However, little information is available about GAs toxicity in kidneys and the underlying mechanism has not been thoroughly elucidated so far. Instead of GAs extract, the renal cytotoxicity of GA(15 : 1), which was isolated from the testa of Ginkgo biloba, was assessed in vitro by using MDCK cells. The action of GA(15 : 1) on cell viability was evaluated by the MTT and neutral red uptake assays. Compared with the control, the cytotoxicity of GA(15 : 1) on MDCK cells displayed a time-and dose-dependent manner, suggesting the cells mitochondria and lysosomes were damaged. It was confirmed that GA(15 : 1) resulted in the loss of cells mitochondrial trans-membrane potential(ΔΨm). In propidium iodide(PI) staining analysis, GA(15 : 1) induced cell cycle arrest at the G0/G1 and G2/M phases, influencing on the DNA synthesis and cell mitosis. Characteristics of necrotic cell death were observed in MDCK cells at the experimental conditions, as a result of DNA agarose gel electrophoresis and morphological observation of MDCK cells. In conclusion, these findings might provide useful information for a better understanding of the GA(15 : 1) induced renal toxicity.展开更多

The structural properties of the adsorbents were characterized by X-ray diffraction(XRD), N_2 sorption(BET) and thermal analysis(TGA). The effects of calcination temperature, calcination atmosphere, and adsorption temperature were studied by the dynamic and static tests. The competitive adsorption mechanisms between thiophene and 1-hexene were studied by in-situ Fourier transform infrared spectroscopy(in-situ FTIR) and temperature-programmed desorption(TPD). CsY adsorbents exhibited high selectivity for thiophene even when a large amount of olefins exist.In-situ FTIR spectra of thiophene and 1-hexene adsorption indicated that both thiophene and 1-hexene were mainly adsorbed on CsY via π-complexation. The higher desorption activated energy and higher adsorption heat of thiophene than 1-hexene obtained by thiophene-TPD and hexene-TPD has revealed that thiophene is adsorbed more strongly in CsY adsorbents than 1-hexene.展开更多

The aging behavior of the alloy was studied by Vickers hardness measurement at 200℃ and 300℃. and the tensile properties of alloy specimens were measured at 20℃, 200℃, 250℃ and 300℃. The microstructure was observed by using optical microscope, SEM and TEM. It was found that the addition of Sc and Zr to 2618 alloy resulted in a primary Al_3(Sc,Zr) phase which could refine the grain because it acts as nuclei of heterogeneous crystallization in the melt during solidification. The secondary Al_3(Sc,Zr) particles were full coherent with matrix and had obvious precipitation hardening effect. They also made the S' phase precipitate more homogeneous. So the strength of alloy increases at both ambient and elevated temperatures without a decrease of ductility. The ductile fracture of alloy occurs by microvoid nucleation, growth and coalescence, so the microvoid coalescence is the dominant fracture mechanism.展开更多

Rats were randomly divided into three groups: sham, cecal ligation and puncture (CLP), and CLP with ginsenoside Rb1. Then, the survival rate, arterial blood pressure, TLR4 mRNA, and TNF-α levels were determined. The liver and lung tissues were stained with hematoxylin-eosin (HE). The overall survival rate of the Rb1 group was significantly higher than that of the CLP group. Mean arterial blood pressure went down in both the CLP and Rb1 groups after CLP, and there was a significant difference both in the sham and Rb1 groups when compared with the CLP group. The Rb1 treatment group had markedly lower TLR4 mRNA expression and TNF-a levels than the CLP group. In the CLP group, pathology showed swelling, degeneration, necrosis, and neutrophii infiltration in the liver and alveolar epithelial cells. However, in the Rb1 group, there was mild degeneration and slight neutrophil infiltration, but no obvious necrosis. Rb1 may improve the survival rate, ameliorate arterial blood pressure, and protect the liver and lung in septic shock rats by downregulating the expression of TLR4 mRNA and inhibiting the production of TNF-α.展开更多

The connection between the simplified model and the target aster-oid is built by considering the positions of equilibrium points. To improve the performance of po-sition matching for the equilibrium points associated with these non-axisymmetric asteroids, a non-axisymmetric triple-particle-linkage model is proposed based on two existing axisymmetric particle- linkage models. The unknown parameters of the simplified model are determined by minimizing the matching error using the nonlinear optimization method. The proposed simplified model is applied for three realistic elongated asteroids, 243 Ida, 433 Eros and (8567) 1996 HW1. The simulation re-sults verify that the current particle-linkage model has better matching accuracy than the two existing particle-linkage models. The comparison, between the simplified model and the polyhedral model, on the topological cases of the equilibrium points and the distribution of gravitational potential further validate the rationality and accuracy of the simplified model.展开更多

