Type 2 diabetes mellitus(T2DM)is a global health concern,with a significant proportion of patients developing cardiovascular complications due to vascular endothelial dysfunction(VED).Arginase 1(ARG1)exacerbates the p...Type 2 diabetes mellitus(T2DM)is a global health concern,with a significant proportion of patients developing cardiovascular complications due to vascular endothelial dysfunction(VED).Arginase 1(ARG1)exacerbates the progression of VED by inhibiting the production of nitric oxide.Malvidin 3-O-glucoside(M3G)is an important anthocyanin in blueberries,which has potential in reducing oxidative stress and improving metabolic dysfunction in various diseases.However,its role in T2DM and VED has yet to be explored.In this study,we investigated the therapeutic effects of M3G on T2DM and VED,focusing on its impact on ARG1 expression and endothelial function.The results showed that M3G administration significantly improved metabolic disorders and restored the aortic relaxation function in T2DM mice.Further research revealed that M3G specifically inhibited the expression of ARG1 in endothelial cells to improve T2DM-related VED.Molecular docking simulations indicated that this effect may occur through direct interaction with the active site of ARG1.However,overexpression of ARG1 in endothelial cells reversed these beneficial effects of M3G.In conclusion,this study provides new insights into the mechanism by which M3G,a dietary anthocyanin,regulates endothelial function and ARG1 activity,offering a promising strategy for integrating functional foods into the prevention and management of T2DM.展开更多
