BACKGROUND Anxiety,depression,and other negative emotions are common among patients with chronic renal failure(CRF).Analyzing the factors related to negative emotions is necessary to provide targeted nursing care.AIM ...BACKGROUND Anxiety,depression,and other negative emotions are common among patients with chronic renal failure(CRF).Analyzing the factors related to negative emotions is necessary to provide targeted nursing care.AIM To explore the correlations among life satisfaction,pleasure levels,and negative emotions in patients with CRF.METHODS One hundred patients with CRF who received therapy at the First Affiliated Hospital of Jinzhou Medical University between December 2022 and February 2025 were included.The Depression,Anxiety,and Stress Scale(DASS-21),Satisfaction with Life Scale(SWLS),and Temporal Experience of Pleasure Scale(TEPS)were used to evaluate negative emotions,life satisfaction,and pleasure level,respectively.Pearson’s correlation coefficient analyzed the correlation between life satisfaction,pleasure level,and negative emotions.Linear regression analysis identified the factors affecting negative emotions.RESULTS The average DASS-21 score among patients with CRF was 51.90±2.30,with subscale scores of 17.90±1.50 for depression,18.53±1.18 for anxiety,and 15.47±2.36 for stress,all significantly higher than the domestic norm(P<0.05).The average SWLS score was 22.17±4.90.Correlation analysis revealed a negative correlation between the SWLS and total DASS-21 scores(P<0.05),but not with the individual depression,anxiety,or stress dimensions.The average TEPS score was 67.80±8.34.TEPS scores were negatively correlated with the DASS-21 score and the stress dimension(P<0.05),but not with depression or anxiety.Linear regression analysis showed that TEPS scores significantly influenced DASS-21 scores(P<0.05).CONCLUSION Patients with CRF experience high levels of negative emotions,which are negatively correlated with life satisfaction and pleasure.Furthermore,pleasure level had an impact on negative emotions.展开更多

Detecting geomagnetic anomalies preceding earthquakes is a challenging yet promising area of research that has gained increasing attention in recent years.This study introduces a novel reconstruction-based modeling ap...Detecting geomagnetic anomalies preceding earthquakes is a challenging yet promising area of research that has gained increasing attention in recent years.This study introduces a novel reconstruction-based modeling approach enhanced by negative learning,employing a Bidirectional Long Short-Term Memory(BiLSTM)network explicitly trained to accurately reconstruct non-seismic geomagnetic signals while intentionally amplifying reconstruction errors for seismic signals.By penalizing the model for accurately reconstructing seismic anomalies,the negative learning approach effectively magnifies the differences between normal and anomalous data.This strategic differentiation enhances the sensitivity of the BiLSTM network,enabling improved detection of subtle geomagnetic anomalies that may serve as earthquake precursors.Experimental validation clearly demonstrated statistically significant higher reconstruction errors for seismic signals compared to non-seismic signals,confirmed through the Mann-Whitney U test with a p-value of 0.0035 for Root Mean Square Error(RMSE).These results provide compelling evidence of the enhanced anomaly detection capability achieved through negative learning.Unlike traditional classification-based methods,negative learning explicitly encourages sensitivity to subtle precursor signals embedded within complex geomagnetic data,establishing a robust basis for further development of reliable earthquake prediction methods.展开更多
OBJECTIVE:To identify the antitumor effects of Gualou Beimu Yin(瓜蒌贝母饮,GLBMY)in triple-negative breast cancer(TNBC)and to explore the underlying mechanisms.METHODS:A mouse model of breast cancer was established an...OBJECTIVE:To identify the antitumor effects of Gualou Beimu Yin(瓜蒌贝母饮,GLBMY)in triple-negative breast cancer(TNBC)and to explore the underlying mechanisms.METHODS:A mouse model of breast cancer was established and treated with GLBMY.Freeze-dried GLBMY powder was used to treat MDA-MB-231 and BT549 cells to assess the therapeutic efficacy of GLBMY against TNBC.Network pharmacology,transcriptomics and metabolomics were employed to identify the potential mechanism of GLBMY in TNBC treatment.Finally,the main regulating genes and proteins in the enriched pathways were validated by Quantitative real-time polymerase chain reaction(q PCR)and Western blotting analysis to confirm its mechanism.RESULTS:GLBMY can inhibit the growth of TNBC through apoptosis and necroptosis pathways and inhibit TNBC lung metastasis by inhibiting epithelialmesenchymal transition(EMT).Network pharmacology has elucidated the most important active ingredients(tubeimoside I,emodin,cucurbitacin,and ursolic acid)and the most critical targets[interleukin-6(IL6),signal transducer and activator of transcription 3(STAT3),mitogen-activated protein kinase 3(MAPK3)]of GLBMY in treating TNBC.RNA-seq revealed that GLBMY affected the nuclear factor kappa-light-chain-enhancer of activated B cells,rat sarcoma virus,and MAPK signalling pathways.Metabolomics revealed that the metabolites mainly affected by GLBMY were L-(+)-lactic acid,isocitric acid,benzoic acid and indoxyl sulfate.Subsequent experiments demonstrated that GLBMY can inhibit EMT in TNBC through the MAPK/ERK pathway and inhibit the proliferation and progression of TNBC through the IL6-STAT signalling pathway.CONCLUSIONS:We confirmed that GLBMY inhibits the development and metastasis of TNBC through the MAPK/Erk and IL6-STAT signalling pathways.GLBMY shows promise as a long-term supplementary or alternative therapy for TNBC,offering new insights for TNBC treatment.展开更多

