Identification of markers of saccharides in Polygonati rhizoma during the steaming process by MALDI-Q-TOF mass spectrometry imaging

Polygonati rhizoma is often used in Chinese medicine and as food.In this study,atmospheric pressure matrixassisted laser desorption ionization and quadruple-time-of-flight(MALDI-Q-TOF)mass spectrometry techniques were applied to P.rhizoma samples from Polygonatum cyrtonema Hua species.Positive ions were mainly detected in the mass range of m/z 200-600,while negative ions were mainly observed in the mass range of m/z 100-450.A total of 263 components were identified and the spatial distribution and changes in saccharides contents during the steaming process of P.rhizoma were investigated.Monosaccharide and disaccharide exhibit a relatively uniform distribution,while the oligosaccharides were mainly found in the bast of fresh P.rhizoma.Although the contents of monosaccharide and disaccharide were increased during steaming,that of trisaccharide,tetrasaccharide,and pentasaccharide were decreased.We used the 5 saccharide types with the greatest variation in content as variables for the principal component analysis(PCA)and cluster analysis.Both PCA and cluster analysis showed that these 5 saccharides can be used as markers in the steaming process of the P.rhizoma.Present study of mass spectrometry imaging provides novel insights into the spatiotemporal accumulation patterns of saccharides in P.rhizoma,improving our understanding of the steaming process. 展开更多

A purified polysaccharide with a galactose backbone(SPR-1,Mw 3,622 Da)was isolated from processed Polygonati Rhizoma with black beans(PRWB)and characterized its chemical properties.The backbone of SPR-1 consisted of[(4)-b-D-Galp-(1]9/4,6)-b-D-Galp-(1/4)-a-D-GalpA-(1/4)-a-D-GalpA-(1/4)-aD-Glcp-(1/4,6)-a-D-Glcp-(1/4)-a/b-D-Glcp,with a branch chain of R1:b-D-Galp-(1/3)-b-D-Galp-(1/connected to the/4,6)-b-D-Galp-(1/via O-6,and a branch chain of R2:a-D-Glcp-(1/6)-a-D-Glcp-(1/connected to the/4,6)-a-D-Glcp-(1/via O-6.Immunomodulatory assays showed that the SPR-1 significantly activated macrophages,and increased secretion of NO and cytokines(i.e.,IL-1b and TNF-a),as well as promoted the phagocytic activities of cells.Furthermore,isothermal titration calorimetry(ITC)analysis and molecular docking results indicated high-affinity binding between SPR-1 and MD2 with the equilibrium dissociation constant(KD)of 18.8 mM.It was suggested that SPR-1 activated the immune response through Toll-like receptor 4(TLR4)signaling and downstream responses.Our research demonstrated that the SPR-1 has a promising candidate from PRWB for the TLR4 agonist to induce immune response,and also provided an easily accessible way that can be used for PR deep processing。 展开更多

Conversion of the closed circular double-stranded supercoiled DNA (pBR 322) to the nicked circular form and linear form was used to investigate the effect of extracts of some Chinese medical herbs on DNA nicking induced by alternariol. Some substances in the extracts of Rhizoma polygonati (RP) and Fructus lycii (FL) were shown to protect DNA from the attack by alternariol.Some substance in the RP may bind to plasmid DNA, and this binding reduces the electrophoretic mobility of DNA. These results indicate that substances from FL and RP may be used as DNA protectors. It is possible that they play an important role in preventing cancer. 展开更多

Huang Jing (黄精 Polygonati Rhizoma, PR) was first documented as a herbal medicine in Ming Yi Bie Lu (《名医别录》 Miscellaneous Records of Famous Physicians) in China. However, there was no comprehensive review on th... Huang Jing (黄精 Polygonati Rhizoma, PR) was first documented as a herbal medicine in Ming Yi Bie Lu (《名医别录》 Miscellaneous Records of Famous Physicians) in China. However, there was no comprehensive review on the botany, traditional uses, and pharmacological effects of PR till now. In this study, the botany, traditional uses including Taoist medicine, and pharmacological effects of PR were reviewed and summarized to provide insights on drug development of PR. In Taoist medicine, PR maintains agerasia and helps prolong human life-span, and is used for fasting (Bigu). In the Zhong Guo Yao Dian (《中国药典》Chinese Pharmacopeia) version 2020, PR exerts replenishing qi and nourishing yin, invigorating the spleen, moistening the lung, and strengthening the kidney. Pharmacological studies show that PR has effects of anti-oxidation, anti-diabetes, anti-osteoporosis, anti-cancer, anti-hyperlipidemia, cardiomyocyte protection, immunomodulatory, and thus can be used for treatment of infertility, anti-microorganisms, and improving sleep and memory. In conclusion, PR may play a potential role for chronic disease management and health preservation and this very role deserves a more in-depth research. 展开更多

