Diseases resulting from exaggerated inflammatory responses present a significant peril to human beings,and its pathogenesis and treatment have not been fully elaborated.Traditional Chinese medicine offers the benefit ...Diseases resulting from exaggerated inflammatory responses present a significant peril to human beings,and its pathogenesis and treatment have not been fully elaborated.Traditional Chinese medicine offers the benefit of minimal adverse effects and favorable outcomes in the management of inflammatory diseases.Polygonum cuspidatum,a type of traditional Chinese medicine,refers to the dried rhizomes and roots of the plant species Polygonum cuspidatum from the Polygonaceae family.It has a slightly bitter taste and exhibits a cold nature,exhibiting therapeutic effects such as heat-clearing and detoxification,blood stasis removal and pain relief,phlegm resolution,and cough alleviation.Research has demonstrated that the bioactive constituents found in Polygonum cuspidatum can impede the inflammatory response through the modulation of pertinent pathways.The anti-inflammatory effects of it have been primarily observed in nonalcoholic fatty liver disease,arthritis,ischemic stroke,parkinson’s disease,ulcerative colitis,and diabetes mellitus.This review is conducted by combining papers from the past decade.The anti-inflammatory properties and mechanisms of Polygonum cuspidatum and its natural compounds are summarized.It provides a new theoretical basis for further research on the clinical pharmacological effects of Polygonum cuspidatum.展开更多

Methods TLC and HPLC were employed for the chemical identification and content determination,respectively. Results A qualitative TLC method and a quantitative HPLC method with piceid as the reference substance were established, respectively. With piceid as the reference substance and ethyl acetate-methanol-formic acid-water ( 19:3:0.5:1) as the mobile phase, a TLC method for the identification of Huzhang from the commonly used crude drugs of the same family was also set up. Conclusion The established TLC method can reasonably appraise the quality of the drug and easily distinguish Huzhang from the other commonly used crude drugs of the same family. Rfd _values, the vitality index, in leaves of K. humilis and P.viviparum grown at 3?980 m were higher than those at 3?200 m. The higher ratio of F v/F o and F v/F m in leaves of K. humilis and P.viviparum indicated that the rate of photosynthetic conversion of light energy increased at higher altitude. Ratios of F v/F o and F v/F m and Rfd _values in K.humilis were higher than that in P.viviparum grown at the same altitude. There were more irregular chloroplasts in leaves of both species grown at higher altitude. Many irregular chloroplasts such as swollen thylakoid, deformed chloroplast envelope, were observed in P.viviparum grown at 3?980 m, but few in K. humilis . These results were discussed in relation to the photosynthetic adaptability of alpine plants and the different adaptive competence between K.humilis and P.viviparum .展开更多
A sensitive and specific reversed-phase high performance liquid chromatography (RP-HPLC) method was established for the simultaneous quantitative determination of eight active components, two stilbenes (resveratrol...A sensitive and specific reversed-phase high performance liquid chromatography (RP-HPLC) method was established for the simultaneous quantitative determination of eight active components, two stilbenes (resveratrol, polydatin) and six flavonoids (rutin, quercilrin, quercetin, luteotin, isoorientin, kaempferol), in Polygonum multiflorum Thunb. The chromatographic separation was performed on a Kromasil C18 column (250 mm^4.6 mm, 5 ~tm), and gradient elution was carried out with water-methanol as the mobile phase at a flow rate of 1 mL/min. Stilbenes and flavonoids were respectively detected at 320 nm and 350 nm with DAD. The correlation coefficients of all the calibration curves were found to be higher than 0.9995. The average recoveries were ranged from 96.8% to 102.5% with RSD less than 4.8% for these components. RP-HPLC was validated to be a robust method for the quantitative determination of active components in Polygonum multiflorum Thunb, and it could be used in the quality control of this traditional medicine.展开更多

