Betaine combined with traditional Chinese medicine ointment to treat skin wounds in microbially infected diabetic mice

BACKGROUND Skin wounds are highly common in diabetic patients,and with increasing types of pathogenic bacteria and antibiotic resistance,wounds and infections in diabetic patients are difficult to treat and heal.AIM To explore the effects of betaine ointment(BO)in promoting the healing of skin wounds and reducing the inflammation and apoptosis of skin cells in microbially infected diabetic mice.METHODS By detecting the minimum inhibitory concentrations(MICs)of betaine and plant monomer components such as psoralen,we prepared BO with betaine as the main ingredient,blended it with traditional Chinese medicines such as gromwell root and psoralen,and evaluated its antibacterial effects and safety in vitro and in vivo.The skin infection wound models of ordinary mice and diabetic mice were constructed,and the OTC drugs mupirocin ointment and Zicao ointment were used as controls to evaluate the antibacterial effects in vivo and the anti-inflammatory and anti-apoptotic effects of BO.RESULTS The MICs of betaine against microorganisms such as Staphylococcus aureus(S.aureus),Candida albicans and Cryptococcus neoformans ranged from 4 to 32μg/mL.Gromwell root and psoralea,both of which contain antimicrobial components,mixed to prepare BO with MICs ranging from 16 to 64μg/mL,which is 32-256 times lower than those of Zicao ointment,although the MIC is greater than that of betaine.After 15 days of treatment with BO for USA300-infected ordinary mice,the wound scab removal rates were 83.3%,while those of mupirocin ointment and Zicao ointment were 66.7%and 0%,respectively,and the differences were statistically significant.In diabetic mice,the wound scab removal rate of BO and mupirolacin ointment was 80.0%,but BO reduced wound inflammation and the apoptosis of skin cells and facilitated wound healing.CONCLUSION The ointment prepared by mixing betaine and traditional Chinese medicine can effectively inhibit common skin microorganisms and has a strong effect on the skin wounds of sensitive or drug-resistant S.aureus-infected ordinary mice and diabetic mice. 展开更多

[Objectives]To observe the clinical efficacy of external application of Scorzonera Herpes Ointment combined with Methylprednisolone Sodium Succinate for Injection in the treatment of herpes zoster oticus(HZO).[Methods]A total of 100 HZO patients admitted to the 988 th Hospital of the Joint Logistics Support Force and Henan Provincial People's Hospital from June 2021 to June 2023 were selected.They were divided into a treatment group and a control group using a random number table method,with 50 cases in each group.Both groups received Methylprednisolone Sodium Succinate for Injection.Additionally,the treatment group was treated with external application of Scorzonera Herpes Ointment,while the control group received acyclovir ointment.Both groups were treated for 10 d.The comparisons included clinical efficacy,total symptom and sign scores,pain level[Visual Analogue Scale(VAS)],time for erythema reduction,cessation of blister formation,scab formation,and scab shedding,incidence of post-herpetic neuralgia(PHN),air conduction hearing threshold,and air-bone gap.[Results]After 10 d of treatment,the total effective rate was 98.00%(49/50)in the treatment group and 84.00%(42/50)in the control group,with a statistically significant difference between the two groups(P<0.05).After 10 d of treatment,the total symptom and sign scores and VAS scores of both groups decreased compared to those before treatment.The treatment group had significantly lower scores than the control group(P<0.05).The treatment group showed significantly shorter time for erythema reduction,cessation of blister formation,scab formation,and scab shedding compared to the control group(P<0.05).During the 1-month follow-up after treatment,no PHN cases occurred in the treatment group,while the incidence of PHN in the control group was 24.00%(12/50),showing a statistically significant difference(P<0.05).After 10 d of treatment,both groups showed reduced air conduction hearing thresholds,and the treatment group exhibited significantly lower air conduction thresholds and air-bone gaps compared to the control group(P<0.05).No statistically significant difference was observed in the air-bone gap before and after treatment in the control group(P>0.05).[Conclusions]The combination of external application of Scorzonera Herpes Ointment and Methylprednisolone Sodium Succinate for Injection can alleviate pain and other discomforts,reduce PHN incidence,shorten disease duration,and improve hearing in HZO patients. 展开更多

