[ Metbod] By employing polyacrylamide gel electrophoresis method, the expressions of AMY and SOD isozymes during seed germination process were analyzed. ~ Result] The main AMY bands remained strong during the whole peri- od and a new band A2 appeared in the middle and late period of seed germination. Some new SOD bands occurred at the early stage, then be- came weak or disappeared in the middle period, and band S6 became intense in the late peried. [Method] Non-denaturing polyacrylamide gel electrophoresis was used to analyze isozyme patterns such as esterase (EST),acid phosphatase (ACP),ATP enzyme (ATPase),amylase (AMY),superoxide dismutase (SOD) and peroxidase (POD) in long-term subculture callus of Emmenopterys henryi Oliv. [Result] The research showed that there were differences among the 6 kinds of isozymes in embryogenic callus and non-embryogenic callus of Emmenopterys henryi Oliv.,and both levels could be taken as the basis for identification,the EST,ACP,and POD of non-embryogenic callus were significantly higher than embryogenic callus. The browning of non-embryogenic callus was non-level in the AMY,SOD and POD isozymes when was compared with normal non-embryogenic callus,while the EST,ACP and ATPase isozymes decreased; When the browning of embryogenic callus was contrasted with normal embryogenic callus,EST isozyme increased and the other 5 kinds of enzymes decreased. The herbal extract was separated by repeated column chromatography over silica gel and celite. The structures were elucidated by spectroscopic analysis. Thirteen compounds were obtained and identified as 7-methoxyl-aristololactam Ⅳ (1), aristololactam Ⅱ (2), aristolochic acid Ⅰ (3), aristololactam AⅡ (4), daucosterol (5), aristololactam Ⅰa (6), N-trans-feruloyl tyramine (7), aristololactam Ⅰ (8), 4β,10β-aromadendranediol (9), aristololide (10), aristolic acid Ⅰ (11), meso-dihydroguaiaretic acid (12), and calopiptin (13). These compounds were obtained from the genus Saruma for the first time, and they provided chemical evidences for the chemotaxonomy of plants of the Aristolochiaceae family. Since aristolochic acids and aristololactams are toxic to kidney, the results of this investigation suggest that it should be cautious to use Saruma henryi as a medicine.展开更多

METHOD: The hot methanol extract of the root bark of A. henryi was subjected to XAD-4 column chromatography eluting with a gradient of methanol in water. The cytotoxicity and anti-inflammatory effects of the MeOH fractions were evaluated on the inhibition on lipopolysaccharide(LPS)-induced nitric oxide, prostaglandin E2, interleukin-1β, and interleukin-6 production in RAW 264.7 macrophages. RESULTS: The 80% MeOH fraction was a better inhibitor of LPS-induced NO, PGE2, IL-1β, and IL-6 production, and expression of inducible nitric oxide synthase(iNOS) at the protein levels in a concentration-dependent manner. CONCLUSION: The 80% MeOH fraction of A. henryi root bark has significant anti-inflammatory activity. This provides a pharmacological basis for clinical application for the treatment of inflammation.展开更多

Four flavonoids were isolated from the extract of F. sarmentosa var. henryi by activity-guided fractionation and they were identified as eriodictyol (1), homoeriodictyol (2), dihydroquercetin (3), and luteolin (4) by spectroscopic means and displayed excellent inhibitory activity against F. graminearum and S. zeicola. Among them, luteolin (4) showed the strongest inhibitory activity with IC50 values of 56.38 and 81.48 mg L^-1, to the fungi, respectively. The in vitro inhibitory activity of F. sarmentosa var. henryi (King) Corner was reported for the first time and the inhibitory action mechanism of luteolin (4) was worthy of further research.展开更多
Chinquapin(Castanea henryi) is a dual-purpose tree species in China valued for as a source of timber and starch.We investigated the effect of four cutting mediums(pure vermiculite;peat:river sand at 3:1 v/v;peat:krasn...Chinquapin(Castanea henryi) is a dual-purpose tree species in China valued for as a source of timber and starch.We investigated the effect of four cutting mediums(pure vermiculite;peat:river sand at 3:1 v/v;peat:krasnozem at 1:1 v/v;and pure krasnozem) and three stem cutting periods(March,May,and July) on rooting performance of C.henryi cuttings.Different cutting periods and cutting mediums greatly influenced the rooting rate of C.henryi,ranging from 3.35 to 77.31%.Principal component analysis indicated that the best combination of cutting period and cutting medium was semi-hardwood cuttings(May cuttings)+krasnozem.Histological evidence indicated that adventitious root initials were present by week 5-6,and that the site of root primordia initiation was observed in the vascular cambium.Stem anatomical structures observed at different periods indicated that a xylem/radius ratio of29.90-37.42% and a fractured phloem fiber ring are indicative of rooting success.The relational model between rooting index and medium properties indicated that nutrient content and porosity significantly influenced callus production.However,pH strongly affected C.henryi root formation,with the Pearson correlation coefficients for May and July cuttings of-0.856 and-0.947,respectively.Our protocol is helpful to achieve mass clone propagation of improved C.henryi genotypes,thus overcoming a common hurdle in chinquapin breeding programs.展开更多

The chlor...Insecticidal activities of the petroleum ether-, chloroform-, ethyl acetate-, and water-soluble fractions of the methanolic extract ofFicus sarmentosa var. henryi were assayed against Musca domestica adults. The chloroform- and ethyl acetate- soluble fractions were the most active with 92.6 and 88.9% mortalities (24 h after treatment) respectively. Therefore, the two fractions were combined and four compounds, isolated from the fractions by activity-guided fractionation, were elucidated as 7-hydroxycoumarin, apigenin, eriodictyol, and quercetin by spectroscopic method and displayed excellent insecticidal activities against adults of M. domestica and 4th instar larva of Aedes albopictus. Among those, 7- hydroxycoumarin showed the strongest insecticidal activities with lethal concentrations (LC50) values of 72.13 μg g^-1 sugar and 4.87 μg mL^-1 (48 h after treatment) against the test insects respectively. The cytoxicities of these compounds on BTI-Tn-5B 1-4 cell were also investigated for the insecticidal mechanism and found that quercetin represented superior inhibitory activity with MTT assay and reactive oxygen species (ROS) against BTI-Tn-5B 1-4 cell, but slightly weaker than that of the positive control (azadirachtin) and significantly greater than the negative control (DMSO only). Meanwhile, eriodictyol demonstrated the strongest effect on the mitochondrial membrane potentials (MMP). In conclusion, based on their comparative toxicities to commercial insecticides and their cytotoxic effects, some of the compounds from the F. sarmentosa have potential as botanical insecticides.展开更多

RESULTS: Seven compounds were obtained and their structures were identified as 10-benzoyl-cycloparvifloralone(1), cycloparvifloralone(2), 2α-hydroxycycloparviforalone(3), henrylactone B(4), merrillianone(5), henrylactone C(6) and 7, 14-ortholactone-3-hydroxyfloridanolide(7). CONCLUSION: Compound 1 is a new sesquiterpene lactone.
