They are synthesized by the elongation of very long fatty acids-4(ELOVL4) enzyme, expressed mainly in the brain, retina, skin, testes, and meibomian gland, where these fatty acids are found(Agbaga et al., 2008). Further, these organs exhibit tissuespecific VLC-PUFA and VLC-SFA biosynthesis and incorporation into complex lipids for specific functions. In the brain, skin, and Meibomian glands, the ELOVL4 mainly makes VLC-SFA, which are incorporated into complex sphingolipids. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).展开更多

文摘Very long chain-saturated and-polyunsaturated fatty acids(VLC-SFA and VLC-PUFA, respectively) are a functionally important class of fatty acids containing 28 carbons or more in their acyl chain. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).

文摘Zinc-ion batteries(ZIBs)are inexpensive and safe, but side reactions on the Zn anode and Zn dendrite growth hinder their practical applications. In this study,1,3,5-triformylphloroglycerol(Tp)and various diamine monomers(p-phenylenediamine(Pa),benzidine(BD),and 4,4"-diamino-p-terphenyl(DATP))were used to synthesize a series of two-dimensional covalent-organic frameworks(COFs).The resulting COFs were named TpPa,TpBD,and TpDATP,respectively, and they showed uniform zincophilic sites, different pore sizes, and high Young's moduli on the Zn anode.Among them,TpPa and TpBD showed lower surface work functions and higher ion transfer numbers, which were conducive to uniform galvanizing/stripping zinc and inhibited dendrite growth.Theoretical calculations showed that TpPa and TpBD had wider negative potential region and greater adsorption capacity for Zn2+than TpDATP,providing more electron donor sites to coordinate with Zn^(2+).Symmetric cells protected by TpPa and TpBD stably cycled for more than 2300 h,whereas TpDATP@Zn and the bare zinc symmetric cells failed after around 150 and200 h.The full cells containing TpPa and TpBD modification layers also showed excellent cycling capacity at 1 A/g.This study provides comprehensive insights into the construction of highly reversible Zn anodes via COF modification layers for advanced rechargeable ZIBs.

文摘Hydrogel scaffolds have numerous potential applications in the tissue engineering field. However, tough hydrogel scaffolds implanted in vivo are seldom reported because it is difficult to balance biocompatibility and high mechanical properties.Inspired by Chinese ramen,we propose a universal fabricating method(printing-P,training-T,cross-linking-C,PTC&PCT)for tough hydrogel scaffolds to fill this gap.First,3D printing fabricates a hydrogel scaffold with desired structures(P).Then, the scaffold could have extraordinarily high mechanical properties and functional surface structure by cycle mechanical training with salting-out assistance(T).Finally, the training results are fixed by photo-cross-linking processing(C).The tough gelatin hydrogel scaffolds exhibit excellent tensile strength of 6.66 MPa(622-fold untreated)and have excellent biocompatibility.Furthermore, this scaffold possesses functional surface structures from nanometer to micron to millimeter, which can efficiently induce directional cell growth.Interestingly, this strategy can produce bionic human tissue with mechanical properties of 10 kPa-10 MPa by changing the type of salt, and many hydrogels, such as gelatin and silk, could be improved with PTC or PCT strategies.Animal experiments show that this scaffold can effectively promote the new generation of muscle fibers, blood vessels, and nerves within 4 weeks, prompting the rapid regeneration of large-volume muscle loss injuries.

文摘Fe-Mo functionally graded materials(FGMs)with different composition-change rates from 100%304 stainless steel to 100%Mo along the composition gradient direction were prepared by electron beam-directed energy deposition(EB-DED)technique,including three samples with composition mutation of 100%,composition change rate of 10%and 30%.Results show that the composition-change rate significantly affects the microstructure and mechanical properties of the samples. In the sample with abrupt change of composition, the sharp shift in composition between 304 stainless steel and Mo leads to a great difference in the microstructure and hardness near the interface between the two materials. With the increase in the number of gradient layers, the composition changes continuously along the direction of deposition height, and the microstructure morphology shows a smooth transition from 304 stainless steel to Mo,which is gradually transformed from columnar crystal to dendritic crystal.Elements Fe,Mo,and other major elements transform linearly along the gradient direction, with sufficient interlayer diffusion between the deposited layers, leading to good metallurgical bonding. The smaller the change in composition gradient, the greater the microhardness value along the deposition direction. When the composition gradient is 10%,the gradient layer exhibits higher hardness(940 HV)and excellent resistance to surface abrasion, and the overall compressive properties of the samples are better, with the compressive fracture stress in the top region reaching 750.05±14 MPa.

