Background Recent studies have suggested a potential role of the oral microbiome in the development of cardiovascular diseases.This study aims to investigate the association between oral microbiota and cardiovascular ...Background Recent studies have suggested a potential role of the oral microbiome in the development of cardiovascular diseases.This study aims to investigate the association between oral microbiota and cardiovascular disease risk,including atrial fibrillation,myocardial infarction,chronic heart failure,and hypertension.Methods We analyzed GWAS data from East Asian populations'oral microbiome,involving 2,017 tongue and 1,915 saliva samples from 2,984 individuals with whole-genome sequencing.Additionally,we sourced cardiovascular disease GWAS data from NBDC,including atrial fibrillation(8,180 cases,28,621 controls),myocardial infarction(14,992 cases,146,214 controls),chronic heart failure(10,540 cases,168,186 controls),and systolic blood pressure(145,505 individuals).Results Several oral microbiota taxa were found to be significantly associated with cardiovascular disease outcomes.Specific microbiota,such as Centipeda,Corynebacterium,and Pseudomonas E,were negatively correlated with heart failure.In contrast,taxa like Neisseria D and Actinomyces were associated with an increased risk of atrial fibrillation and myocardial infarction.Additionally,certain oral microbiota showed correlations with changes in blood pressure,highlighting their potential role in hypertension.Conclusion Our findings suggest that the oral microbiota may influence the development and progression of cardiovascular diseases,providing new insights into the potential impact of oral health on cardiovascular risk.展开更多

Oral squamous cell carcinoma(OSCC)is a prevalent malignancy with high morbidity and mortality.Globally,about 400000 people are affected,often with a poor quality of life.Its high mortality is mainly due to its aggress...Oral squamous cell carcinoma(OSCC)is a prevalent malignancy with high morbidity and mortality.Globally,about 400000 people are affected,often with a poor quality of life.Its high mortality is mainly due to its aggressive growth and tendency to spread.Epithelial-mesenchymal transition(EMT)is a central regulatory hub driving tumor cell migration and invasion by enabling changes in cell characteristics.During EMT,epithelial cells gradually take on mesenchymal traits,gaining mobility and spreading mo re easily.Recent multi-omics studies show that many cancer cells exist in a hybrid or partial-EMT state,which lies between the full epithelial and mesenchymal forms.Cells in this state are especially invasive and metastatic,with high plasticity that promotes tumor progression.This review summarizes the role of partial-EMT in OSCC,with a focus on how it alters the tumor microenvironment(TME),promotes invasion and metastasis,and influences cancer stem cells(CSCs).We also highlight the link between partial-EMT and treatment resistance in OSCC.Based on these insights,we discuss therapeutic strategies targeting partial-EMT to improve outcomes.Targeting partial-EMT may offer promising strategies to enhance treatment effectiveness and improve patient survival and quality of life.展开更多
Chitosan(CS),a natural polymer derived from chitin found in the exoskeletons of crustaceans,has garnered significant interest in the pharmaceutical field due to its unique properties,including biocompatibility and bio...Chitosan(CS),a natural polymer derived from chitin found in the exoskeletons of crustaceans,has garnered significant interest in the pharmaceutical field due to its unique properties,including biocompatibility and biodegradability.In recent years,various studies have reported that CS can affect drug bioavailability,and interestingly,it works as an oral absorption enhancer and inhibitor.This review offers an in-depth analysis of the mechanisms underlying such a phenomenon and supports its application as a pharmaceutical excipient.CS enhances oral drug absorption through various mechanisms,such as interaction with the intestinal mucosa,tight junction modulation,inhibition of efflux transporters,enzyme inhibition,solubility and stability enhancement,and complexation.On the other side,CS exhibits the ability to inhibit the absorption of certain drugs by adsorbing to lipids and sterols,modulating bile acids and gut microbiota,altering drug-cell interaction at the polar interface,and mucus-mediated entrapment and interference.Future potential pharmaceutical research in this field includes elucidating the underneath absorption relevant mechanisms,rational use in formulations as excipient,exploring functional CS derivatives,and developing CS-based drug delivery systems.This comprehensive review highlights CS's versatile and significant role in enhancing and inhibiting oral drug absorption,providing insights into the complexities of drug delivery and the potential of CS to improve therapeutic outcomes.展开更多
The objective of this study was to understand the effect of long-term aconitine(AC)oral administration on the digestive tract and serum metabolism.Subjects consumed either 0.9%Na Cl(n=8)or AC(n=17)gavage designed to r...The objective of this study was to understand the effect of long-term aconitine(AC)oral administration on the digestive tract and serum metabolism.Subjects consumed either 0.9%Na Cl(n=8)or AC(n=17)gavage designed to represent human chronic AC administrations for 13 days.Organ pathology was determined using hematoxylin-eosin staining and immunohistochemistry.Fecal and proximal intestinal content samples were collected to perform shotgun metagenomic sequencing.Serum samples were collected,and untargeted metabolomics was performed.In this study,AC administration induced proximal intestine,liver,and kidney injury.Microbiome composition remained stable after AC exposure,while several microbes presented dynamic alteration.Moreover,AC affected the abundance of the fatty acid biosynthesis rate-limiting gene acc A at day 7.AC induces 30 serum metabolites to significantly change at day 14,including several short-chain acylcarnitines.WGCNA revealed 2 sub-modules associated with the level of several short-chain acylcarnitines.In summary,AC affects the digestive tract and serum metabolism after chronic administration.AC may affect the enrichment of microbial-derived acc A gene.The abundance of serum acylcarnitines detected in the AC group may associate with its anti-heart failure effects.展开更多