Lycoris longituba is a popular ornamental plant native to central eastern China. The decrease in anthocyanin accumulation causes L. longituba petal color fading during flower development, which considerably affects the ornamental value of L. longituba. However, mechanisms underlying anthocyanin biosynthesis inhibition during L. longituba petal development remain unclear. In this study, three LlDFR genes were confirmed to be involved in anthocyanin biosynthesis and LlDFRc exerted the strongest promoting effect on anthocyanin accumulation. According to the correlation analysis results, LlbHLH12 exhibited the strongest negative correlation with LlDFRc. Quantitative real-time PCR analysis showed that LlbHLH12 was highly expressed during the medium bud and full bloom stages of flower development. LlbHLH12 was identified as a member of subgroup XII of bHLH transcription factor family. Subcellular localization and transcriptional activation ability assay revealed that LlbHLH12 was located in the nucleus without transcriptional activation activity. Overexpression of LlbHLH12 in Nicotiana tabacum and L. longituba inhibited anthocyanin accumulation by suppressing the expression of anthocyanin biosynthetic pathway genes. Furthermore, yeast one-hybrid, dual-luciferase, and β-glucuronidase activity assays showed that LlbHLH12 directly bound to the promoters of LlPAL and LlDFRc and suppressed their expression to inhibit anthocyanin biosynthesis. Overall, our study identified a novel bHLH repressor negatively regulating anthocyanin biosynthesis and provided new insights into the molecular mechanisms underlying color fading in L. longituba petals.展开更多
In recent years,the rapid advancement of artificial intelligence(AI)technology has enabled AI-assisted negative screening to significantly enhance physicians'efficiency through image feature analysis and multimoda...In recent years,the rapid advancement of artificial intelligence(AI)technology has enabled AI-assisted negative screening to significantly enhance physicians'efficiency through image feature analysis and multimodal data modeling,allowing them to focus more on diagnosing positive cases.Meanwhile,multispectral imaging(MSI)integrates spectral and spatial resolution to capture subtle tissue features invisible to the human eye,providing high-resolution data support for pathological analysis.Combining AI technology with MSI and employing quantitative methods to analyze multiband biomarkers(such as absorbance differences in keratin pearls)can effectively improve diagnostic specificity and reduce subjective errors in manual slide interpretation.To address the challenge of identifying negative tissue sections,we developed a discrimination algorithm powered by MSI.We demonstrated its efficacy using cutaneous squamous cell carcinoma(cSCC)as a representative case study.The algorithm achieved 100%accuracy in excluding negative cases and effectively mitigated the false-positive problem caused by cSCC heterogeneity.We constructed a multispectral image(MSI)dataset acquired at 520 nm,600 nm,and 630 nm wavelengths.Subsequently,we employed an optimized MobileViT model for tissue classification and performed comparative analyses against other models.The experimental results showed that our optimized MobileViT model achieved superior performance in identifying negative tissue sections,with a perfect accuracy rate of 100%.Thus,our results confirm the feasibility of integrating MSI with AI to exclude negative cases with perfect accuracy,offering a novel solution to alleviate the workload of pathologists.展开更多
This study addresses the optimization of automated yarn handling in textile manufacturing by examining the related suction process through a combined numerical and experimental approach.In particular,a three-dimension...This study addresses the optimization of automated yarn handling in textile manufacturing by examining the related suction process through a combined numerical and experimental approach.In particular,a three-dimensional model of the suction nozzle was coupled with an equivalent linear-elastic beam representation of the yarn,and a Fluent-IDW-Abaqus weakly coupled fluid-structure interaction(FSI)framework was employed to capture the yarn’s release and dynamic response under negative-pressure suction.High-speed imaging experiments validated the simulations,demonstrating excellent agreement in displacements and velocities.According to the results,increasing the initial suction pressure from -0.04 MPa to -0.06 MPa reduces adsorption time by approximately 62% and markedly dampens yarn-end vibrations,enhancing suction performance.Pressures beyond -0.06 MPa,however,induce overshoot and nozzle collisions,increasing the risk of entanglement and mechanical damage.The outcomes of a statistical analysis are also presented to further quantify the interplay among energy consumption,suction efficiency,and operational success under varying pressures,thereby providing a rigorous foundation for the optimal selection of pressure parameters in automated yarn-handling systems.展开更多

The accuracy of the developed simplified approach was verified by numerical simulation model with the same condition. Then, the influence factors, such as, taper angles, the diameter of pile base, surcharge, and pile-soil interface parameters were analyzed and discussed. The results show that the developed simplified approach can calculate NSF of special-shaped pile under surcharge effectively. A limited parametric study indicates that in many practical situations special-shaped piles(such as belled wedge pile shown in this work) offer a design option that is more economical than traditional uniform cross-section piles.展开更多

The experimental re...A series of model experiments of bucket foundations concerning suction installation and negative pressure consolidation in saturated silt were carried out in a cube steel bin at Tianjin University. The experimental results show that the silt inside the bucket has been strengthened by negative pressure, and the strengthening effect decreases with the increase of the distance from the bucket. A three-dimensional numerical model of the experiments was built by means of finite element software ABAQUS with fluid-solid coupling method. The results show that the bearing capacity of the silt inside the bucket foundation increases significantly at the former stage of negative pressure consolidation, while the increasing trend slows down over time. The rotation centers of the bucket foundation and the inner soil region tend to be closer to each other based on the consolidation. The bearing capacity of the bucket foundation is improved effectively with the increase of soil strength. The effects of negative pressure consolidation on the bearing capacity of bucket foundation were also illustrated by an actual offshore wind power project case.展开更多

