Sudden and unforeseen seismic failures of coal mine overburden(OB)dump slopes interrupt mining operations,cause loss of lives and delay the production of coal.Consideration of the spatial heterogeneity of OB dump materials is imperative for an adequate evaluation of the seismic stability of OB dump slopes.In this study,pseudo-static seismic stability analyses are carried out for an OB dump slope by considering the material parameters obtained from an insitu field investigation.Spatial heterogeneity is simulated through use of the random finite element method(RFEM)and the random limit equilibrium method(RLEM)and a comparative study is presented.Combinations of horizontal and vertical spatial correlation lengths were considered for simulating isotropic and anisotropic random fields within the OB dump slope.Seismic performances of the slope have been reported through the probability of failure and reliability index.It was observed that the RLEM approach overestimates failure probability(P_(f))by considering seismic stability with spatial heterogeneity.The P_(f)was observed to increase with an increase in the coefficient of variation of friction angle of the dump materials.Further,it was inferred that the RLEM approach may not be adequately applicable for assessing the seismic stability of an OB dump slope for a horizontal seismic coefficient that is more than or equal to 0.1. 展开更多

Accurate prediction of coal and gas outburst(CGO)hazards is paramount in gas disaster prevention and control.This paper endeavors to overcome the constraints posed by traditional prediction indexes when dealing with CGO incidents under low gas pressure conditions.In pursuit of this objective,we have studied and established a mechanical model of the working face under abnormal stress and the excitation energy conditions of CGO,and proposed a method for predicting the risk of CGO under abnormal stress.On site application verification shows that when a strong outburst hazard level prediction is issued,there is a high possibility of outburst disasters occurring.In one of the three locations where we predicted strong outburst hazards,a small outburst occurred,and the accuracy of the prediction was higher than the traditional drilling cuttings index S and drilling cuttings gas desorption index q.Finally,we discuss the mechanism of CGO under the action of stress anomalies.Based on the analysis of stress distribution changes and energy accumulation characteristics of coal under abnormal stress,this article believes that the increase in outburst risk caused by high stress abnormal gradient is mainly due to two reasons:(1)The high stress abnormal gradient leads to an increase in the plastic zone of the coal seam.After the working face advances,it indirectly leads to an increase in the gas expansion energy that can be released from the coal seam before reaching a new stress equilibrium.(2)Abnormal stress leads to increased peak stress of coal body in front of working face.When coal body in elastic area transforms to plastic area,its failure speed is accelerated,which induces accelerated gas desorption and aggravates the risk of outburst. 展开更多

Legendre polynomial method is well-known in modeling acoustic wave characteristics.This method uses for the mechanical displacements a single polynomial expansion over the entire sandwich layers.This results in a limitation in the accuracy of the field profile restitution.Thus,it can deal with the guided waves in layered sandwich only when the material properties of adjacent layers do not change significantly.Despite the great efforts regarding this issue in the literature,there remain open questions.One of them is:“what is the exact threshold of contrasting material properties of adjacent layers for which this polynomial method cannot correctly restitute the roots of guided waves?”We investigated this numerical issue using the calculated guided phase velocities in 0°/φ/0°-carbon fibre reinforced plastics(CFRP)sandwich plates with gradually increasing angleφ.Then,we approached this numerical problem by varying the middle layer thickness h90°for the 0°/90°/0°-CFRP sandwich structure,and we proposed an exact thickness threshold of the middle layer for the Legendre polynomial method limitations.We showed that the polynomial method fails to calculate the quasi-symmetric Lamb mode in 0°/φ/0°-CFRP whenφ>25°.Moreover,we introduced a new Lamb mode so-called minimum-group-velocity that has never been addressed in literature. 展开更多

This study proposes an alternative calculation mode for stresses on the slip surface(SS).The calculation of the normal stress(NS)on the SS involves examining its composition and expanding its unknown using the Taylor series.This expansion enables the reasonable construction of a function describing the NS on the SS.Additionally,by directly incorporating the nonlinear Generalized Hoke-Brown(GHB)strength criterion and utilizing the slope factor of safety(FOS)definition,a function of the shear stress on the SS is derived.This function considers the mutual feedback mechanism between the NS and strength parameters of the SS.The stress constraints conditions are then introduced at both ends of the SS based on the spatial stress relation of one point.Determining the slope FOS and stress solution for the SS involves considering the mechanical equilibrium conditions and the stress constraint conditions satisfied by the sliding body.The proposed approach successfully simulates the tension-shear stress zone near the slope top and provides an intuitive description of the concentration effect of compression-shear stress of the SS near the slope toe.Furthermore,compared to other methods,the present method demonstrates superior processing capabilities for the embedded nonlinear GHB strength criterion. 展开更多

