They are synthesized by the elongation of very long fatty acids-4(ELOVL4) enzyme, expressed mainly in the brain, retina, skin, testes, and meibomian gland, where these fatty acids are found(Agbaga et al., 2008). Further, these organs exhibit tissuespecific VLC-PUFA and VLC-SFA biosynthesis and incorporation into complex lipids for specific functions. In the brain, skin, and Meibomian glands, the ELOVL4 mainly makes VLC-SFA, which are incorporated into complex sphingolipids. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).展开更多

文摘Spinal cord injury represents a severe form of central nervous system trauma for which effective treatments remain limited.Microglia is the resident immune cells of the central nervous system, play a critical role in spinal cord injury.Previous studies have shown that microglia can promote neuronal survival by phagocytosing dead cells and debris and by releasing neuroprotective and anti-inflammatory factors. However, excessive activation of microglia can lead to persistent inflammation and contribute to the formation of glial scars, which hinder axonal regeneration.Despite this, the precise role and mechanisms of microglia during the acute phase of spinal cord injury remain controversial and poorly understood. To elucidate the role of microglia in spinal cord injury,we employed the colony-stimulating factor 1 receptor inhibitor PLX5622 to deplete microglia. We observed that sustained depletion of microglia resulted in an expansion of the lesion area, downregulation of brain-derived neurotrophic factor, and impaired functional recovery after spinal cord injury.Next,we generated a transgenic mouse line with conditional overexpression of brain-derived neurotrophic factor specifically in microglia. We found that brain-derived neurotrophic factor overexpression in microglia increased angiogenesis and blood flow following spinal cord injury and facilitated the recovery of hindlimb motor function.Additionally,brain-derived neurotrophic factor overexpression in microglia reduced inflammation and neuronal apoptosis during the acute phase of spinal cord injury.Furthermore, through using specific transgenic mouse lines,TMEM119,and the colony-stimulating factor 1 receptor inhibitor PLX73086,we demonstrated that the neuroprotective effects were predominantly due to brain-derived neurotrophic factor overexpression in microglia rather than macrophages. In conclusion, our findings suggest the critical role of microglia in the formation of protective glial scars.Depleting microglia is detrimental to recovery of spinal cord injury, whereas targeting brain-derived neurotrophic factor overexpression in microglia represents a promising and novel therapeutic strategy to enhance motor function recovery in patients with spinal cord injury.
文摘BACKGROUND Dry eye disease(DED)is a multifactorial ocular surface disorder with rising prevalence. It is closely related to systemic health and psychological factors, such as sleep and mood disorders, which significantly impact the quality of life of patients.AIM To explore the correlations between ocular surface function, sleep quality, and anxiety/depression in patients with DED.METHODS This was a cross-sectional investigative study that included 358 patients with DED between January 2022 and January 2025.Ocular surface was assessed using the ocular surface disease index(OSDI),tear film break-up time, fluorescein staining score, and Schirmer I test. The Pittsburgh Sleep Quality Index(PSQI),Self-Rating Anxiety Scale(SAS),and Self-Rating Depression Scale(SDS)were used to evaluate sleep quality and anxiety/depression levels.Correlation and linear regression analyses were used to explore the relationships.RESULTS The mean PSQI score of the patients was 9.94±2.18;the mean SAS score was 47.30±4.90,and the mean SDS score was 50.08±5.52.These suggested a prevalence of sleep and psychological abnormalities. There was a significant correlation between the indicators of ocular surface function(OSDI,tear film break-up time, fluorescein staining, and Schirmer I test)and PSQI,SAS,and SDS scores(P<0.05).Moreover,multiple regression revealed that age≥50 years(β=1.55,P=0.029),PSQI scores(β=0.58,P<0.001),SAS scores(β=0.17,P=0.017),and SDS scores(β=0.15,P=0.019)were independent predictors of the OSDI scores.CONCLUSION Ocular surface function in patients with DED is closely related to sleep quality and anxiety/depression,emphasizing the need for holistic clinical management.

文摘The organization of biological neuronal networks into functional modules has intrigued scientists and inspired engineers to develop artificial systems. These networks are characterized by two key properties.First, they exhibit dense interconnectivity(Braitenburg and Schüz,1998;Campagnola et al.,2022).The strength and probability of connectivity depend on cell type,inter-neuronal distance, and species.Still, every cortical neuron receives input from thousands of other neurons while transmitting output to a similar number of neurons.Second, communication between neurons occurs primarily via chemical or electrical synapses.
文摘Invasive as well as non-invasive neurotechnologies conceptualized to interface the central and peripheral nervous system have been probed for the past decades, which refer to electroencephalography, electrocorticography and microelectrode arrays. The challenges of these mentioned approaches are characterized by the bandwidth of the spatiotemporal resolution, which in turn is essential for large-area neuron recordings(Abiri et al.,2019).

