The preparation of 1-(2'-deoxy-?-D-ribofuranosyl) -1,2,4-triazole-3-carboxamide was also performed through a four-step procedure, protection of 3', 5'-dihydroxyl group of Ribavirin with 1,3-dichloro-1,1,3,3-tetraisopropyldisiloxane (TPDS-Cl), phenoxythio-carbonylation of the 2'-hydroxyl group of 3', 5'-O-TPDS-Ribavirin with phenoxythiocarbonyl-chloride (PTC-Cl), reduction of 2′-O-phenoxythiocarbonyl ester of 3', 5'-O-TPDS-Ribavirin with tri-n-butyltin hydride and AIBN, deprotection of 3', 5'-O-TPDS-Ribavirin with tetrabutylammon-ium fluoride in THF.展开更多
An electroanalytical method for the determination of ammonium ion using a platinized platinum electrode is described.Under optimized analytical conditions,the linear range of the calibration graphs for ammonium ion is...An electroanalytical method for the determination of ammonium ion using a platinized platinum electrode is described.Under optimized analytical conditions,the linear range of the calibration graphs for ammonium ion is 3.0×10^(-5)-5.0×10^(-3)mol·L^(-1)with a detection limit of 5.0×10^(-6)mol·L^(-1).The method has been applied to the determination of ammonium ion in lake water samples and recoveries of 100%–103%are obtained.The results obtained are found to be in good agreement with spectrophotometric results.展开更多
Proteolysis targeting chimeras(PROTACs),which catalytically degrade disease-related proteins,can overcome the limitations of traditional small-molecule inhibitors and thus have revolutionized the field of targeted the...Proteolysis targeting chimeras(PROTACs),which catalytically degrade disease-related proteins,can overcome the limitations of traditional small-molecule inhibitors and thus have revolutionized the field of targeted therapy.Building on this advancement,we present platinated PROTAC[PROTAC-Pt(Ⅳ)],a new class of“dual-action”prodrug that leverages the ubiquitin-proteasome system-mediated degradation capabilities of PROTAC and exhibits the advantages of Pt-based anticancer prodrugs.PROTAC-Pt(Ⅳ)exhibits exceptional cytotoxicity,with half-maximal inhibitory concentration values in the nanomolar range.It outperformed conventional inhibitor-based Pt(Ⅳ)prodrugs by up to three orders of magnitude by efficiently degrading the target protein BRD4 in a range of human cancer cells.PROTAC-Pt(Ⅳ)induces cancer cell death through mechanisms including augmented apoptosis,p21-mediated cell cycle arrest,and immune activation via PD-L1 downregulation.Compared with PROTAC alone,PROTAC-Pt(Ⅳ)more effectively suppressed the growth of tumor xenografts in a mouse model via its altered pharmacokinetic properties.Collectively,the development of PROTAC-Pt(Ⅳ)marks a revolution in dual-action Pt(Ⅳ)anticancer prodrugs and offers a promising avenue for enhanced and targeted cancer therapies.展开更多