[Objective] The aim was to study the variation of leaf characters from different provenance sources of Polygonum multiflorum Thunb,as well as to carry out cluster analysis on P.multiflorum from different provenance so...[Objective] The aim was to study the variation of leaf characters from different provenance sources of Polygonum multiflorum Thunb,as well as to carry out cluster analysis on P.multiflorum from different provenance sources to provide basis for the classification,identification,breeding and improved variety selection of P.multiflorum.[Method] Leaf shape characters of 31 copies of germplasm resources in the major distribution region of the whole country were determined,and the genetic variation of P.multiflorum leaves from different producing areas was analyzed.[Result] The leaf characters of single plant of the same experimental provenance source of P.multiflorum were relatively stable,the variation was mainly found on the single leaf area,1/2 leaf width,leaf width and other indicators;the variation of each leaf character among different provenance sources was obvious,and the variation was mainly found on the single leaf weight,leaf area,1/2 leaf width,leaf length and other indicators.The correlation analysis of each leaf character in P.multiflorum suggested that the single leaf area and single leaf weight showed extremely significant positive correlation with leaf length,1/2 leaf width,leaf width,leaf thickness and leaf stem length,while the single leaf area and single leaf weight showed significant negative correlation with WWR(leaf width/1/2 leaf width)and LWR(leaf length/1/2 leaf length),in addition,several macroscopic leaf characters such as leaf length,1/2 leaf width,leaf width,leaf stem length showed extremely positive correlation.The main component analysis result suggested that the contribution rate of accumulation variance of the front three main components was up to 97.4%,which could better reflect the comprehensive performance of leaf characters of different provenance sources of P.multiflorum.The cluster analysis showed that the experimental 31 copies of P.multiflorum provenance sources should be divided into three classes,the first class was distributed in the Middle,Western of Guizhou,northwestern of Guangxi and western areas with higher altitude;the second class was distributed in Hunan,Hubei,Sichuan,Guangdong and the most area of Guangxi;the third class was distributed in Anhui,Jiangsu and Henan and Shandong.[Conclusion] Cluster analysis of leaf characters indicated that the kinds of provenance sources which the geographical position was closer could be got together.The study had provided a certain basis for the classification of P.multiflorum.展开更多

Eight marker flavonoids were identified and simultaneously quantified, which included myricitrin, hyperoside, galuteolin, avicularin, quercitrin, quercetin, luteolin, and kaempferol. The analysis was performed on an Inertsil ODS-4 column(4.6 mm×150 mm, 5 μm) with gradient elution. The mobile phases were 0.5% aqueous phosphoric acid and acetonitrile. The detection wavelength was 360 nm. The eight marker flavonoids were separated well with good linearity(r20.9991), precision, stability and repeatability. The recovery rate was 95.58%–102.65%. Cluster analysis was employed to analyze 28 batches of samples. The result indicated that this method provides an efficient way to perform quality control as well as a scientific rationale for the Geo-authentication of Polygonum aviculare L.展开更多