文摘Diabetic retinopathy is a prominent cause of blindness in adults, with early retinal ganglion cell loss contributing to visual dysfunction or blindness. In the brain, defects inγ-aminobutyric acid synaptic transmission are associated with pathophysiological and neurodegenerative disorders, whereas glucagon-like peptide-1 has demonstrated neuroprotective effects. However,it is not yet clear whether diabetes causes alterations in inhibitory input to retinal ganglion cells and whether and how glucagon-like peptide-1 protects against neurodegeneration in the diabetic retina through regulating inhibitory synaptic transmission to retinal ganglion cells. In the present study,we used the patch-clamp technique to recordγ-aminobutyric acid subtype A receptor-mediated miniature inhibitory postsynaptic currents in retinal ganglion cells from streptozotocin-induced diabetes model rats.We found that early diabetes(4 weeks of hyperglycemia)decreased the frequency of GABAergic miniature inhibitory postsynaptic currents in retinal ganglion cells without altering their amplitude, suggesting a reduction in the spontaneous release ofγ-aminobutyric acid to retinal ganglion cells.Topical administration of glucagon-like peptide-1 eyedrops over a period of 2 weeks effectively countered the hyperglycemia-induced downregulation of GABAergic mIPSC frequency, subsequently enhancing the survival of retinal ganglion cells.Concurrently, the protective effects of glucagon-like peptide-1 on retinal ganglion cells in diabetic rats were eliminated by topical administration of exendin-9-39,a specific glucagon-like peptide-1 receptor antagonist,or SR95531,a specific antagonist of theγ-aminobutyric acid subtype A receptor.Furthermore, extracellular perfusion of glucagon-like peptide-1 was found to elevate the frequencies of GABAergic miniature inhibitory postsynaptic currents in both ON-and OFF-type retinal ganglion cells. This elevation was shown to be mediated by activation of the phosphatidylinositol-phospholipase C/inositol 1,4,5-trisphosphate receptor/Ca2+/protein kinase C signaling pathway downstream of glucagon-like peptide-1 receptor activation. Moreover, multielectrode array recordings revealed that glucagon-like peptide-1 functionally augmented the photoresponses of ON-type retinal ganglion cells.Optomotor response tests demonstrated that diabetic rats exhibited reductions in visual acuity and contrast sensitivity that were significantly ameliorated by topical administration of glucagon-like peptide-1.These results suggest that glucagon-like peptide-1 facilitates the release ofγ-aminobutyric acid onto retinal ganglion cells through the activation of glucagon-like peptide-1 receptor, leading to the de-excitation of retinal ganglion cell circuits and the inhibition of excitotoxic processes associated with diabetic retinopathy.Collectively, our findings indicate that theγ-aminobutyric acid system has potential as a therapeutic target for mitigating early-stage diabetic retinopathy.Furthermore, the topical administration of glucagon-like peptide-1 eyedrops represents a non-invasive and effective treatment approach for managing early-stage diabetic retinopathy.
文摘BACKGROUND At present, the influencing factors of social function in patients with residual depressive symptoms are still unclear.Residual depressive symptoms are highly harmful, leading to low mood in patients, affecting work and interpersonal communication, increasing the risk of recurrence, and adding to the burden on families.Studying the influencing factors of their social function is of great significance.AIM To explore the social function score and its influencing factors in patients with residual depressive symptoms.METHODS This observational study surveyed patients with residual depressive symptoms(case group)and healthy patients undergoing physical examinations(control group).Participants were admitted between January 2022 and December 2023.Social functioning was assessed using the Sheehan Disability Scale(SDS),and scores were compared between groups.Factors influencing SDS scores in patients with residual depressive symptoms were analyzed by applying multiple linear regression while using the receiver operating characteristic curve, and these RESULTS The SDS scores of the 158 patients with depressive symptoms were 11.48±3.26.Compared with the control group, the SDS scores and all items in the case group were higher.SDS scores were higher in patients with relapse,discon-tinuous medication, drug therapy alone, severe somatic symptoms, obvious residual symptoms, and anxiety scores≥8.Disease history, medication compliance, therapy method, and residual symptoms correlated positively with SDS scores(r=0.354,0.414,0.602,and 0.456,respectively).Independent influencing factors included disease history, medication compliance, therapy method, somatic symptoms, residual symptoms, and anxiety scores(P<0.05).The areas under the curve for predicting social functional impairment using these factors were 0.713,0.559,0.684,0.729,0.668,and 0.628,respectively,with sensitivities of 79.2%,61.8%,76.8%,81.7%,63.6%,and 65.5%and specificities of 83.3%,87.5%,82.6%,83.3%,86.7%,and 92.1%,respectively.CONCLUSION The social function scores of patients with residual symptoms of depression are high. They are affected by disease history, medication compliance, therapy method, degree of somatic symptoms, residual symptoms, and anxiety.

