Firstly, more than 50 efficient crude oil-degrading and biosurfactant producing bacteria were isolated from petroleum-contaminated soil and water in Tianjin Binhai New Area Oil field, China. Twenty-four of them were selected for further study. These strains were identified as belonging Pseudornonas aeruginosa, Bacillus subtilis, Brevibacillus brevis, Achrornobacter sp., Acinetobacter venetianus, Lysinibacillus rnacroides, Klebsiella oxytoca, Stenotrophornonas rhizophila, Rhodococcus sp. and Bacillus thuringiensis. A shake-flask degradation test revealed that 12 of these strains could degrade over 50% of 1% crude oil concentration in 7 d. Of these, 8 strains were able to produce biosurfactants. Furthermore, environmental tolerance experiments indicated that the majority of the strains had the ability to adapt to extreme environments including high temperatures, alkaline en- vironments and high salinity environments. A mixed bacterial agent comprising the strains WB2, W2, W3 and HA was developed based on the environmental tolerance tests and subjected to the pilot-scale degradation test indicating that this bacterial agent could degrade 85.2% of 0.8% crude oil concentration in 60 d. Our results suggest that the application of this mixed agent could remediate crude oil polluted soils in saline and alkaline environments.展开更多

Experimental results showed that when the concentrations of COD and NH4+-N of the influent were 370~910 mg/L and 10~70 mg/L, the corresponding average concentrations of those of effluent were about 80 mg/L and 8 mg/L respectively, which was better than the Level I criteria of the Integrated Wastewater Discharge Standard (GB8978-1996). According to GC-MS analysis of the effluents from both the wastewater treatment plant (WWTP) and the pilot system, there were 68 kinds of persistent organic pollutants in the WWTP effluent, while there were only 32 in that of the pilot system. In addition, the amount of the organics in the effluent of the pilot system reduced by almost 50% compared to that of the WWTP. Moreover, since such wastewater is also not suitable for the growth of microorganisms, the way of wastewater treatment by microorganisms is also greatly limited. For this kind of industrial waste water, apart from the degradation of organic matters, the separation of inorganic salts and waste water is also required to meet the sewage discharge standard. Based on this, the article focuses on the coal chemical high salt wastewater electro-catalytic oxidation pilot study.展开更多

Recombinant trypsin is a non-animal derived protease used as an alternative to animal-derived trypsin. To inactivate recombinant trypsin, a soybean trypsin inhibitor (STI) should be added to the medium. A protocol was first tested in T-flasks and then passaged to 500 mL and 3 L spinner flasks. Cell detachment was completed in 10 - 12 min, and 0.4 g/L STI was added to a 3L spinner, and cells were transferred into a 30 L stirred tank bioreactor. On day 5, the cell density had reached its maximum (around 1.8 × 106 cells/mL). At an MOI of 0.3 with serum-free medium conditions, cell infection yielded a maximal rabies virus titer of 1.82 × 107 FFU/mL at 5 days. All cell culture conditions and virus growth kinetics in serum-free media were investigated. In conclusion, Vero cells were grown on Cytodex 1 with serum-free media and a high amount of rabies virus was obtained. A mouse challenge was used to determine the immune response to an inactivated rabies virus vaccine candidate. Also, we evaluated inactive rabies vaccine candidate safety, and immunogenicity in mice, sheep, horses, and cattle. We found that no horses, sheep, or cattle who were given vaccine IM at 3.2 IU/dose exhibited any clinical sign of disease and all developed high VNA titers (up to 10.03 IU/mL) by 3 - 4 WPI. After the accelerated stability studies, the lyophilized inactivated rabies vaccine candidate showed enough antigenic potency (2.6 IU/mL) in the mouse challenge test. Also, 18-month long-term stability studies showed enough immune response (1.93 IU/mL) on day 14. The activity of the vaccine candidate showed a good immune response and safety criteria that meet WHO requirements. This is the first pilot-scale mammalian cell-based viral rabies vaccine production study in Türkiye that used microcarriers.展开更多

文摘Industrial wastewater from modern industrial production often contains excessive organic hazardous substances or excessive salts, acids and bases, etc. Traditional methods cannot play an effective role in the treatment of such wastewater. Based on this, the article focuses on the coal chemical high salt wastewater electro-catalytic oxidation pilot study.
文摘The upstream process was carried out in an animal component-free medium on Cytodex 1 microcarriers. This is the first pilot-scale mammalian cell-based viral rabies vaccine production study in Türkiye that used microcarriers.

