There were 13 ma...Fifteen cases of fresh fractures have been studied, including 14 cases of tubu-lar bone fractures with age from 28 to 60 years and one case of vertebral fracture ofdorsolumbar region, 16 years of age. There were 13 male and 2 female patients. Theyhad no endocrine disturbances or other bone diseases. The ratio ofhydroxyproline/creatinine (HP/Cr) in these cases increased gradually within 20-30 daysafter fracture and then reached the peak value which was about 3-4 times of normal, thepeak value maintained for a long time. If the trauma was mild or the patient was young,the peak began to decline on the 60th day after fracture. While in those severe cases withprolonged confinement to bed, the peak only began to come down about 110th dayafter fracture. The ratio of HP/Cr decreased rapidly from the peak at first, then pursueda slow course. Generally, the ratio of HP/Cr returned to normal on the 240th to 310thday. Bone union began at a time when peak value started to decline, and with completionof the remodelling process, the HP/Cr value returned to normal.展开更多

A diet containing 50% crude protein and 12% crude lipid was formulated as the basal and control, on which other two protein and lipid contents identical experimental diets were formulated by supplementing the basal with either 0.75% Pro(Pro-0.75) or 0.75% Pro and 0.75% Hyp(Pro+Hyp). Four groups of fish in indoor seawater recirculating systems, 35 individuals each, were fed twice a day to apparent satiation for 10 weeks. The results showed that dietary Pro and Hyp supplementation had no significant effect on growth performance and feed utilization of juvenile turbot(P > 0.05). Total Hyp and collagen concentrations in muscle were significantly increased when dietary Pro and Hyp increased(P < 0.05), and fish fed diet Pro+Hyp showed significantly higher free Hyp content in plasma than those fed other diets(P < 0.05). The expression of P4 H α(I) gene in liver and muscle was significantly up regulated in fish fed diet Pro-0.75 in comparison with control(P < 0.05); however the gene was significantly down regulated in fish fed diet Pro+Hyp in muscle in comparison with fish fed diet Pro-0.75(P < 0.05). It can be concluded that supplement of crystal L-Pro and L-Hyp to high plant protein diets did not show positive effects on growth performance of juvenile turbot, but enhanced total collagen concentrations in muscle.展开更多

In SH medium supplemented with 1 mg/L 2, 4-dichlorophenoxy-acetic acid (2,4-D), 0.5 mg/L kinetin (KT) and 0.2 mg/L naphthalene acetic acid (NAA), the division frequency of protoplastderived cells reached uP to over 60 %, and microcalli were obtained in 5-6 wk. Upon transferring them on agar solidified MS medium plus 2 mg/L indole-3-acetic acid (IAA), shoots were induced. After cultivating them on MS medium with or without IAA, roots were regenerated.Chromosome number of all protoplast-regenerated plants examined were normal (2n=28). The result of this study shows a significant correlation between Vitamin C and Hydroxyproline at level 0.01. This study investigated the effects of hydroxyproline(Hyp)and vitamin C(VC)addition to practical diet on growth and flesh quality of Pacific white shrimp(Litopenaeus vannamei)cultured in low salinity water.The control...This study investigated the effects of hydroxyproline(Hyp)and vitamin C(VC)addition to practical diet on growth and flesh quality of Pacific white shrimp(Litopenaeus vannamei)cultured in low salinity water.The control diet was formulated to contain 200 g/kg fish meal(FM20),and then 14 g/kg coated hydroxyproline(50%)(FM20H),2 g/kg L-ascorbyl-2-phosphate(38%)(FM20C)and their combination(FM20HC)were added to the control diet to form four isoproteic(41.6%)and isolipidic(6.7%)diets.Pacific white shrimp with initial body weight of(1.50±0.05)g were fed the above four diets in low salinity water(salinity 1‰)for 8 weeks.The results showed that the combined supplementation of Hyp and VC(FM20HC)increased the weight gain by 13.4%,and decreased the feed conversion ratio by 0.18(P<0.05),while the individual supplementation(FM20H and FM20C groups)just numerically promoted the growth(P>0.05).In addition,the FM20HC group also showed higher protein and lipid retention than the FM20 group(P<0.05).In flesh quality,the supple-mentation of Hyp(FM20H)increased total collagen and heat-soluble collagen contents,flesh chewiness and free flavor amino acids contents(P<0.05).The supplementation of vC(FM20C)increased the contents of total collagen,heat-insoluble collagen and free flavor amino acids,and decreased flesh lactic acid,malondialdehyde contents and steaming loss(P<0.05).The combined supplementation of Hyp and VC(FM20HC)showed higher total collagen and heat-insoluble collagen contents than the individual supplementation(FM20H and FM20C)(P<0.05),and the myofiber density,flesh hardness and shear force in the FM20HC group were also significantly higher than those in the control(P<0.05).In summary,the combined supplementation of hydroxyproline and vc in practical diet with fishmeal content of 200 g/kg significantly improved the growth performance and flesh quality of L.vannamei cultured in low salinity water.展开更多
Hardness is widely regarded as a critical factor influencing the whole texture of fish flesh.The objective of this study was to elucidate the regulatory mechanism underlying muscle hardness in hybrid bream(BBTB,Megalo...Hardness is widely regarded as a critical factor influencing the whole texture of fish flesh.The objective of this study was to elucidate the regulatory mechanism underlying muscle hardness in hybrid bream(BBTB,Megalobrama amblycephala♀×Culter alburnus♂).A comparison of the physiological features of high hardness(HH)and low hardness(LH)muscle revealed that the former had higher contents of collagen I and lower muscle fiber diameter.Transcriptomic data revealed that the myofiber assembly pathway and the HIF-1 signaling pathway were activated in HH muscle.At the metabolic level,the categories of amino acids and lipids were the principal differentially abundant metabolites between the HH and LH muscle.The detection of amino acid profiles further revealed significant differences in amino acid metabolism between the HH and LH muscles,with the HH muscle having higher levels of amino acids than the LH muscle,especially hydroxypro-line(Hyp).Furthermore,through supplementation of Hyp in BBTB myoblasts,the results indicated that 0.8 mmol/L Hyp increased the proliferation,differentiation,migration,and collagen synthesis of myoblasts.Finally,BBTB was treated with Hyp intraperitoneally for 15 days.The results revealed that 0.1 g/kg Hyp significantly increased muscle hardness,myofiber density,myofibrillar protein synthesis,and HIF-1 protein content.The results obtained in this study indicated that Hyp supplementation promoted collagen synthesis and proliferation of myoblast and muscle fibers in the BBTB,which may be induced by activation of the transcription factor HIF1 and contributes to the impacts of Hyp on improvements in muscle hardness in the BBTB.展开更多
This experiment aimed to evaluate the impact of dietary hydroxyproline(Hyp)supplementation on the muscle quality of juvenile Pacific white shrimp(Litopenaeus vannamei)fed a low fishmeal diet.Six formulated diets inclu...This experiment aimed to evaluate the impact of dietary hydroxyproline(Hyp)supplementation on the muscle quality of juvenile Pacific white shrimp(Litopenaeus vannamei)fed a low fishmeal diet.Six formulated diets included one high fishmeal(HF;25%fishmeal content)and five low fishmeal diets(10%fishmeal content)with 0%,0.2%,0.4%,0.6%and 0.8%Hyp(LFO,LF2,LF4,LF6 and LF8,respectively).Each diet was assigned to four replicates,and 40 shrimp(0.32±0.00 g)per replicate were fed four times a day for 8 weeks.Dietary Hyp supplementation had little effects on growth performance,but increased the contents of Hyp,prolyl 4-hydroxylases(P4Hs),and collagen.The meat yield,springiness,hardness,chewiness,and cohesiveness of muscle were the highest in the LF4 group among the low fishmeal groups(P<0.05).Cooking loss and freezing loss of muscle were the lowest in the LF4 group(P<0.05).Dietary supplementation with 0.4%Hyp increased the myofiber density and decreased the myofiber diameter of muscle(P<0.05).Supplementation of Hyp in the diet up-regulated the mRNA expression of smyhc5,smyhc15,col1a1,col1a2,igf-1f,tgf-β and tor and down-regulated the mRNA expression of smy hc1,smyhc2,smyhc6a(P<0.05).Supplementation of Hyp in the diet up-regulated the protein expression of P-4E-BP1,P-AKT,AKT and P-AKT/AKT(P<0.05).These results suggested that the addition of 0.4%Hyp to low fishmeal diets improved the muscle quality of L.vannamei.展开更多

