目的:构建含MoMLVgag-pol基因的重组表达载体,实现其在NIH3T3细胞中稳定表达。方法:应用RT-PCR方法反转录并扩增gag-pol基因,克隆入真核表达载体pcDNA4/HisMaxA上,构建重组表达载体pcDNA4/HisMaxA-gag-pol,用脂质体法转染NIH3T3细胞,Zeo... 目的:构建含MoMLVgag-pol基因的重组表达载体,实现其在NIH3T3细胞中稳定表达。方法:应用RT-PCR方法反转录并扩增gag-pol基因,克隆入真核表达载体pcDNA4/HisMaxA上,构建重组表达载体pcDNA4/HisMaxA-gag-pol,用脂质体法转染NIH3T3细胞,Zeocin筛选稳定表达细胞株,通过SDS-PAGE分析检测表明,gag-pol基因在NIH3T3细胞实现了表达,产物相对分子质量(kDa)为194.78×103。然后,将逆转录病毒载体导入此细胞系,包装逆转录病毒。用PCR与标记基因补救分析法检测野生型辅助病毒。结果:酶切鉴定的片段大小分别为5.2kb,与预期大小一致,经Zeocin筛选后获得稳定表达细胞株,SDS-PAGE实验表明产物融合蛋白相对分子质量(kDa)为194.78×103,与预期相符。脂质体转染包装细胞,嘌呤霉素加压筛选出高病毒滴度(4.0×106CFU/ml)的细胞克隆,且未检测到辅助病毒。结论:构建的融合表达载体pcDNA4/HisMaxA-gag-pol及其在NIH3T3细胞中的表达,构建成功具有靶向性的逆转录病毒包装细胞系,该细胞系能够包装出高滴度的逆转录病毒,为肝细胞的基因治疗提供了一种新的基因转移系统。 展开更多

通过光学显微镜、扫描电镜观察及能谱分析等方法,分析研究了叠层片式压敏电阻器(MLV,mutilayer varistor)的微观结构.实验结果表明,在半成品叠层片式压敏电阻器的微观结构中ZnO晶粒较小且不均匀,结构较疏松,强度差.而成品由于... 通过光学显微镜、扫描电镜观察及能谱分析等方法,分析研究了叠层片式压敏电阻器(MLV,mutilayer varistor)的微观结构.实验结果表明,在半成品叠层片式压敏电阻器的微观结构中ZnO晶粒较小且不均匀,结构较疏松,强度差.而成品由于进行了低温液相烧结,其微观结构则为较规则的ZnO晶粒,晶粒变大尺寸较为均匀,致密度明显提高.由于在陶瓷叠片上内电极浆料分布不均匀或电极印刷的原因,经过烧结后个别地方可能出现内电极层的缺损,以至观察到的内电极是断续的。同时观察到空洞可影响到内电极扣外电极的连接,以及电极变形等现象。这些缺陷结构的形成与叠层片式压敏电阻器的制作工艺有关. 展开更多

Biological Nitrogen and COD Removal of Nutrient-Rich Wastewater Using Aerobic and Anaerobic Reactors

A preliminary study on nitrogen and organic removal efficiency of a lab-scale system using aerobic and an-aerobic reactors was performed. A simulated wastewater containing elevated levels of nitrogen was used. This paper aims to compare the efficiency of aerobic and anaerobic reactors in achieving nitrogen and chemical oxygen demand (COD) removal of nutrient-rich wastewater. It also presents the start-up experi-mentation conducted on simulated wastewater using two different reactors configured as aerobic and anaero-bic. Start-up experiments were carried out using a 5-liter acrylic aerobic reactor and a 4-liter flask anaerobic reactor containing activated sludge taken from De La Salle University (DLSU) wastewater treatment plant as a source of inoculum. Simulated wastewater was continuously fed to the two reactors and the time course of biomass growth was monitored by measuring the biomass concentration represented by mixed liquor volatile solids (MLVS). The time course of organic pollutant reduction by measuring the chemical oxygen demand (COD) was conducted until steady state condition was reached. On the other hand, COD and nitrogen tests such as Ammonia nitrogen (NH3-N), Nitrite nitrogen (NO2--N), Nitrate nitrogen (NO3--N) were also per-formed using 5 batch aerobic reactors containing different concentrations of wastewater and a single batch anaerobic reactor to see the effect of different feed concentrations in the removal of nitrogen. Preliminary results showed that 98% reduction in COD was obtained in aerobic reactor, as supported by increasing con-centration of MLVS, with a hydraulic retention time (HRT) of 5 hours after 11 days while 34% reduction in COD was obtained in anaerobic reactor with the same HRT after 14 days. 展开更多

