Inhibiting GRK2 is a promising mechanism for the treatment of heart failure (HF). Further development and optimization of inhibitors targeting GRK2 are highly meaningful. Therefore, in order to design GRK2 inhibitors with better performance, the most active molecule was selected as a reference compound from a data set containing 4-pyridylhydrazone derivatives and triazole derivatives, and its scaffold was extracted as the initial scaffold. Then, a powerful optimization-based framework for de novo drug design, guided by binding affinity, was used to generate a virtual molecular library targeting GRK2. The binding affinity of each virtual compound in this dataset was predicted by our developed deep learning model, and the designed potential compound with high binding affinity was selected for molecular docking and molecular dynamics simulation. It was found that the designed potential molecule binds to the ATP site of GRK2, which consists of key amino acids including Arg199, Gly200, Phe202, Val205, Lys220, Met274 and Asp335. The scaffold of the molecule is stabilized mainly by H-bonding and hydrophobic contacts. Concurrently, the reference compound in the dataset was also simulated by docking. It was found that this molecule also binds to the ATP site of GRK2. In addition, its scaffold is stabilized mainly by H-bonding and π-cation stacking interactions with Lys220, as well as hydrophobic contacts. The above results show that the designed potential molecule has similar binding modes to the reference compound, supporting the effectiveness of our framework for activity-focused molecular design. Finally, we summarized the interaction characteristics of general GRK2 inhibitors and gained insight into their molecule-target binding mechanisms, thereby facilitating the expansion of lead to hit compound.展开更多

These methods are also used for deriving mathematical models for BVPs and IVPs associated with isotropic, homogeneous as well as non-homogeneous, non-isotropic continuous matter. In energy methods when applied to IVPs, one constructs energy functional (<i>I</i>) consisting of kinetic energy, strain energy and the potential energy of loads. The first variation of this energy functional (<em>δI</em>) set to zero is a necessary condition for an extremum of <i>I</i>. In this approach one could use <i>δI</i> = 0 directly in constructing computational processes such as the finite element method or could derive Euler’s equations (differential or partial differential equations) from <i>δI</i> = 0, which is also satisfied by a solution obtained from <i>δI</i> = 0. The Euler’s equations obtained from <i>δI</i> = 0 indeed are the mathematical model associated with the energy functional <i>I</i>. In case of BVPs we follow the same approach except in this case, the energy functional <i>I</i> consists of strain energy and the potential energy of loads. In using the principle of virtual work for BVPs and the IVPs, we can also accomplish the same as described above using energy methods. In this paper we investigate consistency and validity of the mathematical models for isotropic, homogeneous and non-isotropic, non-homogeneous continuous matter for BVPs that are derived using energy functional consisting of strain energy and the potential energy of loads. Similar investigation is also presented for IVPs using energy functional consisting of kinetic energy, strain energy and the potential energy of loads. The computational approaches for BVPs and the IVPs designed using energy functional and principle of virtual work, their consistency and validity are also investigated. Classical continuum mechanics (CCM) principles <i>i.e.</i> conservation and balance laws of CCM with consistent constitutive theories and the elements of calculus of variations are employed in the investigations presented in this paper.展开更多

The computational fluid dynamics was used to analyze the flow features of the sloping U-shape notch on the spool, such as mass flow rates, flow coefficients, effiux angles and steady state flow forces under different operating conditions. At last, the reliability of the mathematical model of the flow area for the sloping U-shape notch orifice on the spool was demonstrated by the comparison between the orifice area curve derived and the corresponding experimental data provided by the test. It is presented that the bottom arc of sloping U-shape notch (ABU) should not be omitted when it is required to accurately calculate the orifice area of ABU. Although the theoretical flow area of plain bottom sloping U-shape notch (PBU) is larger than that of ABU at the same opening, the simulated mass flow and experimental flow area of ABU are both larger than these of PBU at the same opening, while the simulated flow force of PBU is larger than that of ABU at the same opening. Therefore, it should be prior to adapt the ABU when designing the spool with proportional character.展开更多

These equations can be solved with numerical methods. Initial and boundary conditions are needed for such of these numerical methods. Predication and simulations for different case studies are major sources for the great importance of these models. Satellite data from different wide ranges of sensors provide observations that indicate system state. So both numerical models and satellite data provide estimation of system states, and between the different estimations it is required the best estimate for system state. Assimilation of observations in numerical weather models with data assimilation techniques provide an improved estimate of system states. In this work, highlights on the mathematical perspective for data assimilation methods are introduced. Least square estimation techniques are introduced because it is considered the basic mathematical building block for data assimilation methods. Stochastic version of least square is included to handle the error in both model and observation. Then the three and four dimensional variational assimilation 3dvar and 4dvar respectively will be handled. Kalman filters and its derivatives Extended, (KF, EKF, ENKF) and hybrid filters are introduced.展开更多