PDZ-LIM基因与人心肌肥厚相互关系初探

目的探讨肥厚型心肌病(HCM)中Enigma家族蛋白的作用及其与心肌发育和功能异常的关联。方法通过复制缺陷腺病毒为载体,将携带PDZ-LIM结构域片段的点突变序列和正常序列转染至et-1诱导的大鼠心肌细胞中,分成实验组和对照组,采用心肌病模... 目的探讨肥厚型心肌病(HCM)中Enigma家族蛋白的作用及其与心肌发育和功能异常的关联。方法通过复制缺陷腺病毒为载体,将携带PDZ-LIM结构域片段的点突变序列和正常序列转染至et-1诱导的大鼠心肌细胞中,分成实验组和对照组,采用心肌病模型建立、心肌细胞形态及Z线结构电镜观察、2D-gel电泳和质谱分析、免疫荧光染色以及统计分析等方法,以阐明PDZ-LIM基因与人心肌肥大的相互关系,并解释其分子生物学机制。结果实验组小鼠心肌细胞平均直径、面积高于对照组,且PDZ-LIM基因的蛋白表达高于对照组,差异有统计学意义(P<0.05)。通过免疫荧光染色和电镜观察,HCM小鼠心肌细胞Z线结构发生变化,介导心脏细胞整体结构变化和Z线内蛋白分布。超微结构透射电子显微镜分析显示,突变型心肌的Z线出现中断和紊乱,M线的形态相对正常。结论HCM心肌细胞PDZ-LIM基因表达增高,在一定程度上能反映出心肌细胞的病理变化,与肥厚型心肌病存在相关性。 展开更多

目的:探究含LIM调宁蛋白同源域蛋白1(LIM and calponin homology domains 1,LIMCH1)在口腔鳞状细胞癌发生发展中的作用及潜在应用价值。方法:利用口腔鳞状细胞癌患者组织样本全转录组测序结合GO(Gene Ontology)、基因共表达网络等生物... 目的:探究含LIM调宁蛋白同源域蛋白1(LIM and calponin homology domains 1,LIMCH1)在口腔鳞状细胞癌发生发展中的作用及潜在应用价值。方法:利用口腔鳞状细胞癌患者组织样本全转录组测序结合GO(Gene Ontology)、基因共表达网络等生物信息学分析方法筛选可能在口腔鳞状细胞癌发生发展中起关键作用的基因,使用12组口腔鳞状细胞癌患者组织样本采用实时定量PCR及Western blotting验证其表达趋势,采用CCK-8、流式细胞术、划痕实验等探究关键基因对口腔鳞状细胞癌细胞系Cal27及口腔潜在恶性病变细胞系DOK生物学行为的影响,并采用TCGA(The Cancer Genome Atlas)中214例口腔鳞状细胞癌患者生存数据分析关键基因与患者临床疾病信息的关联,同时结合基因集富集分析(gene set enrichment analysis,GSEA)的方法分析结果辅以实时定量PCR及Western blot探究关键基因可能的作用机制。结果:测序数据结合生物信息学分析发现LIMCH1在口腔鳞状细胞癌组织中mRNA(P<0.001)及蛋白质(P<0.01)表达显著低于正常对照组织。Cal27中LIMCH1低表达组24 h内划痕愈合面积大于高表达组(P<0.01),72 h内增殖能力高于高表达组(P<0.01),24 h凋亡率高于其高表达组(P<0.05),而DOK中LIMCH1低表达组与其高表达组差异无统计学意义。同时发现LIMCH1低表达与患者较差的远期预后(P=0.013)及较高的淋巴结转移率(P<0.05)相关。对于LIMCH1潜在作用机制的探究中明确其并非通过上皮-间质转化途径影响口腔鳞状细胞癌的发生和发展。结论:LIMCH1可能作为辅助口腔鳞状细胞癌预后判断的潜在生物学标志物,但其具体作用机制仍有待深入探究。 展开更多

LIM基因广泛存在于真核生物中,在调控基因的表达、信号转导和胁迫应答反应等方面具有重要的调控作用。本研究基于紫鸭跖草的转录组测序数据,利用生物信息学工具筛选紫鸭跖草的LIM基因,并分析了LIM蛋白理化性质、亚细胞定位、蛋白结构和系统进化树。结果显示,筛选出16个紫鸭跖草LIM基因家族成员,其编码蛋白的长度在136~536 aa之间,蛋白分子量在14.97~60.39 kD之间,紫鸭跖草LIM蛋白大部分为稳定的碱性亲水蛋白,紫鸭跖草LIM蛋白的二级结构元件以无规卷曲和α-螺旋为主,紫鸭跖草LIM蛋白主要定位于细胞核和细胞质。系统进化树显示,紫鸭跖草LIM蛋白分为6个类群。本研究筛选和分析了紫鸭跖草LIM基因家族成员,为深入研究紫鸭跖草LIM基因家族的生物学功能提供了一定的理论基础。 展开更多

