AIM:To conduct a systematic review and Meta-analysis to determine the differential effects of combined lutein and zeaxanthin supplementation on myopia prevention in teenagers.It also investigates the effects of supple...AIM:To conduct a systematic review and Meta-analysis to determine the differential effects of combined lutein and zeaxanthin supplementation on myopia prevention in teenagers.It also investigates the effects of supplements dosage,intervention duration,and geographical variation on intervention results.METHODS:A systematic search and screening of randomized controlled trials(RCTs)completed between 2014 and 2023 was undertaken using the PubMed,EMBASE,Cochrane Library,and Web of Science databases,in accordance with the PRISMA recommendations.The Cochrane risk of bias method was used to assess the quality of the studies.A Meta-analysis was performed using Stata 17.0 to calculate standardized mean differences(SMDs)and 95%confidence intervals(CIs).Subgroup analyses were performed to look at the effects of different doses,intervention periods,and geographic areas.Additionally,publication bias was assessed using Egger’s test.RESULTS:Ten studies including 1035 participants with myopia were analyzed.Supplementation with lutein and zeaxanthin resulted in a significant reduction in axial length elongation among adolescents in the intervention group(SMD=-0.40,P=0.001),an increase in macular pigment optical density(SMD=0.50,P=0.010),and an enhancement in visual sensitivity(SMD=0.53,P=0.008).Subgroup analyses revealed that participants receiving high doses and those undergoing intervention for more than 12mo exhibited significantly improved outcomes compared to those in the low-dose and short-term groups(high-dose vs low-dose:SMD=-0.41 vs-0.22,P=0.003;>12mo vs 6-12mo:SMD=-0.43 vs-0.23,P=0.004,respectively).Furthermore,Egger’s test indicated no significant publication bias(P=0.094).CONCLUSION:Combined lutein and zeaxanthin supplementation has a significant effect on myopia prevention in adolescents,with more pronounced benefits observed in high-dose and long-term interventions.The findings provide scientific evidence for its use as an adjunctive approach in myopia control.展开更多

The results indicated that DA at the concentration from 0 1?μmol/L to 10?μmol/L inhibited the release of LH and FSH from the pituitary fragments of sexually pre mature or hibernating individuals,and the inhibitory effects enhanced with increasing concentrations of DA. E 2 at 1?μmol/L and 10?μmol/L significantly stimulated the release of LH of sexually pre mature individuals,but inhibited their FSH release at 0 1?μmol/L to 10?μmol/L;T had no obvious effects on their FSH release,but significantly inhibited their LH release at 10?μmol/L. Yellow pigment content (YPC) and carotenoid components (lutein and zeaxanthin) of 270 foxtail millet acces...Kernel color is an important trait for assessing the commercial and nutritional quality of foxtail millet. Yellow pigment content (YPC) and carotenoid components (lutein and zeaxanthin) of 270 foxtail millet accessions, including 50 landraces and 220 improved cultivars, from four different eco-regions in China were surveyed using spectrophotometry and high performance liquid chromatography methods. Results indicated that YPC had rich variance, ranging from 1.91 to 28.54 mg kg-1, with an average value of 17.80 mg kg-1. The average YPC of improved cultivars (18.31 mg kg-1) was significantly higher than that of landraces (15.51 mg kg-l). The YPC in cultivars from the Loess Plateau spring sowing region (LPSSR) was the highest (20.59 mg kg-~), followed by the North China summer sowing region (NCSSR, 18.25 mg kg-1), the northeast spring sowing region (NSSR, 17.25 mg kg-1), and the Inner Mongolia Plateau spring sowing region (IMPSSR, 13.92 mg kg-1). The variation coefficients of YPC in cultivars from NSSR, LPSSR, and IMPSSR were higher than that from NCSSR. A similar carotenoid profile was also obtained for 270 foxtail millet cultivars. Lutein and zeaxanthin accounted for approximately 55-65% of YPC in accessions. The lutein content was higher than zeaxanthin content in all cultivars. The ratio of lutein to zeaxanthin ranged from 1.51 to 6.06 with an average of 3.34. YPC was positively correlated with lutein (r=0.935, P〈0.01), zeaxanthin (r=0.808, P〈0.01 ), and growth duration (t=0.488, P〈0.01 ), whereas it was negatively correlated with grain protein (t=-0.332, P〈0.01) and 1 000-kernel weight (t=-0.153, P〈0.05). Our study is useful for screening and selecting cultivars with high levels of yellow pigment and for enhancing phytochemical concentrations in breeding programs.展开更多

