Risk assessment and residue behavior of a novel pesticide cyetpyrafen and its metabolites from fresh tea leaves to tea infusion

China has limited acaricide options for tea plantations.Cyetpyrafen,a novel domestic acaricide with high efficacy,low toxicity and a negative temperature coefficient,offers an alternative for tea pest control;however,its residue fate in tea remains unclear.This study developed a method to simultaneously detect cyetpyrafen and its metabolites(M-309,M-325-1,and M-409-3)in different tea matrices to investigate their fate.Recoveries of compounds ranged from 73.4 to 106.2%with the relative standard deviations(RSDs)below 12.0%.During tea cultivation,the dissipation half-life of cyetpyrafen was 0.59 d,with M-309 as a major metabolite.The residues of cyetpyrafen and M-309 were affected by different processing stages,especially water loss and high temperatures during fixing,drying and withering.The total processing factors ranged from 1.39 to 1.71 for green tea and 1.48 to 2.28 for black tea(processed from fresh tea leaves sampled at 1,5,and 7 d),respectively.The leaching rates of cyetpyrafen from green tea and black tea into tea infusions were 7.4 and 6%,respectively.The risk associated with cyetpyrafen intake from tea consumption was low,with risk quotient values below 100%.However,theoretical calculation indicated potential harm to non-target organisms from its metabolites.This research provides a reference for the safe and efficient use of cyetpyrafen in tea gardens. 展开更多

Protective effects of vine tea on high-fat diet rat:regulation of ferroptosis pathway and gut microbiota

Vine tea is documented in ancient Chinese books as having the function of promoting blood circulation.However,its effects and mechanisms remain unclear.The aim of this study was to comprehensively investigate the protective potential and mechanisms of vine tea in high-fat diet rat through a combination of in vivo and in vitro experiments.The efficacy of vine tea was evaluated using a high-fat diet rat model and an oxidized low-density lipoprotein-treated cell model,with physiological and biochemical indicators measured in rat serum and cell supernatants.Transcriptomics was utilized to investigate alterations mRNA expression following the administration of dihydromyricetin in cell model.Metabolomics and 16S rRNA sequencing was employed to examine changes in metabolites in the serum and changes in gut microbiota of high-fat diet rats after administering vine tea extract.Vine tea extract and dihydromyricetin can reduce elevated levels of lipids,including total cholesterol and triglycerides,following modeling.Transcriptomic data indicate that dihydromyricetin exerts its effects by regulating ferroptosis signaling pathways.Metabolomic analysis demonstrates that the administration of vine tea extract influences the vitamin K cycle and glutathione,thereby alleviating the progression of ferroptosis.Additionally,16S rRNA sequencing reveals that vine tea extract increases Lactobacillaceae in the gut microbiota,which subsequently affects the levels of lysophosphatidylcholine,a major phospholipid component of oxidized low-density lipoprotein in serum.Our results indicate that vine tea can regulate ferroptosis signaling pathways and increase Lactobacillaceae in the gut microbiota,thereby exhibiting cardiovascular protective effects in high-fat diet rats. 展开更多

The Biological Properties of Carbon Dots-Tea Polyphenols Complex:In Vitro and Cell Experiment

Carbon dots(CDs),nanoscale carbon-based particles ubiquitously present in thermally processed foods such as toasted products,exhibit exceptional structural stability even after gastrointestinal digestion,a property that facilitates their systemic absorption and bioaccumulation.Their persistence in biological systems enables direct interactions with small bioactive molecules,potentially modulating the functional properties of dietary components.This study systematically evaluates the impact of CDs on the antioxidant efficacy of tea polyphenols(TPs),a class of natural antioxidants widely recognized for their health-promoting effects.The structure,antioxidant capacity,and in vitro cytological effects of the CDs-TPs complex were also investigated.Our results demonstrate that CDs and TPs form stable complexes mediated by non-covalent interactions,with hydrogen bonding identified as the dominant mechanism–specifically between the phenolic hydroxyl groups of TPs and fluorescent carbon-rich domains on CDs.These interactions induced a concentration-dependent enhancement in fluorescence intensity,with optimal binding efficiency observed at low CDs/TPs molar ratios(1:1 and 10:1).Structural analyses revealed that TP binding altered the tertiary conformation of CDs,exposing additional luminescent sites and modifying surface charge distribution.Transcriptomic profiling further demonstrated dose-dependent increases in differentially expressed genes under higher CDs/TPs ratios,which were predominantly enriched in oxidative stress response pathways.Mechanistic studies identified the FoxO signaling pathway as a central regulatory axis,with CDs-TPs complexes modulating the expression of antioxidant-related genes(e.g.,SOD2)and downstream effectors involved in redox homeostasis.These findings not only elucidate the structure-function interplay governing CDs-TPs interactions,but also highlight their dual role as modulators of antioxidant activity and potential therapeutic agents,establishing a foundation for developing CD-based composite materials in targeted antioxidant therapies. 展开更多

