We investigated the balance of free radicals in different tissues (liver, heart, brain and muscle) of rats in course of in vivo and in vitro processing by Macrovipera lebetina obtusa (MLO) and Montivipera raddei (MR) snake venoms. Chemiluminescence (ChL) levels were examined in tissue assays after incubation (at 37 &#176C for a period of 10 min) with venom for in vitro experiments and in tissue assays isolated of 10 min after venom injection for in vivo experiments. The TBA-test was also performed to confirm the free radical expression. The activities of antioxidant enzymes (such as superoxide dismutase and glutathione peroxidase) in isolated tissues were detected by spectro-photometry. During the in vitro processing chemiluminescence levels of tissue homogenates significantly decreased, while in course of in vivo intoxication the level of ChL was elevated in brain and liver;lipid peroxidation also increased in brain tissue, but there was no significant balance change in other tissues;the activity of superoxide dismutase mainly correlated with changes of free radical balance during intoxication. On the contrary, the activity of glutathione peroxidase showed the reverse tendencies to change. We suggest that free radicals and their oxidative stresses may play a role in the early stage of intoxication causing the so-named “spreading-effect”, which is very characteristic for the venom of vipers. 展开更多

Objective:To investigate the antioxidant and cytotoxic properties of the most active extracts from Ulva lactuca and Laurencia obtusa against colon and cervical cancer cell lines.Methods:The antioxidant radical scavenging activity of the algal extracts was estimated using the 2,2-diphenyl-1-picrylhydrazyl colorimetric assay.Moreover,the cytotoxic potential of these bioactive extracts was studied against HCT-116 and HeLa cells by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide assay.Wound healing analysis was adopted to evaluate the antimetastatic effects of protein extracts from both algae.Cell cycle analysis was performed by flow cytometry,and apoptotic bodies were detected using 4′,6-diamidino-2-phenylindole fluorescent staining.Amino acids of hydrolyzed protein extract were separated and identified chromatographically by high performance liquid chromatography.Results:The green algae Ulva lactuca had higher lipid content than Laurencia obtusa,whereas the latter had higher protein content with high antioxidant capacity.Protein extracts had significant dose-and time-dependent cytotoxicity and anti-metastatic activity against HCT-116 cells.Protein b extracts of both algae obtained from the chloroform:hexane solvent lipid-free residue caused morphological changes and induced apoptosis of HCT-116 cells.Further analysis revealed that apoptosis induced upon Laurencia obtusa protein b treatment was triggered via the ROS pathway,causing a significant increase in the sub-G_(1) population.Glycine and arginine(5.94 and 5.47μM,respectively)were the most common amino acids in Laurencia obtusa protein b extract.Conclusions:Proteins of Laurencia obtusa and Ulva lactuca could be considered as an adjuvant nutraceutical source of pro-apoptotic and anti-metastatic biomolecules against colon cancer. 展开更多

Caryota obtusa of Arecaceae is a fascinating palm tree native to southwestern China,India,Myanmar,Thailand,Laos and Vietnam.It is an economically important and threatened species and appears as a canopy dominant in some karst areas in Yunnan.We aim to clarify the forest structure,species diversity,population status and regeneration dynamics of C.obtusa in the karst forest ecosystem of Yunnan,China.We established 56 vegetation plots dominated by C.obtusa in 10 counties of southern Yunnan.Based on the plot data,we analyzed the community stratification,floristic composition and C.obtusa’s population structure.We used questionnaires to interview local people and recorded the human activity history in C.obtusa-dominated forests.Caryota obtusa palm forests were distributed on limestone mountain slopes and gullies.There were seven forest community types.The stratification of each community included arborous layer,shrub layer and understory.The communities had rich species composition.For all the plots as a whole of each community type,Shannon–Wiener diversity index of either woody or herbaceous species ranged from 2.1 to 3.8.The diameter at breast height(DBH)-class frequency distribution of C.obtusa was a multimodal type.The regeneration was sporadic and dependent on intermediate natural disturbances.In the current population structure,a number of C.obtusa trees with small DBHs consisted mainly of the forest communities with no or a slight degree of human disturbances.Intensive human activities terribly hindered recruitment of C.obtuse,followed by the medium intensity of human activities. 展开更多