BACKGROUND Polygonum multiflorum is one of the leading causes of herb-induced liver injury in China.HLA-B*35:01 is reported to be a potential biomarker of Polygonum multiflorum-induced liver injury(PM-DILI).However,li...BACKGROUND Polygonum multiflorum is one of the leading causes of herb-induced liver injury in China.HLA-B*35:01 is reported to be a potential biomarker of Polygonum multiflorum-induced liver injury(PM-DILI).However,little is known about the relationship between single-nucleotide polymorphisms(SNPs) and PM-DILI.AIM To identify SNPs that indicate susceptibility to PM-DILI METHODS We conducted a systematic study enrolling 382 participants from four independent hospitals,including 73 PM-DILI patients,118 patients with other drug-induced liver injury(other-DILI) and 191 healthy controls.Whole-exome sequencing was performed for 8 PM-DILI patients and 8 healthy controls who were randomly selected from the above subjects.Nineteen SNPs that showed high frequencies in the 8 PM-DILI patients were selected as candidate SNPs and then screened in 65 PM-DILI patients,118 other-DILI patients and 183 healthy controls using the MassARRAY system.HLA-B high-resolution genotyping was performed for the 73 PM-DILI and 118 other-DILI patients.The Han-MHC database was selected as a population control for HLA-B analysis.P <6.25 x 103 after Bolferroni correction was considered significant.RESULTS The frequencies of rslll686806 in the HLA-A gene,rs1055348 in the HLA-B gene,and rs202047044 in the HLA-DRB1 gene were significantly higher in the PM-DILI group than in the control group [27.2% vs 11.6%,P=1.72×105,odds ratio(OR)=3.96,95% confidence interval(Cl):2.21-7.14;42.5% vs 8.6%,P=1.72×10-19 OR=13.62,95% CI:7.16-25.9;22.9% vs 8.1%,P=4.64×106,OR=4.1,95% CI:2.25-7.47].Only rs1055348 showed a significantly higher frequency in the PM-DILI group than in the other-DILI group(42.5% vs 13.6%,P=1.84×10-10,OR=10.06,95% Cl:5.06-20.0),which suggested that it is a specific risk factor for PM-DILI.rs1055348 may become a tag for HLA-B*35:01 with 100% sensitivity and 97.7% specificity in the PM-DILI group and 100% sensitivity and 98.1% specificity in the other-DILI group.Furthermore,HLA-B*35:01 was confirmed to be associated with PM-DILI with a frequency of 41.1% in the PM-DILI group compared with 11.9%(P=4.30×10-11,OR=11.11,95% CI:5.57-22.19) in the other-DILI group and 2.7%(P=6.22×10-166,OR=62.62,95% Cl:35.91-109.20) in the Han-MHC database.CONCLUSION rslll686806,rs1055348,and rs202047044 are associated with PM-DILI,of which,rs1055348 is specific to PM-DILI.As a tag for HLA-B*35:01,rs1055348 may become an alternative predictive biomarker of PM-DILI.展开更多

A comparative analysis of the antimicrobial an...The present study investigated the chemical composition of ethylacetate extracts from an endophytic actinomycete Streptomyces sp. A0916 and its host Polygonum cuspidatum. A comparative analysis of the antimicrobial and antioxidant properties of the extracts was also conducted. 32 compounds of P. cuspidatum and 23 compounds of Streptomyces sp. A0916 were isolated and identified by GC/MS. Antimicrobial activities of the extracts were evaluated using eight microbial strains(3 Gram-positive bacteria, 3 Gram-negative bacteria, and 2 fungi). The Streptomyces sp. A0916 extracts showed a wide range of antimicrobial activities and presented greater antimicrobial effectiveness than the P. cuspidatum extracts. The minimum inhibitory concentration(MIC) of Streptomyces sp. A0916 extracts against the ampicillin-resistant strain Enterococcus faecium SIIA843 was 32 μg·m L-1. Furthermore, the extracts had greater antimicrobial effect against Gram-positive bacteria than Gram-negative bacteria. Finally, the antioxidant activity of the Streptomyces sp. A0916 extracts was equal to that of the P. cuspidatum extracts. In conclusion, our results suggest that the endophytic actinomycetes of the medicinal plants are an important source of bioactive substances.展开更多
Objective:To investigate the antioxidant and the antibacterial activities of crude extract from aerial part of Polygonum maritimumL.(Polygonaceae)(P.maritimum)and to find new actives biomolecules.Methods:The whole pla...Objective:To investigate the antioxidant and the antibacterial activities of crude extract from aerial part of Polygonum maritimumL.(Polygonaceae)(P.maritimum)and to find new actives biomolecules.Methods:The whole plant was collected from the Rechgoune coast(West of Algeria),and methanolic crude extract of aerial parts ofP.maritimum(PMCE)was prepared.The extract was tested against different bacterial strain and tested for his ability to neutralize free radical(DPPH)and to scavenge the H2O2.Results:PMCE had a very high content of total phenol,which was(352.49依18.03)mg/g dry weight,expressed as gallic acid equivalent.PMCE exhibited excellent antioxidant activity,as measured using DPPH and H2O2 scavenging assays.It also showed a high antibacterial activity against gram-positive bacterial strains:Bacillus cereus,Bacillus subtilisandStaphylococcus aureus with an highest MIC of 120μg/mL.Conclusions:The antioxidant and antibacterial activity of the PMCE is probably due to phenolic compounds present in the extract.The contemporary presence of antioxidant and antibacterialactivities in the PMCE suggests that this plant may be a source of bioactive substances with multifaceted activity.展开更多