Biochemical markers of bone resorption that reflect osteoclast activity and/or collagen degradation provide a new and potentially important clinical tool for the assessment and monitoring of bone metabolism. Assessment of bone resorption can be achieved with measurement of urinary hydroxylysine glycosides, urinary excretion of the collagen pyridinium cross-links, urinary excretion of type I collagen telopeptide breakdown products (cross-linked telopeptides) and urinary hydroxyproline.Conclusion Urinary hydroxyproline has been in use as a marker of bone resorption, but it lacks sensitivity and specificity. It is a modified aminoacid that is a metabolic product of collagen breakdown. Hydroxyproline may be released either free or with fragments of the collagen molecule attached during bone resorption, and it is also liberated by the breakdown of complement and nonskeletal collagen.展开更多

Compelling evidence has beengathered showing the potential antioxidative properties of Hyp. However, the role and mechanism ofHyp in porcine intestinal epithelial cells in response to oxidative stress remains unknown. In this study,small intestinal epithelial cell lines of piglets (IPEC-1) were used to evaluate the protective effects of Hypon 4-hydroxy-2-nonenal (4-HNE)-induced oxidative DNA damage and apoptosis. IPEC-1 pretreated with0.5 to 5 mmol/L Hyp were exposed to 4-HNE (40 mmol/L) in the presence or absence of Hyp. Thereafter,the cells were subjected to apoptosis detection by Hoechst staining, flow cytometry, and Western blot orDNA damage analysis by comet assay, immunofluorescence, and reverse-transcription quantitative PCR(RT-qPCR). Cell apoptosis and the upregulation of cleaved-caspase-3 induced by 4-HNE (40 mmol/L) wereinhibited by 5 mmol/L of Hyp. In addition, 5 mmol/L Hyp attenuated 4-HNE-induced reactive oxygenspecies (ROS) accumulation, glutathione (GSH) deprivation and DNA damage. The elevation in transcriptionof GADD45a (growth arrest and DNA-damage-inducible protein 45 alpha) and GADD45b(growth arrest and DNA-damage-inducible protein 45 beta), as well as the phosphorylation of H2AX(H2A histone family, member X), p38 MAPK (mitogen-activated protein kinase), and JNK (c-Jun N-terminalkinase) in cells treated with 4-HNE were alleviated by 5 mmol/L Hyp. Furthermore, Hyp supplementationincreased the protein abundance of Krüppel like factor 4 (KLF4) in cells exposed to 4-HNE.Suppression of KLF4 expression by kenpaulone impeded the resistance of Hyp-treated cells to DNAdamage and apoptosis induced by 4-HNE. Collectively, our results indicated that Hyp serves to protectagainst 4-HNE-induced apoptosis and DNA damage in IPEC-1 cells, which is partially pertinent with theenhanced expression of KLF4. Our data provides an updated explanation for the nutritional values ofHyp-containing animal products.展开更多
BACKGROUND Growth hormone(GH)plays a crucial role in wound healing and tissue repair in postoperative patients.In particular,colonic anastomosis healing following colorectal surgery is impaired by numerous chemotherap...BACKGROUND Growth hormone(GH)plays a crucial role in wound healing and tissue repair in postoperative patients.In particular,colonic anastomosis healing following colorectal surgery is impaired by numerous chemotherapy agents.AIM To investigate whether GH can improve the healing of a colonic anastomosis following the adverse effects of intraperitoneal administration of 5-fluorouracil(5-FU),bleomycin and cisplatin.METHODS Eighty Wistar rats underwent laparotomy and a 1 cm-resection of the transverse colon,followed by an end-to-end anastomosis under general anesthesia.The rats were blindly allocated into four equal groups and administered a different daily intraperitoneal therapeutic regimen for 6 days.The control group(A)received normal saline.Group B received chemotherapy with 5-FU(20 mg/kg),bleomycin(4 mg/kg)and cisplatin(0.7 mg/kg).Group C received GH(2 mg/kg),and group D received the aforementioned combination chemotherapy and GH,as described.The rats were sacrificed on the 7th postoperative day and the anastomoses were macroscopically and microscop-ically examined.Body weight,bursting pressure,hydroxyproline levels and inflammation markers were measured.RESULTS All rats survived until the day of sacrifice,with no infections or other complications.A decrease in the body weight of group D rats was observed,not statistically significant compared to group A(P=1),but significantly different to groups C(P=0.001)and B(P<0.01).Anastomotic dehiscence rate was not statistically different between the groups.Bursting pressure was not significantly different between groups A and D(P=1.0),whereas group B had a significantly lower bursting pressure compared to group D(P<0.001).All groups had significantly more adhesions than group A.Hydroxyproline,as a measurement of collagen deposition,was significantly higher in group D compared to group B(P<0.05),and higher,but not statistically significant,compared to group A.Significant changes in group D were recorded,compared to group A regarding inflammation(3.450 vs 2.900,P=0.016)and fibroblast activity(2.75 vs 3.25,P=0.021).Neoangiogenesis and collagen deposition were not signifi-cantly different between groups A and D.Collagen deposition was significantly increased in group D compared to group B(P<0.001).CONCLUSION Intraperitoneal administration of chemotherapy has an adverse effect on the healing process of colonic anastomosis.However,GH can inhibit the deleterious effect of administered chemotherapy agents and induce colonic healing in rats.展开更多

