Objective:To analyze the effects of Ageratum conyzoides L.on the monosodium iodoacetate induced osteoarthritis rats.Methods:Thin layer chromatography was performed to analyze the constituents of the babandotan extract leaves.White male Sprague-Dawley rats used in this study were divided into 6 groups:normal control and negative control groups,both given0.5%carboxymethyl cellulose;the positive control group that was given glucosamine and chondroitin suspension(486 mg/200 g B.W.);the 3 dose variation extract groups including dose 1,2,and 3 that were given 40,80,and 160 mg/200 g B.W.respectively on day 29 until50.All the groups were induced with 0.05 mL monosodium iodoacetate(20 mg/mL)on day1,except normal control induced by saline.Measurement of edema volume of rat knees was performed on day 0,8,15,22,29,43,and 50.Hematology data was measured at day 1,29and 50.Serum was collected at day 50 to evaluate TNF-α and MMP-9 by ELISA.Cartilage histopathology was evaluated by staining with H&E and Safranin-o-fast green staining on day 50.Results:The babandotan leaves extract dose 2(80 mg/200 g B.W.)and dose 3(160mg/200 g B.W.)could decrease the edema volume,increase the area and thickness of articular cartilage,and increase proteoglycan level.Particularly,dose 3(160 mg/200 g B.W.)of extract babandotan leaves were able to significantly decrease the number of leukocytes,lymphocytes and udem volume,and decrease TNF alpha and MMP-9 levels.Conclusions:Babandotan leaves extract can recover inflammation and cartilages degradation by inhibiting TNF-αin inflammation processes and MMP-9 in the collagenase reaction in the cartilages. 展开更多

This study is examined the effect of past and present lifestyle habits and nutrition on the os-teo-sono assessment index (OSI) in pre- and post-menopausal females. The subjects were 200 premenopausal females (38.8±10.3years) and 156 postmenopausal females (59.2±5.9 ye- ars). BMD (Body mineral density) was estimated by right-calcaneal OSI using an ultrasonic transmission method with an AOS-100 device (ALOKA). The number of postmenopau- sal fe-males in the close examination and guidance required groups (80 cases: 51.3 %) (OSI < 2.428) was significantly higher than that of premeno-pausal females (44 cases: 22.0 %) (χ2=33.105: P<0.000). In premenopausal females, the proportion of subjects that had not taken vitamin D in the past (in junior high school and high school) was significantly higher in the close examination- guidance required group (OSI < 2.428) than in the normal group (OSI ≧ 2.428). However, in postmenopausal females, there was no signifi-cant difference in past and present lifestyle habits and nutrition between the close exami-nation-guidance required group and the normal group. In premenopausal females, it was deter-mined that the intake of vitamin D during pu-berty increased the absorption of calcium sig-nificantly. 展开更多

In this study, we use a pluripotent mesenchymal stem cell (MSC) model, C3H/10T1/2, to evaluate three calcium phos-phate (CaP) materials, namely the hydroxyapatite (HA), α-tricalcium phosphate (α-TCP) and β-tricalcium phosphate (β-TCP). 10T1/2 cell was chosen as it has advantages over its counterparts in terms of ease of maintenance, free of ethical concerns and also more reproducible results. ALP enzymatic assay, RT-qPCR, DAPI staining and SEM were employed to assess the osteoinductivity of these materials. A good reference material which also acts as a scientific control is necessary for comparisons of results from different experimental batches and hence other materials such as titanium, Nunclon plastic surface, BD Falcon plastic surface and gold coated porous HA were also tested. The results show that ceramics induce a more sustained osteo-differentiation state as compared with plastics. Inductivity was found to be acting in descending order of strength with HA > β-TCP > α-TCP, which is reversed in terms of their impact on proliferation rate (HA TCP) and in vivo osteoinductivity in terms of incidence and quality of bone described previously (HA > β-TCP > α-TCP). These confirm the suitability of using 10T1/2 cells in cell culture assay of osteoinductivity. 展开更多

