Effect of domestically-made levonorgestrel-releasing intrauterine device (LNG, release rate: 6 μg/day) on the endocrine system and menstruation in mondeys was investigated. The results showed that the Estradiol (E2) and progesterone (P) levels were significantly decreased 2 ovulatory cycles after iusertion of the devices as compared with those before insertion in 3 monkeys (P>0. 001) ,suggesting an evident ovulation-suppressing effect. In 1 monkey, the E2 and P levels were the same before and after insertion, and expulsion of device was found in the monkey later. The plasma LNG concentration in 3 animals was 493. 69-454. 60 pg/ml and plasma LNG was not detected in 1 monkey. The hormone level returned to normal level after removal of the devices. 展开更多

Many environmental contaminants could be transmitted fromparents and generate impairments to their progeny. The 2,4,6-tribromophenol(TBP),a novel brominated flame retardant which has been frequently detected in various organisms, was supposed to be bioaccumulated and intergenerational transmitted in human beings.Previous studies revealed that TBP could disrupt thyroid endocrine system in zebrafish larvae. However, there is no available data regarding the parental and transgenerational toxicity of this contaminant. Thus,in this study adult zebrafish were exposed to environmental contaminated levels of TBP for 60 days to investigate the parental and transgenerational impairments on thyroid endocrine system.Chemical analysis verified the bioaccumulation of TBP in tested organs of parents(concentration:liver>gonads>brain)and its transmission into eggs. For adults, increased thyroid hormones, disturbed transcriptions of related genes and histopathological changes in thyroid follicles indicate obvious thyroid endocrine disruptions.Transgenerational effects are indicated by the increased thyroid hormones both in eggs(maternal source)and in developed larvae(newly synthesized),aswell as disrupted transcriptional profiles of key genes in HPT axis. The overall results suggest that the accumulated TBP could be transmitted from parent to offspring and generate thyroid endocrine disruptions in both generations. 展开更多

Short-chain fatty acids, metabolites produced by the fermentation of dietary fiber by gut microbiota, have garnered significant attention due to their correlation with neurodegenerative diseases, particularly Parkinson’s disease. In this review,we summarize the changes in short-chain fatty acid levels and the abundance of short-chain fatty acid-producing bacteria in various samples from patients with Parkinson’s disease, highlighting the critical role of gut homeostasis imbalance in the pathogenesis and progression of the disease.Focusing on the nervous system,we discuss the molecular mechanisms by which short-chain fatty acids influence the homeostasis of both the enteric nervous system and the central nervous system. We identify key processes, including the activation of G protein-coupled receptors and the inhibition of histone deacetylases by short-chain fatty acids.Importantly, structural or functional disruptions in the enteric nervous system mediated by these fatty acids may lead to abnormalα-synuclein expression and gastrointestinal dysmotility, which could serve as an initiating event in Parkinson’s disease.Furthermore,we propose that short-chain fatty acids help establish communication between the enteric nervous system and the central nervous system via the vagal nerve, immune circulation, and endocrine signaling. This communication may shed light on their potential role in the transmission ofα-synuclein from the gut to the brain. Finally,we elucidate novel treatment strategies for Parkinson’s disease that target short-chain fatty acids and examine the challenges associated with translating short-chain fatty acid-based therapies into clinical practice. In conclusion, this review emphasizes the pivotal role of short-chain fatty acids in regulating gut-brain axis integrity and their significance in the pathogenesis of Parkinson’s disease from the perspective of the nervous system. Moreover,it highlights the potential value of short-chain fatty acids in early intervention for Parkinson’s disease.Future research into the molecular mechanisms of short-chain fatty acids and their synergistic interactions with other gut metabolites is likely to advance the clinical translation of innovative short-chain fatty acid-based therapies for Parkinson’s disease. 展开更多