Methods: Thirty-seven ...Objective: The aim of this study was to evaluate the anti-tumor activity and safety of Gemcitabine (GEM) combined with Vinorelbine (NVB) in patients with advanced TNABC after chemotherapy. Methods: Thirty-seven patients with immunohistochemical proved TNABC were enrolled. The patients received 21-day cycles of NVB 25mg/m^2 i.v. with GEM 1000 mg/m^2 i.v. on days 1 and 8. Results: A total of 136 cycles were given to 37 patients(median 4 cycles, ranged 2-6 cycles). The treatment response was evaluable in all patients. Of the 37 patients, 1 received complete remission (CR), 8 received partial remission (PR), 20 had stable disease (SD), 9 had progressive disease (PD). Overall objective response (CR+ PR) were 24.3 %. The median time to progress (TTP) was 6 months (95% CI, 4-6 months). The median overall survival was 24 months (95% CI, 11-37 months). The median 1-year survival rate was (66.24±8.43)%. The median 3-year survival rate was (28.77±11.96)%. The major adverse events were grade Ⅰ-Ⅱ myelosuppression, peripheral neurologic toxicities, nausea and vomiting. Some patients had rash and hepatic dysfunction. A total of 40% of patients experienced flu-like symptoms. Alopecia and diarrhea were rare. Conclusion: The combination of GEM and NVB is an effective and well tolerated regimen for the patients with TNABC.展开更多

In order to ensure the cover-bearing mode, the muddy soil inside the bucket foundation should be reinforced by some soil consolidation methods, such as negative pressure and electro-osmosis. Firstly, tests were conducted to obtain the reasonable current density. Meanwhile, to improve the electro-osmotic speed and effectiveness, other factors such as intermittent power and layout of electrode, were also studied in the tests. Then, the soil reinforcing tests by negative pressure combined with electro-osmosis were performed for the muddy soil consolidation inside the bucket foundation. The results showed that soil reinforcement by negative pressure was quicker and more obvious during the early phase, and electro-osmotic method can affect more range of soil by rational arrangement of electrodes. Compared with negative pressure, the electro-osmotic method was a continuous and relatively slow process of reinforcement, which was complementary to the negative pressure method. The voltage value of electro-osmosis had little effect on the muddy soil reinforcement inside the bucket foundation, and 1.5 A was chosen as the most reasonable current value for scale model testing in the electro-osmotic method.展开更多
The mechanisms of streamer generation and propagation in double-sided pulsed surface dielectric barrier discharge(SDBD)on both sides have been analyzed and investigated by experiment and numerical simulation.The fully...The mechanisms of streamer generation and propagation in double-sided pulsed surface dielectric barrier discharge(SDBD)on both sides have been analyzed and investigated by experiment and numerical simulation.The fully exposed asymmetric SDBD has two discharge processes located on the high voltage electrode(HVE)side and the ground electrode(GE)side.Discharge images of the HVE side and GE side are taken by a digital camera under continuous pulse and ICCD(Intensified Charge Coupled Device)is utilized to diagnose the generation and propagation of streamers in single pulse discharge.In order to understand the physical mechanisms of streamer evolution more deeply,we establish a 2D simulation model and analyze it from the aspects of electron density,ion density,reduced electric field and electron impact ionization source term.The results show that the primary and secondary discharges on the HVE side and the GE side of the double-sided SDBD are composed of positive streamer and negative streamer,respectively.On the HVE side,the accumulation of positive charges on the dielectric surface causes the direction of the electric field to reverse,which is the principal factor for the polarity reversal of the streamer.On the GE side,both the negative charges accumulated on the dielectric surface and the falling voltage are the key factors for the streamer polarity switch.展开更多

