Pro-resolving lipid mediator reduces amyloid-β42–induced gene expression in human monocyte–derived microglia

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease. 展开更多

Rheumatoid arthritis(RA)is a systemic autoimmune disease in which synovial fibroblasts(SFs)maintain chronic inflammation by secreting proinflammatory mediators,leading to joint destruction.While the role of proinflammatory mediators in this process is well-established,the contribution of non-inflammatory regulators in SFs to joint pathology remains poorly understood.In this study,we investigated the non-inflammatory role of SFs in RA using a co-culture model,and found that SFs from RA patients promote apoptosis of human chondrocytes.Mechanistic investigations reveal that SFs can secrete small extracellular vesicles(sEVs),which are taken up by chondrocytes and induce chondrocyte apoptosis in both normal chondrocytes and chondrocytes from patients with RA.sEV-derived miRNA 15-29148 are identified as key signaling molecules mediating the apoptosis effects of chondrocytes.Further studies reveal that SF-derived miRNA 15-29148 targeting CIAPIN1 results in increased chondrocyte apoptosis.We further demonstrate that SF-derived miRNA 15-29148 is transferred to chondrocytes,exacerbating cartilage damage in vivo.Moreover,chondrocyte-specific aptamer-modified polyamidoamine nanoparticles not only ameliorated RA but also prevented its onset.This study suggests that,in RA,the secretion of specific sEV-miRNAs from SFs plays a crucial role in promoting chondrocyte apoptosis,potentially through non-inflammatory regulation,and that sEV-miRNA inhibition in SFs may represent an early preventive treatment strategy for cartilage degradation in RA. 展开更多

Relationship between parenting stress and behavioral and emotional problems in preschool children: A mediation effect analysis

BACKGROUND Emotional reactions,such as anxiety,irritability,and aggressive behavior,have attracted clinical attention as behavioral and emotional problems in preschool-age children.AIM To investigate the current status of family rearing,parental stress,and behavioral and emotional problems of preschool children and to analyze the mediating effect of the current status of family rearing on parental stress and behavioral/emo-tional problems.METHODS We use convenience sampling to select 258 preschool children in the physical examination center of our hospital from October 2021 to September 2023.The children and their parents were evaluated using a questionnaire survey.Pearson's correlation was used to analyze the correlation between child behavioral and emotional problems and parental stress and family rearing,and the structural equation model was constructed to test the mediating effect.RESULTS The score for behavioral/emotional problems of 258 preschool children was(27.54±3.63),the score for parental stress was(87.64±11.34),and the score for parental family rearing was(31.54±5.24).There was a positive correlation between the behavioral and emotional problems of the children and the“hostile/mandatory”parenting style;meanwhile,showed a negative correlation with the“support/participation”parenting style(all P<0.05).The intermediary effect value between the family upbringing of parents in parental stress and children's behavior problems was 29.89%.CONCLUSION Parental family upbringing has a mediating effect between parental stress and behavioral and emotional problems of children.Despite paying attention to the behavioral and emotional problems of preschool-age children,clinical medical staff should provide correct and reasonable parenting advice to their parents to promote the mental health of preschool-age children. 展开更多

Escalation of soya cross-allergy in infants with cow's milk allergy

BACKGROUND Cow's milk allergy(CMA)is a common condition in infants,requiring alternative protein sources in their diets.Soya milk has become a popular substitute,especially in developing countries where it is a more affordable option compared to expensive hypoallergenic feeds for infants with insufficient breast milk supply.However,recent observations have shown an increase in soya cross-allergic reactions among infants with CMA.AIM To determine how often infants diagnosed with CMA also had soya cross-allergy and to examine the symptoms and outcomes of these infants at 2 years of age.METHODS Data from two pediatric centers were analyzed,looking at clinical records of children under 2 years old diagnosed with CMA from August 2015 to July 2023,divided into two four-year periods.RESULTS The records of 432 infants with CMA were analyzed.In the first four-year period from August 2015 to July 2019,142 infants were studied,with 27(19%)found to have soya-protein allergy as well.In the second four-year period,a total of 290 infants were studied,and soya allergy was found in 136 babies(47%).This represents a significant increase(P<0.0001)in cases of soya protein cross-allergy among infants with CMA.The most common symptoms observed were gastroesophageal reflux disorder(39%),followed by failure to thrive,bloody diarrhea,watery diarrhea,and constipation.At 2 years of age,these infants showed significant growth failure compared to infants with CMA only.CONCLUSION In conclusion,this study emphasizes the importance of being cautious when using soy protein in infants with cow's milk protein allergy,especially in areas where cost is a major concern. 展开更多

