Introduction The plants of genus Cistanche G. Beck, which belong to Orobanchaceae family, include about twenty species, and there are four species and one variety are distributed in the northwest part of China. The dried fleshy stem of Cistanche genus plant known as 'Roucongrong' in Chinese traditional medicine, has long been used for kidney deficiency, female infertility, morbid leucorrhea, and neurasthenia. Studies on active components and pharmacological activities show that it possesses significant activities in enhancing potency, anti-fatigue, immuno-modulability, etc, and phenylethanoid glycosides are the major active components. Due to excessive exploitation, its natural resources are facing with exhaustion. In order to improve this situation, the plant has been cultivated on a large scale in the northwest part of China. Up to now, the chemical investigations of the cultivated Cistartcbe salsa have not been undertaken. In this paper we report seven phenylethanoid glycosides from the cultivated Cistanche salsa: echinacoside (1), cistanoside A (2), acteoside (3), isoacteoside (4), 2'-acetylacteoside (5), tubuloside B (6), and eutigoside A (7). Among them, compound 7 isolated fronl family Orobanchaceae was reported previously, and compounds 1, 2, 3, 4, 5 and 6 from the cultivated Cistanche salsa are reported for the first time. 展开更多

Two phenylethanoid glycosides (PhGs) were isolated and purified from the aerial parts of Plantago asiatica for the first time by high performance centrifugal partition chromatography (HPCPC) using ethyl acetate-n-butanol-ethanol-water (0.5:0.5:0.1:1, v/v/v/v). A total of 45.6 mg of compound 1 and 293.8 mg of compound 2 were purified from 1341 mg of the n-butanol extract of P. asiatica. The structures of the two PhGs were tentatively identified as plantamajoside and aeteoside or isoacteoside by eleetrospray ionization multi stage tandem mass spectrometry (ESI-MS^n) in the negative ion mode. 展开更多

In the present study, a method for the quantitative analysis of multi-components by single marker(QAMS) has been developed and validated for the simultaneous determination of echinacoside(ECH), tubuloside A, acteoside, isoacteoside, and2’-acetylacteoside in Cistanches Herba. ECH was used as the internal standard(IS) to obtain the relative correction factors(RCFs) of the other four phenylethanoid glycosides(PhGs);meanwhile, various influencing factors on RCFs were investigated under different conditions. The content of each component was calculated with RCF. The results were compared with those obtained by the external standard method(ESM) to verify the feasibility and accuracy of the established QAMS method. No significant difference was found in the quantitative results of 10 batches of Cistanches Herba between QAMS and ESM. The proposed QAMS method for simultaneous determination of PhGs in Cistanches Herba is accurate and feasible, providing an efficient and economical approach for the quality control of Cistanches Herba. 展开更多

Cymbaria daurica L. is a well-known traditional Mongolian medicine, which has been used to treat diabetesrelated conditions characterized by persistent thirst and hunger, copious urination, and weight loss. We aimed to investigate the protective effects of C. daurica extracts and phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. We discovered phenylethanoid glycosides from n-butanol extract with large content through extraction and separation. We continue to study the protective effects of phenylethanoid glycosides including verbascoside and isoacteoside on INS-1 cells. INS-1 cells were treated with C. daurica, cell viability assay, RNA-seq technology, superoxide dismutase activity and malonaldehyde content, quantitative real time-PCR and Western blot analysis were used to study the protective effects of C. daurica. Cell viability assay resulted that n-butanol extract and verbascoside, isoacteoside showed protective effects of C. daurica. According to the RNA-seq technology to identify the differentially expressed genes in INS-1 cells, the pathway of gene enrich the protective effect of C. daurica on oxidative stress. SOD activity and the content of MDA indicated that C. daurica could enhance the antioxidant capacity of INS-1 cells. Further investigation indicated C. daurica alleviate oxidative stress by inhibiting INS-1 cell apoptosis. C. daurica may play an anti-diabetic role by inhibiting islet cell apoptosis. 展开更多

