Identification of markers of saccharides in Polygonati rhizoma during the steaming process by MALDI-Q-TOF mass spectrometry imaging

Polygonati rhizoma is often used in Chinese medicine and as food.In this study,atmospheric pressure matrixassisted laser desorption ionization and quadruple-time-of-flight(MALDI-Q-TOF)mass spectrometry techniques were applied to P.rhizoma samples from Polygonatum cyrtonema Hua species.Positive ions were mainly detected in the mass range of m/z 200-600,while negative ions were mainly observed in the mass range of m/z 100-450.A total of 263 components were identified and the spatial distribution and changes in saccharides contents during the steaming process of P.rhizoma were investigated.Monosaccharide and disaccharide exhibit a relatively uniform distribution,while the oligosaccharides were mainly found in the bast of fresh P.rhizoma.Although the contents of monosaccharide and disaccharide were increased during steaming,that of trisaccharide,tetrasaccharide,and pentasaccharide were decreased.We used the 5 saccharide types with the greatest variation in content as variables for the principal component analysis(PCA)and cluster analysis.Both PCA and cluster analysis showed that these 5 saccharides can be used as markers in the steaming process of the P.rhizoma.Present study of mass spectrometry imaging provides novel insights into the spatiotemporal accumulation patterns of saccharides in P.rhizoma,improving our understanding of the steaming process. 展开更多

A purified polysaccharide with a galactose backbone(SPR-1,Mw 3,622 Da)was isolated from processed Polygonati Rhizoma with black beans(PRWB)and characterized its chemical properties.The backbone of SPR-1 consisted of[(4)-b-D-Galp-(1]9/4,6)-b-D-Galp-(1/4)-a-D-GalpA-(1/4)-a-D-GalpA-(1/4)-aD-Glcp-(1/4,6)-a-D-Glcp-(1/4)-a/b-D-Glcp,with a branch chain of R1:b-D-Galp-(1/3)-b-D-Galp-(1/connected to the/4,6)-b-D-Galp-(1/via O-6,and a branch chain of R2:a-D-Glcp-(1/6)-a-D-Glcp-(1/connected to the/4,6)-a-D-Glcp-(1/via O-6.Immunomodulatory assays showed that the SPR-1 significantly activated macrophages,and increased secretion of NO and cytokines(i.e.,IL-1b and TNF-a),as well as promoted the phagocytic activities of cells.Furthermore,isothermal titration calorimetry(ITC)analysis and molecular docking results indicated high-affinity binding between SPR-1 and MD2 with the equilibrium dissociation constant(KD)of 18.8 mM.It was suggested that SPR-1 activated the immune response through Toll-like receptor 4(TLR4)signaling and downstream responses.Our research demonstrated that the SPR-1 has a promising candidate from PRWB for the TLR4 agonist to induce immune response,and also provided an easily accessible way that can be used for PR deep processing。 展开更多

Conversion of the closed circular double-stranded supercoiled DNA (pBR 322) to the nicked circular form and linear form was used to investigate the effect of extracts of some Chinese medical herbs on DNA nicking induced by alternariol. Some substances in the extracts of Rhizoma polygonati (RP) and Fructus lycii (FL) were shown to protect DNA from the attack by alternariol.Some substance in the RP may bind to plasmid DNA, and this binding reduces the electrophoretic mobility of DNA. These results indicate that substances from FL and RP may be used as DNA protectors. It is possible that they play an important role in preventing cancer. 展开更多

Huang Jing (黄精 Polygonati Rhizoma, PR) was first documented as a herbal medicine in Ming Yi Bie Lu (《名医别录》 Miscellaneous Records of Famous Physicians) in China. However, there was no comprehensive review on th... Huang Jing (黄精 Polygonati Rhizoma, PR) was first documented as a herbal medicine in Ming Yi Bie Lu (《名医别录》 Miscellaneous Records of Famous Physicians) in China. However, there was no comprehensive review on the botany, traditional uses, and pharmacological effects of PR till now. In this study, the botany, traditional uses including Taoist medicine, and pharmacological effects of PR were reviewed and summarized to provide insights on drug development of PR. In Taoist medicine, PR maintains agerasia and helps prolong human life-span, and is used for fasting (Bigu). In the Zhong Guo Yao Dian (《中国药典》Chinese Pharmacopeia) version 2020, PR exerts replenishing qi and nourishing yin, invigorating the spleen, moistening the lung, and strengthening the kidney. Pharmacological studies show that PR has effects of anti-oxidation, anti-diabetes, anti-osteoporosis, anti-cancer, anti-hyperlipidemia, cardiomyocyte protection, immunomodulatory, and thus can be used for treatment of infertility, anti-microorganisms, and improving sleep and memory. In conclusion, PR may play a potential role for chronic disease management and health preservation and this very role deserves a more in-depth research. 展开更多