Objective:Recent investigations have demonstrated that Polygonum perfoliatum L.can protect against chemical liver injury,but the mechanism behind its efficacy is still unclear.Therefore,we studied the pharmacological ...Objective:Recent investigations have demonstrated that Polygonum perfoliatum L.can protect against chemical liver injury,but the mechanism behind its efficacy is still unclear.Therefore,we studied the pharmacological mechanism at work in P.perfoliatum protection against chemical liver injury.Methods:To evaluate the activity of P.perfoliatum against chemical liver injury,levels of alanine transaminase,lactic dehydrogenase,aspartate transaminase,superoxide dismutase,glutathione peroxidase and malondialdehyde were measured,alongside histological assessments of the liver,heart and kidney tissue.A nontargeted lipidomics strategy based on ultra-performance liquid chromatography quadrupoleorbitrap high-resolution mass spectrometry method was used to obtain the lipid profiles of mice with chemical liver injury and following treatment with P.perfoliatum;these profiles were used to understand the possible mechanisms behind P.perfoliatum's protective activity.Results:Lipidomic studies indicated that P.perfoliatum protected against chemical liver injury,and the results were consistent between histological and physiological analyses.By comparing the profiles of liver lipids in model and control mice,we found that the levels of 89 lipids were significantly changed.In animals receiving P.perfoliatum treatment,the levels of 8 lipids were significantly improved,relative to the model animals.The results showed that P.perfoliatum extract could effectively reverse the chemical liver injury and significantly improve the abnormal liver lipid metabolism of mice with chemical liver injury,especially glycerophospholipid metabolism.Conclusion:Regulation of enzyme activity related to the glycerophospholipid metabolism pathway may be involved in the mechanism of P.perfoliatum's protection against liver injury.展开更多

文摘A sensitive and specific reversed-phase high performance liquid chromatography (RP-HPLC) method was established for the simultaneous quantitative determination of eight active components, two stilbenes (resveratrol, polydatin) and six flavonoids (rutin, quercilrin, quercetin, luteotin, isoorientin, kaempferol), in Polygonum multiflorum Thunb. The chromatographic separation was performed on a Kromasil C18 column (250 mm^4.6 mm, 5 ~tm), and gradient elution was carried out with water-methanol as the mobile phase at a flow rate of 1 mL/min. Stilbenes and flavonoids were respectively detected at 320 nm and 350 nm with DAD. The correlation coefficients of all the calibration curves were found to be higher than 0.9995. The average recoveries were ranged from 96.8% to 102.5% with RSD less than 4.8% for these components. RP-HPLC was validated to be a robust method for the quantitative determination of active components in Polygonum multiflorum Thunb, and it could be used in the quality control of this traditional medicine.
文摘Two new stilbene glycosides (1 and 2), together with nine known compounds (3-11), were isolated from the water extract of Polygonum multiflorum Thunb. The structures of the new compounds were elucidated by their chemical properties and spectroscopic analyses, including extensive 2D NMR experiments. Compound 2 showed strong DNA cleavage activity, and compounds 1, 2 and 10 (2, 3, 4′, 5_tetrahydroxy_ trans _stilbene_2_O_β_ D _glucopyranoside) exhibited significant inhibition of lipid peroxidation.

