Cloning ZDS gene and developing functional mar... ζ (zeta)-carotene desaturase (ZDS) is a key enzyme for carotenoid biosynthesis, demonstrating high association with the yellow pigment (YP) content in wheat grain. Cloning ZDS gene and developing functional markers are important for marker-assisted selection in wheat breeding. In the present study, the full-length DNA sequence of a ZDS gene on wheat chromosome 2A, designated TaZds-A1, was cloned, with 14 exons and 13 introns, and it has an open reading frame (ORF) of 1 707 bp, encoding 568 amino acid residues. A co-dominant functional marker, YP2A-1, was designed based on the polymorphisms of two alleles at the locus, TaZds-Ala and TaZds-Alb, yielding 183- and 179-bp fragments in TaZds-Ala and TaZds-Alb genotypes, respectively. A new QTL for YP content was detected on chromosome 2A, co-segregating with the functional marker YP2A-1 and TaZds-A1; it explained 11.3% of the phenotypic variance for YP content in a doubled haploid (DH) population from Zhongyou 9507/CA9632. Among 217 Chinese wheat cultivars and advanced lines, the average grain YP content of 126 cultivars with TaZds-Alb allele was 7.8% higher than that of 91 cultivars with TaZds- Ala allele. 展开更多

Low glutelincontent gene Lgc1 in rice has a 3.5-kb deletion between two highly similar glutelin genes GluB4 and GluB5, which locates on the short arm of chromosome 2. To improve the selection efficiency in low glutelin-content rice breeding, two molecular markers designated as InDel-Lgc1-1 and InDel-Lgc1-2 were developed to detect the low glutelin-content gene Lgc1. A double PCR detection indicated that combined use of the two markers could easily distinguish the genotypes of Lgc1 from different rice varieties. Therefore, as a simple and low-cost technique, the molecular marker could be widely used to identify different varieties with Lgc1 gene and applied in marker-assisted selection of low glutelin-content rice. 展开更多

Rice blast disease is one of the most devastating diseases of rice(Oryza sativa L.)caused by the fungus Magnaporthe oryzae(M.oryzae),and neck blast is the most destructive phase of this illness.The underlying molecular mechanisms of rice blast resistance are not well known.Thus,we collected 150 rice varieties from different ecotypes in China and assessed the rice blast resistances under the natural conditions that favoured disease development in Jining,Shandong Province,China in 2017.Results showed that 92(61.3%)and 58(38.7%)rice varieties were resistant and susceptible to M.oryzae,respectively.Among the 150 rice varieties screened for the presence of 13 major blast resistance(R)genes against M.oryzae by using functional markers,147 contained one to eight R genes.The relationship between R genes and disease response was discussed by analysing the phenotype and genotype of functional markers.The results showed that the rice blast resistance gene Pita was significantly correlated with rice blast resistance.Our results provided a basis for the further understanding of the distribution of 13 major R genes of rice blast in the germplasm resources of the tested rice varieties,and were meaningful for rice disease resistance breeding. 展开更多

Field resistances of nine accessions of common wild rice (Oryza rufipogon Griff.) and one rice variety (IR24) were evaluated by using nine strains of bacterial blight pathogen (Xanthomonas oryzae pv. oryzae) from the Philippines. IR24 was highly susceptible to all the strains, and six common wild rice accessions resisted all the nine strains, with a resistance frequency of 67%. The accessions Yulin and Wanning were only susceptible to PXO280 and PXO71, respectively. The accession Gaozhou was susceptible to the three strains PXO79, PXO99 and PXO339, whereas resistant to the other six strains. It could be concluded that there is at least one resistance gene in each common wild rice accession. The functional markers of the genes xa5, xa13, Xa21 and Xa27 were used to detect the presence of these resistance genes in the nine tested wild rice accessions, and it was found that four wild rice accessions contained heterozygous xa13. Among the nine common wild rice accessions, five were homozygous for Xa27 and three homozygous for xa27, and the accession Laibin contained neither xa27 nor Xa27. Thirty-one PSM accessions wer... Paw San Hmwe (PSM) rice has been cultivated in many areas of Myanmar for a long time. Strong aroma, good taste and its elongation during cooking are the key characteristics of PSM rice. Thirty-one PSM accessions were genotypically characterized, and their physical grain and cooking quality traits were studied. We used specific gene markers associated with aroma, apparent amylose content (AAC) and alkali spreading value to determine the alleles carried by different PSM accessions. The results revealed that six PSM accessions (PSM10, PSM12, PSM13, PSM21, PSM22 and PSM30) had a 3-bp insertion in Os2AP gene. Gel consistency (GC) allele was predominant among the PSM accessions for gelatinization temperature (GT), however, the phenotype observed was between low and intermediate GT because of the combination of the GC allele with the presence of low GT allele at heterozygous state from the other loci of the SSIla gene. Intermediate to high AAC was observed among the PSM accessions corresponding to the haplotype identified for the single nucleotide polymorphism G/T and the (Cm)n repeat in the Wx gene. The characterization and grouping data of PSM accessions posted benefits to Myanmar seed banks, and our results will help in maintaining the integrity of PSM rice variety. 展开更多