[Objectives]This study aimed to investigate the water extraction-alcohol precipitation technology and content determination method of polysaccharides in Rhizoma Polygonati.[Methods]Taking the content of polysaccharides in the extract as the indicator,the extraction process was optimized using single-factor tests and Box-Behnken Design.The content of polysaccharides extracted was determined by the anthrone-sulfuric acid method.Among the three factors in the test design,the solid/liquid ratio and extraction time had a significant effect(P<0.05,P<0.01),while the extraction temperature had no effect on the content of polysaccharides extracted(P>0.05).[Results]The optimal extraction process for polysaccharides in Rhizoma Polygonati was as follows:liquid/solid ratio 20∶1(mL/g),extraction time 2 h,and extraction temperature 85℃.Under the optimal extraction process,the content of polysaccharides in the extract reached 12.50%.[Conclusions]The extraction process optimized by response surface methodology for polysaccharides in Rhizoma Polygonati is reliable and has practical value,and can be used to guide production practices. 展开更多

Recently,traditional Chinese medicine-based treatment has succeeded in fighting coronavirus disease 2019(COVID-19),and Rhizoma polygonati(Huangjing)has been one of the recommended components.Its processed products play antidiabetic,antiviral,antitumor,antioxidation,antifatigue,antiaging,and immune enhancement roles.The climate in Mount Tai is mild,and the dense forest is suitable for the growth of Rhizome polygonati,which has gradually evolved into a unique specie.Considering the important medicinal value and pleasant taste of Mount Tai-Rhizoma polygonati,various healthy and functional food products,controlled by quality markers with anti-COVID-19 potential,as well as emergency foods can be developed.The study aimed to review current evidence on the nutritional value of Rhizoma polygonati from Mount Tai and its usefulness as a traditional Chinese medicine,source of herbzyme,and potential remediating agent for COVID-19 and food shortage.Most recent findings regarding herbal nanomedicine have revealed that nanoscale chemical compounds are potentially efficient in drug delivery or nanozyme catalysis upon bioprocessing.Nanoflower structure is found in processed Rhizoma polygonati by self-assembly and has wide application in enzymatic events,particularly nanoscale herbzyme.The novel findings regarding Mount Tai-Rhizoma polygonati could enhance its novel applications in chronic and hidden hunger,clinical nanomedicine,and as an anti-COVID-19 agent. 展开更多

Objective To explore the major compound in Polygonati Rhizoma(Huang Jing,黄精)for quality control.Methods The major compound was isolated and analyzed by liquid chromatography-mass spectrometry(LC-MS),and subsequently further identified by nuclear magnetic resonance(NMR).Thin layer chromatography(TLC)was optimized based on the previous methods reported in the Chinese Pharmacopeia(2015 edition).Results The major compound was isolated from the natural material and identified as linoleic acid.A high performance liquid chromatography(HPLC)method with robust linearity(R2=0.9997),specificity,precision,stability,repeatability and recovery was developed for linoleic acid determination.TLC chromatogram was improved significantly after optimization for qualitative analysis.Conclusions The optimized TLC method is practical and can be adopted for quality control of Polygonati Rhizoma(Huang Jing,黄精).The levels of linoleic acid vary between species of Polygonati Rhizoma(Huang Jing,黄精),with Polygonatum cyrtonema Hua(Jiang Xing Huang Jing,姜型黄精)showing the highest contents.This study provides valuable information for quality control of Polygonati Rhizoma(Huang Jing,黄精). 展开更多

Fingerprint Study of Polygonati Rhizoma with Steaming and Exposing to the Sun Alternatively for Different Times

[Objectives]To explore the influence of different times of steaming and exposing to the sun on the fingerprint of Polygonati Rhizoma by studying the HPLC fingerprint of Polygonati Rhizoma processed products with different times of steaming and exposing to the sun,and to provide a basis for the determination of the best processing technology of Polygonati Rhizoma.[Methods]SETSAIL II AQ-C 18(5μm×250 mm×4.6 mm)was used as the column,the column temperature was 30℃,pure water(A)and acetonitrile(B)were eluted gradually,0-10 min,B(5%-10%),10-30 min,B(10%-35%),30-40 min,B(35%-60%),40-45 min,B(60%-100%),flow rate 1 mL/min,absorption wavelength 200 nm.[Results]The relative retained peak area RSDs of the common peaks in the precision,reproducibility and stability tests were all less than 5%.There were 17 common peaks in the fingerprint of nine batches of samples,and the retention time of Peak 2 was basically the same as that of the reference peak of 5-HMF.Peak 4 mainly existed in the chromatogram of Sample 3 to Sample 5,peaks 5 and 11 mainly existed after Sample 3,peaks 9,14 and 16 mainly existed after Sample 6,and peaks 12 and 17 mainly existed after Sample 4.[Conclusions]A total of 17 common peaks were obtained,and the Peak 2 was the designated peak,and the chemical components of each processed product were different. 展开更多