Materials exhibiting broadband nonlinear optical responses are critically important for ultrafast photonics applications,particularly as saturable absorbers(SAs)that facilitate broadband optical pulse generation.In this study,tea polyphenolpolyvinyl alcohol(TP-PVA)composite films are synthesized via a polymer embedding method and employed as SAs to initiate ultrafast pulse operation in fiber lasers.The TP-PVA SA film exhibits excellent broadband saturable absorption performance at wavelengths of 1.0μm,1.5μm,and 2.0μm,with modulation depths of 54.21%,41.41%,and 51.16%,respectively.Stable passively mode-locked pulses with pulse widths of 588 fs,419 fs,and 743 fs are generated in Yb-,Er-,and Tm-doped fiber lasers,respectively.This work confirms the effective performance of TP-PVA as a broadband SA,and establishes a foundation for the integration of novel and sustainable materials within ultrafast photonic systems.The approach paves the way for developing compact broadband ultrafast laser systems operating in the near-infrared spectral region. 展开更多

Understanding the molecular responses of tea leaves to mechanical stress is crucial for elucidating the mechanisms of post-harvest quality formation during oolong tea processing.This study employed an integrated multi-omics strategy to characterize the changes and interactions among metabolomic(MB),transcriptomic(TX),and proteomic(PT)profiles in mechanically stressed tea leaves.Mechanical stress initially activated damage-associated molecular patterns(DAMPs),including Ca^(2+)signaling,jasmonic acid signaling,and glutathione metabolism pathways.These processes subsequently induced quality-related metabolic pathways(QRMPs),particularly α-linolenic acid and phenylalanine metabolism.Upregulated expression of LOX,ADH1,and PAR genes,together with the increased abundance of their encoded proteins,respectively promoted the accumulation of jasmine lactone,benzyl alcohol,and 2-phenylethanol.These findings indicate that mechanical stress influences the metabolite biosynthesis in tea leaves through coordinated molecular responses.This study provides new insights into the molecular mechanisms underlying tea leaf responses to mechanical stress and a foundation for future investigations into how early molecular events may contribute to post-harvest metabolic changes during oolong tea processing. 展开更多

Tea plant(Camellia sinensis(L.)O.Kuntze)is a cold-sensitive leaf-harvesting crop whose growth,yield,and processed tea quality are all inhibited by low temperatures.Therefore,identifying the regulatory genes involved in tea plant growth and freezing tolerance is crucial for genetic improvement.WRKY transcription factors regulate various plant processes,including growth and development,stress responses,and metabolite biosynthesis.However,the molecular network through which WRKY coordinates these pathways in tea plants remains unclear.In this study,we revealed that CsWRKY57L,a cold-inducible WRKY IIc subfamily member,positively regulated freezing tolerance by directly promoting flavonoid accumulation in tea plants.Transient suppression of CsWRKY57L weakened the freezing tolerance of tea plants by reducing flavonoid content and suppressing the C-repeat-binding factor(CBF)-cold-responsive(COR)gene pathway.In contrast,heterologous overexpression of CsWRKY57L in Arabidopsis had the opposite effect.Additionally,overexpression of CsWRKY57L inhibited reproductive development and accelerated senescence in Arabidopsis.Interaction analysis revealed that CsWRKY57L directly binds to the promoters of CsSWEET1a,CsSWEET15,and AtSWEET15,which encode sugar transporters essential for plant reproductive development,and inhibits their transcription.Overall,the study revealed a dual role of CsWRKY57L in promoting freezing tolerance via flavonoid biosynthesis and inhibiting reproductive development by regulating SWEETs expression.This study uncovers a novel mechanism whereby CsWRKY57L coordinately regulates both stress responses and growth in tea plants,providing a molecular basis for breeding low-temperature-tolerant varieties with restricted reproductive development. 展开更多