AIM: To investigate the effects of feeding phenylalanine(Phe) and tyrosine(Tyr) on the accumulation of total phenolic compounds and four phenylethanoid glycosides(PeGs) to a cell suspension culture of the parasitic plant Cistanche deserticola. METHOD: A cell suspension culture of C. deserticola was established and precursors of different concentrations were fed. In each group, the cell was sampled at the 24th day after inoculation. The content of total phenolic compounds and four PeGs compounds were determined using the Folin-Ciocalteu method and an HPLC method, respectively. RESULTS: In the Phe fed cells, the maximum PeGs yield was achieved when Phe was fed at 1.5 mmol·L-1 and the yield reached 1.13 times the control cell concentration. In the Tyr fed cells, the maximum yield of PeGs was 1.60 times of control when 0.75 mmol·L-1 Tyr was fed to the cells. Furthermore, it was found that the salidroside yield was 4.01 times of control group when 5 mmol·L-1 Tyr was fed. CONCLUSION: Tyr is a better precursor for PeGs accumulation compared with Phe, and the rate limiting enzymes might be involved in the Tyr branch. 展开更多

Phenylethanoid glycosides(PhGs)are a group of important natural products widely distributed in medicinal plants and known for their remarkable pharmacological properties.Uridine diphosphate(UDP)glycosyltransferase 79G15(UGT79G15)from Rehmannia glutinosa catalyzes the conversion of osmanthuside A to osmanthuside B,a key intermediate in the PhG biosynthetic pathway,through the formation of a(1→3)glycosidic bond.In this study,we present the crystal structures of UGT79G15 in its apo form,UDP-bound form,and,notably,its ternary complex containing UDP and a mimic acceptor,forsythiaside A,within its active site.Structural and comparative analyses revealed that UGT79G15 possesses a distinctive funnel-shaped acceptor-binding pocket with a small auxiliary cavity capable of accommodating the 4′-hydroxycinnamoyl group of PhGs,explaining the enzyme’s regiospecificity toward the 3′-OH of the acceptor.Additional structural examination and site-directed mutagenesis identified key residues that recognize and stabilize UDP-rhamnose and the sugar acceptor.Among the variants generated,I204W exhibits enhanced catalytic efficiency for osmanthuside A conversion,reaching up to 2.2-fold higher activity than the wild type.This study provides mechanistic insight into the donor specificity and acceptor regioselectivity of PhG 1,3-rhamnosyltransferase and expands the structural understanding of plant UGTs. 展开更多

背景:研究证实肉苁蓉苷A具有抗炎、抗氧化和抗骨质疏松的作用,但其对破骨细胞分化和功能的影响及潜在的机制尚缺少研究。目的:探讨肉苁蓉苷A对核因子κB受体活化因子配体诱导的体外破骨细胞分化和骨吸收功能的影响及作用机制。方法:提... 背景:研究证实肉苁蓉苷A具有抗炎、抗氧化和抗骨质疏松的作用,但其对破骨细胞分化和功能的影响及潜在的机制尚缺少研究。目的:探讨肉苁蓉苷A对核因子κB受体活化因子配体诱导的体外破骨细胞分化和骨吸收功能的影响及作用机制。方法:提取与培养4-6周龄C57BL/6小鼠股骨和胫骨中的骨髓巨噬细胞,采用CCK-8毒性实验检验不同浓度的肉苁蓉苷A(5,10,20,40,80,160μmol/L)对骨髓巨噬细胞的毒性,抗酒石酸酸性磷酸酶染色观察不同浓度的肉苁蓉苷A干预破骨细胞的分化情况,确定药物的有效干预浓度。将实验分为阳性对照组、肉苁蓉苷A低、中、高剂量组(40,80,160μmol/L),细胞贴壁后各组均加入50 ng/mL的核因子κB受体活化因子配体诱导破骨细胞分化,肉苁蓉苷A低、中、高剂量组分别加入相应剂量的肉苁蓉苷A进行干预。采用肌动蛋白环鬼笔环肽染色和DAPI染色检测肉苁蓉苷A对破骨细胞形成的影响;骨磨片甲苯胺蓝染色观察肉苁蓉苷A对破骨细胞骨吸收功能的影响;Western blot检测JNK/MAPK通路上下游蛋白的表达情况;RT-qPCR检测抗酒石酸酸性磷酸酶、DC-STAMP、Nfatc-1、Ctsk和c-Fos等破骨细胞分化和骨吸收功能相关基因的表达情况。结果与结论:①抗酒石酸酸性磷酸酶染色、肌动蛋白环染色和骨陷窝甲苯胺蓝染色结果表明,与阳性对照组相比,肉苁蓉苷A对核因子κB受体活化因子配体诱导的体外破骨细胞的分化和骨吸收功能具有呈剂量依赖性的显著抑制作用;②RT-qPCR结果表明,与阳性对照组相比,肉苁蓉苷A高剂量组和低剂量组的抗酒石酸酸性磷酸酶、DC-STAMP、Nfatc-1、Ctsk和c-Fos等的mRNA表达量均显著降低,且肉苁蓉苷A高剂量组的降低效果更加显著;③Western blot结果显示,高剂量肉苁蓉苷A干预10,20,30和60 min时显著抑制p-JNK蛋白的表达;与阳性对照组相比,肉苁蓉苷A低、中、高剂量组显著抑制Nfatc1和c-Fos蛋白的表达,且呈剂量依赖性;④结果说明,肉苁蓉苷A能够通过降低p-JNK蛋白水平,抑制MAPK通路的激活和破骨细胞关键基因的表达,进而抑制破骨细胞的形成和骨吸收功能。 展开更多