BACKGROUND Mixed tumors of the colon and rectum, composed of a combination of epithelial and endocrine elements of benign and malignant potential are rare neoplasms. These can occur anywhere in the gastrointestinal tract and are often diagnosed incidentally.Though they have been a well-documented entity in the pancreas, where the exocrine-endocrine mixed tumors have been known for a while, recognition and accurate diagnosis of these tumors in the colon and rectum,to date, remains a challenge. This is further compounded by the different terminologies that have been attributed to these lesions over the years adding to increased confusion and misclassification. Therefore, dedicated literature reviews of these lesions in the colon and rectum are inconsistent and are predominantly limited to case reports and case series of limited case numbers.Though, most of these tumors are high grade and of advanced stage, intermediate and low grade lesions of these mixed tumors are also increasingly been reported. There are no established independent consensus based guidelines for the therapeutic patient management of these unique lesions.AIM To provide a comprehensive targeted literature review of these complex mixed tumors in the colon and rectum that chronicles the evolution over time with summarization of historical perspectives of terminology and to further our understanding regarding their pathogenesis including genomic landscape, clinicoradiological features, pathology, treatment, prognosis, the current status of the management of the primary lesions, their recurrences and metastases.METHODS A comprehensive review of the published English literature was conducted using the search engines PubMed,MEDLINE and GOOGLE scholar. The following search terms[“mixed tumors colon”OR mixed endocrine/neuroendocrine tumor/neoplasm/lesion colon OR adenocarcinoma and endocrine/neuroendocrine tumor colon OR mixed adenocarcinoma and endocrine/neuroendocrine carcinoma colon OR Amphicrine tumors OR Collision tumors]were used.Eligibility criteria were defined and all potential relevant items, including full articles and/or abstracts were independently reviewed, assessed and agreed upon items were selected for in-depth analysis.RESULTS In total 237 full articles/abstracts documents were considered for eligibility of which 45 articles were illegible resulting in a total of 192 articles that were assessed for eligibility of which 139 have been selected for reference in this current review. This seminal manuscript is a one stop article that provides a detailed outlook on the evolution over time with summarization of historical perspectives, nomenclature, clinicoradiological features, pathology, treatment, prognosis and the current status of the management of both the primary lesions, their recurrences and metastases.Gaps in knowledge have also been identified and discussed. An important outcome of this manuscript is the justified proposal for a new, simple, clinically relevant,non-ambiguous terminology for these lesions to be referred to as mixed epithelial endocrine neoplasms(MEENs).CONCLUSION MEEN of the colon and rectum are poorly understood rare entities that encompass an extensive range of heterogeneous tumors with a wide variety of combinations leading to tumors of high, intermediate or low grade malignant potential. This proposed new revised terminology of MEEN will solve the biggest hurdle of confusion and misclassification that plagues these rare unique colorectal neoplasms thus facilitating the future design of multi institutional prospective randomized controlled clinical trials to develop and evaluate newer therapeutic strategies that are recommended for continued improved understanding and personal optimization of clinical management of these unique colorectal neoplasms. 展开更多

Objective To measure the endocrine disrupting chemicals (EDCs) in wastewater and evaluate the EDCs removal efficiencies in the municipal wastewater treatment plants (WWTP). Methods A battery of in vitro recombinant yeast bioassays incorporated with exogenous metabolic activation system (rat liver preparation, S9 mix) was conducted to assess the estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), and thyroid receptor (TR) ant/agonistic activities of effluents collected from Datansha WWTP. Results The indirect estrogenic, anti‐androgenic, anti‐progesteronic, and anti‐thyroidic activities were observed in the influent. The removal efficiencies of EDCs were above 74%, suggesting that the present wastewater treatment processes were good enough to remove most of these indirect endocrine disrupting chemicals. Conclusion The incorporation of exogenous metabolic capacity into the test system was valid for the study of indirect effects on ER, AR, PR, and TR. 展开更多