OBJECTIVE:To evaluate the therapeutic effect of a selfformulated Traditional Chinese Medicine(TCM),Shugan Jieyu San(疏肝解郁散),on improvement of liver function and depression alleviation in the patients with triple n...OBJECTIVE:To evaluate the therapeutic effect of a selfformulated Traditional Chinese Medicine(TCM),Shugan Jieyu San(疏肝解郁散),on improvement of liver function and depression alleviation in the patients with triple negative breast cancer(TNBC).METHODS:A total of 60 patients diagnosed with TNBC and depression were enrolled and randomly assigned to either the control group(receiving routine tumor treatment and duloxetine)or the treatment group(receiving the TCM in addition to routine treatment).RESULTS:Both treatment and control groups showed a reduction in depressive symptoms and improved daily living abilities after treatment.However,the treatment group demonstrated significantly better outcomes compared to the control group.Furthermore,the levels of dopamine and serotonin in the serum increased in both groups after 8 weeks of treatment,while the treatment group exhibited superior results.CONCLUSIONS:This TCM showed promising results in reducing depressive symptoms and improving daily abilities in patients with TNBC and comorbid depression,which was verified by the increase in serum levels of dopamine and serotonin,suggesting the potential involvement of these neurotransmitters in the therapeutic effects of this TCM.展开更多
BACKGROUND China has recently encountered severe challenges associated with population aging.Parents of first-generation only children face significant challenges regarding elderly care needs and the associated negati...BACKGROUND China has recently encountered severe challenges associated with population aging.Parents of first-generation only children face significant challenges regarding elderly care needs and the associated negative emotions.AIM To analyze the elderly care needs of first-generation only child parents in China and identify factors that influence negative emotions.METHODS This study used a cross-sectional design.Convenience sampling was used to select 1580 elderly individuals who met the inclusion criteria in a Chinese city between June and September 2022.A questionnaire was administered to collect general information about participants.Depression and anxiety were assessed using the patient health questionnaire-9 and generalized anxiety disorder-7 scale,respectively.A logistic regression analysis was performed to evaluate the relevant correlations.RESULTS Among 1580 first-generation only child parents,1120(70.89%)preferred family based care,324(20.51%)opted for community care,and 136(8.61%)chose institutional care,with 460(29.11%)reporting negative emotions.Significant differences in the distribution of negative emotions among only child parents were observed based on age,marital status,living conditions,disability,type of chronic disease,frailty status,and family support(P<0.05).The regression analysis indicated that disability,type of chronic disease,living environment,frailty status,and level of family support were independent risk factors for negative emotions among parents with only children(P<0.05).CONCLUSION Elderly care for parents of only children is primarily family-based.Independent risk factors for negative emotions in this group include disability,chronic disease type,and living environment.展开更多
BACKGROUND Diabetic foot ulcers(DFUs)are a significant challenge in diabetic care,and the efficacy of negative pressure wound therapy(NPWT)in treating them remains a subject of continuous investigation.AIM To provide ...BACKGROUND Diabetic foot ulcers(DFUs)are a significant challenge in diabetic care,and the efficacy of negative pressure wound therapy(NPWT)in treating them remains a subject of continuous investigation.AIM To provide a comprehensive meta-analysis of the role of NPWT in the manage-ment of DFUs.METHODS A systematic review was performed based on Preferred Reporting Items for Sys-tematic Reviews and Meta-Analyses guidelines,searching databases like PubMed,Embase,Web of Science,and the Cochrane Library.Randomized clinical trials(RCTs)were included to compare NPWT to other dressings for DFUs.Outcomes measured were wound healing time and rate,granulation tissue formation time,amputation rate,and adverse events.Study quality was evaluated using Coch-rane's risk of bias tool.Analyses utilizedχ2,I2,fixed or random-effects models via Stata v17.RESULTS Of the 1101 identified articles,9 RCTs were selected for meta-analysis.Studies spanned from 2005 to 2020 and originated from countries including the United States,Chile,Pakistan,Italy,India,and Germany.Meta-analysis demonstrated a significant improvement in wound healing rate[risk ratio(RR)=1.46,95%CI:1.22-1.76,P<0.01]and a reduction in amputation rate(RR=0.69,95%CI:0.50-0.96,P=0.006)with NPWT.Furthermore,the time for granulation tissue formation was significantly reduced by an average of 19.54 days.However,the incidence of adverse events did not significantly differ between NPWT and control treatments.CONCLUSION NPWT significantly improves wound healing rates and reduces amputation rates in DFUs.It also hastens the formation of granulation tissue.However,the therapy does not significantly alter the risk of adverse events compared to alternate treatments.展开更多

文摘Coulomb drag refers to the phenomenon in which a current driven through one conducting layer induces a voltage nearby,electrically isolated layer sorely through interlayer Coulomb interactions between charge carriers.It has been extensively studied in various systems,including parallel nanowires,double quantum wells,and double-layer graphene.Here,we report the observation of Coulomb drag in a novel system consisting of two graphene layers separated laterally by a 30 nm gap within the material plane,exhibiting behavior distinct from that in vertical graphene heterostructures.Our experiments reveal pronounced negative drag resistances under an out-of-plane magnetic field at the quantum Hall edges,reaching a maximum when the carrier densities in both graphene layers are tuned to the charge neutrality point via gate voltages.Our work establish two separate and spatially closed quantum Hall edge modes as a new platform to explore electronic interaction physics between one dimensional systems.
文摘BACKGROUND Anxiety,depression,and other negative emotions are common among patients with chronic renal failure(CRF).Analyzing the factors related to negative emotions is necessary to provide targeted nursing care.AIM To explore the correlations among life satisfaction,pleasure levels,and negative emotions in patients with CRF.METHODS One hundred patients with CRF who received therapy at the First Affiliated Hospital of Jinzhou Medical University between December 2022 and February 2025 were included.The Depression,Anxiety,and Stress Scale(DASS-21),Satisfaction with Life Scale(SWLS),and Temporal Experience of Pleasure Scale(TEPS)were used to evaluate negative emotions,life satisfaction,and pleasure level,respectively.Pearson’s correlation coefficient analyzed the correlation between life satisfaction,pleasure level,and negative emotions.Linear regression analysis identified the factors affecting negative emotions.RESULTS The average DASS-21 score among patients with CRF was 51.90±2.30,with subscale scores of 17.90±1.50 for depression,18.53±1.18 for anxiety,and 15.47±2.36 for stress,all significantly higher than the domestic norm(P<0.05).The average SWLS score was 22.17±4.90.Correlation analysis revealed a negative correlation between the SWLS and total DASS-21 scores(P<0.05),but not with the individual depression,anxiety,or stress dimensions.The average TEPS score was 67.80±8.34.TEPS scores were negatively correlated with the DASS-21 score and the stress dimension(P<0.05),but not with depression or anxiety.Linear regression analysis showed that TEPS scores significantly influenced DASS-21 scores(P<0.05).CONCLUSION Patients with CRF experience high levels of negative emotions,which are negatively correlated with life satisfaction and pleasure.Furthermore,pleasure level had an impact on negative emotions.
文摘Climate change is one of the most pressing global challenges and spares no nation.Bhutan,as the only carbon negative country in the world,is no exception and being in the Himalayas–a region warming faster than the global average–further intensifies climate risks.This study explores how Bhutan navigates climate change through the perspectives of 41 policy influencers,including politicians,civil servants,and leaders from nongovernment organisations and the media.The findings are compared with scientific literature from the broader Himalayan region to identify shared vulnerabilities(e.g.,glacial lake outburst floods,water scarcity,and agricultural disruptions)and governance challenges(e.g.,financial constraints,geopolitical pressures,and gaps in policy implementation).The study reveals that Bhutan,like its Himalayan neighbours,prioritizes adaptation over mitigation due to immediate climate threats,despite its carbonnegative status.However,Bhutan faces unique pressures in upholding its dual commitments—maintaining 60%forest cover and carbon neutrality—amid competing development needs.While its Gross National Happiness(GNH)framework provides a holistic governance model,the analysis uncovers systemic barriers,including limited financing,transboundary climate risks,and uneven policy execution,which mirror regional struggles.The paper contributes to climate governance discourse by highlighting the risks of symbolic environmental pledges without robust implementation mechanisms,particularly for vulnerable mountain nations.It calls for integrated policies that bridge mitigation and adaptation,stronger regional cooperation,and equitable climate financing.The findings are relevant for policymakers in the Himalayas and international stakeholders advocating context-specific,justiceoriented climate strategies.