[Objectives]This study aimed to investigate the water extraction-alcohol precipitation technology and content determination method of polysaccharides in Rhizoma Polygonati.[Methods]Taking the content of polysaccharides in the extract as the indicator,the extraction process was optimized using single-factor tests and Box-Behnken Design.The content of polysaccharides extracted was determined by the anthrone-sulfuric acid method.Among the three factors in the test design,the solid/liquid ratio and extraction time had a significant effect(P<0.05,P<0.01),while the extraction temperature had no effect on the content of polysaccharides extracted(P>0.05).[Results]The optimal extraction process for polysaccharides in Rhizoma Polygonati was as follows:liquid/solid ratio 20∶1(mL/g),extraction time 2 h,and extraction temperature 85℃.Under the optimal extraction process,the content of polysaccharides in the extract reached 12.50%.[Conclusions]The extraction process optimized by response surface methodology for polysaccharides in Rhizoma Polygonati is reliable and has practical value,and can be used to guide production practices. 展开更多

Recently,traditional Chinese medicine-based treatment has succeeded in fighting coronavirus disease 2019(COVID-19),and Rhizoma polygonati(Huangjing)has been one of the recommended components.Its processed products play antidiabetic,antiviral,antitumor,antioxidation,antifatigue,antiaging,and immune enhancement roles.The climate in Mount Tai is mild,and the dense forest is suitable for the growth of Rhizome polygonati,which has gradually evolved into a unique specie.Considering the important medicinal value and pleasant taste of Mount Tai-Rhizoma polygonati,various healthy and functional food products,controlled by quality markers with anti-COVID-19 potential,as well as emergency foods can be developed.The study aimed to review current evidence on the nutritional value of Rhizoma polygonati from Mount Tai and its usefulness as a traditional Chinese medicine,source of herbzyme,and potential remediating agent for COVID-19 and food shortage.Most recent findings regarding herbal nanomedicine have revealed that nanoscale chemical compounds are potentially efficient in drug delivery or nanozyme catalysis upon bioprocessing.Nanoflower structure is found in processed Rhizoma polygonati by self-assembly and has wide application in enzymatic events,particularly nanoscale herbzyme.The novel findings regarding Mount Tai-Rhizoma polygonati could enhance its novel applications in chronic and hidden hunger,clinical nanomedicine,and as an anti-COVID-19 agent. 展开更多

Objective To explore the major compound in Polygonati Rhizoma(Huang Jing,黄精)for quality control.Methods The major compound was isolated and analyzed by liquid chromatography-mass spectrometry(LC-MS),and subsequently further identified by nuclear magnetic resonance(NMR).Thin layer chromatography(TLC)was optimized based on the previous methods reported in the Chinese Pharmacopeia(2015 edition).Results The major compound was isolated from the natural material and identified as linoleic acid.A high performance liquid chromatography(HPLC)method with robust linearity(R2=0.9997),specificity,precision,stability,repeatability and recovery was developed for linoleic acid determination.TLC chromatogram was improved significantly after optimization for qualitative analysis.Conclusions The optimized TLC method is practical and can be adopted for quality control of Polygonati Rhizoma(Huang Jing,黄精).The levels of linoleic acid vary between species of Polygonati Rhizoma(Huang Jing,黄精),with Polygonatum cyrtonema Hua(Jiang Xing Huang Jing,姜型黄精)showing the highest contents.This study provides valuable information for quality control of Polygonati Rhizoma(Huang Jing,黄精). 展开更多

Fingerprint Study of Polygonati Rhizoma with Steaming and Exposing to the Sun Alternatively for Different Times

