This book presents general theories and applications of structural optimization methods. The first chapter of the book introduces various basic concepts of structural optimization through several illustrative examples. Chapter 2 is devoted to a comprehensive review of math- ematical programming methods, including classical methods such as the simplex method, the (SLP) method, the interior point method and relatively new approaches such as genetic algo- rithms. The third chapter presents the main topic of this book, the SLP methods based on the incremental equations of structures. Some useful techniques in implementation of SLP, including the move limit, scaling and active constraint identifying, are discussed. In chapter 4, classical optimality criterion approaches, including the fully stress approach for truss struc- tures and its extensions for membrane and plate structures, are presented with demonstrative examples. The remaining chapters develop an overview of structural optimization problems, among which are dynamic optimization, multi-criterion optimization, earlier work on the op- timal layouts of plates, as well as some practical issues in general size and shape optimization problems. 展开更多

The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. These optimized base editors will serve as powerful genome-editing tools for basic research or molecular breeding in rice and will provide a reference for the development of superior editing tools for other plants or animals. 展开更多

OBJECTIVE: Exploring the effect of Optimized New Shengmai powder(优化新生脉散方, ONSMP) on myocardial fibrosis in heart failure(HF) based on rat sarcoma(RAS)/rapidly accelerated fibrosarcoma(RAF)/mitogen-activated protein kinase kinase(MEK)/extracellular regulated protein kinases(ERK) signaling pathway. METHODS: Randomized 70 Sprague-Dawley rats into sham(n = 10) and operation(n = 60) groups, then established the HF rat by ligating the left anterior descending branch of the coronary artery. We randomly divided the operation group rats into the model, ONSMP [including low(L), medium(M), and high(H) dose], and enalapril groups. After the 4-week drug intervention, echocardiography examines the cardiac function and calculates the ratios of the whole/left heart to the rat's body weight. Finally, we observed the degree of myocardial fibrosis by pathological sections, determined myocardium collagen(COL) Ⅰ and COL Ⅲ content by enzyme-linked immunosorbent assay, detected the m RNA levels of COL Ⅰ, COL Ⅲ, α-smooth muscle actin(α-SMA), and c-Fos proto-oncogene(c-Fos) by universal real-time, and detected the protein expression of p-RAS, p-RAF, p-MEK1/2, p-ERK1/2, p-ETS-like-1 transcription factor(p-ELK1), p-c-Fos, α-SMA, COL Ⅰ, and COL Ⅲ by Western blot. RESULTS: ONSMP can effectively improve HF rat's cardiac function, decrease cardiac organ coefficient, COL volume fraction, and COL Ⅰ/Ⅲ content, down-regulate the m RNA of COL Ⅰ/Ⅲ, α-SMA and c-Fos, and the protein of p-RAS, p-RAF, p-MEK1/2, p-ERK1/2, p-ELK1, c-Fos, COL Ⅰ/Ⅲ, and α-SMA. CONCLUSIONS: ONSMP can effectively reduce myocardial fibrosis in HF rats, and the mechanism may be related to the inhibition of the RAS/RAF/MEK/ERK signaling pathway. 展开更多

This paper is the second part of the article and is devoted to the construction and analysis of new non-linear optimal weights for WENO interpolation capable of rising the order of accuracy close to discontinuities for data discretized in the cell averages.Thus,now we are interested in analyze the capabilities of the new algorithm when working with functions belonging to the subspace L1\L2 and that,consequently,are piecewise smooth and can present jump discontinuities.The new non-linear optimal weights are redesigned in a way that leads to optimal theoretical accuracy close to the discontinuities and at smooth zones.We will present the new algorithm for the approximation case and we will analyze its accuracy.Then we will explain how to use the new algorithm in multiresolution applications for univariate and bivariate functions.The numerical results confirm the theoretical proofs presented. 展开更多
