Objective:Triple-negative breast cancer(TNBC)is a highly aggressive subtype that lacks targeted therapies,leading to a poorer prognosis.However,some patients achieve long-term recurrence-free survival(RFS),offering va...Objective:Triple-negative breast cancer(TNBC)is a highly aggressive subtype that lacks targeted therapies,leading to a poorer prognosis.However,some patients achieve long-term recurrence-free survival(RFS),offering valuable insights into tumor biology and potential treatment strategies.Methods:We conducted a comprehensive multi-omics analysis of 132 patients with American Joint Committee on Cancer(AJCC)stage III TNBC,comprising 36 long-term survivors(RFS≥8 years),62 moderate-term survivors(RFS:3-8 years),and 34 short-term survivors(RFS<3 years).Analyses investigated clinicopathological factors,whole-exome sequencing,germline mutations,copy number alterations(CNAs),RNA sequences,and metabolomic profiles.Results:Long-term survivors exhibited fewer metastatic regional lymph nodes,along with tumors showing reduced stromal fibrosis and lower Ki67 index.Molecularly,these tumors exhibited multiple alterations in genes related to homologous recombination repair,with higher frequencies of germline mutations and somatic CNAs.Additionally,tumors from long-term survivors demonstrated significant downregulation of the RTK-RAS signaling pathway.Metabolomic profiling revealed decreased levels of lipids and carbohydrate,particularly those involved in glycerophospholipid,fructose,and mannose metabolism,in long-term survival group.Multivariate Cox analysis identified fibrosis[hazard ratio(HR):12.70,95%confidence interval(95%CI):2.19-73.54,P=0.005]and RAC1copy number loss/deletion(HR:0.22,95%CI:0.06-0.83,P=0.026)as independent predictors of RFS.Higher fructose/mannose metabolism was associated with worse overall survival(HR:1.30,95%CI:1.01-1.68,P=0.045).Our findings emphasize the association between biological determinants and prolonged survival in patients with TNBC.Conclusions:Our study systematically identified the key molecular and metabolic features associated with prolonged survival in AJCC stage III TNBC,suggesting potential therapeutic targets to improve patient outcomes.展开更多

The big-tapered profiled ring disk is a key component of engines for rockets and missiles.A new forming technology,as called spinning-rolling process,has been proposed previously for the high performance,high efficien...The big-tapered profiled ring disk is a key component of engines for rockets and missiles.A new forming technology,as called spinning-rolling process,has been proposed previously for the high performance,high efficiency and low-cost manufacturing of the component.Blank design is the key part of plastic forming process design.For spinning-rolling process,the shape and size of the blank play a crucial role in process stability,deformation behavior and dimensional accuracy.So this work proposes a blank design method to determine the geometry structure and sizes of the blank.The mathematical model for calculating the blank size has been deduced based on volume conservation and neutral layer length invariance principle.The FE simulation and corresponding trial production of an actual big-tapered profiled ring disk show that the proposed blank design method is applicative.In order to obtain a preferred blank,the influence rules of blank size determined by different deformation degrees(rolling ratio k)on the spinning-rolling process are revealed by comprehensive FE simulations.Overall considering the process stability,circularity of the deformed ring disk and forming forces,a reasonable range of deformation degree(rolling ratio k)is recommended for the blank design of the new spinning-rolling process.展开更多
In this paper,a double-effect DNN-based Digital Back-Propagation(DBP)scheme is proposed and studied to achieve the Integrated Communication and Sensing(ICS)ability,which can not only realize nonlinear damage mitigatio...In this paper,a double-effect DNN-based Digital Back-Propagation(DBP)scheme is proposed and studied to achieve the Integrated Communication and Sensing(ICS)ability,which can not only realize nonlinear damage mitigation but also monitor the optical power and dispersion profile over multi-span links.The link status information can be extracted by the characteristics of the learned optical fiber parameters without any other measuring instruments.The efficiency and feasibility of this method have been investigated in different fiber link conditions,including various launch power,transmission distance,and the location and the amount of the abnormal losses.A good monitoring performance can be obtained while the launch optical power is 2 dBm which does not affect the normal operation of the optical communication system and the step size of DBP is 20 km which can provide a better distance resolution.This scheme successfully detects the location of single or multiple optical attenuators in long-distance multi-span fiber links,including different abnormal losses of 2 dB,4 dB,and 6 dB in 360 km and serval combinations of abnormal losses of(1 dB,5 dB),(3 dB,3 dB),(5 dB,1 dB)in 360 km and 760 km.Meanwhile,the transfer relationship of the estimated coefficient values with different step sizes is further investigated to reduce the complexity of the fiber nonlinear damage compensation.These results provide an attractive approach for precisely sensing the optical fiber link status information and making correct strategies timely to ensure optical communication system operations.展开更多
A segmented predictor-corrector method is proposed for hypersonic glide vehicles to address the issue of the slow computational speed of obtaining guidance commands using the traditional predictor-corrector guidance m...A segmented predictor-corrector method is proposed for hypersonic glide vehicles to address the issue of the slow computational speed of obtaining guidance commands using the traditional predictor-corrector guidance method.Firstly,an altitude-energy profile is designed,and the bank angle is derived analytically as the initial iteration value for the predictor-corrector method.The predictor-corrector guidance method has been improved by deriving an analytical form for predicting the range-to-go error,which greatly accelerates the iterative speed.Then,a segmented guidance algorithm is proposed.The above analytically predictor-corrector guidance method is adopted when the energy exceeds an energy threshold.When the energy is less than the threshold,the equidistant test method is used to calculate the bank angle command,which ensures guidance accuracy as well as computational efficiency.Additionally,an adaptive guidance cycle strategy is applied to reduce the computational time of the reentry guidance trajectory.Finally,the accuracy and robustness of the proposed method are verified through a series of simulations and Monte-Carlo experiments.Compared with the traditional integral method,the proposed method requires 75%less computation time on average and achieves a lower landing error.展开更多