为探究肉苁蓉苯乙醇苷(PhGs)和光甘草定(Gla)协同抑制皮肤色素沉着作用,通过体外酪氨酸酶活性抑制实验、1,1-二苯基-2-三硝基苯肼(DPPH)和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐阳离子(ABTS+)自由基清除实验,初步筛选了PhGs和Gla... 为探究肉苁蓉苯乙醇苷(PhGs)和光甘草定(Gla)协同抑制皮肤色素沉着作用,通过体外酪氨酸酶活性抑制实验、1,1-二苯基-2-三硝基苯肼(DPPH)和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐阳离子(ABTS+)自由基清除实验,初步筛选了PhGs和Gla的复配质量比。进一步构建了UVB诱导B16F10细胞色素沉着模型,以酪氨酸酶活性、黑色素含量为指标,评价了药物抑制黑色素的生成作用;构建了脂多糖(LPS)诱导HaCaT细胞炎症模型,以抑制白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)释放效果为指标,评价了药物的抗炎作用;构建偶氮二异丁脒盐酸盐(AAPH)诱导HaCaT细胞氧化应激模型,以提升超氧化物歧化酶(SOD)活力、过氧化氢酶(CAT)活力为指标,评价了药物的抗氧化作用。通过上述3种细胞模型综合筛选出最佳的PhGs和Gla复配质量比。结果表明,以m(PhGs)∶m(Gla)=1∶1、5∶1、10∶1制备的PhGs/Gla(1∶1)、PhGs/Gla(5∶1)、PhGs/Gla(10∶1)磷酸盐缓冲溶液(PBS,pH=6.8)在抑制酪氨酸酶活性、抗氧化方面具有较好的抑制作用和协同效应,对酪氨酸酶抑制率分别为94.37%、92.93%和88.06%;对DPPH自由基清除率分别为89.44%、88.72%和88.10%;对ABTS+自由基清除率分别为100.13%、100.01%和99.87%。当质量浓度为25μg/mL时,PhGs/Gla(1∶1)、PhGs/Gla(5∶1)、PhGs/Gla(10∶1)DMEM溶液未对B16F10和HaCaT细胞表现出细胞毒性;PhGs/Gla(1∶1)DMEM溶液的酪氨酸酶活性抑制能力更强(酪氨酸酶活性23.80%),黑色素含量(30.90%)显著降低;PhGs/Gla(1∶1)DMEM溶液抑制IL-6、TNF-α释放效果,以及增强SOD、CAT活力的能力最佳,表现出良好的PhGs和Gla复配协同性能,均优于单一用药,也高于PhGs/Gla(5∶1)、PhGs/Gla(10∶1)DMEM溶液。PhGs和Gla复配通过抑制黑色素生成、抗炎及抗氧化作用,协同发挥改善皮肤色素沉着药效。 展开更多