文摘This mixed-methods study investigated how AI-enhanced English as a Foreign Language(EFL)learning environments influence students’psychological well-being through the mediating roles of motivation and language learning anxiety and the moderating role of trust.Participants were Chinese university students(N=310,62%female,mean age=18.9,SD=0.8),of whom 15 completed interviews to both add to and to clarify the evidence from the surveys.Structural equation modeling results revealed that AI use had significant indirect effects on well-being through increased motivation and reduced language learning anxiety.Trust in AI significantly moderated both paths, amplifying the motivational benefits and anxiety reduction associated with AI use.Thematic analysis supported these results, identifying three experiential themes:(1)motivational empowerment through personalization,(2)anxiety regulation through safe practice and feedback,and(3)trust as the emotional bridge between AI and well-being.The study extends AI psychology applications by empirically linking technology engagement with affective outcomes and underscores the need for human-centered and trust-enhancing design in AI-supported education. From these findings,we conclude that adaptive, transparent, and autonomy-supportive AI systems promote self-determined motivation, emotional safety, and overall psychological health among EFL learners.

文摘Yang Hui was one of the most important authors of mathematical works during the thirteenth century.Mathematical Methods Explaining in Detail The Nine Chapters(Xiangjie jiuzhang suanfa詳解九章算法,1261 CE)is the earliest extant work attributed to Yang Hui.From the thirteenth to the fifteenth century, this work played a crucial role in the circulation and popularization of The Nine Chapters on Mathematical Procedures(Jiuzhang suanshu九章算术).However, the only surviving printed edition of Mathematical Methods is incomplete and contains many mistakes obstructing contemporary researchers'understanding of this work. The "Fangcheng" chapter of The Nine Chapters deals with problems related to solving what today are known as simultaneous sets of linear equations. However, interpreting the text in this chapter of Mathematical Methods and recovering the mathematical practices relating to fangcheng are difficult.Through detailed textual and mathematical analyses, the author of this paper explains Yang Hufs understanding and practice relating to〃the fangcheng method"and"the method of the positive and the negative".This paper includes an appendix that provides a detailed translation of the ambiguous text relating to"the method of the positive and the negative"and gives reasons supporting the interpretation provided here.Yang Hufs understanding of the concepts of"positive"and"negative"and his practice relating to these two concepts may easily be confused with their apparent counterparts in modem mathematics. Also, careful analysis of the mathematical methods in this work reveal that the order of problems in Yang Hufs Reclassifications of Mathematical Methods Explaining in Detail The Nine Chapters([Xiangjie jiuzhang suanfa zuanlei詳解九章算法纂類],namely, the last section of Mathematical Methods)were rearranged according to commentaries to specific methods that appear in Mathematical Methods. Some textual clues referring to the zzprevious question"(qianwen前問)in certain commentaries of Mathematical Methods indeed reflect the order of problems in Reclassifications.Yang Hui made especially detailed commentaries on the problems that he arranged in a sequence that differs with respect to the original order of problems as they appear in the ancient classic work,The Nine Chapters. All these discoveries reveal and serve to prove a close relationship between Yang Hufs Mathematical Methods and his Reclassifications.
文摘Energy methods and the principle of virtual work are commonly used for obtaining solutions of boundary value problems (BVPs) and initial value problems (IVPs) associated with homogeneous, isotropic and non-homogeneous, non-isotropic matter without using (or in the absence of) the mathematical models of the BVPs and the IVPs. Classical continuum mechanics (CCM) principles <i>i.e.</i> conservation and balance laws of CCM with consistent constitutive theories and the elements of calculus of variations are employed in the investigations presented in this paper.

文摘Precise function expression of the flow area for the sloping U-shape notch orifice versus the spool stroke was derived. Therefore, it should be prior to adapt the ABU when designing the spool with proportional character.
文摘The design of evolutionary approaches has a vital role in the recent development of scientific literature. To tackle highly nonlinear complex problems, nonlinear ordinary differential equations, partial differential equations, stochastic differential equations, and many more may called computational algorithms. The rotavirus causes may include severe diarrhea, vomiting, and fever leading to rapid dehydration.By the report of the World Health Organization(WHO),approximately 600,000 children die worldwide each year,80 percent of whom live in developing countries. Two million children are hospitalized each year.In Asia,up to 45 percent of the children hospitalized for diarrhea may be infected with rotavirus. The rotavirus model is categorized into five-compartment like susceptible(S),breastfeeding(M),vaccinated(V),infected(I),and recovered children(R).Positivity, boundedness, equilibria, reproduction number, and stability results are part of the qualitative analysis of the model. After that, the design of the evolutionary approaches on the model predicts the efficiency,visualization,long-term behavior of the disease, and best results of the rotavirus disease. In the end, evolutionary computations are an appropriate tool for double-checking the qualitative examination of the model.