文摘[Objective] The aim was to study the variation of leaf characters from different provenance sources of Polygonum multiflorum Thunb,as well as to carry out cluster analysis on P.multiflorum from different provenance sources to provide basis for the classification,identification,breeding and improved variety selection of P.multiflorum.[Method] Leaf shape characters of 31 copies of germplasm resources in the major distribution region of the whole country were determined,and the genetic variation of P.multiflorum leaves from different producing areas was analyzed.[Result] The leaf characters of single plant of the same experimental provenance source of P.multiflorum were relatively stable,the variation was mainly found on the single leaf area,1/2 leaf width,leaf width and other indicators;the variation of each leaf character among different provenance sources was obvious,and the variation was mainly found on the single leaf weight,leaf area,1/2 leaf width,leaf length and other indicators.The correlation analysis of each leaf character in P.multiflorum suggested that the single leaf area and single leaf weight showed extremely significant positive correlation with leaf length,1/2 leaf width,leaf width,leaf thickness and leaf stem length,while the single leaf area and single leaf weight showed significant negative correlation with WWR(leaf width/1/2 leaf width)and LWR(leaf length/1/2 leaf length),in addition,several macroscopic leaf characters such as leaf length,1/2 leaf width,leaf width,leaf stem length showed extremely positive correlation.The main component analysis result suggested that the contribution rate of accumulation variance of the front three main components was up to 97.4%,which could better reflect the comprehensive performance of leaf characters of different provenance sources of P.multiflorum.The cluster analysis showed that the experimental 31 copies of P.multiflorum provenance sources should be divided into three classes,the first class was distributed in the Middle,Western of Guizhou,northwestern of Guangxi and western areas with higher altitude;the second class was distributed in Hunan,Hubei,Sichuan,Guangdong and the most area of Guangxi;the third class was distributed in Anhui,Jiangsu and Henan and Shandong.[Conclusion] Cluster analysis of leaf characters indicated that the kinds of provenance sources which the geographical position was closer could be got together.The study had provided a certain basis for the classification of P.multiflorum.

文摘[Objective] The research aimed to study the response and accumulation characteristics of Cd in Polygonum pubescens Bl.[Method] Taking Hoagland nutritional solution as the culture substrate,eight kinds of Cd concentrations (0,25,50,100,200,500,800,1 000 mg/L) were set up.[Result] Under low concentrations of Cd (25 and 50 mg/L),the biomass of P.pubescens had no significant difference with CK (P0.05).Under all the test concentrations of Cd,Cd content in the roots and shoots of P.pubescens exceeded 100 mg/kg and the enrichment coefficients of Cd were over 1.With the increasing of Cd concentration,Cd content and Cd accumulation amount in P.pubescens increased significantly.When Cd concentration exceeded 50 mg/L,translocation capacity coefficient was greater than 1.[Conclusion] Polygonum pubescens has high tolerance and accumulation capacity of Cd,and it may has some potential on the phytoremediation of Cd-contaminated soil.

文摘A reliable high performance liquid chromatography method was developed for the quality evaluation of Polygonum aviculare L. Eight marker flavonoids were identified and simultaneously quantified, which included myricitrin, hyperoside, galuteolin, avicularin, quercitrin, quercetin, luteolin, and kaempferol. The analysis was performed on an Inertsil ODS-4 column(4.6 mm×150 mm, 5 μm) with gradient elution. The mobile phases were 0.5% aqueous phosphoric acid and acetonitrile. The detection wavelength was 360 nm. The eight marker flavonoids were separated well with good linearity(r20.9991), precision, stability and repeatability. The recovery rate was 95.58%–102.65%. Cluster analysis was employed to analyze 28 batches of samples. The result indicated that this method provides an efficient way to perform quality control as well as a scientific rationale for the Geo-authentication of Polygonum aviculare L.