目的 探讨低浓度阿托品(ATR)对光学离焦性近视(LIM)豚鼠屈光度、眼轴及巩膜氧化应激的影响。方法 选择造模成功的LIM豚鼠30只,其中右眼为实验眼,左眼为对照眼,将大鼠随机均分为对照组(右眼0.3%玻璃酸钠滴眼1次/天)、ATR1组(右眼ATR滴眼1... 目的 探讨低浓度阿托品(ATR)对光学离焦性近视(LIM)豚鼠屈光度、眼轴及巩膜氧化应激的影响。方法 选择造模成功的LIM豚鼠30只,其中右眼为实验眼,左眼为对照眼,将大鼠随机均分为对照组(右眼0.3%玻璃酸钠滴眼1次/天)、ATR1组(右眼ATR滴眼1次/天)和ATR2组(右眼ATR滴眼2次/天)。比较各组屈光度、眼轴长度、巩膜病理形态、超氧化物歧化酶(SOD)和丙二醛(MDA)含量。结果 与实验前比较,实验后各组左眼屈光度、SOD降低,眼轴长度延长,MDA升高(P<0.05)。实验后,ATR1组和ATR2组右眼屈光度、SOD高于左眼,眼轴长度短于左眼,MDA低于左眼(P<0.05)。实验后,右眼屈光度和SOD ATR2组>ATR1组>对照组,眼轴长度和MDA ATR2组<ATR1组<对照组(P<0.05)。阿托品可有效延缓巩膜病理进展。结论 2次/天低浓度阿托品滴眼可显著延缓LIM豚鼠屈光度、眼轴长度长,延缓巩膜病理进展,并抑制组织内氧化应激。 展开更多

MRI联合血清LIMK2 mRNA FGFR4对上皮性卵巢癌的诊断价值 被引量:1

目的 探讨磁共振成像(MRI)联合LIM激酶2(LIMK2) mRNA、成纤维生长因子受体4(FGFR4)诊断上皮性卵巢癌的价值。方法 选取2020年5月至2023年5月在十堰市太和医院接受诊治的134例卵巢肿瘤患者为研究对象,以病理切片确诊为上皮性卵巢癌的患... 目的 探讨磁共振成像(MRI)联合LIM激酶2(LIMK2) mRNA、成纤维生长因子受体4(FGFR4)诊断上皮性卵巢癌的价值。方法 选取2020年5月至2023年5月在十堰市太和医院接受诊治的134例卵巢肿瘤患者为研究对象,以病理切片确诊为上皮性卵巢癌的患者49例为卵巢癌组,良性肿瘤患者85例为良性组,另选同期体检健康者55例为健康组。实时荧光定量聚合酶链反应(qRT-PCR)和酶联免疫吸附(ELISA)法分别测定LIMK2 mRNA、FGFR4水平,比较各组研究对象LIMK2 mRNA、FGFR4表达水平。所有患者均经过MRI、LIMK2 mRNA、FGFR4诊断,并以病理学结果为金标准,评价各指标单独及联合诊断上皮性卵巢癌效能。结果卵巢癌组和良性组患者LIMK2 mRNA和FGFR4水平高于健康组,差异有统计学意义(P<0.05);卵巢癌组患者LIMK2 mRNA和FGFR4水平高于良性组(P<0.05)。LIMK2 mRNA、FGFR4和MRI诊断上皮性卵巢癌的结果均与病理诊断一致性较高(Kappa=0.632、0.684、0.661,P<0.001);三者联合诊断Kappa值为0.770。三者联合诊断灵敏度较单独诊断灵敏度高(P<0.05)。结论 上皮性卵巢癌患者血清LIMK2 mRNA、FGFR4上调,MRI联合血清LIMK2 mRNA、FGFR4诊断上皮性卵巢癌效果较好。 展开更多