Aims: The microglia activity of rat brain following exposure of theMacrovipera lebetina obtusa venom was investi-gated. Methods: Histochemical analysis of brain microcirculatory bed staining by Ca2+ ATPase method for variable doses after intraperitoneal injections given for different time periods was used. The hemorrhagic activity of snake venom metalloproteinases was tested. Toxicological data were calculated using Behrens and Miller-Tainter methods. Surface, size of brain microglial cells (MGCs) and staining intensity were quantified using ImageJ software. Results: The vasodestructive action of the venom resulted in changes in ATPase activity. The intensity of staining of rat brain microcirculatory bed was venom dose-, and time-dependent. Increased activity of MGCs in hemorrhagic loci of different regions of venom affected brain was also demonstrated. Conclusion: The activation of microglia and changes of its form, size, and position strongly correlates with hemorrhage-induced cerebrovascular damage. 展开更多

Objective:To study the possible effects of Cerithidea obtusa extract as antidiabetic and to screen the secondary metabolites presence.Methods:Antidiabetic activity of Cerithidea obtusa extract was measured in vitro usingα-glucosidase inhibition method.Whereas,secondary metabolites screening was measured qualitatively.Results:The methanol extract had antidiabetic activity(IC_(50)=36.40 mg/mL).However,the control drug acarbose had significantly higher antidiabetic activity(IC_(50)=0.32 mg/mL).Secondary metabolites screening showed the presence of alkaloids,flavonoids,triterpenoids and saponins.Conclusions:The methanol extract had antidiabetic activity and the presence of alkaloids,flavonoids and triterpenoids might contribute to the activity. 展开更多

【目的】林窗是森林中树木死亡倒伏后形成的空隙,可以改变森林的空间结构与林下植被生物多样性的组成,林窗的形成对森林群落生物多样性的动态变化及保护具有重要意义。【方法】以庐山国家级自然保护区的日本柳杉(Cryptomeria japonica)... 【目的】林窗是森林中树木死亡倒伏后形成的空隙,可以改变森林的空间结构与林下植被生物多样性的组成,林窗的形成对森林群落生物多样性的动态变化及保护具有重要意义。【方法】以庐山国家级自然保护区的日本柳杉(Cryptomeria japonica)与日本扁柏(Chamaecyparis obtusa)人工林为研究对象,通过对2种人工林林窗内植物多样性的采样调查,分别探讨2种人工林林窗内物种多样性与系统发育多样性,并比较不同人工林下植物多样性谱系结构的差异。【结果】(1)日本扁柏人工林林窗内共有48种植物,隶属于26科40属;日本柳杉人工林林窗内共有114种植物,隶属于47科77属,不同面积日本柳杉林林窗的植物种类及多度均高于日本扁柏林林窗;(2)不同面积林窗下2种人工林的物种多样性存在差异,但均为中林窗内植物多样性最高;(3)日本柳杉林林窗下的植物多样性谱系结构在中小林窗面积下趋于聚集;日本扁柏林林窗下的植物多样性谱系结构在中等与大面积林窗下呈现集聚趋势。【结论】庐山地区不同人工林林窗内植物多样性组成存在明显差异,中等面积林窗有利于提高日本柳杉与日本扁柏人工林群落物种多样性与系统发育多样性。 展开更多