文摘this paper,we propose a class of smoothing-regularization methods for solving the mathematical programming with vanishing constraints. These methods include the smoothing-regularization method proposed by Kanzow et al.in[Comput.Optim.Appl.,2013,55(3):733-767]as a special case.Under the weaker conditions than the ones that have been used by Kanzow et al.in 2013,we prove that the Mangasarian-Fromovitz constraint qualification holds at the feasible points of smoothing-regularization problem. We also analyze that the convergence behavior of the proposed smoothing-regularization method under mild conditions,i.e.,any accumulation point of the stationary point sequence for the smoothing-regularization problem is a strong stationary point. Finally, numerical experiments are given to show the efficiency of the proposed methods.
文摘In this paper, we investigate some new traveling wave solutions to Vakhnenko-Parkes equation via three modified mathematical methods. The derived solutions have been obtained including periodic and solitons solutions in the form of trigonometric, hyperbolic, and rational function solutions. The graphical representations of some solutions by assigning particular values to the parameters under prescribed conditions in each solutions and comparing of solutions with those gained by other authors indicate that these employed techniques are more effective, efficient and applicable mathematical tools for solving nonlinear problems in applied science.

文摘Modal parameters can accurately characterize the structural dynamic properties and assess the physical state of the structure. Therefore,it is particularly significant to identify the structural modal parameters according to the monitoring data information in the structural health monitoring(SHM)system,so as to provide a scientific basis for structural damage identification and dynamic model modification. In view of this, this paper reviews methods for identifying structural modal parameters under environmental excitation and briefly describes how to identify structural damages based on the derived modal parameters. The paper primarily introduces data-driven modal parameter recognition methods(e.g.,time-domain,frequency-domain,and time-frequency-domain methods, etc.),briefly describes damage identification methods based on the variations of modal parameters(e.g.,natural frequency, modal shapes, and curvature modal shapes, etc.)and modal validation methods(e.g.,Stability Diagram and Modal Assurance Criterion,etc.).The current status of the application of artificial intelligence(AI)methods in the direction of modal parameter recognition and damage identification is further discussed.Based on the pre-vious analysis, the main development trends of structural modal parameter recognition and damage identification methods are given to provide scientific references for the optimized design and functional upgrading of SHM systems.
文摘Environmental systems including our atmosphere oceans, biological… etc. can be modeled by mathematical equations to estimate their states. Kalman filters and its derivatives Extended, (KF, EKF, ENKF) and hybrid filters are introduced.

文摘As pivotal supporting technologies for smart manufacturing and digital engineering,model-based and data-driven methods have been widely applied in many industrial fields, such as product design, process monitoring, and smart maintenance. While promising, both methods have issues that need to be addressed. For example,model-based methods are limited by low computational accuracy and a high computational burden, and data-driven methods always suffer from poor interpretability and redundant features. To address these issues, the concept of data-model fusion(DMF)emerges as a promising solution.DMF involves integrating model-based methods with data-driven methods by incorporating big data into model-based methods or embedding relevant domain knowledge into data-driven methods.Despite growing efforts in the field of DMF,a unanimous definition of DMF remains elusive, and a general framework of DMF has been rarely discussed. This paper aims to address this gap by providing a thorough overview and categorization of both data-driven methods and model-based methods.Subsequently, this paper also presents the definition and categorization of DMF and discusses the general framework of DMF.Moreover, the primary seven applications of DMF are reviewed within the context of smart manufacturing and digital engineering. Finally, this paper directs the future directions of DMF.

文摘The separation-of-variable(SOV)methods,such as the improved SOV method, the variational SOV method, and the extended SOV method, have been proposed by the present authors and coworkers to obtain the closed-form analytical solutions for free vibration and eigenbuckling of rectangular plates and circular cylindrical shells.By taking the free vibration of rectangular thin plates as an example, this work presents the theoretical framework of the SOV methods in an instructive way,and the bisection–based solution procedures for a group of nonlinear eigenvalue equations.Besides, the explicit equations of nodal lines of the SOV methods are presented, and the relations of nodal line patterns and frequency orders are investigated. It is concluded that the highly accurate SOV methods have the same accuracy for all frequencies, the mode shapes about repeated frequencies can also be precisely captured, and the SOV methods do not have the problem of missing roots as well.

文摘With the continuous increase of aeroengine flight ceiling(>20 km),the thin atmosphere at high altitudes and the size effect all cause the compressor component inlet Reynolds number to decrease rapidly to a critical value(approximately 2.0×10^(5)),and the significant transition process on the blade/endwall surface leads to the sharp degradation of compressor performance, which seriously affects the engine fuel consumption and working stability at high altitudes. In this paper, the research progress on the internal flow mechanism and flow control methods of axial compressors at low Reynolds numbers is reviewed from the aspects of quantification and prediction of performance variation, flow loss mechanism related to separation and transition, efficient transition control and flow field organization. The development trend of the low-Reynolds-number effect of axial flow compressors is noted, and the difficulties and application prospects of aerodynamic design and efficient flow control methods for compressors under low Reynolds numbers at high altitudes are discussed.