Advanced DriverAssistance Systems(ADAS)technologies can assist drivers or be part of automatic driving systems to support the driving process and improve the level of safety and comfort on the road.Traffic Sign Recognition System(TSRS)is one of themost important components ofADAS.Among the challengeswith TSRS is being able to recognize road signs with the highest accuracy and the shortest processing time.Accordingly,this paper introduces a new real time methodology recognizing Speed Limit Signs based on a trio of developed modules.Firstly,the Speed Limit Detection(SLD)module uses the Haar Cascade technique to generate a new SL detector in order to localize SL signs within captured frames.Secondly,the Speed Limit Classification(SLC)module,featuring machine learning classifiers alongside a newly developed model called DeepSL,harnesses the power of a CNN architecture to extract intricate features from speed limit sign images,ensuring efficient and precise recognition.In addition,a new Speed Limit Classifiers Fusion(SLCF)module has been developed by combining trained ML classifiers and the DeepSL model by using the Dempster-Shafer theory of belief functions and ensemble learning’s voting technique.Through rigorous software and hardware validation processes,the proposedmethodology has achieved highly significant F1 scores of 99.98%and 99.96%for DS theory and the votingmethod,respectively.Furthermore,a prototype encompassing all components demonstrates outstanding reliability and efficacy,with processing times of 150 ms for the Raspberry Pi board and 81.5 ms for the Nano Jetson board,marking a significant advancement in TSRS technology. 展开更多

To analyze the differences in the transport and distribution of different types of proppants and to address issues such as the short effective support of proppant and poor placement in hydraulically intersecting fractures,this study considered the combined impact of geological-engineering factors on conductivity.Using reservoir production parameters and the discrete elementmethod,multispherical proppants were constructed.Additionally,a 3D fracture model,based on the specified conditions of the L block,employed coupled(Computational Fluid Dynamics)CFD-DEM(Discrete ElementMethod)for joint simulations to quantitatively analyze the transport and placement patterns of multispherical proppants in intersecting fractures.Results indicate that turbulent kinetic energy is an intrinsic factor affecting proppant transport.Moreover,the efficiency of placement and migration distance of low-sphericity quartz sand constructed by the DEM in the main fracture are significantly reduced compared to spherical ceramic proppants,with a 27.7%decrease in the volume fraction of the fracture surface,subsequently affecting the placement concentration and damaging fracture conductivity.Compared to small-angle fractures,controlling artificial and natural fractures to expand at angles of 45°to 60°increases the effective support length by approximately 20.6%.During hydraulic fracturing of gas wells,ensuring the fracture support area and post-closure conductivity can be achieved by controlling the sphericity of proppants and adjusting the perforation direction to control the direction of artificial fractures. 展开更多

目的研究新生儿持续性肺动脉高压(PPHN)模型大鼠肺组织中缺氧诱导因子1α(Hif1α)、四个半LIM结构域蛋白1(Fhl1)的表达,初步探讨Hif1α、Fhl1与PPHN发生发展的关系。方法妊娠第19天,将6只孕鼠随机分为PPHN模型组和对照组,每组3只,给予P... 目的研究新生儿持续性肺动脉高压(PPHN)模型大鼠肺组织中缺氧诱导因子1α(Hif1α)、四个半LIM结构域蛋白1(Fhl1)的表达,初步探讨Hif1α、Fhl1与PPHN发生发展的关系。方法妊娠第19天,将6只孕鼠随机分为PPHN模型组和对照组,每组3只,给予PPHN模型组低氧(吸氧浓度10%~12%)联合吲哚美辛干预孕鼠建立PPHN大鼠模型,给予对照组正常氧浓度(吸氧浓度21%)。妊娠第22天,麻醉孕鼠并迅速剖腹取出新生鼠,采集新生鼠血液、心脏及肺组织标本。采用新生鼠血浆脑钠肽(BNP)浓度、右心室肥厚指数、肺小动脉中膜厚度百分比(MT%)、肺小动脉中膜面积百分比(MA%)评价PPHN模型是否成功建立;应用Western blot技术监测两组新生大鼠肺组织中Hif1α、Fhl1蛋白表达变化。结果与对照组比较,PPHN模型组肺小动脉管壁增厚,血管腔变窄,肺小动脉MT%、MA%、右心室肥厚指数、血浆BNP浓度均升高(P<0.01);PPHN模型组Hif1α、Fhl1蛋白表达水平均增加(P<0.05)。结论PPHN模型大鼠肺组织Hif1α、Fhl1蛋白表达均增加,提示Hif1α和Fhl1可能与PPHN的发生发展密切相关。 展开更多

