Eight sesquiterpenes were isolated from Ainsliaea glabra.Among them,a new guaianolide sesquiterpenoid,named ainsglaolide A(1),and seven known sesquiterpenoids 2~8 were identified.Compound 1 was a novel guaianolide sesquiterpenoid featuring a rare six-membered lactone ring formed between C-4 and C-15.The new compound structure was elucidated by HRESI-MS and NMR spectroscopy,as well as electrostatic circular dichroism(ECD)calculations were used to further confirm the absolute of 1.Anti-inflammatory activity tests were conducted on the separated compounds,indicating that compound 1 had significant inhibitory effect on NLRP3 inflammasome with an IC_(50) value of 1.86μmol/L. 展开更多

Mechanistic insights into Smilax Glabra and Semen Coicis for gout and hyperuricemia treatment:a network pharmacology and molecular docking approach

To elucidate the mechanisms underlying the therapeutic effects of the herbal medicine pair Smilax Glabra and Semen Coicis in treating gout and hyperuricemia,a comprehensive analysis was conducted using network pharmacology and molecular docking methods.Disease-associated targets for gout and hyperuricemia were identified from the GeneCards,OMIM,Disgenet,and TTD databases,while the key active components and their corresponding targets for Smilax Glabra and Semen Coicis were obtained from the TCSMP database.The intersection of these targets enabled the construction of a protein-protein interaction(PPI)network,which was subsequently visualized and analyzed.Core targets were further subjected to Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses to elucidate the biological processes and pathways involved.Molecular docking was then employed to validate the reliability of the interactions between the active components and the identified targets.The analysis revealed that Smilax Glabra and Semen Coicis contained 15 bioactive components that interacted with 393 potential targets,while gout and hyperuricemia were associated with 660 targets in total.The primary active compounds implicated in treating these conditions included diosgenin,quercetin,and naringenin,which were found to interact with crucial hub targets such as BCL2,CASP3,and MAPK3.These interactions suggested that the herbal medicine pair modulated several biological processes,including gland development and the regulation of body fluid levels,through pathways involving membrane rafts,membrane microdomains,and nuclear receptor activities.Enrichment analyses highlighted their involvement in multiple signaling pathways,such as EGFR tyrosine kinase inhibitor resistance,phospholipase D signaling,and platelet activation.Molecular docking confirmed the strong binding affinities between the hub genes and the major active components,supporting their potential role in therapeutic efficacy.This study demonstrated that Smilax Glabra and Semen Coicis might offer a promising therapeutic strategy for gout and hyperuricemia by targeting multiple molecular components,biological functions,and pathways.The findings underscored the unique potential of traditional Chinese medicine(TCM)in managing complex diseases by leveraging synergistic effects across diverse biological mechanisms. 展开更多

Based on the meteorological data of 24 automatic stations in Xinfeng County,combined with the climatic conditions for the growth of Morinda officinalis F.C.How and Sarcandra glabra(Thunb.)Nakai,the climate suitability zoning indicators of M.officinalis and S.glabra in Xinfeng County were constructed by relevant statistical methods,and the climate suitability zoning indicators were divided into four grades:most suitable,suitable,sub-suitable and unsuitable.According to the results of expert scores,the relevant growth impact factors were assigned weight values.Based on geographic information system(GIS),the climate suitability zoning of M.officinalis and S.glabra planting in Xinfeng County was carried out.The results show that most areas of Xinfeng County were suitable for planting M.officinalis and S.glabra.Among them,the most suitable planting areas of M.officinalis were mainly distributed in most of Huilong Town,from Jiangnan Community of Fengcheng Street to Yuntianhai area of Meikeng Town,Matouwantian Village and areas to the south,and areas to the south of Zhutong Village,Fengcheng Street.The most suitable areas for S.glabra planting were mainly distributed in Puchang Village of Huilong Town,the northeast of Huangzhai Town,the south of Fengcheng Street,and the central-southern part of Matou Town.The zoning results provide a scientific basis for the rational layout of M.officinalis and S.glabra planting in Xinfeng County. 展开更多

