BACKGROUND Depression is a prevalent affective disorder,but its pathophysiology remains unclear.Dysfunction in the gamma-aminobutyric acid(GABA)-ergic system may contribute to its onset.Recently,antidepressants(e.g.,brexanolone,zuranolone)targeting the GABA-A receptor were introduced.The zona incerta(ZI),an inhibitory subthalamic region mainly composed of GABAergic neurons,has been implicated in emotional regulation.Deep brain stimulation of the ZI in humans affects anxiety and depression symptoms,while activation of ZI neurons in mice can either worsen or alleviate anxiety.Currently,there is no direct evidence linking GABAergic neurons in the ZI to depression-like behaviors in rodents.AIM To explore the relationship between GABAergic neurons in the ZI and depression-like behaviors in mice.METHODS A chronic restraint stress(CRS)model was utilized to induce depression in mice.Whole-cell patch-clamp recordings assessed the excitability changes of GABAergic neurons in the ZI.Additionally,chemogenetic techniques were employed to modulate ZI GABAergic neurons.The performance of the mice in behavioral tests for depression and anxiety was observed.RESULTS The findings indicated that GABAergic neurons in the ZI were closely associated with depression-like behaviors in mice.Twenty-eight days after the CRS model was established,depression-like and anxiety-like behaviors were observed in the mice.The excitability of GABAergic neurons in the ZI was reduced.Chemogenetic activation of these neurons alleviated CRS-induced depression-like and anxiety-like behaviors.Conversely,inhibition of GABAergic neurons in the ZI led to changes in emotion-related behavioral outcomes in mice.CONCLUSION Activity of GABAergic neurons in the ZI was closely associated with depression-like phenotypes in mice,suggesting that these neurons could be a potential therapeutic target for treating depression. 展开更多

Parkinson’s disease(PD)is the second most common and fastest-growing neurodegenerative disorder.In recent years,it has been recognized that neurotransmitters other than dopamine and neuronal systems outside the basal ganglia are also related to PD pathogenesis.However,little is known about whether and how the caudal zona incerta(ZIc)regulates parkinsonian motor symptoms.Here,we showed that specific glutamatergic but not GABAergic ZIc^(VgluT2) neurons regulated these symptoms.ZIc^(VgluT2) neuronal activation induced time-locked parkinsonian motor symptoms.In mouse models of PD,the ZIc^(VgluT2) neurons were hyperactive and inhibition of their activity ameliorated the motor deficits.ZIc^(VgluT2) neurons monosynaptically projected to the substantia nigra pars reticulata.Incerta-nigral circuit activation induced parkinsonian motor symptoms.Together,our findings provide a direct link between the ZIc,its glutamatergic neurons,and parkinsonian motor symptoms for the first time,help to better understand the mechanisms of PD,and supply a new important potential therapeutic target for PD. 展开更多

The zona incerta(ZI)is involved in various functions and may serve as an integrative node of the circuits for global behavioral modulation.However,the long-range connectivity of different sectors in the mouse ZI has not been comprehensively mapped.Here,we obtained whole-brain images of the input and output connections via fluorescence micro-optical sectioning tomography and viral tracing.The principal regions in the input-output circuits of ZI GABAergic neurons were topologically organized.The 3D distribution of cortical inputs showed rostro-caudal correspondence with different ZI sectors,while the projection fibers from ZI sectors were longitudinally organized in the superior colliculus.Clustering results show that the medial and lateral ZI are two different major functional compartments,and they can be further divided into more subdomains based on projection and input connectivity.This study provides a comprehensive anatomical foundation for understanding how the ZI is involved in integrating different information,conveying motivational states,and modulating global behaviors. 展开更多

