Alterations in hepatic metabolism of adult male rats following exposure to hydroxyprogesterone during embryonic development

Aim:To investigate the effect of in utero exposure to hydroxyprogesterone(HP)on liver metabolism in adult male albino rats.Methods:Pregnant Wistar strain albino rats were exposed to supra-normal levels(10 mg/kg and 25 mg/kg)of HP on days 1,7 and 14 of pregnancy.The male pups were maintained under controlled conditions and the rats were killed 90 days after birth.The liver tissue was immediately excised,weighed and used for biochemical assays. Results:The activity levels of succinate dehydrogenase(SDH),glutamate dehydrogenase(GDH),glucose-6-phosphate dehydrogenase(G-6-PDH),malate dehydrogenase(MDH)and aminotransaminases were significantly increased in the livers of rats exposed to HP during embryonic development.The lactate dehydrogenase(LDH)activity level was significantly decreased in the liver of experimental rats.Furthermore,there was a significant elevation of activity levels of antioxidant enzymes(glutathione S-transferase[GST] and catalase[CAT])with an increased lipid peroxidation in the hepatic tissue of experimental rats compared with the control group.Conclusion:The results of the present study suggest that there is an increase in the oxidative metabolism,antioxidative mechanism and levels of lipid peroxidation in rats exposed to HP during embryonic development.The increased aminotransaminase activities in these rats reveal tissue damage and disruption of mitochondrial integrity.(Asian J Androl 2006 Jul;8:463-467) 展开更多

Background:Preterm birth can cause morbidity and mortality in addition to trauma or loss to the mother,and present a major financial burden to the family.This risk may be minimized by timely intervention.Objective:To understand the safety and efficacy of hydroxyprogesterone in decreasing the incidence of preterm birth,neonatal complications,and maternal mortality,based on evidence obtained from the Indian scientific literature.Methods:Relevant articles were searched for and retrieved from the databases PubMed,medrXiv,Embase,Cochrane,biorXiv,and arXiv.Results:A total of 62 relevant articles were assessed,and of these,59 articles did not meet the eligibility criteria.In total,3 articles which satisfied the criteria and 2 articles sourced from cross references were included in the study.Across these 5 articles,a total of 536 patients received hydroxyprogesterone caproate or placebo for preterm birth.Conclusion:In all five articles,recurrence rate of preterm birth was reduced in the treated group compared to control group.The safety profile of progesterone tends to be better than that of the placebo.The findings of our analysis of individual patient data(IPD)summarizes various clinical trial data,which can assure clinicians that vaginal progesterone administration is effective and well tolerated. 展开更多

Preterm parturition is the consequence of pathological signals that activate the common pathway of parturition and considered as a syndrome.Many risk factors for spontaneous preterm birth(sPTB)have been identified.Two significant risk factors for sPTB are history of prior sPTB and short cervical length at midtrimester.17 hydroxyprogesterone caproate,vaginal progesterone,cerclage,and pessary have all been studied for prevention of sPTB.Difference in patient populations likely contributes to the conflicting study results.Further studies are needed to establish strategies in prevention of sPTB in singleton as well as multiple pregnancies. 展开更多

Hydroxyprogesterone Co. (EP) was injected i.m. into female Wistar rats, different strains of mice of both sex and Syrian golden hamsters with doses of 5--100 times the human contraceptive dose once or twice a month for 10--32 times. In some experiments EP was combined with whole-body 3 Gy gamma ray radiation once or twice. Results show that EP has obvious carcinogenicity and can enhance the gamma ray carcinogenicity, thereby increasing the tumor incidence or the ratio of malignant to benign turnouts. Carcinogenic mechanisms of EP have also been studied. 展开更多
