Here, we described a simple and efficient method for constructing a normalized cDNA library from a high oil content cultivar of sesame Zhongzhi 14, during its oil accumulation stages. It combined switching mechanism at 5?end of RNA transcript (SMART) technique and duplex-specific nuclease (DSN) normalization methods. Double-stranded cDNAs were synthesized from mRNAs, processed by normalization and Sfi I restriction endonuclease, and finally the cDNAs were ligated to pDNR-LIB vector. The ligation mixture was transformed into Escherichia coli DH10B by electroporation. The capacity of the library was 1.0?06 clones in this library. Gel electrophoresis results indicated the fragments ranged from 700 to 2 000 bp, with the average size of 1 800 bp. Random picking clones showed that the recombination rate was 100%. The results showed that the cDNA library constructed successfully was a full-length library with high quality, and could be used to screen the genes related to development of oil synthesis.展开更多

The normal paraffin hydrocarbon distribution patterns of six crude oils were built and compared. The cluster analysis on the normal paraffin hydrocarbon concentration was conducted for classification and some ratios of oils were used for oils comparison. The results indicated: there was a clear difference within different crude oils in different oil fields and a small difference between the crude oils in the same oil platform. The normal paraffin hydrocarbon distribution pattern and ratios, as well as the cluster analysis on the nomad paraffin hydrocarbon concentration can have a better differentiation result for the crude oils with small difference than the original gas chromatogram.展开更多
In order to analyze the normal deviatoric stress that viscous-elastic fluid acting on the residual oil under the situation of different flooding conditions and different permeabilities, Viscous-elastic fluid flow equa...In order to analyze the normal deviatoric stress that viscous-elastic fluid acting on the residual oil under the situation of different flooding conditions and different permeabilities, Viscous-elastic fluid flow equation is established in the micro pore by choosing the continuity equation, motion equation and the upper-convected Maxwell constitutive equation, the flow field is computed by using numerical analysis, the forces that driving fluid acting on the residual oil in micro pore are got, and the influence of flooding conditions, pore width and viscous-elasticity of driving fluid on force is compared and analyzed. The results show that: the more viscous-elasticity of driving fluid increases, the greater the normal deviatoric stress acting on the residual oil increases;using constant pressure gradient flooding, the lager the pore width is, the greater normal deviatoric stress acting on the residual oil will be.展开更多