为解决目前中药质控体系中存在的质控标准合理性欠佳且难以体现药材整体优劣水平等“瓶颈”性问题。以地黄药材为例,加强地黄“效-物”相关度,针对地黄的多种功效,以“化学成分群”作为指标,建立系统、合理的控制标准。将中药有效性进... 为解决目前中药质控体系中存在的质控标准合理性欠佳且难以体现药材整体优劣水平等“瓶颈”性问题。以地黄药材为例,加强地黄“效-物”相关度,针对地黄的多种功效,以“化学成分群”作为指标,建立系统、合理的控制标准。将中药有效性进行完整的表达,建立整体性的中药质控指标,科学全面地反映地黄的内在质量,提升中药及其产品质量标准,以期对中药高质量发展提供参考。该研究根据鲜地黄、生地黄中主要化学成分-苷类物质在干预肾阴虚模型过程中的“效-物”关系,构成“药效-化学成分群”结构;建立“化学成分群指纹图谱”,基于道地产区不同产地不同储藏年限的地黄,对2个活性部位进行定量表征。环烯醚萜苷部位化学成分群指纹图谱中选出12个特征峰,苯乙醇苷部位化学成分群指纹图谱中选出13个特征峰。参照含量测定的计算方法,引入质控系数(Q),建立数学模型。以梓醇、地黄苷D和毛蕊花糖苷作为指标参照峰,计算各Q,Q_(Z)=0.17×A_总/A_(min),Q_(D)=0.085×A_总/A_(min),Q_(M)=0.017×A_总/A_(min),定义当检测样品为环烯醚萜苷部位时,算得梓醇Q_(Z)≥0.57,或地黄苷D Q_(D)≥0.69时,样品环烯醚萜苷活性部位合格;当检测样品苯乙醇苷为部位时,计算毛蕊花糖苷Q_(M)≥0.019时,样品苯乙醇苷活性部位合格。且Q越大则苷类部位含量越高,认为Q在反映样品真伪的基础上同时可反映样品的优劣程度。根据Q评价提示贮藏时间6年内的地黄质量较佳,且贮藏年限越短质量越优。同年样品之间进行比较,显示Q_(Z)、Q_(D)、Q_(M)3个指标参照评价结果基本平行一致。通过构建地黄中“干预调控肾阴虚-环烯醚萜苷”和“干预调控肾阴虚-苯乙醇苷”的“药效-化学成分群”结构关系,成功建立Q,该建立办法为其他中药及其产品质量评价标准提升提供参考。 展开更多

沧江糙苏化学成分及体外抗肿瘤细胞活性研究

目的 对沧江糙苏的化学成分及其体外抗肿瘤细胞活性进行研究。方法 通过正反相硅胶柱、Sephadex LH-20凝胶柱、HPLC等色谱技术对沧江糙苏95%乙醇水提取物及90%石油醚-10%三氯甲烷混合液提取物的化学成分进行分离,运用核磁共振、质谱、... 目的 对沧江糙苏的化学成分及其体外抗肿瘤细胞活性进行研究。方法 通过正反相硅胶柱、Sephadex LH-20凝胶柱、HPLC等色谱技术对沧江糙苏95%乙醇水提取物及90%石油醚-10%三氯甲烷混合液提取物的化学成分进行分离,运用核磁共振、质谱、红外光谱、紫外光谱等方法对化合物进行分析鉴定,通过CCK-8法进行化合物抗肿瘤细胞活性的筛选。结果 从沧江糙苏提取物中分离鉴定得18个化合物,具体为:salvialoside A(1)、毛蕊花糖苷(2)、木犀草素-7-O-β-D-葡萄糖苷(3)、alyssonoside(4)、8-O-乙酰山栀苷甲酯(5)、5,9-epi-7,8-didehydropens-temoside(6)、胡麻属苷(7)、十四烷酸(8)、胡萝卜苷(9)、compound VI(10)、乙基-β-D-糖苷(11)、6,9-epi-8-O-乙酰山栀苷甲酯(12)、山栀苷甲酯(13)、5,9-epi-phlomiol(14)、phlomiol(15)、5,9-epi-penstemoside(16)、落叶松脂醇-9-O-β-D-葡萄糖苷(17)、巴西木素(18)。不同浓度(40、8、1.6、0.32、0.064μmol/L)化合物14、15、16对HCT-15细胞有一定的抑制作用,随着药物浓度增加,抑制率增加,其中化合物14的半数抑制浓度为(5.86±0.77)μmol/L。结论 compound VI、落叶松脂醇-9-O-β-D-葡萄糖苷、巴西木素(分别为化合物10、17、18)为该属首次分离得到的化合物。化合物1~18为首次从沧江糙苏中分离。化合物14可抑制HCT-15细胞增殖,具有一定的抗肿瘤活性。 展开更多