Objective:A TLCS method was established for the determination of the content of Shenbei Beigua ointment,and the product quality of six samples from two formulations was evaluated.Methods:The determination method was thin-layer chromatography scanning(TLCS),using a developing solvent composed of ethyl acetate–methanol–strong ammonia water(17:2:1).The plates were heated at 105°C for 5 minutes,then sprayed with a mixture of dilute bismuth potassium iodide and 1%ferric chloride in ethanol(10:1),and scanned at a wavelength of 500 nm.Results:Peimine showed good linearity in the concentration range of 0.21–2.1μg with a correlation coefficient of r=0.9997,and Peiminine also exhibited good linearity in the same range with r=0.9995.The accuracy was≥95.0%,and the relative standard deviation(RSD)was≤5.0%(n=6).Conclusion:This method allows for the simultaneous determination of peimine and peiminine,providing a reliable reference for the quality control of the product. 展开更多

BACKGROUND Postpartum hemorrhoids are common complications in women after childbirth,with an incidence rate of 37.9%.Progesterone levels increase during pregnancy,which can lead to constipation.Abdominal pressure surges sharply during delivery,which can block the venous return of the rectum and result in hemorrhoids,pain,and edema.Tongyangxiao lotion and Ziyu ointment,traditional Chinese medicinal preparations,have the efficacy of subduing swelling,relieving itching,and promoting healing;however,their combined application in treating postpartum hemorrhoids is less studied.AIM To investigate effects of Tongyangxiao lotion sitz bath therapy combined with Ziyu ointment on postpartum hemorrhoidal pain,edema,anxiety,and depression.METHODS A retrospective analysis was conducted at Hebei Hospital of Traditional Chinese Medicine on 268 patients with hemorrhoids(September 2023 to February 2025).They were divided according to treatment method into control(wet compress with magnesium sulfate+Ziyu ointment)and observation(Tongyangxiao lotion sitz bath therapy+Ziyu ointment)groups(n=134 in each group).Degree of pain,comfort level,compliance behavior with medical care,psychological state,therapeutic effect,hemorrhoid edema score,incidence of adverse reactions,and hemorrhoid diameter were compared.RESULTS No statistically significant difference was observed in the diameter of the hemorrhoids or edema between the two groups before treatment.Hemorrhoid diameter and degree of edema were significantly smaller in the observation group than those in the control group 14 days post treatment.No statistically significant difference was observed in the scores of the General Comfort Questionnaire(GCQ)dimensions and participants’compliance before treatment.All GCQ dimension scores were significantly higher in the observation group than those in the control group 7 and 14 days post treatment.The compliance rate was significantly higher in the observation group than that of the control group.No statistically significant difference was observed in the incidence of adverse reactions between the two groups(P>0.05).CONCLUSION Tongyangxiao lotion combined with Ziyu ointment can improve patients’surface edema,postpartum hemorrhoid pain,treatment adherence,and comfort,reducing negative emotions.This is relatively safe and worthy of clinical promotion. 展开更多

Intraocular ointment is conventionally placed on the eye to prevent infection after cataract surgery.The purpose of this study is to report a case and conduct a systematic review of a rare occurrence of the entry of intraocular ointment after cataract surgery.PubMed,Scopus,Embase,CNKI,WANFANG data,China Science and Technology Journal Database and Chinese Medical Journal Full-text Database were systematically searched from their commencement to 30 th October 2023,and 19 literatures were screened out and 31 cases of intraocular ointment after surgery were collected.Among the 31 patients,the age of presentation ranged from 55 to 87 years with a median of 73,males accounted for 45.2%and females accounted for 32.3%.The length of the incision was generally 3.2 mm.Most of the patients detected ointment within 3 days post-operation and presented without complications(45.2%).The most common ocular manifestations were corneal edema,glaucoma and uveitis.Early postoperative follow-up is very important.Presence of anterior chamber ointment is a rare complication after cataract surgery,but it can lead to severe vision loss if not detected and treated on time.When patients complain of foreign body sensation in the in the eye after cataract surgery,ophthalmologists need to take a kin interest and examine the eye for early detection of ointment for appropriate intervention and prevent further complications. 展开更多