[Objectives]To explore the influence of different times of steaming and exposing to the sun on the fingerprint of Polygonati Rhizoma by studying the HPLC fingerprint of Polygonati Rhizoma processed products with different times of steaming and exposing to the sun,and to provide a basis for the determination of the best processing technology of Polygonati Rhizoma.[Methods]SETSAIL II AQ-C 18(5μm×250 mm×4.6 mm)was used as the column,the column temperature was 30℃,pure water(A)and acetonitrile(B)were eluted gradually,0-10 min,B(5%-10%),10-30 min,B(10%-35%),30-40 min,B(35%-60%),40-45 min,B(60%-100%),flow rate 1 mL/min,absorption wavelength 200 nm.[Results]The relative retained peak area RSDs of the common peaks in the precision,reproducibility and stability tests were all less than 5%.There were 17 common peaks in the fingerprint of nine batches of samples,and the retention time of Peak 2 was basically the same as that of the reference peak of 5-HMF.Peak 4 mainly existed in the chromatogram of Sample 3 to Sample 5,peaks 5 and 11 mainly existed after Sample 3,peaks 9,14 and 16 mainly existed after Sample 6,and peaks 12 and 17 mainly existed after Sample 4.[Conclusions]A total of 17 common peaks were obtained,and the Peak 2 was the designated peak,and the chemical components of each processed product were different. 展开更多

Rhizome,a modified subterranean plant stem,has been used in China for centuries as a Traditional Chinese Medicine(TCM).Polygonati Rhizome(PR)is a well-known species in the Rhizome family.Previous studies show that PR might be beneficial in the treatment of osteoporosis,cancer,diabetes,and many other diseases because of its properties such as antioxidant property.Thus cultivated PR strains were developed to fulfil the increasing demands.However,the metabolite profile differences between wild-type and cultivated PR remain largely unknown.Here,we performed unbiased and untargeted quantitative mass spectrometry-based metabolomics on 1 wild strain and 3 cultivated strains.A total of 1126 metabolites were identified on all 4 strains.Analyses of these data with unsupervised and supervised approaches identified common metabolites in all 4 strains as well as strain-specific metabolites.Next,we conducted a metabolomic-wide pairwise correlation analysis on metabolite abundance and discovered 577 significantly correlated metabolite-metabolite pairs by Pearson’s correlation test.Importantly,KEGG enrichment analysis indicated that phenylpropanoids biosynthesis was the most differentially expressed pathway between cultivated and wild-type PR.By contrast,common metabolites on all 4 strains might suggest shared therapeutic targets.In summary,the systematic,untargeted metabolomics profiling of 4 PR strains and comprehensive data analyses provide invaluable resources to the understanding of the metabolite landscape and shed light on potential therapeutic applications for the treatment of diseases. 展开更多

为研制一款具有一定饱腹感及低血糖生成指数(glycemic index,GI)的黄精代餐粉,以黄精为主要原料,通过单因素试验筛选原料配比,响应面法优化代餐粉制作工艺,得到最佳配方为黄精粉添加量7.40 g、车前子壳粉添加量5.94 g、山楂粉添加量5.0... 为研制一款具有一定饱腹感及低血糖生成指数(glycemic index,GI)的黄精代餐粉,以黄精为主要原料,通过单因素试验筛选原料配比,响应面法优化代餐粉制作工艺,得到最佳配方为黄精粉添加量7.40 g、车前子壳粉添加量5.94 g、山楂粉添加量5.06 g、奇亚籽粉添加量4 g、燕麦粉添加量6 g、罗汉果甜苷添加量0.025 g。对代餐粉进行品质分析,包括冲调特性、水分含量、色度分析以及稳定餐后血糖调节作用和饱腹指数等指标。测得分散稳定性指数为2.41%,溶解度为39.0%,白度平均值为40.17。代餐粉的GI平均值为24.21,属于低GI值食物。此代餐粉的饱腹感在摄入后大约15 min时达到峰值,能在1 h内维持较好的饱腹感,随后饱腹作用逐渐降低,2 h后恢复至初始状态。 展开更多