The changes and gastric digestive properties of gel were investigated for oxidiz ed myofibrillar protein(MP)(0–1 mmol/L H_(2)O_(2))treated by 30%substitution of NaCl with KCl,MgCl_(2),and KCl/MgCl_(2).Mild oxidation ...The changes and gastric digestive properties of gel were investigated for oxidiz ed myofibrillar protein(MP)(0–1 mmol/L H_(2)O_(2))treated by 30%substitution of NaCl with KCl,MgCl_(2),and KCl/MgCl_(2).Mild oxidation level(0.3 mmol/L H_(2)O_(2))facilitated the protein unfolding,and MgCl_(2)obviously intervened the protein selfassembly by forming the salt bridge and more disulfide bonds.At the same oxidation concentration,the sodium-reduced MP substituted with KCl formed dense network structure with better gel strength(increased by 6%–18%)and water retention in comparision to the gel without sodium reduction.The KCl-substituted gel chewing work was also relatively higher among the low-sodium samples at the first chewing cycle.Reversely,the relatively soft structure and lower gel viscosity in simulated oral chewing were found in MgCl_(2)-substituted gel.These changes decreased the contact limit between pepsin and protein for better gastric disintegration or pepsin diffusivity,and increased the proportion of small molecular weight peptides(25.83%,<5 kDa).Therefore,partial substitution of NaCl with other salt replacers could influence the physicochemical properties of low-sodium MP suspension or gel under different oxidation degree and the subsequent gastric digestion characteristics.展开更多
Oral immunization is an alternative or supplementary approach that can significantly improve dog vaccination coverage,especially for free-roaming dogs.Safe and effective oral rabies vaccines for dogs are still being s...Oral immunization is an alternative or supplementary approach that can significantly improve dog vaccination coverage,especially for free-roaming dogs.Safe and effective oral rabies vaccines for dogs are still being sought.In our previous studies,we generated a genetically modified rabies virus(RABV) ERA strain,rERAG_(333E),containing a mutation from arginine(Arg,R) to glutamic acid(Glu,E) at residue 333 of the G protein(G_(333E)).Our previous results demonstrated that rERAG_(333E) was safe for adult mice and dogs,and oral vaccination with rERAG_(333E) induced a strong and long-lasting protective immune response in dogs.Here,we further investigated the safety and immunogenicity of rERAG_(333E) in nontarget species,including suckling mice,rhesus monkeys,foxes,raccoon dogs,piglets,goats,and sheep.Suckling mice studies demonstrated that the G_(333E) mutation significantly reduced the virulence of the ERA strain.All of the suckling mice aged 10 days and above survived and showed no apparent signs of disease after intracerebral inoculation with rERAG_(333E).Animal studies demonstrated that rERAG_(333E) was safe in rhesus monkeys,foxes,raccoon dogs,piglets,goats,and sheep.None of those animals inoculated orally with 10 times the intended field dose of rERAG_(333E) showed abnormal clinical signs before and after the booster immunization with Rabvac 3,an inactivated rabies vaccine.Meanwhile,oral inoculation with rERAG_(333E) induced strong neutralizing antibody(NA) responses to RABV in rhesus monkeys,foxes,raccoon dogs,and piglets.These results demonstrated that rERAG_(333E) has the potential to serve as a safe oral rabies vaccine for dogs.展开更多
This narrative review examines recent advances in salivary biomarkers for oral squamous cell carcinoma(OSCC),a major subtype of oral cancer with persistently low five-year survival rates due to delayed diagnosis.Saliv...This narrative review examines recent advances in salivary biomarkers for oral squamous cell carcinoma(OSCC),a major subtype of oral cancer with persistently low five-year survival rates due to delayed diagnosis.Saliva has emerged as a noninvasive diagnostic medium capable of reflecting both local tumor activity and systemic physiological changes.Various salivary biomarkers,including microRNAs,cytokines,proteins,metabolites,and exosomes,have been linked to oncogenic signaling pathways involved in tumor progression,immune modulation,and therapeutic resistance.Advances in quantitative polymerase chain reaction,mass spectrometry,and next-generation sequencing have enabled comprehensive biomarker profiling,while point-of-care detection systems and saliva-based omics platforms are accelerating clinical translation.Remaining challenges include variability in salivary composition,lack of standardized collection protocols,and insufficient validation across large patient cohorts.This review highlights the mechanistic relevance,diagnostic potential,and translational challenges of salivary biomarkers in OSCC.展开更多

