NUP62 alleviates senescence and promotes the stemness of human dental pulp stem cells via NSD2-dependent epigenetic reprogramming 被引量:1

Stem cells play a crucial role in maintaining tissue regenerative capacity and homeostasis.However,mechanisms associated with stem cell senescence require further investigation.In this study,we conducted a proteomic analysis of human dental pulp stem cells(HDPSCs)obtained from individuals of various ages.Our findings showed that the expression of NUP62 was decreased in aged HDPSCs.We discovered that NUP62 alleviated senescence-associated phenotypes and enhanced differentiation potential both in vitro and in vivo.Conversely,the knocking down of NUP62 expression aggravated the senescence-associated phenotypes and impaired the proliferation and migration capacity of HDPSCs.Through RNA-sequence and decoding the epigenomic landscapes remodeled induced by NUP62 overexpression,we found that NUP62 helps alleviate senescence in HDPSCs by enhancing the nuclear transport of the transcription factor E2F1.This,in turn,stimulates the transcription of the epigenetic enzyme NSD2.Finally,the overexpression of NUP62 influences the H3K36me2 and H3K36me3 modifications of anti-aging genes(HMGA1,HMGA2,and SIRT6).Our results demonstrated that NUP62 regulates the fate of HDPSCs via NSD2-dependent epigenetic reprogramming. 展开更多

Aiming at the problems of low detection efficiency and difficult positioning of traditional steel surface defect detection methods,a lightweight steel surface defect detection model based on you only look once version 7(YOLOv7)is proposed.First,a cascading style sheets(CSS)block module is proposed,which uses more lightweight operations to obtain redundant information in the feature map,reduces the amount of computation,and effectively improves the detection speed.Secondly,the improved spatial pyramid pooling with cross stage partial convolutions(SPPCSPC)structure is adopted to ensure that the model can also pay attention to the defect location information while predicting the defect category information,obtain richer defect features.In addition,the convolution operation in the original model is simplified,which significantly reduces the size of the model and helps to improve the detection speed.Finally,using efficient intersection over union(EIOU)loss to focus on high-quality anchors,speed up convergence and improve positioning accuracy.Experiments were carried out on the Northeastern University-defect(NEU-DET)steel surface defect dataset.Compared with the original YOLOv7 model,the number of parameters of this model was reduced by 40%,the frames per second(FPS)reached 112,and the average accuracy reached 79.1%,the detection accuracy and speed have been improved,which can meet the needs of steel surface defect detection. 展开更多

Objective This study aimed to explore the mechanism of obtaining yang from yin in Yougui pill against aging based on network pharmacology and molecular docking technology.Methods The active components and targets of Yougui Pill were obtained by searching the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database and the Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine(BATMEN-TCM)database,and kidney deficiency syndrome-related targets were obtained in the Symptom Mapping(SymMap)Database,a traditional Chinese medicine(TCM)syndrome correlation database.The protein–protein interaction(PPI)network was constructed by using the STRING11.5 database.Then,we used CytoScape3.9.0 software to construct the network of TCM–active components–potential targets,and the core TCM components and core targets of Yougui Pill for the treatment of kidney deficiency were obtained.The function analysis of Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway enrichment analysis were performed in the Database for Annotation,Visualization,and Integrated Discovery(DAVID).Finally,preliminary verification was performed with the help of molecular docking technology.Results A total of 147 active components of 9 drugs of Yougui Pill(Fuzi[Aconiti Lateralis Radix Praeparata],Shudihaung[Rehmanniae Radix Praeparata],Gouqi[Lycii Fructus],Shanyao[Rhizoma Dioscoreae],Shanzhuyu[Corni Fructus],Tusizi[Cuscutae Semen],Danggui[Angelicae Sinensis Radix],Duzhong[Eucommiae Cortex],Lujiaojiao[Cervi Cornus Colla])were obtained,corresponding to 233 targets.A total of 2,235targets related to kidney deficiency syndrome and 43 potential therapeutic targets were obtained after the intersection.The core TCM components mainly included quercetin,kaempferol,diosgenin,β-carotene,etc.The core targets involved Trp53(Tp53),Akt1,Pparg,Nr3c1,App,Casp8,Mapk1,Cav1,and Ctnnb1.A total of 27biological processes,10 cellular components,and 11 molecular functions were obtained by gene function enrichment analysis,mainly related to the regulation of gene expression,cell apoptosis and proliferation,and the response to estrogen.A total of 51 KEGG signaling pathways,mainly involving a variety of cancer pathways,apoptosis pathways,longevity regulation pathways,etc.Conclusion Yougui Pill can play a role in preventing and treating kidney deficiency syndrome through multiple targets and pathways. 展开更多