In recent years, the use of new biomarkers in different phases of the diagnosis andtreatment of several diseases has allowed substantial improvement in clinicalpractice. The use of donor-derived cell-free DNA (dd-cfDNA) in organ transplantationhas led to significant progress in the treatment of post-transplantoutcomes, particularly after kidney transplantation. In addition, the use of ddcfDNAin organ transplantation has led to significant advancements in posttransplantoutcome monitoring. The aim of this study is to review many of therecent studies on the use of this biomarker and to evaluate its most relevantadvantages and limitations. dd-cfDNA is released from several types of cells ofthe transplanted organ, most often from endothelial cells and this happens in thecase of organ damage, most often rejection. Its presence in the bloodstream of therecipients is an important sign of graft damage;its principal advantage is in theavoidance of invasive tools such as renal biopsy. Additionally, several studiesreported that the finding of dd-cfDNA in the serum may precede histologicalabnormalities;its utility in the diagnosis of subclinical rejection is extremelyimportant. Among the principal limitations of this tool are the difficulty in distinguishingdifferent forms of graft damage. According to several studies this toolhas several limitations in diagnosing T-cell mediated rejection. This study tested a chain mediation model on whether hopelessness and nostalgia play a mediating role in psychache and suicidal ideation of firefighters.A total of 652 firefighters participated in the survey(male=94.94%;mean age=23.71 years,SD=4.18 years).The firefighters completed the Chinese Revised Psychache Scale(PAS),Beck Hopelessness Scale(BHS),Southampton Nostalgia Scale(SNS),and Columbia-Suicide Severity Rating Scale(CSSRS).The path analysis results indicated that psychache positively predicted suicidal ideation.Hopelessness plays a significant mediating role between psychache and suicidal ideation further strengthening this relationship.In contrast,nostalgia mediates and weakened the relationship between psychache and suicidal ideation.Hopelessness and nostalgia jointly constructed a chain mediating effect between psychache and suicidal ideation,for higher suicidal ideation with higher hopelessness and lower nostalgia.The findings align with the Three-Step Theory of Suicide,which proposes that suicidal ideation results from the combination of pain and hopelessness and that connectedness is a key protective factor against escalating ideation.Therefore,interventions to reduce suicidal ideation in firefighters should aim to enhance their nostalgia,while reducing sense of hopelessness. 展开更多

From health literacy to organ donation and transplantation awareness:The path through self-control

BACKGROUND Organ donation and transplantation are vital for saving lives,yet awareness remains limited.Health literacy,the ability to understand and use health information,and self-control,which influences decision-making,are crucial factors in promoting awareness.Understanding their relationship can guide efforts to increase participation in organ donation.AIM To investigate the effect of health literacy on organ donation and transplantation awareness,with self-control acting as a mediator,and to emphasize its importance.METHODS The study comprised 345 adults recruited from five different cities in Türkiye.The sample included 208 women(60.29%)and 137 men(39.71%).The age range of the sample group varies between 18 and 49(median age=24.13;SD=8.81).Selfreport data were collected including psychometric measures assessing organ donation and transplantation awareness,health literacy,and self-control.Reliability analysis,descriptive statistics,correlational analysis,and mediational analysis of the data were carried.RESULTS The analysis showed that health literacy was positively correlated with organ donation and transplantation awareness(r=0.340)and self-control(r=0.376).Self-control was positively related to organ donation and transplantation awareness(r=0.531).Mediating analysis supported a mediation model where health literacy was found to influence organ donation and transplantation awareness via self-control(bootstrap=0.158,95%confidence interval=0.105-0.215).CONCLUSION The results indicate that self-control mediates the positive relationship between health literacy and organ donation and transplantation awareness.Enhancing individuals’health literacy may therefore increase awareness of organ donation and transplantation through improvements in self-control. 展开更多

This letter critically examines a recent study by Zhang et al investigating the mediating role of overweight in the association between depression and new-onset diabetes among middle-aged and older adults.The study provides com-pelling evidence that overweight mediates approximately 61%of this relationship,suggesting that depression may contribute to diabetes by influencing behaviors that lead to weight gain.This aligns with the understanding that depression can impact appetite regulation and physical activity.While the study employs a longitudinal design and robust statistical methods,limitations such as reliance on self-reported data and body mass index measurements warrant consideration.This analysis emphasizes the need for integrated interventions that address both mental and metabolic health for effective diabetes prevention.Future research should further explore the interplay of lifestyle factors,biological pathways,and social determinants in the development of this complex relationship.Ultimately,an integrated approach targeting both behavioral and biological components is crucial for the prevention and management of new-onset diabetes. 展开更多