Background:The regulatory mechanisms governing vasculogenic mimicry(VM)in oral squamous cell carcinoma(OSCC)remain largely undefined.This study aimed to identify critical factors and elucidate the epigenetic mechanism...Background:The regulatory mechanisms governing vasculogenic mimicry(VM)in oral squamous cell carcinoma(OSCC)remain largely undefined.This study aimed to identify critical factors and elucidate the epigenetic mechanisms underlying VM in OSCC.Methods:Bioinformatics analysis was performed utilizing single-cell RNA-seq,bulk RNA-seq,and histone H3 lysine 27 acetylation(H3K27ac)Chromatin Immunoprecipitation(ChIP)-seq data obtained from The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)databases.ChIP-qPCR was used to validate the binding of ETS transcription factor ELK4(ELK4)to the dihydrofolate reductase(DHFR)enhancer.In vitro VM formation and invasion of OSCC cells were assessed using Matrigel-based tube formation and Transwell assays,respectively.Results:Elevated expression of VM-related genes predicts unfavorable prognosis in OSCC patients.High-dimensional weighted gene co-expression network analysis(hdWGCNA)identified epithelial subcluster C4 as most strongly associated with VM and metastasis.Three co-expression modules within this subcluster exhibited significant positive correlations with both phenotypic traits.Among the 30 eigengenes from the three modules,DHFR emerged as a key regulator of VM and metastasis.Knockdown or inhibition of DHFR significantly suppressed VM formation and invasion in OSCC cells.Mechanistically,ELK4 activated DHFR transcription through direct binding to its enhancer.DHFR overexpression rescued VM and invasion impairment induced by ELK4 knockdown.Conclusion:DHFR was a pivotal enhancer-regulated gene driving VM and metastasis in OSCC.ELK4 directly binds to DHFR enhancer regions to activate its transcription,thereby promoting these malignant phenotypes.These findings identified the ELK4/DHFR axis as a promising therapeutic target for anti-angiogenic intervention in OSCC.展开更多
Objective:To explore the application effect of digital intraoral scanning impression technique in oral implant restoration for periodontitis patients and analyze its impact on patients’Visual Analogue Scale(VAS)score...Objective:To explore the application effect of digital intraoral scanning impression technique in oral implant restoration for periodontitis patients and analyze its impact on patients’Visual Analogue Scale(VAS)scores.Methods:A total of 80 periodontitis patients who received implant restoration in our hospital from May 2023 to May 2025 were selected as research subjects.They were randomly divided into an observation group and a control group using a random number table method,with 40 cases in each group.The observation group used the digital intraoral scanning impression technique to obtain impressions,while the control group used the traditional silicone rubber impression technique.The impression-taking time,the number of prostheses try-ins,implant survival rate,periodontal health indicators(probing depth,gingival index,bleeding index),and VAS scores(pain during treatment and comfort after restoration)were compared between the two groups.Results:The observation group was superior to the control group in terms of impression-taking time,the number of prostheses try-ins,and implant survival rate(p<0.05).Six months after restoration,the improvement in periodontal health indicators in the observation group was significantly better than that in the control group(p<0.05).In addition,the pain VAS score of the observation group during treatment was lower than that of the control group,and the comfort VAS score after restoration was higher than that of the control group(p<0.05).Conclusion:Digital intraoral scanning impression technology can effectively enhance the efficiency and success rate of implant restoration in periodontitis patients,improve periodontal health,alleviate patients’discomfort during treatment,and increase post-restoration comfort,demonstrating high clinical application value.展开更多
Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to enviro...Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to environmental changes upon entry into oral biofilms are reviewed.Supra-gingival biofilms are characterized by acidic pH,the presence of bacterial enzymes,and the development of hypoxia in deeper layers.Sub-gingival biofilms are slightly alkaline,with hypoxia occurring over their entire depth.Upon entering biofilms,negatively charged,pH-and/or hypoxia-responsive nanocarriers become positively charged.This charge reversal leads to electrostatic double-layer attraction between positively charged nanocarriers towards negatively charged,waterfilled channel walls in biofilms,enhancing their accumulation in a biofilm.Degradation of bacterial enzyme-responsive nanocarriers causes in-biofilm release of antimicrobial cargo,yielding higher local antimicrobial concentrations than can be achieved through their direct,oral administration without harming soft tissues.Enhanced antibiofilm activity after in-biofilm antimicrobial release from biofilm-responsive micelles and liposomes has been demonstrated in vitro towards single-species Streptococcus mutans and Staphylococcus aureus biofilms or in vivo using specific-pathogen-free rodents inoculated with selected pathogens.This preferential antibacterial activity regulated the microbial composition of ex vivo human oral biofilm towards a more healthy microbiome composition.Although clinical confirmation is limited,the potential benefits of stimuli-responsive,antimicrobial-loaded nanocarriers for oral biofilm control and microbiome restoration are worth further investigation towards clinical translation.展开更多
Osmanthus fragrans is most famous for its strong aroma,and different varieties have different degrees of fragrance and color.Fragrance and color are important factors affecting the ornamental quality of O.fragrans.Ter...Osmanthus fragrans is most famous for its strong aroma,and different varieties have different degrees of fragrance and color.Fragrance and color are important factors affecting the ornamental quality of O.fragrans.Terpenoids are important secondary metabolites in plants,with β-carotene(C40)being the major pigment substance and linalool(C10)being the key aromatic component in O.fragrans.The geranylgeranyl pyrophosphate synthase genes(GGPPSs)play important roles in secondary metabolism in plants.However,the functions of the GGPPS family in floral color and fragrance formation has rarely been reported in O.fragrans.In this study,24 Of GGPPS genes were identified and classified into two subfamilies.The Of GGPPSs showed tissue-specific expression and Of GGPPS13 had highest expression in flowers.The Of GGPPS13 protein was localized to chloroplasts.The transcriptome data of Of GGPPS13 was verified by q RT-PCR and the expression level in‘Wanyingui'with strong aroma was higher than that in‘Zhuangyuanhong'with deep color at different flower development stages.Transient overexpression of Of GGPPS13 in O.fragrans petals showed that Of GGPPS13 increased the β-carotene content,the main color substance of O.fragrans,but decreased the linalool content,the main volatile organic compound(VOC)in the floral aroma of O.fragrans.Of GGPPS13 was indicated as the critical gene related to terpenoid synthesis in the floral aroma and color formation in O.fragrans.Our findings provide gene resources on the GGPPS gene family for further revealing the molecular regulation mechanism of the floral color and aroma formation in O.fragrans.展开更多

