RNA polymerase transcriptional pausing represents a major checkpoint in transcription in bacteria and metazoans,but it is unknown whether this phenomenon occurs in plant organelles.Here,we report that transcriptional ...RNA polymerase transcriptional pausing represents a major checkpoint in transcription in bacteria and metazoans,but it is unknown whether this phenomenon occurs in plant organelles.Here,we report that transcriptional pausing occurs in chloroplasts.We found that mTERF5 specifically and positively regulates the transcription of chloroplast psbEFLJ in Arabidopsis thaliana that encodes four key subunits of photosystem II.We found that mTERF5 causes the plastid-encoded RNA polymerase(PEP)complex to pause at psbEFLJ by binding to the+30 to+51 region of double-stranded DNA.Moreover,we revealed that mTERF5 interacts with pTAC6,an essential subunit of the PEP complex,although pTAC6 is not involved in the transcriptional pausing at psbEFLJ.We showed that mTERF5 recruits additional pTAC6 to the transcriptionally paused region of psbEFLJ,and the recruited pTAC6 proteins could be assembled into the PEP complex to regulate psbEFLJ transcription.Taken together,our findings shed light on the role of transcriptional pausing in chloroplast transcription in plants.展开更多
Lesions on the DNA template can impact transcription via distinct regulatory pathways.Ionizing radiation(IR)as the mainstay modality for many malignancies elicits most of the cytotoxicity by inducing a variety of DNA ...Lesions on the DNA template can impact transcription via distinct regulatory pathways.Ionizing radiation(IR)as the mainstay modality for many malignancies elicits most of the cytotoxicity by inducing a variety of DNA damages in the genome.How the IR treatment alters the transcription cycle and whether it contributes to the development of radioresistance remain poorly understood.Here,we report an increase in the paused RNA polymerase II(RNAPII),as indicated by the phosphorylation at serine 5 residue of its C-terminal domain,in recurrent nasopharyngeal carcinoma(NPC)patient samples after IR treatment and cultured NPC cells developing IR resistance.Reducing the pool of paused RNAPII by either inhibiting TFIIH-associated CDK7 or stimulating the positive transcription elongation factor b,a CDK9-CycT1 heterodimer,attenuates IR resistance of NPC cells.Interestingly,the poly(ADP-ribosyl)ation of CycT1,which disrupts its phase separation,is elevated in the IR-resistant cells.Mutation of the major poly(ADP-ribosyl)ation sites of CycT1 decreases RNAPII pausing and restores IR sensitivity.Genome-wide chromatin immunoprecipitation followed by sequencing analyses reveal that several genes involved in radiation response and cell cycle control are subject to the regulation imposed by the paused RNAPII.Particularly,we identify the NIMA-related kinase NEK7 under such regulation as a new radioresistancefactor,whose downregulation results in the increased chromosome instability,enabling the development of IR resistance.Overall,our results highlight a novel link between the alteration in the transcription cycle and the acquisition of IR resistance,opening up new opportunities to increase the efficacy of radiotherapy and thwart radioresistance in NpC.展开更多

The data for this study were collected from two E...This paper examined the pedagogical employment of pauses in classroom interactions in EFL reading lessons in senior high school using a conversation analytic approach. The data for this study were collected from two EFL classes at the same Chinese high school, each taught by a different teacher. The length and context of pauses naturally occurring in the whole lessons were analyzed in terms of reading lessons in EFL classrooms. Findings were as follows. Appropriately prolonged pauses combined with teachers' explicit interventions can enhance the quality and quantity of students' answers and promote their involvement, maximizing their learning opportunity. These results offer valuable insights for recommendations regarding classroom interactions for teaching reading in English lessons in senior high school.展开更多

Effects of reduced frequency, stop angle, and pause duration have been studied on a thin supercritical airfoil undergoing a pitch-pause-return motion, which is one of the classic maneuvers introduced by the AIAA Fluid...Effects of reduced frequency, stop angle, and pause duration have been studied on a thin supercritical airfoil undergoing a pitch-pause-return motion, which is one of the classic maneuvers introduced by the AIAA Fluid Dynamics Technical Committee. Experiments were conducted in a low-speed wind tunnel at both a constant mean angle of attack and an oscillation amplitude with a reduced frequency ranging from 0.01 to 0.12. The desired stop angles of the airfoil were set to occur during the upstroke motion. The unsteady pressure distribution on the airfoil was measured for below, near, and beyond static stall conditions. Results showed that the reduced frequency and stop angle were the dominant contributors to the time lag in the flowfield. For stop angles in both belowand post-stall regions, the time for the flowfield to reach its steady state conditions, known as the time lag, decreased as the reduced frequency was increased. However, in the static-stall region and for a certain value of reduced frequency, a resonance phenomenon was observed, and a minimum time lag was achieved. The pressure distribution in this condition was shown to be highly influenced by this phenomenon.展开更多

The Bacillus subtilis phage φ29 is an excellent model system to investigate the DNA packaging mechanism ...During the assembly of many viruses, a powerful molecular motor packages the genome into a preassembled capsid. The Bacillus subtilis phage φ29 is an excellent model system to investigate the DNA packaging mechanism because of its highly efficient in vitro DNA packaging activity and the development of a single-molecule packaging assay. Here we make use of structural and biochemical experimental data to build a physical model of DNA packaging by the φ29 DNA packaging motor. Based on the model, various dynamic behaviours such as the packaging rate, pause frequency and slip frequency under different ATP concentrations, ADP concentrations, external loads as well as capsid fillings are studied by using Monte Carlo simulation. Good agreement is obtained between the simulated and available experimental results. Moreover, we make testable predictions that should guide future experiments related to motor function.展开更多

Electrochemical CO_(2) reduction reaction(CO_(2) RR)exhibits remarkable potential in producing valuable chemicals with renewable energy.Operating CO_(2) RR in acidic media is beneficial to solve the issue of low carbo...Electrochemical CO_(2) reduction reaction(CO_(2) RR)exhibits remarkable potential in producing valuable chemicals with renewable energy.Operating CO_(2) RR in acidic media is beneficial to solve the issue of low carbon utilization brought by(bi)carbonate formation at the cathode.Suppressing the competing hydrogen evolution reaction and achieving stable CO_(2) RR performance remains challenging.Herein,we constructed a 3-dimensional Cu(3D-Cu)gas diffusion electrode(GDE)to achieve efficient C_(2)H_(4) production with a partial current density(j C_(2)H_(4))of over 470 mA cm^(2) and a Faradaic efficiency(FE C_(2)H_(4)) of 40%.With pause electrolysis,the decay rate of the j C_(2)H_(4) is only half that of the traditional constant electrolysis.The GDE after constant electrolysis was found to suffer from severe salt formation,leading to the decreased activity and poor stability.展开更多