Wollastonite, a mineral of wide industrial applications was synthesised from rice husk ash silica and limestone. A number of raw batches consisting of these starting materials, in 1:1 molar ratio, were heat treated to produce it through solid state reaction from 900℃ to 1300℃. The conducted reaction was monitored by XRD step by step. Amount of Wollastonite formed at every temperature was also studied to some extent. Analyses of the obtained data indicated that the target mineral formation was quite effective and almost proportional to a rise in temperature up to 1200℃. The results from both, XRD and chemical analysis were found in fair agreement with one another 展开更多

In this paper, a photoelectric signal obtained scheme via equator triangle pattern engraved on rotor is discussed and the mathematic model is deduced in the case which is deflexion between rotor axis and the coordinate frame of case. The deflexion error and coupling error under the situation are analyzed. Finally, three methods of engraving based on the spherical triangle pattern are presented. The error models of various methods are built up and the simulation curves are provided respectively. We have done the primary experiments on the surface of rotor using this method. It can be seen from the enlarged figures that the rim of the pattern is smooth and the demand of sensor resolution is satisfied by and large. The magnetic properties and microstructure of diffusion annealed [Ta/Nd/NdFeB/Nd/Ta]thin films have been investigated. The films were deposited on Si substrate with various thickness ratio of Nd/NdFeB layer (R=0~3.3), then diffused and crystallized by annealing at 650℃ for 10 min. The film without Nd layer showed soft magnetic behavior and high content of a-Fe phase. The films with R > =1 showed good hard magnetic properties with the high coercivity of about 20 kOe. 展开更多

Short-duration heavy rainfall(SHR),as delineated by the National Meteorological Center of the China Me-teorological Administration,is characterized by hourly rainfall amounts no less than 20.0 mm.SHR is one of the most common convective weather phenomena that can cause severe damage.Short-range forecasting of SHR is an important part of operational severe weather prediction.In the present study,an improved objective SHR forecasting scheme was developed by adopting the ingredients-based methodology and using the fuzzy logic approach.The 1.0°×1.0°National Centers for Environmental Prediction(NCEP)final analysis data and the ordinary rainfall(0.1-19.9 mm h-1)and SHR observational data from 411 stations were used in the improved scheme.The best lifted index,the total precipitable water,the 925 hPa specific humidity(Q 925),and the 925 hPa divergence(DIV 925)were selected as predictors based on objective analysis.Continuously distributed membership functions of predictors were obtained based on relative frequency analysis.The weights of predictors were also objectively determined.Experiments with a typhoon SHR case and a spring SHR case show that the main possible areas could be captured by the improved scheme.Verification of SHR forecasts within 96 hours with NCEP global forecasts 1.0°×1.0°data initiated at 08:00 Beijing Time during the warm seasons in 2015 show the results were improved from both deterministic and probabilistic perspectives.This study provides an objectively feasible choice for short-range guidance forecasts of SHR.The scheme can be applied to other convective phenomena. 展开更多