为了解山东滨州某猪场PRRSV的流行及变异情况,将该猪场采集的疑似PRRSV阳性病料进行RT-PCR检测,检测出的阳性样品用Marc-145细胞进行分离,设计了9对扩增引物用于分离株(BZ4-19)的全基因扩增,将扩增出的全基因序列进行核苷酸及其推导的... 为了解山东滨州某猪场PRRSV的流行及变异情况,将该猪场采集的疑似PRRSV阳性病料进行RT-PCR检测,检测出的阳性样品用Marc-145细胞进行分离,设计了9对扩增引物用于分离株(BZ4-19)的全基因扩增,将扩增出的全基因序列进行核苷酸及其推导的氨基酸序列分析。最终成功在Marc-145细胞上分离到1株RespPRRS MLV-like毒株,全基因扩增结果显示其全长为15412 bp(不含polyA尾);核苷酸同源性分析说明,其与美洲经典株VR2332及RespPRRS MLV(疫苗株)同源性最高,分别为99.4%和99.8%。将BZ4-19分离株与VR2332、RespPRRS MLV的氨基酸序列进行比较分析揭示,VR2332与RespPRRS MLV共有30处氨基酸差异,在这30处差异中,分离株与VR2332有7处一致,与RespPRRS MLV有23处一致。在鉴别VR2332与RespPRRS MLV的9处可能与毒力相关的氨基酸位点时,分离株有7处与VR2332相同,由此推测本研究分离到的毒株属于疫苗株RespPRRS MLV返强毒株,我国具有上述分子变化的PRRSV的流行应被关注。 展开更多

Porcine reproductive and respiratory syndrome(PRRS)is one of the most significant diseases affecting the pig industry worldwide.The PRRSV mutation rate is the highest among the RNA viruses.To date,NADC30-like PRRSV and highly pathogenic PRRSV(HP-PRRSV)are the dominant epidemic strains in China;however,commercial vaccines do not always provide sufficient cross-protection,and the reasons for insufficient protection are unclear.This study isolated a wild-type NADC30-like PRRSV,SX-YL1806,from Shaanxi Province.Vaccination challenge experiments in piglets showed that commercial modified live virus(MLV)vaccines provided good protection against HP-PRRSV.However,it could not provide sufficient protection against the novel strain SXYL1806.To explore the reasons for this phenomenon,we compared the genomic homology between the MLV strain and HP-PRRSV or NADC30-like PRRSV and found that the MLV strain had a lower genome similarity with NADC30-like PRRSV.Serum neutralization assay showed that MLV-immune serum slightly promoted the homologous HP-PRRSV replication and significantly promoted the heterologous NADC30-like PRRSV strain replication in vitro,suggesting that antibody-dependent enhancement(ADE)might also play a role in decreasing MLV protective efficacy.These findings expand our understanding of the potential factors affecting the protective effect of PRRSV MLV vaccines against the NADC30-like strains. 展开更多