BACKGROUND Depression is a leading global health concern with high suicide rates and recurrence.Cognitive models suggest that mental pain and automatic thoughts are central to depression's impact.The hypothesis is that self-compassion will be negatively associated with mental pain,mediated by automatic thoughts.AIM To determine the mediating role of automatic thoughts in the relationship between self-compassion and mental pain in individuals with depression.METHODS This cross-sectional study included 389 inpatients with depression from Tianjin Anding Hospital.Participants completed the Self-Compassion Scale-Chinese Version(SCS-C),Automatic Thought Questionnaire(ATQ),and Orbach&Mikulincer Mental Pain Scale-Chinese Version(OMMP).Data were analyzed using Pearson correlations,multiple linear regressions,and mediation analysis.RESULTS The SCS-C total score was 68.95±14.89,ATQ was 87.02±28.91,and OMMP was 129.01±36.74.Correlation analysis showed mental pain was positively associated with automatic thoughts(r=0.802,P<0.001)and negatively with selfcompassion(r=-0.636,P<0.001).Regression analysis indicated automatic thoughts(β=0.623,P<0.001)and self-compassion(β=-0.301,P<0.001)significantly predicted mental pain.Mediation analysis confirmed automatic thoughts partially mediated the relationship between self-compassion and mental pain(ab=-0.269,95%CI:-0.363 to-0.212).CONCLUSION Self-compassion is inversely related to mental pain in depression,with automatic thoughts playing a mediating role.These findings suggest potential therapeutic targets for alleviating mental pain in depressed patients. 展开更多

Background:The Teaching Personal and Social Responsibility(TPSR)model in physical education(PE)has been shown to promote Social and emotional competence(SEC).However,the underlying mechanisms through which TPSR enhances SEC,particularly in university students within the Chinese context,remain unclear.This study aims to explore the effects of TPSR and the mediating roles of self-efficacy and grit in improving SEC.Methods:71 Chinese university students were in the TPSR group,and 39 in the Traditional Teaching Model(TTM)control group,assessed before and after a 14-week intervention.The Adapted Social and Emotional Competence Scale(ASECS),General Self-Efficacy Scale(GSES),and Short Grit Scale(SGS)were used for measurement.A mixed-design ANOVA assessed TPSR’s effects,with post-hoc t-tests for pre-post differences and mediation analysis for underlying mechanisms.Results:The mixed-design ANOVA revealed a significant interaction between time and intervention on SEC.The TPSR group showed significant improvement from pre-test to post-test(t(70)=−2.63,p=0.011,Cohen’s d=−0.31),whereas the TTM control group did not(t(38)=1.40,p=0.170,Cohen’s d=0.22).The TPSR group also showed a significant increase in self-efficacy(t(70)=−3.67,p<0.001,Cohen’s d=−0.44),while no change was observed in the TTM group(t(38)=0.62,p=0.540,Cohen’s d=0.10).No significant effects were found for grit(F=0.342,p=0.560).Mediation analysis confirmed that self-efficacy significantly mediated the effects of the TPSR intervention on SEC(95%CI:0.0277 to 0.2897).Conclusion:The TPSR intervention led to significant improvements in university students’SEC and self-efficacy,with no significant changes observed in grit.Self-efficacy served as a mediator in the relationship between the intervention and SEC. 展开更多

This study examines how organizational support influences the career adaptability of novice university teachers in Guangdong,China,and the mediating role of teacher self-efficacy.Drawing on social cognitive theory and organizational support theory,we hypothesized that organizational support would positively predict career adaptability through self-efficacy.A cross-sectional survey was conducted with 326 novice teachers(with 1–3 years of teaching experience)from 12 universities in Guangdong.Data were analyzed using correlation analysis,hierarchical regression,and bootstrap.Results showed that:(1)organizational support was positively associated with both self-efficacy(r=0.62,P<0.001)and career adaptability(r=0.58,P<0.001);(2)self-efficacy fully mediated the relationship between organizational support and career adaptability(indirect effect=0.24,95%CI[0.18,0.31]),with a partial reduction in the direct effect fromβ=0.35 toβ=0.17(P<0.05);(3)female teachers reported higher self-efficacy than males(P<0.05),and teachers with 2 years of experience showed significantly higher adaptability than those with 1 year(P<0.05).The findings highlight the critical role of self-efficacy in translating organizational support into adaptability,providing empirical evidence for universities to design targeted support strategies. 展开更多

