BACKGROUND Gastric cancer is a major global health concern,often diagnosed at advanced stages,leading to poor prognosis.Proximal and distal gastric cancers exhibit distinct clinicopathological features.AIM To investig...BACKGROUND Gastric cancer is a major global health concern,often diagnosed at advanced stages,leading to poor prognosis.Proximal and distal gastric cancers exhibit distinct clinicopathological features.AIM To investigate the diagnostic value of hematological and inflammatory markers in differentiating proximal and distal gastric cancers and to evaluate their association with clinical outcomes.METHODS A retrospective cohort study was conducted on 150 patients diagnosed with gastric adenocarcinoma through histopathological analysis.Patients were categorized into proximal gastric cancer and distal gastric cancer groups.Laboratory parameters were analyzed.RESULTS Of the 150 patients,84 had proximal gastric cancer and 66 had distal gastric cancer.Dysphagia was significantly more common in the proximal gastric cancer group,while anemia and higher platelet-to-lymphocyte ratio values were observed in the distal gastric cancer group(P=0.031).Tumor stage and neutrophil-to-lymphocyte ratio emerged as independent predictors of all-cause mortality.No significant differences were found in other laboratory or biochemical parameters between the groups.CONCLUSION Proximal and distal gastric cancers demonstrate distinct clinical and laboratory profiles.The platelet-to-lymphocyte ratio may serve as a valuable marker in differentiating cancer localization,while the neutrophil-to-lymphocyte ratio is a prognostic indicator for mortality.These findings highlight the potential of hematological markers in optimizing diagnosis and treatment strategies for gastric cancer.展开更多

Background Cotton is a significant crop for fiber production;however,seed shape-related traits have been less investigated in comparison to fiber quality.Comprehending the genetic foundation of traits associated with ...Background Cotton is a significant crop for fiber production;however,seed shape-related traits have been less investigated in comparison to fiber quality.Comprehending the genetic foundation of traits associated with seed shape is crucial for improving the seed and fiber quality in cotton.Results A total of 238 cotton accessions were evaluated in four different environments over a period of two years.Traits including thousand grain weight(TGW),aspect ratio(AR),seed length,seed width,diameter,and roundness demonstrated high heritability and significant genetic variation,as indicated by phenotypic analysis.The association analysis involved 145 simple sequence repeats(SSR)markers and identified 50 loci significantly associated with six traits related to seed shape.The markers MON_DPL0504aa and BNL2535ba were identified as influencing multiple traits,including aspect ratio and thousand grain weight.Notably,markers such as HAU2588a and MUSS422aa had considerable influence on seed diameter and roundness.The identified markers represented an average phenotypic variance between 3.92%for seed length and 16.54%for TGW.Conclusions The research finds key loci for seed shape-related traits in cotton,providing significant potential for marker-assisted breeding.These findings establish a framework for breeding initiatives focused on enhancing seed quality,hence advancing the cotton production.展开更多
BACKGROUND Familial Mediterranean fever(FMF)is an autosomal recessive autoinflammatory disorder marked by recurrent episodes of fever and serositis.Resistin,a proinflammatory cytokine,may play a role in FMF pathogenes...BACKGROUND Familial Mediterranean fever(FMF)is an autosomal recessive autoinflammatory disorder marked by recurrent episodes of fever and serositis.Resistin,a proinflammatory cytokine,may play a role in FMF pathogenesis by promoting the release of interleukin-1beta,tumour necrosis factor alpha,and interleukin-6.AIM To evaluate serum resistin levels in children with FMF during acute attacks and remission,and to assess its potential as a biomarker for disease activity and progression.METHODS A case-control study was conducted involving 40 pediatric patients with FMF and 40 age-and sex-matched healthy controls.Serum resistin and inflammatory markers—including total leukocyte count(TLC),erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),serum amyloid A(SAA),and fibrinogen—were measured using enzyme-linked immunosorbent assay and standard assays.RESULTS No significant differences were found in age or sex between FMF patients and controls.Among FMF patients,fever was the most prevalent symptom(95%),followed by abdominal pain(75%).The most frequently detected genetic mutation was M694I,followed by M694V,E148Q,M680I,and V726A.Compound heterozygous mutations,including M694I/V726A and M694I/M694V,were equally represented.During acute attacks,FMF patients exhibited significantly elevated levels of TLC,ESR,CRP,SAA,and fibrinogen compared to attack-free periods and controls.Serum resistin levels were markedly higher during acute attacks and showed a strong positive correlation with other acute inflammatory markers.Receiver operating characteristic curve analysis demonstrated high sensitivity and specificity of resistin as a potential biomarker for FMF.CONCLUSION Resistin is significantly elevated in children with FMF during acute episodes and correlates with established inflammatory markers.These findings support its potential role as a non-invasive biomarker for disease activity and severity in pediatric FMF.展开更多
BACKGROUND Metastatic colorectal cancer(mCRC)is a global health challenge with a poor prognosis.Prognostic markers are critical for survival prediction.METHODS This multicenter,retrospective study measured baseline ca...BACKGROUND Metastatic colorectal cancer(mCRC)is a global health challenge with a poor prognosis.Prognostic markers are critical for survival prediction.METHODS This multicenter,retrospective study measured baseline carcinoembryonic antigen and carbohydrate antigen 19-9 levels to calculate a TMI as the geometric mean of values normalized to their upper limits of normal.Receiver operating characteristic curve analysis assessed TMI’s prognostic accuracy,and patients were stratified into high-TMI(≥1.39)and low-TMI(<1.39)groups.The primary endpoint was overall survival(OS),with progression-free survival and treatment response as secondary endpoints.RESULTS The study included 305 mCRC patients with a median follow-up of 22.9 months.The median OS for high-TMI patients was 29.5 months,significantly lower than the 45.6 months observed in the low-TMI group(P=0.02).The 2-year OS rates for the high-and low-TMI groups were 59.4%and 72.9%,respectively.Median progression-free survival was also shorter for the high-TMI group(14.0 vs 16.0 months,P=0.84).High TMI is an independent prognostic factor for worse OS.CONCLUSION TMI is a simple,cost-effective prognostic tool for mCRC,with high TMI associated with poorer survival outcomes.展开更多