AIM: To evaluate the antifibrotic effect of different doses of recombinant human Gamma-Interferon (IFN-gamma) in two rat models of hepatic fibrosis, and to observe its effect on moderate chronic hepatitis B virus fibr...AIM: To evaluate the antifibrotic effect of different doses of recombinant human Gamma-Interferon (IFN-gamma) in two rat models of hepatic fibrosis, and to observe its effect on moderate chronic hepatitis B virus fibrosis. METHODS: Hepatic fibrosis was successfully induced in 150 and 196 rats by subcutaneous injection of carbon tetrachloride (CCl4) and intraperitoneal injection of dimethylnitrosamine (DMN), respectively. Each of the two model groups was divided into: (1) fibrotic model group; (2) colchicine treatment group (0.1 mg/kg/day, gastrogavage for 8 weeks); (3) high-dose IFN-gamma group (15 MU/kg per day, i.m. for 8 weeks); (4) medium-dose IFN-gamma group (5 MU/kg daily, i.m. for 8 weeks); and (5) Y low-dose IFN-gamma group (1.67 MU/kg daily, i.m. for 8 weeks). Another group of 10 rats without any treatment was used as normal controls. At the end of the experiment, semi-quantitative histopathological scores of inflammation and fibrosis, liver alpha smooth muscle actin (alpha-SMA) expression level, liver hydroxyl proline content and serum hyaluronic acid levels were compared. And 47 medium chronic hepatitis B viral fibrosis patients were studied. They were given IFN-gamma treatment, 100 MU/day i.m. for the first three months and 100 MU qod i.m. for the next six months. Semi-quantitative pathological scores of inflammation and fibrosis and serum hepatic fibrosis indices were compared within the 9 months. RESULTS: In animal experiment, the pathological fibrosis scores and liver hydroxyl proline content were found to be significantly lower in rats treated with different doses of IFN-gamma as compared with rats in fibrotic model group induced by either CCl4 or DMN, in a dose-dependent manner. For CCl4-induced model, pathological fibrosis scores in high, medium and low doses IFN-gamma groups were 5.10 +/- 2.88, 7.70 +/- 3.53 and 8.00 +/- 3.30, respectively, but the score was 14.60 +/- 7.82 in fibrotic model group. Hydroxyl proline contents were 2.83 +/- 1.18, 3.59 +/- 1.22 and 4.80 +/- 1.62, in the three IFN-gamma groups, and 10.01 +/- 3.23 in fibrotic model group. The difference was statistically significant (P【0.01). Similar results were found in DMN-induced model. Pathological fibrosis scores were 6.30 +/- 0.48, 8.10 +/- 2.72 and 8.30 +/- 2.58, in high, medium and low doses IFN-gamma groups, and 12.60 +/- 3.57 in fibrotic model group. Hydroxyl proline contents were 2.72 +/- 0.58, 3.14 +/- 0.71 and 3.62 +/- 1.02, in the three IFN-gamma groups, and 12.79 +/- 1.54 in fibrotic model group. The difference was statistically significant (P【0.01).Serum hepatic fibrosis indices decreased significantly in the 47 patients after IFN-gamma treatment (HA: 433.38 +/- 373.00 vs 281.57 +/- 220.48; LN: 161.22 +/- 41.02 vs 146 +/- 35 +/- 44. 67; PC III: 192.59 +/- 89.95 vs 156.98 +/- 49.22; C-I: 156.30 +/- 44.01 vs 139.14 +/- 34.47) and the differences between the four indices were significant (P 【0.05). Thirty-three patients received two liver biopsies, one before and one after IFN-gamma treatment. In thirty of 33 patients IFN-gamma had better effects according to semi-quantitative pathological scores (8.40 +/- 5.83 vs 5.30 +/- 4.05, P【0.05). CONCLUSION: All the three doses of IFN-gamma are effective in treating rat liver fibrosis induced by either CCl4 or DMN, the higher the dose, the better the effect. And IFN-gamma is effective for patients with moderate chronic hepatitis B viral fibrosis.展开更多

The liver functions and serological fibrotic markers were observed before and after treatment, 12 cases in the treated group were examined with liver biopsy.RESULTS Three hundreds nineteen recipe could remarkably decreased serum ALT level and total bilirubin and significantly improve serum albumin and A/G ratio. Its effects were better than Dahuang Zhachong pill. Before treatment, patients' serum monamine oxidase activities, tissue inhibitor of metalloproteinase (TIMP)-1, procollagen type Ⅲ and laminin were all higher than those of health peoples. These levels decreased remarkably after treatment, and urine hydroxyproline level increased significantly (P<0.001-0.05). Compared with the control, the improvement in treated group was better than that in the control except TIMP-1. According to the scoring system for staging of chronic hepatitis, the fibrotic extents of 7 cases among 12 cases examined by liver biopsy decreased remarkably (1 case decreased by 3 scores, 5 by 2 scores, 1 by 1 score).CONCLUSION Fuzheng Huayu 319 recipe had good therapeutic effects on chronic hepatitis B, it could reverse the development of liver fibrosis to some extent. In general its effects were better than that of Dahuang Zhachong pill.展开更多

A II degree scald model was established in rats, which was subsequently treated with chitosan and carboxymethyl chitosan solution, respectively. The concentration of bradykinin and 5-hydroxytryptophan was detected by assaying enzyme-linked immunosorbent. Healing condition was observed and pathological sections were made to determine the healing effect of chitosan and carboxymethyl chitosan. Results showed that the concentration of bradykinin and 5-hydroxytryptophan peaked at the third hour post-wound in all groups, while the concentration of hydroxyproline peaked at the seventh day post-wound in both chitosan and carboxymethyl chitosan group. The concentration of bradykinin and 5-hydroxytryptophan of carboxymethyl chitosan group was significantly lower than that of control(P < 0.05), while that of chitosan group was similar to that of control(P > 0.05). These findings indicated that carboxymethyl chitosan reduced the concentration of algogenic substances, resulting in analgesia. During the whole recovery process, the hydroxyproline concentration in chitosan and carboxymethyl chitosan group on day 3 and 7 was significantly higher than that of control(P < 0.01); however the significance of such a highness decreased on day 14(P < 0.05). These findings indicated that chitosan and carboxymethyl chitosan accelerated tissue repair. Meanwhile, chitosan performed better in healing than carboxymethyl chitosan in both decrustation and healing time. In conclusion, carboxymethyl chitosan showed significant analgesis and wound-healing promotion effect, but chitosan only showed wound-healing promotion effect.展开更多