Supercritical antisolvent precipitation was applied to prepare lutein/HPMCP nano-capsule. The effects of several operating parameters on the yield, lutein loading, encapsulation efficiency, particle size and particle size distribution of the nanocapsule were investigated. The mean diameter of nanocapsules ranged from 163 nm to 219 nm under appropriate experimental conditions. The result of scanning electron microscope showed that the nanocapsules were nearly spherical. The highest yield reached 95.35% when the initial concentration of lutein was saturated. The highest lutein loading of 15.80% and encapsulation efficiency of 88,41% were obtained under the conditions of 11 MPa, 40℃ and CHPMCP: Clutein= 5:1. The results may promote the application of lutein in food industry.展开更多

Methods: The extract was administered at doses of 30, 60 and 120 mg/kg by oral gavage, acutely (one time, 45 min before mating test) or subchronically (daily for 10 days) in sexually potent and sexually sluggish/impotent rats. Sexual behavior, serum levels of luteinizing hormone (LH) and testosterone (T) were evaluated in treated rats and compared with controls receiving vehicle. The effect of the extract on central dopaminergic neurotransmission was assessed in the nucleus accumbens using a microdialysis technique. Results: In sexually potent rats, both acute and subchronic treatment with the extract dosed at 30 and 60 mg/kg reduced mount latency and intromission latency. In sluggish/impotent rats, the acutely administered extract at the dose of 60 mg/kg shortened ejaculation latency, whereas subchronically administered at the doses of 30 and 60 mg/kg, reduced mount, intromission and ejaculation latencies, increasing also the percentage of mounting and ejaculating rats. The extract dosed at 60 mg/kg significantly increased LH and T following acute and subchronic administration and increased 3,4-dihydroxyphenylacetic acid levels in the nucleus accumbens, 30 min after the acute administration. Conclusion: The improvement in both appetitive and consummatory components of sexual behavior observed in male rats treated with the extract could be ascribed to increased serum T level in parallel with the activation of the central dopaminergic system.展开更多

Vitreoscilla hemoglobin is an oxygen-binding protein that promotes oxygen delivery and reduces oxygen consumption under low oxygen conditions to increase the effi ciency of cell respiration and metabolism.In this stud...Vitreoscilla hemoglobin is an oxygen-binding protein that promotes oxygen delivery and reduces oxygen consumption under low oxygen conditions to increase the effi ciency of cell respiration and metabolism.In this study,we introduced a Vitreoscilla hemoglobin gene(vgb)into Chlorella vulgaris by Agrobacterium tumefaciens-mediated transformation(ATMT).PCR analysis confi rmed that the vgb gene was successfully integrated into the Chlorella vulgaris genome.Analysis of biomass obtained in shake fl asks revealed transformant biomass concentrations as high as 3.28 g/L,which was 38.81% higher than that of the wild-type strain.Lutein content of transformants also increased slightly.Further experiments recovered a maximum lutein yield of 2.91 mg/L from the transformants,which was 36.77% higher than that of the wild-type strain.The above results suggest that integrated expression of the vgb gene may improve cell growth and lutein yield in Chlorella vulgaris,with applications to lutein production from Chlorella during fermentation.展开更多

Methods: Thirty male rats were divided into five groups: group 1 (untreated) and groups 2, 3, 4 and 5 were fed for 30 days with 5%, 10%, 15% and 30% crude garlic, respectively. Testes and accessory organs were weighed and some markers were assessed. Light and electron microscopy observations were also performed. Results: A significant decrease was observed in the body weight of groups 4 (14%; P 〈 0.01) and 5 (20%; P 〈 0.01); of the prostate weight in group 5 (29.1%; P 〈 0.05) and of seminal vesicle weight in groups 3 (14.4%; P 〈 0.01), 4 (18.3%; P 〈 0.01) and 5 (27.3%; P 〈 0.01). In contrast, testis and epididymis weights were unchanged. In epididymis tissue, the alpha glucosidase activity and the spermatozoa density were unchanged. The treatment resulted in a significant decrease in testosterone serum levels in groups 3 (77.3%; P 〈 0.01), 4 (77.3%; P 〈 0.01) and 5 (90.9%; P 〈 0.01), associated with a significant increase in LH serum levels (P 〈 0.01). Testicular histology showed a dose-dependent increase in the percentage of empty seminiferous tubules. Moreover, testicular function was affected; a significant decrease in phosphatase acid activity (P 〈 0.01) and testosterone (P 〈 0.05) contents were observed. Conclusion: Crude garlic consumption during 1 month reduced testosterone secretion and altered spermatogenesis at 10%, 15% and 30% doses.展开更多