黄精是传统的食药同源物质,富含多糖、皂苷、黄酮等多种营养与功效成分,具有增强免疫功能、抗疲劳、调节血糖、调节肠道菌群等功效。随着大健康产品需求的增长,黄精提取物常应用于中成药和保健食品的配伍。讨论了黄精活性成分提取工艺,... 黄精是传统的食药同源物质,富含多糖、皂苷、黄酮等多种营养与功效成分,具有增强免疫功能、抗疲劳、调节血糖、调节肠道菌群等功效。随着大健康产品需求的增长,黄精提取物常应用于中成药和保健食品的配伍。讨论了黄精活性成分提取工艺,并从生产实际和科学研究2个方面进行了小结与展望。总结提取方法发现:目前对于黄精中主要活性成分的提取,尤其是工业化生产过程中,大多仍采用传统的回流提取法,虽成本低但损耗多、效率低。重点介绍了超声波协同酶提取等新方法在多糖、皂苷类成分提取中的优势,新方法的得率可达传统方法的1.5~3.3倍。指出黄精中特有的小极性高异黄酮现有提取方法的不足。建议根据目标物的性质组合或开发出针对性的提取技术,改良的提取工艺可提高黄精活性成分的得率。 展开更多

目的基于网络药理学的方法探讨黄精治疗肝癌的作用机制。方法本研究基于TCMSP数据库获取黄精活性成分及其靶标,同时利用Genecards数据库筛选肝癌相关靶点。运用Cytoscape软件构建“药物-成分-靶点”相互作用的网络模型,并通过STRING数... 目的基于网络药理学的方法探讨黄精治疗肝癌的作用机制。方法本研究基于TCMSP数据库获取黄精活性成分及其靶标,同时利用Genecards数据库筛选肝癌相关靶点。运用Cytoscape软件构建“药物-成分-靶点”相互作用的网络模型,并通过STRING数据库构建蛋白质互作(PPI)网络,识别核心靶点。利用Metascape平台对核心靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。最后,采用分子对接技术评估关键活性成分与核心靶蛋白的结合潜能。结果获得黄精的主要成分9个,靶点87个,疾病靶点19097个。通过PPI网络获得了10个核心基因。KEGG和GO富集分析表明黄精通过调控多条通路发挥作用。分子对接显示,黄精主要活性成分与核心靶点具有较强结合亲和力。结论本研究展示了黄精通过多靶点和多信号通路治疗肝癌。 展开更多

Rhizome,a modified subterranean plant stem,has been used in China for centuries as a Traditional Chinese Medicine(TCM).Polygonati Rhizome(PR)is a well-known species in the Rhizome family.Previous studies show that PR might be beneficial in the treatment of osteoporosis,cancer,diabetes,and many other diseases because of its properties such as antioxidant property.Thus cultivated PR strains were developed to fulfil the increasing demands.However,the metabolite profile differences between wild-type and cultivated PR remain largely unknown.Here,we performed unbiased and untargeted quantitative mass spectrometry-based metabolomics on 1 wild strain and 3 cultivated strains.A total of 1126 metabolites were identified on all 4 strains.Analyses of these data with unsupervised and supervised approaches identified common metabolites in all 4 strains as well as strain-specific metabolites.Next,we conducted a metabolomic-wide pairwise correlation analysis on metabolite abundance and discovered 577 significantly correlated metabolite-metabolite pairs by Pearson’s correlation test.Importantly,KEGG enrichment analysis indicated that phenylpropanoids biosynthesis was the most differentially expressed pathway between cultivated and wild-type PR.By contrast,common metabolites on all 4 strains might suggest shared therapeutic targets.In summary,the systematic,untargeted metabolomics profiling of 4 PR strains and comprehensive data analyses provide invaluable resources to the understanding of the metabolite landscape and shed light on potential therapeutic applications for the treatment of diseases. 展开更多