文摘Very long chain-saturated and-polyunsaturated fatty acids(VLC-SFA and VLC-PUFA, respectively) are a functionally important class of fatty acids containing 28 carbons or more in their acyl chain. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).
文摘Background Acute myocardial infarction(AMI),one of the most critical cardiovascular emergencies, carries a high mortality rate due to progressive cardiac dysfunction caused by myocardial ischemia.Percutaneous coronary intervention(PCI)is an effective treatment for AMI,rapidly restoring blood flow, reducing myocardial injury, and mitigating adverse remodeling. However, reperfusion injury and ventricular remodeling post-PCI may still lead to heart failure.Recent studies highlighted the cardioprotective effects of sodium-glucose cotransporter-2(SGLT-2)inhibitors,particularly dapagliflozin, which improves myocardial energetics, reduces inflammation, and attenuates adverse remodeling.However,high-quality evidence on its early application following primary PCI in AMI re-mains limited. In this study,we aimed to evaluated the clinical utility of dapagliflozin in AMI management.Meth-ods This study retrospectively analyzed the clinical data of 100 patients with AMI undergoing emergency PCI ad-mitted to our hospital between June 2022 and June 2024.They were randomly divided into observation group(n=50)and control group(n=50).The control group received standard post-PCI medication(antiplatelet and lipid-low-ering therapy),while the observation group received additional dapagliflozin.Cardiac function parameters,serolog-ical markers, and the incidence of major adverse cardiovascular events(MACE)were compared between the two groups. Results After 14 days of treatment, observation group exhibited significantly lower left ventricular end-diastolic diameter(LVEDD)and left ventricular end-systolic diameter(LVESD)(P<0.05)and higher left ventricu-lar ejection fraction(LVEF)(P<0.05)compared to control group.Additionally, the level of myocardial injury mark-ers[creatine kinase-MB(CK-MB),cardiac troponin I(cTnI),and brain natriuretic peptide(BNP)]were significant-ly lower in observation group(P<0.05).However,there was no significant difference in the incidence of MACE be-tween the two groups(P>0.05).Conclusions The combination of dapagliflozin with standard post-PCI therapy improves cardiac function and reduces the level of myocardial injury markers in AMI patients, though it does not significantly affect the short-term incidence of adverse cardiovascular events.

文摘Zinc-ion batteries(ZIBs)are inexpensive and safe, but side reactions on the Zn anode and Zn dendrite growth hinder their practical applications. In this study,1,3,5-triformylphloroglycerol(Tp)and various diamine monomers(p-phenylenediamine(Pa),benzidine(BD),and 4,4"-diamino-p-terphenyl(DATP))were used to synthesize a series of two-dimensional covalent-organic frameworks(COFs).The resulting COFs were named TpPa,TpBD,and TpDATP,respectively, and they showed uniform zincophilic sites, different pore sizes, and high Young's moduli on the Zn anode.Among them,TpPa and TpBD showed lower surface work functions and higher ion transfer numbers, which were conducive to uniform galvanizing/stripping zinc and inhibited dendrite growth.Theoretical calculations showed that TpPa and TpBD had wider negative potential region and greater adsorption capacity for Zn2+than TpDATP,providing more electron donor sites to coordinate with Zn^(2+).Symmetric cells protected by TpPa and TpBD stably cycled for more than 2300 h,whereas TpDATP@Zn and the bare zinc symmetric cells failed after around 150 and200 h.The full cells containing TpPa and TpBD modification layers also showed excellent cycling capacity at 1 A/g.This study provides comprehensive insights into the construction of highly reversible Zn anodes via COF modification layers for advanced rechargeable ZIBs.