Dan-Shao-Hua-Xian ( DSHX) cap sule, a traditional Chinese herbal compound, has shown marked preventive effects on hepatic fibrosis in rats in our previous studies. The present study was designed to further investigate its therapeutic actions on hepatic fibrosis in rats and its possible mechanisms. METHODS: Eighty male Wistar rats were randomly divid- ed into normal control group, hepatic fibrosis group, non DSHX-treated group, low-dose-treated group, and high dose-treated group. The rat models of hepatic fibrosis were established by subcutaneous injecton of CC14 , drinking al- cohol , giving diet of hyperliprosis and hypoprotein for 8 weeks. The two DSHX-treated groups were treated respec- tively with low dose (0.5 g/kg) and high dose (1.0 g/kg) of DSHX capsule p. o. everyday for 8 weeks. At the end of the experiment, liver indexes were calculated in each group in addition to the levels of the serum hyaluronic acid and alanine aminotransferase. Their degree of hepatic fibrosis and urinary excretion of hydroxyproline and expression of collagen , were detected. RESULTS: Comparison of the indexes of the hepatic fibro- sis group and non-DSHX-treated group revealed that the liver indexes, levels of serum hyaluronic acid and alanine aminotransferase, and stage of hepatic fibrosis were all sig- nificantly reduced in the two DSHX treated groups. The urinary excretion of hydroxyproline was increased and the expression of collagen I and HI in liver tissue was lessened. These alterations were more obviously observed in the high-dose-treated group. CONCLUSION: DSHX capsule has certain therapeutic effect on hepatic fibrosis in rats.展开更多
Objective:To investigate the antifibrotic effects of Chrysanthemum indicum ethanol extract(CIEE)against activated hepatic stellate cells(HSC)and thioacetamide(TAA)-induced hepatofibrosis in rats.Methods:Cell viability...Objective:To investigate the antifibrotic effects of Chrysanthemum indicum ethanol extract(CIEE)against activated hepatic stellate cells(HSC)and thioacetamide(TAA)-induced hepatofibrosis in rats.Methods:Cell viability and proliferation of HSC-T6 cells were measured using MTT assay.Primary HSCs were used to study morphology.TAA(200 mg/kg)was used to induced hepatic fibrosis in rats.CIEE(100 and 500 mg/kg)and silymarin(50 mg/kg)were administered orally.Liver functions including alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels were measured using commercial kits.Liver sections and fibrotic biomarker expression were measured using hematoxylin and eosin staining and real-time polymerase chain reaction.Results:In vitro study revealed that CIEE(0.1,0.25,and 0.5 mg/mL)inhibited the proliferation of activated HSCs exposed to transforming growth factor(TGF)-β and restored the activated primary HSC morphology.In in vivo studies,TAA-induced increase in liver/body weight ratio(5.46±0.26)was significantly reduced(4.13±0.22)by CIEE(P<0.05 at 500 mg/kg).CIEE(100 and 500 mg/kg)improved the liver functions by significantly attenuating changes in alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels(P<0.05).Further,CIEE(100 and 500 mg/kg)ameliorated the histological changes in liver tissue and TGF-β expression significantly(P<0.05)in TAA-induced rats.Conclusions:CIEE significantly protects against TAA-induced liver damage in rats and can be used in the treatment of liver fibrosis.展开更多

The control group (n = 10) had an end- to-end ...AIM: To investigate the influence of remote ischemic preconditioning (RIPC) on anastomotic integrity. METHODS: Sixty male Wistar rats were randomized to six groups. The control group (n = 10) had an end- to-end ileal anastomosis without RIPC. The preconditioned groups (n = 34) varied in time of ischemia and time of reperfusion. One group received the amino acid L-arginine before constructing the anastomosis (n = 9). On postoperative day 4, the rats were re-laparotomized, and bursting pressure, hydroxyproline concentration, intra-abdominal adhesions, and a histological score concerning the mucosat ischemic injury were collected. The data are given as median (range).RESULTS: On postoperative day 4, median bursting pressure was 124 mmHg (60-146 mmHg) in the control group. The experimental groups did not show a statistically significant difference (P 〉 0.05). Regarding the hydroxyproline concentration, we did not find any significant variation in the experimental groups. We detected significantly less mucosal injury in the RIPC groups. Furthermore, we assessed more extensive intra-abdominal adhesions in the preconditioned groups than in the control group. CONCLUSION: RIPC directly before performing small bowel anastomosis does not affect anastomotic stability in the early period, as seen in ischemic preconditioning.展开更多
文摘Fifteen cases of fresh fractures have been studied, including 14 cases of tubu-lar bone fractures with age from 28 to 60 years and one case of vertebral fracture ofdorsolumbar region, 16 years of age. There were 13 male and 2 female patients. Theyhad no endocrine disturbances or other bone diseases. The ratio ofhydroxyproline/creatinine (HP/Cr) in these cases increased gradually within 20-30 daysafter fracture and then reached the peak value which was about 3-4 times of normal, thepeak value maintained for a long time. If the trauma was mild or the patient was young,the peak began to decline on the 60th day after fracture. While in those severe cases withprolonged confinement to bed, the peak only began to come down about 110th dayafter fracture. The ratio of HP/Cr decreased rapidly from the peak at first, then pursueda slow course. Generally, the ratio of HP/Cr returned to normal on the 240th to 310thday. Bone union began at a time when peak value started to decline, and with completionof the remodelling process, the HP/Cr value returned to normal.