文摘Detecting geomagnetic anomalies preceding earthquakes is a challenging yet promising area of research that has gained increasing attention in recent years.This study introduces a novel reconstruction-based modeling approach enhanced by negative learning,employing a Bidirectional Long Short-Term Memory(BiLSTM)network explicitly trained to accurately reconstruct non-seismic geomagnetic signals while intentionally amplifying reconstruction errors for seismic signals.By penalizing the model for accurately reconstructing seismic anomalies,the negative learning approach effectively magnifies the differences between normal and anomalous data.This strategic differentiation enhances the sensitivity of the BiLSTM network,enabling improved detection of subtle geomagnetic anomalies that may serve as earthquake precursors.Experimental validation clearly demonstrated statistically significant higher reconstruction errors for seismic signals compared to non-seismic signals,confirmed through the Mann-Whitney U test with a p-value of 0.0035 for Root Mean Square Error(RMSE).These results provide compelling evidence of the enhanced anomaly detection capability achieved through negative learning.Unlike traditional classification-based methods,negative learning explicitly encourages sensitivity to subtle precursor signals embedded within complex geomagnetic data,establishing a robust basis for further development of reliable earthquake prediction methods.

文摘OBJECTIVE:To identify the antitumor effects of Gualou Beimu Yin(瓜蒌贝母饮,GLBMY)in triple-negative breast cancer(TNBC)and to explore the underlying mechanisms.METHODS:A mouse model of breast cancer was established and treated with GLBMY.Freeze-dried GLBMY powder was used to treat MDA-MB-231 and BT549 cells to assess the therapeutic efficacy of GLBMY against TNBC.Network pharmacology,transcriptomics and metabolomics were employed to identify the potential mechanism of GLBMY in TNBC treatment.Finally,the main regulating genes and proteins in the enriched pathways were validated by Quantitative real-time polymerase chain reaction(q PCR)and Western blotting analysis to confirm its mechanism.RESULTS:GLBMY can inhibit the growth of TNBC through apoptosis and necroptosis pathways and inhibit TNBC lung metastasis by inhibiting epithelialmesenchymal transition(EMT).Network pharmacology has elucidated the most important active ingredients(tubeimoside I,emodin,cucurbitacin,and ursolic acid)and the most critical targets[interleukin-6(IL6),signal transducer and activator of transcription 3(STAT3),mitogen-activated protein kinase 3(MAPK3)]of GLBMY in treating TNBC.RNA-seq revealed that GLBMY affected the nuclear factor kappa-light-chain-enhancer of activated B cells,rat sarcoma virus,and MAPK signalling pathways.Metabolomics revealed that the metabolites mainly affected by GLBMY were L-(+)-lactic acid,isocitric acid,benzoic acid and indoxyl sulfate.Subsequent experiments demonstrated that GLBMY can inhibit EMT in TNBC through the MAPK/ERK pathway and inhibit the proliferation and progression of TNBC through the IL6-STAT signalling pathway.CONCLUSIONS:We confirmed that GLBMY inhibits the development and metastasis of TNBC through the MAPK/Erk and IL6-STAT signalling pathways.GLBMY shows promise as a long-term supplementary or alternative therapy for TNBC,offering new insights for TNBC treatment.