油楠(Sindora glabra)作为极具开发潜力的能源植物,在分子水平上的研究仅局限于ISSR分子标记。本研究利用Trinity软件对油楠转录组数据进行组装,共得到总长为48307806 bp的283998条Unigenes。利用MISA软件寻找到97443个含有SSR的重复基... 油楠(Sindora glabra)作为极具开发潜力的能源植物,在分子水平上的研究仅局限于ISSR分子标记。本研究利用Trinity软件对油楠转录组数据进行组装,共得到总长为48307806 bp的283998条Unigenes。利用MISA软件寻找到97443个含有SSR的重复基元。SSR位点中主要重复序列为单核苷酸和二核苷酸,分别占总SSR位点的67.68%和22.56%,其次是三核苷酸,占8.54%。利用Primer 3.0设计引物,并从中随机选择200对引物进行PCR扩增,其中有13对扩增出清晰条带,多态性高,重复性好。油楠SSR分子标记的开发对于研究其遗传多样性、重要性状基因定位及分子标记辅助选择育种等起到重要的推动作用。 展开更多

Total glucosides of Rhizoma Smilacis Glabrae(RSG)are selective immunosuppressants that exhibit primary efficacy in the treatment of rheumatoid arthritis through targeted inhibition of activated T cells.In this study,we aimed to investigate the potential application of RSG in the treatment of psoriasis and elucidate its mechanism of action and material basis.Our findings revealed significant improvements upon administration of RSG in an imiquimod(IMQ)-induced psoriasis model.These improvements were characterized by a remarkable increase in the number of tail scales in mice and a substantial amelioration of skin erythema,ulceration,and flaking.By transcriptome sequencing and T-cell flow sorting assay,we identified notable effects of RSG on the modulation of various cellular processes.Specifically,RSG prominently down-regulated the Th17/Treg ratio in damaged skin tissues and reduced the proportion of G2 phase cells.Furthermore,RSG exhibited a stimulatory effect on the proliferation and differentiation of epithelial cells.Of particular interest,we discovered thatβ-sitosterol,sitostenone,stigmasterol,smiglanin,and cinchonain Ib displayed potent inhibitory effects on the IL-17-mediated inflammatory response in HaCaT cells.In summary,our study highlights the therapeutic potential of RSG in the treatment of psoriasis,attributed to its ability to regulate the Th17/Treg balance.These findings contribute to the development of new indications for RSG and provide a solid theoretical foundation for further exploration in this field. 展开更多

Sarcandra glabra(Thunb.)Nakai is a perennial evergreen herb categorised within the Sarcandra Gardner genus under the Chloranthaceae family.Indigenous to tropical and subtropical regions of East Asia and India,this species is extensively distributed across China,particularly in the southern regions(Sichuan,Yunnan,and Jiangxi).In addition to its high ornamental value,S.glabra has a rich history of use in traditional Chinese medicine,evident through its empirical prescriptions for various ailments like pneumonia,dysentery,fractures,bruises,numbness,amenorrhea,rheumatism,and other diseases.Besides,modern pharmacological studies have revealed various biological activities,such as antitumour,anti-bacterial,anti-viral anti-inflammatory and immunomodulatory effects.The diverse chemical constituents of S.glabra have fascinated natural product researchers since the 1900s.To date,over 400 compounds including terpenoids,coumarins,lignans,flavonoids,sterols,anthraquinones,organic acids,and organic esters have been isolated and characterised,some featuring unprecedented structures.This review comprehensively examines the current understanding of S.glabra’s phytochemistry and pharmacology,with emphasis on the chemistry and biosynthesis of its unique chemotaxonomic marker,the lindenane-type sesquiterpenoids. 展开更多

This study was conducted to comprehensively evaluate the effects of salicylic acid and sodium molybdate on cold tolerance of an ornamental plant Bougainvillea glabra and to provide a theoretical guidance for landscape maintenance.B.glabra plants were treated with 0.5 mmol/L salicylic acid and 2.0 μmol/L alone or in combination,and then exposed to low temperature stress before physiological indices were measured.The results showed that all salicylic acid and sodium molybdate treatments reduced the relative conductivity and malondialdehyde( MDA) content of B.glabra to varying extents under the stress of low temperature,and more significant effect was achieved by using the two agents in combination.Oxygen free radicals production rate increased with decreasing temperature from 20 to 6 ℃,but declined with temperature decreasing from 3 to-3 ℃.The SOD activity of the control( CK) was significantly lower than that of other treatments at 0 and-3 ℃.The treatments with salicylic acid and sodium molybdate alone and in combination increased POD activity of B.glabra plants,especially at 0 ℃,as the POD activity of treatments T1,T2 and T3 was significantly higher than that of CK at 0 ℃.In addition,under low temperature stress,the contents of soluble sugar,starch and proline increased initially and decreased subsequently with temperature decreasing.The soluble sugar content at 3 ℃,starch and proline contents at 0 and-3 ℃ in treatments with salicylic acid and sodium molybdate alone and in combination were significantly higher than those of CK.All above results proved that salicylic acid and sodium molybdate are able to improve cold tolerance of B.glabra,and better effect can be achieved by using them together. 展开更多