文摘Background Recent studies have suggested a potential role of the oral microbiome in the development of cardiovascular diseases.This study aims to investigate the association between oral microbiota and cardiovascular disease risk,including atrial fibrillation,myocardial infarction,chronic heart failure,and hypertension.Methods We analyzed GWAS data from East Asian populations'oral microbiome,involving 2,017 tongue and 1,915 saliva samples from 2,984 individuals with whole-genome sequencing.Additionally,we sourced cardiovascular disease GWAS data from NBDC,including atrial fibrillation(8,180 cases,28,621 controls),myocardial infarction(14,992 cases,146,214 controls),chronic heart failure(10,540 cases,168,186 controls),and systolic blood pressure(145,505 individuals).Results Several oral microbiota taxa were found to be significantly associated with cardiovascular disease outcomes.Specific microbiota,such as Centipeda,Corynebacterium,and Pseudomonas E,were negatively correlated with heart failure.In contrast,taxa like Neisseria D and Actinomyces were associated with an increased risk of atrial fibrillation and myocardial infarction.Additionally,certain oral microbiota showed correlations with changes in blood pressure,highlighting their potential role in hypertension.Conclusion Our findings suggest that the oral microbiota may influence the development and progression of cardiovascular diseases,providing new insights into the potential impact of oral health on cardiovascular risk.