文摘Spinal cord injury represents a severe form of central nervous system trauma for which effective treatments remain limited.Microglia is the resident immune cells of the central nervous system, play a critical role in spinal cord injury.Previous studies have shown that microglia can promote neuronal survival by phagocytosing dead cells and debris and by releasing neuroprotective and anti-inflammatory factors. However, excessive activation of microglia can lead to persistent inflammation and contribute to the formation of glial scars, which hinder axonal regeneration.Despite this, the precise role and mechanisms of microglia during the acute phase of spinal cord injury remain controversial and poorly understood. To elucidate the role of microglia in spinal cord injury,we employed the colony-stimulating factor 1 receptor inhibitor PLX5622 to deplete microglia. We observed that sustained depletion of microglia resulted in an expansion of the lesion area, downregulation of brain-derived neurotrophic factor, and impaired functional recovery after spinal cord injury.Next,we generated a transgenic mouse line with conditional overexpression of brain-derived neurotrophic factor specifically in microglia. We found that brain-derived neurotrophic factor overexpression in microglia increased angiogenesis and blood flow following spinal cord injury and facilitated the recovery of hindlimb motor function.Additionally,brain-derived neurotrophic factor overexpression in microglia reduced inflammation and neuronal apoptosis during the acute phase of spinal cord injury.Furthermore, through using specific transgenic mouse lines,TMEM119,and the colony-stimulating factor 1 receptor inhibitor PLX73086,we demonstrated that the neuroprotective effects were predominantly due to brain-derived neurotrophic factor overexpression in microglia rather than macrophages. In conclusion, our findings suggest the critical role of microglia in the formation of protective glial scars.Depleting microglia is detrimental to recovery of spinal cord injury, whereas targeting brain-derived neurotrophic factor overexpression in microglia represents a promising and novel therapeutic strategy to enhance motor function recovery in patients with spinal cord injury.

文摘Emerging evidence of the beneficial effects of defatted rice bran(DFRB)on gut health has advanced the development of fermented defatted rice bran as a potential functional food. However, less is known about its effects and underlying mechanisms on gut health. In this study,a mouse model together with fecal microbiota transplantation(FMT)was utilized to study the effects and mechanisms of fermented DFRB(FR)on gut barrier function. We found that FR improved the intestinal morphology, gut tight junction proteins, mucin, antimicrobial peptides, and interleukin 22(IL-22)and promoted the gut Clostridium butyricum and butyrate.Notably, correlation analysis indicated gut C.butyricum and butyrate were two FR-induced effectors that improved gut health.FMT results suggested that C.butyricum, butyrate, and fecal microbiota from the FR group all reduced prolyl hydroxylase 2(PHD2)expression by activating peroxisome proliferator-activated receptor gamma(PPARγ)in the mouse colon. This decrease in gut PHD2 subsequently upregulated the hypoxia-inducible factor-1 alpha(HIF-1α)expression,which in turn increased the expression of its targeted downstream tight junction proteins, mucin and antimicrobial peptides, and colonic IL-22 secretion.Overall,FR-derived C.butyricum and butyrate might improve gut barrier function through the HIF-1 signaling pathway, which provides a reference for the application of fermented DFRB as a potential functional food for improving of gut barrier function.