However, metabolic regulation in mechanobiology remains largely unexplored. Here, we identified glucose transporter 1(GLUT1)—the primary glucose transporter in various cells—as a novel mechanosensitive gene in orthodontic tooth movement(OTM). Using an in vivo rat OTM model, we demonstrated the specific induction of Glut1 proteins on the compressive side of a physically strained periodontal ligament. This transcriptional activation could be recapitulated in in vitro cultured human periodontal ligament cells(PDLCs), showing a time-and dose-dependent mechanoresponse. Importantly, application of GLUT1 specific inhibitor WZB117 greatly suppressed the efficiency of orthodontic tooth movement in a mouse OTM model, and this reduction was associated with a decline in osteoclastic activities. A mechanistic study suggested that GLUT1 inhibition affected the receptor activator for nuclear factor-κ B Ligand(RANKL)/osteoprotegerin(OPG)system by impairing compressive force-mediated RANKL upregulation. Consistently, pretreatment of PDLCs with WZB117 severely impeded the osteoclastic differentiation of co-cultured RAW264.7 cells. Further biochemical analysis indicated mutual regulation between GLUT1 and the MEK/ERK cascade to relay potential communication between glucose uptake and mechanical stress response. Together, these cross-species experiments revealed the transcriptional activation of GLUT1 as a novel and conserved linkage between metabolism and bone remodelling.展开更多

DATA SOURCES: A computer-based online search of Medline database was undertaken to identify articles about the metabolic pathway of MPTP and its mechanism in inducing Parkinson disease published in English between January 1996 and August 2004, the keywords were "MPTP, Parkinson disease". Meanwhile, Chinese relevant articles published between January 2000 and August 2004 were searched in Wanfang database with the keywords of "MPTP, Parkinson disease". STUDY SELECTION: More than 300 relevant literatures were retrieved, and the full-texts were further searched, those about the establishment of animal models, molecular mechanism of MPTP neurotoxicity and the metabolism were selected, and the obviously repetitive ones, case report and reviews were excluded, finally 18 of them were selected for summarization. DATA EXTRACTION: The 18 literatures were categorized according to MPTP induced animal models of Parkinson disease, mechanism of MPTP in inducing apoptosis in models of Parkinson disease, role of dopamine in the neurotoxic mechanism of MPTP, the role of reactive oxygen species and nitric oxide in the neurotoxicity of MPTP. DATA SYNTHESIS: Animal models of Parkinson disease induced by MPTP can not only produce the clinical characters of Parkinson disease, also duplicate the main biochemical and pathological changes of Parkinson disease. The metabolic pathway of MPTP and its mechanism in inducing Parkinson disease included producing free oxygen and nitric oxide, damaging mitochondrial respiratory chain, and inducing apoptosis, etc. which could all lead to the degeneration and loss of dopaminergic neurons. CONCLUSION : Although some aspects of the models of Parkinson disease are different from that in human beings, we can still know the neurodegeneration of Parkinson disease through studying the molecular mechanism of MPTP.展开更多

(NH4)2 HPO4 at room temperature and atmospheric pressure. Nanocomposites of HA and partially stabilized zirconia (HA/PSZ) were sintered at atmospheric pressure and 1300 for 2h in air. The preparation techniques, structure and me- chanical properties of these materials were characterized. The addition of nanosized PSZ reinforcing phase to HA may lead to an improvement of the macro and micro mechanical properties and not affect its biocompatibility and bioactivity. The bending strength, fracture toughness and nano-hardness were near to or greater than those for human cortical bone and human tooth (dentine and enamel). The composite was incubated in a fresh human plasma which confirmed the bioactivity of nanosized HA/PSZ materials. The bonding reaction between HA/PSZ ceramic and the plasma proteins was found, and he- matopoietic cell phosphatase (HCP) layers formed on surface of each composite incubated in human plasma for two weeks. The diameter of a single HCP globule was less than 100 nm. Furthermore, the precipitating mechanism investigation was carried out through a comparative experiment in this paper.展开更多

With the A1 additions from 2.5 wt pct to 5 wt pct, the siliceous grain boundary phase in hot pressed samples was eliminated because of the formation of Al2O3 particles. It was shown that Al and SiO2 reacted at 860℃. During the reaction, A1 atoms were mainly transferred to Al2O3 particles, and to some extent, diffused into MoSi2 grains. Both the toughness and strength of Al containing composites exceeded those of pure MoSi2 material. Bending strength and fracture toughness reach the highest value of 350 MPa, 4.05 MPa.m1/2, respectively, at ambient temperature when Al addition was of 3.5 wt pct.展开更多