Dingzhou eye ointment gained significant popularity from the late Qing dynasty to the Republican era.Its legacy persists through three prominent families:Bai Jingyu(白敬宇),Ma Yinglong(马应龙)and Zhang Qizhu(张齐珠).Each family’s Dingzhou eye ointment is similar,with comparable ingredients,dosages,and usage methods,suggesting a common origin.While many well-established Dingzhou eye ointment brands claim their origins date back to the late Ming dynasty,the origins of its formulation are likely much older.In ancient Chinese medical texts,there were proto-formulations of similar eye ointments recorded during the Jin and Yuan dynasties,with comparable external and standardized eye medicine formulations appearing in the Ming and Qing dynasties.By the late Qing dynasty,Dingzhou eye ointment had become a popular mainstream topical eye treatment in traditional Chinese medicine.From a broader geographically perspective,the composition aligns with that of similar treatments used in Greco-Roman medicine,which combined minerals such as calamine with aromatic ingredients to create eye medicines.The principles behind the composition of Dingzhou eye ointment are in accord with the concept of the four humours in Arabian medicine.It is possible that the Hui ethnic group was responsible for the introduction of Dingzhou eye ointment to China,indicating cultural exchange between Chinese and foreign medicine practices. 展开更多

Objective:To evaluate the clinical effect of combined therapy with Diosmin Tablets and Mayinglong Musk Hemorrhoids Ointment for patients with acute hemorrhoids.Methods:A total of 50 patients with acute hemorrhoids who visited the hospital from January 2023 to January 2025 were selected as samples and randomly divided into two groups.Group A was treated with Diosmin Tablets combined with Mayinglong Musk Hemorrhoids Ointment,while Group B was treated with Mayinglong Musk Hemorrhoids Ointment only.The efficacy,wound recovery time,perianal pain score,hemorrhoid symptom score,and stress indicators were compared between the two groups.Results:The efficacy of Group A was higher than that of Group B(P<0.05).The postoperative perianal edema time and wound healing time in Group A were shorter than those in Group B,and the Visual Analog Scale(VAS)score was lower than that in Group B(P<0.05).The hemorrhoid symptom score in Group A was lower than that in Group B(P<0.05).The stress level in Group A was lower than that in Group B(P<0.05).Conclusion:The combination therapy of Diosmin Tablets and Mayinglong Musk Hemorrhoids Ointment for postoperative treatment of acute hemorrhoids can effectively relieve perianal pain,shorten the duration of hemorrhoids,and is highly feasible. 展开更多

BACKGROUND Excessive oxidative stress plays a key role in the development of diabetic complications,including impaired ulcer healing.Previous studies have shown that fish scale ointment can promote wound healing.AIM To preliminarily investigate the effect of fish scale ointment on wound healing in a diabetic foot ulcer(DFU)rat model by examining its regulation of the nuclear factor E2-related factor 2(Nrf2)pathway and induction of ferroptosis.METHODS Fish scale ointment(collagen product)was prepared from 500 g of silver carp scales.A diabetic rat model was induced by high-fat and high-sugar feeding combined with intraperitoneal streptozotocin injections.For the DFU rat model,ulcer wounds were created by removing dorsal foot hair and cutting the skin to the fascia.The diabetic rats were randomized into five groups:Model,fish scale collagen(FSC),control+liproxstatin-1(Lip-1),model+Lip-1,and FSC+Lip-1.In each group,treatments were administered once daily by topical application and intraperitoneal injection for 14 days.Wound healing was evaluated on days 7 and 14 after treatment.Hematoxylin and eosin staining was used to assess wound injury and capillary formation.Basic fibroblast growth factor(bFGF)and CD31 levels in wound tissue were measured by immunohistochemistry.Additionally,malondialdehyde(MDA),glutathione(GSH),ferroptosis-associated genes,and iron ion concentrations were quantified using assay kits.Protein levels of Nrf2,heme oxygenase-1(HO-1),and glutathione peroxidase 4(GPX4)were determined using Western blotting.RESULTS Compared with the control group,the model group showed slower wound healing,reduced angiogenesis,decreased bFGF and CD31 levels,increased iron ion concentration and MDA levels,reduced GSH levels,and decreased Nrf2,HO-1,and GPX4 protein expression(all P<0.05).The FSC,model+Lip-1,and FSC+Lip-1 groups showed increased wound healing and angiogenesis,elevated bFGF and CD31 expression,lowered iron ion concentration and MDA levels,increased GSH levels,and enhanced Nrf2,HO-1,and GPX4 protein levels compared with the model group(P<0.05).Improvements were more pronounced in the FSC+Lip-1 group compared with the FSC group(P<0.05).CONCLUSION Fish scale ointment promotes angiogenesis and wound healing in DFU rat models by inhibiting ferroptosis,possibly through the activation of the Nrf2 pathway. 展开更多