文摘Wearable sensors have revolutionized health monitoring by transitioning from clinical diagnostics to continuous,real-time applications in daily life.The oral cavity,rich in saliva containing over 1,000 biomarkers that reflect systemic health(e.g.,glucose,cortisol,and inflammatory markers)[1],offers the advantage of non-invasive sampling.Its superior environmental stability and strong connection to key physiological processes make it an ideal candidate in the field of digital medicine,serving as a natural gateway to personalized health monitoring.Therefore,the oral cavity represents not only a convenient sampling site but also a strategic interface for realizing the vision of continuous,personalized digital health monitoring.

文摘Oral squamous cell carcinoma(OSCC)is a prevalent malignancy with high morbidity and mortality.Globally,about 400000 people are affected,often with a poor quality of life.Its high mortality is mainly due to its aggressive growth and tendency to spread.Epithelial-mesenchymal transition(EMT)is a central regulatory hub driving tumor cell migration and invasion by enabling changes in cell characteristics.During EMT,epithelial cells gradually take on mesenchymal traits,gaining mobility and spreading mo re easily.Recent multi-omics studies show that many cancer cells exist in a hybrid or partial-EMT state,which lies between the full epithelial and mesenchymal forms.Cells in this state are especially invasive and metastatic,with high plasticity that promotes tumor progression.This review summarizes the role of partial-EMT in OSCC,with a focus on how it alters the tumor microenvironment(TME),promotes invasion and metastasis,and influences cancer stem cells(CSCs).We also highlight the link between partial-EMT and treatment resistance in OSCC.Based on these insights,we discuss therapeutic strategies targeting partial-EMT to improve outcomes.Targeting partial-EMT may offer promising strategies to enhance treatment effectiveness and improve patient survival and quality of life.

文摘Chitosan(CS),a natural polymer derived from chitin found in the exoskeletons of crustaceans,has garnered significant interest in the pharmaceutical field due to its unique properties,including biocompatibility and biodegradability.In recent years,various studies have reported that CS can affect drug bioavailability,and interestingly,it works as an oral absorption enhancer and inhibitor.This review offers an in-depth analysis of the mechanisms underlying such a phenomenon and supports its application as a pharmaceutical excipient.CS enhances oral drug absorption through various mechanisms,such as interaction with the intestinal mucosa,tight junction modulation,inhibition of efflux transporters,enzyme inhibition,solubility and stability enhancement,and complexation.On the other side,CS exhibits the ability to inhibit the absorption of certain drugs by adsorbing to lipids and sterols,modulating bile acids and gut microbiota,altering drug-cell interaction at the polar interface,and mucus-mediated entrapment and interference.Future potential pharmaceutical research in this field includes elucidating the underneath absorption relevant mechanisms,rational use in formulations as excipient,exploring functional CS derivatives,and developing CS-based drug delivery systems.This comprehensive review highlights CS's versatile and significant role in enhancing and inhibiting oral drug absorption,providing insights into the complexities of drug delivery and the potential of CS to improve therapeutic outcomes.