文摘This study was conducted to investigate the effect of dietary proline(Pro), and Pro and hydroxyproline(Hyp) in combination on the growth performance, total Hyp and collagen concentrations of tissues, and prolyl 4-hydroxylase α(I)(P4H α(I)) gene expression in juvenile turbot feeding high plant protein diets. A diet containing 50% crude protein and 12% crude lipid was formulated as the basal and control, on which other two protein and lipid contents identical experimental diets were formulated by supplementing the basal with either 0.75% Pro(Pro-0.75) or 0.75% Pro and 0.75% Hyp(Pro+Hyp). Four groups of fish in indoor seawater recirculating systems, 35 individuals each, were fed twice a day to apparent satiation for 10 weeks. The results showed that dietary Pro and Hyp supplementation had no significant effect on growth performance and feed utilization of juvenile turbot(P > 0.05). Total Hyp and collagen concentrations in muscle were significantly increased when dietary Pro and Hyp increased(P < 0.05), and fish fed diet Pro+Hyp showed significantly higher free Hyp content in plasma than those fed other diets(P < 0.05). The expression of P4 H α(I) gene in liver and muscle was significantly up regulated in fish fed diet Pro-0.75 in comparison with control(P < 0.05); however the gene was significantly down regulated in fish fed diet Pro+Hyp in muscle in comparison with fish fed diet Pro-0.75(P < 0.05). It can be concluded that supplement of crystal L-Pro and L-Hyp to high plant protein diets did not show positive effects on growth performance of juvenile turbot, but enhanced total collagen concentrations in muscle.
文摘An efficient protocol for plant regeneration from protoplasts of hydroxyproline(HYP)resistant cell line of Onoblychis viciaefolia was established. In SH medium supplemented with 1 mg/L 2, 4-dichlorophenoxy-acetic acid (2,4-D), 0.5 mg/L kinetin (KT) and 0.2 mg/L naphthalene acetic acid (NAA), the division frequency of protoplastderived cells reached uP to over 60 %, and microcalli were obtained in 5-6 wk. Upon transferring them on agar solidified MS medium plus 2 mg/L indole-3-acetic acid (IAA), shoots were induced. After cultivating them on MS medium with or without IAA, roots were regenerated.Chromosome number of all protoplast-regenerated plants examined were normal (2n=28). The protoplast-derived calli and plants grew vigorously on the medium containing 10 mmol/L HYP.

文摘A total of 16 Friesian cows were used in the study to determine the peripheral concentration of hydroxyproline(HYPRO) and 3 methyl histidine(3 MEHIS).The objectives of the experiment were to find out the relationship between collagen and myosin degradation and uterine involution in the postpartum cow.The results showed tht the mean concentrations of HYPRO and 3 MEHIS were 13.47±2.38 and 18.35±2.77 n mole/mL,respectively,for the cows during the days 60~90 postpartum.And for the cows immediately after calving the concentrations of HYPRO and 3 HEHIS reduced from 34.29 and 28.06 nmole/mL on the day one after calving to the normal lever of 13.23 and 17.97 n mole on the day 50 postpartum,respectively.There was a good correlation between the peripheral concentrations of both HYPRO and 3 MEHIS and the diameters of the involuting uterus(r=0.8128).

文摘The urinary concentrations of hydroxyproline (HYPRO)and 3 -methyl histidine (3 - MEHIS) were de- termined in 16 Chinese-Holstein cows. The objectives of the experiment were to find out thhe relationship between collagen and myosin ddegradation and uterine involution in the postpartum cow. The results in the experiment showed that the mean concentrations of HYPRO and 3-MEHIS were 138.32±22.99 and 37.09 ±3.90 nmol·mL-1,respectively,for the cows during the days between 60-90 postpartum,and for the cows immediately after calving the concentrations of HYPRO and 3 -MEHIS reduced from 284.30 and 65.48 nmol ·mL-1 on the day one after calving to the normal level of 109.18 and 33.51 nmol·mL^-1 on the day 50 post- partum,respectively. There was a good correlation between the urinary concentrations of both HYPRO and 3 -MEHIS and the diameters of the involuting uterus (r = 0. 79).
文摘Vitamin C is a water soluble vitamin found in many natural sources, citric fruits, vegetables, particularly as antioxidants and as a factor to catabolism and metabolism. The aim of this article is to correlate the level of vitamin C and the hydroxyproline level in urine samples among young palestinian children from age 5 - 14 years. Materials and Methods: Urine samples of 34 individuals of both sexes, their ages ranged from 5 - 14 years were collected and analyzed for determination of Urinary Hydroxyproline by using Modified Neuman and Logan Method. The result of this study shows a significant correlation between Vitamin C and Hydroxyproline at level 0.01. The conclusion of this study was that low vitamin C intake was found among young palestinian children from age 5 - 14 years, Vitamin C should be supplemented in the drinks taken by young palestinian children from age 5 - 14 years as well as further research and investigation with large samples required to include all children from middle and southern Palestine.

文摘This study investigated the effects of hydroxyproline(Hyp)and vitamin C(VC)addition to practical diet on growth and flesh quality of Pacific white shrimp(Litopenaeus vannamei)cultured in low salinity water.The control diet was formulated to contain 200 g/kg fish meal(FM20),and then 14 g/kg coated hydroxyproline(50%)(FM20H),2 g/kg L-ascorbyl-2-phosphate(38%)(FM20C)and their combination(FM20HC)were added to the control diet to form four isoproteic(41.6%)and isolipidic(6.7%)diets.Pacific white shrimp with initial body weight of(1.50±0.05)g were fed the above four diets in low salinity water(salinity 1‰)for 8 weeks.The results showed that the combined supplementation of Hyp and VC(FM20HC)increased the weight gain by 13.4%,and decreased the feed conversion ratio by 0.18(P<0.05),while the individual supplementation(FM20H and FM20C groups)just numerically promoted the growth(P>0.05).In addition,the FM20HC group also showed higher protein and lipid retention than the FM20 group(P<0.05).In flesh quality,the supple-mentation of Hyp(FM20H)increased total collagen and heat-soluble collagen contents,flesh chewiness and free flavor amino acids contents(P<0.05).The supplementation of vC(FM20C)increased the contents of total collagen,heat-insoluble collagen and free flavor amino acids,and decreased flesh lactic acid,malondialdehyde contents and steaming loss(P<0.05).The combined supplementation of Hyp and VC(FM20HC)showed higher total collagen and heat-insoluble collagen contents than the individual supplementation(FM20H and FM20C)(P<0.05),and the myofiber density,flesh hardness and shear force in the FM20HC group were also significantly higher than those in the control(P<0.05).In summary,the combined supplementation of hydroxyproline and vc in practical diet with fishmeal content of 200 g/kg significantly improved the growth performance and flesh quality of L.vannamei cultured in low salinity water.

文摘Hardness is widely regarded as a critical factor influencing the whole texture of fish flesh.The objective of this study was to elucidate the regulatory mechanism underlying muscle hardness in hybrid bream(BBTB,Megalobrama amblycephala♀×Culter alburnus♂).A comparison of the physiological features of high hardness(HH)and low hardness(LH)muscle revealed that the former had higher contents of collagen I and lower muscle fiber diameter.Transcriptomic data revealed that the myofiber assembly pathway and the HIF-1 signaling pathway were activated in HH muscle.At the metabolic level,the categories of amino acids and lipids were the principal differentially abundant metabolites between the HH and LH muscle.The detection of amino acid profiles further revealed significant differences in amino acid metabolism between the HH and LH muscles,with the HH muscle having higher levels of amino acids than the LH muscle,especially hydroxypro-line(Hyp).Furthermore,through supplementation of Hyp in BBTB myoblasts,the results indicated that 0.8 mmol/L Hyp increased the proliferation,differentiation,migration,and collagen synthesis of myoblasts.Finally,BBTB was treated with Hyp intraperitoneally for 15 days.The results revealed that 0.1 g/kg Hyp significantly increased muscle hardness,myofiber density,myofibrillar protein synthesis,and HIF-1 protein content.The results obtained in this study indicated that Hyp supplementation promoted collagen synthesis and proliferation of myoblast and muscle fibers in the BBTB,which may be induced by activation of the transcription factor HIF1 and contributes to the impacts of Hyp on improvements in muscle hardness in the BBTB.