Wi...This study was performed to assess serum testosterone alterations induced by paradoxical sleep deprivation (PSD) and to verify their attenuation during sleep recovery (SR) based on different durations and ages. Wistar male rats aged 12 weeks for the younger group and 20 weeks for the elder group were randomly distributed into one of the following groups: a control group (cage and platform), 3-day SD, 5-day SD, 7-day SD, 1-day SR, 3-day SR and 5-day SR groups. For PSD, the modified multiple platform method was used to specifically limit rapid eye movement (REM) sleep. Differences in the testosterone and luteinizing hormone levels between the younger group and the elder group according to duration of PSD and SR recovery were analysed. Testosterone continued to fall during the sleep deprivation period in a time-dependent manner in both the younger (P=-0.001, correlation coefficient r=-0.651) and elder groups (P=0.001, correlation coefficient r=-0.840). The elder group showed a significantly lower level of testosterone compared with the younger group after PSD. Upon SR after 3 days of PSD, the testosterone level continued to rise for 5 days after sleep recovery in the younger group (P=0.013), whereas testosterone concentrations failed to recover until day 5 in the elder group. PSD caused a more detrimental effect on serum testosterone in the elder group compared to the younger group with respect to decreases in luteinizing hormone (LH) levels. The replenishment of serum testosterone level was prohibited in the elder group suggesting that the effects of SD/SR may be age-dependent. The mechanism by which SD affects serum testosterone and how age may modify the process are still unclear.展开更多
Hormone naive advanced prostate cancer is subdivided into two disease states: biochemical recurrence and traditional M 1 (metastatic) prostate cancer and characterized by no prior hormonal therapy or androgen depri...Hormone naive advanced prostate cancer is subdivided into two disease states: biochemical recurrence and traditional M 1 (metastatic) prostate cancer and characterized by no prior hormonal therapy or androgen deprivation therapy (ADT). In biochemical recurrence/ prostate-specific antigen (PSA) recurrence, men should be risk-stratified based on their PSA doubling time, the Gleason score and the timing of the recurrence. In general, only men who are at high risk should be considered for early/immediate ADT although this is best done using shared decision with the patient. The type of ADT to be used in biochemical recurrence ranging from oral-only peripheral blockade (peripheral androgen deprivation) to complete hormonal therapy (combined androgen blockade [CAB]) remains in debate owing to lack of randomized controlled trials (RCT). However, there is good RCT support for use of intermittent hormonal therapy (IHT). There is also limited research on biomarker response (PSA and testosterone decline) to predict prognosis. On the other hand, in the setting of M1 hormone naive prostate cancer, there are many more RCT's to inform our decisions. CAB and gonadotrophin-releasing hormone antagonists perhaps provide a slight efficacy advantage while IHT may be slightly inferior with minimal M1 disease. The PSA nadir at 7 months after starting ADT is a powerful prognostic tool for M1 patients. There is growing recognition that serum testosterone (T) control while on ADT is linked to the development of castrate-resistant prostate cancer. Especially for a M 1 patient, maintaining a serum T below 20-30 ng d1-1 prolongs the response to ADT. Novel oral agents (abiraterone and enzalutamide) may soon find use in hormone naive disease and may alter the treatment landscape. Despite over 75 years of experience with ADT, many questions remain, and the field continues to evolve.展开更多

Methods An E-waste dismantling area in Taizhou of Zhejiang Province was selected as the research site. One hundred and fifty two samples were collected from the groundwater, soil, rice, corn, chicken, and pork in the dismantling area. The effects of the multi-media lead pollution on the male blood lead and sex hormone levels of FSH, LH, and T, as well as the correlation with confounding factors, were studied. Results The blood lead concentrations in the males aged under 31, from 31 to 45 and from 46 to 60 were 98.55, 100.23, and 101.45 I~/L, respectively. Of all the environmental media lead exposures, the groundwater, rice and soil were main contributing factors to the lead accumulation in humans. FSH and LH levels increased with the age while the T levels decreased with the age instead. There was a significant correlation between the FSH and LH levels and wearing masks. Conclusion There was correlation between the FSH, LH, and T levels, and the mean values of lead concentrations in environmental media, and the sex hormone levels were correlated with the confounding factor of wearing masks.展开更多
