Taking advantage of this fact, we may find out whether there are microtubule organizing centers in the interphasic nuclei and whether nuclear membrane does play any role in the formation of microtubules, if the isolated nuclei are mixed with a solution of tubulin and let the tubulin in the mixture polymerize. 目的了解异染色质相关蛋白1-γ(heterochro-Matin-associated protein 1,HP1-γ)在食管癌发生发展各阶段的表达情况,并初步探讨利用激光扫描细胞仪分析免疫组化结果较传统光镜下评分方法的优势。方法选用含各级别食管癌病变组织的组织芯...目的了解异染色质相关蛋白1-γ(heterochro-Matin-associated protein 1,HP1-γ)在食管癌发生发展各阶段的表达情况,并初步探讨利用激光扫描细胞仪分析免疫组化结果较传统光镜下评分方法的优势。方法选用含各级别食管癌病变组织的组织芯片,采用免疫组化方法检测其中HP1-γ的表达,并通过激光扫描细胞仪对免疫组化结果进行定量分析。将激光扫描细胞仪分析结果和传统光镜下评分结果进行相关性分析。结果HP1-γ蛋白主要表达于胞核。在正常食管组织、中-重度异型增生、原位癌、食管鳞癌的阳性率分别为37.5%(3/8),100%(21/21)、100%(7/7)和23.7%(9/38),且在正常食管、中-重度异型增生加原位癌、食管鳞癌组织中的表达差异有统计学意义(P<0.01)。激光扫描细胞仪分析结果与传统光镜下评分结果具有高度相关性(P<0.01)。结论HP1-γ蛋白可能在食管正常细胞向恶性细胞转化的过程中起到了一定的抵抗作用。与传统人工评分方法相比,激光扫描细胞仪在分析免疫组化结果时具有高通量、高度客观、准确的优点。展开更多