文摘The objective of this study was to understand the effect of long-term aconitine(AC)oral administration on the digestive tract and serum metabolism.Subjects consumed either 0.9%Na Cl(n=8)or AC(n=17)gavage designed to represent human chronic AC administrations for 13 days.Organ pathology was determined using hematoxylin-eosin staining and immunohistochemistry.Fecal and proximal intestinal content samples were collected to perform shotgun metagenomic sequencing.Serum samples were collected,and untargeted metabolomics was performed.In this study,AC administration induced proximal intestine,liver,and kidney injury.Microbiome composition remained stable after AC exposure,while several microbes presented dynamic alteration.Moreover,AC affected the abundance of the fatty acid biosynthesis rate-limiting gene acc A at day 7.AC induces 30 serum metabolites to significantly change at day 14,including several short-chain acylcarnitines.WGCNA revealed 2 sub-modules associated with the level of several short-chain acylcarnitines.In summary,AC affects the digestive tract and serum metabolism after chronic administration.AC may affect the enrichment of microbial-derived acc A gene.The abundance of serum acylcarnitines detected in the AC group may associate with its anti-heart failure effects.
文摘After the“Ballad Movement”in the early 20th century revealed and systematized the Liang Zhu legend,numerous literary works had created a rich body of classic reinterpretations.This paper argues that Yang Yinshen’s A Storm stands as the earliest modern literary creation that actively embraced the folk Liang Zhu legend.By situating the ancient story of Zhu Yingtai in a contemporary context,the play replaces the traditional core values of“loyalty and righteousness”with modern themes of pursuing free love,amplifying women’s autonomy.Through a modern tragic love story,it achieves a classic reinterpretation of the classical folk legend.The“revised narrative”in A Storm primarily involves derivative adaptations,incorporating contemporary revisions to characters and themes.Its three-act structure also forms the foundational framework for subsequent reinterpretations of the Liang Zhu legend.The play demonstrates a conscious absorption of the rebellious spirit in folk literature,incorporates dialectal speech,and provides valuable insights into how modern literature can utilize oral traditions.

文摘The changes and gastric digestive properties of gel were investigated for oxidiz ed myofibrillar protein(MP)(0–1 mmol/L H_(2)O_(2))treated by 30%substitution of NaCl with KCl,MgCl_(2),and KCl/MgCl_(2).Mild oxidation level(0.3 mmol/L H_(2)O_(2))facilitated the protein unfolding,and MgCl_(2)obviously intervened the protein selfassembly by forming the salt bridge and more disulfide bonds.At the same oxidation concentration,the sodium-reduced MP substituted with KCl formed dense network structure with better gel strength(increased by 6%–18%)and water retention in comparision to the gel without sodium reduction.The KCl-substituted gel chewing work was also relatively higher among the low-sodium samples at the first chewing cycle.Reversely,the relatively soft structure and lower gel viscosity in simulated oral chewing were found in MgCl_(2)-substituted gel.These changes decreased the contact limit between pepsin and protein for better gastric disintegration or pepsin diffusivity,and increased the proportion of small molecular weight peptides(25.83%,<5 kDa).Therefore,partial substitution of NaCl with other salt replacers could influence the physicochemical properties of low-sodium MP suspension or gel under different oxidation degree and the subsequent gastric digestion characteristics.

文摘Oral immunization is an alternative or supplementary approach that can significantly improve dog vaccination coverage,especially for free-roaming dogs.Safe and effective oral rabies vaccines for dogs are still being sought.In our previous studies,we generated a genetically modified rabies virus(RABV) ERA strain,rERAG_(333E),containing a mutation from arginine(Arg,R) to glutamic acid(Glu,E) at residue 333 of the G protein(G_(333E)).Our previous results demonstrated that rERAG_(333E) was safe for adult mice and dogs,and oral vaccination with rERAG_(333E) induced a strong and long-lasting protective immune response in dogs.Here,we further investigated the safety and immunogenicity of rERAG_(333E) in nontarget species,including suckling mice,rhesus monkeys,foxes,raccoon dogs,piglets,goats,and sheep.Suckling mice studies demonstrated that the G_(333E) mutation significantly reduced the virulence of the ERA strain.All of the suckling mice aged 10 days and above survived and showed no apparent signs of disease after intracerebral inoculation with rERAG_(333E).Animal studies demonstrated that rERAG_(333E) was safe in rhesus monkeys,foxes,raccoon dogs,piglets,goats,and sheep.None of those animals inoculated orally with 10 times the intended field dose of rERAG_(333E) showed abnormal clinical signs before and after the booster immunization with Rabvac 3,an inactivated rabies vaccine.Meanwhile,oral inoculation with rERAG_(333E) induced strong neutralizing antibody(NA) responses to RABV in rhesus monkeys,foxes,raccoon dogs,and piglets.These results demonstrated that rERAG_(333E) has the potential to serve as a safe oral rabies vaccine for dogs.