AIM: To compare serum Lutein and Zeaxanthin (L/Z) concentrations between patients with nonproliferative diabetic retinopathy (NDR) and normal subjects,and to explore the effect of L/Z supplementation on serum L/Z leve...AIM: To compare serum Lutein and Zeaxanthin (L/Z) concentrations between patients with nonproliferative diabetic retinopathy (NDR) and normal subjects,and to explore the effect of L/Z supplementation on serum L/Z level and visual function in NDR patients METHODS: Subjects were divided into three groups: 30 NDR patients supplied with Lutein 6mg/d and Zeaxanthin 0.5mg/d for three months (DR Group),30 NDR patients without L/Z supplementation (DR Control Group) and 30 normal subjects (Control Group).Serum L/Z concentrations were measured by liquid high-resolution chromatography (HPLC).Visual acuity was recorded at baseline,1 month,2 months and 3 months post initial supplementation.Serum L/Z concentration were measured at baseline,1 month and 2 months post initial supplementation.Contrast sensitivity (CS) and fovea thickness were recorded at baseline and 3 months post initial supplementation.RESULTS: Mean serum lutein concentrations in DR group were 0.0686±0.0296μg/mL and zeaxanthin concentration was 0.0137±0.0059μg/mL.The L/Z level of DR group was significantly lower compared to the control group( lutein: 0.2302±0.1308μg/mL,zeaxanthin: 0.0456±0.0266μg/m,P = 0.000).The concentration of lutein and zeaxanthin in the DR control group at base line was 0.0714±0.0357μg/mL and 0.0119±0.0072μg/mL,respectively.There was no significant change of L/Z concentration in the DR control group during the study.Serum L/Z concentrations of DR group increased significantly after supplementation(F=109.124,P=0.000;F=219.207,P=0.000).Visual acuity improved significantly after medication.Compared with pre-medication,the average CS values of 1.5cpd,3cpd and 6cpd after three months increased significantly (P =0.030,0.013,0.008) and the foveal thickness decreased.(P =0.05) CONCLUSION: Serum L/Z concentrations in DR patients are significantly lower than those in normal subjects,and L/Z intake can improve the visual acuity,CS and macular edema in DR patients,suggesting that L/Z supplementation might be targeted as potential potential therapeutic agents in treating NDR.展开更多
文摘AIM:To conduct a systematic review and Meta-analysis to determine the differential effects of combined lutein and zeaxanthin supplementation on myopia prevention in teenagers.It also investigates the effects of supplements dosage,intervention duration,and geographical variation on intervention results.METHODS:A systematic search and screening of randomized controlled trials(RCTs)completed between 2014 and 2023 was undertaken using the PubMed,EMBASE,Cochrane Library,and Web of Science databases,in accordance with the PRISMA recommendations.The Cochrane risk of bias method was used to assess the quality of the studies.A Meta-analysis was performed using Stata 17.0 to calculate standardized mean differences(SMDs)and 95%confidence intervals(CIs).Subgroup analyses were performed to look at the effects of different doses,intervention periods,and geographic areas.Additionally,publication bias was assessed using Egger’s test.RESULTS:Ten studies including 1035 participants with myopia were analyzed.Supplementation with lutein and zeaxanthin resulted in a significant reduction in axial length elongation among adolescents in the intervention group(SMD=-0.40,P=0.001),an increase in macular pigment optical density(SMD=0.50,P=0.010),and an enhancement in visual sensitivity(SMD=0.53,P=0.008).Subgroup analyses revealed that participants receiving high doses and those undergoing intervention for more than 12mo exhibited significantly improved outcomes compared to those in the low-dose and short-term groups(high-dose vs low-dose:SMD=-0.41 vs-0.22,P=0.003;>12mo vs 6-12mo:SMD=-0.43 vs-0.23,P=0.004,respectively).Furthermore,Egger’s test indicated no significant publication bias(P=0.094).CONCLUSION:Combined lutein and zeaxanthin supplementation has a significant effect on myopia prevention in adolescents,with more pronounced benefits observed in high-dose and long-term interventions.The findings provide scientific evidence for its use as an adjunctive approach in myopia control.
文摘This review focuses on the diagnostic algorithm for nonobstructive azoospermia(NOA),a significant male factor contributing to infertility.NOA,characterized by the absence of sperm in the ejaculate,requires a systematic diagnostic approach to identify reversible conditions,genetic factors,and prognosis for achieving pregnancy.The diagnostic pathway involves semen analysis and a comprehensive evaluation for hormonal deficiencies,anatomical abnormalities,and genetic factors.The importance of medical history,physical examination,endocrine evaluation,imaging,and genetic testing is emphasized.This review highlights the significance of differentiating NOA from obstructive azoospermia(OA)and outlines key considerations for effective management,including surgical sperm retrieval and assisted reproductive techniques.Testicular biopsy is discussed as a definitive method to distinguish obstructive cases from nonobstructive cases,providing valuable prognostic information.Overall,a thorough and systematic diagnostic approach is essential for the effective management of men suspected with NOA,offering insights into potential treatment options and reproductive outcomes.