Peel color is an important appearance quality of melons that significantly affects consumer preferences.In this study,a near-isogenic line NIL-G(dark green peel)was generated from B8(grey-green peel)and B15(white peel...Peel color is an important appearance quality of melons that significantly affects consumer preferences.In this study,a near-isogenic line NIL-G(dark green peel)was generated from B8(grey-green peel)and B15(white peel).The F_2 population constructed by crossing NIL-G and B15 was used to study the inheritance pattern of peel color,and bulked-segregant analysis sequencing(BSA-seq)was employed to identify the interval in which the target gene was located.Genetic analysis showed that a dominant gene controls the dark green peel trait at maturity.BSAseq and molecular markers were used to localize the candidate gene in a 263.7 kb interval of chromosome 4,which contained the CmAPRR2 gene with known functions.Moreover,allelic sequence analysis revealed four SNP variations of the CmAPRR2 gene in B15,of which SNP.G614331A was located at the junction of the 6th exon and 6th intron.The G-to-A mutation caused alternative splicing of the transcript of CmAPRR2 in B15,generating two transcripts(CmAPRR2-A and CmAPRR2-B)with premature termination codons.Furthermore,the Kompetitive Allele Specific PCR(KASP)marker,APRR2-G/A,was developed based on this SNP and shown to co-segregate with the peel color phenotype in the F_(2) population.Compared to white-peel B15,the expression level of CmAPRR2 in dark green peel NIL-G was higher at each growth stage.Therefore,CmAPRR2 may be the key gene controlling the fruit color of melons.This study identified a novel allelic variant of CmAPRR2 that leads to white peel formation in mature melons.We also provides a theoretical basis for further research on the gene regulatory mechanism of melon peel colors,which promotes using molecular marker-assisted selection to modify melon peel colors in the future.展开更多
BACKGROUND An increasing number of studies to date have found preoperative magnetic resonance imaging(MRI)features valuable in predicting the prognosis of rectal cancer(RC).However,research is still lacking on the cor...BACKGROUND An increasing number of studies to date have found preoperative magnetic resonance imaging(MRI)features valuable in predicting the prognosis of rectal cancer(RC).However,research is still lacking on the correlation between preoperative MRI features and the risk of recurrence after radical resection of RC,urgently necessitating further in-depth exploration.AIM To investigate the correlation between preoperative MRI parameters and the risk of recurrence after radical resection of RC to provide an effective tool for predicting postoperative recurrence.METHODS The data of 90 patients who were diagnosed with RC by surgical pathology and underwent radical surgical resection at the Second Affiliated Hospital of Bengbu Medical University between May 2020 and December 2023 were collected through retrospective analysis.General demographic data,MRI data,and tumor markers levels were collected.According to the reviewed data of patients six months after surgery,the clinicians comprehensively assessed the recurrence risk and divided the patients into high recurrence risk(37 cases)and low recurrence risk(53 cases)groups.Independent sample t-test andχ2 test were used to analyze differences between the two groups.A logistic regression model was used to explore the risk factors of the high recurrence risk group,and a clinical prediction model was constructed.The clinical prediction model is presented in the form of a nomogram.The receiver operating characteristic curve,Hosmer-Lemeshow goodness of fit test,calibration curve,and decision curve analysis were used to evaluate the efficacy of the clinical prediction model.RESULTS The detection of positive extramural vascular invasion through preoperative MRI[odds ratio(OR)=4.29,P=0.045],along with elevated carcinoembryonic antigen(OR=1.08,P=0.041),carbohydrate antigen 125(OR=1.19,P=0.034),and carbohydrate antigen 199(OR=1.27,P<0.001)levels,are independent risk factors for increased postoperative recurrence risk in patients with RC.Furthermore,there was a correlation between magnetic resonance based T staging,magnetic resonance based N staging,and circumferential resection margin results determined by MRI and the postoperative recurrence risk.Additionally,when extramural vascular invasion was integrated with tumor markers,the resulting clinical prediction model more effectively identified patients at high risk for postoperative recurrence,thereby providing robust support for clinical decision-making.CONCLUSION The results of this study indicate that preoperative MRI detection is of great importance for predicting the risk of postoperative recurrence in patients with RC.Monitoring these markers helps clinicians identify patients at high risk,allowing for more aggressive treatment and monitoring strategies to improve patient outcomes.展开更多
One of agriculture’s major challenges is the low efficiency of phosphate(Pi)use,which leads to increased costs,harmful environmental impacts,and the depletion of phosphorus(P)resources.The TaPHT1;6 gene,which encodes...One of agriculture’s major challenges is the low efficiency of phosphate(Pi)use,which leads to increased costs,harmful environmental impacts,and the depletion of phosphorus(P)resources.The TaPHT1;6 gene,which encodes a high-affinity Pi transporter(PHT),plays a crucial role in Pi absorption and transport.In this study,the promoter and coding regions of three TaPHT1;6 gene copies on chromosomes 5A,5B,and 5D were individually amplified and sequenced from 167 common wheat(Triticum aestivum L.)cultivars.Sequence analysis revealed 16 allelic variation sites within the promoters of TaPHT1;6-5B among these cultivars,forming three distinct haplotypes:Hap1,Hap2,and Hap3.Field trials were conducted over two years to compare wheat genotypes with these haplotypes,focusing on assessing plant dry weight,grain yield,P content,Pi fertilizer absorption efficiency,and Pi fertilizer utilization efficiency.Results indicated that Hap3 represented the favored Pi-efficient haplotype.Dual-luciferase reporter assay demonstrated that the Hap3 promoter,carrying the identified allelic variation sites,exhibited higher gene-driven capability,leading to increased expression levels of the TaPHT1;6-5B gene.We developed a distributed cleaved amplified polymorphic site marker(dCAPS-571)to distinguish Hap3 from the other two haplotypes based on these allelic variation sites,presenting an opportunity for breeding Pi-efficient wheat cultivars.This study successfully identified polymorphic sites on TaPHT1;6-5B associated with Pi efficiency and developed a functional molecular marker to facilitate future breeding endeavors.展开更多
