Saussurea inversa is one of the Tibetan medicine with developed aerenchyma.In this study,we investigated the function of the ShCTR1 gene related to the formation of aerenchyma.The study of the occurrence and formation...Saussurea inversa is one of the Tibetan medicine with developed aerenchyma.In this study,we investigated the function of the ShCTR1 gene related to the formation of aerenchyma.The study of the occurrence and formation of aerenchyma in S.inversa has certain theoretical significance in revealing the special mechanism of alpine plants adapting to the environment.The results of yeast two-hybrid experiments showed that S.inversa ShCTR1 interacted with the ShRBOH1,which is a key producer of reactive oxygen species(ROS),and it was hypothesized that ShCTR1 regulation of aerenchyma formation in S.inversa mainly mediated by ROS.Aerenchyma induction and inhibition experiments were carried out on S.inversa seedlings,it was shown that ROS inducer(AT)and Ethene(ETH)induced the formation of aerenchyma in S.inversa well in roots,stems,and leaves,and Ethylene inhibitor(1-MCP)and ROS inhibitor DPI(diphenylene iodonium)inhibited the formation of aerenchyma of S.inversa in different degrees,which proved that ROS and ethylene(ET)were positively correlated with the formation of aerenchyma.Meanwhile,AT and ETH well induced the expression of the ShCTR1 gene,and 1-MCP and DPI inhibited the expression of the ShCTR1 gene to different degrees,which proved that ROS and ET were also positively correlated with the expression of the ShCTR1 gene.ShCTR1 was transferred into tobacco(Nicotiana tabacum L.),then repeat the above experiment.It was shown that positive rate and porosity were lowest and highest after DPI and AT treatments,respectively,it showed a positive correlation,which proved that the ShCTR1 gene was positively regulating the formation of aerenchyma.Overexpression of the ShCTR1 gene increased the content of ROS,consequently,the regulation of aerenchyma formation by the ShCTR1 gene is mainly mediated by ROS.It suggested that the formation of aerenchyma in S.inversa was related to hypoxia in the alpine environment,and ShCTR1 interacts with ShRBOH1 to positively regulate the formation of aerenchyma mainly mediated by ROS.展开更多
Hidradenitis suppurativa(HS),also known as acne inversa,is a chronic inflammatory skin disorder characterized by recurrent inflammatory nodules,abscesses,sinus tracts(tunnels),and hypertrophic scarring.The condition o...Hidradenitis suppurativa(HS),also known as acne inversa,is a chronic inflammatory skin disorder characterized by recurrent inflammatory nodules,abscesses,sinus tracts(tunnels),and hypertrophic scarring.The condition often causes significant psychological distress and markedly reduces quality of life.Although its pathogenesis remains incompletely understood,multiple contributing factors-including genetic predisposition,immune dysregulation,hormonal influences,and lifestyle factors-have been implicated.The disease’s recurrence,complexity,and poorly defined pathogenesis make effective treatment particularly challenging.While lifestyle modifications,weight loss,smoking cessation,topical therapies,and oral antibiotics may be effective in mild cases,moderate-to-severe HS remains difficult to manage.This narrative review summarizes recent advances in therapeutic strategies for HS based on published literature and clinical trials.Our goal is to update clinicians on current treatment developments and support improved disease management to enhance the quality of life for individuals affected by HS.展开更多
Familial acne inversa (AI) is an autoinflammatory disorder that affects hair follicles and is caused by loss-of-function mutations in y-secretase component genes.We and other researchers showed that nicastrin (NCSTN) ...Familial acne inversa (AI) is an autoinflammatory disorder that affects hair follicles and is caused by loss-of-function mutations in y-secretase component genes.We and other researchers showed that nicastrin (NCSTN) is the most frequently mutated gene in familial AI.In this study,we generated a keratin 5-Cre-driven epidermis-specific Ncstn conditional knockout mutant in mice.We determined that this mutant recapitulated the major phenotypes of AI,including hyperkeratosis of hair follicles and inflammation.In Ncstnflox/flox;K5-Cre mice,the IL-36a expression level markedly increased starting from postnatal day 0 (P0),and this increase occurred much earlier than those of TNF-α,IL-23A,IL-1 3,and TLR4.RNA-Seq analysis indicated that Sprr2d,a member of the small proline-rich protein 2 family,in the skin tissues of the Ncstnflox/flox,;K5-Cre mice was also upregulated on P0.Quantitative reverse-transcription polymerase chain reaction showed that other Sprr2 genes had a similar expression pattern.Our findings suggested that IL-36a might be a key inflammatory cytokine in the pathophysiology of AI and implicate malfunction of the skin barrier in the pathogenesis of AI.展开更多
Objective:This study was performed to estimate the prevalence of hidradenitis suppurativa(HS)in China and describe the demographic and clinical features of HS in the Chinese population.Methods:This multicenter clinic-...Objective:This study was performed to estimate the prevalence of hidradenitis suppurativa(HS)in China and describe the demographic and clinical features of HS in the Chinese population.Methods:This multicenter clinic-based cross-sectional study was conducted in dermatology clinics within 19 hospitals located in 15 cities/provinces across China from September to October 2020.The prevalence of HS was calculated as the percentage of patients with HS among all visitors at participating clinics during the 1-month study period.HS was independently diagnosed by two certified dermatologists at each site.All visitors at participating clinics were screened,and all patients with HS were invited to participate in a questionnaire survey to elucidate the demographics and clinical features of HS in the Chinese population.Results:We identified 92 patients with HS among 274,742 visitors at participating clinics.The prevalence rate was 0.03349%or 33.49 per 100,000 population(95%confidence interval,26.64–40.32),and the female:male ratio was 1.0:4.7.Conclusion:The overall prevalence of HS in China was lower than that in Western and other Asian countries with a predominance of male patients.展开更多