文摘Dear Editor,I would like to congratulate Mamsen et al.i on their extensive and scientifically valuable work.I analyzed their raw data presented in Table 1 of the original article from a different perspective and discovered an effect not mentioned in the article.My analysis showed that luteinizing hormone(LH)levels are significantly lower in patients at high infertility risk(HIR),whose testes lack A dark(Ad)spermatogonia and display an abnormal ratio of germ cells per crosssectional tubule(G/T).

文摘Objective:To examine the effect of an neurokinin 3 receptor(NK3R)agonist,senktide,on neuronal nitric oxide synthase(nNOS)activation in the median eminence-arcuate nucleus(ME-ARC)and preoptic area(POA)regions of the hypothalamus across proestrus,diestrus,and ovariectomized states in female rats and its correlation with luteinizing hormone(LH)secretion.Methods:Adult female Sprague-Dawley rats were examined for proestrus and diestrus phases of the estrous cycle.Female rats were categorized into proestrus and diestrus groups,and each was further divided into four subgroups(n=4).In both the diestrus and proestrus categories,Group 1 was the control group.Groups 2,3,and 4 received senktide(100μg/kg-1),NK3R antagonist SB222200(10 mg/kg-1),and SB222200 followed by senktide,respectively.To evaluate the effect of sex steroids on NK3R agonist-induced nNOS activation,female rats underwent bilateral ovariectomy and were divided into four groups(n=3).Group 1 served as the control.Group 2 received a subcutaneous injection of 17β-estradiol 3-benzoate(E2,3μg/rat).Group 3 received E2 and progesterone(30μg/rat).Group 4 was administered senktide(100μg/kg).Female rats from each group were sacrificed,blood was collected for LH ELISA,and hypothalamic tissues were collected for Western blotting.Results:Senktide increased nNOS phosphorylation in the ME-ARC during both the proestrus and diestrus phases.In the POA,senktide increased nNOS phosphorylation only during the diestrus phase.In ovariectomized rats,senktide activated nNOS independent of sex steroid levels.Senktide also increased serum LH concentration in diestrus and ovariectomized female rats.Conclusions:Senktide,an NK3R agonist,activates nNOS in the POA and ME-ARC regions of the hypothalamus in a phase dependent manner.The activation of nNOS by senktide suggests a potential mechanism by which neurokinin B triggers nNOS activation in the ARC and POA regions and regulates GnRH/LH secretion.

文摘To understand the regulatory mechanisms of gonadotropin secretion in Rana rugulosa ,this study investigated the effects of dopamine (DA),estradiol (E 2) and testosterone (T) on the in vitro release of luteinizing hormone (LH) and follicle stimulating hormone (FSH) from the pituitary fragments of female Rana rugulosa using a static incubation system and radio immunoassay (RIA). The results indicated that DA at the concentration from 0 1?μmol/L to 10?μmol/L inhibited the release of LH and FSH from the pituitary fragments of sexually pre mature or hibernating individuals,and the inhibitory effects enhanced with increasing concentrations of DA. E 2 at 1?μmol/L and 10?μmol/L significantly stimulated the release of LH of sexually pre mature individuals,but inhibited their FSH release at 0 1?μmol/L to 10?μmol/L;T had no obvious effects on their FSH release,but significantly inhibited their LH release at 10?μmol/L. Neither E 2 nor T,at the concentration from 0 1?μmol/L to 100?μmol/L,had obvious effects on the release of LH and FSH of hibernating individuals. The data suggest that DA and sexual steroids may have direct regulatory actions on LH and FSH release at the pituitary level in Rana rugulosa ,and the action of sexual steroids may relate to the gonadal development stages (seasons).

