BACKGROUND Lotus plumule and its active components have demonstrated inhibitory effects on gastric cancer(GC).However,the molecular mechanism of lotus plumule against GC remains unclear and requires further investigat...BACKGROUND Lotus plumule and its active components have demonstrated inhibitory effects on gastric cancer(GC).However,the molecular mechanism of lotus plumule against GC remains unclear and requires further investigation.AIM To identify the key hub genes associated with the anti-GC effects of lotus plumule.METHODS This study investigated the potential targets of traditional Chinese medicine for inhibiting GC using weighted gene co-expression network analysis and bio-informatics.Initially,the active components and targets of the lotus plumule and the differentially expressed genes associated with GC were identified.Sub-sequently,a protein-protein interaction network was constructed to elucidate the interactions between drug targets and disease-related genes,facilitating the identification of hub genes within the network.The clinical significance of these hub genes was evaluated,and their upstream transcription factors and down-stream targets were identified.The binding ability of a hub gene with its down-stream targets was verified using molecular docking technology.Finally,molecular docking was performed to evaluate the binding affinity between the active ingredients of lotus plumule and the hub gene.RESULTS This study identified 26 genes closely associated with GC.Machine learning analysis and external validation narrowed the list to four genes:Aldo-keto reductase family 1 member B10,fructose-bisphosphatase 1,protein arginine methyltransferase 1,and carbonic anhydrase 9.These genes indicated a strong correlation with anti-GC activity.CONCLUSION Lotus plumule exhibits anti-GC effects.This study identified four hub genes with potential as novel targets for diagnosing and treating GC,providing innovative perspectives for its clinical management.展开更多

The percentages of normal plumules after 4~C treatments for 7 d, 9 d, 11 d, and 14 d, as well as the cold stress tolerance index (CSTI) and the withering index (WI) of rice seedling were investigated. A total of five single-effect QTLs, each for percentages of normal plumules after 4℃treatments for 9 d, 11 d and 14 d, and CSTI and WI, respectively were identified. The QTLs for the percentages of normal plumules after low temperature treatments for g d, 11 d and 14 d were on chromosomes 4, 2 and 11, accounting for 14.1%, 17.3% and 21.5% of the phenotypic variation, respectively. QTLs for CSTI and WI were on chromosomes 10 and 1, respectively. Two pairs of epistatic loci were identified, but none of the epistatic loci involved the single-effect QTLs. The RM528-RM340 interval on chromosome 6 interacted with the RM278-RM3919B interval on chromosome 9 for CSTI, and the epistatic interaction accounted for 17.7% of the phenotypic variation. A pair of epistatic loci was identified for WI, the RM246-RM5461 interval on chromosome 1 interacted with the ISA-RM447 interval on chromosome 8, which accounted for 22.6% of the phenotypic variation.展开更多
Seed plumules comprise multiple developing tissues and are key sites for above-ground plant organ morphogenesis.Here,the spatial expression of genes in developing rice seed plumules was characterized by single-cell tr...Seed plumules comprise multiple developing tissues and are key sites for above-ground plant organ morphogenesis.Here,the spatial expression of genes in developing rice seed plumules was characterized by single-cell transcriptome sequencing in Zhongjiazao 17,a popular Chinese indica rice cultivar.Of 15 cell clusters,13 were assigned to cell types using marker genes and cluster-specific genes.Marker genes of multiple cell types were expressed in several clusters,suggesting a complex developmental system.Some genes for signaling by phytohormones such as abscisic acid were highly expressed in specific clusters.Various cis-elements in the promoters of genes specifically expressed in cell clusters were calculated,and some key hormone-related motifs were frequent in certain clusters.Spatial expression patterns of genes involved in rapid seed germination,seedling growth,and development were identified.These findings enhanced our understanding of cellular diversity and specialization within plumules of rice,a monocotyledonous model crop.展开更多
Lotus(Nelumbo nucifera Gaertn),a kind of perennial aquatic plant,is widely cultivated and consumed by people in Asian countries.Lotus plumule flavonoids(LPF)have been recognized as a hypoglycemic agent.LPF was optimal...Lotus(Nelumbo nucifera Gaertn),a kind of perennial aquatic plant,is widely cultivated and consumed by people in Asian countries.Lotus plumule flavonoids(LPF)have been recognized as a hypoglycemic agent.LPF was optimally obtained using novel ultrasonic-microwave assisted synergistic extraction(UMSE)method by response surface methodology(RSM)on the basis of the results of single-factor experiments.Furthermore,the hypoglycemic activity of LPF was investigated by measuring the body weight,fasting blood glucose(FBG)level,and oral glucose tolerance test(OGTT)and analyzing the physiological indexes in streptozotocin-diabetic mice model.The optimum extraction conditions consisted of microwave power 355 W,ultrasonic power 423 W,extraction time 15 min,solid-liquid ratio 1:40,ultrasound/interval time 1/0,and ethanol concentration 70%with the maximum LPF yield of 2.62%.LPF supplementation significantly decreased the body weight,FBG,OGTT,serum total cholesterol(TC),serum total triglycerides(TG),and insulin levels,indicating the antidiabetic activity of LPF.This research verified that the UMSE technique was highly efficient to extract LPF to the maximum extent and the flavonoids from L.plumule exhibited hypoglycemic activity,which showed broad development and application prospects.展开更多