Objective To evaluate the endocrine disrupting effects of cadmium (Cd) using OECD enhanced TG407 test guideline. Methods Sprague-Dawley (SD) rats were randomly divided into six groups and accordingly administered with 0, 1, 2.5, 5, 10, 20 mg/kg.BW/day of Cd by gavage for 28 days. Body weight, food consumption, hematology, biochemistry, sex hormone levels, urinary β2-microglobulin, organ weights and histopathology and estrous cycle were detected. Results Cd could significantly decrease animals' body weight (P〈0.05). Serum luteinizing hormone (LH) at 10-20 mg/kg.BW groups and testosterone (T) at 2.5 and 10 mg/kg.BW groups decreased significantly (P〈0.05). However, no statistically significant change was found in urinary β2-microglobulin among Cd-treatment groups (P〉0.05). Endpoints related to female reproduction including uterus weight and histopathological change at 10-20 mg/kg.BW groups showed significant increase (P〈0.05). While among male rats in 2.5, 10, 20 mg/kg.BW groups, weight of prostate, thyroids, and seminal vesicle glands significantly decreased (P〈0.05). Moreover, no histopathological change was observed in kidney. Conclusion Results suggested that Cd can cause endocrine disrupting effects in SD rats. Comparing with possible renal toxicity of Cd, its toxicity on endocrine system was more sensitive. 展开更多

Immune checkpoint inhibitors(ICIs),a novel anti-tumor therapeutic modality, are monoclonal antibodies targeting certain immune checkpoints(ICs)that reactivate T cells to achieve anti-tumor immunity by targeting, binding, and blocking ICs.Targeted inhibitory antibodies against the ICs cytotoxic T-lymphocyte antigen and programmed death receptor-1 have demonstrated efficacy and durable anti-tumor activity in patients with cancer.ICs may prevent autoimmune reactions. However,ICIs may disrupt ICs properties and trigger autoimmune-related adverse reactions involving various organ systems including the cardiovascular, pulmonary, gastrointestinal, renal, musculoskeletal, dermal, and endocrine systems.Approximately 10%of patients with damage to target organs such as the thyroid, pituitary, pancreas, and adrenal glands develop endocrine system immune-related adverse events(irAEs)such as thyroid dysfunction, pituitary gland inflammation, diabetes mellitus, and primary adrenal insufficiency. However, the symptoms of immunotherapy-associated endocrine system irAEs may be nonspecific and similar to those of other treatment-related adverse reactions, and failure to recognize them early may lead to death.Timely detection and treatment of immunotherapy-associated endocrine irAEs is essential to improve the efficacy of immunotherapy, prognosis, and the quality of life of patients. This study aimed to review the mechanisms by which ICIs cause endocrine irAEs providing guidance for the development of appropriate management protocols. Here,we discuss(1)the biological mechanisms of ICs in tumorigenesis and progression, focusing on cytotoxic T-lymphocyte antigen and programmed cell death-1/programmed cell death-ligand 1;and(2)the epidemiology, clinical symptoms, diagnosis, and treatment of four immunotherapy-related endocrine complications. 展开更多

The widespread use of agrochemicals and its possible negative impacts on humans and the environment remains a highly topical and controversial issue. Of particular concern here are chemicals that can disturb an organism's endocrine system. Referring to the precautionary principle, the objective of the research project start2 is to develop management strategies that help reduce possible risks for httmans and the environment of endocrine active agrochemicals. As an empirical basis we combined qualitative and quantitative methods and initiated two expert dialogues. Our findings underpin the well known fact that the use of agrochemicals is complex and dependent on more than the individual attitudes of farmers. Agricultural plant protection management is the collective product of a complex agricultural system, which encompasses treatment, marketing, industry and agricultural consultation as well as agricultural teaching institutions and relevant scientific disciplines. Successful management strategies for risk reduction in the context of hormonally active agrochemicals thus need to involve all actors within this system. The paper proposes a strategy that promotes participation by enabling learning processes among these actors. 展开更多