本试验旨在深入了解如何改善商品化疫苗的免疫程序来防制猪繁殖与呼吸综合征病毒,作者采用灭活疫苗(INV)、改性活疫苗(MLV)或INV和MLV联合免疫,制定不同的免疫程序并对免疫效果进行比较。试验1的分组、免疫及攻毒情况为:21只仔猪平均分... 本试验旨在深入了解如何改善商品化疫苗的免疫程序来防制猪繁殖与呼吸综合征病毒,作者采用灭活疫苗(INV)、改性活疫苗(MLV)或INV和MLV联合免疫,制定不同的免疫程序并对免疫效果进行比较。试验1的分组、免疫及攻毒情况为:21只仔猪平均分为3组,试验A组在2.5、3.5和6.5月龄时免疫商品化的INV疫苗;试验B组在1.5、2.5、5.5和6.5月龄时免疫INV疫苗;试验C组为对照组,不做任何免疫。在7.5月龄时,所有试验猪都接种PRRSV并观察25d。试验2的分组、免疫及攻毒情况为:32只仔猪平均分为4组,试验A、B、C 3组在1.5月龄时免疫商品化的MLV疫苗,而试验D组作为对照。4.5月龄时,试验A和C组免疫INV疫苗,而试验B组猪免疫MLV,30d后,试验C组猪再次免疫INV疫苗。在6.5月龄时,所有猪按照试验1进行病毒接种。作者评价了这2个试验的总抗体、中和抗体(NA)和细胞介导的免疫反应(CMI),攻毒后测定病毒血症。试验1结果显示,在第2次及后续免疫后,免疫INV疫苗可诱导高IFN-γ反应。与未免疫对照组相比,攻毒后免疫INV疫苗猪中和抗体产生的更快。试验2结果显示,再次免疫INV疫苗诱导的中和抗体反应与再次免疫MLV引起的反应类似,然而,只有C组在攻毒后中和抗体滴度显著增加。联合免疫程序(MLN+INV)诱导细胞介导的免疫反应优于单独免疫MLV疫苗。试验结果表明,首次免疫时,MLV疫苗可以诱导防制PRRSV的最高免疫反应,此后,INV疫苗的再次免疫被认为是诱导显著增强作用的最好选择。 展开更多

Study for Interrelationship Between Human Herpes Simplex Virus Type 2 and Leukemia

Two different strains of human herpes simplex virus type 2 (HHSV-2) and /or murine leukemia virus (MLV) were inoculated into newborn mice. In mice of mixed infection with MLV plus HHSV-2(333) or MLV plus HHSV-2(Wu)incidences of leukemia were respectively 79.16% and 75.00%,and in those inoculated with MLV alone were 41.02% and 44.74% in the experiment for 3 months. The differences of incidences between the two conditions were highly significant (p<0.01). On the contrary, no leukemia was detected in mice that were inoculated with HHSV-2 (333) or HHSV-2 (Wu) alone and in controls. Leukemia developing time of mice infected with two viruses was earlier than with MLV alone. It is obvious that the induction of murine leukemia resulted from infection of MLV and was independent of HHSV-2 (333) and HHSV-2(Wu). Both risen incidences and shortened latencies of murine leukemia were in close relationship with infection of HHSV-2(333) and HHSV-2(Wu). 展开更多

病毒的有效清除是生物技术中首先关注的问题。由于净化设备上只是简单地附加新单元操作的做法带来的是产品低效回收和生产成本的增加 ,因而在现存的操作中有效清除病毒是一个非常值得研究的课题。为此研究了由絮凝和超滤法从鼠科白血病... 病毒的有效清除是生物技术中首先关注的问题。由于净化设备上只是简单地附加新单元操作的做法带来的是产品低效回收和生产成本的增加 ,因而在现存的操作中有效清除病毒是一个非常值得研究的课题。为此研究了由絮凝和超滤法从鼠科白血病毒(murineleukemiavirus ,MLV)和老鼠微小病毒 (minutevirusofmice ,MVM )中清除中国鼠卵巢病毒 (CHO)悬浮物。这些病毒是被美国食品和药物管理部推荐清除的。MLV是包膜病毒 ,MVM是非包膜病毒。MLV的粒径在 80~ 1 3 0nm ,MVM的粒经在 1 8~ 2 4nm。超滤中 ,进料流中含有这些病毒的CHO细胞成份。先采用两种阳离子絮凝剂来絮凝进料悬浮物 ,然后进行中空纤维膜过滤。渗滤物病毒清除水平由TCID5 0分析法确定。用孔径为 0 .1 μm的膜研究病毒清理。 1 0 5倍过量的MVM病毒的清理由絮凝法完成 ,而 1 0 0倍过量的低量清理由非絮凝法完成。同时获得大约 1 0 0 0倍过量的MLV的清理。然而絮凝剂的加入引起过量的渗滤流。 展开更多

