The component E (CE)'s pattern of action with tumor cellular DNA at the molecular pharmacological level was investigated by macromolecular synthesis experiment (MSE) and human DNA interaction system established in our laboratory. The experiments demonstrated that, in vitro, the agent could markedly inhibit the incorporation of 3H-TdR into the cellular DNA, and the IC50 in P388 leukemia cell and in SGC-7901 cell was 17.86 μg/ml and 110.4 μg/ml, respectively. The agent, at mg/ml level, could produce an intercalation reversion pattern with DNA within a short time (2 hours). But when the interval was prolonged for over 4 hours, the action changed to intercalation irreversible pattern. According to these observations, the authors infer that CE interacts with DNA in two ways - directly and indirectly. The indirect action, especially in low concentrations, probably plays the major role. The authors have also compared the interaction of CE with those of components (CB3 and CD1), extracted from FCTM by the same methods, and found that CE is the most active agent against the DNA of cancer cells among the extracts from FCTM.展开更多

Self-assembly is the combination of active ingredients into aggregates through noncovalent bonds, such as hydrogen bonding, electrostatic interactions, ionic interactions, and hydrophobic interactions. The complex properties and special structures of CMM components create the basis for selfassembly. The self-assembled materials formed after CMM compatibility is an important part of the material basis for the efficacy of TCM, which can help explain the scientific connotations of CMM compatibility. This review summarizes the self-assembly phenomenon from the perspective of drug pair combinations in recent decades and explains the scientific connotation of CMM compatibility about the material basis, pharmacodynamic changes, and mechanism of action, providing new ideas and methods for the study of TCM.展开更多

However, PMR-associated hepatotoxicity is becoming a safety issue. In our previous in vivo study, an interaction between stilbenes and anthraquinones has been discovered and a hypothesis is proposed that the interaction between stilbene glucoside-enriching fraction and emodin may contribute to the side effects of PMR. To further support our previous in vivo results in rats, the present in vitro study was designed to evaluate the effects of 2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-D-glucopyranoside(TSG) on the cellular absorption and human liver microsome metabolism of emodin. The obtained results indicated that the absorption of emodin in Caco-2 cells was enhanced and the metabolism of emodin in human liver microsomes was inhibited after TSG treatment. The effects of the transport inhibitors on the cellular emodin accumulation were also examined. Western blot assay suggested that the depressed metabolism of emodin could be attributed to the down-regulation of UDP-glucuronosyltransferases(UGTs) 1A8, 1A10, and 2B7. These findings definitively demonstrated the existence of interaction between TSG and emodin, which provide a basis for a better understanding of the underlying mechanism for PMR-induced liver injury.展开更多