Inspired by the modulation mechanism of neuroendocrine-immune system(NEIs),a novel structure of artificial neural network(ANN) named NEI-NN and its learning method are presented. The NEI-NN includes two parts,i.e.,positive subnetwork(PSN) and negative sub-network(NSN).The neuron functions of PSN and NSN are designed according to the increased and decreased secretion functions of hormone, respectively. In order to make the novel neural network learn quickly, the novel neuron based on some characteristics of NEIs is also redesigned.Besides the normal input signals, two control signals are considered in the proposed solution. One is the enable/disable signal, and the other is the slope control signal. The former can modify the structure of NEI-NN,and the later can regulate the evolutionary speed of NEINN.The NEI-NN can obtain the optimized network structure by using error back-propagation(BP) learning algorithm.Since the modeling of the beam pumping unit is very difficult by using the conventional method, the modeling of bean bump unit is chosen to examine the performance of the NEI-NN.The experiment results show that the optimized structure and learning speed of NEI-NN are better than those of the conventional neural network. 展开更多

Suicide is defined as the act of a person attempting to take their own life by causing death.Suicide is a complex phenomenon that is influenced by a multitude of factors, including psychosocial, cultural, and religious aspects,as well as genetic, biochemical, and environmental factors. From a biochemical perspective,it is crucial to consider the communication between the endocrine, immune, and nervous systems when studying the etiology of suicide.Several pathologies involve the bidirectional communication between the peripheral activity and the central nervous system by the action of molecules such as cytokines, hormones, and neurotransmitters. These humoral signals, when present in optimal quantities, are responsible for maintaining physiological homeostasis, including mood states.Stress elevates the cortisol and proinflammatory cytokines levels and alter neurotransmitters balance, thereby increasing the risk of developing a psychiatric disorder and subsequently the risk of suicidal behavior. This review provides an integrative perspective about the neurochemical, immunological, and endocrinological disturbances associated with suicidal behavior, with a particular focus on those alterations that may serve as potential risk markers and/or indicators of the state preceding such a tragic act. 展开更多

Endocrine disorders frequently lead to metabolic disturbances that significantly affect liver function.Understanding the complex interplay between hormonal imbalances and liver dysfunction is essential for advancing targeted therapeutic strategies. This comprehensive review explores the pathophysiological mechanisms linking major endocrine disorders to liver disease, with a focus on the roles of the thyroid, parathyroid, pancreas, adrenal glands, and sex hormones.Thyroid dysfunction is associated with alterations in liver enzyme levels and metabolic regulation, often resulting in hepatic steatosis or cholestasis.Hyperparathyroidism and consequent hypercalcemia have been linked to hepatic calcifications.Insulin resistance, both hepatic and peripheral, contributes to excessive lipid accumulation in the liver, exacerbating steatotic changes.Adrenal gland disorders, particularly in the setting of chronic liver disease, impair cortisol metabolism and may worsen hepatic injury.Additionally, sex hormones such as estrogen and testosterone modulate the progression of liver fibrosis and influence the development of metabolic syndrome. The intricate relationship between endocrine and hepatic systems underscores the need for a multidisciplinary approach in the management of liver disease.Addressing underlying hormonal disturbances may enhance patient outcomes and prevent further hepatic deterioration.Future research should prioritize integrative therapeutic strategies that concurrently target endocrine and liver dysfunction. 展开更多

Transgender individuals often undergo gender-affirming hormonal therapy(GAHT)to align their physical characteristics with their gender identity, which introduces unique challenges in the management of critically ill patients. In the setting of critical illness, the interactions between GAHT and the body's endocrine response are complex.GAHT can influence the hypothalamic-pituitary-adrenal axis, sex hormone levels, and metabolic parameters, potentially complicating the clinical picture. For example, estrogen therapy in transgender women increases the risk of venous thromboembolism, which is further exacerbated by the immobility and hypercoagulable state often present in critically ill patients.Testosterone therapy in transgender men can lead to erythrocytosis, increasing the risk of thromboembolic events during critical illness. The potential for drug interactions, particularly with medications used in the intensive care unit, also requires careful consideration.Monitoring hormone levels and adjusting GAHT in the acute setting are crucial, although evidence-based guidelines are lacking. The need for individualized care and vigilant monitoring of endocrine and metabolic parameters is paramount to improve outcomes in this vulnerable population. 展开更多

