Four new 3,4-seco-labdane diterpenoids,nudiflopenes J-M,were isolated from the leaves of Callicarpa nudiflora along with six known compounds.The structures of these diterpenoids were determined by comprehensive spectroscopic analysis.All the isolated compounds were evaluated for their inhibitory effects on NO production in LPS-stimulated RPMs and RAW264.7 cells.The results suggest that nudiflopenes J-M and other four known compounds showed significant inhibitory effects against NO production comparable to the positive control dexamethasone. 展开更多

Objective:To evaluate the antidiabetic effect of Callicarpa nudiflora extract in streptozotoc-ininduced diabetic rats.Methods:The chemical constituents in Callicarpa nudiflora extract were identified by UPLCQ-TOF-MS.Callicarpa nudiflora extract(0.15 and 0.3 g/kg)was orally administered to streptozotocin-induced diabetic rats for 42 d.The effects of Callicarpa nudiflora extract on body weight,blood glucose,serum insulin,total cholesterol,triglyceride,LDL-C and HDL-C were investigated,and its effect on liver and pancreatic pathology was assessed by histopathological analysis.Moreover,the expression levels of adenosine 5’-monophosphate-activated protein kinase(AMPK),glucose transporter type 4(GLUT4),phospho-AMPK/AMPK,and p-acetylcoA carboxylase(P-ACC)/ACC in the skeletal muscles and liver were determined by reverse transcription-polymerase chain reaction,Western blotting,and immunohistochemistry.Results:A total of 34 compounds,including 8 iridoids,14 phenylpropanoids,and 12 flavonoids,were identified from Callicarpa nudiflora extract.Callicarpa nudiflora extract significantly reduced blood glucose and significantly restored all other biochemical parameters to near normal levels,including serum insulin,total cholesterol,triglyceride,LDL-C,and HDL-C.Callicarpa nudiflora extract improved insulin resistance and reversed the damage in the liver and pancreas caused by diabetes.Furthermore,Callicarpa nudiflora extract increased the expression levels of phospho-AMPK,GLUT4,P-ACC,and insulin receptor substrate-1 and decreased the expression level of PPAR r in diabetic rats.Conclusions:Callicarpa nudiflora extract improved oral glucose tolerance,lipid metabolism,insulin resistance,and reversed the diabetes-related damage in the liver and pancreas by activating the AMPK-ACC pathway. 展开更多

Objective: To study the effect of the extracts from Callicarpa nudiflora on wound healing and the preparation of ointment. Methods: Totally 50 KM mice were randomly divided into five groups, which were negative control group ,positive control group, high, middle and low dose groups of Callicarpa nudiflora ointment. Mice ear swelling test was conducted to investigate the effect of Callicarpa nudiflora induced by xylene, Bacteriostasis experiments were conducted to observe the bacteriostasis effects of Callicarpa nudiflora on five indicator bacteria, i.e. Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Bacillus subtilis, and Staphylococcus aureus in vitro. A total of 30 new zealand white rabbit with half male and half female were randomly divided into five groups, which were negative control group, positive control group, high, middle and low groups of Callicarpa nudiflora ointment. The rabbits were treated with sterilizing blade and treated with external medicine to observe the effect of the extracts from Callicarpa nudiflora on wound healing. Orthogonal test was used to determine the preparation method of Callicarpa nudiflora ointment,Physical properties, centrifugal stratification test, heat and cold resistance test were conducted. Results:Bacteriostasis experiments in vitro showed high dose group of Callicarpa nudiflora extracts got 16.8 cm antimicrobial zone of Pseudomonas aeruginosa. The experimental results showed that the ear swelling degree of mice induced by xylene was significantly reduced in the middle and high dose groups. All the treatment groups could promote skin healing of rabbit significantly compared with negative control group. There were significantly differences between all the treatment groups and negative control group on the wound healing rate. The optimum preparation process was determined by orthogonal screening test. The optimal formula was as follows: Twain-801.2 g, glycerin monostearate 4.7 g, glycerinum 0.9 g, liquid paraffin 10.8 g, Span801.4 g,solid paraffin 1.4 g,vaseline 0.9 g. The emulsification temperature was 65 ℃,and emulsification time was 25 min. Conclusions: The extract of Callicarpa nudiflora can be used as external preparation and the preparation is in compliance with Chinese pharmacopoeia 2015. 展开更多