Customer segmentation according to load-shape profiles using smart meter data is an increasingly important application to vital the planning and operation of energy systems and to enable citizens’participation in the...Customer segmentation according to load-shape profiles using smart meter data is an increasingly important application to vital the planning and operation of energy systems and to enable citizens’participation in the energy transition.This study proposes an innovative multi-step clustering procedure to segment customers based on load-shape patterns at the daily and intra-daily time horizons.Smart meter data is split between daily and hourly normalized time series to assess monthly,weekly,daily,and hourly seasonality patterns separately.The dimensionality reduction implicit in the splitting allows a direct approach to clustering raw daily energy time series data.The intraday clustering procedure sequentially identifies representative hourly day-unit profiles for each customer and the entire population.For the first time,a step function approach is applied to reduce time series dimensionality.Customer attributes embedded in surveys are employed to build external clustering validation metrics using Cramer’s V correlation factors and to identify statistically significant determinants of load-shape in energy usage.In addition,a time series features engineering approach is used to extract 16 relevant demand flexibility indicators that characterize customers and corresponding clusters along four different axes:available Energy(E),Temporal patterns(T),Consistency(C),and Variability(V).The methodology is implemented on a real-world electricity consumption dataset of 325 Small and Medium-sized Enterprise(SME)customers,identifying 4 daily and 6 hourly easy-to-interpret,well-defined clusters.The application of the methodology includes selecting key parameters via grid search and a thorough comparison of clustering distances and methods to ensure the robustness of the results.Further research can test the scalability of the methodology to larger datasets from various customer segments(households and large commercial)and locations with different weather and socioeconomic conditions.展开更多
Background There is scarce data about comparisons between geriatric assessment tools in patients with aortic stenosis(AS).We aimed to describe the geriatric profile of patients with AS undergoing transcatheter aortic ...Background There is scarce data about comparisons between geriatric assessment tools in patients with aortic stenosis(AS).We aimed to describe the geriatric profile of patients with AS undergoing transcatheter aortic valve implantation(TAVI)and to analyze the ability of different tools for predicting clinical outcomes in this context.Methods This was a single center retrospective registry including patients with AS undergoing TAVI and surviving to hospital discharge.The primary endpoint was all-cause mortality or need for urgent readmission one year after TAVI.Results A total of 377 patients were included(mean age of 80.4 years).Most patients were independent or mildly dependent,with an optimal cognitive status.The proportion of frailty ranged from 17.6%to 49.8%.A total of 20 patients(5.3%)died and 110/377 patients(29.2%)died or were readmitted during follow up.Overall,most components of the geriatric assessment showed an association with clinical outcomes.Disability for instrumental activities showed a significant association with mortality and a strong association with the rate of mortality or readmission.The association between frailty and clinical outcomes was higher for short physical performance battery(SPPB),essential frailty toolset(EFT)and the frailty index based on comprehensive geriatric assessment(IF-VIG)and lower for Fried criteria and FRAIL scale.Conclusions AS patients from this series presented a good physical performance,optimal cognitive status and a reasonably low prevalence of frailty.The best predictive ability was observed for disability for instrumental activities and frailty as measured by the EFT,SPPB and the IF-VIG.展开更多
Degenerative cervical myelopathy is a common cause of spinal cord injury,with longer symptom duration and higher myelopathy severity indicating a worse prognosis.While numerous studies have investigated serological bi...Degenerative cervical myelopathy is a common cause of spinal cord injury,with longer symptom duration and higher myelopathy severity indicating a worse prognosis.While numerous studies have investigated serological biomarkers for acute spinal cord injury,few studies have explored such biomarkers for diagnosing degenerative cervical myelopathy.This study involved 30 patients with degenerative cervical myelopathy(51.3±7.3 years old,12 women and 18 men),seven healthy controls(25.7±1.7 years old,one woman and six men),and nine patients with cervical spondylotic radiculopathy(51.9±8.6 years old,three women and six men).Analysis of blood samples from the three groups showed clear differences in transcriptomic characteristics.Enrichment analysis identified 128 differentially expressed genes that were enriched in patients with neurological disabilities.Using least absolute shrinkage and selection operator analysis,we constructed a five-gene model(TBCD,TPM2,PNKD,EIF4G2,and AP5Z1)to diagnose degenerative cervical myelopathy with an accuracy of 93.5%.One-gene models(TCAP and SDHA)identified mild and severe degenerative cervical myelopathy with accuracies of 83.3%and 76.7%,respectively.Signatures of two immune cell types(memory B cells and memory-activated CD4^(+)T cells)predicted levels of lesions in degenerative cervical myelopathy with 80%accuracy.Our results suggest that peripheral blood RNA biomarkers could be used to predict lesion severity in degenerative cervical myelopathy.展开更多