Toppling failure of rock mass/soil slope is an important geological and environmental problem.Clarifying its failure mechanism under different conditions has great significance in engineering.The toppling failure of a cutting slope occurred in a hydropower station in Kyushu,Japan illustrates that the joint characteristic played a significant role in the occurrence of rock slope tipping failure.Thus,in order to consider the mechanical properties of jointed rock mass and the influence of geometric conditions,a simplified analytical approach based on the limit equilibrium method for modeling the flexural toppling of cut rock slopes is proposed to consider the influence of the mechanical properties and geometry condition of jointed rock mass.The theoretical solution is compared with the numerical solution taking Kyushu Hydropower Station in Japan as one case,and it is found that the theoretical solution obtained by the simplified analysis method is consistent with the numerical analytical solution,thus verifying the accuracy of the simplified method.Meanwhile,the Goodman-Bray approach conventionally used in engineering practice is improved according to the analytical results.The results show that the allowable slope angle may be obtained by the improved Goodman-Bray approach considering the joint spacing,the joint frictional angle and the tensile strength of rock mass together. 展开更多

RKDG Methods with Multi-resolution WENO Limiters for Solving Steady-State Problems on Triangular Meshes

In this paper, we design high-order Runge-Kutta discontinuous Galerkin (RKDG) methods with multi-resolution weighted essentially non-oscillatory (multi-resolution WENO) limiters to compute compressible steady-state problems on triangular meshes. A troubled cell indicator extended from structured meshes to unstructured meshes is constructed to identify triangular cells in which the application of the limiting procedures is required. In such troubled cells, the multi-resolution WENO limiting methods are used to the hierarchical L^(2) projection polynomial sequence of the DG solution. Through using the RKDG methods with multi-resolution WENO limiters, the optimal high-order accuracy can be gradually reduced to first-order in the triangular troubled cells, so that the shock wave oscillations can be well suppressed. In steady-state simulations on triangular meshes, the numerical residual converges to near machine zero. The proposed spatial reconstruction methods enhance the robustness of classical DG methods on triangular meshes. The good results of these RKDG methods with multi-resolution WENO limiters are verified by a series of two-dimensional steady-state problems. 展开更多

This study investigated the physicochemical properties,enzyme activities,volatile flavor components,microbial communities,and sensory evaluation of high-temperature Daqu(HTD)during the maturation process,and a standard system was established for comprehensive quality evaluation of HTD.There were obvious changes in the physicochemical properties,enzyme activities,and volatile flavor components at different storage periods,which affected the sensory evaluation of HTD to a certain extent.The results of high-throughput sequencing revealed significant microbial diversity,and showed that the bacterial community changed significantly more than did the fungal community.During the storage process,the dominant bacterial genera were Kroppenstedtia and Thermoascus.The correlation between dominant microorganisms and quality indicators highlighted their role in HTD quality.Lactococcus,Candida,Pichia,Paecilomyces,and protease activity played a crucial role in the formation of isovaleraldehyde.Acidic protease activity had the greatest impact on the microbial community.Moisture promoted isobutyric acid generation.Furthermore,the comprehensive quality evaluation standard system was established by the entropy weight method combined with multi-factor fuzzy mathematics.Consequently,this study provides innovative insights for comprehensive quality evaluation of HTD during storage and establishes a groundwork for scientific and rational storage of HTD and quality control of sauce-flavor Baijiu. 展开更多

In situ stress inversion using nonlinear stress boundaries achieved by the bubbling method 被引量:1

Due to the heterogeneity of rock masses and the variability of in situ stress,the traditional linear inversion method is insufficiently accurate to achieve high accuracy of the in situ stress field.To address this challenge,nonlinear stress boundaries for a numerical model are determined through regression analysis of a series of nonlinear coefficient matrices,which are derived from the bubbling method.Considering the randomness and flexibility of the bubbling method,a parametric study is conducted to determine recommended ranges for these parameters,including the standard deviation(σb)of bubble radii,the non-uniform coefficient matrix number(λ)for nonlinear stress boundaries,and the number(m)and positions of in situ stress measurement points.A model case study provides a reference for the selection of these parameters.Additionally,when the nonlinear in situ stress inversion method is employed,stress distortion inevitably occurs near model boundaries,aligning with the Saint Venant's principle.Two strategies are proposed accordingly:employing a systematic reduction of nonlinear coefficients to achieve high inversion accuracy while minimizing significant stress distortion,and excluding regions with severe stress distortion near the model edges while utilizing the central part of the model for subsequent simulations.These two strategies have been successfully implemented in the nonlinear in situ stress inversion of the Xincheng Gold Mine and have achieved higher inversion accuracy than the linear method.Specifically,the linear and nonlinear inversion methods yield root mean square errors(RMSE)of 4.15 and 3.2,and inversion relative errors(δAve)of 22.08%and 17.55%,respectively.Therefore,the nonlinear inversion method outperforms the traditional multiple linear regression method,even in the presence of a systematic reduction in the nonlinear stress boundaries. 展开更多