[目的]探究高静压处理的压力、时间和切片厚度对鲜切肉苁蓉品质和苯乙醇苷提取量的影响。[方法]采用单因素结合响应面试验的方法优化高静压处理在肉苁蓉中的应用参数。[结果]单因素优化结果显示:随着压力从100 MPa升高到500 MPa和处理时间从10 min延长到25 min,肉苁蓉色差值、硬度和苯乙醇苷提取量增加;随着切片厚度增加(2~10 mm),肉苁蓉色差增加,但硬度和苯乙醇苷提取量降低;三者最优参数为200~300 MPa、15~25 min、2~6 mm。响应面优化结果显示,压力对苯乙醇苷提取量的影响更为显著;最优条件为压力294.341 MPa、时间14.658 min、切片厚度3.644 mm,此时苯乙醇苷提取量较高,为(5.35±0.21) mg/g。[结论]高静压处理的压力作为主要因素改善了肉苁蓉的品质,并促进了苯乙醇苷的提取,而切片厚度的减薄效应和处理时间的适度延长作为次要因素影响苯乙醇苷提取量。 展开更多

目的基于SIRT2-C/EBPβ-AREG轴探讨肉苁蓉苯乙醇苷(CPhGs)通过调控成骨分化改善骨质疏松的作用机制。方法采用卵巢切除法建立小鼠骨质疏松模型。分别采用micro-CT、钙黄绿素双标记和苏木素-伊红染色评估骨小梁数量/厚度、骨形成率和组... 目的基于SIRT2-C/EBPβ-AREG轴探讨肉苁蓉苯乙醇苷(CPhGs)通过调控成骨分化改善骨质疏松的作用机制。方法采用卵巢切除法建立小鼠骨质疏松模型。分别采用micro-CT、钙黄绿素双标记和苏木素-伊红染色评估骨小梁数量/厚度、骨形成率和组织形态。采用Western blotting、免疫共沉淀检测SIRT2-C/EBPβ-AREG轴相关蛋白SIRT2、C/EBPβ、AREG表达。采用实时定量PCR检测成骨分化标志基因OCN、OPN、RUNX2、C/EBPβ、AREG mRNA表达。结果CPhGs干预显著改善卵巢切除小鼠骨小梁微结构并促进骨形成,其机制涉及激活SIRT2介导的C/EBPβ去乙酰化,进而上调AREG表达。细胞实验证实,CPhGs通过增强SIRT2与C/EBPβ的相互作用,显著提高MC3T3-E1细胞中碱性磷酸酶活性及OCN、OPN、RUNX2等成骨基因表达,且该效应可被SIRT2敲低逆转。结论CPhGs通过SIRT2-C/EBPβ-AREG轴调控成骨分化进程,为中药活性成分治疗骨质疏松症提供了新的分子靶点和理论依据。 展开更多

目的采用智能感官结合化学计量学方法对春秋两季节采收肉苁蓉Cistanche deserticola进行品质评价,为肉苁蓉的生产和质量控制提供参考。方法利用色差仪测定肉苁蓉样品的色度值,利用电子鼻测定肉苁蓉的气味,采用HPLC测定肉苁蓉的环烯醚萜... 目的采用智能感官结合化学计量学方法对春秋两季节采收肉苁蓉Cistanche deserticola进行品质评价,为肉苁蓉的生产和质量控制提供参考。方法利用色差仪测定肉苁蓉样品的色度值,利用电子鼻测定肉苁蓉的气味,采用HPLC测定肉苁蓉的环烯醚萜和苯乙醇苷含量,最后测定其多糖含量,采用皮尔逊相关性分析、正交-偏最小二乘法判别分析等方法进行综合分析,比较春季和秋季采收肉苁蓉的质量。结果秋季和春季采收肉苁蓉药材气味差异主要在氮氧化合物和萜烯类成分。颜色上春季采收肉苁蓉的色度值L^(*)和b^(*)高于秋季,说明春季的明亮色和黄色高于秋季,秋季采收肉苁蓉颜色更深。质地上秋季采收肉苁蓉硬度值显著低于春季,秋季采收肉苁蓉质地更为柔软;苯乙醇苷类和环烯醚萜类含量二者差异显著,秋季采收肉苁蓉的京尼平苷酸、马钱苷酸、松果菊苷、肉苁蓉苷A和毛蕊花糖苷含量均显著高于春季,分别是春季采收肉苁蓉的2.1、4.1、3.1、3.4、1.4倍。多糖含量为秋季采收肉苁蓉低于春季,但二者差异不显著。结论秋季采收肉苁蓉较春季在颜色上更深,质地上更柔软,主要活性成分苯乙醇苷类和环烯醚萜类成分含量更高。研究结果可为不同季节采收肉苁蓉的品质评价提供依据。 展开更多