BACKGROUND Disorders of gut-brain interaction(DGBI)are common,but knowledge about their physiopathology is still poor,nor valid tools have been used to evaluate them in childhood.AIM To develop a psycho-gastroenterolo...BACKGROUND Disorders of gut-brain interaction(DGBI)are common,but knowledge about their physiopathology is still poor,nor valid tools have been used to evaluate them in childhood.AIM To develop a psycho-gastroenterological questionnaire(PGQ)to assess the psycho-gastroenterological profile and social characteristics of a pediatric population with and without DGBI.METHODS One hundred and nineteen Italian children(age 11-18)were included:28 outpatient patients with DGBI(Rome IV criteria)and 91 healthy controls.They filled the PGQ,faces pain scale revised(FPS-R),Bristol stool chart,ga-strointestinal symptoms rating scale,state-trait anxiety inventory,Toronto alexithymia scale 20,perceived self-efficacy in the management of negative emotions and expression of positive emotions(APEN-G,APEP-G),irritable bowel syndrome-quality of life questionnaire,school performances,tobacco use,early life events,degree of digital-ization.RESULTS Compared to controls,patients had more medical examinations(35%of them went to the doctor more than five times),a higher school performance(23%vs 13%,P<0.05),didn’t use tobacco(never vs 16%,P<0.05),had early life events(28%vs 1%P<0.05)and a higher percentage of pain classified as 4 in the FPS-R during the examination(14%vs 7%,P<0.05).CONCLUSION Pediatric outpatients with DGBI had a higher prevalence of early life events,a lower quality of life,more medical examinations rising health care costs,lower anxiety levels.展开更多
Phosphorus(Pi)plays a crucial role in the growth and development of plants.Membrane lipid regulation is one of the main mechanisms underlying plant adaptation to Pi deficiency.Previously,the high tolerance to low-Pi s...Phosphorus(Pi)plays a crucial role in the growth and development of plants.Membrane lipid regulation is one of the main mechanisms underlying plant adaptation to Pi deficiency.Previously,the high tolerance to low-Pi stress was justified in an elite line,MSDZ 109,which was obtained from Malus mandshurica.To better understand the mechanism underlying high adaptation to low-Pi stress,currently,lipidomic and transcriptomic analysis,as well as CRISPR/Cas9 and MmGDPD1-overexpressing methodologies were comprehensively integrated into a strategy for elucidating the high tolerance to low-Pi stress.Totally,770 differential metabolites were identified from the roots between the low-Pi and stress-free,belonging to 21 sub-classes of lipid compounds.Fatty acids(FA)constituted the predominant lipid component,accounting for approximately 50%-60%of the total lipids,and triglycerides(TAG)ranked the second,comprising around 12%of the total,consecutively followed by phosphatidylcholine(PC)and diacylglycerol(DAG)with approximately 10%and 8%of the total,respectively.The synchronous transcriptomic analysis revealed a significant up-regulation of genes related to glycerophospholipid and glycerolipid metabolism,specifically those(e.g.,MmGDPD1,MmDGDG1,MmMGDG1,MmSQDG,etc.)involved in phospholipid and galactosyl synthesis in response to low-Pi stress.GUS fusing reporter assay showed that MmGDPD1 promoter induced GUS gene expression and demonstrated initiation activity.Based on expression analysis,a dual-luciferase reporter assay,as well as yeast one-hybrid(Y1H)identification,MmPHR1 was justified to bind with the MmGDPD1 promoter and positively regulate plant tolerance to low-Pi stress.To further elucidate the role of MmGDPD1,CRISPR/Cas9 and MmGDPD1-overexpressing vectors were successfully introduced into apple(‘Royal Gala')calli.Interestingly,the MmGDPD1-KO line calli exhibited the remarkable decreases in the contents of phosphodiesterase(PDE),activity,as well as the contents of total Pi,and Pi in comparison with those of the wild type.Conversely,MmGDPD1-OE ones demonstrated the significant elevation in Pi accumulations,further justifying its potential role in Pi remobilization in apple.Therefore,MmGDPD1 substantially involves elevating low-Pi tolerance via promoting Pi release in M.mandshurica.展开更多
This study describes the use of the weighted multiplicative algebraic reconstruction technique(WMART)to obtain vertical ozone profiles from limb observations performed by the scanning imaging absorption spectrometer f...This study describes the use of the weighted multiplicative algebraic reconstruction technique(WMART)to obtain vertical ozone profiles from limb observations performed by the scanning imaging absorption spectrometer for atmospheric chartography(SCIAMACHY).This technique is based on SaskMART(the combination of the multiplicative algebraic reconstruction technique and SaskTRAN radiative transfer model),which was originally developed for optical spectrometer and infrared imaging system(OSIRIS)data.One of the objectives of this study was to obtain consistent ozone profiles from the two satellites.In this study,the WMART algorithm is combined with a radiative transfer model(SCIATRAN),as well as a set of measurement vectors comprising five Hartley pairing vectors(HPVs)and one Chappuis triplet vector(CTV),to retrieve ozone profiles in the altitude range of 10–69 km.Considering that the weighting factors in WMART have a significant effect on the retrievals,we propose a novel approach to calculate the pair/triplet weighting factors using wavelength weighting functions.The results of the application of the proposed ozone retrieval scheme are compared with the SCIAMACHY v3.5 ozone product by University of Bremen and validated against profiles derived from other passive satellite observations or measured by ozonesondes.Between 18 and 55 km,the retrieved ozone profiles typically agree with data from the SCIAMACHY ozone product within 5%for tropics and middle latitudes,whereas a negative deviation exists between 35 and 50 km for northern high latitudes,with a deviation of less than 10%above 50 km.Comparison of the retrieved profiles with microwave limb sounder(MLS)v5.0 indicates that the difference is within±5%between 18 and 55 km,and an agreement within 10%is achieved in other altitudes for tropics and middle latitudes.Comparison of the retrieved profiles with OSIRIS v7.1 indicates that the average deviation is within±5%between 20 and 59 km,and difference of approximately 10%is achieved below 20 km.Compared with ozonesondes data,a general validity of the retrievals is no more than 5%between 15 and 30 km.展开更多
