Despite demonstrating significant anti-tumor potential as an artemisinin derivative,artesunate faces delivery efficiency challenges due to low water solubility and insufficient targeting specificity.To improve the del...Despite demonstrating significant anti-tumor potential as an artemisinin derivative,artesunate faces delivery efficiency challenges due to low water solubility and insufficient targeting specificity.To improve the delivery efficiency,we engineered three artesunate(ART) derivatives,AC_(15)-L(linear),AC_(15)-B(branched),and AC_(15)-C(cyclic) with distinct aliphatic chain architectures.Unexpectedly,we observed that AC_(15)-C exhibited superior cytotoxicity against 4T1 breast cancer cells,and had the highest binding affinity for Lon protease 1(LONP1)(-72.6 kcal/mol).Subsequently,disulfide bond-containing lipid-PEG(DSPESS-PEG2K) modified chain architecture-engineered ART derivatives nanoassemblies(NAs) were developed to mitigate solubility-related limitations while enhancing targeting precision.Molecular docking and experimental validation demonstrated that ART derivatives inhibited LONP1 through hydrophobic interactions while preserved Fe^(2+)-mediated Fenton-like reaction activity.In vitro and in vivo evaluations demonstrated that AC_(15)-C NAs outperformed free ART and other NAs,suppressing 4T1 tumor growth via dual action:LONP1-directed mitochondrial proteostasis collapse and reactive oxygen species(ROS) amplification through Fe^(2+)-ART interactions.This study elucidated a novel anti-tumor mechanism of ART through the rational design of derivatives with spatially configured aliphatic chains,and developed reductionresponsive NAs to provide an advanced delivery strategy.展开更多
More than two decades ago, object-oriented representation of AEC (architecture engineering and construction) projects started to offer the promise of seamless communication of semantic data models between computer-b...More than two decades ago, object-oriented representation of AEC (architecture engineering and construction) projects started to offer the promise of seamless communication of semantic data models between computer-based systems used from the design stage to the operation of the facilities. BIM (building information modelling) emerged and appeared as a means to store all relevant data generated during the life-cycle of the facilities. But this upstream view of the built environment, arising from the design and construction stages, extended to the downstream operations where building and industrial facilities appeared more and more as huge dynamic data producers and concentrators while being operated. This created new challenges leading to what is referred to as ISCs (intelligent and smart constructions). The current state of the art is that final constructions still contain various and increasingly versatile control and service systems, which are hardly standardised, and not interconnected among themselves. Monitoring, maintenance and services are done by specialised companies, each responsible of different systems, which are relying on customised software and techniques to meet specific user needs and are based on monolithic applications that require manual configuration for specific uses, maintenance and support. We demonstrate in this paper that the early promises of integration across the actors and along the life-time of facilities have gone a long way but will only be delivered through enhanced standardisation of computerized models, representations, services and operations still not yet fully accomplished 25 years after work started.展开更多

Its trend is gradually changed from client to web, and so many people start to research the 3D graphics engine technology on the web. WebGL and HTML5 rise in recent years and WebGL solves two problems of interactive 3D application on the web perfectly. Firstly, it implements the interactive 3D web application by JavaScript without any browser plug-in components. Secondly, it makes graphics rendering using the underlying graphics hardware, which is united, standard and cross-platform OpenGL interface. However, it is very difficult for 3D application web programmer to understand multifarious details. Therefore, a 3D engine based on WebGL comes into being. HVAC engineering, as an important component of architecture, affects not only the comfort of houses but also the quality of our lives. Therefore, people must pay more attention to the design of HVAC engineering and give full consideration to heating, indoor air circulation and service life. As BIM technology plays a key role in HVAC engineering design and can greatly improve the scientific engineering design, BIM technology needs to be applied to heating and cooling resistance thermal engineering design, drawing, scheme aided design, computer aided design and other activities.

Therefore, the construction industry must pay more attention to the construction technology of deep foundation pit support, and use more scientific methods to improve this technology more effectively, so that this technology can be widely popularized and used in the construction industry. In addition, although the deep foundation pit support technology has developed well, there will inevitably be some problems in the development process.