文摘This experiment aimed to evaluate the impact of dietary hydroxyproline(Hyp)supplementation on the muscle quality of juvenile Pacific white shrimp(Litopenaeus vannamei)fed a low fishmeal diet.Six formulated diets included one high fishmeal(HF;25%fishmeal content)and five low fishmeal diets(10%fishmeal content)with 0%,0.2%,0.4%,0.6%and 0.8%Hyp(LFO,LF2,LF4,LF6 and LF8,respectively).Each diet was assigned to four replicates,and 40 shrimp(0.32±0.00 g)per replicate were fed four times a day for 8 weeks.Dietary Hyp supplementation had little effects on growth performance,but increased the contents of Hyp,prolyl 4-hydroxylases(P4Hs),and collagen.The meat yield,springiness,hardness,chewiness,and cohesiveness of muscle were the highest in the LF4 group among the low fishmeal groups(P<0.05).Cooking loss and freezing loss of muscle were the lowest in the LF4 group(P<0.05).Dietary supplementation with 0.4%Hyp increased the myofiber density and decreased the myofiber diameter of muscle(P<0.05).Supplementation of Hyp in the diet up-regulated the mRNA expression of smyhc5,smyhc15,col1a1,col1a2,igf-1f,tgf-β and tor and down-regulated the mRNA expression of smy hc1,smyhc2,smyhc6a(P<0.05).Supplementation of Hyp in the diet up-regulated the protein expression of P-4E-BP1,P-AKT,AKT and P-AKT/AKT(P<0.05).These results suggested that the addition of 0.4%Hyp to low fishmeal diets improved the muscle quality of L.vannamei.
文摘Purpose The purpose of this study is to review the urine products of bone breakdown as markers of bone resorption and usefulness of urinary hydroxyproline.Data Related researches published in 1985 -2000 were systematically reviewed.Results Bone markers could be used for early diagnosis of bone metabolic diseases. Biochemical markers of bone resorption that reflect osteoclast activity and/or collagen degradation provide a new and potentially important clinical tool for the assessment and monitoring of bone metabolism. Assessment of bone resorption can be achieved with measurement of urinary hydroxylysine glycosides, urinary excretion of the collagen pyridinium cross-links, urinary excretion of type I collagen telopeptide breakdown products (cross-linked telopeptides) and urinary hydroxyproline.Conclusion Urinary hydroxyproline has been in use as a marker of bone resorption, but it lacks sensitivity and specificity. It is a modified aminoacid that is a metabolic product of collagen breakdown. Hydroxyproline may be released either free or with fragments of the collagen molecule attached during bone resorption, and it is also liberated by the breakdown of complement and nonskeletal collagen.