微创入路单髁置换术与全膝关节表面置换术治疗膝关节单间室骨性关节炎患者的临床效果对比

目的探讨膝关节单间室骨性关节炎(KOA)患者应用微创入路单踝置换术(UKA)与全膝关节表面置换术(TKA)的临床效果。方法回顾性分析青海省人民医院2014年4月-2019年4月收治的25例KOA患者的临床资料,根据手术方式的不同将其分为对照组(n=13)... 目的探讨膝关节单间室骨性关节炎(KOA)患者应用微创入路单踝置换术(UKA)与全膝关节表面置换术(TKA)的临床效果。方法回顾性分析青海省人民医院2014年4月-2019年4月收治的25例KOA患者的临床资料,根据手术方式的不同将其分为对照组(n=13)和观察组(n=12)。对照组采用TKA治疗,观察组采用微创入路UKA治疗。对比两组的临床指标、疼痛程度、膝关节屈曲度、膝关节功能及并发症发生情况。结果观察组手术、住院时间均短于对照组,术中出血量少于对照组,术后3 d血红蛋白下降量低于对照组,组间差异有统计学意义(P<0.05)。术后2周,观察组疼痛视觉模拟评分为(3.62±0.46)分,低于对照组的(5.12±0.53)分,差异有统计学意义(P<0.05);术后6个月,观察组膝关节屈曲度为(112.02±8.63)°,大于对照组的(97.11±7.27)°,差异有统计学意义(P<0.05)。整体分析结果显示,两组术前、术后6个月、术后12个月、术后24个月的美国特种外科医院膝关节功能量表(HSS)评分交互、组间、时间对比,差异有统计学意义(P<0.05);进一步两两比较,术前、术后24个月,两组HSS评分对比,组间差异无统计学意义(P>0.05);术后6个月、术后12个月,观察组HSS评分均高于对照组,组间差异有统计学意义(P<0.05)。两组并发症发生率对比,差异无统计学意义(P>0.05)。结论KOA患者采用TKA与微创入路UKA治疗膝关节功能的远期恢复效果接近,但微创入路UKA治疗患者的疼痛程度更低,膝关节屈曲度的恢复速度更快,且膝关节功能的近期恢复效果更明显。 展开更多

The reinforcement of calcium phosphate materials with silk fibroin (SF) has been one of the strategies to overcome the brittleness. However, the lack of osteoinductivity may still restrict their further use. This study aimed to investigate the biocompatibility and osteogenesis capacity of a novel Semaphorin 3A-loaded chitosan microspheres/SF/a-tricalcium phosphate composite (Sema3A CMs/SF/a-TCP) in vitro. Sema3A was first incorporated into CMs, and the Sema3A CMs/SF/a-TCP composite was then prepared. The morphology of the CMs was observed using SEM. The in vitro release kinetics, cytotoxicity, and cell compatibility were evaluated, and the real-time quantitative polymerase chain reaction (RT-qPCR) and activity of alkaline phosphatase (ALP) were used to evaluate the osteogenesis capacity of the composite. The in vitro release of Sema3A from the Sema3A CMs/SF/a-TCP composite showed a temporally controlled manner. The extract of the Sema3A CMs/SF/a-TCP composite presented no obvious side effect on the MC3T3-E1 cell proliferation, nor promote cell proliferation. The MC3T3-E1 cells were well-spread and presented an elongated shape on the Sema3A CMs/SF/a-TCP composite surface; the ALP activity and the osteogenic-related gene expression were higher than those seeded on the surface of the CMs/SF/a-TCP and SF/a-TCP composites. In conclusion, Sema3A CMslSF/a-TCP has excellent biocompatibility and contributes to the osteoblastic differentiation of MC3T3-E1 cells. 展开更多