[Method] The conventional method and medium dilution method were used for bacterial, mold and yeast counting in sample recovery test. [Result] The medium dilution method (1:10 test solution, 0.5 ml/plate) could effectively eliminate the inhibition effect of the Bupi Qiangli Ointment, and the recovery of Staphylococcus aureus was greater than 70% in the 3 batches of samples; and the conventional method exhibited the recoveries of E. coil, Bacillus subtilis, Candida albicans and Aspergillus greater than 70% in the 3 batches of samples. [Conclusion] Due to Bupi Qiangli Ointment has strongly antibacterial effect on Staphylococcus au- reus, the medium dilution method was used for bacterial counting, and the conventional method was used for mold and yeast counting; and the conventional method was used for controlled bacterium examination of E. coll. 展开更多

Objective:To evaluate the efficacy and safety of sodium fusidate ointment and mupirocin ointment in the treatment of bacteria-infected skin disease.Methods:The search terms“sodium fusidate ointment”,“mupirocin ointment”,“bacterial infection”,etc.were searched in the database of PubMed,EMbase,Cochrane,Web of Science,Wanfang,VIP,and CNKI.The search time was from inception to November 2019.Randomized controlled trials of sodium fusidate ointment and mupirocin ointment for the treatment of bacteria-infected skin disease were collected.Two studies independently performed literature screening,data extraction,and literature quality evaluation.Meta analysis was performed using RevMan 5.3 software.Results:A total of 14 literature were included,involving 1,825 patients,916 in the experimental group,and 909 in the control group.Meta analysis results showed that the total clinical effective rate of the experimental group(RR=1.12,95%CI(1.06,1.19),P<0.0001),degree of improvement in pruritus score(MD=−1.21,95%CI(−1.78,−0.64),P<0.0001),degree of improvement of eczema area and severity index score(MD=−2.47,95%CI(−3.92,−1.02),P=0.0008),and sensitivity rate of drug sensitivity test(RR=1.35,95%CI(1.24,1.46),P<0.00001)were better than those of control group.The incidence of adverse reactions(RR=0.18,95%CI(0.09,0.39),P<0.00001)was significantly smaller than that of control group.But the bacterial clearance rate(RR=1.22,95%CI(0.98,1.52),P=0.08)compared with the control group,the difference was not statistically significant.Conclusion:Sodium fusidate ointment is better than mupirocin ointment in the treatment of bacteria-infected skin disease,and it helps to improve the severity of disease and itching,and has good safety,which is worthy of clinical promotion. 展开更多