文摘Advances in the identification of molecular biomarkers and the development of targeted therapies have enhanced the prognosis of patients with advanced gastric cancer.Several established biomarkers have been widely integrated into routine clinical diagnostics of gastric cancer to guide personalized treatment.Human epidermal growth factor receptor 2(HER2)was the first molecular biomarker to be used in gastric cancer with trastuzumab being the first approved targeted therapy for HER2-positive gastric cancer.Programmed death-ligand 1 positivity and microsatellite instability can guide the use of immunotherapies, such as pembrolizumab and nivolumab.More recently, zolbetuximab has been approved for patients with claudin 18.2-positive diseases in some countries.More targeted therapies, including savolitinib for MET-positive patients, are currently under clinical investigation. However, the clinical application of these diagnostic approaches could be hampered by many existing challenges, including invasive and costly sampling methods, variability in immunohistochemistry interpretation, high costs and long turnaround times for next-generation sequencing, the absence of standardized and clinically validated diagnostic cut-off values for some biomarkers, and tumor heterogeneity.Novel testing and analysis techniques, such as artificial intelligence-assisted image analysis and multiplex immunohistochemistry, and emerging therapeutic strategies, including combination therapies that integrate immune checkpoint inhibitors with targeted therapies, offer potential solutions to some of these challenges. This article reviews recent progress in gastric cancer testing, outlines current challenges, and explores future directions for biomarker testing and targeted therapy for gastric cancer.

文摘The mechanical behavior of cohesive soil is sensitized to drying-wetting cycles under confinements. However, the hydromechanical coupling effect has not been considered in current constitutive models.A macro-micro analysis scheme is proposed in this paper to investigate the soil deformation behavior under the coupling of stress and drying-wetting cycles.A new device is developed based on CT(computerized tomography)workstation to apply certain normal and shear stresses on a soil specimen during drying-wetting cycles.A series of tests are conducted on a type of loess with various coupling of stress paths and drying-wetting cycles. At macroscopic level, stress sensor and laser sensor are used to acquire stress and strain, respectively. The shear and volumetric strain increase during the first few drying-wetting cycles and then become stable. The increase of the shear stress level or confining pressure would cause higher increase rate and the value of shear strain in the process of drying-wetting cycles. At microscopic level, the grayscale value(GSV)of CT scanning image is characterized as the proportion of soil particles to voids.A fabric state parameter is proposed to characterize soil microstructures under the influence of stress and drying-wetting cycle.Test results indicate that the macroand micro-responses show high consistence and relevance. The stress and drying-wetting cycles would both induce collapse of the soil microstructure, which dominants degradation of the soil mechanical properties. The evolution of the macro-mechanical property of soil exhibits a positive linear relationship with the micro-evolution of the fabric state parameter.

文摘With the depletion of fossil fuels and increasing environmental concerns, the development of renewable energy, such as wave energy, has become a critical component of global energy strategies. However, challenges persist in the field testing methodologies for wave energy converters(WECs).In this paper,a numerical wave field of the Dawanshan Island Sea Area in Zhuhai City is constructed based on the MIKE21 SW wave model and by using an NCEP wind field driving model. In conjunction with the IEC-62600-100 standard,by taking site testing of the“Wanshan”wave energy converter on which a sea trial has been conducted in Dawanshan Island of Zhuhai city as an example, research on-site testing method for a wave energy converter has been carried out.The wave measurement position for the“Wanshan”converter was determined by combining statistically analyzed field data with a validated numerical wave model.By comparing a valid wave height at the position where a wave rider is located with a valid wave height at the position where the“Wanshan”wave energy converter is situated, the correlation coefficient between simulation and observed data reached 0.90,with a root-mean-square error of 0.19.The representativeness of wave measurement data during site testing is verified and can be used as a basis for calculating the input energy of the“Wanshan”wave energy converter.