To illuminate the similarities and differences between wild and cultivated Sarcandra glabra(S.glabra),we performed a comprehensively study on 26 batches of cultivated S.glabra and 2 batches of wild S.glabra.Chemical constituents and distribution characteristics of roots,stems and leaves in both wild and cultivated S.glabra were investigated through UHPLC-TOF-MS method.The result revealed that there were significant differences between roots,stems and leaves in S.glabra.And the chemical contents in the root part were less or even absence than those in leaf and stem,which suggested the root organ could be excluded as medicine.Meanwhile,the chemical contents of stems and leaves in cultivated S.glabra was sightly higher than that of wild samples.Therefore,cultivated S.glabra may have a high potential for substitution of wild S.glabra without affecting its pharmaceutical properties.In summary,our study could provide important information to the molecular basis for quality control of S.glabra. 展开更多

Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chromatography/diode array detector(HPLC/DAD).The conditions of MAE were optimized by an orthogonal experiment,and then the authentication and validation of the chromatographic fingerprint were conducted.Nine peaks were identified as common peaks in the fingerprint chromatograms,and isofraxidin was considered as a reference compound and quantified.Relative standard deviations of retention time and peak area of each component were less than 3%and 8%,respectively.Similarity and difference analysis were conducted by use of PCA and relation coefficient.Twenty batches of sarcandra glabra(Thunb.)samples from two different producing areas could be classified into two different groups in the PCA model.The results showed that MAE-HPLC/DAD method was simple,efficient and stable for the study of complex chromatographic fingerprint of sarcandra glabra(Thunb.),which could provide more reliable and precise information for quality evaluation. 展开更多

对缙云山草珊瑚8个天然居群结实特征的20项表型性状的变异性进行了研究,结果表明:各表型性状在居群内与居群间都存在很大的变异性,其中种实产量、果序数和果序分枝的变异程度较高,而果实和种子的特征指数变异程度较低,同类性状果实的变... 对缙云山草珊瑚8个天然居群结实特征的20项表型性状的变异性进行了研究,结果表明:各表型性状在居群内与居群间都存在很大的变异性,其中种实产量、果序数和果序分枝的变异程度较高,而果实和种子的特征指数变异程度较低,同类性状果实的变异程度要大于种子;平均变异系数最大的性状是四分枝果序百分比(1.477),最小的是种子长宽比(0.041);变异程度最大的居群是居群6(0.564),最小的是居群4(0.292);各居群间除果序分枝总数差异不显著、果序数和二分枝果序百分比差异显著外,其它17项表型指标均达到极显著水平,表明缙云山草珊瑚各居群间已经发生了明显的形态分化;居群内和居群间变异分别占总变异的60.7%和39.3%,说明居群内变异是表型变异主要来源,但居群间变异也不容忽视;欧氏距离聚类将所有居群划归为两大类群,主要与居群的群落类型与干扰程度有关;表型性状的Pearson相关性聚类显示草珊瑚同类性状在表型分化过程中具有很强的相关性。 展开更多

An ultra-performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-qTOF-MS/MS) method was developed and validated for the simultaneous determination of glycyrrhizin and glycyrrhetic acid. These analytes were separated on a reverse phase C18 column using a mobile phase of acetonitrile:2% acetic acid in water (75:25, v/v) with a flow rate of 200 μL/min. The qTOF-MS was operated under multiple reaction monitoring (MRM) mode using the electrospray ionization (ESI) technique with positive ion polarity. A comparison of three different extraction techniques i.e. accelerated solvent extraction (ASE), extraction under ultrasonic waves (USW) and the classical extraction by percolation (CE) method was done and quantification of these extracts was also carried out by the proposed method. 展开更多