In marker-assisted breeding for bacterial blight(BB) resistance in rice, three major resistance genes, viz., Xa21, xa13 and xa5, are routinely deployed either singly or in combinations. As efficient and functional markers are yet to be developed for xa13 and xa5, we have developed simple PCR-based functional markers for both the genes. For xa13, we designed a functional PCR-based marker, xa13-prom targeting the In Del polymorphism in the promoter of candidate gene Os8N3 located on chromosome 8 of rice. With respect to xa5, a multiplex-PCR based functional marker system, named xa5 FM, consisting of two sets of primer pairs targeting the 2-bp functional nucleotide polymorphism in the exon II of the gene TFIIA5(candidate for xa5), has been developed. Both xa13-prom and xa5 FM can differentiate the resistant and susceptible alleles for xa13 and xa5, respectively, in a co-dominant fashion. Using these two functional markers along with the already reported functional PCR-based marker for Xa21(p TA248), we designed a single-tube multiplex PCR based assay for simultaneous detection of all the three major resistance genes and demonstrated the utility of the multiplex marker system in a segregating population. 展开更多

The aim of this study was to assess the prevalence of sleep-disordered breathing (SDB) symptoms among primary schoolchildren, and to objectively determine the influence of SDB on the intra-oral environment through the analysis of saliva. A questionnaire survey was conducted among approximately 400 children from a primary school in Hiroshima. Parents were asked to complete the questionnaire and provide their contact information if they allowed the collection of saliva samples from their children. Thirty-eight chil- dren agreed to participate in the saliva study. Habit- ual snoring and cessation of breathing during sleep were found in approximately 8% and 1% of children, respectively. The present results showed significant correlations between snoring and mouth breathing. A significant association between excessive daytime sle- epiness (EDS) and learning problems was found. Fur- thermore, among children between the ages of 7 and 12 years, those with EDS and learning problems tended to be older. SDB symptom scores were statisti- cally significant only in relation to EDS. The present study also demonstrated significantly higher levels of salivary IgA and cortisol in children with sleep-re- lated disorders. The present study determined the pre- valence and characteristics of SDB among Japanese primary schoolchildren and their effects on the oral environment. Approximately 8% of primary school- children with habitual snoring might need to be care- fully monitored for SDB symptoms and immune sta- tus to ensure proper psychological and physical deve- lopment. 展开更多

Soybean mosaic virus(SMV)is a worldwide disease significantly decreasing soybean yield and seed quality.In this study,a genome-wide association study(GWAS)for SMV-SC3 resistance was conducted by using a deep re-sequencing dataset of 547 soybean accessions.A total of 11,405 SNPs and 1566 InDels were significantly associated with disease index(DI)at seedling stage and eight yield-and seed quality-related traits to SC3 resistance under eight environments.Among these genetic loci,952 SNPs and 118 InDels were firstly identified to control SC3 resistance,and 52.42% and 42.37% of them were pleiotropic loci across multiple environments.Notably,the 8.47-8.89 Mb genomic region on chromosome 18 was firstly discovvered to associate with DI at seedling stage and four related traits at adult stage across multiple environments.Furthermore,the causal gene Gm18GRSC3 was identified and validated in this stable and pleiotropic locus for resistance to SC3 via positive and negative transgenic strategies.Overexpression of Gm18GRSC3 significantly decreased the accumulation of SC3 in transgenic soybean hairy roots,while silencing of Gm18GRSC3 significantly increased SC3 accumulation in soybean leaves.A functional marker,FM18GSC3,was developed based on the allelic variation of Gm18GRSC3,and the detection efficiency reached to 76% in another 100 soybean accessions.These findings provide valuable genetic loci and a functional gene for the improvement of SMV resistance in soybean. 展开更多