This paper begins with the overthrow of the concept of combining ability in crossbreeding by the concept of heritability.The reason is that general combining ability changes with the number and kind of pure strains in the foundation stock and hence special combining ability changes also,so that work with different kinds of pure strains in the foundation stock cannot be compared.Hence combining ability is useless as a parameter to predict the amount of heterosis expected in the next generation.On the other hand,since each cross has a separate heritability,it can be applied to a cross population just as successfully as in purebreeding.Since the same concept holds in both cases,resort to any other concept would be superfluous.That's why combining ability must be rejected.Another reason(not given in the full text)is,an infinite number of pure strains would be required in the foundation stock for its results to be comparable with those of the heritability theory,which disposes of its utility altogether.The main content of the thesis is then the centennial enigma of heterosis can be resolved by Descarte's theoretic method of deduction.Accordingly we start from the definition of heterosis.H=F¡-MP,where H is heterosis,F,is the first generation offspring,MP is the mean of the parents or midparent,and from the use of a binomial random variable and its extention to the multinomial case derive the basic relations of heterosis with its components.Starting with second degree statistics,we obtain Vn=Vr,-2cov(F,,MP)+Vup,where V and cov stand for variance and covariance.The equations of heterosis are v„=(1/2)Na²+(1/4)Nd’+Vr(F,)=additive dominance F,epistasis Vup=(1/2)Na’+(1/2)V1,additive parental epistasis V„=(1/4)Nd’+V(F)+(1/2)V1,dominance F,epistasis parental epistasis.where N is number of genes controlling a trait,a=(P1-P,)12,d is deviation from midparent,while the variance components are all indicated by their names under the repective terms.It turns out that all these can be easily computed from the data so that the problem becomes a simple one which any college student may solve.In other words,the right answers are found when the right questions are asked.Who had ever shown that the heritability principle is inapplicable in crossbreeding,e.g.,in a crossing of two pure strains?From this cue arose the realization that the F,of a cross of two pure strains must also be a Mendelian population,with p and q both equal to 1/2 which simplifies the algebra outright.This Heritability Theory of Heterosis,or HTH in capital letters,re-sts on 2 initial anguments:1)Since 0.5+0.5=1,crossing two pure strains gives a population which is only a special case of pure-breeding,thereforea heritability coefficient must exist for the F1;2)Our problem reduces to that of finding that coefficient;the an-swer is given by the additive component divided by Ve.,i.e.,(1/2)No'1 Vp..which is readily found from the solution of the het-erosis equations.Thus the elemnal enigma of heterosis is resolved!This happened at the end of the 20th century.We now come to the second point of the discovery,the new genetic parameter crossheritability which will rise in size with the increase of the number of times it's used and form the link between breeding and evolution.The advent of the Age of Evolution Engineering in the 21st century marks a totally new era,showing that artificial will ultimately supercede natural selection,with the long span of time element eliminated.For agriculture at least,it means there is no limit to the increase of food supply by the new method,with the concentra-tion of desirable genes by hybridization in place of the old theory of their fixation.Genetic gain is achieved through artificial selec-tion,with an 80%saving of time,labor and cost by adoption of the new method.Applied to a further increase in all kinds of agri-cultural products including hybrd rice,it means that a huge eacalation,in fact a New Green Revolution,on a much langer scale than that of any such before,is in view,provided it is adopted in our research and educational institutions as early as possible,ere its spread elsewhere.The possibilities from the evolution point of view can only be pictured by science fiction. 展开更多

With a total investment of 300 million yuan, more than 1000 geologists were involved annually to conduct mapping at a grid of one line to every four km along the Kunlun Mts-Altyn Shan-Tanggula Shah, Kokoxili-Qiangtang, Gangdese, Yalun Zangbu River, and finally to the Himalayan ranges. The extensive mapping culminated in a total of 112 maps covering an area of 152 square kilometers. Based on this success, the China Geological Survey launched another program, the "Integration and Comprehensive Study of Basic Geological Survey Result(s) in the Qinghai-Tibet Plateau". The program lasted from 2006 to 2010 and has yielded copious research results, making big strides in upgrading our understanding of the Qinghai-Tibet Plateau. The achievements are manifold. Around 100 maps have been drafted, including the 1:1500000 geological map, tectonic, tectono-magmatic, metamorphic rocks, Precambrian geology, lithofacies palaeogeography, Cenozoic geology, geophysical-geochemical maps and others. 展开更多

A brief description will be provided for the writer's life and his works, and then I will discuss Captain Ahab's paranoid personality through his whaling capturing process. Then, I will discuss obtaining principles of Captain Ahab, Mate Starbuck, Crew Ishmael, and reveal the Captain's tragic causes and reflected thinking. Spatial characteristics of Thomson scattering spectra are studied for an electron moving in the circularly polarized laser field in the presence of a strong uniform magnetic field. The results show that the angular distributions of the spectra with respect to the azimuthal and polar angles exhibit different symmetries, respectively, which depend on the fields and electron parameters sensitively and significantly. Moreover, for relatively large parameters such as high laser intensity, high magnetic resonance parameter as well as large initial momentum of electron, the two lobes in spectra tend to the laser-propagating direction so that the radiation can be collimated in the forward direction. Furthermore, an important finding is that by choosing the appropriate fields and initial momentum of electron, the high frequency part of the Thomson scattering spectra can reach the frequency range of soft x-ray,in which a high radiation power per solid angle as ~10^11 a.u. can be obtained. 展开更多