In order to save manpower and time costs,and to achieve simultaneous detection of multiple animal-derived components in meat and meat products,this study used multiple nucleotide polymorphism(MNP)marker technology bas...In order to save manpower and time costs,and to achieve simultaneous detection of multiple animal-derived components in meat and meat products,this study used multiple nucleotide polymorphism(MNP)marker technology based on the principle of high-throughput sequencing,and established a multi-locus 10 animalderived components identification method of cattle,goat,sheep,donkey,horse,chicken,duck,goose,pigeon,quail in meat and meat products.The specific loci of each species could be detected and the species could be accurately identified,including 5 loci for cattle and duck,3 loci for sheep,9 loci for chicken and horse,10 loci for goose and pigeon,6 loci for quail and 1 locus for donkey and goat,and an adulteration model was established to simulate commercially available samples.The results showed that the method established in this study had high throughput,good repeatability and accuracy,and was able to identify 10 animalderived components simultaneously with 100%repeatability accuracy.The detection limit was 0.1%(m/m)in simulated samples of chicken,duck and horse.Using the method established in this study to test commercially available samples,4 samples from 14 commercially available samples were detected to be inconsistent with the labels,of which 2 did not contain the target ingredient and 2 were adulterated with small amounts of other ingredients.展开更多
BACKGROUND Colorectal cancer(CRC)is a prevalent malignant neoplasm characterized by subtle early manifestations.AIM To investigate the correlation among serum lipid profiles,the triglyceride-glucose(TyG)index,and the ...BACKGROUND Colorectal cancer(CRC)is a prevalent malignant neoplasm characterized by subtle early manifestations.AIM To investigate the correlation among serum lipid profiles,the triglyceride-glucose(TyG)index,and the atherosclerotic index(AI)in patients with CRC.Furthermore,it explored the clinical diagnostic utility of combining serum lipids with cancer antigens in the context of CRC.METHODS A retrospective analysis encompassed 277 patients with CRC and 1034 healthy individuals.RESULTS Following propensity score matching,patients with CRC exhibited significantly reduced levels of serum triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol,and low-density lipoprotein cholesterol(LDL-C),as well as a diminished TyG index.Conversely,they displayed elevated AI levels compared to their healthy counterparts.Patients in advanced stages exhibited lower serum levels of TG,TC,and LDL-C compared to those in early stages.Patients with positive lymph node metastasis demonstrated reduced levels of TG,LDL-C,and the TyG index.Receiver operating characteristic analysis revealed that the combination of the TyG index,carcinoembryonic antigen,and carbohydrate antigen 19-9 yielded the highest positive prediction rate for CRC at 75.3%.CONCLUSION Preoperative serum lipid profiles exhibit a robust association with patients with CRC.The concurrent assessment of multiple serum lipids and cancer antigens effectively enhances the diagnostic accuracy for CRC.展开更多
BACKGROUND Non-ST segment elevation myocardial infarction(NSTEMI)poses significant challenges in clinical management due to its diverse outcomes.Understanding the prognostic role of hematological parameters and derive...BACKGROUND Non-ST segment elevation myocardial infarction(NSTEMI)poses significant challenges in clinical management due to its diverse outcomes.Understanding the prognostic role of hematological parameters and derived ratios in NSTEMI patients could aid in risk stratification and improve patient care.AIM To evaluate the predictive value of hemogram-derived ratios for major adverse cardiovascular events(MACE)in NSTEMI patients,potentially improving clinical outcomes.METHODS A prospective,observational cohort study was conducted in 2021 at the Internal Medicine Clinic of the University Hospital in Tuzla,Bosnia and Herzegovina.The study included 170 patients with NSTEMI,who were divided into a group with MACE and a control group without MACE.Furthermore,the MACE group was subdivided into lethal and non-lethal groups for prognostic analysis.Alongside hematological parameters,an additional 13 hematological-derived ratios(HDRs)were monitored,and their prognostic role was investigated.RESULTS Hematological parameters did not significantly differ between non-ST segment elevation myocardial infarction(NSTEMI)patients with MACE and a control group at T1 and T2.However,significant disparities emerged in HDRs among NSTEMI patients with lethal and non-lethal outcomes post-MACE.Notably,neutrophil-to-lymphocyte ratio(NLR)and platelet-to-lymphocyte ratio(PLR)were elevated in lethal outcomes.Furthermore,C-reactive protein-to-lymphocyte ratio(CRP/Ly)at T1(>4.737)demonstrated predictive value[odds ratio(OR):3.690,P=0.024].Both NLR at T1(>4.076)and T2(>4.667)emerged as significant predictors,with NLR at T2 exhibiting the highest diagnostic performance,as indicated by an area under the curve of 0.811(95%CI:0.727-0.859)and OR of 4.915(95%CI:1.917-12.602,P=0.001),emphasizing its important role as a prognostic marker.CONCLUSION This study highlights the significant prognostic value of hemogram-derived indexes in predicting MACE among NSTEMI patients.During follow-up,NLR,PLR,and CRP/Ly offer important insights into the inflammatory processes underlying cardiovascular events.展开更多