目的探究川渝特色黑豆汁炮制对滇黄精化学成分及体外抗氧化活性的影响。方法采用UPLC-Q-Exactive Orbitrap MS技术结合主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA),对生黄精、黑豆汁制黄精(制黄精)成分进行分析。通过DPPH、OH-、... 目的探究川渝特色黑豆汁炮制对滇黄精化学成分及体外抗氧化活性的影响。方法采用UPLC-Q-Exactive Orbitrap MS技术结合主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA),对生黄精、黑豆汁制黄精(制黄精)成分进行分析。通过DPPH、OH-、ABTS自由基清除实验开展体外抗氧化活性研究,结合灰色关联度法解析生、制黄精主要差异化学成分与体外抗氧化活性的谱效关系。结果鉴定出生、制黄精共95种化学成分,显著差异成分46种,包括氨基酸及衍生物9种、核苷及衍生物4种、有机酸类12种、脂肪酸类1种、黄酮类6种、生物碱类2种、糖类1种、其它成分11种。自由基清除实验表明,炮制后黄精体外抗氧化能力呈质量浓度依赖性趋势增强。相关性分析表明,5-羟甲基糠醛、6,7-二羟基-4-甲基香豆素、黄精碱A、大豆苷元等12种化学成分与体外抗氧化活性密切相关。结论川渝特色黑豆汁炮制明显影响黄精的化学成分及抗氧化活性,其中小分子糖类、苷元类、生物碱类与有机酸类成分升高,部分氨基酸类与苷类成分降低,抗氧化活性增强。该研究为后续探究黄精黑豆汁炮制前后药效变化与饮片质量评价奠定基础。 展开更多

该研究旨在制作一款黄精乌鸡汤,并结合感官评价、网络药理学和动物实验探究其免疫调节作用机制。首先利用数据库筛选黄精乌鸡汤与免疫力交集靶点并进行富集分析,其次制备黄精乌鸡汤并进行感官评价,最后构建环磷酰胺(cyclophosphamide,C... 该研究旨在制作一款黄精乌鸡汤,并结合感官评价、网络药理学和动物实验探究其免疫调节作用机制。首先利用数据库筛选黄精乌鸡汤与免疫力交集靶点并进行富集分析,其次制备黄精乌鸡汤并进行感官评价,最后构建环磷酰胺(cyclophosphamide,CTX)诱导的免疫抑制小鼠模型,并灌胃不同剂量的黄精乌鸡汤冻干粉水溶液,评估其对小鼠免疫功能的影响。结果显示,共筛选出206个黄精乌鸡汤提高免疫力的潜在作用靶点,富集于趋化因子、T细胞受体、磷脂酰肌醇3激酶等信号通路。感官评价结果表明,制作所得的黄精乌鸡汤汤体澄清透亮,呈浅琥珀色,香气浓郁,汤鲜味浓且回甘。动物实验结果显示,与正常(Control)组相比,模型(CTX)组小鼠相对体质量极显著下降,同时脾脏指数、胸腺指数及血清免疫球蛋白A(immunoglobulin A,IgA)、免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)水平和血常规指标[白细胞(white blood cell,WBC)、红细胞(red blood cell,RBC)、血红蛋白(hemoglobin,HGB)和淋巴细胞(lymphocyte,LYMPH)]水平均显著降低(P<0.05,P<0.01)。而黄精乌鸡汤各组均能不同程度缓解CTX诱导的相对体质量下降,并显著提升上述免疫器官指数、免疫球蛋白水平及血常规指标。 展开更多

目的探究黄精皂苷对卵巢癌细胞恶性生物学行为作用机制。方法以卵巢癌细胞RMG-1为模型,分为对照组、SRP低剂量组(50μM)、SRP高剂量组(100μM)、SRP组、Fer-1组、SRP+Fer-1组、过表达(overexpression,ov)-TERT组以及SRP+ov-TERT组。通过... 目的探究黄精皂苷对卵巢癌细胞恶性生物学行为作用机制。方法以卵巢癌细胞RMG-1为模型,分为对照组、SRP低剂量组(50μM)、SRP高剂量组(100μM)、SRP组、Fer-1组、SRP+Fer-1组、过表达(overexpression,ov)-TERT组以及SRP+ov-TERT组。通过CCK-8、划痕实验、Transwell、流式细胞术、商业试剂盒及免疫印迹法检测细胞活力、迁移、侵袭、凋亡、铁死亡相关指标及端粒酶逆转录酶(telomerase reverse transcriptase,TERT)、溶质载体家族7成员11(solute carrier family 7 member 11,11SLC7A11)和谷胱甘肽过氧化物酶(glutathione peroxidase,GPX)4蛋白表达。结果与对照组相比,SRP低、高剂量组的细胞迁移率和侵袭率更低(P<0.05),并降低细胞活力、提高细胞凋亡率(P<0.05)。SRP组显著提高细胞内的铁含量与活性氧水平(P<0.001),同时降低SLC7A11与GPX4蛋白表达(P<0.01)。SRP呈剂量依赖性下调TERT蛋白表达水平及端粒酶活性(P<0.001)。ov-TERT可逆转SRP对端粒酶活性、细胞活力、迁移与侵袭率(P<0.05),且拮抗SRP引起的铁蓄积、活性氧水平升高以及SLC7A11与GPX4表达下调(P<0.05)。结论SRP可通过调控TERT表达抑制卵巢癌细胞的迁移、侵袭以及细胞活力,并促进细胞凋亡,延缓卵巢癌进一步发展。 展开更多