文摘Flower color is an essential trait in ornamental plant breeding. Lycoris longituba is a popular ornamental plant native to central eastern China. The decrease in anthocyanin accumulation causes L. longituba petal color fading during flower development, which considerably affects the ornamental value of L. longituba. However, mechanisms underlying anthocyanin biosynthesis inhibition during L. longituba petal development remain unclear. In this study, three LlDFR genes were confirmed to be involved in anthocyanin biosynthesis and LlDFRc exerted the strongest promoting effect on anthocyanin accumulation. According to the correlation analysis results, LlbHLH12 exhibited the strongest negative correlation with LlDFRc. Quantitative real-time PCR analysis showed that LlbHLH12 was highly expressed during the medium bud and full bloom stages of flower development. LlbHLH12 was identified as a member of subgroup XII of bHLH transcription factor family. Subcellular localization and transcriptional activation ability assay revealed that LlbHLH12 was located in the nucleus without transcriptional activation activity. Overexpression of LlbHLH12 in Nicotiana tabacum and L. longituba inhibited anthocyanin accumulation by suppressing the expression of anthocyanin biosynthetic pathway genes. Furthermore, yeast one-hybrid, dual-luciferase, and β-glucuronidase activity assays showed that LlbHLH12 directly bound to the promoters of LlPAL and LlDFRc and suppressed their expression to inhibit anthocyanin biosynthesis. Overall, our study identified a novel bHLH repressor negatively regulating anthocyanin biosynthesis and provided new insights into the molecular mechanisms underlying color fading in L. longituba petals.

文摘In recent years,the rapid advancement of artificial intelligence(AI)technology has enabled AI-assisted negative screening to significantly enhance physicians'efficiency through image feature analysis and multimodal data modeling,allowing them to focus more on diagnosing positive cases.Meanwhile,multispectral imaging(MSI)integrates spectral and spatial resolution to capture subtle tissue features invisible to the human eye,providing high-resolution data support for pathological analysis.Combining AI technology with MSI and employing quantitative methods to analyze multiband biomarkers(such as absorbance differences in keratin pearls)can effectively improve diagnostic specificity and reduce subjective errors in manual slide interpretation.To address the challenge of identifying negative tissue sections,we developed a discrimination algorithm powered by MSI.We demonstrated its efficacy using cutaneous squamous cell carcinoma(cSCC)as a representative case study.The algorithm achieved 100%accuracy in excluding negative cases and effectively mitigated the false-positive problem caused by cSCC heterogeneity.We constructed a multispectral image(MSI)dataset acquired at 520 nm,600 nm,and 630 nm wavelengths.Subsequently,we employed an optimized MobileViT model for tissue classification and performed comparative analyses against other models.The experimental results showed that our optimized MobileViT model achieved superior performance in identifying negative tissue sections,with a perfect accuracy rate of 100%.Thus,our results confirm the feasibility of integrating MSI with AI to exclude negative cases with perfect accuracy,offering a novel solution to alleviate the workload of pathologists.
文摘This study addresses the optimization of automated yarn handling in textile manufacturing by examining the related suction process through a combined numerical and experimental approach.In particular,a three-dimensional model of the suction nozzle was coupled with an equivalent linear-elastic beam representation of the yarn,and a Fluent-IDW-Abaqus weakly coupled fluid-structure interaction(FSI)framework was employed to capture the yarn’s release and dynamic response under negative-pressure suction.High-speed imaging experiments validated the simulations,demonstrating excellent agreement in displacements and velocities.According to the results,increasing the initial suction pressure from -0.04 MPa to -0.06 MPa reduces adsorption time by approximately 62% and markedly dampens yarn-end vibrations,enhancing suction performance.Pressures beyond -0.06 MPa,however,induce overshoot and nozzle collisions,increasing the risk of entanglement and mechanical damage.The outcomes of a statistical analysis are also presented to further quantify the interplay among energy consumption,suction efficiency,and operational success under varying pressures,thereby providing a rigorous foundation for the optimal selection of pressure parameters in automated yarn-handling systems.
文摘The global transition to carbon neutrality is an urgent and multifaceted challenge that requires the deployment of renewable energy technologies and negative emission solutions(NETs)to reduce greenhouse gas emissions across all sectors.This is a review article that looks at the contemporary environment of renewable technologies,such as solar,wind,biomass,hydropower,and geothermal,and how they might help to decarbonize the power sector and their combination with NETs.The paper also looks at the prospects of carbon capture,utilization,and storage,afforestation and reforestation,soil carbon sequestration,ocean-based,and enhanced weathering as some of the methods of offsetting the residual emissions.The article also outlines the economic,policy,and social factors required to have these solutions scaled up,such as the need to have good policy frameworks,invest in innovation,and the need to have the people on board.Lastly,it also gives the future perspective of having a carbon-neutral global economy,and it highlights that technology must be enhanced,more cooperation between countries must be established,and a holistic,open-ended way of attaining carbon neutrality.

文摘Negative Poisson’s ratio materials and structures exhibit lateral expansion under tensile loading,demonstrating significant mechanical advantages over conventional materials.This study systematically investigated three typical two-dimensional negative Poisson’s ratio metamaterial structures(Concave honeycomb,Anti-chiral,and Anti-chiral concave honeycomb hybrid structures)through both experimental tests and numerical analysis.The test specimens were fabricated using selective laser melting(SLM)additive manufacturing technology,and the experimental test was conducted with the use of a DIC strain measurement system.The numerical studies were performed considering both static tensile loading and dynamic impact loading with different strain rates.The deformation behaviors,failure process,negative Poisson’s ratio effects,and energy absorption capacity of the three different metamaterial structures are systematically investigated,and the associated mechanical mechanisms are thoroughly revealed.Results and findings of this work could provide valuable guidance for the engineering design and application of negative Poisson’s ratio metamaterials and structures.