The decreasing of hazard ratio for cancer incidence (HRCVD) in the range of twenty native tissues (lung, liver, brain, hematologic, neuroendocrine renal, pancreas, lymphoid, bladder, colon, lip-oral-head-neck, leukocy...The decreasing of hazard ratio for cancer incidence (HRCVD) in the range of twenty native tissues (lung, liver, brain, hematologic, neuroendocrine renal, pancreas, lymphoid, bladder, colon, lip-oral-head-neck, leukocytic, rectum and anus, thyroid, soft tissue, prostate, skin, ovarian, breast, uterine) as much, as decreasing of the level of cardiovascular pathology (CVD) in the host before malignization, have been described by C.F. Bell et al. in 2023. Earlier, in 2022, the decreasing of 5-year mortality from cancer in similar range of tissues discussed by us as the inverse dependence from the content of stem CD34 markers in tissues before malignization, with example of population in England. In present article we investigate the interrelation between both data more thoroughly, using accessible and more representative populations level of the data. The analysis shows that high level of HRCVD is able to predicts only high cancer death for tissue sites in the beginning of the range, being applied to the referent data of cancer cases and deaths in estimated population of USA 2024. Along with this, an increasing the content of CD34 stem marker in the native tissues of the same range was favorite for increasing of cancer’s cases at the end of the range, diminishing, in parallel, the signs of vasculo-endothelial pathology, i.e. HR CVD. Thus, the cases (incidence) of cancer depend directly rather from content of CD34, which preexisted in native sites, than that from HRCVD. Further analysis shows that CD34 content averaged over twenty cites dominates over that CD2 marker of total T-cells more than 7 times, in oppose to their ratio in the blood. The enhancement of stem CD34 marker in the range of tissues is accompanied by unidirectional rising of its maturing derivatives, vasculo-endothelial CD31 and total T-cells CD2 markers, which contents relate positively to increasing of cancer death in US population 2024. The increase of CD34 decreases cancer mortality (death: cases) in sites, but indirectly, rather due to enhancement of the denominator. The high HRCVD (more than 1.0) in range of 20 tissues, concerns of those of them, which have had highest mitotic activity (by Ki67), but lowest “stemness” (by CD34), “vascularity” (by CD31), cancer’s incidence (cases) and the worse results of therapy. Oppositely, the normal tissue with lowest HRCVD (below 1.0) and Ki67, but highest CD34, CD31, and cancer incidence (cases) are more sensitive to treatment. Thus, the residential hematopoietic “stemness” in native tissues acts as natural protectors for cardio-vascular system and promoter for cancer incidence in them. The steady and irreversible exhaustion of current regenerative resource (CRR) of BM, which assumed by us as a product of CD34 number and average telomeres length, manifests itself in acceleration of non-malignant CVD and deceleration of malignancy in population +70 (in term the death per 105), according to data extracted from WHO Mortality Database. The similar deficit of CD34 arises artificially during cytotoxic treatment of cancer, when rapid waste of local CRR forces malignant cells to search more “stemness” cites. The competition between malignant and native tissues of the host for scanty CRR seems to be the most important factor for evaluation and prediction of prevalence, curability, and long-term results in oncology.展开更多

This cross-sectional study, conducted in Cotonou among 35 azoospermic men, aimed to assess the role of biochemical analysis of seminal fluid in determining the etiological diagnosis. The results revealed significant heterogeneity in biochemical profiles. Most of the patients had normal levels of alpha-glucosidase, fructose, and zinc, indicating normal accessory gland function and suggesting non-obstructive azoospermia. However, a significant subgroup exhibited abnormalities in these markers, suggesting a possible obstructive azoospermia. The heterogeneity in biochemical profiles observed in this study highlights the need for further studies to better characterize azoospermia in Benin.展开更多

Discourse markers(DMs)in many languages have been investigated from various theoretical perspectives and within diverse methodological paradigms for decades.Studies within the field of DMs are accelerating rapidly;how...Discourse markers(DMs)in many languages have been investigated from various theoretical perspectives and within diverse methodological paradigms for decades.Studies within the field of DMs are accelerating rapidly;however,few state-of-the-art reviews have been conducted to systematically inform scholars of the current landscape of DM research.This investigation conducted a comprehensive review of 2,183 studies from 2009 to 2022,leveraging CiteSpace for bibliometric analysis.The study aims to illuminate emerging trends and focal areas within DM research and lay the groundwork for further interdisciplinary exploration and collaboration in this area.The analysis reveals a growing interest in DM studies,identifies leading contributors,including countries,universities,and scholars,highlights the most cited journals and references shaping the field,and delineates prevalent research themes and topics,as well as methodological approaches employed.These findings indicate that DM research has evolved from a linguistic inquiry into an interdisciplinary domain employing varied methodologies such as qualitative and/or quantitative,experimental,and synchronic and/or diachronic.This study can provide important implications for future research in terms of the pragmatic turn,interdisciplinary nature,expansion of data from English to various other languages,and diversified methodologies.展开更多
文摘In this article,we critically appraise the study by Wu et al,which investigated the prognostic value of preoperative inflammatory and nutritional markers in colorectal cancer patients with peritoneal metastasis.This retrospective cohort study included 133 patients.The findings indicate that patients with high neutrophil-to-lymphocyte ratios(NLRs)or lower hemoglobin(Hb)levels have significantly shorter overall survival(OS)than those with lower NLRs or normal Hb levels,respectively.Furthermore,age,carbohydrate antigen 199 levels,the NLR,Hb,and the peritoneal cancer index were identified as independent prognostic factors for OS.A nomogram was subsequently developed,demon-strating its ability to accurately predict the OS of colorectal cancer patients with peritoneal metastasis.This study introduces a potentially valuable prognostic tool for these patients.However,further validation in multicenter cohorts is needed to confirm its clinical applicability,to assess its limitations,and to elucidate the underlying mechanism of these identified biomarkers.

