MicroRNA-29c表达与胃癌临床病理特征的关系研究

目的:探讨miR-29c的表达与胃癌临床病理特征之间的关系,预测miR-29c靶基因生物学功能。方法:从癌症基因组图谱(TCGA)数据库下载胃癌微小RNA表达及临床数据,分析miR-29c在胃癌及正常胃黏膜组织中的表达情况。收集胃癌及正常组织石蜡标本... 目的:探讨miR-29c的表达与胃癌临床病理特征之间的关系,预测miR-29c靶基因生物学功能。方法:从癌症基因组图谱(TCGA)数据库下载胃癌微小RNA表达及临床数据,分析miR-29c在胃癌及正常胃黏膜组织中的表达情况。收集胃癌及正常组织石蜡标本,制作成组织芯片,使用miRNAscope技术检测miR-29c在胃癌及正常组织中的表达情况,通过卡方检验分析miR-29c表达与胃癌临床病理特征的关系。TargetScan、Pictar、miRTarbase和Starbase数据库预测miR-29c的靶基因,并对潜在靶基因进行富集分析。结果:TCGA数据分析显示,与正常组织相比(n=41),miR-29c在胃癌组织(n=434)中表达显著下调(P<0.001)。miRNAscope检测结果显示,与正常组织相比,miR-29c在胃癌组织中表达下调(P<0.001),其表达水平与胃癌患者TNM分期、淋巴结转移、组织学分型显著相关(P<0.05)。数据库预测得到18个潜在靶基因,主要富集在DNA甲基化转移酶活性等生物学功能,以及癌症相关miRNAs等信号通路。结论:miR-29c在胃癌组织中低表达,与胃癌患者TNM分期、淋巴结转移、组织学分型相关,有望成为判断胃癌预后的潜在标记物及分子治疗的新靶点。 展开更多

Objective:To explore the relationship between the expression of miR-29c and clinicopathological characteristics of gastric cancer and predict the biological functions of target genes of miR-29c.Methods:The expression and clinical data of microRNA of gastric cancer were downloaded from The Cancer Genome Atlas(TCGA)to analyze the expression of miR-29c in gastric cancer tissues and normal gastric mucosal tissues.Paraffin specimens of gastric cancer and normal tissues were collected to make tissue microarray.The expression of miR-29c in gastric cancer tissues and normal tissues were detected by miRNAscope technique.The relationship between the expression of miR-29c and clinicopathological characteristics of gastric cancer was analyzed by chi-square test.Target genes of miR-29c were predicted by TargetScan,Pictar,miRTarbase and Starbase databases,and enrichment analysis were performed on potential target genens.Results:TCGA analysis showed that the expression of miR-29c was significantly lower in gastric cancer tissue(n=434)than normal tissues(n=41,P<0.001).miRNAscope analysis showed that the expression of miR-29c was significantly lower in gastric cancer tissue than normal tissues(P<0.001),and was significantly associated with TNM staging,lymph node metastasis and histological type of gastric cancer patients(P<0.05).Database predicted 18 potential target genes that were enriched in DNA methyltransferase activity and cancer-associated miRNA signal pathways.Conclusion:miR-29c was expressed at a low level in gastric cancer tissue and associated with TNM staging,lymph node metastasis and histological type,and could become a potential prognosis marker of gastric cancer and a new target of molecular treatment. 展开更多