文摘Two novel flavone glycosides, 3′_hydroxy_5,4′_dimethoxy_6,7_methylenedioxyflavone_3_O_[β_D_xylopyranosyl(1→6)]_β_D_glucopyranoside (1) and 5,4′_dimethoxy_3′_isopropenylacetyl_6,7_methylenedioxyflavone_3_O_[β_D_xylopyranoxyl(1→6)]_β_D_glucopyranoside (2), were isolated from Polygonum sphaerostachyum Meisn., and their structures were deduced by spectral and chemical methods. This is the first report on the isolation of flavone glycosides from P. sphaerostachyum.
文摘Three new anthraquinones, emodin-8-β-D-(2"-O-coumarate)glucoside 1, emodin-8-β-D-(6'-O-acetyl)glucoside 2 and physicon-8-β-D-(6'-O-acetyl)glucoside 3, were isolated from the roots of Polygonum cillinerve and their structures were established by spectroscopic methods. The biological activity indicated that compound 1 had the scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals (the IC50 = 8.5 μmol/L), and compound 1-3 showed no activities against HL-60 and BCJC-823 cells by MTT method in vitro.

文摘BACKGROUND Polygonum multiflorum is one of the leading causes of herb-induced liver injury in China.HLA-B*35:01 is reported to be a potential biomarker of Polygonum multiflorum-induced liver injury(PM-DILI).However,little is known about the relationship between single-nucleotide polymorphisms(SNPs) and PM-DILI.AIM To identify SNPs that indicate susceptibility to PM-DILI METHODS We conducted a systematic study enrolling 382 participants from four independent hospitals,including 73 PM-DILI patients,118 patients with other drug-induced liver injury(other-DILI) and 191 healthy controls.Whole-exome sequencing was performed for 8 PM-DILI patients and 8 healthy controls who were randomly selected from the above subjects.Nineteen SNPs that showed high frequencies in the 8 PM-DILI patients were selected as candidate SNPs and then screened in 65 PM-DILI patients,118 other-DILI patients and 183 healthy controls using the MassARRAY system.HLA-B high-resolution genotyping was performed for the 73 PM-DILI and 118 other-DILI patients.The Han-MHC database was selected as a population control for HLA-B analysis.P <6.25 x 103 after Bolferroni correction was considered significant.RESULTS The frequencies of rslll686806 in the HLA-A gene,rs1055348 in the HLA-B gene,and rs202047044 in the HLA-DRB1 gene were significantly higher in the PM-DILI group than in the control group [27.2% vs 11.6%,P=1.72×105,odds ratio(OR)=3.96,95% confidence interval(Cl):2.21-7.14;42.5% vs 8.6%,P=1.72×10-19 OR=13.62,95% CI:7.16-25.9;22.9% vs 8.1%,P=4.64×106,OR=4.1,95% CI:2.25-7.47].Only rs1055348 showed a significantly higher frequency in the PM-DILI group than in the other-DILI group(42.5% vs 13.6%,P=1.84×10-10,OR=10.06,95% Cl:5.06-20.0),which suggested that it is a specific risk factor for PM-DILI.rs1055348 may become a tag for HLA-B*35:01 with 100% sensitivity and 97.7% specificity in the PM-DILI group and 100% sensitivity and 98.1% specificity in the other-DILI group.Furthermore,HLA-B*35:01 was confirmed to be associated with PM-DILI with a frequency of 41.1% in the PM-DILI group compared with 11.9%(P=4.30×10-11,OR=11.11,95% CI:5.57-22.19) in the other-DILI group and 2.7%(P=6.22×10-166,OR=62.62,95% Cl:35.91-109.20) in the Han-MHC database.CONCLUSION rslll686806,rs1055348,and rs202047044 are associated with PM-DILI,of which,rs1055348 is specific to PM-DILI.As a tag for HLA-B*35:01,rs1055348 may become an alternative predictive biomarker of PM-DILI.