文摘BACKGROUND Gastric cancer is a major global health concern,often diagnosed at advanced stages,leading to poor prognosis.Proximal and distal gastric cancers exhibit distinct clinicopathological features.AIM To investigate the diagnostic value of hematological and inflammatory markers in differentiating proximal and distal gastric cancers and to evaluate their association with clinical outcomes.METHODS A retrospective cohort study was conducted on 150 patients diagnosed with gastric adenocarcinoma through histopathological analysis.Patients were categorized into proximal gastric cancer and distal gastric cancer groups.Laboratory parameters were analyzed.RESULTS Of the 150 patients,84 had proximal gastric cancer and 66 had distal gastric cancer.Dysphagia was significantly more common in the proximal gastric cancer group,while anemia and higher platelet-to-lymphocyte ratio values were observed in the distal gastric cancer group(P=0.031).Tumor stage and neutrophil-to-lymphocyte ratio emerged as independent predictors of all-cause mortality.No significant differences were found in other laboratory or biochemical parameters between the groups.CONCLUSION Proximal and distal gastric cancers demonstrate distinct clinical and laboratory profiles.The platelet-to-lymphocyte ratio may serve as a valuable marker in differentiating cancer localization,while the neutrophil-to-lymphocyte ratio is a prognostic indicator for mortality.These findings highlight the potential of hematological markers in optimizing diagnosis and treatment strategies for gastric cancer.
文摘The retrospective cohort study by Zuo et al investigates the interplay between microsatellite instability(MSI)status,nutritional indicators,and inflammatory profiles in colorectal cancer(CRC).Analyzing 56 patients,the study reveals that MSI-high tumors are associated with significantly lower serum albumin,body mass index,and absolute lymphocyte counts,alongside elevated neutrophil-tolymphocyte ratios compared to microsatellite stable tumors.These findings highlight distinct immunological and nutritional profiles in MSI-high CRC,suggesting potential clinical utility in risk stratification and personalized treat-ment.While the study underscores the importance of MSI status in CRC mana-gement,its single-center design and limited sample size warrant validation through multicenter trials.This article contextualizes these findings within the broader landscape of CRC research,emphasizing the need for integrating bio-markerdriven strategies into clinical practice to optimize outcomes for patients with differing MSI statuses.

文摘Background Cotton is a significant crop for fiber production;however,seed shape-related traits have been less investigated in comparison to fiber quality.Comprehending the genetic foundation of traits associated with seed shape is crucial for improving the seed and fiber quality in cotton.Results A total of 238 cotton accessions were evaluated in four different environments over a period of two years.Traits including thousand grain weight(TGW),aspect ratio(AR),seed length,seed width,diameter,and roundness demonstrated high heritability and significant genetic variation,as indicated by phenotypic analysis.The association analysis involved 145 simple sequence repeats(SSR)markers and identified 50 loci significantly associated with six traits related to seed shape.The markers MON_DPL0504aa and BNL2535ba were identified as influencing multiple traits,including aspect ratio and thousand grain weight.Notably,markers such as HAU2588a and MUSS422aa had considerable influence on seed diameter and roundness.The identified markers represented an average phenotypic variance between 3.92%for seed length and 16.54%for TGW.Conclusions The research finds key loci for seed shape-related traits in cotton,providing significant potential for marker-assisted breeding.These findings establish a framework for breeding initiatives focused on enhancing seed quality,hence advancing the cotton production.