文摘Kernel color is an important trait for assessing the commercial and nutritional quality of foxtail millet. Yellow pigment content (YPC) and carotenoid components (lutein and zeaxanthin) of 270 foxtail millet accessions, including 50 landraces and 220 improved cultivars, from four different eco-regions in China were surveyed using spectrophotometry and high performance liquid chromatography methods. Results indicated that YPC had rich variance, ranging from 1.91 to 28.54 mg kg-1, with an average value of 17.80 mg kg-1. The average YPC of improved cultivars (18.31 mg kg-1) was significantly higher than that of landraces (15.51 mg kg-l). The YPC in cultivars from the Loess Plateau spring sowing region (LPSSR) was the highest (20.59 mg kg-~), followed by the North China summer sowing region (NCSSR, 18.25 mg kg-1), the northeast spring sowing region (NSSR, 17.25 mg kg-1), and the Inner Mongolia Plateau spring sowing region (IMPSSR, 13.92 mg kg-1). The variation coefficients of YPC in cultivars from NSSR, LPSSR, and IMPSSR were higher than that from NCSSR. A similar carotenoid profile was also obtained for 270 foxtail millet cultivars. Lutein and zeaxanthin accounted for approximately 55-65% of YPC in accessions. The lutein content was higher than zeaxanthin content in all cultivars. The ratio of lutein to zeaxanthin ranged from 1.51 to 6.06 with an average of 3.34. YPC was positively correlated with lutein (r=0.935, P〈0.01), zeaxanthin (r=0.808, P〈0.01 ), and growth duration (t=0.488, P〈0.01 ), whereas it was negatively correlated with grain protein (t=-0.332, P〈0.01) and 1 000-kernel weight (t=-0.153, P〈0.05). Our study is useful for screening and selecting cultivars with high levels of yellow pigment and for enhancing phytochemical concentrations in breeding programs.

文摘Lutein was nano-encapsuled with hydroxypropylmethyl cellulose phthalate (HPMCP) to maintain its bioactivity and to avoid thermal/light degradation. Supercritical antisolvent precipitation was applied to prepare lutein/HPMCP nano-capsule. The effects of several operating parameters on the yield, lutein loading, encapsulation efficiency, particle size and particle size distribution of the nanocapsule were investigated. The mean diameter of nanocapsules ranged from 163 nm to 219 nm under appropriate experimental conditions. The result of scanning electron microscope showed that the nanocapsules were nearly spherical. The highest yield reached 95.35% when the initial concentration of lutein was saturated. The highest lutein loading of 15.80% and encapsulation efficiency of 88,41% were obtained under the conditions of 11 MPa, 40℃ and CHPMCP: Clutein= 5:1. The results may promote the application of lutein in food industry.
文摘Aim: To investigate the influence of an extract obtained from five Chinese medicinal plants on sexual behavior of adult male rats. Methods: The extract was administered at doses of 30, 60 and 120 mg/kg by oral gavage, acutely (one time, 45 min before mating test) or subchronically (daily for 10 days) in sexually potent and sexually sluggish/impotent rats. Sexual behavior, serum levels of luteinizing hormone (LH) and testosterone (T) were evaluated in treated rats and compared with controls receiving vehicle. The effect of the extract on central dopaminergic neurotransmission was assessed in the nucleus accumbens using a microdialysis technique. Results: In sexually potent rats, both acute and subchronic treatment with the extract dosed at 30 and 60 mg/kg reduced mount latency and intromission latency. In sluggish/impotent rats, the acutely administered extract at the dose of 60 mg/kg shortened ejaculation latency, whereas subchronically administered at the doses of 30 and 60 mg/kg, reduced mount, intromission and ejaculation latencies, increasing also the percentage of mounting and ejaculating rats. The extract dosed at 60 mg/kg significantly increased LH and T following acute and subchronic administration and increased 3,4-dihydroxyphenylacetic acid levels in the nucleus accumbens, 30 min after the acute administration. Conclusion: The improvement in both appetitive and consummatory components of sexual behavior observed in male rats treated with the extract could be ascribed to increased serum T level in parallel with the activation of the central dopaminergic system.

文摘Mesocarp color is an important agronomic trait of Cucurbita maxima and is determined mainly by the contents and compositions of the carotenoids.The two inbred lines with significant differences in fruit flesh color were used in the study,the orange’312-1’and white’98-2’.Changes in seven carotenoid contents and compositions in the flesh of fruit produced by inbred lines’312-1’and’98-2’were analyzed during fruit development.The expression of eight key carotenoid biosynthesis genes in the fruit flesh were investigated during five fruit development stage in two inbred lines.As the flesh color intensified,the orange flesh color of’312-1’was determined mainly by the increased contents of lutein,β-carotene and zeaxanthin and the lack of carotenoid accumulation led to the formation of white flesh in’98-2’fruit.The expression of the LCY-e and CHYb genes was significantly stronger in’312-1’than in’98-2’,and their expression was strongly correlated with lutein andβ-carotene contents during fruit development.This study provides a deep understanding of the molecular mechanism of carotenoid biosynthesis in fruit flesh and provides a basis for additional studies on the highly refined improvement of squash quality.