In recent years,forest therapy has become a popular method for improving human health.However,guided forest therapy is not always easily accessible,and forest walking is a more convenient and feasible alterna-tive.Therefore,it is important to determine whether forest walking has the same effect as guided forest therapy.To investigate this,we conducted a campus forest-based study in which 247 university students were randomly assigned to participate in either forest walking or guided forest therapy activities.The study measured physical and psychological interventions in participants,while controlling for the inten-sity of physical activity.The findings indicated that both approaches were effective in promoting stress relief and physical and mental recovery among university students.No significant difference in effectiveness was observed between the two approaches.Furthermore,we constructed a mediation model that combines the biophilia hypothesis,stress reduction theory,and attention restoration theory to investigate the psychological mechanisms underlying the restorative effects of forest activities.Our findings indi-cate that an increase in nature connectedness significantly predicts a reduction in state anxiety.This effect is medi-ated by perceived restorativeness and a combination chain of mediators from perceived restorativeness to mood.This study presents a justification for selecting forest walking as a means of stress relief when guided forest therapy is unavail-able.Additionally,it enhances our comprehension of how forests contribute to the restorative effects experienced by individuals. 展开更多

Background:Recent scholarly attention has increasingly focused on filial piety beliefs'impact on youth's psychological development.However,the mechanisms by which filial piety indirectly influences adolescent autonomy through depression and well-being remain underexplored.This study aimed to test a sequential mediation model among filial piety beliefs,depression,well-being,and autonomy in Taiwan region of China university students.Methods:A total of 566 Taiwan region of China undergraduate and graduate students,comprising 390 females and 176 males,and including 399 undergraduates and 167 graduate students,were recruited through convenience sampling.Data were collected via an online questionnaire.Validated instruments were employed,including the Filial Piety Scale(FPS),the Center for Epidemiological Studies Depression Scale(CES-D),the Chinese Well-being Inventory(CHI),and the Adolescent Autonomy Scale-Short Form(AAS-SF).Statistical analyses included group comparisons,correlation analyses,and structural equation modeling to examine the hypothesized relationships and mediation effects.Results:The results revealed that filial piety beliefs exerted a significant positive impact on adolescent autonomy,with depression and well-being serving as key mediators in this relationship.A sequential mediation effect was confirmed through structural equation modeling(β=0.052,95%CI[0.028,0.091]),with good model fit indices(x^(2)/df=4.25,RMSEA=0.076,CFI=0.968),supporting the hypothesized pathway from filial piety to autonomy via depression and well-being.In terms of demographic differences,male students showed significantly higher autonomy than females(p<0.001);students from single-parent families reported significantly higher depression levels than those from two-parent families(p<0.05);and graduate students exhibited significantly higher autonomy and well-being than undergraduates(p<0.05).Conclusions:These findings underscore not only the importance of filial piety beliefs for developing youth autonomy but also the critical role that mental health factors,such as depression and well-being,play in this process.The study concludes with a discussion of both theoretical implications and practical recommendations.These include strategies to foster reciprocal filial piety,strengthen parent-child relationships,and promote mental health.Additionally,the study outlines its limitations and proposes directions for future research. 展开更多

BACKGROUND Several studies have suggested a close link between depression,overweight,and new-onset diabetes,particularly among middle-aged and older populations;however,the causal associations remain poorly understood.AIM To investigate the role of overweight in mediating the association between depression and new-onset diabetes in middle-aged and older populations.METHODS Data of 9426 individuals aged≥50 years from the 1998-2016 Health and Retirement Study database were analyzed.Weighted logistic regression was employed to obtain odds ratios(ORs)and 95%confidence intervals(95%CIs)for depression and new-onset diabetes in the middle-aged and older populations.Mediation analysis and the Sobel test were used to test the mediating effects of overweight between depression and the risk of new-onset diabetes.RESULTS New-onset diabetes was identified in 23.6%of the study population.Depression was significantly associated with new-onset diabetes(OR:1.18,95%CI:1.03-1.35,P value:0.014).Further adjustment for overweight attenuated the effect of depression on new-onset diabetes to 1.14(95%CI:1.00-1.30,P=0.053),with a significant mediating effect(P of Sobel test=0.003).The mediation analysis demonstrated that overweight accounted for 61%in depression for the risk of new-onset diabetes,with overweight having a partially mediating role in the depression-to-diabetes pathway.CONCLUSION New-onset diabetes was not necessarily a direct complication of depression;rather,depression led to behaviors that increase the risk of overweight and,consequently,new-onset diabetes. 展开更多