日本扁柏是优良用材、荒山绿化和园林观赏树种 ,世界各国广为引种栽培 ,已成为我国长江中下游地区中、高山地带的重要造林树种。对其地理分布、生物生态学特性、栽培技术等进行了研究总结。结果表明 ,日本扁柏是一个喜温暖湿润的浅根性... 日本扁柏是优良用材、荒山绿化和园林观赏树种 ,世界各国广为引种栽培 ,已成为我国长江中下游地区中、高山地带的重要造林树种。对其地理分布、生物生态学特性、栽培技术等进行了研究总结。结果表明 ,日本扁柏是一个喜温暖湿润的浅根性树种 ,生长快 ,适应性强 ,繁殖容易 ,病虫害少。适合中性至微酸性土壤 ,可耐 - 2 5℃低温。较喜光 ,幼时稍耐庇荫。春季日均温大于 10℃时开始萌动 ,超过 15℃时开始生长 ;生长具明显的坡向和坡位效应 ;树高速生期为 10~ 2 0 a,>5 0 cm·a-1;胸径速生期 2 0~ 30 a,>0 .6cm· a-1;材积速生期 30~ 40 a。可进行种子或扦插繁殖 ,结实有丰歉年 ,种子发芽率 30 %~ 40 %。造林密度为 2 5 0 0~ 45 0 0株· hm-2 ,以不超过 30 0 0株· hm-2为好。目前我国引种栽培日本扁柏的主要问题是 :( 1)遗传基础狭窄 ,需引进更多的种源和家系 ;( 2 )大多为纯林 。 展开更多

本文以日本扁柏人工林为研究对象,通过对间伐林分和不间伐林分的生产结构、光能分布、光合速率、枝叶生长等季节变化的观测,进而计算了光能利用效率的季节变化。通过比较研究,证明了生产结构影响光能利用效率,而密度影响群落生产结构。... 本文以日本扁柏人工林为研究对象,通过对间伐林分和不间伐林分的生产结构、光能分布、光合速率、枝叶生长等季节变化的观测,进而计算了光能利用效率的季节变化。通过比较研究,证明了生产结构影响光能利用效率,而密度影响群落生产结构。合理的生产结构可以使光能在群落内合理分配、各层叶的光合潜力充分发挥、提高群落的光能利用效率。为了标定林分生产结构的合理性,本文提出了相对光合时间数和相对光能利用率的概念及数学公式。绘制了生产结构与功能图。对合理的生产结构与合理密度的关系进行了论述。 展开更多

RAPD作为一简易的DNA分子标记,在得以广泛应用的同时,存在着随机扩增的DNA条带并不遵行显性遗传模式,以及受PCR反应条件影响大等现象。以5个日本扁柏Chamaecyparisobtusa优树无性系及其部分双列杂交的12组合(各含30个体)F1代为材料,探讨RAPD标记的子代遗传及分离特征。研究结果表明:优树无性系中供试的46个引物中,14个引物扩增了42条多态的片段,筛选了其中7个引物扩增得来的14条片段进行分析,在子代中均能找到其对应的片段,且这些片段符合孟德尔的遗传分离规律。说明RAPD可作为遗传标记在日本扁柏中用于遗传分析。 展开更多

Often facilitated by human-mediated pathways,aquatic invasive species are a threat to the health and biodiversity of global ecosystems.We present a novel approach incorporating survey data of watercraft movement in a social network analysis to reconstruct potential pathways of aquatic invasive species spread between lakes.As an example,we use the green alga Nitellopsis obtusa,also known as starry stonewort,an aquatic invasive species affecting the Great Lakes region in the United States and Canada.The movement of algal fragments via human-mediated pathways(i.e.,watercraft)has been hypothesized as the primary driver of starry stonewort invasion.We used survey data collected at boat ramps during the 2013 and 2014 openwater seasons to describe the flow of watercraft from Lake Koronis,where N.obtusa was first detected in Minnesota,to other lakes in the state.Our results suggest that the risk of N.obtusa expansion is not highly constrained by geographic proximity and management efforts should consider highly connected lakes.Estimating human movement via network analysis may help to explain past and future routes of aquatic invasive species infestation between lakes and can improve evidence-based prevention and control efforts. 展开更多