文摘This narrative review examines recent advances in salivary biomarkers for oral squamous cell carcinoma(OSCC),a major subtype of oral cancer with persistently low five-year survival rates due to delayed diagnosis.Saliva has emerged as a noninvasive diagnostic medium capable of reflecting both local tumor activity and systemic physiological changes.Various salivary biomarkers,including microRNAs,cytokines,proteins,metabolites,and exosomes,have been linked to oncogenic signaling pathways involved in tumor progression,immune modulation,and therapeutic resistance.Advances in quantitative polymerase chain reaction,mass spectrometry,and next-generation sequencing have enabled comprehensive biomarker profiling,while point-of-care detection systems and saliva-based omics platforms are accelerating clinical translation.Remaining challenges include variability in salivary composition,lack of standardized collection protocols,and insufficient validation across large patient cohorts.This review highlights the mechanistic relevance,diagnostic potential,and translational challenges of salivary biomarkers in OSCC.
文摘The post-hoc analysis of the PERSEO registry by Minardi,et al.[1]offers critical insights into antithrombotic management for elderly patients(≥80 years)on oral anticoagulation undergoing percutaneous coronary intervention.While the authors effectively demonstrate the vulnerability of this population,characterized by substantial comorbidity burden and markedly elevated rates of mortality,ischemic events,and bleeding at one year compared with younger cohorts,their findings raise important questions about contemporary practice patterns and their alignment with evidence-based guidelines.
文摘Objective:To systematically sort out the application forms and effects of digital health intervention technologies in oral health management,and provide references for the digital development of stomatology.Methods:By reviewing relevant domestic and foreign studies and clinical practices,this paper summarizes and analyzes the main application forms of digital health interventions,including digital health education,intelligent detection equipment,telemedicine platforms,oral health big data platforms,and school-hospital collaborative screening robots.Results:Studies have shown that digital health interventions can effectively improve the public’s oral health knowledge level,optimize personal health behaviors,enhance clinical diagnosis efficiency,reduce overall medical costs,and promote the innovation and upgrading of oral health management models.Conclusion:Digital health intervention represents an inevitable trend in the future development of stomatology.In the future,it is still necessary to improve data security and privacy protection,technology adaptability and popularity,as well as relevant policies and norms,to give full play to its potential value.

文摘Background:The regulatory mechanisms governing vasculogenic mimicry(VM)in oral squamous cell carcinoma(OSCC)remain largely undefined.This study aimed to identify critical factors and elucidate the epigenetic mechanisms underlying VM in OSCC.Methods:Bioinformatics analysis was performed utilizing single-cell RNA-seq,bulk RNA-seq,and histone H3 lysine 27 acetylation(H3K27ac)Chromatin Immunoprecipitation(ChIP)-seq data obtained from The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)databases.ChIP-qPCR was used to validate the binding of ETS transcription factor ELK4(ELK4)to the dihydrofolate reductase(DHFR)enhancer.In vitro VM formation and invasion of OSCC cells were assessed using Matrigel-based tube formation and Transwell assays,respectively.Results:Elevated expression of VM-related genes predicts unfavorable prognosis in OSCC patients.High-dimensional weighted gene co-expression network analysis(hdWGCNA)identified epithelial subcluster C4 as most strongly associated with VM and metastasis.Three co-expression modules within this subcluster exhibited significant positive correlations with both phenotypic traits.Among the 30 eigengenes from the three modules,DHFR emerged as a key regulator of VM and metastasis.Knockdown or inhibition of DHFR significantly suppressed VM formation and invasion in OSCC cells.Mechanistically,ELK4 activated DHFR transcription through direct binding to its enhancer.DHFR overexpression rescued VM and invasion impairment induced by ELK4 knockdown.Conclusion:DHFR was a pivotal enhancer-regulated gene driving VM and metastasis in OSCC.ELK4 directly binds to DHFR enhancer regions to activate its transcription,thereby promoting these malignant phenotypes.These findings identified the ELK4/DHFR axis as a promising therapeutic target for anti-angiogenic intervention in OSCC.
文摘Objective:To explore the application effect of digital intraoral scanning impression technique in oral implant restoration for periodontitis patients and analyze its impact on patients’Visual Analogue Scale(VAS)scores.Methods:A total of 80 periodontitis patients who received implant restoration in our hospital from May 2023 to May 2025 were selected as research subjects.They were randomly divided into an observation group and a control group using a random number table method,with 40 cases in each group.The observation group used the digital intraoral scanning impression technique to obtain impressions,while the control group used the traditional silicone rubber impression technique.The impression-taking time,the number of prostheses try-ins,implant survival rate,periodontal health indicators(probing depth,gingival index,bleeding index),and VAS scores(pain during treatment and comfort after restoration)were compared between the two groups.Results:The observation group was superior to the control group in terms of impression-taking time,the number of prostheses try-ins,and implant survival rate(p<0.05).Six months after restoration,the improvement in periodontal health indicators in the observation group was significantly better than that in the control group(p<0.05).In addition,the pain VAS score of the observation group during treatment was lower than that of the control group,and the comfort VAS score after restoration was higher than that of the control group(p<0.05).Conclusion:Digital intraoral scanning impression technology can effectively enhance the efficiency and success rate of implant restoration in periodontitis patients,improve periodontal health,alleviate patients’discomfort during treatment,and increase post-restoration comfort,demonstrating high clinical application value.