Objective:To study the effects of Traditional Chinese Medicine Jianpi Chushi decoction and ointment on chronic eczema.Methods:DNCB acetone solution was used to sensitize the skin of back and ears of 36 rats in order to establish chronic eczema model.A total of 36 rats were divided into four groups of 9 randomly including oral medicine group,external inunctum group,combination therapy group,and model control group respectively.Besides,the blank group of 4 healthy rats were set.The oral medicine group was given Traditional Chinese Medicine Jianpi Chushi decoction [(Poria cocos,Chinese yam,Cortex dictamni,Zaocys dhumnade,Rhizoma atractylodis,Pericarpium citri reticulatae,Scutellaria baicalensis,Radix Sophorae Flavescentis,Raw Radix Paeoniae Alba,Licorice roots(Northwest Origin)] by gastric infusion(1.6 g/m L·5 m L/d); the external inunctum group was given Qingpeng ointment on the skin,the combination therapy group was given Jianpi Chushi decoction by gastric infusion and Qingpeng ointment combination therapy.The model control group was given normal saline(NS) of the same volume by gastric infusion and vaseline on skin.Continuous administration 15 d and stopped for 3 d.The thickness difference and weight difference of left and right ear of every group were measured and the degree of ear swelling were evaluated.The CD4^+ and CD8^+ content and the IL-2,IL-4 level of serum were detected,and the inflammatory cells counts of back skin were recorded.Results:After treatment,the degree of ear swelling of oral medicine group,external inunctum group and combination therapy group significant decreased compared with model control group(P<0.05).The CD4^+,CD8^+ cell content and IL-2 level of oral medicine group,external inunctum group,combination therapy group and model control group significant decreased compared with blank group,and IL-2 level and the inflammatory cells count increased.After 15 d of treatment,the CD4^+,CD8^+ cell content and IL-2 level of serum of oral medicine group,external inunctum group and combination therapy group raised and the IL-4 level and the inflammatory cells count had significant decreased compared with model control group,and the effect of combination therapy group was more obvious(P<0.05).Conclusions:Qral Jianpi Chushi decoction could treat chronic eczema effectively,and oral Chinese medicine combined with ointment could enhance and speed up the efficacy. 展开更多

BACKGROUND Aplasia cutis congenita (ACC) in newborns is a condition in which congenital defects or hypoplasia is present in part of the epidermis,dermis and even subcutaneous tissue (including muscle and bones).First reported by Cordon in 1767,ACC is a rare disease with a low incidence of 1/100000 to 3/10000.Currently,there are 500 cases reported worldwide.ACC can be accompanied by other malformations.The onset mechanism of the disease remains unknown but is thought to be correlated to factors such as genetics,narrow uterus,foetal skin and amniotic membrane adhesion,use of teratogenic drugs in early pregnancy and viral infection.CASE SUMMARY In August 2018,we treated a newborn with ACC on the left lower limbs using a combination of ionic silver dressing and moist exposed burn ointment (MEBO) and achieved a satisfactory treatment outcome.The skin defects were observed on the external genitals and on areas from the left foot to 3/4 of the upper left side.Subcutaneous tissue and blood vessels were observed in the regions with skin defects.The following treatments were provided.First,the wound was rinsed with 0.9% sodium chloride solution followed by disinfection with povidone-iodine twice.And then MEBO was applied to the wound at a thickness of approximately 1 mm.After applying ionic silver dressing,the wound was covered with sterile gauze.The wound dressing was replaced every 2-3 d.At the 4-mo follow-up,the treatment outcome was satisfactory.There was minimal scar tissue formation,and limb function was not impaired.CONCLUSION The combination of ionic silver dressing and MEBO to ACC is helpful. 展开更多

BACKGROUND The most effective treatment for knee joint pain is total knee arthroplasty(TKA),but the risk of pain and swelling in patients after surgery is high.Ice application,ankle pump exercise and non-steroidal anti-inflammatory painkillers are the primary clinical treatments after surgery.However,long-term use of nonsteroidal anti-inflammatory pain relievers can easily cause gastrointestinal damage.Traditional Chinese medicine(TCM)ointments and tuina therapy integrate TCM and manipulation,which effectively promotes the penetration of TCM into the skin lesions,improves local blood circulation and inflammatory reaction and has good long-term effects on patients.AIM To evaluate the efficacy of TCM ointment combined with tuina therapy in the treatment of pain and swelling after TKA.METHODS The randomized controlled clinical trial enrolled 80 patients who underwent TKA via the same procedure.The patients were randomly divided among the treatment group(n=40)and the control group(n=40).The control group was given an analgesia pump in addition to oral painkillers as the postoperative intervention.The treatment group received TCM ointment with tuina therapy in addition to the analgesia pump and oral painkillers in the postoperative period.The following variables were recorded 3 d before surgery and 3 d,7 d and 14 d after surgery:Visual analogue scale(VAS)score;skin temperature;circumferences at 15 cm above and below the patella;maximum active knee flexion angle;and the knee injury and Osteoarthritis Outcome score(KOOS).RESULTS After treatment,VAS was significantly lower in the treatment group than the control group at 7 d(t=7.536,P<0.001)and 14 d(t=8.563,P<0.001).The skin temperature of participants in the treatment group was significantly lower than that in the control group at 7 d(t=2.968,P=0.004)and 14 d(t=4.423,P<0.001).The circumference values of the two positions in the treatment group were lower than those in the control group at 7 d[t=2.315,P=0.023(above);t=2.121,P=0.037(below)]and 14 d[t=2.374,P=0.020(above);t=2.095,P=0.039(below)].After 14 d of treatment,the maximum active knee flexion angle and KOOS of the two groups were significantly improved but were significantly higher in the treatment group(P<0.05 for both).CONCLUSION TCM ointment and tuina therapy have significant advantages over standard care in the treatment of pain and swelling after TKA.This additional treatment may improve knee function but additional studies are needed to confirm our observations. 展开更多