Objective:To establish the quality standard of Li folk medicine callicarpa nudiflora powder by modern analytical method.Methods:TLC was used to identify the powder.Five heavy metals and harmful elements(Pb,Cd,As,Hg and Cu)in the powder were determined by flame atomic absorption spectrometry and hydride atomic fluorescence spectrometry.The content of luteolin and Mullein glycoside in the powder was determined by HPLC.Results:the fluorescent spots of TLC identification of powder were clear,and the separation of the two main components was good.It has good specificity,reproducibility and durability,which can be used for the identification of the powder.The inspection items of heavy metals for the identification of powder were established.It was stipulated that:according to the determination method,the lead should not exceed 5 mg/kg;the cadmium should not exceed 0.3 mg/kg;the arsenic should not exceed 2 mg/kg;the mercury should not exceed 0.2 mg/kg;the copper should not exceed 20 mg/kg.The content of luteolin and Mullein glycoside in the powder were determined by HPLC and the content inspection item of the powder was established.It was stipulated that the content of luteolin and Mullein glycoside in the powder should not be less than 0.48%and 5.83%,which can be used for the content limit inspection of the powder.Conclusion:the quality standard method established by our institute is advanced and operable,and can be used for the quality control of the powder. 展开更多

目的探讨裸花紫珠(CN)对反复性骨骼肌损伤大鼠修复作用及机制。方法SD大鼠随机分为对照组、模型组及150、300、600 mg/kg CN治疗组。模型组与CN治疗组建立胫前肌反复性损伤模型,CN组从造模首日灌胃给药,分别于第8天(早期)和第22天(晚期... 目的探讨裸花紫珠(CN)对反复性骨骼肌损伤大鼠修复作用及机制。方法SD大鼠随机分为对照组、模型组及150、300、600 mg/kg CN治疗组。模型组与CN治疗组建立胫前肌反复性损伤模型,CN组从造模首日灌胃给药,分别于第8天(早期)和第22天(晚期)取材,通过HE染色、免疫荧光先验证CN的治疗效果;为明确其药效物质基础,采用UPLC-HRMS表征CN主要化学成分并筛选活性成分;基于活性成分开展网络药理学分析,预测其作用靶点与通路;最终通过天狼猩红染色、Western blot对预测的TGF-β1/Smad通路及Ⅰ/Ⅲ型胶原蛋白进行验证。结果CN促进大鼠反复性胫前肌损伤的早期肌源性修复并改善其修复晚期纤维化。UPLC-HRMS鉴定出901种原型成分,筛选出44种潜在活性成分;网络药理学预测得到116个成分-疾病共同靶点,TGF-β信号通路显著富集。实验验证:在早期(第8天)CN干预显著减轻肌肉肿胀,增加肌管数量、Pax7+肌卫星细胞及MYOD1表达,同时上调Ⅰ/Ⅲ型胶原蛋白、α-SMA+细胞数及TGF-β1、Smad2、Smad3、p-Smad2/3蛋白表达;晚期(第22天)CN治疗降低骨骼肌萎缩及纤维化程度,下调Ⅰ/Ⅲ型胶原蛋白、α-SMA+细胞数及TGF-β1/Smad通路活性。结论CN通过双向调控TGF-β1/Smad通路促进反复性骨骼肌损伤修复:早期激活该通路加速细胞外基质沉积,支持肌源性再生;晚期抑制通路活性减轻过度纤维化,改善修复效果。 展开更多

裸花紫珠研究动态及热点:基于CiteSpace和VOSviewer可视化分析

目的基于文献计量学方法,分析裸花紫珠的研究现状、热点及发展趋势。方法以裸花紫珠为关键词,分别从中国知网(CNKI)和Web of Science(WOS)核心合集数据库中检索近20年的中英文文献。运用Excel、CiteSpace和VOSviewer软件对裸花紫珠中英... 目的基于文献计量学方法,分析裸花紫珠的研究现状、热点及发展趋势。方法以裸花紫珠为关键词,分别从中国知网(CNKI)和Web of Science(WOS)核心合集数据库中检索近20年的中英文文献。运用Excel、CiteSpace和VOSviewer软件对裸花紫珠中英文文献的总发文量、期刊发文量、高被引文献、合作网络、关键词等进行可视化分析。结果共纳入文献451篇,其中中文文献397篇,英文文献54篇。裸花紫珠年发文量整体呈上升趋势,发文作者及机构以谷陟欣(海南九芝堂药业有限公司)为主,发文国家以中国为主。刊文量最多的中英文期刊分别为《中草药》(13篇)和Phytochemistry(5篇)。中英文高被引文献主要围绕化学成分、抗炎、抑菌、止血等药理作用及机制研究。裸花紫珠研究热点集中于活性成分、药理作用及机制、临床应用方面。结论裸花紫珠研究在国内取得了一定进展,未来须加强国际合作,其研究趋势可能为深化药理作用及机制、规范临床试验、开发新剂型提供参考。 展开更多