目的:探讨玉竹多糖通过调节肠道菌群改善小鼠高脂血症的作用。方法:90只雄性C57BL/6J小鼠随机分成正常组、模型组、辛伐他汀组、玉竹多糖低、中、高剂量组(100、200、400 mg·kg^(-1)),每组15只,除正常组外均采用高脂饮食造模,辛伐... 目的:探讨玉竹多糖通过调节肠道菌群改善小鼠高脂血症的作用。方法:90只雄性C57BL/6J小鼠随机分成正常组、模型组、辛伐他汀组、玉竹多糖低、中、高剂量组(100、200、400 mg·kg^(-1)),每组15只,除正常组外均采用高脂饮食造模,辛伐他汀组和玉竹多糖组分别采用辛伐他汀和玉竹多糖灌胃12周。采用自动生化仪测定血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C),苏木素-伊红(HE)染色观察肝脏和附睾脂肪的组织形态,油红O染色检测肝脏脂质蓄积情况,并通过16S rRNA对肠道菌群进行测序分析。结果:与正常组比较,模型组小鼠体质量显著上升(P<0.01),血清TC,TG和LDL-C含量明显上升(P<0.05,P<0.01),肝脏指数和附睾脂肪系数明显上调(P<0.05),肝脏脂肪蓄积、附睾脂肪细胞体积增大,小鼠肠道菌群α多样性和β多样性降低,模型组的别样棒菌属(Allobaculum)相对丰度显著增高(P<0.01);与模型组比较,玉竹多糖能显著抑制小鼠体质量的增加(P<0.01),降低小鼠血清TC、TG和LDL-C含量(P<0.05,P<0.01)、肝脏指数和附睾脂肪系数(P<0.05),减轻肝脂肪蓄积,减小附睾脂肪细胞体积,提高小鼠肠道菌群α多样性和β多样性,降低包括别样棒菌属(Allobaculum)、丹毒丝菌属(Erysipelotrichaceae)、梭菌属(Clostridium)等厚壁菌门(Firmicutes)的丰度(P<0.01),增加艾克曼菌属(Akkermansia)和布劳特氏菌属(Blautia)等相对丰度(P<0.01)。结论:玉竹多糖可通过调节肠道菌群的多样性和物种组成改善高脂饲料诱导的小鼠高脂血症。 展开更多

目的 基于网络药理学和实验探究生、熟黄精及其有效成分治疗缺血性脑卒中(IS)大鼠的保护作用及分子机制。方法 运用网络药理学方法构建生、熟黄精治疗IS的“关键成分-核心靶点-通路”网络图。将56只大鼠随机分为假手术组、模型组、尼莫... 目的 基于网络药理学和实验探究生、熟黄精及其有效成分治疗缺血性脑卒中(IS)大鼠的保护作用及分子机制。方法 运用网络药理学方法构建生、熟黄精治疗IS的“关键成分-核心靶点-通路”网络图。将56只大鼠随机分为假手术组、模型组、尼莫地平组(20 mg/kg)、生黄精多糖低剂量组(0.1 g/kg)、生黄精多糖高剂量组(0.4 g/kg)、熟黄精多糖低剂量组(0.1 g/kg)、熟黄精多糖高剂量组(0.4 g/kg)。除手术组外,其余组采用线栓法建立大鼠IS模型。造模成功后,灌胃给药,每日1次,连续7 d。按照Zea Longa评分标准对各组大鼠给药前及给药后0.5、1、3、5、7 d的Longa神经功能进行评分;给药7 d后,取脑组织,计算脑含水量;ELISA法检测大鼠血清中白细胞介素(IL)-1β、IL-6和肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)、泛素羧基端酯酶L1(UCH-L1)、神经元特异性烯醇化酶(NSE)和神经胶质纤维酸性蛋白(GFAP)含量;Western blot检测脑组织中磷酸化Janus激酶2/Janus激酶2(p-JAK2/JAK2)、磷酸化信号转导和转录激活因子3/信号转导和转录激活因子3(p-STAT3/STAT3)和磷酸化核因子κB/核因子κB(p-NF-κB/NF-κB)、B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)和半胱氨酸蛋白酶-3(Caspase-3)的蛋白表达水平;RT-qPCR法检测Bcl-2、Bax和Caspase-3的mRNA相对表达水平。结果 网络药理学结果表明,蛋白激酶B α、细胞色素P450家族2亚家族D成员6和STAT3等为生、熟黄精治疗IS的共同潜在靶点;雌激素受体α、二肽基肽酶-4和雄激素受体等为生黄精治疗IS的特有靶点;Caspase-3、代谢型谷氨酸受体1和溶质载体家族1成员2等为熟黄精治疗IS的潜在靶点;氧化应激反应和炎症反应信号转导等为生、熟黄精治疗IS的共同信号通路。与假手术组相比,模型组大鼠脑含水量、神经功能评分均升高(P<0.01);血清IL-1β、IL-6、TNF-α、MDA、UCH-L1、NSE和GFAP水平均升高(P<0.05,P<0.01),血清CAT和SOD水平显著降低(P<0.01);脑组织中p-JAK2/JAK2、p-STAT3/STAT3和p-NF-κB/NF-κB蛋白表达水平升高(P<0.05),Bax和Caspase-3的mRNA和蛋白表达水平升高(P<0.05),Bcl-2的mRNA和蛋白表达水平降低(P<0.05)。与模型组相比,生黄精多糖低、高剂量组和熟黄精多糖低、高剂量组大鼠的神经功能评分在给药治疗5 d后降低(P<0.01);生黄精多糖组大鼠血清IL-1β、IL-6、TNF-α水平降低(P<0.05,P<0.01),熟黄精多糖组大鼠血清MDA、UCH-L1水平降低(P<0.05,P<0.01),生黄精多糖高剂量组及熟黄精多糖组血清SOD水平升高(P<0.05);熟黄精多糖高剂量组脑组织中p-JAK2/JAK2、p-STAT3/STAT3和p-NF-κB/NF-κB蛋白表达水平降低(P<0.05,P<0.01),Bax和Caspase-3 mRNA表达水平降低(P<0.05,P<0.01);生、熟黄精多糖高剂量组脑组织中Bcl-2 mRNA表达水平升高(P<0.05)。结论 生、熟黄精多糖可改善IS大鼠的神经损伤,其机制可能通过减少炎症因子表达、改善氧化应激、降低神经损害标志物表达水平,并调控JAK-STAT/NF-κB信号通路,从而减轻脑组织炎症反应。 展开更多