Heat stress reduces theanine content in tea plants,but the underlying molecular mechanism remains unclear.In this study,a temperature gradient treatment(20℃,25℃,30℃,and 35℃)was performed to unveil the effect of heat stress on biosynthesis and accumulation of theanine.We found that heat stress induced metabolic changes,characterized by decreased theanine content and increased catechin levels.In addition,heat stress up-regulated the expression of the class B heat shock transcription factor gene CsHSFB2c,while significantly suppressing the transcription of key theanine biosynthetic genes CsTS1 and CsGS1.Functional studies showed that silencing CsHSFB2c increased theanine content,while its overexpression significantly decreased theanine levels.Consistent with these changes,silencing CsHSFB2c upregulated the expression of CsTS1 and CsGS1,while overexpression of CsHSFB2c downregulated their expression.Yeast one-hybrid(Y1H)and dual-luciferase reporter gene(Dual-LUC)assays showed that CsHSFB2c directly binds to the promoters of CsTS1 and CsGS1 and inhibits their expression.These results demonstrate that CsHSFB2c mediates heat-induced suppression of theanine biosynthesis by directly inhibiting the expression of CsTS1 and CsGS1.This study provides a theoretical basis for improving the heat resistance and quality of tea plants via molecular breeding. 展开更多

[Method] The amino acid contents of fermented and unfermented the leaves of large-leaf Pu’er tea variety produced in Yunnan Mengku, were measured before and after the treatment with high-intensity pulsed electric field (HPEF). [Result] The difference of amino acid contents was notable between fermented and unfermented tea leaves produced in the same year. The amino acid content of unfermented tea leaves was larger than that in fermented tea leaves. The amino acid content in tea leaves decreased over time during storage. HPEF treatment under al the conditions improved the amino acid content in tea leaves. [Conclusion] This experiment provides theoretical foundation for the application of HPEF in processing of Pu’er tea leaves. 展开更多

Our previous study found that large-leaf yellow tea(LYT)had interesting hypoglycemic activity in high-fat diet-induced obese mice and highly safety in healthy mice. To study the anti-diabetic potential of LYT, the present study further investigated the preventive effects and mechanisms of action of LYT administration on diabetes and diabetic nephropathy in high-fat diet plus streptozotocin-induced diabetic mice. Results showed that LYT infusions(1/100 and 1/50, m/V)as drinking fluid for 4 weeks reduced diabetic polydipsia and polyuria, enhanced glucose tolerance and insulin sensitivity, and lowered fasting blood glucose level. The underlying mechanisms involve downregulation of gluconeogenesis(lower protein levels of TXNIP and FBP and enzyme activity of FBP), upregulation of lipid catabolism(higher protein levels of CPT-1α and PPARα), downregulation of lipogenesis(lower protein level of SREBP-1), and modification of the structure and abundance of gut microbiota to modulate metabolic homeostasis. Moreover, LYT administration prevented diabetic nephropathy, possibly due to reduced glucose-caused osmotic diuresis and lowered levels of renal PKC-β2, NLRP3 as well as membrane PKC-α, AQP2 and glycosylated AQP2 proteins. Taken together, LYT exhibits the activities in alleviating diabetic symptoms, ameliorating glucose and lipid dysmetabolism and fatty liver, and preventing diabetic nephropathy in diabetic mice. These activities may be explored for the prevention and treatment of diabetes in humans. 展开更多