文摘Nanotechnology has provided thousands of novel nano-antimicrobials possessing features uncommon in clinically available antimicrobials.Here,nanocarriers loaded with conventional antimicrobials and responding to environmental changes upon entry into oral biofilms are reviewed.Supra-gingival biofilms are characterized by acidic pH,the presence of bacterial enzymes,and the development of hypoxia in deeper layers.Sub-gingival biofilms are slightly alkaline,with hypoxia occurring over their entire depth.Upon entering biofilms,negatively charged,pH-and/or hypoxia-responsive nanocarriers become positively charged.This charge reversal leads to electrostatic double-layer attraction between positively charged nanocarriers towards negatively charged,waterfilled channel walls in biofilms,enhancing their accumulation in a biofilm.Degradation of bacterial enzyme-responsive nanocarriers causes in-biofilm release of antimicrobial cargo,yielding higher local antimicrobial concentrations than can be achieved through their direct,oral administration without harming soft tissues.Enhanced antibiofilm activity after in-biofilm antimicrobial release from biofilm-responsive micelles and liposomes has been demonstrated in vitro towards single-species Streptococcus mutans and Staphylococcus aureus biofilms or in vivo using specific-pathogen-free rodents inoculated with selected pathogens.This preferential antibacterial activity regulated the microbial composition of ex vivo human oral biofilm towards a more healthy microbiome composition.Although clinical confirmation is limited,the potential benefits of stimuli-responsive,antimicrobial-loaded nanocarriers for oral biofilm control and microbiome restoration are worth further investigation towards clinical translation.

文摘Osmanthus fragrans is most famous for its strong aroma,and different varieties have different degrees of fragrance and color.Fragrance and color are important factors affecting the ornamental quality of O.fragrans.Terpenoids are important secondary metabolites in plants,with β-carotene(C40)being the major pigment substance and linalool(C10)being the key aromatic component in O.fragrans.The geranylgeranyl pyrophosphate synthase genes(GGPPSs)play important roles in secondary metabolism in plants.However,the functions of the GGPPS family in floral color and fragrance formation has rarely been reported in O.fragrans.In this study,24 Of GGPPS genes were identified and classified into two subfamilies.The Of GGPPSs showed tissue-specific expression and Of GGPPS13 had highest expression in flowers.The Of GGPPS13 protein was localized to chloroplasts.The transcriptome data of Of GGPPS13 was verified by q RT-PCR and the expression level in‘Wanyingui'with strong aroma was higher than that in‘Zhuangyuanhong'with deep color at different flower development stages.Transient overexpression of Of GGPPS13 in O.fragrans petals showed that Of GGPPS13 increased the β-carotene content,the main color substance of O.fragrans,but decreased the linalool content,the main volatile organic compound(VOC)in the floral aroma of O.fragrans.Of GGPPS13 was indicated as the critical gene related to terpenoid synthesis in the floral aroma and color formation in O.fragrans.Our findings provide gene resources on the GGPPS gene family for further revealing the molecular regulation mechanism of the floral color and aroma formation in O.fragrans.

文摘The oral cavity is a complex physiological community encompassing a wide range of microorganisms.Dysbiosis of oral microbiota can lead to various oral infectious diseases,such as periodontitis and tooth decay,and even affect systemic health,including brain aging and neurodegenerative diseases.Recent studies have highlighted how oral microbes might be involved in brain aging and neurodegeneration,indicating potential avenues for intervention strategies.In this review,we summarize clinical evidence demonstrating a link between oral microbes/oral infectious diseases and brain aging/neurodegenerative diseases,and dissect potential mechanisms by which oral microbes contribute to brain aging and neurodegeneration.We also highlight advances in therapeutic development grounded in the realm of oral microbes,with the goal of advancing brain health and promoting healthy aging.