BACKGROUND Psoriasis is a chronic inflammatory condition related to an increased athero-sclerotic cardiovascular disease(ASCVD)risk.AIM To investigate whether lipoprotein(a)[Lp(a)]levels are increased in patients with...BACKGROUND Psoriasis is a chronic inflammatory condition related to an increased athero-sclerotic cardiovascular disease(ASCVD)risk.AIM To investigate whether lipoprotein(a)[Lp(a)]levels are increased in patients with psoriasis.METHODS A comprehensive literature search up to January 30,2025 was conducted utilizing PubMed and Cochrane Library databases.A qualitative synthesis and a meta-analysis on Lp(a)mean differences(MD)between psoriasis cases and healthy controls(HC)was performed.The protocol of this meta-analysis has been re-gistered to PROSPERO(No.CRD420250652465).RESULTS Eighteen studies with 1650 psoriasis patients and 1621 HC were eligible for qua-litative synthesis.Pooled analysis from 16 studies(1401 psoriasis patients and 1320 HC)demonstrated that psoriasis patients had significantly higher Lp(a)levels compared with the HC group(MD:6.72 mg/dL,95%CI:4.32-9.12,P<0.00001,I2=71%).Sensitivity analyses according to the region of origin was also performed.The pooled analysis of the European sub-population showed a pronounced increase in Lp(a)levels in 189 patients with psoriasis vs 178 HC(MD:15.86 mg/dL,95%CI:5.79-25.92,P<0.002,I2=79%),while the pooled analysis on the Asian sub-population demonstrated a smaller but still significant difference in Lp(a)levels between 1177 psoriasis patients and 1127 HC(MD:4.95 mg/dL,95%CI:2.99-6.92,P<0.00001,I2=58%).CONCLUSION Our findings suggest that Lp(a)levels are significantly elevated in psoriasis patients,further adding to their ASCVD risk.展开更多
Human endogenous retroviruses(HERVs)comprise approximately 8%of the human genome,integrated into the dynamic regulatory network of cellular potency during early embryonic development.In recent studies,resurgent the tr...Human endogenous retroviruses(HERVs)comprise approximately 8%of the human genome,integrated into the dynamic regulatory network of cellular potency during early embryonic development.In recent studies,resurgent the transcriptional activity of HERVs has been frequently observed in many types of human cancers,suggesting their potential functions in the occurrence and progression of malignancy.However,a dedicated web resource for querying the relationship between the activation of HERVs and cancer development is lacking.Here,we construct a database to explore the sequence information,expression profiles,survival prognosis,and genetic interactions of HERVs in diverse cancer types.Our database currently contains RNA sequencing data of 580 HERVs across 16,246 samples,including that of 6478 tumoral and 634 normal tissues,932 cancer cell lines,as well as 151 early embryonic and 8051 human adult tissues.The primary goal is to provide an easily accessible and user-friendly database for professionals in the fields of bioinformatics,pathology,pharmacology,and related areas,enabling them to efficiently screen the activity of HERVs of interest in normal and cancerous tissues and evaluate the clinical relevance.The ERVcancer database is available at http://kyuanlab.com/ervcancer/.展开更多
Traditional manufacturing processes for lightweight curved profiles are often associated with lengthy procedures,high costs,low efficiency,and high energy consumption.In order to solve this problem,a new staggered ext...Traditional manufacturing processes for lightweight curved profiles are often associated with lengthy procedures,high costs,low efficiency,and high energy consumption.In order to solve this problem,a new staggered extrusion(SE)process was used to form the curved profile of AZ31 magnesium alloy in this paper.The study investigates the mapping relationship between the curvature,microstructure,and mechanical properties of the formed profiles by using different eccentricities of the die.Scanning electron microscopy(SEM)and electron backscatter diffraction techniques are employed to examine the effects of different eccentricity values(e)on grain morphology,recrystallization mechanisms,texture,and Schmid factors of the products.The results demonstrate that the staggered extrusion method promotes the deep refinement of grain size in the extruded products,with an average grain size of only 15%of the original billet,reaching 12.28μm.The tensile strength and elongation of the curved profiles after extrusion under the eccentricity value of 10 mm,20 mm and 30 mm are significantly higher than those of the billet,with the tensile strength is increased to 250,270,235 MPa,and the engineering strain elongation increased to 10.5%,12.1%,15.9%.This indicates that staggered extrusion enables curvature control of the profiles while improving their strength.展开更多
In-depth study of the components of polymyxins is the key to controlling the quality of this class of antibiotics.Similarities and variations of components present significant analytical challenges.A two-dimensional(2...In-depth study of the components of polymyxins is the key to controlling the quality of this class of antibiotics.Similarities and variations of components present significant analytical challenges.A two-dimensional(2D)liquid chromatography-mass spectrometry(LC-MS)method was established for screening and comprehensive profiling of compositions of the antibiotic colistimethate sodium(CMS).A high concentration of phosphate buffer mobile phase was used in the first-dimensional LC system to get the components well separated.For efficient and high-accuracy screening of CMS,a targeted method based on a self-constructed high resolution(HR)mass spectrum database of CMS components was established.The database was built based on the commercial MassHunter Personal Compound Database and Library(PCDL)software and its accuracy of the compound matching result was verified with six known components before being applied to genuine sample screening.On this basis,the unknown peaks in the CMS chromatograms were deduced and assigned.The molecular formula,group composition,and origins of a total of 99 compounds,of which the combined area percentage accounted for more than 95%of CMS components,were deduced by this 2D-LC-MS method combined with the MassHunter PCDL.This profiling method was highly efficient and could distinguish hundreds of components within 3 h,providing reliable results for quality control of this kind of complex drugs.展开更多