文摘In distributed fusion, when one or more sensors are disturbed by faults,a common problem is that their local estimations are inconsistent with those of other fault-free sensors. Most of the existing fault-tolerant distributed fusion algorithms, such as the Covariance Union(CU)and Faulttolerant Generalized Convex Combination(FGCC),are only used for the point estimation case where local estimates and their associated error covariances are provided.A treatment with focus on the fault-tolerant distributed fusions of arbitrary local Probability Density Functions(PDFs)is lacking. For this problem,we first propose Kullback–Leibler Divergence(KLD)and reversed KLD induced functional Fuzzy c-Means(FCM)clustering algorithms to soft cluster all local PDFs,respectively. On this basis, two fault-tolerant distributed fusion algorithms of arbitrary local PDFs are then developed. They select the representing PDF of the cluster with the largest sum of memberships as the fused PDF.Numerical examples verify the better fault tolerance of the developed two distributed fusion algorithms.

文摘Surface deformation calculations due to environmental loading typically rely on the Preliminary Reference Earth Model(PREM),which assumes a homogeneous and isotropic Earth structure, leading to noticeable errors. To enhance accuracy, the high-precision crustal model CRUST 1.0 is used to refine calculations of regional surface deformation caused by hydrological and non-tidal atmospheric loading. The improved model is applied to 27 Global Navigation Satellite System(GNSS)reference stations in the first phase of the Crustal Movement Observation Network of China(CMONOC),considering their geographical locations.Green's functions are employed to compute surface deformation at each site. Results indicate relative discrepancies of 11.78%and 14.14%for non-tidal atmospheric and hydrological loading compared to PREM,with vertical deformation differences reaching an average of 18.95%.Additionally,the distinct spatial distribution characteristics of the relative differences in each direction indicate that the improved RPREM model is more responsive to the mass variations derived from Gravity Recovery and Climate Experiment(GRACE).The results suggest that the improved PRREM model demonstrates higher sensitivity to loading variations than the PREM model.Utilizing the enhanced method of calculating surface deformation through the utilization of Green's function at the site could effectively reduce the calculation error caused by regional structure, leading to enhanced uniformity and isotropy of PREM.

文摘BACKGROUND Adolescents with an evening chronotype are at higher risk for mental health problems, such as depression and anxiety. However, the mechanisms underlying this relationship are not well understood.Sleep disturbances and impaired social functioning may mediate this association.AIM To examine the mediating roles of sleep quality and social functioning in the relationship between chronotype and mental health in adolescents.METHODS A cross-sectional survey was conducted among 381 adolescents(mean age 14.3 years;61.9%male)using the Morningness-Eveningness Questionnaire,Hamilton Depression Rating Scale,Hamilton Anxiety Scale,Pittsburgh Sleep Quality Index, and Social Disability Screening Schedule.Descriptive statistics,Pearson correlation analyses, and chain mediation analysis using the bootstrap method were performed.RESULTS Chronotype was significantly associated with both depression and anxiety.Sleep quality and social functioning significantly mediated these relationships. The indirect effect on depression was-0.52,accounting for 69.3%of the total effect. For anxiety, the indirect effect was -0.39, accounting for 79.3%. No significant direct effect of chronotype on depressionor anxiety was found after accounting for the mediators.CONCLUSIONSleep quality and social functioning mediate the association between chronotype and adolescent mental health. These findings highlight the importance of addressing sleep disturbances and social functioning impairments ininterventions aimed at improving mental well-being among adolescents with evening chronotypes.
文摘[Objectives]This study was conducted to investigate the mechanism of continuous cropping obstacles in Polygonatum odoratum.[Methods]Three treatments were established:continuous cropping(two consecutive crops),first-crop control(with Phaseolus vulgaris as the preceding crop),and blank control. The effects of continuous cropping on the functional diversity of soil microorganisms, soil enzyme activities, and soil nutrient coordination in the rhizosphere soil of P.odoratum during different growth stages were investigated.[Results](1)Continuous cropping increased the carbon source metabolic capacity,Shannon diversity index, and richness of rhizosphere soil microorganisms by 3.2%-14.7%,0.9%-3.5%,and 1.3%-12.5%,respectively,but the differences were not significant.(2)Principal component analysis indicated that during the middle stage of rhizome expansion, continuous cropping significantly altered the characteristics of microbial carbon metabolism, and the microbial communities utilizing carbohydrates, amino acids, polymers, carboxylic acids and amines as carbon sources exhibited vigorous metabolism.(3)Continuous cropping significantly reduced the activities of urease, polyphenol oxidase, and acid phosphatase in rhizosphere soil, with decreases of 24.4%-39.5%,3.2%-14.8%,and 7.9%-18.2%,respectively.The activities of catalase and invertase sometimes exceeded and sometimes fell below those of the first crop, showing no consistent pattern.(4)Under continuous cropping conditions, nutrient imbalance occurred in the rhizosphere soil, characterized by nitrogen deficiency and phosphorus surplus.(5)Grey correlation analysis indicated that available phosphorus content,alkali-hydrolyzable nitrogen content and polyphenol oxidase activity in rhizosphere soil were the main factors influencing microbial functional diversity.[Conclusions]This study provides a theoretical basis for understanding the formation mechanism of continuous cropping obstacles in P.odoratum.