Defensive behaviors induced by innate fear or Pavlovian fear conditioning are crucial for animals to avoid threats and ensure survival.The zona incerta(ZI)has been demonstrated to play important roles in fear learning and fear memory,as well as modulating auditory-induced innate defensive behavior.However,whether the neuronal subtypes in the ZI and specific circuits can mediate the innate fear response is largely unknown.Here,we found that somatostatin(SST)-positive neurons in the rostral ZI of mice were activated by a visual innate fear stimulus.Optogenetic inhibition of SST-positive neurons in the rostral ZI resulted in reduced flight responses to an overhead looming stimulus.Optogenetic activation of SST-positive neurons in the rostral ZI induced fear-like defensive behavior including increased immobility and bradycardia.In addition,we demonstrated that manipulation of the GABAergic projections from SST-positive neurons in the rostral ZI to the downstream nucleus reuniens(Re)mediated fear-like defensive behavior.Retrograde trans-synaptic tracing also revealed looming stimulus-activated neurons in the superior colliculus(SC)that projected to the Re-projecting SST-positive neurons in the rostral ZI(SC-ZIrSST-Re pathway).Together,our study elucidates the function of SST-positive neurons in the rostral ZI and the SC-ZIrSST-Re tri-synaptic circuit in mediating the innate fear response. 展开更多

AIM:To evaluate different promising magnetic resonance imaging(MRI) methods at 7.0 Tesla(T) for the pre-stereotactic visualization of the zona incerta(ZI).METHODS:Two neuroradiologists qualitatively and quantitatively examined T2-turbo spin-echo(T2-TSE),T1-weighted gradient-echo,as well as FLASH2D-T2Star and susceptibility-weighted imaging(SWI) for the visualization of the ZI at 7.0 T MRI.Delineation and image quality for the ZI were independently examined using a 6-scale grading system.Inter-rater reliability using Cohen's kappa coefficient(κ) were assessed.Contrast-tonoise ratios(CNR),and signal-to-noise ratios(SNR) for the ZI were calculated for all sequences.Differences in delineation,SNR,and CNR between the sequences were statistically assessed using a paired t-test.For the anatomic validation the coronal FLASH2D-T2Star images were co-registered with a stereotactic atlas(Schaltenbrand-Wahren).RESULTS:The rostral part of the ZI(rZI) could easily be identified and was best and reliably visualized in the coronal FLASH2D-T2Star images.The caudal part was not definable in any of the sequences.No major artifacts in the rZI were observed in any of the scans.FLASH2D-T2Star and SWI imaging offered significant higher CNR values for the rZI compared to T2-TSE images(P > 0.05).The co-registration of the coronal FLASH2D-T2Star images with the stereotactic atlas schema(Schaltenbrand-Wahren) confirmed the correct localization of the ZI in all cases.CONCLUSION:FLASH2D-T2Star imaging(particularly coronal view) provides the reliable and currently optimal visualization of the rZI at 7.0 T.These results can facilitate a better and more precise targeting of the caudal part of the ZI than ever before. 展开更多

为明确荒漠植物囊果草生物学特性及其种群分布格局的生态适应意义,以北疆地区温性荒漠中的特有植物囊果草为研究对象,对其生物学特性、种子传播方式及野外种群的分布格局进行了观测。结果表明,囊果草具膀胱状膜质果囊和膨大的不规则块... 为明确荒漠植物囊果草生物学特性及其种群分布格局的生态适应意义,以北疆地区温性荒漠中的特有植物囊果草为研究对象,对其生物学特性、种子传播方式及野外种群的分布格局进行了观测。结果表明,囊果草具膀胱状膜质果囊和膨大的不规则块状根茎,每株平均具果囊8.29个,果囊底部和侧面具网状脉络,且底部脉络较粗,不易破裂,顶部无脉络但具蓝紫斑,成熟时易破裂,每个果囊内平均包含球形种子2.82个。囊果草的花具有暂时闭合特性。囊果草为风滚式传播植物,一方面,地上果囊随风滚动时释放内部种子完成远距离传播,随着果囊体积的增大,果囊在不同粗糙程度微地形条件下的移动距离呈线性递增趋势,随地面粗糙度的增加,果囊最短移动距离显著减小(P<0.05),散布的种子数量则显著增加(P<0.05),且果囊移动距离与散布种子数量之间显著负相关(r=0.30,P<0.05);另一方面,地下块状根茎以营养生殖的方式在原位萌发。囊果草种群的分布格局与其生殖方式密切相关。在小尺度范围内(0~50m),随着空间尺度的扩大,囊果草种群幼苗和成株的分布格局表现为从均匀分布到聚集分布再到随机分布的特点;在中等尺度范围内(3.0~17.5m),幼苗和成株的空间关联性表现为正关联,随着空间尺度的递增,二者之间的关联性逐渐减小且关联性不显著。 展开更多