文摘BACKGROUND Familial Mediterranean fever(FMF)is an autosomal recessive autoinflammatory disorder marked by recurrent episodes of fever and serositis.Resistin,a proinflammatory cytokine,may play a role in FMF pathogenesis by promoting the release of interleukin-1beta,tumour necrosis factor alpha,and interleukin-6.AIM To evaluate serum resistin levels in children with FMF during acute attacks and remission,and to assess its potential as a biomarker for disease activity and progression.METHODS A case-control study was conducted involving 40 pediatric patients with FMF and 40 age-and sex-matched healthy controls.Serum resistin and inflammatory markers—including total leukocyte count(TLC),erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),serum amyloid A(SAA),and fibrinogen—were measured using enzyme-linked immunosorbent assay and standard assays.RESULTS No significant differences were found in age or sex between FMF patients and controls.Among FMF patients,fever was the most prevalent symptom(95%),followed by abdominal pain(75%).The most frequently detected genetic mutation was M694I,followed by M694V,E148Q,M680I,and V726A.Compound heterozygous mutations,including M694I/V726A and M694I/M694V,were equally represented.During acute attacks,FMF patients exhibited significantly elevated levels of TLC,ESR,CRP,SAA,and fibrinogen compared to attack-free periods and controls.Serum resistin levels were markedly higher during acute attacks and showed a strong positive correlation with other acute inflammatory markers.Receiver operating characteristic curve analysis demonstrated high sensitivity and specificity of resistin as a potential biomarker for FMF.CONCLUSION Resistin is significantly elevated in children with FMF during acute episodes and correlates with established inflammatory markers.These findings support its potential role as a non-invasive biomarker for disease activity and severity in pediatric FMF.
文摘BACKGROUND Metastatic colorectal cancer(mCRC)is a global health challenge with a poor prognosis.Prognostic markers are critical for survival prediction.METHODS This multicenter,retrospective study measured baseline carcinoembryonic antigen and carbohydrate antigen 19-9 levels to calculate a TMI as the geometric mean of values normalized to their upper limits of normal.Receiver operating characteristic curve analysis assessed TMI’s prognostic accuracy,and patients were stratified into high-TMI(≥1.39)and low-TMI(<1.39)groups.The primary endpoint was overall survival(OS),with progression-free survival and treatment response as secondary endpoints.RESULTS The study included 305 mCRC patients with a median follow-up of 22.9 months.The median OS for high-TMI patients was 29.5 months,significantly lower than the 45.6 months observed in the low-TMI group(P=0.02).The 2-year OS rates for the high-and low-TMI groups were 59.4%and 72.9%,respectively.Median progression-free survival was also shorter for the high-TMI group(14.0 vs 16.0 months,P=0.84).High TMI is an independent prognostic factor for worse OS.CONCLUSION TMI is a simple,cost-effective prognostic tool for mCRC,with high TMI associated with poorer survival outcomes.

文摘Peel color is an important appearance quality of melons that significantly affects consumer preferences.In this study,a near-isogenic line NIL-G(dark green peel)was generated from B8(grey-green peel)and B15(white peel).The F_2 population constructed by crossing NIL-G and B15 was used to study the inheritance pattern of peel color,and bulked-segregant analysis sequencing(BSA-seq)was employed to identify the interval in which the target gene was located.Genetic analysis showed that a dominant gene controls the dark green peel trait at maturity.BSAseq and molecular markers were used to localize the candidate gene in a 263.7 kb interval of chromosome 4,which contained the CmAPRR2 gene with known functions.Moreover,allelic sequence analysis revealed four SNP variations of the CmAPRR2 gene in B15,of which SNP.G614331A was located at the junction of the 6th exon and 6th intron.The G-to-A mutation caused alternative splicing of the transcript of CmAPRR2 in B15,generating two transcripts(CmAPRR2-A and CmAPRR2-B)with premature termination codons.Furthermore,the Kompetitive Allele Specific PCR(KASP)marker,APRR2-G/A,was developed based on this SNP and shown to co-segregate with the peel color phenotype in the F_(2) population.Compared to white-peel B15,the expression level of CmAPRR2 in dark green peel NIL-G was higher at each growth stage.Therefore,CmAPRR2 may be the key gene controlling the fruit color of melons.This study identified a novel allelic variant of CmAPRR2 that leads to white peel formation in mature melons.We also provides a theoretical basis for further research on the gene regulatory mechanism of melon peel colors,which promotes using molecular marker-assisted selection to modify melon peel colors in the future.
文摘BACKGROUND An increasing number of studies to date have found preoperative magnetic resonance imaging(MRI)features valuable in predicting the prognosis of rectal cancer(RC).However,research is still lacking on the correlation between preoperative MRI features and the risk of recurrence after radical resection of RC,urgently necessitating further in-depth exploration.AIM To investigate the correlation between preoperative MRI parameters and the risk of recurrence after radical resection of RC to provide an effective tool for predicting postoperative recurrence.METHODS The data of 90 patients who were diagnosed with RC by surgical pathology and underwent radical surgical resection at the Second Affiliated Hospital of Bengbu Medical University between May 2020 and December 2023 were collected through retrospective analysis.General demographic data,MRI data,and tumor markers levels were collected.According to the reviewed data of patients six months after surgery,the clinicians comprehensively assessed the recurrence risk and divided the patients into high recurrence risk(37 cases)and low recurrence risk(53 cases)groups.Independent sample t-test andχ2 test were used to analyze differences between the two groups.A logistic regression model was used to explore the risk factors of the high recurrence risk group,and a clinical prediction model was constructed.The clinical prediction model is presented in the form of a nomogram.The receiver operating characteristic curve,Hosmer-Lemeshow goodness of fit test,calibration curve,and decision curve analysis were used to evaluate the efficacy of the clinical prediction model.RESULTS The detection of positive extramural vascular invasion through preoperative MRI[odds ratio(OR)=4.29,P=0.045],along with elevated carcinoembryonic antigen(OR=1.08,P=0.041),carbohydrate antigen 125(OR=1.19,P=0.034),and carbohydrate antigen 199(OR=1.27,P<0.001)levels,are independent risk factors for increased postoperative recurrence risk in patients with RC.Furthermore,there was a correlation between magnetic resonance based T staging,magnetic resonance based N staging,and circumferential resection margin results determined by MRI and the postoperative recurrence risk.Additionally,when extramural vascular invasion was integrated with tumor markers,the resulting clinical prediction model more effectively identified patients at high risk for postoperative recurrence,thereby providing robust support for clinical decision-making.CONCLUSION The results of this study indicate that preoperative MRI detection is of great importance for predicting the risk of postoperative recurrence in patients with RC.Monitoring these markers helps clinicians identify patients at high risk,allowing for more aggressive treatment and monitoring strategies to improve patient outcomes.