文摘The presence of endogenous neural stem/progenitor cells in the adult mammalian brain suggests that the central nervous system can be repaired and regenerated after injury. However, whether it is possible to stimulate neurogenesis and reconstruct cortical layers II to VI in non-neurogenic regions, such as the cortex, remains unknown. In this study,we implanted a hyaluronic acid collagen gel loaded with basic fibroblast growth factor into the motor cortex immediately following traumatic injury. Our findings reveal that this gel effectively stimulated the proliferation and migration of endogenous neural stem/progenitor cells,as well as their differentiation into mature and functionally integrated neurons.Importantly, these new neurons reconstructed the architecture of cortical layers II to VI,integrated into the existing neural circuitry, and ultimately led to improved brain function. These findings offer novel insight into potential clinical treatments for traumatic cerebral cortex injuries.

文摘The acceleration of global urbanization has caused habitat loss, fragmentation, and decrease of habitat quality, often leading to a decline in biodiversity. However, most previous urbanization studies focused on taxonomic diversity, with relatively less research on functional and phylogenetic diversity. In this study,we examined the phylogenetic and functional diversity and underlying influencing factors of bird communities in 37 urban parks in Nanjing,China. We conducted a systematic survey of bird communities in Nanjing urban parks and selected six park characteristics that are generally considered to affect bird diversity.Model selection based on corrected Akaike Information Criterion(AICc)and model averaging showed that park area, habitat diversity and building index(a proxy for the degree of urbanization)were significant factors affecting avian phylogenetic and functional diversity in Nanjing urban parks.Specifically, habitat diversity and park area were positively correlated with bird diversity, while the building index was negatively correlated with bird diversity. Moreover, the phylogenetic and functional structures of urban bird communities exhibited a clustered pattern, indicating that environmental filtering might play a role in shaping community composition. In addition, building index had certain impact on the construction of bird phylogenetic communities in urban parks. Our results suggest that expanding park areas, increasing habitat diversity and reducing building indexes may be effective measures to increase the avian phylogenetic and functional diversity in our system.

文摘Amyloid-like proteins are critical for interfacial adhesion across various marine organisms and bacteria. However, the specific contributions of different functional residues remain unclear.Herein,we introduce an approach to deconstruct and mimic these residues using synthetic homopolymers and random copolymers with phenyl, amino, carboxyl, and hydroxyl functional groups using reversible addition-fragmentation chain transfer(RAFT)polymerization. The resulting polymers, designed with comparable molecular weights(M_(n):10–20 kDa)and narrow dispersities(PDI<1.3),mimic the diverse surface chemistry of amyloid-like proteins, enabling systematic investigation of their adhesive properties. The interfacial adhesion forces of different polymer films were quantified using atomic force microscopy(AFM)with a colloidal probe.Remarkably copolymers with multiple functional groups demonstrated significantly enhanced adhesion compared to homopolymers,a trend corroborated by macroscopic shear strength and stability tests. These results highlight that the synergistic effects of multiple functional groups are crucial for achieving universal interfacial adhesion of macromolecules, offering insights into protein adhesion mechanisms, and guiding polymer-based interfacial modifications.