BACKGROUND Hepatitis B cirrhosis(HBC)is a chronic disease characterized by irreversible diffuse liver damage and aggravated by intestinal microbial imbalance and metabolic dysfunction.Although the relationship between certain single probiotics and HBC has been explored,the impact of the complex ready-to-eat Lactobacillus paracasei N1115(LP N1115)supplement on patients with HBC has not been determined.AIM To compare the changes in the microbiota,inflammatory factor levels,and liver function before and after probiotic treatment in HBC patients.METHODS This study included 160 HBC patients diagnosed at the General Hospital of Ningxia Medical University between October 2018 and December 2020.Patients were randomly divided into an intervention group that received LP N1115 supplementation and routine treatment and a control group that received routine treatment only.Fecal samples were collected at the onset and conclusion of the 12-wk intervention period.The structure of the intestinal microbiota and the levels of serological indicators,such as liver function and inflammatory factors,were assessed.RESULTS Following LP N1115 intervention,the intestinal microbial diversity significantly increased in the intervention group(P<0.05),and the structure of the intestinal microbiota was characterized by an increase in the proportions of probiotic microbes and a reduction in harmful bacteria.Additionally,the intervention group demonstrated notable improvements in liver function indices and significantly lower levels of inflammatory factors(P<0.05).CONCLUSION LP N1115 is a promising treatment for ameliorating intestinal microbial imbalance in HBC patients by modulating the structure of the intestinal microbiota,improving liver function,and reducing inflammatory factor levels. 展开更多

The functional role of glutamatergic(vGluT2)neurons in the pedunculopontine nucleus(PPN)in modulating motor activity remains controversial.Here,we demonstrated that the activity of vGluT2 neurons in the rostral PPN is correlated with locomotion and ipsilateral head-turning.Beyond these motor functions,we found that these rostral PPN-vGluT2 neurons remarkably respond to salient stimuli.Furthermore,we systematically traced the upstream and downstream projections of these neurons and identifed two downstream projections from these neurons to the caudal pontine reticular nucleus/anterior gigantocellular reticular nucleus(PnC/GiA)and the zona incerta(ZI).Our fndings indicate that the projections to the PnC/GiA inhibit movement,consistent with‘pause-and-play’behavior,whereas those to the ZI promote locomotion,and others respond to a new‘pause-switch-play’pattern.Collectively,these fndings elucidate the multifaceted infuence of the PPN on motor functions and provide a robust theoretical framework for understanding its physiological and potential therapeutic implications. 展开更多

目的探索未定带区(ZI)γ-氨基丁酸(GABA)能神经元对七氟醚和丙泊酚麻醉-觉醒的调控作用。方法将48只雄性C57BL/6J小鼠分为8组(6只/组)。研究七氟醚麻醉时,化学遗传学实验分为hM3Dq组(注射携带hM3Dq的腺相关病毒)和mCherry组(注射仅携带m... 目的探索未定带区(ZI)γ-氨基丁酸(GABA)能神经元对七氟醚和丙泊酚麻醉-觉醒的调控作用。方法将48只雄性C57BL/6J小鼠分为8组(6只/组)。研究七氟醚麻醉时,化学遗传学实验分为hM3Dq组(注射携带hM3Dq的腺相关病毒)和mCherry组(注射仅携带mCherry的对照病毒);光遗传学实验分为ChR2组(注射携带ChR2的腺相关病毒)和GFP组(注射仅携带GFP的对照病毒),注射剂量均为100 nL。研究丙泊酚麻醉时,化学遗传学实验分为hM3Dq组和mCherry组;光遗传学实验分为ChR2组和GFP组。上述各组处理方法同七氟醚。通过化学遗传学和光遗传学技术激活ZI区GABA能神经元,观察其对七氟醚和丙泊酚的麻醉诱导及觉醒的调节作用,通过脑电监测观察其在七氟醚麻醉维持中的作用。结果在七氟醚麻醉中,与mCherry组比,hM3Dq组麻醉诱导时间显著缩短(P<0.05),与GFP组比,ChR2组麻醉诱导时间显著缩短(P<0.01),而两实验中麻醉觉醒时间差异均无统计学意义。在丙泊酚麻醉中,hM3Dq组比mCherry组麻醉诱导时间显著缩短(P<0.05),ChR2组比GFP组麻醉诱导时间显著缩短(P<0.01),而两实验中麻醉觉醒时间差异均无统计学意义。光遗传激活ZI区GABA能神经元没有改变七氟醚麻醉维持时的脑电频谱。结论ZI区GABA能神经元能够促进七氟醚和丙泊酚的麻醉诱导,而对麻醉维持及觉醒无调节作用。 展开更多

