and overlooks the pipeline's safety requirements.Considering the coal pillar recovery rate and pipeline's safety requirements,two new shaped coal pillar design approaches for subsurface pipelines were developed.Firstly,the deformation limitations for measuring pipeline safety are categorized into two:no deformation is permitted,and deformation is acceptable within elastic limits.Subsequently,integrating the key stratum theory(KST)and cave angle,a fishbone-shaped coal pillar design approach that does not permit pipeline deformation is established.Meanwhile,combined with the ground subsidence and the pipeline's elastic deformation limit,a grille-shaped coal pillar design approach that accepts deformation pipelines within elastic limits is established.Those two new approaches clarify parameters including mined width,coal pillar width and mined length.Finally,the case study shows that the designed mined width,coal pillar width and mined length of the fishbone-shaped coal pillar are 90,80,and 130 m,while those of the grille-shaped are 320,370,and640 m.Compared with the conventional method,the fishbone-shaped and grille-shaped coal pillar design approaches recovered coal pillar resources of 2.65×10~6and 5.81×10~6t on the premise of meeting the pipeline safety requirements,and the recovery rates increased by 20.5%and 45.0%,with expenditures representing only 56.46%and 20.02%of the respective benefits.These new approaches provide managers with diverse options for protecting pipeline safety while promoting coal pillar recovery,which is conducive to the harmonic mining of gas-coal resources. 展开更多

App reviews are crucial in influencing user decisions and providing essential feedback for developers to improve their products.Automating the analysis of these reviews is vital for efficient review management.While traditional machine learning(ML)models rely on basic word-based feature extraction,deep learning(DL)methods,enhanced with advanced word embeddings,have shown superior performance.This research introduces a novel aspectbased sentiment analysis(ABSA)framework to classify app reviews based on key non-functional requirements,focusing on usability factors:effectiveness,efficiency,and satisfaction.We propose a hybrid DL model,combining BERT(Bidirectional Encoder Representations from Transformers)with BiLSTM(Bidirectional Long Short-Term Memory)and CNN(Convolutional Neural Networks)layers,to enhance classification accuracy.Comparative analysis against state-of-the-art models demonstrates that our BERT-BiLSTM-CNN model achieves exceptional performance,with precision,recall,F1-score,and accuracy of 96%,87%,91%,and 94%,respectively.Thesignificant contributions of this work include a refined ABSA-based relabeling framework,the development of a highperformance classifier,and the comprehensive relabeling of the Instagram App Reviews dataset.These advancements provide valuable insights for software developers to enhance usability and drive user-centric application development. 展开更多

Objective To explore the effect of acupuncture on the endoplasmic reticulum stress IRE l-CHOP signal pathway and the expression levels of related apoptosis genes in pancreatic tissue of rats with STZ-induced diabetes mellitus. Methods Eighty SD rats were selected, among which, 10 were fed with normal diet and taken as blank control group ("blank group" for short), the other 70 rats were fed with high fat and high glucose food and injected with small dosage of STZ intraperitoneally, and 30 models were established successfully. The models were randomly divided into acupuncture treatment group (~'acupuncture group" for short), model control group ("model group" for short) and western medicine group (metformin) with 10 rats in each group. After treatment for 4 weeks, RBG, and the expression levels of CHOP, IRE 2, Bax, Bcl-2 protein and mRNA in pancreatic tissue of rats were determined. Result Compared with the conditions before treatment, RBG reduced obviously in the acupuncture group and western medicine group (P〈0.05), and there was no statistical difference in RBG in the model group and blank group (P〉0.05); the expression levels of CHOP, IRE 1 and Bax proteins in pancreatic tissue of rats in the model group were significantly higher than those in the blank group (P〈0.05), the expression results in the acupuncture group and western medicine group were significantly lower than those in model group (P〈0.05); the expression levels of CHOP, IRE 1 and Bax mRNAs in pancreatic tissue of rats in the model group were significantly higher than those in the blank group (P〈0.01), the expression results in the acupuncture group and western medicine group were significantly lower than those in the model group (P〈0.01); the expression level of Bcl-2 protein in the model group was significantly lower than that in the blank group (P〈0.05), the expression results in the acupuncture group and western medicine group were significantly higher than that in the model group (P〈0.02); there was no statistical difference between the acupuncture group and western medicine group (P〉0.05); the expression level of Bcl-2 mRNA in the model group was significantly lower than that in the blank group (P〈0.02), the expression results in the acupuncture group and western medicine group were significantly higher than that in the model group (P〈0.01); there was no statistical difference between the acupuncture group and western medicine group (P〉0.05). Conclusion There was endoplasmic reticulum stress in the pancreatic tissue of rats with STZ-induced diabetes mellitus, and IRE 1-CHOP signal pathway was the main pathway for inhibiting the endoplasmic reticulum stress in the pancreatic tissue. Acupuncture can improve the endoplasmic reticulum stress in pancreatic tissue, inhibit apoptosis genes, and protect the pancreatic tissue through IRE 1-CHOP signal pathway. 展开更多

As the trend to use the latestmachine learning models to automate requirements engineering processes continues,security requirements classification is tuning into the most researched field in the software engineering community.Previous literature studies have proposed numerousmodels for the classification of security requirements.However,adopting those models is constrained due to the lack of essential datasets permitting the repetition and generalization of studies employing more advanced machine learning algorithms.Moreover,most of the researchers focus only on the classification of requirements with security keywords.They did not consider other nonfunctional requirements(NFR)directly or indirectly related to security.This has been identified as a significant research gap in security requirements engineering.The major objective of this study is to propose a security requirements classification model that categorizes security and other relevant security requirements.We use PROMISE_exp and DOSSPRE,the two most commonly used datasets in the software engineering community.The proposed methodology consists of two steps.In the first step,we analyze all the nonfunctional requirements and their relation with security requirements.We found 10 NFRs that have a strong relationship with security requirements.In the second step,we categorize those NFRs in the security requirements category.Our proposedmethodology is a hybridmodel based on the ConvolutionalNeural Network(CNN)and Extreme Gradient Boosting(XGBoost)models.Moreover,we evaluate the model by updating the requirement type column with a binary classification column in the dataset to classify the requirements into security and non-security categories.The performance is evaluated using four metrics:recall,precision,accuracy,and F1 Score with 20 and 28 epochs number and batch size of 32 for PROMISE_exp and DOSSPRE datasets and achieved 87.3%and 85.3%accuracy,respectively.The proposed study shows an enhancement in metrics values compared to the previous literature studies.This is a proof of concept for systematizing the evaluation of security recognition in software systems during the early phases of software development. 展开更多