应用AB-8大孔树脂吸附蒸制黄精醇提物,通过乙醇溶液梯度洗脱得到Fr1~Fr4不同极性组分;比较淀粉消化酶抑制、自由基清除、胰岛素抵抗(IR)HepG2细胞葡萄糖消耗和ROS形成等指标,分析该提取物的降血糖和抗氧化等活性。研究结果表明:Fr1~Fr4... 应用AB-8大孔树脂吸附蒸制黄精醇提物,通过乙醇溶液梯度洗脱得到Fr1~Fr4不同极性组分;比较淀粉消化酶抑制、自由基清除、胰岛素抵抗(IR)HepG2细胞葡萄糖消耗和ROS形成等指标,分析该提取物的降血糖和抗氧化等活性。研究结果表明:Fr1~Fr4均能明显抑制α-淀粉酶和α-葡萄糖苷酶活性,有效淬灭DPPH和ABTS自由基,然而对羟基自由基的清除能力较弱;其中Fr2和Fr3的上述活性较为显著。Fr2和Fr3明显提高了IR-HepG2细胞葡萄糖消耗量,显著降低了细胞内ROS形成,尤其是在120μg/mL的Fr3作用下葡萄糖消耗量与正常组、Metformin阳性对照组较为接近,ROS水平恢复至正常组。研究结果表明蒸制黄精醇提物主要以中等极性组分Fr3发挥着降血糖和抗氧化作用。 展开更多

目的:建立熵权法和灰色关联度分析质量评价模型,对吉林省不同产地的玉竹质量进行综合评价,为其进一步研究与开发提供依据。方法:按照2020年版《中华人民共和国药典》玉竹检测项,测定吉林省不同产地玉竹药材的水分、浸出物及多糖含量,在... 目的:建立熵权法和灰色关联度分析质量评价模型,对吉林省不同产地的玉竹质量进行综合评价,为其进一步研究与开发提供依据。方法:按照2020年版《中华人民共和国药典》玉竹检测项,测定吉林省不同产地玉竹药材的水分、浸出物及多糖含量,在此基础上利用灰色关联分析方法,以水分、浸出物和多糖含量为指标,以相对关联度为测度,运用熵权法对各品质指标进行权重分析,构建熵权法结合灰色关联度的质量评价模型。结果:吉林省8个产地获得的玉竹药材相对关联度值的范围为0.265~0.580,质量优劣次序为:龙井>磐石>公主岭>舒兰>图们>扶余>九台>柳河。结论:灰色关联度分析通过比较各因素数据序列之间的几何相似性或差异程度,来判断各因素对玉竹品质的影响程度;熵权法基于信息熵的原理得到较为准确的权重分配,该综合评价方法可以在常规检测指标的基础上分析评价玉竹以及其他的中药材进行等级评级。 展开更多