Infectious agents causing aborted fetus problems in domestic pigs were investigated in this study. More than 10 different infectious agents were known to cause abortion in swine and the major eight viruses among them were inspected. One hundred twelve samples of aborted fetuses from nine provinces in South Korea were collected during April to November, 2013 in this study for the diagnosis of infectious agents causing abortions in pigs. Eight major infection viruses were examined in this study mainly using various diagnostic kits and reverse transcriptase polymerase chain reaction (RT-PCR). Positive rate of the detection differed from each viruses. In this study, the main focus was the porcine reproductive and respiratory syndrome virus (PRRSV), which took the second large portion in the positive rate of detection, and then its ORF5 gene was compared with modified live virus (MLV) vaccine strain to figure out the influence of vaccine on disease. Between four positive samples' sequence, two of them were 99.9%-100% similar to MLV vaccine strain and two other samples were 88.6%-92.7% similar. Similarity rate of the sequences between the vaccine and virus from aborted fetuses are very crucial, because it implies that abortion in swine can be made due to the usage of vaccine not only by the infection of field virus, and if MLV vaccine actually do have an impact on the infection, usage of the vaccine should be reconsidered. 展开更多

猪繁殖与呼吸综合征病毒通过阻碍STAT1/STAT2核转位抑制Ⅰ型干扰素的信号转导

Ⅰ型干扰素(IFNs),IFN-α/β诱导抗病毒应答,在天然免疫抗病毒感染中起着重要作用。猪繁殖与呼吸综合征病毒(PRRSV)抑制Ⅰ型干扰素的合成,但其是否抑制IFN的信号转导尚未明确。本研究结果发现PRRSV干扰IFN的信号转导通路。用PRRSV VR2385株感染MARC-145细胞,经IFN-α处理的感染细胞中,干扰素刺激基因ISG15和ISG56的转录、信号转导蛋白及转录激活因子(STAT)2水平均显著低于未经过IFN-α处理过的对照感染细胞。在PRRSV感染的细胞中,IFN诱导的STAT1和STAT2磷酸化及其异源二聚体的形成都未受影响,但大部分的STAT1/STAT2/IRF9的三聚体仍存在于PRRSV感染细胞的细胞质中,表明三聚体的核易位受到了阻碍。PRRSV VR2385感染的细胞中过度表达的NSP1β抑制了ISG15和ISG56的表达,并阻止STAT1的核转位,这些都表明NSP1β可能是抑制IFN信号转导的病毒蛋白。PRRSV感染的猪肺泡巨噬细胞(PAMs)也抑制INF-α刺激的ISG基因和STAT2蛋白的表达。相反,已获批上市的的低致病性疫苗株Ingelvac PRRSV MLA感染PAMs,不需要添加外源的IFN就能活化包括趋化因子和抗病毒细胞因子等IFN诱导基因的表达,且检测不出其对IFN的信号转导产生影响。这些发现都说明PRRSV通过阻碍ISGF3的核转位阻止Ⅰ型干扰素的活化和信号转导通路。 展开更多

When the signal is noised by the multiplicative noise, it is difficult to remove. The paper presents an improved filter to remove multiplicative noise by changing the multiplicative noise to the additive noise, and then using the MLV-like to remove the additive noise. The simulation results show that the performance is better than Minimum Coefficient of Variation (MCV) filter and MLV filter. Both one-dimension and image experiments demonstrate its theoretical performance. 展开更多

Laccases stabilization with phosphatidylcholine liposomes

Enzymes are globular proteins whose catalytic function is due to their three dimensional structure. For this reason, stability strategies make use of compounds that avoid dismantling or distorting protein 3D structures. This study is concerned with the use of microencapsulation techniques to optimize enzyme stabilization. Laccases were embedded in phophatidylcholine liposomes and their encapsulation capacity was assessed. Their enzymatic activity and stability were analyzed, comparing free-enzymes, enzymes in liposomes, and the lipid fraction separated from the aqueous fraction. An increase in their encapsulation efficiency was found at higher lipid/laccase ratios. Relative activity of enzyme-containing vesicles has also been shown to be retained much more than that of free native enzymes. The loss of activity of laccases entrapped in the vesicles in the total stability process is lower than 10% compared with 40% to 60% of loss of free-laccases after heating the samples for 3 days. Laccase stabilization could be of interest to future textile or cosmetic applications because of their potential for environmentally friendly oxidation technologies. 展开更多