It has been confirmed by basic pharmacodynamic experiments that Baimai ointment has some therapeutic effects on stroke. However, the detailed evaluations of Baimai ointment on clinically treating stroke patients for function recovery are still insufficient with little reports. OBJECTIVE: To observe the efficiency and safety of Baimai ointment on alleviating hypermyotonia following cerebral infarction DESIGN: A randomized, parallel and controlled experiment. SETTING: Department of Neurology, Beijing Aerospace General Hospital. PARTICIPANTS: A total of 120 patients with acute front-cycle cerebral infarction were selected from Department of Neurology, Beijing Aerospace General Hospital from July to December 2006 and met the diagnosis criteria modified by Forth National Seminar on Cerebrovascular Disease. The experiment had got confirmed consent from local ethic committee. All 120 patients with cerebral infarction were randomly divided into experimental group (n =80; 32 in the acute phase, 32 in the recovery phase and 16 in the sequela phase) and control group (n =40; 16 in the acute phase, 16 in the recovery phase and 8 in the sequela phase). METHODS: Within 4 weeks after the patients were enrolled in this study, no other traditional Chinese medicines containing similar components were adopted besides routine supporting therapy. The Baimai ointment being consisted of jianghuang, Roudoukou, Gansong, Yangqishi, Gancao, She, shannai, Zanghuixiang, Zangchangpu, Huajiao and dianhua was used by trial group (20 g/ampoule, Tibet Lingzhiqizheng Tibetan Drug Factory; batch number: 050403), 2-3 times per day, the ointment was moderately coated traveling parts of Baimai. MAIN OUTCOME MEASURES: The evaluation of National Institutes of Health Stroke Scale (NIHSS), Modified Rankin Scale and Modified Ashworth Scale were performed before administration, 2 and 4 weeks after administration. RESULTS: All 120 patients with acute front-cycle cerebral infarction were involved in the final analysis. ① Evaluation of therapeutic effects: After 4 weeks, the comparison of overall muscular tension and muscular tension in acute phase between the two groups (acute phase, chronic phase and recovery phase) demonstrated that the experimental group improved significantly (Z = - 2.25, - 2.06, P 〈 0.05). After 2 weeks, the change of muscular tension was not significantly different from baseline (P 〉 0.05). Whereas after 4 weeks, the change of muscular tension in acute phase was significantly different from baseline (P 〈 0.05); however, the change of muscular tension in recovery phase and sequelae phase was not significantly different from baseline (P 〉 0.05). After the treatment, the overall NIHSS and disability score of both groups did not change significantly (P 〉 0.05). ② Adverse reaction and side effect: There were two adverse events in experimental groups. One patient underwent petechia, and the symptom disappeared following stopping medication which did not reappear after remedication. The other patient complained of a pain coming from the place where the medication was used, and the symptom went away without treatment. Two cases of adverse events were not significantly correlated with the usage of medication, so both of which could not be confirmed to be adverse reactions. CONCLUSION: Baimai ointment achieves some therapeutic effects on treating hypermyotonia of cerebral infarction patients, especially significantly preventing the hypermyotonia in acute phase with good safety. 展开更多