文摘The present study investigated the chemical composition of ethylacetate extracts from an endophytic actinomycete Streptomyces sp. A0916 and its host Polygonum cuspidatum. A comparative analysis of the antimicrobial and antioxidant properties of the extracts was also conducted. 32 compounds of P. cuspidatum and 23 compounds of Streptomyces sp. A0916 were isolated and identified by GC/MS. Antimicrobial activities of the extracts were evaluated using eight microbial strains(3 Gram-positive bacteria, 3 Gram-negative bacteria, and 2 fungi). The Streptomyces sp. A0916 extracts showed a wide range of antimicrobial activities and presented greater antimicrobial effectiveness than the P. cuspidatum extracts. The minimum inhibitory concentration(MIC) of Streptomyces sp. A0916 extracts against the ampicillin-resistant strain Enterococcus faecium SIIA843 was 32 μg·m L-1. Furthermore, the extracts had greater antimicrobial effect against Gram-positive bacteria than Gram-negative bacteria. Finally, the antioxidant activity of the Streptomyces sp. A0916 extracts was equal to that of the P. cuspidatum extracts. In conclusion, our results suggest that the endophytic actinomycetes of the medicinal plants are an important source of bioactive substances.

文摘Objective:To investigate the antioxidant and the antibacterial activities of crude extract from aerial part of Polygonum maritimumL.(Polygonaceae)(P.maritimum)and to find new actives biomolecules.Methods:The whole plant was collected from the Rechgoune coast(West of Algeria),and methanolic crude extract of aerial parts ofP.maritimum(PMCE)was prepared.The extract was tested against different bacterial strain and tested for his ability to neutralize free radical(DPPH)and to scavenge the H2O2.Results:PMCE had a very high content of total phenol,which was(352.49依18.03)mg/g dry weight,expressed as gallic acid equivalent.PMCE exhibited excellent antioxidant activity,as measured using DPPH and H2O2 scavenging assays.It also showed a high antibacterial activity against gram-positive bacterial strains:Bacillus cereus,Bacillus subtilisandStaphylococcus aureus with an highest MIC of 120μg/mL.Conclusions:The antioxidant and antibacterial activity of the PMCE is probably due to phenolic compounds present in the extract.The contemporary presence of antioxidant and antibacterialactivities in the PMCE suggests that this plant may be a source of bioactive substances with multifaceted activity.
文摘The glycosylation of hydroxylcoumarin was investigated by using suspension cultures of hairy roots of Polygonum multiflorum. Two new coumarin glucosides (3 and 4) were biosysthesized by regioselectively glycosylation of corresponding substrates (1 and 2) in the system. The structures of two products were identified as 7-hydroxy-4-methylcoumarin 5-O-β-D-glucopyranoside and 7- hydroxy-3,4-dimethylcoumarin 5-O-β-D-glucopyranoside on the ground of chemical and spectroscopic methods, respectively. 2007 Rong Min Yu. 文摘Objective:Recent investigations have demonstrated that Polygonum perfoliatum L.can protect against chemical liver injury,but the mechanism behind its efficacy is still unclear.Therefore,we studied the pharmacological mechanism at work in P.perfoliatum protection against chemical liver injury.Methods:To evaluate the activity of P.perfoliatum against chemical liver injury,levels of alanine transaminase,lactic dehydrogenase,aspartate transaminase,superoxide dismutase,glutathione peroxidase and malondialdehyde were measured,alongside histological assessments of the liver,heart and kidney tissue.A nontargeted lipidomics strategy based on ultra-performance liquid chromatography quadrupoleorbitrap high-resolution mass spectrometry method was used to obtain the lipid profiles of mice with chemical liver injury and following treatment with P.perfoliatum;these profiles were used to understand the possible mechanisms behind P.perfoliatum's protective activity.Results:Lipidomic studies indicated that P.perfoliatum protected against chemical liver injury,and the results were consistent between histological and physiological analyses.By comparing the profiles of liver lipids in model and control mice,we found that the levels of 89 lipids were significantly changed.In animals receiving P.perfoliatum treatment,the levels of 8 lipids were significantly improved,relative to the model animals.The results showed that P.perfoliatum extract could effectively reverse the chemical liver injury and significantly improve the abnormal liver lipid metabolism of mice with chemical liver injury,especially glycerophospholipid metabolism.Conclusion:Regulation of enzyme activity related to the glycerophospholipid metabolism pathway may be involved in the mechanism of P.perfoliatum's protection against liver injury.