Larger-leaf yellow tea(LYT)is a characteristic type of Chinese tea produced in Huoshan County,Anhui Province,which is made by mature leaves with stems.According to recent report,LYT showed competitive effects in anti-hyperglycemia in comparison to other teas such as green or black tea.However,the bioactive compounds of LYT are still undiscovered so far.For this purpose,5 fractions of LYT were prepared by sequential extraction.The in vitro bioassay results indicated that the ethyl acetate fraction of LYT had the strongest inhibitory effects onα-glucosidase andα-amylase.Fluorescence-quenching analysis and proteinbinding test revealed that the compounds of ethyl acetate fraction could inhibitα-glucosidase andα-amylase activities through binding to enzymes or other mechanisms.All chromatographic peaks of high-performance liquid chromatography(HPLC)of ethyl acetate fraction were separated and collected.The purified compounds were identified by liquid chromatography-mass spectrometry(LC-MS),and subsequently screened by calculating their inhibition ratio onα-glucosidase at the real concentration in LYT infusion.The results showed that(-)-epigallocatechin gallate,(-)-gallocatechin gallate,caffeine,N-ethyl-2-pyrrolidone-substituted flavan-3-ols were effective inhibitors forα-glucosidase. 展开更多

Analysis on Antibacterial, Detumescent and Antioxidant Activity of Huoshan Large-leaf Yellow Tea

[Objectives] To provide experimental basis for the effective development and utilization of Huoshan large-leaf yellow tea resources and the screening of safe and effective active ingredients of large-leaf yellow tea. ... [Objectives] To provide experimental basis for the effective development and utilization of Huoshan large-leaf yellow tea resources and the screening of safe and effective active ingredients of large-leaf yellow tea. [Methods] The active substances of Huoshan large-leaf yellow tea were extracted by hot-water extraction, and the freeze-dried powder of Huoshan large-leaf yellow tea was obtained by freeze drying. The antibacterial activity of the extract was preliminarily confirmed using the Oxford cup method, and its antimicrobial spectrum was analyzed using 14 strains. A xylene-induced mouse auricle swelling test was carried out to detect the swelling inhibition rate of the extract and analyze its in-vitro detumescent activity. Then, the antioxidant activity of the extract was identified through a DPPH free radical scavenging capacity test and a ferric reducing antioxidant power assay. [Results] The extract had significant inhibitory effects on various bacteria. The extract could effectively inhibit the growth of Escherichia coli, Staphylococcus aureus, Enterococcus hirae, Acinetobacter baumannii, Klebsiella pneumonia, Pseudomonas aeruginosa, Bacillus subtilis , and other strains. The diameter of the inhibition zone increased with the increase of sample concentration. The extract had a significant inhibitory effect on auricle swelling induced by xylene in mice. When the concentration of the drug reached 1.0 mg/mL, its inhibition rate on mouse auricle swelling reached 55.2% ( P <0.01), slightly lower than the swelling inhibition rate of the aspirin group (66.52%, P <0.01). The results of the antioxidant test showed that large-leaf yellow tea extract also had strong activity. Within the concentration range of 0.1-1.0 mg/mL, its DPPH radical scavenging rate increased with the increase of sample concentration. Within the concentration range of 0.1-1.0 mg/mL, its DPPH radical scavenging rate increased with the increase of sample concentration. When the concentration reached 1.0 mg/mL, the scavenging rate reached 69.75%. The Fe 3+ -reduction capacity of the extract also increased with the increase of sample concentration within the concentration range of 0.1-2.5 mg/mL. When the concentration was 2.5 mg/mL, the reducing power of the extract reached 1.43±0.04. However, its DPPH free radical scavenging rate and reducing power were slightly lower than the capacity of V C at the same concentration. [Conclusions] The extract of Huoshan large-leaf yellow tea obtained by hot-water extraction had strong activity in many aspects, including inhibiting the growth of various microbes, subsiding swelling in vitro and resisting oxidation. These experimental results provide certain guiding significance for the basic research of Huoshan large-leaf yellow tea extract, as well as experimental data support for the subsequent development of functional foods and drugs of Huoshan large-leaf yellow tea. 展开更多