文摘Objective:Triple-negative breast cancer(TNBC)is a highly aggressive subtype that lacks targeted therapies,leading to a poorer prognosis.However,some patients achieve long-term recurrence-free survival(RFS),offering valuable insights into tumor biology and potential treatment strategies.Methods:We conducted a comprehensive multi-omics analysis of 132 patients with American Joint Committee on Cancer(AJCC)stage III TNBC,comprising 36 long-term survivors(RFS≥8 years),62 moderate-term survivors(RFS:3-8 years),and 34 short-term survivors(RFS<3 years).Analyses investigated clinicopathological factors,whole-exome sequencing,germline mutations,copy number alterations(CNAs),RNA sequences,and metabolomic profiles.Results:Long-term survivors exhibited fewer metastatic regional lymph nodes,along with tumors showing reduced stromal fibrosis and lower Ki67 index.Molecularly,these tumors exhibited multiple alterations in genes related to homologous recombination repair,with higher frequencies of germline mutations and somatic CNAs.Additionally,tumors from long-term survivors demonstrated significant downregulation of the RTK-RAS signaling pathway.Metabolomic profiling revealed decreased levels of lipids and carbohydrate,particularly those involved in glycerophospholipid,fructose,and mannose metabolism,in long-term survival group.Multivariate Cox analysis identified fibrosis[hazard ratio(HR):12.70,95%confidence interval(95%CI):2.19-73.54,P=0.005]and RAC1copy number loss/deletion(HR:0.22,95%CI:0.06-0.83,P=0.026)as independent predictors of RFS.Higher fructose/mannose metabolism was associated with worse overall survival(HR:1.30,95%CI:1.01-1.68,P=0.045).Our findings emphasize the association between biological determinants and prolonged survival in patients with TNBC.Conclusions:Our study systematically identified the key molecular and metabolic features associated with prolonged survival in AJCC stage III TNBC,suggesting potential therapeutic targets to improve patient outcomes.

文摘Laser twin-arc GTAW(LTA-GTAW)process has been developed by using the synergic interaction effects of laser and a coupled arc in a weld pool to achieve higher energy efficiency.In this study,bead-on-plate welding was conducted on 8-mm-thick Q235B work-pieces to investigate the variation of hybrid arc profile,the influence of hybrid arc profile on weld forming,microstructure and mech-anical properties of the joint during the LTA-GTAW process.The influence of Laser-GTAW and LTA-GTAW methods on weld surface appearance,heat input per unit length,and weld metal microstructure were also demonstrated systematically.The LTA-GTAW can make the distribution of arc energy more reasonable in welding depth and width.When defocus is 0,I_(f)is 330 A,I_(b)is 240 A,laser power is 2.4 kW,and spacing between heat sources of tungsten electrode is 10 mm,the weld shape is better.Compared with Laser-GTAW,LTA-GTAW can achieve lower heat input at the same penetration depth,and the microstructure of the weld is refined.The tensile strength of the welded joint is 121.8%of the base material,and the fracture mode of the welded joint is ductile fracture,the comprehensive mechanical properties are better.

文摘The big-tapered profiled ring disk is a key component of engines for rockets and missiles.A new forming technology,as called spinning-rolling process,has been proposed previously for the high performance,high efficiency and low-cost manufacturing of the component.Blank design is the key part of plastic forming process design.For spinning-rolling process,the shape and size of the blank play a crucial role in process stability,deformation behavior and dimensional accuracy.So this work proposes a blank design method to determine the geometry structure and sizes of the blank.The mathematical model for calculating the blank size has been deduced based on volume conservation and neutral layer length invariance principle.The FE simulation and corresponding trial production of an actual big-tapered profiled ring disk show that the proposed blank design method is applicative.In order to obtain a preferred blank,the influence rules of blank size determined by different deformation degrees(rolling ratio k)on the spinning-rolling process are revealed by comprehensive FE simulations.Overall considering the process stability,circularity of the deformed ring disk and forming forces,a reasonable range of deformation degree(rolling ratio k)is recommended for the blank design of the new spinning-rolling process.

文摘In this paper,a double-effect DNN-based Digital Back-Propagation(DBP)scheme is proposed and studied to achieve the Integrated Communication and Sensing(ICS)ability,which can not only realize nonlinear damage mitigation but also monitor the optical power and dispersion profile over multi-span links.The link status information can be extracted by the characteristics of the learned optical fiber parameters without any other measuring instruments.The efficiency and feasibility of this method have been investigated in different fiber link conditions,including various launch power,transmission distance,and the location and the amount of the abnormal losses.A good monitoring performance can be obtained while the launch optical power is 2 dBm which does not affect the normal operation of the optical communication system and the step size of DBP is 20 km which can provide a better distance resolution.This scheme successfully detects the location of single or multiple optical attenuators in long-distance multi-span fiber links,including different abnormal losses of 2 dB,4 dB,and 6 dB in 360 km and serval combinations of abnormal losses of(1 dB,5 dB),(3 dB,3 dB),(5 dB,1 dB)in 360 km and 760 km.Meanwhile,the transfer relationship of the estimated coefficient values with different step sizes is further investigated to reduce the complexity of the fiber nonlinear damage compensation.These results provide an attractive approach for precisely sensing the optical fiber link status information and making correct strategies timely to ensure optical communication system operations.

文摘A segmented predictor-corrector method is proposed for hypersonic glide vehicles to address the issue of the slow computational speed of obtaining guidance commands using the traditional predictor-corrector guidance method.Firstly,an altitude-energy profile is designed,and the bank angle is derived analytically as the initial iteration value for the predictor-corrector method.The predictor-corrector guidance method has been improved by deriving an analytical form for predicting the range-to-go error,which greatly accelerates the iterative speed.Then,a segmented guidance algorithm is proposed.The above analytically predictor-corrector guidance method is adopted when the energy exceeds an energy threshold.When the energy is less than the threshold,the equidistant test method is used to calculate the bank angle command,which ensures guidance accuracy as well as computational efficiency.Additionally,an adaptive guidance cycle strategy is applied to reduce the computational time of the reentry guidance trajectory.Finally,the accuracy and robustness of the proposed method are verified through a series of simulations and Monte-Carlo experiments.Compared with the traditional integral method,the proposed method requires 75%less computation time on average and achieves a lower landing error.