目的 观察调肝理肺汤对慢性阻塞性肺疾病急性加重期(AECOPD)患者的临床疗效、中医证候积分、慢性阻塞性肺疾病评估测试量表(CAT)评分、改良英国医学研究委员会呼吸困难量表(mMRC)评分、肺通气功能、血气指标以及炎症指标的影响。方法 选... 目的 观察调肝理肺汤对慢性阻塞性肺疾病急性加重期(AECOPD)患者的临床疗效、中医证候积分、慢性阻塞性肺疾病评估测试量表(CAT)评分、改良英国医学研究委员会呼吸困难量表(mMRC)评分、肺通气功能、血气指标以及炎症指标的影响。方法 选取AECOPD患者92例,按随机数字表法分为两组。对照组采用西医常规治疗,观察组在对照组基础上加服调肝理肺汤。评价两组患者临床疗效、中医证候积分、CAT评分、mMRC评分,比较两组治疗前后肺通气功能[用力肺活量(FVC)、第1秒用力呼气容积(FEV_(1))、第1秒用力呼气容积/用力肺活量比值(FEV_(1)/FVC)]、血气指标[动脉血氧分压(PaO_(2))、氧合指数(OI)、肺泡动脉血氧分压差(PA-aDO_(2))]、炎症指标[C反应蛋白(CRP)、降钙素原(PCT)]差异。结果 观察组治疗后的中医证候积分、CAT评分、mMRC评分较对照组降低更明显(P<0.05);与对照组比较,观察组治疗后FEV_(1)/FVC明显升高(P<0.05),FVC、FEV_(1)两组治疗后均有所改善;与对照组相比,观察组治疗后PaO_(2)、OI水平明显改善(P<0.05),PAaDO_(2)明显降低;观察组PCT、CRP下降幅度较对照组更明显(P<0.05)。结论 调肝理肺汤治疗AECOPD可以显著改善患者的临床症状,改善肺通气功能,降低机体炎症水平。 展开更多

目的:探究依洛尤单抗联合沙库巴曲缬沙坦钠对急性心肌梗死(AMI)患者介入治疗后心功能及主要不良心血管事件(MACE)发生情况的影响。方法:共90例AMI患者,均为2023年12月—2024年7月常熟市第一人民医院收治,按随机数字表法将其分为两组,均... 目的:探究依洛尤单抗联合沙库巴曲缬沙坦钠对急性心肌梗死(AMI)患者介入治疗后心功能及主要不良心血管事件(MACE)发生情况的影响。方法:共90例AMI患者,均为2023年12月—2024年7月常熟市第一人民医院收治,按随机数字表法将其分为两组,均于本院行经皮冠状动脉介入术(PCI)治疗,对照组(n=45)给予沙库巴曲缬沙坦钠治疗,观察组(n=45)在此基础上给予依洛尤单抗治疗。治疗时间为3个月。对比两组临床疗效、心功能、心肌损伤标志物及MACE发生情况。结果:治疗后,观察组总有效率(97.78%)高于对照组(82.22%)(P<0.05)。治疗后,观察组左心室射血分数(53.37±1.42)%、每搏输出量(58.26±4.13)mL、心排血量(5.03±0.48)L/min,分别高于对照组的(48.92±1.76)%、(49.67±3.34)mL、(4.51±0.46)L/min,左室舒张末期内径(47.13±3.42)mm、N末端B型利钠肽原(581.93±72.25)ng/L,分别低于对照组的(52.27±5.15)mm、(842.74±81.94)ng/L(P<0.05)。观察组MACE发生率(2.22%)低于对照组(17.78%)(P<0.05)。结论:依洛尤单抗联合沙库巴曲缬沙坦钠可减轻AMI介入治疗患者心肌损伤,促进心功能改善,且可有效减少MACE发生风险。 展开更多