Transcutaneous electrical acupoint stimulation(TEAS)is a kind of physical therapy that use electric cur-rent through the electrodes placed on the surface of acupoints to produce clinical effects in the human body,which is characterized by less adverse reaction and convenient operation.It has been widely used in the treatment of various diseases.This review introduces six major clinical applications of TEAS,named analgesia,regulation of gastrointestinal function,improvement of reproductive function,enhancement of cognitive function,promotion of limb function recovery and relief of fatigue.Besides,TEAS has been ap-plied to the treatment of other chronic diseases such as hypertension and diabetes,achieving satisfactory clinical effects.However,two crucial challenges are encountered in the development of TEAS.One is the lack of standardization in the selection of parameters such as waveform,frequency,intensity and stimula-tion duration.The other is the limitation on the flexibility in the acupoint selection.This review analyzes key issues that need to be addressed in the current clinical application of TEAS,such as the selection of parameters and acupoints,and this review provides a certain reference value for optimizing regimens of TEAS and promoting its development and application. 展开更多

Pyrrolizidine alkaloids(PAs)and their N-oxides(PANOs)are phytotoxins produced by various plant species and have been emerged as environmental pollutants.The sorption/desorption behaviors of PAs/PANOs in soil are crucial due to the horizontal transfer of these natural products from PA-producing plants to soil and subsequently absorbed by plant roots.This study firstly investigated the sorption/desorption behaviors of PAs/PANOs in tea plantation soils with distinct characteristics.Sorption amounts for seneciphylline(Sp)and seneciphylline-N-oxide(SpNO)in three acidic soils ranged from 2.9 to 5.9μg/g and 1.7 to 2.8μg/g,respectively.Desorption percentages for Sp and SpNO were from 22.2%to 30.5%and 36.1%to 43.9%.In the mixed PAs/PANOs systems,stronger sorption of PAs over PANOs was occurred in tested soils.Additionally,the Freundlich models more precisely described the sorption/desorption isotherms.Cation exchange capacity,sand content and total nitrogen were identified as major influencing factors by linear regression models.Overall,the soils exhibiting higher sorption capacities for compounds with greater hydrophobicity.PANOs were more likely to migrate within soils and be absorbed by tea plants.It contributes to the understanding of environmental fate of PAs/PANOs in tea plantations and provides basic data and clues for the development of PAs/PANOs reduction technology. 展开更多

[Objectives]To investigate the optimal preparation process for lemon-flavored solid instant tea.[Methods]The formulation process was optimized through a single-factor combined orthogonal test,utilizing large-leaf Pu'er tea as the primary raw material and perfume lemon as the auxiliary ingredient.Sensory scores and the effectiveness of the spray drying process served as the key indicators for this investigation.[Results]The optimal formulation for lemon-flavored solid instant tea was established as follows:15 g of Pu'er tea,25 g of perfume lemon,a solid-liquid ratio of 1:20(g/mL),an extraction temperature of 90℃,an extraction duration of 30 min,and air inlet and outlet temperatures of 200℃and 80℃,respectively.This formulation produced the most favorable flavor profile for the perfume lemon-flavored solid instant tea,which was characterized by a stable texture and a sensory evaluation score of 91.97 points.[Conclusions]This study has the potential to enhance the development and utilization of large-leaf Pu'er tea and perfume lemon resources. 展开更多

Effects of ultrasonic-assisted extraction on bioactive compounds,volatile flavors and antioxidant activities of vine tea water extracts