目的分析裸花紫珠调控M2型巨噬细胞极化对糖尿病足溃疡大鼠伤口愈合及磷脂酰肌醇-3-激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白(PI3K/AKT/mTOR)通路的影响。方法选取40只大鼠,将其中10只作为对照组(生理盐水外敷),剩余30只用于建立糖尿病... 目的分析裸花紫珠调控M2型巨噬细胞极化对糖尿病足溃疡大鼠伤口愈合及磷脂酰肌醇-3-激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白(PI3K/AKT/mTOR)通路的影响。方法选取40只大鼠,将其中10只作为对照组(生理盐水外敷),剩余30只用于建立糖尿病足溃疡大鼠模型。最终建模成功27只大鼠,并分为模型组(生理盐水外敷)、裸花紫珠组(裸花紫珠外敷)、裸花紫珠联合抑制剂组(裸花紫珠外敷联合M2型巨噬细胞标志蛋白CD206抑制剂),每组9只。比较各组大鼠创面愈合率,碱性成纤维生长因子(bFGF)、血管内皮生长因子(VEGF)水平,诱导型一氧化氮合酶(iNOS)、CD16/32、CD206、PI3K、AKT、mTOR蛋白相对表达量,PI3K、AKT、mTOR的mRNA表达量。结果干预后第3、7、14天,与对照组大鼠相比,各组大鼠创面愈合率、VEGF、bFGF、CD206相对表达量以及PI3K、AKT、mTOR的mRNA表达量和蛋白相对表达量均降低,iNOS、CD16/32相对表达量升高,差异有统计学意义(P<0.05);与模型组大鼠相比,裸花紫珠组、裸花紫珠联合抑制剂组大鼠创面愈合率、VEGF、bFGF、CD206相对表达量以及PI3K、AKT、mTOR的mRNA表达量和蛋白相对表达量均升高,iNOS、CD16/32相对表达量降低,差异有统计学意义(P<0.05);与裸花紫珠组大鼠相比,裸花紫珠联合抑制剂组大鼠创面愈合率、VEGF、bFGF、CD206相对表达量以及PI3K、AKT、mTOR的mRNA表达量和蛋白相对表达量均降低,iNOS、CD16/32相对表达量升高,差异有统计学意义(P<0.05)。结论裸花紫珠可促进M2型巨噬细胞极化,提高糖尿病足溃疡大鼠伤口愈合率以及VEGF、bFGF水平,激活PI3K/AKT/mTOR信号通路。 展开更多

裸花紫珠联合地塞米松保留灌肠治疗急性放射性直肠损伤的疗效观察

目的:比较使用裸花紫珠(Callicarpa nudiflora)联合地塞米松(dexamethasone)与单药地塞米松进行保留灌肠治疗急性放射性直肠损伤的疗效差异。方法:收集2021年11月—2023年6月在江西省人民医院接受放疗且出现急性放射性直肠损伤的100例... 目的:比较使用裸花紫珠(Callicarpa nudiflora)联合地塞米松(dexamethasone)与单药地塞米松进行保留灌肠治疗急性放射性直肠损伤的疗效差异。方法:收集2021年11月—2023年6月在江西省人民医院接受放疗且出现急性放射性直肠损伤的100例盆腔恶性肿瘤患者,并将其随机分为2组:对照组(50例)和治疗组(50例)。依据美国肿瘤放疗肿瘤研究组(Radiation Therapy Oncology Group,RTOG)制定的急性放射性损伤分级标准,剔除各组中急性放射性损伤评分等级≤1的患者,并制定后续药物保留灌肠的实施方案:对照组(45例)采用单药地塞米松保留灌肠;治疗组(47例)采用裸花紫珠联合地塞米松保留灌肠。然后记录对照组和治疗组患者在灌肠前和灌肠2周时的临床症状、排便频率以及大便常规检测中白细胞和红细胞的计数情况,以比较上述2种药物保留灌肠方案治疗急性放射性直肠损伤的疗效差异。结果:治疗组与对照组之间药物保留灌肠方案的疗效具有显著差异(χ^(2)=11.37,P=0.003,P<0.05),地塞米松联合裸花紫珠保留灌肠的有效率要显著高于单药地塞米松保留灌肠(82.2%vs 62.2%)。结论:裸花紫珠联合地塞米松保留灌肠可以显著改善急性放射性直肠损伤患者的临床症状,提高其生活质量,为临床上优化急性放射性损伤治疗方案提供了新的思路。 展开更多