目的:探讨早期非小细胞肺癌(NSCLC)经PD-1抑制剂与立体定向体部放疗(SABR)联合治疗的疗效观察。方法:选取154例NSCLC患者作为研究对象。按照随机数字表法,将其分为观察组和对照组,各77例。对照组给予SABR治疗,观察组给予PD-1抑制剂联合S... 目的:探讨早期非小细胞肺癌(NSCLC)经PD-1抑制剂与立体定向体部放疗(SABR)联合治疗的疗效观察。方法:选取154例NSCLC患者作为研究对象。按照随机数字表法,将其分为观察组和对照组,各77例。对照组给予SABR治疗,观察组给予PD-1抑制剂联合SABR治疗。比较两组患者临床疗效、肿瘤标志物水平、免疫功能、生活质量及不良反应。结果:观察组ORR(45.45%)及DCR(84.42%)显著高于对照组(19.48%、54.55%)(均P<0.05)。治疗后,观察组CEA、CYFRA21-1及VEGF水平显著低于对照组(均P<0.05)。观察组CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)及NK水平显著高于对照组(均P<0.05);CD8^(+)显著低于对照组(P<0.05)。观察组生活质量改善率(80.52%)显著高于对照组(46.75%,P<0.05)。观察组总不良反应率(10.39%)显著低于对照组(22.08%,P<0.05)。结论:PD-1抑制剂与SABR联合应用能够显著提高早期NSCLC患者治疗效果。 展开更多

抗除草剂品种与对应的除草剂结合使用是解决水稻草害问题的一种重要方法,利用功能标记辅助选择可以加快抗除草剂新品种选育。为了快速改良现有水稻品种除草剂抗性,本研究开发了抗除草剂基因的功能标记,利用抗咪唑啉酮类除草剂的水稻种质... 抗除草剂品种与对应的除草剂结合使用是解决水稻草害问题的一种重要方法,利用功能标记辅助选择可以加快抗除草剂新品种选育。为了快速改良现有水稻品种除草剂抗性,本研究开发了抗除草剂基因的功能标记,利用抗咪唑啉酮类除草剂的水稻种质HR1对扬航糯1号进行了改良。序列比对分析结果表明,本研究中抗除草剂品种乙酰乳酸合成酶基因(ALS)编码的第548位氨基酸由色氨酸突变为甲硫氨酸(W548M),赋予了水稻对ALS抑制剂类除草剂的抗性。为加快育种进程,根据ALS基因序列特异性开发了基于Tetra-primer ARMS-PCR技术原理的功能标记ALS1642。该标记可以准确鉴定基因型,且与苗期的除草剂抗性表型一致。利用该标记对扬航糯1号与HR1杂交衍生的遗传分离群体进行抗除草剂基因筛选,获得了含抗除草剂基因的水稻新品系。研究结果表明ALS1642可以广泛用于W548M突变基因型的标记辅助选择和抗咪唑啉酮类除草剂水稻新品种的培育。本研究在改良水稻品种除草剂抗性和提高育种效率方面具有重要意义。 展开更多

目的探讨腹腔镜卵巢癌根治术对早期卵巢癌患者血清肿瘤标志物的影响。方法按照随机数字表法将82例早期卵巢癌患者分成2组,每组41例。对照组实施传统开腹手术,观察组实施腹腔镜卵巢癌根治术。比较2组围术期指标、血清肿瘤标志物、免疫功... 目的探讨腹腔镜卵巢癌根治术对早期卵巢癌患者血清肿瘤标志物的影响。方法按照随机数字表法将82例早期卵巢癌患者分成2组,每组41例。对照组实施传统开腹手术,观察组实施腹腔镜卵巢癌根治术。比较2组围术期指标、血清肿瘤标志物、免疫功能、生活质量及并发症。结果与对照组相比,观察组手术耗时明显缩短,术后排气时间缩短,住院时间显著缩短,术中出血量也显著降低,淋巴结清扫数量明显增多,视觉模拟评分法(VAS)评分显著降低,差异有统计学意义(P<0.05);术后观察组癌胚抗原(CEA)水平低于对照组,T淋巴细胞CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)水平高于对照组,CD8^(+)低于对照组,健康调查简表(SF-36)评分高于对照组;术后并发症少于照组,差异有统计学意义(P<0.05)。结论腹腔镜卵巢癌根治术治疗早期卵巢癌患者疗效确切,有助于改善患者围术期指标,减轻疼痛程度,降低肿瘤标志物水平,增强免疫功能,患者生活质量高,且并发症少,术后恢复速度快,值得临床借鉴。 展开更多