文摘Customer segmentation according to load-shape profiles using smart meter data is an increasingly important application to vital the planning and operation of energy systems and to enable citizens’participation in the energy transition.This study proposes an innovative multi-step clustering procedure to segment customers based on load-shape patterns at the daily and intra-daily time horizons.Smart meter data is split between daily and hourly normalized time series to assess monthly,weekly,daily,and hourly seasonality patterns separately.The dimensionality reduction implicit in the splitting allows a direct approach to clustering raw daily energy time series data.The intraday clustering procedure sequentially identifies representative hourly day-unit profiles for each customer and the entire population.For the first time,a step function approach is applied to reduce time series dimensionality.Customer attributes embedded in surveys are employed to build external clustering validation metrics using Cramer’s V correlation factors and to identify statistically significant determinants of load-shape in energy usage.In addition,a time series features engineering approach is used to extract 16 relevant demand flexibility indicators that characterize customers and corresponding clusters along four different axes:available Energy(E),Temporal patterns(T),Consistency(C),and Variability(V).The methodology is implemented on a real-world electricity consumption dataset of 325 Small and Medium-sized Enterprise(SME)customers,identifying 4 daily and 6 hourly easy-to-interpret,well-defined clusters.The application of the methodology includes selecting key parameters via grid search and a thorough comparison of clustering distances and methods to ensure the robustness of the results.Further research can test the scalability of the methodology to larger datasets from various customer segments(households and large commercial)and locations with different weather and socioeconomic conditions.
文摘Background There is scarce data about comparisons between geriatric assessment tools in patients with aortic stenosis(AS).We aimed to describe the geriatric profile of patients with AS undergoing transcatheter aortic valve implantation(TAVI)and to analyze the ability of different tools for predicting clinical outcomes in this context.Methods This was a single center retrospective registry including patients with AS undergoing TAVI and surviving to hospital discharge.The primary endpoint was all-cause mortality or need for urgent readmission one year after TAVI.Results A total of 377 patients were included(mean age of 80.4 years).Most patients were independent or mildly dependent,with an optimal cognitive status.The proportion of frailty ranged from 17.6%to 49.8%.A total of 20 patients(5.3%)died and 110/377 patients(29.2%)died or were readmitted during follow up.Overall,most components of the geriatric assessment showed an association with clinical outcomes.Disability for instrumental activities showed a significant association with mortality and a strong association with the rate of mortality or readmission.The association between frailty and clinical outcomes was higher for short physical performance battery(SPPB),essential frailty toolset(EFT)and the frailty index based on comprehensive geriatric assessment(IF-VIG)and lower for Fried criteria and FRAIL scale.Conclusions AS patients from this series presented a good physical performance,optimal cognitive status and a reasonably low prevalence of frailty.The best predictive ability was observed for disability for instrumental activities and frailty as measured by the EFT,SPPB and the IF-VIG.

文摘Degenerative cervical myelopathy is a common cause of spinal cord injury,with longer symptom duration and higher myelopathy severity indicating a worse prognosis.While numerous studies have investigated serological biomarkers for acute spinal cord injury,few studies have explored such biomarkers for diagnosing degenerative cervical myelopathy.This study involved 30 patients with degenerative cervical myelopathy(51.3±7.3 years old,12 women and 18 men),seven healthy controls(25.7±1.7 years old,one woman and six men),and nine patients with cervical spondylotic radiculopathy(51.9±8.6 years old,three women and six men).Analysis of blood samples from the three groups showed clear differences in transcriptomic characteristics.Enrichment analysis identified 128 differentially expressed genes that were enriched in patients with neurological disabilities.Using least absolute shrinkage and selection operator analysis,we constructed a five-gene model(TBCD,TPM2,PNKD,EIF4G2,and AP5Z1)to diagnose degenerative cervical myelopathy with an accuracy of 93.5%.One-gene models(TCAP and SDHA)identified mild and severe degenerative cervical myelopathy with accuracies of 83.3%and 76.7%,respectively.Signatures of two immune cell types(memory B cells and memory-activated CD4^(+)T cells)predicted levels of lesions in degenerative cervical myelopathy with 80%accuracy.Our results suggest that peripheral blood RNA biomarkers could be used to predict lesion severity in degenerative cervical myelopathy.
文摘BACKGROUND Dyslipidemia was strongly linked to stroke,however the relationship between dyslipidemia and its components and ischemic stroke remained unexplained.AIM To investigate the link between longitudinal changes in lipid profiles and dyslipidemia and ischemic stroke in a hypertensive population.METHODS Between 2013 and 2014,6094 hypertension individuals were included in this,and ischemic stroke cases were documented to the end of 2018.Longitudinal changes of lipid were stratified into four groups:(1)Normal was transformed into normal group;(2)Abnormal was transformed into normal group;(3)Normal was transformed into abnormal group;and(4)Abnormal was transformed into abnormal group.To examine the link between longitudinal changes in dyslipidemia along with its components and the risk of ischemic stroke,we utilized multivariate Cox proportional hazards models with hazard ratio(HR)and 95%CI.RESULTS The average age of the participants was 62.32 years±13.00 years,with 329 women making up 54.0%of the sample.Over the course of a mean follow-up of 4.8 years,143 ischemic strokes happened.When normal was transformed into normal group was used as a reference,after full adjustments,the HR for dyslipidemia and ischemic stroke among abnormal was transformed into normal group,normal was transformed into abnormal group and abnormal was transformed into abnormal Wei CC et al.Dyslipidemia changed and ischemic stroke WJCC https://www.wjgnet.com 2 February 6,2025 Volume 13 Issue 4 group were 1.089(95%CI:0.598-1.982;P=0.779),2.369(95%CI:1.424-3.941;P<0.001)and 1.448(95%CI:1.002-2.298;P=0.047)(P for trend was 0.233),respectively.CONCLUSION In individuals with hypertension,longitudinal shifts from normal to abnormal in dyslipidemia-particularly in total and low-density lipoprotein cholesterol-were significantly associated with the risk of ischemic stroke.