目的 系统探究蒸制次数对黄精性状、多糖和5-羟甲基糠醛(5-hydroxymethylfurfural,5-HMF)水平的影响,关联黄精多糖抗氧化活性揭示黄精传统炮制过程的演变规律。方法 采用色差仪量化黄精样品的颜色参数,测定黄精多糖和5-HMF的含量,全面... 目的 系统探究蒸制次数对黄精性状、多糖和5-羟甲基糠醛(5-hydroxymethylfurfural,5-HMF)水平的影响,关联黄精多糖抗氧化活性揭示黄精传统炮制过程的演变规律。方法 采用色差仪量化黄精样品的颜色参数,测定黄精多糖和5-HMF的含量,全面评价黄精多糖的抗氧化活性。运用多元统计分析对数据进行降维、分类解析和相关性分析,构建综合评价体系对黄精各蒸制阶段的质量进行全面评估。结果 黄精的颜色特征随蒸制次数的不同而显著区分。“九蒸九制”过程中黄精多糖含量先降后升再趋于稳定,而5-HMF含量先升后趋于稳定。黄精“九蒸九制”过程划分为初期、中期和后期3个阶段。黄精多糖的抗氧化能力随着蒸制次数增加呈现先上升后下降的趋势,颜色与多糖、5-HMF含量及抗氧化活性之间的相关性显著(P<0.05、0.01)。综合评价体系显示,“六蒸六制”黄精在各项指标上综合得分最高。结论 颜色可作为判断黄精炮制程度的有效手段。“六蒸六制”后,黄精色泽变黑、麻舌感消失、味甜而微苦,结合抗氧化活性,推测“六蒸六制”或为黄精炮制的理想终点。 展开更多

目的:研究黄精、昆布、覆盆子等9种药食同源复方组合物水提液对小鼠抗疲劳作用的影响。方法:将健康的昆明种小鼠随机分为空白对照组(等量蒸馏水)、阳性对照组(人参蜂王浆口服液0.62 g/mL)和复方黄精组合物的水提液低、中、高(0.21、0.62... 目的:研究黄精、昆布、覆盆子等9种药食同源复方组合物水提液对小鼠抗疲劳作用的影响。方法:将健康的昆明种小鼠随机分为空白对照组(等量蒸馏水)、阳性对照组(人参蜂王浆口服液0.62 g/mL)和复方黄精组合物的水提液低、中、高(0.21、0.62、1.85 g/mL)剂量组,每组10只,雌雄各半,连续灌胃30 d,灌胃期间,记录各组小鼠体重;试验末期测定小鼠的力竭游泳时间、脏器指数,检测血清尿素氮含量、肝糖原含量、肌糖原含量。结果:与空白对照组比较,复方黄精组合物的水提液对小鼠体重的影响无统计学差异(P>0.05),且小鼠脏器方面未发现异常(P>0.05),复方黄精组合物的水提液能显著提升小鼠的耐力,延长其力竭游泳的时间(P<0.05),降低尿素氮含量(P<0.01),提高小鼠肝糖原和肌糖原含量(P<0.01)。结论:复方黄精组合物的水提液具有改善运动性疲劳的作用。 展开更多

目的评价人参-黄精不同配比对SPS诱导的小鼠PTSD样行为的改善作用,并利用生物信息学及动物实验探究其作用机制。方法人参-黄精以1∶0(RH1)、2∶1(RH2)、1∶2(RH3)、0∶1(RH4)四种配比(总共60 g)进行提取,UPLC-UV法检测不同配比人参-黄... 目的评价人参-黄精不同配比对SPS诱导的小鼠PTSD样行为的改善作用,并利用生物信息学及动物实验探究其作用机制。方法人参-黄精以1∶0(RH1)、2∶1(RH2)、1∶2(RH3)、0∶1(RH4)四种配比(总共60 g)进行提取,UPLC-UV法检测不同配比人参-黄精的中药有效成分人参皂苷Rg1、Rb1及多糖提取率;建立SPS小鼠模型,分别灌胃给药RH1、RH2、RH3及RH4(400 mg·kg^(-1))14 d后,行为学评价PTSD样行为;ELISA测定血清CORT、IL-1β及IL-10含量;生物信息学分析筛选潜在靶点;Western blot检测海马Calpain-1、PSD95和BDNF蛋白表达。结果SPS小鼠经人参-黄精不同配比提取物治疗后,能改善SPS小鼠PTSD样行为,RH2效果最佳,降低CORT及IL-1β水平,提高IL-10含量。筛选PTSD疾病潜在作用靶点160个,分别富集在抑制突触损伤、发挥抗氧化特性及抑制神经炎症等功能。RH2能阻止SPS诱导的Calpain-1、PSD95、BDNF的表达下降和GluN2B的升高。分子对接显示,人参皂苷Rg1和Rb1与Calpain-1、PSD95、BDNF分别有较强的结合力。结论人参-黄精以2∶1的配比能更有效阻止SPS诱导的小鼠PTSD样行为,其作用可能与调控Calpain-1、PSD95、BDNF和GluN2B等靶标相关。 展开更多