目的比较经椎弓根旁外侧入路与经椎弓根入路椎体成形术治疗骨质疏松性胸腰椎压缩骨折的疗效及术后近期残留腰背部疼痛发生率的差异。方法纳入自2019-03—2020-05采用经皮椎体成形术治疗的58例新发单节段骨质疏松性胸腰椎压缩骨折,其中2... 目的比较经椎弓根旁外侧入路与经椎弓根入路椎体成形术治疗骨质疏松性胸腰椎压缩骨折的疗效及术后近期残留腰背部疼痛发生率的差异。方法纳入自2019-03—2020-05采用经皮椎体成形术治疗的58例新发单节段骨质疏松性胸腰椎压缩骨折,其中29例经椎弓根旁外侧入路(椎弓根旁外侧入路组),29例经椎弓根穿刺入路(椎弓根入路组),比较两组手术时间、骨水泥注入量、骨水泥渗漏率,以及术后疼痛VAS评分、术后近期残余腰背痛发生率、术后伤椎后凸角。结果两组手术时间、骨水泥渗漏率差异无统计学意义(P>0.05),但椎弓根旁外侧入路组骨水泥灌注量多于椎弓根入路组,差异有统计学意义(P<0.05)。所有患者均获得随访,随访时间3~12个月,平均6.5个月。两组术后疼痛均有明显缓解,椎弓根旁外侧入路组术后1 d、1周疼痛VAS评分比椎弓根入路组低,差异有统计学意义(P<0.05)。两组术后伤椎后凸角均较术前明显改善,但两组术后伤椎后凸角差异无统计学意义(P>0.05)。两组术后近期均有患者残余腰背部疼痛,椎弓根入路组术后1 d、术后1周残余腰背部疼痛发生率高于椎弓根旁外侧入路组,差异有统计学意义(P<0.05),两组术后1个月残余腰背部疼痛发生率差异无统计学意义(P>0.05)。结论经椎弓根旁外侧入路与经椎弓根入路椎体成形术治疗骨质疏松性胸腰椎压缩骨折均能获得满意疗效,但是经椎弓根旁外侧入路术后近期残留腰背痛发生率明显低于经椎弓根入路。 展开更多

Introduction: Pseudarthrosis (PSA) of the diaphysis of long bones still remains a current problem, despite improvements in the treatment of these fractures. Our study aims to study the epidemiological and therapeutic aspects of PSA of the diaphysis of long bones. Method: This retrospective work concerns 30 cases of non-union of the diaphysis of long bones treated in the orthopedic and trauma surgery department at Donka National Hospital, during a period of 18 months from January 1, 2019 to June 30, 2020. Results: We recruited 30 patients, 80% of whom were male, with an average age of 39.9 years. Public road accidents (AVP) represented the main cause of fractures of the diaphysis of long bones 87%, they were open in 25 cases or 83%. The fractures were located in the middle 1/3 of the diaphysis of the long bones in 50% of cases. Treatment of initial fractures was traditional in 21 cases, orthopedic in 2 cases and surgical in 7 cases. It was aseptic nonunion in 28 cases (93%) and septic nonunion in 2 cases. They were hypertrophic in 7 cases, slightly hypertrophic in 5 cases, oligotrophic in 11 cases, atrophic in 6 cases and with bone defect in 1 case. The treatment was based on osteosynthesis including 16 cases of screwed “PV” plate: 7 cases of centromedullary “ECM” nailing, 2 cases of external fixator, 1 case of broaching and 4 cases of Plastering. The results according to ASAMI criteria on an anatomical level were excellent in 19 cases, good in 3 cases and poor in 3 cases, with a union rate of 76%. And 5 patients undergoing consolidation. Conclusion: Based on the literature data and the experience of our department, the true treatment of PSA requires correct management of the initial fracture without forgetting the interest in preventing AVP which appears to be an element essential, making it possible to reduce the incidence of fractures of the diaphysis. 展开更多

全身低载高频振动参数交互作用下松质骨力学行为的模拟研究 被引量:1

目的:研究在全身低载高频振动(low-magnitude high-frequency vibration,LMHFV)参数交互作用下松质骨的力学行为,以为临床治疗废用性骨质疏松提供理论指导。方法:选取股骨中段松质骨通过Mimics软件进行三维重建,并将其导入Comsol软件中... 目的:研究在全身低载高频振动(low-magnitude high-frequency vibration,LMHFV)参数交互作用下松质骨的力学行为,以为临床治疗废用性骨质疏松提供理论指导。方法:选取股骨中段松质骨通过Mimics软件进行三维重建,并将其导入Comsol软件中建立三维流固耦合有限元模型。设定12种振动方案(振动加速度幅值a为0.015×g、0.02×g、0.03×g,振动频率f为30、45、60、100 Hz)进行模拟研究,分析LMHFV参数交互作用下松质骨的流体力学微环境、应力及变形位移的分布规律。结果:骨小梁表面骨髓流速及其变形位移随a增加而增大,随f增加而减小。在振动方案(0.015×g/0.02×g/0.03×g,30 Hz)下,骨小梁产生的变形位移平均值较高。在LMHFV参数范围(0.02×g~0.03×g,30~35 Hz)内,骨小梁的von Mises应力值较高。结论:全身LMHFV可显著改善松质骨的受力及流体力学环境,临床上可通过合理调控LMHFV参数来提高力介导的成骨细胞生物活性,进而促进骨生成。 展开更多