文摘One of agriculture’s major challenges is the low efficiency of phosphate(Pi)use,which leads to increased costs,harmful environmental impacts,and the depletion of phosphorus(P)resources.The TaPHT1;6 gene,which encodes a high-affinity Pi transporter(PHT),plays a crucial role in Pi absorption and transport.In this study,the promoter and coding regions of three TaPHT1;6 gene copies on chromosomes 5A,5B,and 5D were individually amplified and sequenced from 167 common wheat(Triticum aestivum L.)cultivars.Sequence analysis revealed 16 allelic variation sites within the promoters of TaPHT1;6-5B among these cultivars,forming three distinct haplotypes:Hap1,Hap2,and Hap3.Field trials were conducted over two years to compare wheat genotypes with these haplotypes,focusing on assessing plant dry weight,grain yield,P content,Pi fertilizer absorption efficiency,and Pi fertilizer utilization efficiency.Results indicated that Hap3 represented the favored Pi-efficient haplotype.Dual-luciferase reporter assay demonstrated that the Hap3 promoter,carrying the identified allelic variation sites,exhibited higher gene-driven capability,leading to increased expression levels of the TaPHT1;6-5B gene.We developed a distributed cleaved amplified polymorphic site marker(dCAPS-571)to distinguish Hap3 from the other two haplotypes based on these allelic variation sites,presenting an opportunity for breeding Pi-efficient wheat cultivars.This study successfully identified polymorphic sites on TaPHT1;6-5B associated with Pi efficiency and developed a functional molecular marker to facilitate future breeding endeavors.

文摘In order to save manpower and time costs,and to achieve simultaneous detection of multiple animal-derived components in meat and meat products,this study used multiple nucleotide polymorphism(MNP)marker technology based on the principle of high-throughput sequencing,and established a multi-locus 10 animalderived components identification method of cattle,goat,sheep,donkey,horse,chicken,duck,goose,pigeon,quail in meat and meat products.The specific loci of each species could be detected and the species could be accurately identified,including 5 loci for cattle and duck,3 loci for sheep,9 loci for chicken and horse,10 loci for goose and pigeon,6 loci for quail and 1 locus for donkey and goat,and an adulteration model was established to simulate commercially available samples.The results showed that the method established in this study had high throughput,good repeatability and accuracy,and was able to identify 10 animalderived components simultaneously with 100%repeatability accuracy.The detection limit was 0.1%(m/m)in simulated samples of chicken,duck and horse.Using the method established in this study to test commercially available samples,4 samples from 14 commercially available samples were detected to be inconsistent with the labels,of which 2 did not contain the target ingredient and 2 were adulterated with small amounts of other ingredients.

文摘BACKGROUND Colorectal cancer(CRC)is a prevalent malignant neoplasm characterized by subtle early manifestations.AIM To investigate the correlation among serum lipid profiles,the triglyceride-glucose(TyG)index,and the atherosclerotic index(AI)in patients with CRC.Furthermore,it explored the clinical diagnostic utility of combining serum lipids with cancer antigens in the context of CRC.METHODS A retrospective analysis encompassed 277 patients with CRC and 1034 healthy individuals.RESULTS Following propensity score matching,patients with CRC exhibited significantly reduced levels of serum triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol,and low-density lipoprotein cholesterol(LDL-C),as well as a diminished TyG index.Conversely,they displayed elevated AI levels compared to their healthy counterparts.Patients in advanced stages exhibited lower serum levels of TG,TC,and LDL-C compared to those in early stages.Patients with positive lymph node metastasis demonstrated reduced levels of TG,LDL-C,and the TyG index.Receiver operating characteristic analysis revealed that the combination of the TyG index,carcinoembryonic antigen,and carbohydrate antigen 19-9 yielded the highest positive prediction rate for CRC at 75.3%.CONCLUSION Preoperative serum lipid profiles exhibit a robust association with patients with CRC.The concurrent assessment of multiple serum lipids and cancer antigens effectively enhances the diagnostic accuracy for CRC.