囊果草(Leontice incerta Pall.)为生长于天山北坡荒漠和低山山坡的小檗科(Berberidaceae)多年生早春开花植物,具有较高的观赏价值、饲用价值和生态价值,为探究该物种的有性生殖特征,并为后续进行栽培育种等工作提供理论依据,作者利用... 囊果草(Leontice incerta Pall.)为生长于天山北坡荒漠和低山山坡的小檗科(Berberidaceae)多年生早春开花植物,具有较高的观赏价值、饲用价值和生态价值,为探究该物种的有性生殖特征,并为后续进行栽培育种等工作提供理论依据,作者利用光学显微镜和石蜡制片技术,对该物种大/小孢子发生和雌/雄配子体发育进行了观察。结果表明,(1)花药具4室,药壁由5层细胞组成,发育为基本型。药室内壁在发育后期具纤维状加厚现象;腺质型绒毡层具2核或多核现象。(2)小孢子母细胞减数分裂时胞质分裂为同时型;小孢子四分体为四面体型,被胼胝质壁所包围,游离小孢子形成后胼胝质壁逐渐消失。成熟花粉多为2细胞型,偶见3细胞型。(3)雌蕊由1心皮组成,子房1室,倒生胚珠,具双珠被,厚珠心。大孢子四分体呈线形排列,最终发育为7细胞8核的蓼型胚囊,助细胞具发达的漏斗形丝状器。这些特点说明,囊果草大/小孢子发生和雌/雄配子体发育正常,未见败育现象,表现出较原始的发育特征。研究结果丰富了小檗科植物的胚胎学资料,为该物种及其近缘物种的生殖生物学研究积累了理论基础。 展开更多

未定带GABA能神经元活性降低介导脂多糖诱导的小鼠运动能力受损 被引量:1

目的:通过激活未定带(ZI)γ-氨基丁酸(GABA)能神经元,观察脂多糖(LPS)处理后小鼠运动能力损伤的缓解情况,探讨GABA能神经元在中枢神经系统感染模型小鼠运动功能损伤过程中的作用。方法:利用腹膜腔注射LPS方法制备C57BL/6小鼠中枢神经系... 目的:通过激活未定带(ZI)γ-氨基丁酸(GABA)能神经元,观察脂多糖(LPS)处理后小鼠运动能力损伤的缓解情况,探讨GABA能神经元在中枢神经系统感染模型小鼠运动功能损伤过程中的作用。方法:利用腹膜腔注射LPS方法制备C57BL/6小鼠中枢神经系统感染模型,利用旷场实验与转轮实验检测小鼠运动功能的变化,免疫荧光染色观察小鼠ZI神经元c-Fos和离子钙结合适配器分子1(Iba-1)的表达。利用立体定位技术将rAAV-DIO-hM3Dq-mCherry注射至成年雄性VGAT-Cre转基因小鼠双侧ZI区,等待3周病毒表达充分后,进行腹腔注射LPS制备神经炎症感染模型,行为检测30 min前腹腔注射N-氧化氯氮平(CNO)激活GABA能神经元,利用旷场实验与转轮实验检测小鼠运动功能的变化。结果:旷场实验与转轮实验结果显示,与对照组相比,LPS组小鼠活动度显著降低(P<0.05),小鼠ZI内c-Fos阳性蛋白表达显著下调,同时Iba-1阳性蛋白表达显著增加;并且化学遗传结果显示,与LPS组相比,激活小鼠ZI部位GABA能神经元后LPS小鼠活动能力显著增强(P<0.05)。结论:神经炎症状态小鼠ZI部位GABA能神经元活性显著降低,运动能力受损,化学遗传激活ZI部位GABA能神经元显著改善了炎症状态小鼠的运动能力。 展开更多