文摘A simplified approach was proposed to analyze the negative skin friction calculation of special-shaped pile considering pile-soil interaction under surcharge. Based on the concentric cylinder shearing theory, considering the changes of pile shape(such as, taper angle and diameters of pile base, etc.), the load-transfer of special-shaped pile was built. The accuracy of the developed simplified approach was verified by numerical simulation model with the same condition. Then, the influence factors, such as, taper angles, the diameter of pile base, surcharge, and pile-soil interface parameters were analyzed and discussed. The results show that the developed simplified approach can calculate NSF of special-shaped pile under surcharge effectively. A limited parametric study indicates that in many practical situations special-shaped piles(such as belled wedge pile shown in this work) offer a design option that is more economical than traditional uniform cross-section piles.

文摘A series of model experiments of bucket foundations concerning suction installation and negative pressure consolidation in saturated silt were carried out in a cube steel bin at Tianjin University. The experimental results show that the silt inside the bucket has been strengthened by negative pressure, and the strengthening effect decreases with the increase of the distance from the bucket. A three-dimensional numerical model of the experiments was built by means of finite element software ABAQUS with fluid-solid coupling method. The results show that the bearing capacity of the silt inside the bucket foundation increases significantly at the former stage of negative pressure consolidation, while the increasing trend slows down over time. The rotation centers of the bucket foundation and the inner soil region tend to be closer to each other based on the consolidation. The bearing capacity of the bucket foundation is improved effectively with the increase of soil strength. The effects of negative pressure consolidation on the bearing capacity of bucket foundation were also illustrated by an actual offshore wind power project case.

文摘Objective: The aim of this study was to evaluate the anti-tumor activity and safety of Gemcitabine (GEM) combined with Vinorelbine (NVB) in patients with advanced TNABC after chemotherapy. Methods: Thirty-seven patients with immunohistochemical proved TNABC were enrolled. The patients received 21-day cycles of NVB 25mg/m^2 i.v. with GEM 1000 mg/m^2 i.v. on days 1 and 8. Results: A total of 136 cycles were given to 37 patients(median 4 cycles, ranged 2-6 cycles). The treatment response was evaluable in all patients. Of the 37 patients, 1 received complete remission (CR), 8 received partial remission (PR), 20 had stable disease (SD), 9 had progressive disease (PD). Overall objective response (CR+ PR) were 24.3 %. The median time to progress (TTP) was 6 months (95% CI, 4-6 months). The median overall survival was 24 months (95% CI, 11-37 months). The median 1-year survival rate was (66.24±8.43)%. The median 3-year survival rate was (28.77±11.96)%. The major adverse events were grade Ⅰ-Ⅱ myelosuppression, peripheral neurologic toxicities, nausea and vomiting. Some patients had rash and hepatic dysfunction. A total of 40% of patients experienced flu-like symptoms. Alopecia and diarrhea were rare. Conclusion: The combination of GEM and NVB is an effective and well tolerated regimen for the patients with TNABC.

文摘Cover-bearing-type bucket foundation for offshore wind turbines has been paid more and more attention due to its low cost and great bearing capacity. In order to ensure the cover-bearing mode, the muddy soil inside the bucket foundation should be reinforced by some soil consolidation methods, such as negative pressure and electro-osmosis. Firstly, tests were conducted to obtain the reasonable current density. Meanwhile, to improve the electro-osmotic speed and effectiveness, other factors such as intermittent power and layout of electrode, were also studied in the tests. Then, the soil reinforcing tests by negative pressure combined with electro-osmosis were performed for the muddy soil consolidation inside the bucket foundation. The results showed that soil reinforcement by negative pressure was quicker and more obvious during the early phase, and electro-osmotic method can affect more range of soil by rational arrangement of electrodes. Compared with negative pressure, the electro-osmotic method was a continuous and relatively slow process of reinforcement, which was complementary to the negative pressure method. The voltage value of electro-osmosis had little effect on the muddy soil reinforcement inside the bucket foundation, and 1.5 A was chosen as the most reasonable current value for scale model testing in the electro-osmotic method.