文摘BACKGROUND Disorders of gut-brain interaction(DGBI)are common,but knowledge about their physiopathology is still poor,nor valid tools have been used to evaluate them in childhood.AIM To develop a psycho-gastroenterological questionnaire(PGQ)to assess the psycho-gastroenterological profile and social characteristics of a pediatric population with and without DGBI.METHODS One hundred and nineteen Italian children(age 11-18)were included:28 outpatient patients with DGBI(Rome IV criteria)and 91 healthy controls.They filled the PGQ,faces pain scale revised(FPS-R),Bristol stool chart,ga-strointestinal symptoms rating scale,state-trait anxiety inventory,Toronto alexithymia scale 20,perceived self-efficacy in the management of negative emotions and expression of positive emotions(APEN-G,APEP-G),irritable bowel syndrome-quality of life questionnaire,school performances,tobacco use,early life events,degree of digital-ization.RESULTS Compared to controls,patients had more medical examinations(35%of them went to the doctor more than five times),a higher school performance(23%vs 13%,P<0.05),didn’t use tobacco(never vs 16%,P<0.05),had early life events(28%vs 1%P<0.05)and a higher percentage of pain classified as 4 in the FPS-R during the examination(14%vs 7%,P<0.05).CONCLUSION Pediatric outpatients with DGBI had a higher prevalence of early life events,a lower quality of life,more medical examinations rising health care costs,lower anxiety levels.

文摘Phosphorus(Pi)plays a crucial role in the growth and development of plants.Membrane lipid regulation is one of the main mechanisms underlying plant adaptation to Pi deficiency.Previously,the high tolerance to low-Pi stress was justified in an elite line,MSDZ 109,which was obtained from Malus mandshurica.To better understand the mechanism underlying high adaptation to low-Pi stress,currently,lipidomic and transcriptomic analysis,as well as CRISPR/Cas9 and MmGDPD1-overexpressing methodologies were comprehensively integrated into a strategy for elucidating the high tolerance to low-Pi stress.Totally,770 differential metabolites were identified from the roots between the low-Pi and stress-free,belonging to 21 sub-classes of lipid compounds.Fatty acids(FA)constituted the predominant lipid component,accounting for approximately 50%-60%of the total lipids,and triglycerides(TAG)ranked the second,comprising around 12%of the total,consecutively followed by phosphatidylcholine(PC)and diacylglycerol(DAG)with approximately 10%and 8%of the total,respectively.The synchronous transcriptomic analysis revealed a significant up-regulation of genes related to glycerophospholipid and glycerolipid metabolism,specifically those(e.g.,MmGDPD1,MmDGDG1,MmMGDG1,MmSQDG,etc.)involved in phospholipid and galactosyl synthesis in response to low-Pi stress.GUS fusing reporter assay showed that MmGDPD1 promoter induced GUS gene expression and demonstrated initiation activity.Based on expression analysis,a dual-luciferase reporter assay,as well as yeast one-hybrid(Y1H)identification,MmPHR1 was justified to bind with the MmGDPD1 promoter and positively regulate plant tolerance to low-Pi stress.To further elucidate the role of MmGDPD1,CRISPR/Cas9 and MmGDPD1-overexpressing vectors were successfully introduced into apple(‘Royal Gala')calli.Interestingly,the MmGDPD1-KO line calli exhibited the remarkable decreases in the contents of phosphodiesterase(PDE),activity,as well as the contents of total Pi,and Pi in comparison with those of the wild type.Conversely,MmGDPD1-OE ones demonstrated the significant elevation in Pi accumulations,further justifying its potential role in Pi remobilization in apple.Therefore,MmGDPD1 substantially involves elevating low-Pi tolerance via promoting Pi release in M.mandshurica.

文摘This study describes the use of the weighted multiplicative algebraic reconstruction technique(WMART)to obtain vertical ozone profiles from limb observations performed by the scanning imaging absorption spectrometer for atmospheric chartography(SCIAMACHY).This technique is based on SaskMART(the combination of the multiplicative algebraic reconstruction technique and SaskTRAN radiative transfer model),which was originally developed for optical spectrometer and infrared imaging system(OSIRIS)data.One of the objectives of this study was to obtain consistent ozone profiles from the two satellites.In this study,the WMART algorithm is combined with a radiative transfer model(SCIATRAN),as well as a set of measurement vectors comprising five Hartley pairing vectors(HPVs)and one Chappuis triplet vector(CTV),to retrieve ozone profiles in the altitude range of 10–69 km.Considering that the weighting factors in WMART have a significant effect on the retrievals,we propose a novel approach to calculate the pair/triplet weighting factors using wavelength weighting functions.The results of the application of the proposed ozone retrieval scheme are compared with the SCIAMACHY v3.5 ozone product by University of Bremen and validated against profiles derived from other passive satellite observations or measured by ozonesondes.Between 18 and 55 km,the retrieved ozone profiles typically agree with data from the SCIAMACHY ozone product within 5%for tropics and middle latitudes,whereas a negative deviation exists between 35 and 50 km for northern high latitudes,with a deviation of less than 10%above 50 km.Comparison of the retrieved profiles with microwave limb sounder(MLS)v5.0 indicates that the difference is within±5%between 18 and 55 km,and an agreement within 10%is achieved in other altitudes for tropics and middle latitudes.Comparison of the retrieved profiles with OSIRIS v7.1 indicates that the average deviation is within±5%between 20 and 59 km,and difference of approximately 10%is achieved below 20 km.Compared with ozonesondes data,a general validity of the retrievals is no more than 5%between 15 and 30 km.