The copper-coated iron oxide nanoparticles with core-shell were produced by deposition of a Cu shell on Fe3 O4 NPs through reduction of Cu2 tions in solution using NaB H4. Subsequently, the organosulfur compound, bis-(2,4,4-trimethylpentyl)-dithiophosphinic acid(b-TMP-DTPA), was used to form self-assembled monolayer in order to modify sorbent's surface via covalent bonding between Cu and thiol(–SH)terminal groups. The prepared magnetic nanoparticles were characterized by using Fourier transform infrared(FT-IR) spectroscopy, scanning electron microscopy(SEM), transmission electron microscope(TEM), vibrating sample magnetometer(VSM) and thermo gravimetric analysis(TGA). Then, the application of this new sorbent was investigated to extract the steroid drugs in ointment samples with the aid of ultrasound. An external magnetic field was applied to collect the magnetic nanoparticles(MNPs). The extracted analytes were desorbed using acetonitrile. The obtained extraction solution was analyzed by HPLC-UV. The main affecting factors on the extraction efficiency including p H, sonication time, amount of sorbent, salt concentration, and desorption conditions were optimized in detail. Under the optimum conditions, good linearity was obtained in the range of 2.5–250.0 mg/L with reasonable linearity(R24 0.99) and the limits of detection(LODs) ranged between 0.5 and 1.0 mg/L(based on S/N ? 3).Repeatability(intra-day precision) based on five replicates and preconcentration factors were calculated to be 3.6%–4.7% and 87–116, respectively. Relative recoveries in ointment samples at two spiked levels of the target analytes were obtained in the range of 90.0%–103.2%. The results illustrated that the Fe3 O4@Cu@ b-TMP-DTPA NPs have the capability of extraction of steroid drugs from ointment samples. 展开更多

The purpose of this study was to design an innovative topical ointment containing betamethasone dipropionate loaded nanostructured lipid carrier (BD-NLC) for the treatment of atopic dermatitis (AD). BD-loaded NLC was produced with precirol ATO 5 and oleic oil (OA) by melt emulsification method. Effects of surfactant concentration, amount of solid lipid and liquid lipid on skin retention and skin penetration were investigated by in vitro percutaneous permeation experiment. The optimized BD-NLC showed a homogeneous particle size of 169.1 nm (with PI = 0.195), negatively charged surface (-23.4 mV) and high encapsulation efficiency (85%). Particle morphology assessed by TEM revealed a spherical shape. In vitro skin permeation study was carried out to investigate the percutaneous behaviors of W/O ointment with BD-NLC and Carbopol emulgel ointment with BD-NLC. W/O ointment with BDNLC showed high skin retention (35.43 μg/g) and low penetration (0.87 μg/ml). In vitro drug release studies were carried out to demonstrate the drug releasing properties of the two ointments. W/O ointment with BD-NLC showed an advantage for skin retention as it was better for drug release. The tissue distribution test suggested that BD distribution was skin > muscle > blood. Self-made topical ointment in mice showed no skin irritation. The animal experiments indicated that BD-loaded NLC ointment was effective and safe for topical use. 展开更多

OBJECTIVE: To investigate the influence of heated moxa sticks( "moxibustion") and Daiwenjiu ointment(DO) on changes in temperature and blood perfusion volume on the skin of the backs of healthy subjects.METHODS: DO was spread on the left side of the body, and the right side of the body was treated with a heated moxa stick. Images denoting blood perfusion and body temperature were collected 7-8 cm lateral to the spinous process of the sixth thoracic vertebra using laser speckle contrast imaging(LSCI). Data obtained from eight-frame images were analyzed and used to calculate the mean blood perfusion volume. Simultaneously,blood-perfusion images were collected from the body surface and used to compare the change in blood flow on the body surface and the actual position of imaging.RESULTS: After moxibustion, a rapid increase in blood perfusion volume and body temperature was noted in the local skin surface. The maximum blood perfusion volume and body temperature was noted at 20 min(P<0.05). At 80-110 min after the spreading of DO, a gradual increase was noted in blood perfusion volume(P<0.05) and body temperature(P<0.05) compared with the baseline level. The maximum blood perfusion volume was at 110 min.CONCLUSION: Using LSCI, these data revealed a rapid and sharp increase in blood perfusion volume and body temperature after treatment with moxibustion, but the respective changes seen in the DO group were gradual and moderate. 展开更多