Background:Ampelopsis grossedentata,vine tea,which is the tea alternative beverages in China.In vine tea processing,a large amount of broken tea is produced,which has low commercial value.Methods:This study investigates the influence of different extraction methods(room temperature water extraction,boiling water extraction,ultrasonic-assisted room temperature water extraction,and ultrasonic-assisted boiling water extraction,referred to as room temperature water extraction(RE),boiling water extraction(BE),ultrasonic assistance at room temperature water extraction(URE),and ultrasonic assistance in boiling water extraction(UBE))on the yield,dihydromyricetin(DMY)content,free amino acid composition,volatile aroma components,and antioxidant properties of vine tea extracts.Results:A notable influence of extraction temperature on the yield of vine tea extracts(P<0.05),with BE yielding the highest at 43.13±0.26%,higher than that of RE(34.29±0.81%).Ultrasound-assisted extraction significantly increased the DMY content of the extracts(P<0.05),whereas DMY content in the RE extracts was 59.94±1.70%,that of URE reached 66.14±2.78%.Analysis revealed 17 amino acids,with L-serine and aspartic acid being the most abundant in the extracts,nevertheless ultrasound-assisted extraction reduced total free amino acid content.Gas chromatography-mass spectrometry analysis demonstrated an increase in the diversity and quantity of compounds in the vine tea water extracts obtained through ultrasonic-assisted extraction.Specifically,69 and 68 volatile compounds were found in URE and UBE extracts,which were higher than the number found in RE and BE extracts.In vitro,antioxidant activity assessments revealed varying antioxidant capacities among different extraction methods,with RE exhibiting the highest DPPH scavenging rate,URE leading in ABTS•+free radical scavenging,and BE demonstrating superior ferric ion reducing antioxidant activity.Conclusion:The findings suggest that extraction methods significantly influence the chemical composition and antioxidant properties of vine tea extracts.Ultrasonic-assisted extraction proved instrumental in elevating the DMY content in vine tea extracts,thereby enriching its flavor profile while maintaining its antioxidant properties. 展开更多

目的 探讨NOP2/Sun RNA甲基转移酶2(NSUN2)通过介导TEA结构域转录因子1(TEAD1)5-甲基胞嘧啶(m5C)甲基化修饰对胃癌(gastric cancer, GC)细胞糖酵解的影响及其机制。方法 实时荧光定量PCR(qRT-PCR)检测人胃癌(GC)组织中NSUN2和TEAD1 mRN... 目的 探讨NOP2/Sun RNA甲基转移酶2(NSUN2)通过介导TEA结构域转录因子1(TEAD1)5-甲基胞嘧啶(m5C)甲基化修饰对胃癌(gastric cancer, GC)细胞糖酵解的影响及其机制。方法 实时荧光定量PCR(qRT-PCR)检测人胃癌(GC)组织中NSUN2和TEAD1 mRNA表达水平,Pearson相关分析两者相关性。qRT-PCR和蛋白质印迹法(Western blot)检测人正常胃黏膜上皮细胞GES-1及胃癌AGS、HGC-27、SNU-1和MKN-45等细胞系中NSUN2表达水平。将NSUN2干扰慢病毒(sh-NSUN2)及其阴性对照sh-NC、TEAD1过表达质粒(oe-TEAD1)及其阴性对照(Vector)分别或同时转染至HGC-27细胞中,CCK-8检测细胞增殖活性。葡萄糖摄取、乳酸含量以及三磷酸腺苷(ATP)检测试剂盒评估细胞糖酵解。Western blot检测细胞中糖酵解途径相关蛋白(HK2、LDHA、GLUT1)表达水平。qRT-PCR和Western blot检测细胞中TEAD1 mRNA和蛋白表达水平。RNA测序(RNA-seq)和RNA甲基化免疫共沉淀实验筛选NSUN2的5-甲基胞嘧啶(m5C)修饰靶点,同时采用斑点杂交和双荧光素酶报告基因实验验证其调控功能。结果 与癌旁组织比较,GC组织中NSUN2和TEAD1 mRNA表达水平均显著升高(均P<0.05),且两者呈正相关(r=0.3266,P<0.05)。与GES-1细胞系比较,胃癌细胞系(AGS、HGC-27、SNU-1和MKN-45)中NSUN2 mRNA和蛋白表达水平显著升高,其中在HGC-27细胞系中表达最高(均P<0.05)。沉默NSUN2可显著降低HGC-27细胞增殖活性和糖酵解,下调HK2、LDHA和GLUT1蛋白表达水平,同时细胞中TEAD1 mRNA和蛋白表达水平显著降低(均P<0.05)。TEAD1是NSUN2调控的潜在靶点,沉默NSUN2通过m5C修饰的方式降低TEAD1的表达(P<0.05)。然而,过表达TEAD1可明显逆转沉默NSUN2对HGC-27细胞增殖活性和糖酵解的抑制作用。结论 NSUN2通过m5C依赖性的方式增强TEAD1的表达进而促进GC细胞的糖酵解。 展开更多