Endocrine disorders are increasingly recognized as potentially reversible causes of secondary cardiomyopathies, yet they often remain underdiagnosed in clinical practice. These conditions-including thyroid dysfunction, acromegaly, pheochromocytoma, diabetes mellitus, adrenal disorders, among others-can significantly alter cardiac structure and function through hormonal excess, metabolic remodeling, and neurohumoral activation.Hyperthyroidism may lead to high-output heart failure(HF)and atrial fibrillation, while hypothyroidism is associated with diastolic dysfunction, pericardial effusion, and accelerated atherosclerosis.Acromegaly promotes biventricular hypertrophy and myocardial fibrosis via insulin-like growth factor 1 overproduction.Pheochromocytoma triggers catecholamine-induced cardiomyopathy, resembling Takotsubo syndrome and carrying a high risk of mortality if left untreated.Diabetes induces a distinct phenotype of cardiomyopathy, affecting both systolic and diastolic function through microvascular injury and oxidative stress.Recognizing these endocrine etiologies is crucial,as targeted hormonal therapies-such as antithyroid agents, somatostatin analogs,or adrenalectomy-can reverse or significantly mitigate cardiac dysfunction.Comprehensive endocrine screening in patients with unexplained cardiomyopathy is therefore essential. This review synthesizes current knowledge on the pathophysiological mechanisms, clinical manifestations, and therapeutic strategies for endocrine cardiomyopathies and proposes a diagnostic algorithm for early recognition.Emerging biomarkers, such as galectin-3 in diabetic heart disease, may further enhance diagnostic accuracy and risk stratification. The interplay between endocrine and cardiovascular systems offers a unique opportunity for early intervention, potentially preventing progression to irreversible HF. 展开更多

BACKGROUND Pancreas transplantation(PT)has emerged as a critical therapeutic intervention for patients with type 1 diabetes mellitus(T1DM).This procedure restores neuroendocrine communication, which is essential for optimal pancreatic function and insulin regulation. The recovery process involves multiple phases, including neural regeneration, revascularization, and the re-establishment of synaptic connections, all of which contribute to the restoration of both endocrine and neurological functions.AIM To systematically examine the mechanisms underlying neurological recovery following PT,to explore the role of endocrine factors in restoring neurofunctional integrity, and to evaluate the impact of immunosuppressive therapy on nerve regeneration and its clinical outcomes.METHODS A comprehensive literature search was conducted across international databases such as PubMed,Web of Science, and Cochrane Library to identify studies addressing PT,neurological recovery, and endocrine regulation.Inclusion criteria encompassed randomized controlled trials, cohort studies, and systematic reviews. The review focused on the neurogenic mechanisms activated post-transplantation,the effect of glycemic control on nerve repair, and the implications of immunosuppressive drugs on the process of neurological recovery.RESULTS A total of 211 articles were initially identified through the literature search across international databases such as PubMed,Web of Science, and Cochrane Library.Following a detailed evaluation and the application of inclusion and exclusion criteria,56 articles were further reviewed, and 8 were selected for the final analysis.Additionally,a comprehensive patent search yielded 168 patents, out of which 6 were selected for further examination. These sources, including both journal literature and patents, offer significant insights into the mechanisms of neurological recovery and endocrine function following PT,with an emphasis on nerve regeneration, glycemic control, and the impact of immunosuppressive therapy.CONCLUSION PT represents a promising intervention for restoring both endocrine and neurological functions in patients with T1DM.Glycemic control, neural regeneration, and the restoration of neuroendocrine signaling are key components of successful recovery. While the procedure yields substantial improvements in nerve function, challenges persist, particularly in patients with long-standing diabetes or severe neuropathy. The dual impact of immunosuppressive drugs on immune suppression and neurotoxicity necessitates careful management.Future research should focus on refining immunosuppressive protocols and exploring advanced therapeutic options, including stem cell-based interventions,to enhance neural regeneration and further improve clinical outcomes. 展开更多