目的:在慢性瘙痒模型上检测小鼠的痒行为和焦虑、抑郁样行为,并观察未定带腹侧区(ZIv)内γ-氨基丁酸(GABA)能神经元的激活情况,为其参与痒觉信息的调控提供形态学证据。方法:应用二苯基环丙烯酮(DCP)在谷氨酸脱羧酶67-绿色荧光蛋白(GAD6... 目的:在慢性瘙痒模型上检测小鼠的痒行为和焦虑、抑郁样行为,并观察未定带腹侧区(ZIv)内γ-氨基丁酸(GABA)能神经元的激活情况,为其参与痒觉信息的调控提供形态学证据。方法:应用二苯基环丙烯酮(DCP)在谷氨酸脱羧酶67-绿色荧光蛋白(GAD67-GFP)基因敲入小鼠上建立慢性瘙痒模型,然后运用视频跟踪系统检测痒行为,以验证模型制备是否成功。运用高架十字迷宫实验、悬尾实验检测慢性痒模型小鼠的焦虑、抑郁样行为。利用GAD67-GFP小鼠观察未定带(ZI)各区域内GABA能神经元的分布情况,并结合免疫荧光染色方法分别观察对照组和DCP组小鼠ZIv内GABA能神经元与FOS的双标情况。结果:在GAD67-GFP小鼠的脑切片上可观察到GABA能神经元在未定带各亚区均有分布,且更集中于ZIv。DCP组小鼠抓挠次数显著高于对照组小鼠(P<0.001)。悬尾实验中DCP组小鼠完全不动时间显著高于对照组小鼠,出现抑郁样不动状态。高架十字迷宫实验结果显示DCP组小鼠与对照组相比,进入十字交叉区域的次数和时间占比均明显减少,表现出焦虑样行为。免疫荧光染色结果显示,DCP组小鼠ZIv内FOS阳性细胞数量明显高于对照组,且在该区域观察到大量的FOS与GABA能神经元的共标细胞。结论:小鼠未定带以GABA能神经元分布为主,且更集中于ZIv;慢性痒状态下,ZIv内GABA能神经元激活,为ZIv内GABA能神经元参与慢性痒及其伴发的负性情绪提供了形态学证据。 展开更多

目的:利用囊泡膜谷氨酸转运体2(VGluT2)⁃ires⁃cre、谷氨酸脱羧酶2(GAD2)⁃ires⁃cre和羟色胺转运体(Sert)⁃ires⁃cre小鼠结合重组狂犬病毒介导的逆行跨单级突触追踪技术,研究未定带(ZI)和中脑导水管周围灰质腹外侧区(vlPAG)内特异性神经元... 目的:利用囊泡膜谷氨酸转运体2(VGluT2)⁃ires⁃cre、谷氨酸脱羧酶2(GAD2)⁃ires⁃cre和羟色胺转运体(Sert)⁃ires⁃cre小鼠结合重组狂犬病毒介导的逆行跨单级突触追踪技术,研究未定带(ZI)和中脑导水管周围灰质腹外侧区(vlPAG)内特异性神经元之间的纤维联系。方法:首先将辅助病毒注入转基因小鼠右侧vlPAG内,两周过后将重组狂犬病毒(RV)注入相同区域。一周后,在激光共聚焦显微镜下观察RV标记的突触前神经元在ZI内的分布。结果:将RV注入vlPAG后,在VGluT2⁃ires⁃cre小鼠ZI的吻尾方向上均可观察到逆标的突触前神经元分布;GAD2⁃ires⁃cre小鼠逆标神经元只见于ZI吻侧部,但Sert⁃ires⁃cre小鼠ZI内少见RV逆行标记的突触前神经元。结论:ZI神经元发出投射至vlPAG,作用于其中的谷氨酸能神经元或GABA能神经元,该通路可能参与了疼痛或恐惧样行为的调控。 展开更多
