Border-associated macrophages are located at the interface between the brain and the periphery, including the perivascular spaces, choroid plexus, and meninges. Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment. 展开更多

The endpoint carbon content in the converter is critical for the quality of steel products, and accurately predicting this parameter is an effective way to reduce alloy consumption and improve smelting efficiency. However, most scholars currently focus on modifying methods to enhance model accuracy, while overlooking the extent to which input parameters influence accuracy. To address this issue,in this study,a prediction model for the endpoint carbon content in the converter was developed using factor analysis(FA)and support vector machine(SVM)optimized by improved particle swarm optimization(IPSO).Analysis of the factors influencing the endpoint carbon content during the converter smelting process led to the identification of 21 input parameters.Subsequently,FA was used to reduce the dimensionality of the data and applied to the prediction model. The results demonstrate that the performance of the FA-IPSO-SVM model surpasses several existing methods, such as twin support vector regression and support vector machine. The model achieves hit rates of 89.59%,96.21%,and 98.74%within error ranges of±0.01%,±0.015%,and±0.02%,respectively.Finally,based on the prediction results obtained by sequentially removing input parameters, the parameters were classified into high influence(5%-7%),medium influence(2%-5%),and low influence(0-2%)categories according to their varying degrees of impact on prediction accuracy. This classi-fication provides a reference for selecting input parameters in future prediction models for endpoint carbon content. 展开更多

BACKGROUND Endoscopic submucosal dissection(ESD)is a minimally invasive, safe, and efficient treatment technique for patients diagnosed with early esophageal cancer. However, postoperative disease recurrence remains an important clinical challenge because it negatively alters patient prognosis and quality of life.As such, identification of relevant risk factors for recurrence can help optimize postoperative management strategies.AIM To assess factors that contribute to the risk for disease recurrence after ESD for early esophageal cancer.METHODS Clinical data from 210 patients diagnosed with early stage esophageal cancer, who underwent ESD at the authors’center between March 2012 and March 2025,were retrospectively collected and analyzed.Patients were categorized into 2 groups according to postoperative disease recurrence:Recurrence(n=30),and without recurrence(n=180).Disease recurrence was defined as the appearance of new tumor lesions or pathologically confirmed tumor recurrence during the postoperative follow-up period.Risk factors associated with postoperative recurrence were identified using univariate and multivariate logistic regression analyses.RESULTS During the follow-up period,30 patients experienced tumor recurrence, corresponding to a recurrence rate of 14.19%.Multivariate analysis revealed that poor differentiation was a significant potential cause of esophageal cancer recurrence[odds ratio(OR)=1.782,95%confidence interval(CI):1.154-2.196;P<0.001].Tumors infiltrating the submucosa were more likely to recur than those penetrating the lamina propria or muscularis mucosa(OR=1.573,95%CI:1.073-2.481;P<0.001).Furthermore,inability to completely resect the tumor greatly increased the likelihood of recurrence(OR=2.189,95%CI:1.193-3.125;P=0.001).Tumor diameter≥2 cm was an independent risk factor for postoperative recurrence(OR=1.981,95%CI:1.482-2.862;P=0.005).CONCLUSION Recurrence of early esophageal cancer after ESD is largely influenced by the degree of differentiation, depth of lesion invasion, complete resection status of the tumor, and tumor diameter. 展开更多

BACKGROUND Dental implants are widely used to replace missing teeth.Currently, clinicians assess osseointegration success by measuring the implant’s stability within the bone and monitoring the marginal tissue height.Diabetes, especially type 2 diabetes mellitus(T2DM),has been reported to impair implant healing, drastically reducing implant success rates.AIM To analyze the high-risk factors for inflammatory response and prognosis after dental implantation in patients with T2DM,and provide strong evidence for reducing the incidence of postimplant peri-implantitis(PI).METHODS We performed a retrospective review of 146 patients with T2DM who had dental implants placed at Tianjin Fifth Central Hospital, between September 2021 and September 2023,which was regarded as the observation group.Moreover,60 ageand gender-matched individuals with normal blood glucose levels served as the control group. The general information, postoperative periodontal indices, and levels of inflammatory factors were comprehensively analyzed and compared.Furthermore, the incidence of postimplant PI was counted, and multivariate logistic regression was used to identify the determinants of postimplant PI.RESULTS In terms of the periodontal indices, the probing depth, modified sulcus bleeding index, and marginal bone loss in the observation cohorts began to increase significantly at 6 months and 3 months, respectively, after the completion of dental implant restoration. The T2DM cases demonstrated significantly elevated counts of leukocytes, lymphocytes, and neutrophils compared to the controls at 24 hours postoperatively. Moreover, the TNF-α,IL-1β,and IL-6 concentrations started to increase significantly in the gingival crevicular fluid 3 months after the completion of dental implant restoration in both cohorts, with the observation group exhibiting higher levels than the controls at each time point.63 out of the 146 cases developed PI.Multivariate logistic regression analysis indicated that high glycosylated hemoglobin levels, smoking, daily tooth-brushing frequency of less than once, and the anterior tooth as the implant site independently contributed to postimplant PI in T2DM cases, while a tooth-brushing duration of≥3 minutes was a protective factor.CONCLUSION Patients with T2DM are at risk of developing PI following dental implantation.Clinically,it is necessary to enhance the identification of risk factors for postimplant PI,improve risk prediction, prevention, and control, and formulate targeted intervention countermeasures to reduce the occurrence of postimplant PI. 展开更多