文摘Oxidative stress has been confirmed in relation to intestinal mucosa damage and multiple bowel diseases.Hydroxyproline (Hyp) is an imino acid abundant in sow's milk. Compelling evidence has beengathered showing the potential antioxidative properties of Hyp. However, the role and mechanism ofHyp in porcine intestinal epithelial cells in response to oxidative stress remains unknown. In this study,small intestinal epithelial cell lines of piglets (IPEC-1) were used to evaluate the protective effects of Hypon 4-hydroxy-2-nonenal (4-HNE)-induced oxidative DNA damage and apoptosis. IPEC-1 pretreated with0.5 to 5 mmol/L Hyp were exposed to 4-HNE (40 mmol/L) in the presence or absence of Hyp. Thereafter,the cells were subjected to apoptosis detection by Hoechst staining, flow cytometry, and Western blot orDNA damage analysis by comet assay, immunofluorescence, and reverse-transcription quantitative PCR(RT-qPCR). Cell apoptosis and the upregulation of cleaved-caspase-3 induced by 4-HNE (40 mmol/L) wereinhibited by 5 mmol/L of Hyp. In addition, 5 mmol/L Hyp attenuated 4-HNE-induced reactive oxygenspecies (ROS) accumulation, glutathione (GSH) deprivation and DNA damage. The elevation in transcriptionof GADD45a (growth arrest and DNA-damage-inducible protein 45 alpha) and GADD45b(growth arrest and DNA-damage-inducible protein 45 beta), as well as the phosphorylation of H2AX(H2A histone family, member X), p38 MAPK (mitogen-activated protein kinase), and JNK (c-Jun N-terminalkinase) in cells treated with 4-HNE were alleviated by 5 mmol/L Hyp. Furthermore, Hyp supplementationincreased the protein abundance of Krüppel like factor 4 (KLF4) in cells exposed to 4-HNE.Suppression of KLF4 expression by kenpaulone impeded the resistance of Hyp-treated cells to DNAdamage and apoptosis induced by 4-HNE. Collectively, our results indicated that Hyp serves to protectagainst 4-HNE-induced apoptosis and DNA damage in IPEC-1 cells, which is partially pertinent with theenhanced expression of KLF4. Our data provides an updated explanation for the nutritional values ofHyp-containing animal products.
文摘BACKGROUND Growth hormone(GH)plays a crucial role in wound healing and tissue repair in postoperative patients.In particular,colonic anastomosis healing following colorectal surgery is impaired by numerous chemotherapy agents.AIM To investigate whether GH can improve the healing of a colonic anastomosis following the adverse effects of intraperitoneal administration of 5-fluorouracil(5-FU),bleomycin and cisplatin.METHODS Eighty Wistar rats underwent laparotomy and a 1 cm-resection of the transverse colon,followed by an end-to-end anastomosis under general anesthesia.The rats were blindly allocated into four equal groups and administered a different daily intraperitoneal therapeutic regimen for 6 days.The control group(A)received normal saline.Group B received chemotherapy with 5-FU(20 mg/kg),bleomycin(4 mg/kg)and cisplatin(0.7 mg/kg).Group C received GH(2 mg/kg),and group D received the aforementioned combination chemotherapy and GH,as described.The rats were sacrificed on the 7th postoperative day and the anastomoses were macroscopically and microscop-ically examined.Body weight,bursting pressure,hydroxyproline levels and inflammation markers were measured.RESULTS All rats survived until the day of sacrifice,with no infections or other complications.A decrease in the body weight of group D rats was observed,not statistically significant compared to group A(P=1),but significantly different to groups C(P=0.001)and B(P<0.01).Anastomotic dehiscence rate was not statistically different between the groups.Bursting pressure was not significantly different between groups A and D(P=1.0),whereas group B had a significantly lower bursting pressure compared to group D(P<0.001).All groups had significantly more adhesions than group A.Hydroxyproline,as a measurement of collagen deposition,was significantly higher in group D compared to group B(P<0.05),and higher,but not statistically significant,compared to group A.Significant changes in group D were recorded,compared to group A regarding inflammation(3.450 vs 2.900,P=0.016)and fibroblast activity(2.75 vs 3.25,P=0.021).Neoangiogenesis and collagen deposition were not signifi-cantly different between groups A and D.Collagen deposition was significantly increased in group D compared to group B(P<0.001).CONCLUSION Intraperitoneal administration of chemotherapy has an adverse effect on the healing process of colonic anastomosis.However,GH can inhibit the deleterious effect of administered chemotherapy agents and induce colonic healing in rats.
文摘This study aims to explore the molecular aspects of the wound-healing potential of medicinal plants through preclinical and clinical research.This review focuses on the theoretical sup-port and therapeutic effects of traditional herbal plants,herbal formulations,and active com-pounds in herbal medicine.It provides new insights into the management of diabetic wound healing with herbal medicine.A comprehensive literature review was conducted using key-words such as“herbal remedies”“diabetics”“wounds”“bioactive”“medicinal plant”and“growth factor”across several literature databases,namely,Wiley Online Library,Elsevier,Springer,PubMed,and Google Scholar.The available literature provides a basis for tradition-al remedies found to be effective in healing wounds by targeting key molecules involved in wound pathology,including collagen I&III,vascular endothelial growth factor(VEGF),tu-mor necrosis factor-alpha(TNF-α),nuclear factor kappa B(NF-κB),transforming growth fac-tor(TGF)-β1,hydroxyproline(collagen component),superoxide dismutase(SOD),and Cata-lase(CAT).Numerous studies have investigated the presence of bioactive compounds in many plants for their wound-healing effects.The results underscore the potential of various plants in wound healing,highlighting the need for further pharmacological research before clinical application.These findings support the conventional use of herbal remedies and pro-vide the basis for future research into the development of new therapeutic alternatives for wound healing.
文摘AIM: To evaluate the antifibrotic effect of different doses of recombinant human Gamma-Interferon (IFN-gamma) in two rat models of hepatic fibrosis, and to observe its effect on moderate chronic hepatitis B virus fibrosis. METHODS: Hepatic fibrosis was successfully induced in 150 and 196 rats by subcutaneous injection of carbon tetrachloride (CCl4) and intraperitoneal injection of dimethylnitrosamine (DMN), respectively. Each of the two model groups was divided into: (1) fibrotic model group; (2) colchicine treatment group (0.1 mg/kg/day, gastrogavage for 8 weeks); (3) high-dose IFN-gamma group (15 MU/kg per day, i.m. for 8 weeks); (4) medium-dose IFN-gamma group (5 MU/kg daily, i.m. for 8 weeks); and (5) Y low-dose IFN-gamma group (1.67 MU/kg daily, i.m. for 8 weeks). Another group of 10 rats without any treatment was used as normal controls. At the end of the experiment, semi-quantitative histopathological scores of inflammation and fibrosis, liver alpha smooth muscle actin (alpha-SMA) expression level, liver hydroxyl proline content and serum hyaluronic acid levels were compared. And 47 medium chronic hepatitis B viral fibrosis patients were studied. They were given IFN-gamma treatment, 100 MU/day i.m. for the first three months and 100 MU qod i.m. for the next six months. Semi-quantitative pathological scores of inflammation and fibrosis and serum hepatic fibrosis indices were compared within the 9 months. RESULTS: In animal experiment, the pathological fibrosis scores and liver hydroxyl proline content were found to be significantly lower in rats treated with different doses of IFN-gamma as compared with rats in fibrotic model group induced by either CCl4 or DMN, in a dose-dependent manner. For CCl4-induced model, pathological fibrosis scores in high, medium and low doses IFN-gamma groups were 5.10 +/- 2.88, 7.70 +/- 3.53 and 8.00 +/- 3.30, respectively, but the score was 14.60 +/- 7.82 in fibrotic model group. Hydroxyl proline contents were 2.83 +/- 1.18, 3.59 +/- 1.22 and 4.80 +/- 1.62, in the three IFN-gamma groups, and 10.01 +/- 3.23 in fibrotic model group. The difference was statistically significant (P【0.01). Similar results were found in DMN-induced model. Pathological fibrosis scores were 6.30 +/- 0.48, 8.10 +/- 2.72 and 8.30 +/- 2.58, in high, medium and low doses IFN-gamma groups, and 12.60 +/- 3.57 in fibrotic model group. Hydroxyl proline contents were 2.72 +/- 0.58, 3.14 +/- 0.71 and 3.62 +/- 1.02, in the three IFN-gamma groups, and 12.79 +/- 1.54 in fibrotic model group. The difference was statistically significant (P【0.01).Serum hepatic fibrosis indices decreased significantly in the 47 patients after IFN-gamma treatment (HA: 433.38 +/- 373.00 vs 281.57 +/- 220.48; LN: 161.22 +/- 41.02 vs 146 +/- 35 +/- 44. 67; PC III: 192.59 +/- 89.95 vs 156.98 +/- 49.22; C-I: 156.30 +/- 44.01 vs 139.14 +/- 34.47) and the differences between the four indices were significant (P 【0.05). Thirty-three patients received two liver biopsies, one before and one after IFN-gamma treatment. In thirty of 33 patients IFN-gamma had better effects according to semi-quantitative pathological scores (8.40 +/- 5.83 vs 5.30 +/- 4.05, P【0.05). CONCLUSION: All the three doses of IFN-gamma are effective in treating rat liver fibrosis induced by either CCl4 or DMN, the higher the dose, the better the effect. And IFN-gamma is effective for patients with moderate chronic hepatitis B viral fibrosis.
文摘AIM To investigate clinic effects of Fuzheng Huayu 319 recipe (319 recipe) on liver fibrosis in chronic hepatitis B.METHODS Ninety-five patients with chronic hepatitis B were divide into the treated (63 cases) and control (32 cases) group, and orally administrated with 0.5g 319 capsule or 0.5g Dahuang Zhachong pill tid for 3 months, respectively. The liver functions and serological fibrotic markers were observed before and after treatment, 12 cases in the treated group were examined with liver biopsy.RESULTS Three hundreds nineteen recipe could remarkably decreased serum ALT level and total bilirubin and significantly improve serum albumin and A/G ratio. Its effects were better than Dahuang Zhachong pill. Before treatment, patients' serum monamine oxidase activities, tissue inhibitor of metalloproteinase (TIMP)-1, procollagen type Ⅲ and laminin were all higher than those of health peoples. These levels decreased remarkably after treatment, and urine hydroxyproline level increased significantly (P<0.001-0.05). Compared with the control, the improvement in treated group was better than that in the control except TIMP-1. According to the scoring system for staging of chronic hepatitis, the fibrotic extents of 7 cases among 12 cases examined by liver biopsy decreased remarkably (1 case decreased by 3 scores, 5 by 2 scores, 1 by 1 score).CONCLUSION Fuzheng Huayu 319 recipe had good therapeutic effects on chronic hepatitis B, it could reverse the development of liver fibrosis to some extent. In general its effects were better than that of Dahuang Zhachong pill.