文摘The mechanisms of streamer generation and propagation in double-sided pulsed surface dielectric barrier discharge(SDBD)on both sides have been analyzed and investigated by experiment and numerical simulation.The fully exposed asymmetric SDBD has two discharge processes located on the high voltage electrode(HVE)side and the ground electrode(GE)side.Discharge images of the HVE side and GE side are taken by a digital camera under continuous pulse and ICCD(Intensified Charge Coupled Device)is utilized to diagnose the generation and propagation of streamers in single pulse discharge.In order to understand the physical mechanisms of streamer evolution more deeply,we establish a 2D simulation model and analyze it from the aspects of electron density,ion density,reduced electric field and electron impact ionization source term.The results show that the primary and secondary discharges on the HVE side and the GE side of the double-sided SDBD are composed of positive streamer and negative streamer,respectively.On the HVE side,the accumulation of positive charges on the dielectric surface causes the direction of the electric field to reverse,which is the principal factor for the polarity reversal of the streamer.On the GE side,both the negative charges accumulated on the dielectric surface and the falling voltage are the key factors for the streamer polarity switch.
文摘An Electromagnetic (EM) radiation in dispersion less free space vacuum is represented by a photon, with corpuscular and wave nature. The discussions, for the past century aimed at the nature of photon inside a media having dispersion in the refraction property, other than free space. What about its nature if the space be of refractive index which is negative, is discussed in this paper. We call mechanical momentum, wave-momentum, and try to match our present theories with intriguing property of this ‘photon’ or pulse carrying EM energy packet, and more so we try to find its property energy, momentum inside a media a positive refractive media, and if the media show a negative refractive index behavior, then these queries are profound, and suitable explanations to these classical concepts of corpuscular-wave nature of photon inside these media are quest for the scientists dealing with these materials having negative index of refraction. Here some of this counterintuitive nature of corpuscular-wave nature of photon inside negative indexed material is brought out, with possible ‘new definition’ of its ‘wave-momentum’, the concept of ‘reactive energy’ inside negative indexed material, along with possible ‘new wave equation’. These definitions and expressions of ‘wave-momentum’ and ‘reactive energy’ pertaining to negative indexed material are new and discussed and derived by classical means.
文摘OBJECTIVE:To evaluate the therapeutic effect of a selfformulated Traditional Chinese Medicine(TCM),Shugan Jieyu San(疏肝解郁散),on improvement of liver function and depression alleviation in the patients with triple negative breast cancer(TNBC).METHODS:A total of 60 patients diagnosed with TNBC and depression were enrolled and randomly assigned to either the control group(receiving routine tumor treatment and duloxetine)or the treatment group(receiving the TCM in addition to routine treatment).RESULTS:Both treatment and control groups showed a reduction in depressive symptoms and improved daily living abilities after treatment.However,the treatment group demonstrated significantly better outcomes compared to the control group.Furthermore,the levels of dopamine and serotonin in the serum increased in both groups after 8 weeks of treatment,while the treatment group exhibited superior results.CONCLUSIONS:This TCM showed promising results in reducing depressive symptoms and improving daily abilities in patients with TNBC and comorbid depression,which was verified by the increase in serum levels of dopamine and serotonin,suggesting the potential involvement of these neurotransmitters in the therapeutic effects of this TCM.

文摘BACKGROUND China has recently encountered severe challenges associated with population aging.Parents of first-generation only children face significant challenges regarding elderly care needs and the associated negative emotions.AIM To analyze the elderly care needs of first-generation only child parents in China and identify factors that influence negative emotions.METHODS This study used a cross-sectional design.Convenience sampling was used to select 1580 elderly individuals who met the inclusion criteria in a Chinese city between June and September 2022.A questionnaire was administered to collect general information about participants.Depression and anxiety were assessed using the patient health questionnaire-9 and generalized anxiety disorder-7 scale,respectively.A logistic regression analysis was performed to evaluate the relevant correlations.RESULTS Among 1580 first-generation only child parents,1120(70.89%)preferred family based care,324(20.51%)opted for community care,and 136(8.61%)chose institutional care,with 460(29.11%)reporting negative emotions.Significant differences in the distribution of negative emotions among only child parents were observed based on age,marital status,living conditions,disability,type of chronic disease,frailty status,and family support(P<0.05).The regression analysis indicated that disability,type of chronic disease,living environment,frailty status,and level of family support were independent risk factors for negative emotions among parents with only children(P<0.05).CONCLUSION Elderly care for parents of only children is primarily family-based.Independent risk factors for negative emotions in this group include disability,chronic disease type,and living environment.

文摘BACKGROUND Diabetic foot ulcers(DFUs)are a significant challenge in diabetic care,and the efficacy of negative pressure wound therapy(NPWT)in treating them remains a subject of continuous investigation.AIM To provide a comprehensive meta-analysis of the role of NPWT in the manage-ment of DFUs.METHODS A systematic review was performed based on Preferred Reporting Items for Sys-tematic Reviews and Meta-Analyses guidelines,searching databases like PubMed,Embase,Web of Science,and the Cochrane Library.Randomized clinical trials(RCTs)were included to compare NPWT to other dressings for DFUs.Outcomes measured were wound healing time and rate,granulation tissue formation time,amputation rate,and adverse events.Study quality was evaluated using Coch-rane's risk of bias tool.Analyses utilizedχ2,I2,fixed or random-effects models via Stata v17.RESULTS Of the 1101 identified articles,9 RCTs were selected for meta-analysis.Studies spanned from 2005 to 2020 and originated from countries including the United States,Chile,Pakistan,Italy,India,and Germany.Meta-analysis demonstrated a significant improvement in wound healing rate[risk ratio(RR)=1.46,95%CI:1.22-1.76,P<0.01]and a reduction in amputation rate(RR=0.69,95%CI:0.50-0.96,P=0.006)with NPWT.Furthermore,the time for granulation tissue formation was significantly reduced by an average of 19.54 days.However,the incidence of adverse events did not significantly differ between NPWT and control treatments.CONCLUSION NPWT significantly improves wound healing rates and reduces amputation rates in DFUs.It also hastens the formation of granulation tissue.However,the therapy does not significantly alter the risk of adverse events compared to alternate treatments.

文摘Necrotizing fasciitis(NF)is a rapidly progressing,life-threatening soft tissue infection,with upper limb NF posing a particularly serious threat to patient survival and quality of life.Negative pressure wound therapy(NPWT)has shown considerable advantages in accelerating wound healing and mitigating functional impairment.A retrospective study by Lipatov et al.demonstrated that NPWT significantly reduced the time needed for wound closure preparation while enhancing the success rate of local repair.Despite its benefits,certain limitations highlight the need for further optimization.This paper investigates the potential for personalized dynamic regulation of NPWT,its integration with adjunctive therapies,and the role of multidisciplinary collaboration.Furthermore,it explores the incorporation of advanced technologies such as artificial intelligence,imaging modalities,and biomaterials,presenting novel pathways for the personalized management and global standardization of NF treatment.