裸花紫珠提取物体外抗乙型肝炎病毒作用及机制探究

本研究以裸花紫珠(Callicarpae Nudiflorae Folium,CNF)的水提取物(CNF water extract,WCNF)和醇提取物(CNF ethanol extract,ECNF)作用于HepG2.2.15细胞,体外评价CNF提取物对HBV的抑制作用,同时结合网络药理学探究CNF抗HBV的潜在作用... 本研究以裸花紫珠(Callicarpae Nudiflorae Folium,CNF)的水提取物(CNF water extract,WCNF)和醇提取物(CNF ethanol extract,ECNF)作用于HepG2.2.15细胞,体外评价CNF提取物对HBV的抑制作用,同时结合网络药理学探究CNF抗HBV的潜在作用机制。体外实验采用CCK-8法测定CNF提取物对细胞的毒性;酶联免疫吸附(enzyme-linked immunosorbent assay,ELISA)法检测乙型肝炎病毒e抗原(hepatitis B e antigen,HBeAg)和乙型肝炎病毒表面抗原(hepatitis B surface antigen,HBsAg)分泌量,实时荧光定量PCR(real-time fluorescence quantitative PCR,RT-PCR)法测定HBV DNA及关键靶点基因表达量。通过文献调研、TCMSP数据库检索CNF化学成分、Gene Cards数据库检索HBV相关靶点开展网络药理学研究;运用String数据库构建蛋白互作网络,DAVID数据库进行GO功能及KEGG通路富集分析,Cytoscape软件构建成分-靶点-通路网络可视化并进行拓扑学分析,AutoDock Vina软件进行CNF抗HBV关键成分与核心靶点的分子对接分析,并通过RT-PCR实验对关键靶点进行实验验证。体外实验结果表明CNF水醇提取物均可抑制HbeAg和HBsAg的分泌量,抑制HBV DNA的复制水平,并且水提物的抑制作用明显地强于醇提物。网络药理学结果显示,熊果酸、槲皮素、木犀草素、咖啡酸等化合物为CNF抗HBV的关键活性成分,肿瘤蛋白53(tumor protein 53,TP53)、白介素-6(interleukin-6,IL-6)、肿瘤坏死因子(tumor necrosis factor,TNF)、热休克蛋白90Alpha家族A类成员1(heat shock protein 90 alpha family class A member 1,HSP90AA1)为核心靶点,涉及磷脂酰肌醇-3-激酶/蛋白激酶B(phosphatidylinositol 3-kinase/protein kinase B,PI3K/AKT)、丝裂原蛋白活化激酶(mitogen activated protein kinase,MAPK)等信号通路。此外,实验结果显示CNF提取物可抑制HBV关键基因IL-6、TNF及HSP90AA1表达并提高TP53基因的mRNA表达水平。以上研究揭示了CNF通过多成分、多靶点、多途径发挥抗HBV的作用。后续将以本研究为基础,进一步对其药效物质基础及作用机制进行深入探讨,为CNF抗HBV的临床应用及新药开发提供坚实的理论依据。 展开更多