文摘Vitreoscilla hemoglobin is an oxygen-binding protein that promotes oxygen delivery and reduces oxygen consumption under low oxygen conditions to increase the effi ciency of cell respiration and metabolism.In this study,we introduced a Vitreoscilla hemoglobin gene(vgb)into Chlorella vulgaris by Agrobacterium tumefaciens-mediated transformation(ATMT).PCR analysis confi rmed that the vgb gene was successfully integrated into the Chlorella vulgaris genome.Analysis of biomass obtained in shake fl asks revealed transformant biomass concentrations as high as 3.28 g/L,which was 38.81% higher than that of the wild-type strain.Lutein content of transformants also increased slightly.Further experiments recovered a maximum lutein yield of 2.91 mg/L from the transformants,which was 36.77% higher than that of the wild-type strain.The above results suggest that integrated expression of the vgb gene may improve cell growth and lutein yield in Chlorella vulgaris,with applications to lutein production from Chlorella during fermentation.
文摘Aim: To investigate the effects of crude garlic on adult male rat reproductive functions. Methods: Thirty male rats were divided into five groups: group 1 (untreated) and groups 2, 3, 4 and 5 were fed for 30 days with 5%, 10%, 15% and 30% crude garlic, respectively. Testes and accessory organs were weighed and some markers were assessed. Light and electron microscopy observations were also performed. Results: A significant decrease was observed in the body weight of groups 4 (14%; P 〈 0.01) and 5 (20%; P 〈 0.01); of the prostate weight in group 5 (29.1%; P 〈 0.05) and of seminal vesicle weight in groups 3 (14.4%; P 〈 0.01), 4 (18.3%; P 〈 0.01) and 5 (27.3%; P 〈 0.01). In contrast, testis and epididymis weights were unchanged. In epididymis tissue, the alpha glucosidase activity and the spermatozoa density were unchanged. The treatment resulted in a significant decrease in testosterone serum levels in groups 3 (77.3%; P 〈 0.01), 4 (77.3%; P 〈 0.01) and 5 (90.9%; P 〈 0.01), associated with a significant increase in LH serum levels (P 〈 0.01). Testicular histology showed a dose-dependent increase in the percentage of empty seminiferous tubules. Moreover, testicular function was affected; a significant decrease in phosphatase acid activity (P 〈 0.01) and testosterone (P 〈 0.05) contents were observed. Conclusion: Crude garlic consumption during 1 month reduced testosterone secretion and altered spermatogenesis at 10%, 15% and 30% doses.
文摘This study was performed to assess serum testosterone alterations induced by paradoxical sleep deprivation (PSD) and to verify their attenuation during sleep recovery (SR) based on different durations and ages. Wistar male rats aged 12 weeks for the younger group and 20 weeks for the elder group were randomly distributed into one of the following groups: a control group (cage and platform), 3-day SD, 5-day SD, 7-day SD, 1-day SR, 3-day SR and 5-day SR groups. For PSD, the modified multiple platform method was used to specifically limit rapid eye movement (REM) sleep. Differences in the testosterone and luteinizing hormone levels between the younger group and the elder group according to duration of PSD and SR recovery were analysed. Testosterone continued to fall during the sleep deprivation period in a time-dependent manner in both the younger (P=-0.001, correlation coefficient r=-0.651) and elder groups (P=0.001, correlation coefficient r=-0.840). The elder group showed a significantly lower level of testosterone compared with the younger group after PSD. Upon SR after 3 days of PSD, the testosterone level continued to rise for 5 days after sleep recovery in the younger group (P=0.013), whereas testosterone concentrations failed to recover until day 5 in the elder group. PSD caused a more detrimental effect on serum testosterone in the elder group compared to the younger group with respect to decreases in luteinizing hormone (LH) levels. The replenishment of serum testosterone level was prohibited in the elder group suggesting that the effects of SD/SR may be age-dependent. The mechanism by which SD affects serum testosterone and how age may modify the process are still unclear.