The mechanisms of the extension are analyzed in this paper. Lithiumization diminishes the impurities and hydrogen recycling to the lowest level. After lithiumization a high density up to 7×1019 m-3 was obtained easily by strong gas puffing with ordinary ohmic discharge alone. More attractively we found that metal Li-coating exhibited the effects of wall stabilization. The low qα limit with higher density was extended by a factor of 1.5-2 in comparison with that for boronization, and 1.2 for siliconization. Siliconization not only extended stable operating region significantly by itself, but also provided a good target plasma for other experiments of raising density limit. Core fuelling schemes are favourable especially for siliconized wall with a higher level of medium-Z impurity (Z = 14). After siliconization the maximum density near to 1020 m-3 was achieved by a combination of supersonic molecule beam injection and multipellet injection. The new defined slope of Hugill limit illustrating more clearly the situation under low qα and high ne discharges was created to indicate the new region extended by combining IP ramp-up with core fuelling. The slope with a large Murakami coefficient increased by a factor of 50-60 %.展开更多

CONCLUSION : Although some aspects of the models of Parkinson disease are different from that in human beings, we can still know the neurodegeneration of Parkinson disease through studying the molecular mechanism of MPTP.
文摘Nanocrystalline hydroxyapatite (HA) powders have been synthesized by precipitation using Ca(NO3)2 4H2O and? Furthermore, the precipitating mechanism investigation was carried out through a comparative experiment in this paper.

文摘Puerarin is a major active ingredient of the traditional Chinese plant medicine,Radix Puerariae, and commonly used in the treatment of myocardial and cerebral ischemia. However, the effects of puerarin on neuropathic pain are still unclear. In this study,a neuropathic pain animal model was created by partial sciatic nerve ligation.Puerarin(30 or 60 mg/kg) was intraperitoneally injected once a day for 7 days.Mechanical allodynia and thermal hyperalgesia were examined at 1 day after model establishment.Mechanical threshold and paw withdrawal latency markedly increased in a dose-dependent manner in puerarin-treated rats, especially at 7 days after model establishment. At 7 days after model establishment, quantitative real-time reverse transcriptase-polymerase chain reaction results showed that puerarin administration reversed m RNA expression of transient receptor potential vanilloid 1(Trpv1) and transient receptor potential ankyrin 1(Trpa1) in a dose-dependent manner in dorsal root ganglion neurons after peripheral nerve injury. These results suggest that puerarin dose-dependently ameliorates neuropathic pain by suppressing Trpv1 and Trpa1 up-regulation in dorsal root ganglion of neuropathic pain rats.
文摘Under the assumption that the growth order of the free term to satisfy the natural growth condition with respect to gradient of the generalized solutions, the maximum principle is proved for the bounded generalized solution of quasi_linear elliptic equations.

文摘Broad beans were divided into six groups and implanted with N+ beam of 30 KeV, 8 × 1016/cm2 per time for various radiating times respectively. Besides the statistics of its vigor of germination, the M1 root-tip cells of these broad beans were systematically analyzed on their changes in mitotic percentage, morphology and behavior of chromosomes, along with the structure o f cytoskeletons, including microtubule and intermediate filament. Based on all results of these studies, our opinions have been expressed in the report on the mechanism of low-energy N+ beams effecting on higher dicotyledons such as broad beau.
文摘The effects of Al additions on the microstructure and mechanical properties of MoSi2 have been studied. Bending strength and fracture toughness reach the highest value of 350 MPa, 4.05 MPa.m1/2, respectively, at ambient temperature when Al addition was of 3.5 wt pct.
文摘Stable operating region in the HL-1M tokamak has been extended by means of wall conditioning, core fuelling and current control techniques. The slope with a large Murakami coefficient increased by a factor of 50-60 %.

Our opinions are presented as follows:
文摘Through the development in the past 20 years, the compliant mechanism has become an important branch of modern mechanisms and is widely applied in such fields as precision engineering and robotics. Although a series of design methods and theories of compliant mechanism have emerged, its design problem is still facing challenge. A comprehensive overview of design methods is provided for compliant mechanisms used for precision applications, usually called flexure mechanisms. Several main approaches to analysis and design of flexure mechanisms, including pseudo-rigid-body model, structural matrix model,