To assess the deformation of a coal bunker and propose effective preventative measures for such,a real-time monitoring system was designed. Moreover, methods were proposed for monitoring the coal-bunker inclination, settlement, groundwater level, temperature, and material level.By using a vector autoregression model and time-series data for the coal-bunker deformation,a long-term equilibrium relationship was derived between the coal-bunker inclination and settlement and their influencing factors. The dynamic effects of each factor on the coal-bunker inclination and settlement and how the contribution of each factor changes were revealed by using an impulse response function and variance decomposition. The results show that the coal-bunker inclination and settlement vary hysteretically.Groundwater level has constant and small negative effects on the coal-bunker inclination and settlement, and temperature has continuous and significant negative effects;however, material level has constant positive effects on the coal-bunker settlement and slightly influences the eastward inclination while significantly influencing the northward inclination of the coal bunker. Also, unbalanced loading has little influence on the coal-bunker settlement and eastward inclination but significant influence on its northward inclination. Therefore, monitoring of the coal-bunker deformation should be strengthened in summer, and sudden changes in material level and unbalanced loading should be avoided during production. Moreover, the coal-bunker inclination can be adjusted by unbalanced loading in the reverse direction. 展开更多

Multicollinearity in factor analysis has negative effects, including unreliable factor structure, inconsistent loadings, inflated standard errors, reduced discriminant validity, and difficulties in interpreting factors. It also leads to reduced stability, hindered factor replication, misinterpretation of factor importance, increased parameter estimation instability, reduced power to detect the true factor structure, compromised model fit indices, and biased factor loadings. Multicollinearity introduces uncertainty, complexity, and limited generalizability, hampering factor analysis. To address multicollinearity, researchers can examine the correlation matrix to identify variables with high correlation coefficients. The Variance Inflation Factor (VIF) measures the inflation of regression coefficients due to multicollinearity. Tolerance, the reciprocal of VIF, indicates the proportion of variance in a predictor variable not shared with others. Eigenvalues help assess multicollinearity, with values greater than 1 suggesting the retention of factors. Principal Component Analysis (PCA) reduces dimensionality and identifies highly correlated variables. Other diagnostic measures include the condition number and Cook’s distance. Researchers can center or standardize data, perform variable filtering, use PCA instead of factor analysis, employ factor scores, merge correlated variables, or apply clustering techniques for the solution of the multicollinearity problem. Further research is needed to explore different types of multicollinearity, assess method effectiveness, and investigate the relationship with other factor analysis issues. 展开更多

BACKGROUND: In the early period of orthotopic liver transplantation (OLT), initial poor graft function (IPGF) is one of the complications which leads to primary graft non-function (PGNF) in serious cases. This study set out to establish the clinical risk factors resulting in IPGF after OLT. METHODS: Eighty cases of OLT were analyzed. The IPGF group consisted of patients with alanine aminotransferase (ALT) and/or aspartate aminotransferase (AST) above 1500 IU/L within 72 hours after OLT, while those in the non-IPGF group had values below 1500 IU/L. Recipient-associated factors before OLT analyzed were age, sex, primary liver disease and Child-Pugh classification; factors analyzed within the peri-operative period were non-heart beating time (NHBT), cold ischemia time (CIT), rewarming ischemic time (RWIT), liver biopsy at the end of cold ischemia; and factors analyzed within 72 hours after OLT were ALT and/or AST values. A logistic regression model was applied to filter the possible factors resulting in IPGF. RESULTS: Donor NHBT, CIT and RWIT were significantly longer in the IPGF group than in the non-IPGF group; in the logistic regression model, NHBT was the risk factor leading to IPGF (P < 0.05), while CIT and RWIT were possible risk factors. In one case in the IPGF group, PGNF appeared with moderate hepatic steatosis. CONCLUSIONS: Longer NHBT is an important risk factor leading to IPGF, while serious steatosis in the donor liver, CIT and RWIT are potential risk factors. 展开更多

An unsaturated clay slope, with various sloping angles and a thickness of 14 m, consists of backfill, slope soil and residual soil. Slide interfaces were determined by geophysical approaches and the original slope was reconstructed. Sub-slope masses were classified based on the varieties of sloping angle. A force recursive principle was proposed to calculate the stability coefficient of the sub-slope masses. The influencing factors such as sloping angle, water content, hydrostatic pressure, seismic force as well as train load were analyzed. The range and correlation of the above-mentioned factors were discussed and coupled wave equations were established to reflect the relationships between unit weight, cohesion, internal frictional angle, and water content, as well as between internal frictional angle and cohesion. The sensitivity analysis of slope stability was carried out and susceptive factors were determined when the factors were taken as independent and dependent variables respectively. The results show that sloping angle, water content and earthquake are the principal susceptive factors influencing slope stability. The impact of hydrostatic pressure on slope stability is similar to the seismic force in quantity. Train load plays a small role in slope stability and its influencing only reaches the roadbed and its neighboring slope segment. If the factors are taken as independent variables, the influencing extent of water content and cohesion on slope stability can be weakened and train load can be magnified. 展开更多

Objective: To determine the independent prognostic factors in the recurrence of colonic carcinoma after curative resection. Methods: Two hundred and one patients undergoing curative resections for colonic carcinoma were investigated by univariate and Cox multivariate regression analyses. Ten factors contributed to the rate were analyzed. Results: Dukes stages, obstruction, postoperative chemotherapy as well as the growth manner of the tumor were significantly associated with the recurrence rate of colonic carcinoma (P<0.05) by univariate analysis, while Dukes stages, obstruction, and postoperative chemotherapy were significant factors by the multivariate analysis. Conclusion: Dukes stages, obstruction, and postoperative chemotherapy are independent prognostic factors in the recurrence of colonic carcinoma. 展开更多