文摘BACKGROUND Non-ST segment elevation myocardial infarction(NSTEMI)poses significant challenges in clinical management due to its diverse outcomes.Understanding the prognostic role of hematological parameters and derived ratios in NSTEMI patients could aid in risk stratification and improve patient care.AIM To evaluate the predictive value of hemogram-derived ratios for major adverse cardiovascular events(MACE)in NSTEMI patients,potentially improving clinical outcomes.METHODS A prospective,observational cohort study was conducted in 2021 at the Internal Medicine Clinic of the University Hospital in Tuzla,Bosnia and Herzegovina.The study included 170 patients with NSTEMI,who were divided into a group with MACE and a control group without MACE.Furthermore,the MACE group was subdivided into lethal and non-lethal groups for prognostic analysis.Alongside hematological parameters,an additional 13 hematological-derived ratios(HDRs)were monitored,and their prognostic role was investigated.RESULTS Hematological parameters did not significantly differ between non-ST segment elevation myocardial infarction(NSTEMI)patients with MACE and a control group at T1 and T2.However,significant disparities emerged in HDRs among NSTEMI patients with lethal and non-lethal outcomes post-MACE.Notably,neutrophil-to-lymphocyte ratio(NLR)and platelet-to-lymphocyte ratio(PLR)were elevated in lethal outcomes.Furthermore,C-reactive protein-to-lymphocyte ratio(CRP/Ly)at T1(>4.737)demonstrated predictive value[odds ratio(OR):3.690,P=0.024].Both NLR at T1(>4.076)and T2(>4.667)emerged as significant predictors,with NLR at T2 exhibiting the highest diagnostic performance,as indicated by an area under the curve of 0.811(95%CI:0.727-0.859)and OR of 4.915(95%CI:1.917-12.602,P=0.001),emphasizing its important role as a prognostic marker.CONCLUSION This study highlights the significant prognostic value of hemogram-derived indexes in predicting MACE among NSTEMI patients.During follow-up,NLR,PLR,and CRP/Ly offer important insights into the inflammatory processes underlying cardiovascular events.
文摘The decreasing of hazard ratio for cancer incidence (HRCVD) in the range of twenty native tissues (lung, liver, brain, hematologic, neuroendocrine renal, pancreas, lymphoid, bladder, colon, lip-oral-head-neck, leukocytic, rectum and anus, thyroid, soft tissue, prostate, skin, ovarian, breast, uterine) as much, as decreasing of the level of cardiovascular pathology (CVD) in the host before malignization, have been described by C.F. Bell et al. in 2023. Earlier, in 2022, the decreasing of 5-year mortality from cancer in similar range of tissues discussed by us as the inverse dependence from the content of stem CD34 markers in tissues before malignization, with example of population in England. In present article we investigate the interrelation between both data more thoroughly, using accessible and more representative populations level of the data. The analysis shows that high level of HRCVD is able to predicts only high cancer death for tissue sites in the beginning of the range, being applied to the referent data of cancer cases and deaths in estimated population of USA 2024. Along with this, an increasing the content of CD34 stem marker in the native tissues of the same range was favorite for increasing of cancer’s cases at the end of the range, diminishing, in parallel, the signs of vasculo-endothelial pathology, i.e. HR CVD. Thus, the cases (incidence) of cancer depend directly rather from content of CD34, which preexisted in native sites, than that from HRCVD. Further analysis shows that CD34 content averaged over twenty cites dominates over that CD2 marker of total T-cells more than 7 times, in oppose to their ratio in the blood. The enhancement of stem CD34 marker in the range of tissues is accompanied by unidirectional rising of its maturing derivatives, vasculo-endothelial CD31 and total T-cells CD2 markers, which contents relate positively to increasing of cancer death in US population 2024. The increase of CD34 decreases cancer mortality (death: cases) in sites, but indirectly, rather due to enhancement of the denominator. The high HRCVD (more than 1.0) in range of 20 tissues, concerns of those of them, which have had highest mitotic activity (by Ki67), but lowest “stemness” (by CD34), “vascularity” (by CD31), cancer’s incidence (cases) and the worse results of therapy. Oppositely, the normal tissue with lowest HRCVD (below 1.0) and Ki67, but highest CD34, CD31, and cancer incidence (cases) are more sensitive to treatment. Thus, the residential hematopoietic “stemness” in native tissues acts as natural protectors for cardio-vascular system and promoter for cancer incidence in them. The steady and irreversible exhaustion of current regenerative resource (CRR) of BM, which assumed by us as a product of CD34 number and average telomeres length, manifests itself in acceleration of non-malignant CVD and deceleration of malignancy in population +70 (in term the death per 105), according to data extracted from WHO Mortality Database. The similar deficit of CD34 arises artificially during cytotoxic treatment of cancer, when rapid waste of local CRR forces malignant cells to search more “stemness” cites. The competition between malignant and native tissues of the host for scanty CRR seems to be the most important factor for evaluation and prediction of prevalence, curability, and long-term results in oncology.
文摘Azoospermia, defined as a complete absence of spermatozoa in the ejaculate, is a relatively common condition among infertile men. Establishing the etiological diagnosis can be challenging and often requires various investigations. This cross-sectional study, conducted in Cotonou among 35 azoospermic men, aimed to assess the role of biochemical analysis of seminal fluid in determining the etiological diagnosis. The results revealed significant heterogeneity in biochemical profiles. Most of the patients had normal levels of alpha-glucosidase, fructose, and zinc, indicating normal accessory gland function and suggesting non-obstructive azoospermia. However, a significant subgroup exhibited abnormalities in these markers, suggesting a possible obstructive azoospermia. The heterogeneity in biochemical profiles observed in this study highlights the need for further studies to better characterize azoospermia in Benin.
文摘Colorectal cancer(CRC)is the third most common malignancy and the second leading cause of cancer-related mortality worldwide,responsible for approximately 900000 deaths annually.Inflammation and malnutrition significantly influence patients'responses to treatment.Markers such as serum albumin concentration,the prognostic nutritional index,nutritional risk index(NRI),geriatric NRI,and the systemic immune-inflammation index enable the early identification of high-risk patients,facilitating timely interventions that can improve survival and reduce morbidity.A comprehensive understanding and application of these markers allow for better risk stratification in CRC patients,optimizing their management and outcomes.