目的 研究表明微核糖核酸(microRNA,MiRNA)和Kruppel样因子10(Kruppel-like factor10, KLF10)调控骨质疏松症的进程。研究旨在探讨miR-26b/KLF10信号调控骨髓间充质干细胞(bone marrow-derived mesenchymal stem cells, BMSCs)分化的机... 目的 研究表明微核糖核酸(microRNA,MiRNA)和Kruppel样因子10(Kruppel-like factor10, KLF10)调控骨质疏松症的进程。研究旨在探讨miR-26b/KLF10信号调控骨髓间充质干细胞(bone marrow-derived mesenchymal stem cells, BMSCs)分化的机制。方法 流失鉴定分离的大鼠BMSCs,不同剂量的补肾活血方处理BMSCs,检测其^([1-2])增殖能力,碱性磷酸酶活性和茜素红染色检测BMSCs的成骨分化潜能。qPCR检测不同剂量的补肾活血方处理BMSCs后,miR-26b和KLF10的表达水平。mirtarBase数据库检测KLF10 mRNA的3’非翻译区(untranslated regions, 3'UTR)上是否有miR-26b的结合位点,双荧光素酶报告实验检测miR-26b对KLF10启动子活性的影响。qPCR检测miR-26b/KLF10信号对BMSCs增殖和成骨分化标志物的表达的影响。Western blot检查Wnt/β-catenin信号是否被激活。结果 流式分析显示分离的原代细胞大部分是BMSCs。补肾活血方显著促进骨髓间充质干细胞的增殖,补肾活血方显著提高了BMSCs中碱性磷酸酶的水平和成骨分化的潜能。补肾活血方促进BMSCs中miR-26b和KLF10的表达。在KLF10 mRNA的3’UTR上有miR-26b的结合位点。在BMSCs中,miR-26b抑制KLF10的3’UTR荧光素酶活性。此外,miR-26b也抑制了KLF10的表达水平。CCK8(Cell Counting Kit-8)显示KLF10促进BMSCs增殖,qPCR显示miR-26b显著抑制成骨分化标志物的表达。而KLF10能够逆转miR-26b的抑制效应,提示miR-26b/KLF10信号介导了补肾活血方在BMSCs增殖和成骨分化中的效应,且补肾活血方通过激活Wnt/β-Catenin信号通路促进成骨。结论 补肾活血方促进BMSCs的成骨由miR-26b/KLF10/Wnt/β-Catenin信号轴调控。 展开更多

目的 探究硫酸氨基葡萄糖联合美洛昔康对膝骨关节炎患者血清成纤维细胞生长因子-2(fibroblast growth factor-2, FGF-2)、转化生长因子-β(transforming growth factor-β, TGF-β)、胰岛素样生长因子-1(insulin-like growth factor-1, ... 目的 探究硫酸氨基葡萄糖联合美洛昔康对膝骨关节炎患者血清成纤维细胞生长因子-2(fibroblast growth factor-2, FGF-2)、转化生长因子-β(transforming growth factor-β, TGF-β)、胰岛素样生长因子-1(insulin-like growth factor-1, IGF-1)水平及膝关节运动功能的影响。方法 选择2021年6月-2023年6月就诊的膝骨关节炎114例,以随机数字表法分为联合组和美洛昔康组各57例。美洛昔康组予美洛昔康片治疗,联合组在美洛昔康组基础上加用硫酸氨基葡萄糖胶囊治疗,均治疗6周后观察疗效,比较2组治疗前、治疗6周后膝关节运动功能、炎性因子、生长因子水平及治疗期间安全性。结果 治疗6周后,联合组总有效率为91.23%(52/57)高于美洛昔康组的73.68%(42/57)(P<0.05)。治疗6周后,2组5次坐立试验、2.4 m起立行走试验所需时间短于治疗前,且联合组短于美洛昔康组(P<0.05,P<0.01);西安大略和麦克马斯特大学骨关节炎指数评分及血清前列腺素E2、白细胞介素-17、基质金属蛋白酶-3水平低于治疗前,且联合组低于美洛昔康组(P<0.05,P<0.01)。治疗6周后,2组血清FGF-2、TGF-β、IGF-1水平均高于治疗前,且联合组高于美洛昔康组(P<0.05,P<0.01)。2组治疗期间总不良反应发生率比较差异无统计学意义(P>0.05)。结论 硫酸氨基葡萄糖联合美洛昔康可有效升高膝骨关节炎患者血清FGF-2、TGF-β、IGF-1水平,延缓软骨退行性病变,控制机体炎症反应,进而有效改善患者膝关节运动功能,疗效显著,安全性良好。 展开更多