文摘Hydrogel scaffolds have numerous potential applications in the tissue engineering field. However, tough hydrogel scaffolds implanted in vivo are seldom reported because it is difficult to balance biocompatibility and high mechanical properties.Inspired by Chinese ramen,we propose a universal fabricating method(printing-P,training-T,cross-linking-C,PTC&PCT)for tough hydrogel scaffolds to fill this gap.First,3D printing fabricates a hydrogel scaffold with desired structures(P).Then, the scaffold could have extraordinarily high mechanical properties and functional surface structure by cycle mechanical training with salting-out assistance(T).Finally, the training results are fixed by photo-cross-linking processing(C).The tough gelatin hydrogel scaffolds exhibit excellent tensile strength of 6.66 MPa(622-fold untreated)and have excellent biocompatibility.Furthermore, this scaffold possesses functional surface structures from nanometer to micron to millimeter, which can efficiently induce directional cell growth.Interestingly, this strategy can produce bionic human tissue with mechanical properties of 10 kPa-10 MPa by changing the type of salt, and many hydrogels, such as gelatin and silk, could be improved with PTC or PCT strategies.Animal experiments show that this scaffold can effectively promote the new generation of muscle fibers, blood vessels, and nerves within 4 weeks, prompting the rapid regeneration of large-volume muscle loss injuries.

文摘Fe-Mo functionally graded materials(FGMs)with different composition-change rates from 100%304 stainless steel to 100%Mo along the composition gradient direction were prepared by electron beam-directed energy deposition(EB-DED)technique,including three samples with composition mutation of 100%,composition change rate of 10%and 30%.Results show that the composition-change rate significantly affects the microstructure and mechanical properties of the samples. In the sample with abrupt change of composition, the sharp shift in composition between 304 stainless steel and Mo leads to a great difference in the microstructure and hardness near the interface between the two materials. With the increase in the number of gradient layers, the composition changes continuously along the direction of deposition height, and the microstructure morphology shows a smooth transition from 304 stainless steel to Mo,which is gradually transformed from columnar crystal to dendritic crystal.Elements Fe,Mo,and other major elements transform linearly along the gradient direction, with sufficient interlayer diffusion between the deposited layers, leading to good metallurgical bonding. The smaller the change in composition gradient, the greater the microhardness value along the deposition direction. When the composition gradient is 10%,the gradient layer exhibits higher hardness(940 HV)and excellent resistance to surface abrasion, and the overall compressive properties of the samples are better, with the compressive fracture stress in the top region reaching 750.05±14 MPa.
文摘Lithium-ion capacitors(LICs)combine the high power dens-ity of electrical double-layer capacitors with the high energy density of lithium-ion batteries. However, they face practical limitations due to the narrow operating voltage window of their activated carbon(AC)cathodes. We report a scalable thermal treatment strategy to develop high-voltage-tolerant AC cathodes.Through controlled thermal treatment of commer-cial activated carbon(Raw-AC)under a H_(2)/Ar atmosphere at 400-800℃,the targeted reduction of degradation-prone functional groups can be achieved while preserving the critical pore structure and increasing graph-itic microcrystalline ordering. The AC treated at 400℃(HAC-400)had a significant increase in specific capacity(96.0 vs.75.1 mAh/g at 0.05 A/g)and better rate capability(61.1 vs.36.1 mAh/g at 5 A/g)in half-cell LICs,along with an 83.5%capacity retention over 7400 cycles within an extended voltage range of 2.0-4.2 V in full-cell LICs.Scalability was demonstrated by a 120 g batch production, enabling fabrication of pouch-type LICs with commercial hard carbon anodes that delivered a higher energy density of 28.3 Wh/kg at 1 C,and a peak power density of 12.1 kW/kg compared to devices using raw AC.This simple,industry-compatible approach may be used for producing ad-vanced cathode materials for practical high-performance LICs.

文摘Emerging evidence of the beneficial effects of defatted rice bran(DFRB)on gut health has advanced the development of fermented defatted rice bran as a potential functional food. However, less is known about its effects and underlying mechanisms on gut health. In this study,a mouse model together with fecal microbiota transplantation(FMT)was utilized to study the effects and mechanisms of fermented DFRB(FR)on gut barrier function. We found that FR improved the intestinal morphology, gut tight junction proteins, mucin, antimicrobial peptides, and interleukin 22(IL-22)and promoted the gut Clostridium butyricum and butyrate.Notably, correlation analysis indicated gut C.butyricum and butyrate were two FR-induced effectors that improved gut health.FMT results suggested that C.butyricum, butyrate, and fecal microbiota from the FR group all reduced prolyl hydroxylase 2(PHD2)expression by activating peroxisome proliferator-activated receptor gamma(PPARγ)in the mouse colon. This decrease in gut PHD2 subsequently upregulated the hypoxia-inducible factor-1 alpha(HIF-1α)expression,which in turn increased the expression of its targeted downstream tight junction proteins, mucin and antimicrobial peptides, and colonic IL-22 secretion.Overall,FR-derived C.butyricum and butyrate might improve gut barrier function through the HIF-1 signaling pathway, which provides a reference for the application of fermented DFRB as a potential functional food for improving of gut barrier function.