SDS PAGE analysis of c...The changes of chlorophyll_protein complexes and photosynthetic activities of chloroplast isolated from lotus ( Nelumbo nucifera Gaertn.) seeds germinating under illumination were studied. SDS PAGE analysis of chlorophyll_protein complexes showed that there was only the light harvesting chlorophyll a/b protein complex from PSⅡ (LHCⅡ) precursor in chloroplast from lotus seeds germinated for 2 to 6 days, while LHC Ⅱ 1, and the chlorophyll_protein complex of PSⅠ (CPⅠ) appeared on the 8th day of germination and PSⅡ reaction center complex appeared later. Studies on the polypeptides composition of the chloroplast revealed the following results: 1) Small amount of the 27 kD polypeptide was synthesized in invisible light; 2) The 30 kD polypeptide existed previously in the plumules of the dry seeds; 3) The amount of the 30 kD polypeptide was more than any other polypeptides before germination and decreased gradually throughout germination, while the 27 kD polypeptide changed in the opposite way; 4) In the process of germination, measurement of the electron transport rate and the fluorescence induction kinetics at room temperature showed that PSⅡ activities and efficiency of primary light energy transformation were only experimentally measurable after 7 days of germination and gradually increased afterwards. At the same time, the chl a/b ratio rose from the lower value to normal; 5) The changes of chloroplast membrane components and its functions are concomitant in concert with that of the ultrastructure of chloroplast membranes during germination, as shown in our earlier work . The results have proved again that a different developmental pathway of chloroplast is likely to exist in the lotus plumules, which might provide an important clue for N. nucifera in having an unique position in the phylogeny of the angiosperm.展开更多

[Method] The ...[Objective] This study aimed to investigate the effect of growth regulators on the camellia (Camellia oleifera) bud stock cutting and to improve its survival rate by culturing thick and strong plumule. [Method] The effects of the different growth regulators 911 rooting agent, GA3 , ABT rooting powder on the germination ratio, the average germinating speed, the diameter of plumule and the survival rate of grafting shoots of camellia were compared and analyzed. [Result] The three growth regulators could significantly improve the growth potential of the plumules. The germination rate, plumule stem and survival rates of grafted plumules treated by the three growth regulators were much better than that of control. The germinating of camellia seeds treated with 911 rooting agent, GA3 or ABT rooting powder was 5, 7 or 9 d earlier than control; the diameter of embryo was 45%, 40% or 50% larger and the survival rate was 8%, 12% or 9% higher than that of control respectively. [Conclusion] The three hormones promoted the germination, the development of the roots including the lateral root and the fibrous, and improved the survival rate of cuttings. Different concentrations of leaf (1%, 3% and 5%) and bark (1%, 3% and 5%) aqueous extract of A. latifolia were used to study radicle and plumule growth of Vigna unguiculata. The leaf and bark aqueous extract have shown minimum allelopathic effect on V. unguiculata and suppressed/stimulated radicle and plumule growth under different concentrations. The wide distribution of Anogeissus latifolia in a subtropical region of Garhwal Himalaya, provides good fodder fuel wood and timber to the villagers. Therefore introducing Anogeissus latifolia in agricultural field and maintaining the existed trees at boundaries of agricultural fields can be useful to the villagers requirements for the villages and wherever the possible allopathic effect exit can be minimized by lopping the branches of trees and use of naturally flecked off bark for fuel wood purposes.展开更多