文摘Analgesis and wound healing effect of chitosan and carboxymethyl chitosan on scalded rats were investigated. A II degree scald model was established in rats, which was subsequently treated with chitosan and carboxymethyl chitosan solution, respectively. The concentration of bradykinin and 5-hydroxytryptophan was detected by assaying enzyme-linked immunosorbent. Healing condition was observed and pathological sections were made to determine the healing effect of chitosan and carboxymethyl chitosan. Results showed that the concentration of bradykinin and 5-hydroxytryptophan peaked at the third hour post-wound in all groups, while the concentration of hydroxyproline peaked at the seventh day post-wound in both chitosan and carboxymethyl chitosan group. The concentration of bradykinin and 5-hydroxytryptophan of carboxymethyl chitosan group was significantly lower than that of control(P < 0.05), while that of chitosan group was similar to that of control(P > 0.05). These findings indicated that carboxymethyl chitosan reduced the concentration of algogenic substances, resulting in analgesia. During the whole recovery process, the hydroxyproline concentration in chitosan and carboxymethyl chitosan group on day 3 and 7 was significantly higher than that of control(P < 0.01); however the significance of such a highness decreased on day 14(P < 0.05). These findings indicated that chitosan and carboxymethyl chitosan accelerated tissue repair. Meanwhile, chitosan performed better in healing than carboxymethyl chitosan in both decrustation and healing time. In conclusion, carboxymethyl chitosan showed significant analgesis and wound-healing promotion effect, but chitosan only showed wound-healing promotion effect.
文摘BACKGROUND: Hepatic fibrosis is the common patholo gical change in various chronic liver diseases,and its major cause is the imbalance between the production and degra dation of the extracellular matrix, which is mainly com posed of collagens. Dan-Shao-Hua-Xian ( DSHX) cap sule, a traditional Chinese herbal compound, has shown marked preventive effects on hepatic fibrosis in rats in our previous studies. The present study was designed to further investigate its therapeutic actions on hepatic fibrosis in rats and its possible mechanisms. METHODS: Eighty male Wistar rats were randomly divid- ed into normal control group, hepatic fibrosis group, non DSHX-treated group, low-dose-treated group, and high dose-treated group. The rat models of hepatic fibrosis were established by subcutaneous injecton of CC14 , drinking al- cohol , giving diet of hyperliprosis and hypoprotein for 8 weeks. The two DSHX-treated groups were treated respec- tively with low dose (0.5 g/kg) and high dose (1.0 g/kg) of DSHX capsule p. o. everyday for 8 weeks. At the end of the experiment, liver indexes were calculated in each group in addition to the levels of the serum hyaluronic acid and alanine aminotransferase. Their degree of hepatic fibrosis and urinary excretion of hydroxyproline and expression of collagen , were detected. RESULTS: Comparison of the indexes of the hepatic fibro- sis group and non-DSHX-treated group revealed that the liver indexes, levels of serum hyaluronic acid and alanine aminotransferase, and stage of hepatic fibrosis were all sig- nificantly reduced in the two DSHX treated groups. The urinary excretion of hydroxyproline was increased and the expression of collagen I and HI in liver tissue was lessened. These alterations were more obviously observed in the high-dose-treated group. CONCLUSION: DSHX capsule has certain therapeutic effect on hepatic fibrosis in rats.
文摘Objective:To investigate the antifibrotic effects of Chrysanthemum indicum ethanol extract(CIEE)against activated hepatic stellate cells(HSC)and thioacetamide(TAA)-induced hepatofibrosis in rats.Methods:Cell viability and proliferation of HSC-T6 cells were measured using MTT assay.Primary HSCs were used to study morphology.TAA(200 mg/kg)was used to induced hepatic fibrosis in rats.CIEE(100 and 500 mg/kg)and silymarin(50 mg/kg)were administered orally.Liver functions including alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels were measured using commercial kits.Liver sections and fibrotic biomarker expression were measured using hematoxylin and eosin staining and real-time polymerase chain reaction.Results:In vitro study revealed that CIEE(0.1,0.25,and 0.5 mg/mL)inhibited the proliferation of activated HSCs exposed to transforming growth factor(TGF)-β and restored the activated primary HSC morphology.In in vivo studies,TAA-induced increase in liver/body weight ratio(5.46±0.26)was significantly reduced(4.13±0.22)by CIEE(P<0.05 at 500 mg/kg).CIEE(100 and 500 mg/kg)improved the liver functions by significantly attenuating changes in alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels(P<0.05).Further,CIEE(100 and 500 mg/kg)ameliorated the histological changes in liver tissue and TGF-β expression significantly(P<0.05)in TAA-induced rats.Conclusions:CIEE significantly protects against TAA-induced liver damage in rats and can be used in the treatment of liver fibrosis.

文摘AIM: To investigate the influence of remote ischemic preconditioning (RIPC) on anastomotic integrity. METHODS: Sixty male Wistar rats were randomized to six groups. The control group (n = 10) had an end- to-end ileal anastomosis without RIPC. The preconditioned groups (n = 34) varied in time of ischemia and time of reperfusion. One group received the amino acid L-arginine before constructing the anastomosis (n = 9). On postoperative day 4, the rats were re-laparotomized, and bursting pressure, hydroxyproline concentration, intra-abdominal adhesions, and a histological score concerning the mucosat ischemic injury were collected. The data are given as median (range).RESULTS: On postoperative day 4, median bursting pressure was 124 mmHg (60-146 mmHg) in the control group. The experimental groups did not show a statistically significant difference (P 〉 0.05). Regarding the hydroxyproline concentration, we did not find any significant variation in the experimental groups. We detected significantly less mucosal injury in the RIPC groups. Furthermore, we assessed more extensive intra-abdominal adhesions in the preconditioned groups than in the control group. CONCLUSION: RIPC directly before performing small bowel anastomosis does not affect anastomotic stability in the early period, as seen in ischemic preconditioning.