抗癌平丸联合SOX化疗方案治疗晚期胃癌的临床研究

目的观察抗癌平丸联合SOX化疗方案治疗晚期胃癌的临床效果。方法将2022年2月—2024年10月安庆市第一人民医院收治106例晚期胃癌患者采用随机数字表法分为对照组和治疗组,每组各53例。对照组患者采用SOX化疗方案,第1天静脉滴注注射用奥... 目的观察抗癌平丸联合SOX化疗方案治疗晚期胃癌的临床效果。方法将2022年2月—2024年10月安庆市第一人民医院收治106例晚期胃癌患者采用随机数字表法分为对照组和治疗组,每组各53例。对照组患者采用SOX化疗方案,第1天静脉滴注注射用奥沙利铂,130 mg/m^(2)加入250 mL 5%葡萄糖溶液,持续滴注2 h;同时口服替吉奥胶囊,体表面积(BSA)<1.25 m^(2)患者40 mg/次;1.25 m^(2)≤BSA<1.50 m^(2)患者50 mg/次;BSA≥1.50 m^(2)患者60 mg/次,2次/d;第1~14天口服替吉奥胶囊,第15~21天停药,21 d为1个周期,共治疗4个周期。治疗组在对照组基础上饭后0.5 h口服抗癌平丸,0.5~1.0 g/次,3次/d,连续服用84 d。观察两组患者临床疗效,比较治疗前后两组患者卡氏功能状态量表(KPS)评分和生命质量测定量表(FACT-G)评分,血清糖类抗原72-4(CA72-4)、CA19-9、癌胚抗原(CEA)和CA125水平,及外周血CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)和自然杀伤细胞(NK)水平。结果治疗后,治疗组患者疾病控制率为84.91%,明显高于对照组的67.92%(P<0.05)。治疗后,两组KPS评分、FACT-G评分明显高于治疗前(P<0.05),且治疗组评分明显高于对照组(P<0.05)。治疗后,两组外周血CD3^(+)、CD4^(+)/CD8^(+)、CD4^(+)、NK均低于治疗前,而CD8^(+)高于治疗前(P<0.05),且治疗组外周血免疫功能指标明显好于对照组(P<0.05)。治疗后,两组患者血清CA72-4、CEA、CA19-9、CA125均低于治疗前(P<0.05),且治疗组血清胃癌肿瘤标志物水平明显低于对照组(P<0.05)。治疗组患者不良反应总发生率为18.87%,低于对照组的39.62%(P<0.05)。结论抗癌平丸联合SOX化疗方案治疗晚期胃癌可抑制血清肿瘤标志物表达,改善生活质量,提高疾病控制率,减少毒副反应,对免疫功能影响较小。 展开更多

小偃麦衍生系抗旱基因功能分子标记检测及效应分析

为进一步明确小偃麦衍生系抗旱等位基因组成,采用5个小麦抗旱基因TaNRX-B1、TaDreb-B1、TaPYL1-1B、TaBADH-A1和TaDRO-5B的功能分子标记检测了175份小偃麦衍生系,并结合萌发期13个抗旱指标的抗旱系数和综合评价D值,分析了各等位基因的... 为进一步明确小偃麦衍生系抗旱等位基因组成,采用5个小麦抗旱基因TaNRX-B1、TaDreb-B1、TaPYL1-1B、TaBADH-A1和TaDRO-5B的功能分子标记检测了175份小偃麦衍生系,并结合萌发期13个抗旱指标的抗旱系数和综合评价D值,分析了各等位基因的抗旱效应。结果显示,5个抗旱等位基因在小偃麦衍生系中的频率总体偏低,其中,TaDreb-B1a的频率最高,为38.86%;TaDRO-5B基因的Hap-5B-I的频率最低,为5.14%。在117份中等以上抗旱性衍生系中,2份携带4个抗旱等位基因的衍生系CH6310(TaDreb-B1a/TaBADH-A1b/TaPYL1-1B^(In-442)/Hap-5B-I)和CH1539(TaNRX-B1a/TaDreb-B1a/TaBADH-A1b/Hap5B-I)均为强抗旱种质;13份强抗旱衍生系未检测到任何抗旱等位基因,可能存在未知的抗旱相关基因。携带抗旱等位基因TaDreb-Bla、TaPYL1-1B^(In-442)和Hap-5B-I种质的多数指标抗旱系数均值高于携带其干旱敏感等位基因的衍生系,且抗旱等位基因与多个指标抗旱系数呈显著正相关,抗旱效应较高。因此,抗旱基因TaDreb-B1、TaPYL1-1B和TaDRO-5B的功能标记能够有效用于小偃麦衍生系抗旱基因型检测及分子标记辅助选择育种。 展开更多