目的探讨长链非编码RNA(lncRNA)核旁斑组装转录本1(NEAT1)通过调节细胞焦亡调控人骨髓间充质干细胞(hBMSCs)成骨分化的作用。方法培养hBMSCs 7 d诱导细胞成骨分化,并分为对照组(常规培养)、成骨分化组(成骨分化诱导)、pcD-NEAT1组(转染N... 目的探讨长链非编码RNA(lncRNA)核旁斑组装转录本1(NEAT1)通过调节细胞焦亡调控人骨髓间充质干细胞(hBMSCs)成骨分化的作用。方法培养hBMSCs 7 d诱导细胞成骨分化,并分为对照组(常规培养)、成骨分化组(成骨分化诱导)、pcD-NEAT1组(转染NEAT1过表达质粒)、pcD-null组(转染NEAT1过表达质粒阴性对照)、成骨分化+CML组[成骨分化诱导联合NOD样受体家族蛋白3(NLRP3)炎性小体激活剂(Nε)-羧甲基赖氨酸(CML)处理]、成骨分化+CML+pcD-NEAT1组(成骨分化诱导联合pcD-NEAT1与CML处理)。经茜素红染色检测细胞矿化程度;碱性磷酸酶(ALP)活性检测试剂盒检测ALP活性;CCK-8检测各组细胞存活率,高倍镜下观察细胞形态变化。TUNEL实验检测细胞凋亡率。qRT-PCR检测NEAT1的表达。Western blot检测IL-1β、IL-18、NLRP3、cleaved-caspase 1(cleaved-CASP1)、gasdermin D、Runt-相关转录因子2(RUNX2)、ALP、骨桥蛋白(OPN)的表达。结果与对照组比较,成骨分化组细胞的矿化程度增加,ALP活性升高(P<0.05),NEAT1和RUNX2、ALP、OPN蛋白表达均上调(P<0.05)。与pcD-null组比较,pcD-NEAT1组细胞的矿化程度增加,ALP活性升高(P<0.05),NEAT1和RUNX2、ALP、OPN蛋白表达均上调(P<0.05)。与成骨分化组比较,成骨分化+CML组细胞矿化程度减轻,ALP活性降低(P<0.05),细胞存活率降低(P<0.05),细胞凋亡率增加(P<0.05),细胞膜出现破裂,细胞膨大变形,NLRP3和IL-1β、IL-18、cleaved-CASP1、gasdermin D蛋白表达均显著上调(P<0.05),而RUNX2、ALP、OPN蛋白表达均显著下调(P<0.05)。与成骨分化+CML组比较,成骨分化+CML+pcD-NEAT1组细胞矿化程度增加,ALP活性升高(P<0.05),RUNX2、ALP、OPN蛋白表达均显著上调(P<0.05),细胞存活率增加(P<0.05),细胞凋亡率降低(P<0.05),细胞膜较完整且细胞形态正常,NLRP3和IL-1β、IL-18、cleaved-CASP1、gasdermin D蛋白表达均显著下调(P<0.05)。结论NEAT1过表达通过抑制NLRP3炎性小体介导的细胞焦亡促进hBMSCs的成骨分化。 展开更多