文摘BACKGROUND Psoriasis is a chronic inflammatory condition related to an increased athero-sclerotic cardiovascular disease(ASCVD)risk.AIM To investigate whether lipoprotein(a)[Lp(a)]levels are increased in patients with psoriasis.METHODS A comprehensive literature search up to January 30,2025 was conducted utilizing PubMed and Cochrane Library databases.A qualitative synthesis and a meta-analysis on Lp(a)mean differences(MD)between psoriasis cases and healthy controls(HC)was performed.The protocol of this meta-analysis has been re-gistered to PROSPERO(No.CRD420250652465).RESULTS Eighteen studies with 1650 psoriasis patients and 1621 HC were eligible for qua-litative synthesis.Pooled analysis from 16 studies(1401 psoriasis patients and 1320 HC)demonstrated that psoriasis patients had significantly higher Lp(a)levels compared with the HC group(MD:6.72 mg/dL,95%CI:4.32-9.12,P<0.00001,I2=71%).Sensitivity analyses according to the region of origin was also performed.The pooled analysis of the European sub-population showed a pronounced increase in Lp(a)levels in 189 patients with psoriasis vs 178 HC(MD:15.86 mg/dL,95%CI:5.79-25.92,P<0.002,I2=79%),while the pooled analysis on the Asian sub-population demonstrated a smaller but still significant difference in Lp(a)levels between 1177 psoriasis patients and 1127 HC(MD:4.95 mg/dL,95%CI:2.99-6.92,P<0.00001,I2=58%).CONCLUSION Our findings suggest that Lp(a)levels are significantly elevated in psoriasis patients,further adding to their ASCVD risk.

文摘Human endogenous retroviruses(HERVs)comprise approximately 8%of the human genome,integrated into the dynamic regulatory network of cellular potency during early embryonic development.In recent studies,resurgent the transcriptional activity of HERVs has been frequently observed in many types of human cancers,suggesting their potential functions in the occurrence and progression of malignancy.However,a dedicated web resource for querying the relationship between the activation of HERVs and cancer development is lacking.Here,we construct a database to explore the sequence information,expression profiles,survival prognosis,and genetic interactions of HERVs in diverse cancer types.Our database currently contains RNA sequencing data of 580 HERVs across 16,246 samples,including that of 6478 tumoral and 634 normal tissues,932 cancer cell lines,as well as 151 early embryonic and 8051 human adult tissues.The primary goal is to provide an easily accessible and user-friendly database for professionals in the fields of bioinformatics,pathology,pharmacology,and related areas,enabling them to efficiently screen the activity of HERVs of interest in normal and cancerous tissues and evaluate the clinical relevance.The ERVcancer database is available at http://kyuanlab.com/ervcancer/.

文摘Traditional manufacturing processes for lightweight curved profiles are often associated with lengthy procedures,high costs,low efficiency,and high energy consumption.In order to solve this problem,a new staggered extrusion(SE)process was used to form the curved profile of AZ31 magnesium alloy in this paper.The study investigates the mapping relationship between the curvature,microstructure,and mechanical properties of the formed profiles by using different eccentricities of the die.Scanning electron microscopy(SEM)and electron backscatter diffraction techniques are employed to examine the effects of different eccentricity values(e)on grain morphology,recrystallization mechanisms,texture,and Schmid factors of the products.The results demonstrate that the staggered extrusion method promotes the deep refinement of grain size in the extruded products,with an average grain size of only 15%of the original billet,reaching 12.28μm.The tensile strength and elongation of the curved profiles after extrusion under the eccentricity value of 10 mm,20 mm and 30 mm are significantly higher than those of the billet,with the tensile strength is increased to 250,270,235 MPa,and the engineering strain elongation increased to 10.5%,12.1%,15.9%.This indicates that staggered extrusion enables curvature control of the profiles while improving their strength.

文摘In-depth study of the components of polymyxins is the key to controlling the quality of this class of antibiotics.Similarities and variations of components present significant analytical challenges.A two-dimensional(2D)liquid chromatography-mass spectrometry(LC-MS)method was established for screening and comprehensive profiling of compositions of the antibiotic colistimethate sodium(CMS).A high concentration of phosphate buffer mobile phase was used in the first-dimensional LC system to get the components well separated.For efficient and high-accuracy screening of CMS,a targeted method based on a self-constructed high resolution(HR)mass spectrum database of CMS components was established.The database was built based on the commercial MassHunter Personal Compound Database and Library(PCDL)software and its accuracy of the compound matching result was verified with six known components before being applied to genuine sample screening.On this basis,the unknown peaks in the CMS chromatograms were deduced and assigned.The molecular formula,group composition,and origins of a total of 99 compounds,of which the combined area percentage accounted for more than 95%of CMS components,were deduced by this 2D-LC-MS method combined with the MassHunter PCDL.This profiling method was highly efficient and could distinguish hundreds of components within 3 h,providing reliable results for quality control of this kind of complex drugs.