文摘In distributed fusion, when one or more sensors are disturbed by faults,a common problem is that their local estimations are inconsistent with those of other fault-free sensors. Most of the existing fault-tolerant distributed fusion algorithms, such as the Covariance Union(CU)and Faulttolerant Generalized Convex Combination(FGCC),are only used for the point estimation case where local estimates and their associated error covariances are provided.A treatment with focus on the fault-tolerant distributed fusions of arbitrary local Probability Density Functions(PDFs)is lacking. For this problem,we first propose Kullback–Leibler Divergence(KLD)and reversed KLD induced functional Fuzzy c-Means(FCM)clustering algorithms to soft cluster all local PDFs,respectively. On this basis, two fault-tolerant distributed fusion algorithms of arbitrary local PDFs are then developed. They select the representing PDF of the cluster with the largest sum of memberships as the fused PDF.Numerical examples verify the better fault tolerance of the developed two distributed fusion algorithms.

文摘Magnetically responsive microstructured functional surface(MRMFS),capable of dynamically and reversibly switching the surface topography under magnetic actuation, provides a wireless, noninvasive, and instantaneous way to accurately control the microscale engineered surface. In the last decade, many studies have been conducted to design and optimize MRMFSs for diverse applications, and significant progress has been accomplished. This review comprehensively presents recent advancements and the potential prospects in MRMFSs.We first classify MRMFSs into one-dimensional linear array MRMFSs,two-dimensional planar array MRMFSs,and dynamic self-assembly MRMFSs based on their morphology.Subsequently,an overview of three deformation mechanisms, including magnetically actuated bending deformation, magnetically driven rotational deformation, and magnetically induced self-assembly deformation, are provided.Four main fabrication strategies employed to create MRMFSs are summarized, including replica molding,magnetization-induced self-assembly,laser cutting, and ferrofluid-infused method.Furthermore, the applications of MRMFS in droplet manipulation, solid transport, information encryption, light manipulation, triboelectric nanogenerators, and soft robotics are presented. Finally, the challenges that limit the practical applications of MRMFSs are discussed, and the future development of MRMFSs is proposed.

文摘Surface deformation calculations due to environmental loading typically rely on the Preliminary Reference Earth Model(PREM),which assumes a homogeneous and isotropic Earth structure, leading to noticeable errors. To enhance accuracy, the high-precision crustal model CRUST 1.0 is used to refine calculations of regional surface deformation caused by hydrological and non-tidal atmospheric loading. The improved model is applied to 27 Global Navigation Satellite System(GNSS)reference stations in the first phase of the Crustal Movement Observation Network of China(CMONOC),considering their geographical locations.Green's functions are employed to compute surface deformation at each site. Results indicate relative discrepancies of 11.78%and 14.14%for non-tidal atmospheric and hydrological loading compared to PREM,with vertical deformation differences reaching an average of 18.95%.Additionally,the distinct spatial distribution characteristics of the relative differences in each direction indicate that the improved RPREM model is more responsive to the mass variations derived from Gravity Recovery and Climate Experiment(GRACE).The results suggest that the improved PRREM model demonstrates higher sensitivity to loading variations than the PREM model.Utilizing the enhanced method of calculating surface deformation through the utilization of Green's function at the site could effectively reduce the calculation error caused by regional structure, leading to enhanced uniformity and isotropy of PREM.

文摘Gauss radial basis functions(GRBF)are frequently employed in data fitting and machine learning.Their linear independence property can theoretically guarantee the avoidance of data redundancy. In this paper, one of the main contributions is proving this property using linear algebra instead of profound knowledge. This makes it easy to read and understand this fundamental fact. The proof of linear independence of a set of Gauss functions relies on the constructing method for one-dimensional space and on the deducing method for higher dimensions.Additionally, under the condition of preserving the same moments between the original function and interpolating function, both the interpolating existence and uniqueness are proven for GRBF in one-dimensional space. The final work demonstrates the application of the GRBF method to locate lunar olivine.By combining preprocessed data using GRBF with the removing envelope curve method,a program is created to find the position of lunar olivine based on spectrum data, and the numerical experiment shows that it is an effective scheme.