文摘Hormone naive advanced prostate cancer is subdivided into two disease states: biochemical recurrence and traditional M 1 (metastatic) prostate cancer and characterized by no prior hormonal therapy or androgen deprivation therapy (ADT). In biochemical recurrence/ prostate-specific antigen (PSA) recurrence, men should be risk-stratified based on their PSA doubling time, the Gleason score and the timing of the recurrence. In general, only men who are at high risk should be considered for early/immediate ADT although this is best done using shared decision with the patient. The type of ADT to be used in biochemical recurrence ranging from oral-only peripheral blockade (peripheral androgen deprivation) to complete hormonal therapy (combined androgen blockade [CAB]) remains in debate owing to lack of randomized controlled trials (RCT). However, there is good RCT support for use of intermittent hormonal therapy (IHT). There is also limited research on biomarker response (PSA and testosterone decline) to predict prognosis. On the other hand, in the setting of M1 hormone naive prostate cancer, there are many more RCT's to inform our decisions. CAB and gonadotrophin-releasing hormone antagonists perhaps provide a slight efficacy advantage while IHT may be slightly inferior with minimal M1 disease. The PSA nadir at 7 months after starting ADT is a powerful prognostic tool for M1 patients. There is growing recognition that serum testosterone (T) control while on ADT is linked to the development of castrate-resistant prostate cancer. Especially for a M 1 patient, maintaining a serum T below 20-30 ng d1-1 prolongs the response to ADT. Novel oral agents (abiraterone and enzalutamide) may soon find use in hormone naive disease and may alter the treatment landscape. Despite over 75 years of experience with ADT, many questions remain, and the field continues to evolve.

文摘Objective To study the effects of environmental multi-media lead pollution on blood lead and sex hormone levels among lead exposed males engaged in E-waste dismantling, and the correlation between confounding factors and sex hormone levels. Methods An E-waste dismantling area in Taizhou of Zhejiang Province was selected as the research site. One hundred and fifty two samples were collected from the groundwater, soil, rice, corn, chicken, and pork in the dismantling area. The effects of the multi-media lead pollution on the male blood lead and sex hormone levels of FSH, LH, and T, as well as the correlation with confounding factors, were studied. Results The blood lead concentrations in the males aged under 31, from 31 to 45 and from 46 to 60 were 98.55, 100.23, and 101.45 I~/L, respectively. Of all the environmental media lead exposures, the groundwater, rice and soil were main contributing factors to the lead accumulation in humans. FSH and LH levels increased with the age while the T levels decreased with the age instead. There was a significant correlation between the FSH and LH levels and wearing masks. Conclusion There was correlation between the FSH, LH, and T levels, and the mean values of lead concentrations in environmental media, and the sex hormone levels were correlated with the confounding factor of wearing masks.
文摘AIM: To compare serum Lutein and Zeaxanthin (L/Z) concentrations between patients with nonproliferative diabetic retinopathy (NDR) and normal subjects,and to explore the effect of L/Z supplementation on serum L/Z level and visual function in NDR patients METHODS: Subjects were divided into three groups: 30 NDR patients supplied with Lutein 6mg/d and Zeaxanthin 0.5mg/d for three months (DR Group),30 NDR patients without L/Z supplementation (DR Control Group) and 30 normal subjects (Control Group).Serum L/Z concentrations were measured by liquid high-resolution chromatography (HPLC).Visual acuity was recorded at baseline,1 month,2 months and 3 months post initial supplementation.Serum L/Z concentration were measured at baseline,1 month and 2 months post initial supplementation.Contrast sensitivity (CS) and fovea thickness were recorded at baseline and 3 months post initial supplementation.RESULTS: Mean serum lutein concentrations in DR group were 0.0686±0.0296μg/mL and zeaxanthin concentration was 0.0137±0.0059μg/mL.The L/Z level of DR group was significantly lower compared to the control group( lutein: 0.2302±0.1308μg/mL,zeaxanthin: 0.0456±0.0266μg/m,P = 0.000).The concentration of lutein and zeaxanthin in the DR control group at base line was 0.0714±0.0357μg/mL and 0.0119±0.0072μg/mL,respectively.There was no significant change of L/Z concentration in the DR control group during the study.Serum L/Z concentrations of DR group increased significantly after supplementation(F=109.124,P=0.000;F=219.207,P=0.000).Visual acuity improved significantly after medication.Compared with pre-medication,the average CS values of 1.5cpd,3cpd and 6cpd after three months increased significantly (P =0.030,0.013,0.008) and the foveal thickness decreased.(P =0.05) CONCLUSION: Serum L/Z concentrations in DR patients are significantly lower than those in normal subjects,and L/Z intake can improve the visual acuity,CS and macular edema in DR patients,suggesting that L/Z supplementation might be targeted as potential potential therapeutic agents in treating NDR.