本研究旨在筛选对鸭疫里氏杆菌具有良好抑菌效果的中药。试验对广东梅州地区的疑似患病鸭采样并分离病原菌,对分离菌株进行形态学观察、革兰染色特性观察、生化特性鉴定和PCR鉴定,并分析其ompA基因序列,同时测定紫锥菊和裸花紫珠等中药... 本研究旨在筛选对鸭疫里氏杆菌具有良好抑菌效果的中药。试验对广东梅州地区的疑似患病鸭采样并分离病原菌,对分离菌株进行形态学观察、革兰染色特性观察、生化特性鉴定和PCR鉴定,并分析其ompA基因序列,同时测定紫锥菊和裸花紫珠等中药提取物对鸭疫里氏杆菌的最低抑菌浓度(MIC)和最小杀菌浓度(MBC)。结果表明,分离获得4株鸭疫里氏杆菌,所有分离菌株均能在血平板上生长为光滑的灰白色圆形菌落,革兰染色为阴性,生化鉴定结果显示各菌株均不发酵多种糖类,不产生硫化氢,不能液化明胶,过氧化氢酶试验阳性。PCR鉴定获得大小为707 bp的目的条带。药敏试验结果显示,紫锥菊、裸花紫珠、板蓝根、大黄和大青叶对鸭疫里氏杆菌的MIC值均为15.60 mg/mL,黄芩、黄芪和金银花对鸭疫里氏杆菌的MIC值均为125.00 mg/mL;紫锥菊、裸花紫珠、板蓝根和大青叶对鸭疫里氏杆菌的MBC值均为31.25 mg/mL,大黄对鸭疫里氏杆菌的MBC值为62.50 mg/mL,黄芩、黄芪和金银花对鸭疫里氏杆菌的MBC值均为250.00 mg/mL。结果提示,紫锥菊、裸花紫珠、板蓝根、大黄和大青叶对鸭疫里氏杆菌具有良好的体外抑菌效果。 展开更多

目的 探索裸花紫珠颗粒用于慢性阻塞性肺疾病(chronic obstructive pulmonary disease, COPD)急性加重期患者治疗对肺功能及炎性指标降钙素原(procalcitonin, PCT)、C反应蛋白(C-reactive protein, CRP)、血清淀粉样蛋白A(serum amyloid... 目的 探索裸花紫珠颗粒用于慢性阻塞性肺疾病(chronic obstructive pulmonary disease, COPD)急性加重期患者治疗对肺功能及炎性指标降钙素原(procalcitonin, PCT)、C反应蛋白(C-reactive protein, CRP)、血清淀粉样蛋白A(serum amyloid A,SAA)水平的影响。方法 选取COPD急性加重期患者108例随机均分为对照组与观察组,两组患者均给予吸氧、雾化吸入、抗生素、噻托溴铵粉吸入剂(18μg/次,1次/d)等常规治疗,观察组在此基础上再给予裸花紫珠颗粒(3g/次,3次/d)治疗,均治疗4周;比较两组患者治疗前后第1秒用力呼气容积(forced expiratory volume in one second, FEV1)、FEV1/用力呼气量(forced expiratory volume, FVC)比值,血清炎性指标白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、PCT、CRP和SAA,6分钟步行距离(6-minute walking distance, 6MWD)及疾病改善率。结果 治疗后4周,观察组轻度和中度患者的FEV1和FEV1/FVC改善情况均优于对照组(P<0.05),PCT、CRP、SAA、IL-6及TNF-α均低于对照组(P<0.05);观察组患者治疗后2周和4周6MWD均长于对照组(P<0.05);观察组患者治疗后1周、2周和4周的疾病改善率分别为40.74%、72.22%和96.30%,均高于对照组(P<0.05)。结论 对COPD急性加重期患者加用裸花紫珠颗粒治疗,能够抑制炎症因子释放,改善心肺功能,提高整体疗效。 展开更多

目的探讨裸花紫珠叶抗肿瘤活性部位的提取工艺。方法采用MTT比色法对裸花紫珠叶醇提物的不同溶剂萃取物进行抗肿瘤活性测定。选择料液比、提取次数、提取时间、乙醇浓度作为影响因素,以总黄酮含量为指标,采用Box-Behnken设计优化活性部... 目的探讨裸花紫珠叶抗肿瘤活性部位的提取工艺。方法采用MTT比色法对裸花紫珠叶醇提物的不同溶剂萃取物进行抗肿瘤活性测定。选择料液比、提取次数、提取时间、乙醇浓度作为影响因素,以总黄酮含量为指标,采用Box-Behnken设计优化活性部位的提取工艺。结果乙酸乙酯萃取部位(EAE)对人乳腺癌MCF-7细胞、卵巢癌OVCAR-3细胞和宫颈癌C-33A细胞的增殖具有抑制作用,其IC_(50)值分别为(38.96±2.42)μg/mL、(5.15±3.07)μg/mL、(26.33±1.46)μg/mL(72 h)。活性部位EAE最佳提取工艺为:料液比为1∶20,提取次数为2次,提取时间为2 h,乙醇浓度为70%。结论裸花紫珠叶乙酸乙酯萃取部位具有一定的抗肿瘤活性,其提取工艺经Box-Behnken设计优化后具有更高的提取率,且工艺稳定,可预测性好,值得推广应用。 展开更多
